Catecholamines extracted from tissues are readily measured by fluorescence following trihydroxyindole derivatization. A complicating factor however, has been the variably high background fluorescence levels obtained which adversely affect the precision of the assay. Employing EDTA is shown to reduce the high background fluorescence to a consistently low level, and to improve the accuracy of the trihydroxyindole method. It is suggested that high background fluorescence readings obtained by previous workers were due to metal ions extracted from tissue and carried over into the derivatization procedure.Measurement of tissue catecholamines by fluorescent methods is improved by the addition of EDTA which gives a consistently low background with a concommitant increase in accuracy.  相似文献

Detection of targets that move within visual clutter is a common task for animals searching for prey or conspecifics, a task made even more difficult when a moving pursuer needs to analyze targets against the motion of background texture (clutter). Despite the limited optical acuity of the compound eye of insects, this challenging task seems to have been solved by their tiny visual system. Here we describe neurons found in the male hoverfly,Eristalis tenax, that respond selectively to small moving targets. Although many of these target neurons are inhibited by the motion of a background pattern, others respond to target motion within the receptive field under a surprisingly large range of background motion stimuli. Some neurons respond whether or not there is a speed differential between target and background. Analysis of responses to very small targets (smaller than the size of the visual field of single photoreceptors) or those targets with reduced contrast shows that these neurons have extraordinarily high contrast sensitivity. Our data suggest that rejection of background motion may result from extreme selectivity for small targets contrasting against local patches of the background, combined with this high sensitivity, such that background patterns rarely contain features that satisfactorily drive the neuron.  相似文献

Due to relatively high rates of strongly selected deleterious mutations, directional selection on favorable alleles (causing hitchhiking effects on linked neutral polymorphisms) is expected to occur while a deleterious mutation-selection balance is present in a population. We analyze this interaction of directional selection and background selection and study their combined effects on neutral variation, using a three-locus model in which each locus is subjected to either deleterious, favorable, or neutral mutations. Average heterozygosity is measured by simulations (1) at the stationary state under the assumption of recurrent hitchhiking events and (2) as a transient level after a single hitchhiking event. The simulation results are compared to theoretical predictions. It is shown that known analytical solutions describing the hitchhiking effect without background selection can be modified such that they accurately predict the joint effects of hitchhiking and background on linked, neutral variation. Generalization of these results to a more appropriate multilocus model (such that background selection can occur at multiple sites) suggests that, in regions of very low recombination rates, stationary levels of nucleotide diversity are primarily determined by hitchhiking, whereas in regions of high recombination, background selection is the dominant force. The implications of these results on the identification and estimation of the relevant parameters of the model are discussed.  相似文献

The Optical Plankton Counter (OPC) has been used in oceanicand fresh waters to estimate zooplankton abundance and biovolume.However, it is not clear whether the OPC can produce accurateestimates of zooplankton abundance and biovolume in waters withhigh detritus. In order to test the capability of the OPC toestimate zooplankton abundance and biovolume in Chesapeake Bay,two sets of laboratory experiments were conducted using waterwith high detritus concentrations collected from the upper Choptankestuary of Chesapeake Bay and laboratory cultured Artemia. Ourresults suggest that the OPC is able to produce accurate estimatesof zooplankton biovolume after correcting for the influenceof background detritus in all the detritus concentrations used,but accurate estimates of zooplankton abundance only in waterwith background detritus <100 particles l–1. The relationshipbetween light attenuation and OPC background particle concentrationsprovides a useful way to estimate OPC background particle concentrationswhen direct OPC background particle measurements are not available.Light attenuation corrected OPC particle abundance and particlevolume gave accurate estimates of zooplankton abundance andbiovolume. However, the accuracy of the corrected OPC measurementsby the estimated background particle concentrations was notas high as the corrected OPC measurements by the direct backgroundparticle measurements.  相似文献

Although the identification of specific genes that regulate apoptosis has been a topic of intense study, little is known of the role that background genetic variance plays in modulating cell death. Using germ cells from inbred mouse strains, we found that apoptosis in mature (metaphase II) oocytes is affected by genetic background through at least two different mechanisms. The first, manifested in AKR/J mice, results in genomic instability. This is reflected by numerous DNA double-strand breaks in freshly isolated oocytes, causing a high apoptosis susceptibility and impaired embryonic development following fertilization. Microinjection of Rad51 reduces DNA damage, suppresses apoptosis and improves embryonic development. The second, manifested in FVB mice, results in dramatic dimorphisms in mitochondrial ultrastructure. This is correlated with cytochrome c release and a high apoptosis susceptibility, the latter of which is suppressed by pyruvate treatment, Smac/DIABLO deficiency, or microinjection of 'normal' mitochondria. Therefore, background genetic variance can profoundly affect apoptosis in female germ cells by disrupting both genomic DNA and mitochondrial integrity.  相似文献

Summary The pars intermedia of Sarotherodon mossambicus (Tilapia mossambica) contains two cell types which can be differentiated at both the light and electron microscopic level. The predominant cell type is lead haematoxyline positive, and has been shown to be the MSH producing cell type by means of immunocytochemical staining at the ultrastructural level. The changes in cellular and nuclear volume, as well as the results of stereological measurements on the cytoplasmic organelles, show that the activity of MSH cells is high on a black background and low on a white background or in total darkness. In blinded fish under a normal day-night regime the activity of the MSH cell is as high as that in black adapted fish, whereas the activity is low when the blinded fish are kept in total darkness. From the observed differences in activity of the MSH cells between the experimental groups, it is concluded that the MSH cells are not activated by the absence of reflected light, but by a high ratio between direct and reflected light. A second light-sensitive organ, supposedly the pineal gland, is also involved in the background response of the MSH producing cells.  相似文献

It is demonstrated that the structured coalescent model can readily be extended to include phenomena such as partial selfing and background selection through the use of an approximation based on separation of time scales. A model that includes these phenomena, as well as geographic subdivision and linkage to a polymorphism maintained either by local adaptation or by balancing selection, is derived, and the expected coalescence time for a pair of genes is calculated. It is found that background selection reduces coalescence times within subpopulations and allelic classes, leading to a high degree of apparent differentiation. Extremely high levels of subpopulation differentiation are also expected for regions of the genome surrounding loci important in local adaptation. These regions will be wider the stronger the local selection, and the higher the selfing rate.  相似文献

The interaction of background disease processes with environmental induced diseases has long been an issue of considerable interest and debate with respect to its impact on risk assessment. Whether and to what extent these processes should be considered independent or additive to background has been the principal focus of debate. The concept of hormesis, a biphasic dose response characterized by a low dose stimulation and a high dose inhibition, as framed within the context of post-conditioning, reveal the occurrence of a third type of “background” possibility, that of “subtraction to background”. This novel application of the hormesis concept, which is framed within the biological context of post-conditioning adaptive processes, offers considerable implications for the assessment of aging and environmental risk assessment.  相似文献

The impact of multilocus inheritance on the power of candidate gene association studies and the parameters derived therefrom is considered. Both case-control and family-based control designs are included. When the background heritability H (i.e., residual correlation) among sibs for the disease in question is high, the power of multiplex affected sibships versus singletons is diminished compared with the situation when background heritability is low. Thus, multiplex families for association studies are most advantageous for low heritability disorders. Estimates of genotype relative risk (GRR) are also distorted in multiplex sibships compared with singletons based on background heritability. In a case-control study, GRR is inflated in multiplex families, with the degree of inflation decreasing with background heritability. By contrast, in a TDT (parental control) study, GRR is deflated in multiplex families, with the degree of deflation increasing with background heritability (and no deflation when H is 0). Considering identical twins, the difference in genotype frequencies between concordant versus discordant MZ twin pairs for the candidate gene is large when the heritability is low, but modest when the heritability is high.  相似文献

The relationship between noise and human performance is a crucial topic in ergonomic research. However, the brain dynamics of the emotional arousal effects of background noises are still unclear. The current study employed meaningless speech noises in the n-back working memory task to explore the changes of event-related potentials (ERPs) elicited by the noises with low arousal level vs. high arousal level. We found that the memory performance in low arousal condition were improved compared with the silent and the high arousal conditions; participants responded more quickly and had larger P2 and P3 amplitudes in low arousal condition while the performance and ERP components showed no significant difference between high arousal and silent conditions. These findings suggested that the emotional arousal dimension of background noises had a significant influence on human working memory performance, and that this effect was independent of the acoustic characteristics of noises (e.g., intensity) and the meaning of speech materials. The current findings improve our understanding of background noise effects on human performance and lay the ground for the investigation of patients with attention deficits.  相似文献

The non-random distribution of DNA breakage in pulsed-field gel electrophoresis (PFGE) experiments poses a problem of proper subtraction of the background damage to obtain a fragment-size distribution due to radiation only. As been pointed out by various authors, a naive bin-to-bin subtraction of the background signal will not result in the right DNA mass distribution histogram, and may even result in negative values. Previous more systematic subtraction methods have been based mainly on random breakage, appropriate for low-LET radiation but problematic for high LET. Moreover, an investigation is needed whether the background breakage itself is random or non-random. Previously a new generalized formalism based on stochastic processes for the subtraction of the background damage in PFGE experiments for any LET and any background was proposed, and as now applied it to a set of PFGE data for Fe ions. We developed a Monte Carlo algorithm to compare the naïve subtraction procedure in artificial data sets to the result produced by the new formalism. The simulated data corresponded to various cases, involving non-random (high-LET) or random radiation breakage and random or non-random background breakage. The formalism systematically gives better results than naïve bin-by-bin subtraction in all these artificial data sets.  相似文献

Exposure to elevated levels of background predation risk is known to shape the behavioural response of prey organisms to known and unknown predation threats. However, less is known regarding the effects of background predation risk on predator recognition learning. Here, we test the potential effects of elevated background predation risk on the strength and retention of learned predator recognition in juvenile convict cichlids (Amatitlania nigrofasciata). In a series of laboratory trials, we exposed shoals of juvenile cichlids to conditions of elevated (vs. low) levels of background risk and then conditioned them to recognize a novel predator odour (rainbow trout, Oncorhynchus mykiss). The results of our first experiment demonstrate that despite showing reduced response intensities during initial conditioning (due to risk allocation), conditioned cichlids from high vs. low background risk show similar intensities of learned recognition when tested 24 h post‐conditioning. Moreover, elevated levels of background risk induced a predator avoidance response among unconditioned cichlids (due to induced neophobia). Our second experiment demonstrates that while we find no difference in the strength of learning when tested 24 h post‐conditioning, retention of acquired recognition is enhanced among cichlids from the high background predation risk treatment. Together, our results highlight the complex interacting effects past experience plays in shaping the response to acute predation threats.  相似文献

Various strains of mouse hepatitis virus (MHV) exhibit different pathogenic phenotypes. Infection with the A59 strain of MHV induces both encephalitis and hepatitis, while the highly neurovirulent JHM strain induces a fatal encephalitis with little, if any, hepatitis. The pathogenic phenotype for each strain is determined by the genetic composition of the viral genome, as well as the host immune response. Using isogenic recombinant viruses with A59 background genes differing only in the spike gene, we have previously shown that high neurovirulence is associated with the JHM spike protein, the protein responsible for attachment to the host cell receptor (J. J. Phillips, M. M. Chua, G. F. Rall, and S. R. Weiss, Virology 301:109-120, 2002). Using another set of isogenic recombinant viruses with JHM background genes expressing either the JHM or A59 spike, we have further investigated the roles of viral genes in pathogenesis. Here, we demonstrate that the high neurovirulence of JHM is associated with accelerated spread through the brain and a heightened innate immune response that is characterized by high numbers of infiltrating neutrophils and macrophages, suggesting an immunopathogenic component to neurovirulence. While expression of the JHM spike is sufficient to confer a neurovirulent phenotype, as well as increased macrophage infiltration, background genes contribute to virulence as well, at least in part, by dictating the extent of the T-cell immune response.  相似文献

The modern research in biophysics proved that water is a most essential target, through which the biological effects of ionizing and non-ionizing radiations are realized. Therefore, there is no doubt about the strong dependency of non-ionizing radiation-induced effect on the level of background radiation. The noisy threshold regime, where even a small set of presynaptic neurons can significantly affect postsynaptic spike-timing, is suggested as a key requisite for computation in neurons with high variability. It also has been proposed that signals under the noisy conditions are successfully transferred by a few strong synapses and/or by an assembly of nearly synchronous synaptic activities. We analytically investigate the impact of a transient signaling input on a leaky integrate-and-fire postsynaptic neuron that receives background noise near the threshold regime. The signaling input models a single strong synapse or a set of synchronous synapses, while the background noise represents a lot of weak synapses. We find an analytic solution that explains how the first-passage time (ISI) density is changed by transient signaling input. The analysis allows us to connect properties of the signaling input like spike timing and amplitude with postsynaptic first-passage time density in a noisy environment. Based on the analytic solution, we calculate the Fisher information with respect to the signaling input’s amplitude. For a wide range of amplitudes, we observe a non-monotonic behavior for the Fisher information as a function of background noise. Moreover, Fisher information non-trivially depends on the signaling input’s amplitude; changing the amplitude, we observe one maximum in the high level of the background noise. The single maximum splits into two maximums in the low noise regime. This finding demonstrates the benefit of the analytic solution in investigating signal transfer by neurons.  相似文献

To test whether radioactive contamination reduced the abundance of mammals, and whether species differed in susceptibility to radiation, we censused mammals by counting tracks in the snow along 161 100-m line transects around Chernobyl during February 2009. The abundance of mammal tracks was negatively related to level of background radiation, independent of the statistical model, with effects of radiation accounting for a third of the variance. The effect of radiation differed significantly among species. There was a positive relationship between abundance of predators and abundance of prey, modified by the level of background radiation because the number of predators increased disproportionately with the number of prey at high levels of radiation. These findings suggest that predatory mammals aggregate in areas with abundant prey, especially when prey are exposed to high levels of radiation. This study emphasizes the negative effects of level of background radiation on the abundance of mammals and predator–prey interactions.  相似文献

Past research has shown, separately, that endogenous location cues and high perceptual load search tasks increase the specificity of attentional deployment to task-relevant regions of the visual field, while complex task-irrelevant backgrounds greatly resembling task-relevant stimuli reduce it. Here, we investigated in the same study whether the perceptual load created by an endogenously cued set of task-relevant stimuli determines whether a surrounding complex background of similar task-irrelevant stimuli would interfere with search. Our results show that high perceptual load protects against interference from a complex background of similar but task-irrelevant stimuli, situated just beyond the boundaries of the task-relevant set. Furthermore, our findings demonstrate that search characteristics do not change when the relevant set is restricted attentionally to a smaller delineated area, even in the presence of a background. Finally, we found that the efficacy of endogenous location cueing is not dependent on the type of search task that occurs in the cued area. Our findings also reveal that alternative attention-directing strategies, such as guided search and signal detection, may be employed in such tasks in the absence of endogenous location cueing.  相似文献
