The suitability of differentGracilaria spp. and twoSargassum species for bioconversion to methane was determined through bioassays of methane yield.Gracilaria species and strains were excellent feedstocks for high methane yields, ranging from 0.28 to 0.40 m3 kg–1 volatile solids added. These yields ranged from 58 to 95% of theoretical stoichiometric yields. Methane yields were highly correlated with acid soluble carbohydrate components of theGracilaria spp. BothSargassum fluitans andS. pteropleuron were poor feedstocks, with methane yields ranging from 0.12 to 0.19 m3 kg–1 volatile solids added, 27 to 46% of theoretical stoichiometric yields, respectively. The various tissue types of theseSargassum species were also poor feedstocks for anaerobic digestion to methane. While there is no clear explanation for the low methane yields, the twoSargassum spp. appear to contain a high proportion of an insoluble, non-extractable component which may not be available as a substrate for bioconversion to methane.  相似文献

Summary Degradative changes in tissue during protoplast isolation were a contributing factor to low protoplast yields in the saltsensitive Grevillea arenaria (R. Brown) and the salt-tolerant Grevillea ilicifolia (R. Brown). Protein and malondialdehyde content decreased significantly during the protoplast isolation procedure. Acid and neutral proteases were identified, and high acid protease activities were correlated to low protoplast yields. Acid phosphatase, catalase, polyphenol oxidase and lipoxygenase activities increased in both Grevillea species with cell wall digestion. High activities of catalase and low levels of polyphenol oxidase were correlated with high protoplast yields. Levels of acid phosphatase and lipoxygenase were not good indicators of final protoplast yields. The addition of the anti-oxidant, reduced glutathione, and the acid protease inhibitor, pepstatin A, significantly increased protoplast yields. Strategies were identified to minimize deleterious degradative effects during the isolation of protoplasts, including strict pH control, testing a number of cell wall digestion enzymes, and the addition of anti-oxidative metabolites and protease inhibitors.  相似文献

Climate warming is projected to affect forest water yields but the effects are expected to vary. We investigated how forest type and age affect water yield resilience to climate warming. To answer this question, we examined the variability in historical water yields at long‐term experimental catchments across Canada and the United States over 5‐year cool and warm periods. Using the theoretical framework of the Budyko curve, we calculated the effects of climate warming on the annual partitioning of precipitation (P) into evapotranspiration (ET) and water yield. Deviation (d) was defined as a catchment's change in actual ET divided by P [AET/P; evaporative index (EI)] coincident with a shift from a cool to a warm period – a positive d indicates an upward shift in EI and smaller than expected water yields, and a negative d indicates a downward shift in EI and larger than expected water yields. Elasticity was defined as the ratio of interannual variation in potential ET divided by P (PET/P; dryness index) to interannual variation in the EI – high elasticity indicates low d despite large range in drying index (i.e., resilient water yields), low elasticity indicates high d despite small range in drying index (i.e., nonresilient water yields). Although the data needed to fully evaluate ecosystems based on these metrics are limited, we were able to identify some characteristics of response among forest types. Alpine sites showed the greatest sensitivity to climate warming with any warming leading to increased water yields. Conifer forests included catchments with lowest elasticity and stable to larger water yields. Deciduous forests included catchments with intermediate elasticity and stable to smaller water yields. Mixed coniferous/deciduous forests included catchments with highest elasticity and stable water yields. Forest type appeared to influence the resilience of catchment water yields to climate warming, with conifer and deciduous catchments more susceptible to climate warming than the more diverse mixed forest catchments.  相似文献

Growth yields were determined with Acetobacterium woodii strain NZva 16 on hydrogen and CO2, formate, methanol, vanillate, ferulate and fructose in mineral medium in the absence and presence of 0.05% yeast extract. Yeast extract was not essential for growth but enhanced growth yields by 25–100% depending on the substrate fermented. Comparison of yields on formate or methanol allowed calculation of an energy yield in the range of 1.5–2 mol ATP per mol acetate formed during homoacetate fermentation of A. woodii. In the presence of 6 mM caffeate, growth yields were determined with the substrates formate or methanol. Caffeate was reduced to hydrocaffeate and increased growth yields were obtained. An ATP yield of about 1 mol per mol of caffeate reduced was calculated. Cytochromes were not detectable in cell free extracts or membrane preparations.  相似文献

Trees constitute promising renewable feedstocks for biorefinery using biochemical conversion, but their recalcitrance restricts their attractiveness for the industry. To obtain trees with reduced recalcitrance, large-scale genetic engineering experiments were performed in hybrid aspen blindly targeting genes expressed during wood formation and 32 lines representing seven constructs were selected for characterization in the field. Here we report phenotypes of five-year old trees considering 49 traits related to growth and wood properties. The best performing construct considering growth and glucose yield in saccharification with acid pretreatment had suppressed expression of the gene encoding an uncharacterized 2-oxoglutarate-dependent dioxygenase (2OGD). It showed minor changes in wood chemistry but increased nanoporosity and glucose conversion. Suppressed levels of SUCROSE SYNTHASE, (SuSy), CINNAMATE 4-HYDROXYLASE (C4H) and increased levels of GTPase activating protein for ADP-ribosylation factor ZAC led to significant growth reductions and anatomical abnormalities. However, C4H and SuSy constructs greatly improved glucose yields in saccharification without and with pretreatment, respectively. Traits associated with high glucose yields were different for saccharification with and without pretreatment. While carbohydrates, phenolics and tension wood contents positively impacted the yields without pretreatment and growth, lignin content and S/G ratio were negative factors, the yields with pretreatment positively correlated with S lignin and negatively with carbohydrate contents. The genotypes with high glucose yields had increased nanoporosity and mGlcA/Xyl ratio, and some had shorter polymers extractable with subcritical water compared to wild-type. The pilot-scale industrial-like pretreatment of best-performing 2OGD construct confirmed its superior sugar yields, supporting our strategy.  相似文献

Protoplasts were isolated from root cortical explants of thirteen cultivars of Pisum sativum. The roots were tested for starch in the cortical tissues. There was a considerable range in the average protoplast yields and the coefficient of variation of those yields from the thirteen cultivars. Cultivars with a higher incidence of starchy roots had lower average explam protoplast yields and greater variation in their protoplast yields. Significantly higher protoplast yields were observed from explants with no starch or few starchy cells. Increasing the germination time of the peas increased the starch content in the seedling roots and decreased the protoplast yields.  相似文献

In glasshouse experiments with low levels of soil applied phosphorus (P), yields of four annual pasture legumes (Medicago polymorpha, Trifolium subterraneum, T. balansae, Ornithopus compressus) increased with increasing P concentration in the seed. In a further experiment, M. polymorpha cv. Serena was grown at the same plant density from seed of two P concentrations and two seed sizes when two levels of finely ground superphosphate were applied to the soil. Higher P concentrations in the seed increased yields of dried tops by about 30% for the first harvest (21 days), 20% for the second harvest (52 days), and 9% at maturity (103 days), and seed yields by 11%. Larger seeds increased yields of dried tops by between 6–46% for the first two harvests but at maturity yields of dried tops and seed were unaffected by seed size. None of the interactions were statistically significant (P>0.05), except for the first harvest when two interactions (P concentration in the seed × seed size (i.e. P content in seed), and P applied to the soil × P concentration in the seed × seed size) were significant at P<0.05 level. In a field experiment, Trifolium subterraneum clover seed (two cultivars) of the same size but with two different P concentrations was sown at the same plant density and two levels of granulated (0.2–5 mm) superphosphate were applied to the soil surface. The higher level of superphosphate increased dried herbage yields of the dense clover swards by three- to four-fold 90 and 120 days after sowing. The higher P concentration in the seed increased yields of dried herbage by between 50 to 25%, depending on the level of P applied to the soil and the harvest date.  相似文献

Summary Penicillium vermiculatum deglycosylated lanatosid C to acetyldigoxin and Arthrobacter citreus deacetylated the latter to digoxin. Both reactions were carried out in a short time with high yields. Lanatosid A was transformed by P. vermiculatum simultaneously to acetyldigitoxin and digitoxin. A. citreus transformed lanatosid A directly to digitoxin and lanatosid C to digoxin, but the reactions took a long time and the yields were low.  相似文献

Summary Aspergillus niger was grown in batch culture containing various initial concentrations of sodium phosphate buffer (pH 6.5). A wild-type strain of A. niger and a transformed strain producing hen egg-white lysozyme were studied. The maximum cell yield was attained in medium not supplemented with phosphate. In those cultures acidification of the medium resulted in a minimum of pH 2.0 before reverting to near neutrality. Increasing the initial levels of phosphate buffer reduced the fall in pH but lowered cell yields. Secreted levels of lysozyme were maximal in the 50–100 mm range of added phosphate buffer although mycelial yields were reduced by one third of mycelial yields in medium unsupplemented with phosphate buffer. Cellulose content as well as alpha (α) and beta (β) celluloses were evaluated in 21 seaweed species belonging to different classes growing in Indian waters. The greatest yields of cellulose (crude) and β-cellulose were obtained from Caulerpa taxifolia (approx. 11.0% and 5.2%, respectively), whilst α-cellulose (approx. 8.2%) was the greatest in Padina tetrastromatica. The lowest cellulose (crude), α- and β- contents were recorded from the calcareous red alga Liagora ceranoides (approx. 0.85%, 0.62% and 0.18%, respectively). There was no variation in the yields of cellulose in the brown algae, whilst wide variations in the yields were found in the green and red algae.  相似文献

Plots in a field infested with Globodera rostochiensis and Verticillium dahliae were treated in 1971 with methyl bromide, dazomet, aldicarb, benomyl or aldicarb + benomyl and potatoes (cv. Pentland Dell) were grown in four consecutive years. In 1971, all treatments delayed the development of haulm symptoms, decreased soil populations of G. rostochiensis and increased yields. In the second year yields were increased by all treatments except dazomet whereas in the third and fourth years only benomyl or aldicarb + benomyl increased yields. Soil populations of V. dahliae were decreased by methyl bromide in 1971 but not by other treatments or in later years. In 1976, Pentland Dell and Pentland Crown were grown on an infested field in plots fumigated with methyl bromide. Fumigation slightly delayed shoot emergence but increased plant height, ground cover and the size and persistence of the leaves; development of haulm symptoms was delayed and yield increased. Fumigating with methyl bromide at five times the normal rate was more beneficial to Pentland Dell than to Pentland Crown. In a comparison of 10 cultivars in 1975, symptoms developed sooner and yields were smaller on a field infested with G. rostochiensis and V. dahliae than on a non-infested field and on both sites Maris Peer plants died soonest and yielded least. Pentland Crown yielded most on the non-infested field and Maris Piper on the infested field.  相似文献

Mathematical methods of experimental design were used to determine the optimal concentrations of nutrient medium components, aeration conditions, and pH providing for the maximum biomass yields, as well as fumarase and aspartase activities, during submerged cultivation ofErwinia sp. The data showed that different concentrations of carbon source (molasses) and pH of the nutrient medium were required to reach the maximum yields of fumarase and aspartase. Calculations suggested that the combination of these optimized factors would result in 3.2-, 3.4-, and 3.8-fold increases in theErwinia sp. biomass, aspartase activity, and fumarase activity yields, respectively. The experimental data were consistent with these estimates to 80% accuracy.  相似文献

In a field experiment on infested sandland from 1955 to 1967, numbers of Heterodera avenae increased only in 1957 and then decreased under both continuous and rotational spring oats (1955-62) and subsequently under spring barley (1963-67). Fluctuations in numbers of Pratylenchus neglectus and Trichodorus primitivus were recorded from 1963 to 1967. Cereal grain yields were generally unsatisfactory, but were consistently higher for cereals grown in rotation rather than continuously. Extra nitrogen fertilizer gave slightly improved yields with higher post-cropping numbers of H. avenae.  相似文献

Desulfotomaculum orientis (strain Singapore 1) was grown autotrophically with H2+CO2 and sulfate, thiosulfate or sulfite as electron acceptor in sulfide- and pH-controlled continuous culture. Under sulfate-limiting conditions real growth yields of up to 9.7 g cell dry mass per mol sulfate were obtained. Electron acceptor limitation resulted in the excretion of up to 14.5 mmol acetate per liter, formed by reduction of CO2 with H2. Acetate production was not coupled to an increase of growth yields: under hydrogen-limiting conditions only 1.6 mmol acetate per liter was produced, and even higher growth yields of up to 12,4 g cell dry mass per mol sulfate were obtained. With thiosulfate or sulfite as electron acceptor growth yields increased up to 17.9 g cell dry mass per mol electron acceptor. Growth yields were not simply correlated with the growth rate, and did not allow the determination of maintenance coefficients and the extrapolation to maximal yields at infinite growth rate (Y max). The maximal growth rates (max) with sulfate and thiosulfate were 0.090 and 0.109 h-1, respectively, if cells were grown continuously in sulfidostat culture under nonlimiting conditions.The net energy yield of sulfate reduction and the energy requirement for the activation of sulfate by Desulfotomaculum orientis are discussed.  相似文献

Alcohol yields of 6.5% were obtained with Saccharomyces cerevisiae in lactasehydrolyzed acid whey permeate containing 30–35% total solids. Maximum alcohol yields obtained with Kluyveromyces fragilis were 4.5% in lactase-hydrolyzed acid whey permeate at a solids concentration of 20% and 3.7% in normal permeate at a solids concentration of 10%. Saccharomyces cerevisiae efficiently converted the glucose present in lactase-hydrolyzed whey permeates containing 5–30% total solids (2–13% glucose) to alcohol. However, the galactose, which comprised about half the available carbohydrate in lactase-hydrolyzed whey, was not utilized by S. cerevisiae, so that even though alcohol yields were higher when this organism was used, the process was wasteful in that a substantial proportion of the substrate was not fermented. For the process to become commercially feasible, an efficient means of rapidly converting both the galactose and glucose to alcohol must be found.  相似文献

Preparation of o-palmitoyl alkyl lactates with methyl, ethyl, propyl, isopropyl and butyl lactates were attempted in a complex esterification reaction using lipases as catalysts. Compared to lactic acid, alkyl lactates were found to be less inhibitory in nature as they resulted in slightly better yields at shake-flask level. Of the alkyl lactates tested, butyl lactate showed better esterification. Porcine pancreas lipase gave higher yields of esters than Rhizomucor miehei lipase (Lipozyme IM20).  相似文献

Spore yields were measured for various fungal entomopathogens grown in six nutritionally different liquid media with low and high carbon concentrations (8 and 36 g l–1, respectively) at carbon-to-nitrogen (C:N) ratios of 10:1, 30:1 and 50:1. Six fungi were tested: two Beauveria bassiana strains, three Paecilomyces fumosoroseus strains and one Metarhizium anisopliae strain. Spore yields were examined after 2, 4 or 7 days growth. In general, highest spore yields were obtained in media containing 36 g/l and a C:N ratio of 10:1. After 4 days growth, highest spore yields were measured in the three Paecilomyces isolates (6.9–9.7 × 108 spores ml–1). Spore production by the B. bassiana isolates was variable with one isolate producing high spore yields (12.2 × 108 spores ml–1) after 7 days growth. The M. anisopliae isolate produced low spore concentrations under all conditions tested. Using a commercial production protocol, a comparison of spore yields for the coffee berry borer P. fumosoroseus and a commercial B. bassiana isolate showed that highest spore concentrations (7.2 × 108 spores ml–1) were obtained with the P. fumosoroseus isolate 2-days post-inoculation. The ability of the P. fumosoroseus strain isolated from the coffee berry borer to rapidly produce high concentrations of spores prompted further testing to determine the desiccation tolerance of these spores. Desiccation studies showed that ca. 80% of the liquid culture produced P. fumosoroseus spores survived the air-drying process. The virulence of freshly produced, air-dried and freeze-dried coffee berry borer P. fumosoroseus blastospores preparations were tested against silverleaf whiteflies (Bemisia argentifolii). While all preparations infected and killed B. argentifolii, fresh and air-dried preparations were significantly more effective. These results suggest that screening potential fungal biopesticides for amenability to liquid culture spore production can aid in the identification of commercially viable isolates. In this study, P. fumosoroseus was shown to possess the production and stabilization attributes required for commercial development.  相似文献

Long-term application of sewage sludge resulted in soil cadmium (Cd) and zinc (Zn) contamination in a pot experiment conducted to phytoextract Cd/Zn repeatedly using Sedum plumbizincicola and Apium graceolens in monoculture or intercropping mode eight times. Shoot yields and soil physicochemical properties changed markedly with increasing number of remediation crops when the two plant species were intercropped compared with the unplanted control soil and the two monoculture treatments. Changes in soil microbial indices such as average well colour development, soil enzyme activity and soil microbial counts were also significantly affected by the growth of the remediation plants, especially intercropping with S. plumbizincicola and A. graveolens. The higher yields and amounts of Cd taken up indicated that intercropping of the hyperaccumulator and the vegetable species may be suitable for simultaneous agricultural production and soil remediation, with larger crop yields and higher phytoremediation efficiencies than under monoculture conditions.  相似文献

Bamboo and Miscanthus species are perennial low-input plants that are excellent candidates for bioenergy feedstock production. Biological characteristics, dry matter yields and fuel properties of the bamboo and Miscanthus have been studied. Genotype growth characteristics were determined by measurements of plant height, tillering, tuft diameter, and shoot diameter. To date, comparisons of biomass yields of bamboo and Miscanthus have not been previously reported in the literature. Bamboo and Miscanthus species were collected and previous articles describing the productivity of bamboo and Miscanthus were examined. Genotypes differed in plant height, tillering, tuft diameter, and shoot diameter. Nitrogen, temperature, water and plant density have effects on mature stands biomass production, which ranged from 5.9 to 49.5 tonnes/ha/yr for bamboo and 3.2 to 49.0 tonnes/ha/yr for Miscanthus. With such biomass yields, bamboo and Miscanthus should be considered as two very promising plants for biomass production in Zhejiang, China in the near future.  相似文献

The effect of inoculation of potatoes with isolates ofPseudomonas putida andTrichoderma sp. suspension and in preparations with kaolin and peat carriers on growth of potatoes, yields and physiological proporties of tubers was investigated on seven locations of the potato-rye and sugar-beet regions. In 57.5% of experiments increased yields could be detected. More favorable results were obtained in the sugar-beet region and on commercially utilized fields when using suspensions of the isolates. Variability of the results can be partially explained by soil-climatic factors affecting colonization of the rhizosphere by the microorganisms and their effect on the environment. Physiological properties and the quality of the harvested tubers were comparable with those of the control. The results are discussed from the point of view of a possible inoculation of potatoes with rhizobacteria and, in the presence ofTrichoderma sp., improvement of their growth and yields.  相似文献
