A series of experiments was conducted to assess net CO2assimilationand growth responses to waterlogging of grafted and seedlingtrees in the genus Annona. Seedlings of A. glabra, A. muricataandA. squamosa L., and scions of ‘Gefner’ atemoya(A. squamosaxA. cherimola Mill.), ‘49-11’ (‘Gefner’atemoyaxA. reticulata L.), ‘4-5’ (‘Priestley’atemoyaxA. reticulata), A. reticulata grafted onto either A.glabra, A. reticulata orA. squamosa rootstocks were floodedfor up to 60 d. Soil anaerobiosis occurred on the third dayof flooding. Seedlings ofA. glabra and A. muricata, and thescions ‘49-11’, ‘Gefner’ atemoya, andA. reticulata grafted onto A. glabra rootstock were consideredflood tolerant based on their ability to survive and grow inflooded conditions. Scions of the normally flood-sensitive A.reticulata, ‘Gefner’ atemoya, and ‘49-11’tolerated root waterlogging when grafted onto the flood-tolerantspecies, A. glabra. In contrast, flooding of A. squamosa seedlingsand rootstocks, and A. reticulata rootstocks greatly reducedgrowth and net CO2assimilation rates, and resulted in 20–80%tree mortality. Stem anatomical responses to long-term flooding(12 continuous months) were assessed in seedlings of A. glabraand A. muricata, and trees of ‘49-11’ grafted ontoA. glabra. Flooded trees developed hypertrophied stem lenticels,particularly in A. glabra, and enlarged xylem cells resultingin thicker stems with reduced xylem density. Flooding did notincrease air spaces in pre-existing xylem near the pith or inxylem tissue that was formed during flooding. Thus, flood tolerancedid not involve aerenchyma formation in the stem. This study was conducted with Lolium temulentum, Festuca pratensis,and the two hybrids L. multiflorum x F. pratensis ‘Elmet’and L. perenne x F. pratensis ‘Prior’. In a comparisonof various durations (7–42 d) of pretreatment at 4 or7 °C the highest yield of microspore-derived callus of L.temulentum was obtained after pretreatment of spikes at 7 °Cfor 28 d, conditions which also proved optimal for panicle pretreatmentwith F. pratensis. For ‘Elmet’, durations of 21–42d were optimal, and for ‘Prior’ the responses tendedto decline with increasing duration. Haploids (2n =24) of the common tetraploid (2n=48) potato (SolanumtuberosumL.) provide promising material for attacking many problemsconcerned with the genetics, cytogenetics and breeding of thisspecies. Interspecific 4xx2xcrosses betweenSolanum tuberosumgp.Andigenaorgp.Tuberosumcultivars as pistillate parents andSolanum tuberosumgp.Phurejaassource of pollen (hereafter ‘pollinator’) have beenused to produce maternally derived haploids through parthenogenesis.This paper discusses the nature of the ‘pollinator’effect in haploid extraction. The ‘pollinator’ hada significant effect on haploid frequencies following 4xx2xcrosses.The ‘pollinator’ effect seems to operate via theendosperm, in which haploid (n=2x) embryos are associated withhexaploid endosperm. Increasing ß-amylase activity in wheat (Triticum aestlvum,var. Star) seedling shoot tissues was consistently accompaniedby the development of a characteristic polymorphism of the enzyme,as shown by electrophoresis employing amylopectin-containingpolyacrylamide gels. Very young shoot tissue contained one principalform of the enzyme (ß1), whereas two other major forms(ß2, ß3) appeared complementary to thisupon further growth. In vitro incubation experiments indicatedthat the polymorphism arose via a probably proteolytic conversionof ß1 into ß2 and ß3. The conversioninvolved neither an activation of ß-amylase nor asignificant modification of ß-amylase component plvalues. The electrophoretic ß-amylase patterns ofsubcellular leaf compartments suggested that ß1 issynthesized in the cytoplasm of leaf mesophyfi cells and thatthe other forms arise upon transfer of this ‘primary’form into the vacuole. The development of shoot ß-amylaseactivity did not require light, but appeared to be under thenegative control of the chloroplast and was stimulated by mineralnutrients. No clear relationship between ß-amylaseactivity and starch metabolism was evident, since the leaf activitywas largely absent from mesophyll protoplasts, could not beunequivocally demonstrated in the mesophyll chioroplasts, anddeveloped regardless of whether the tissues contained significantamounts of starch or not. The effects of different applied NO3 concentrations onextension growth and final length and area of leaves 1–4of five cereals and six pasture grasses of temperate originwere examined. Increased applied NO3 in the range 0.1–0.5.0mol m–3 caused decreased duration of growth but increasedgrowth rate and final length of leaves 2–4 of the cerealsAvena saliva, Hordeum vulgare, Secale cereale, x Triticosecaleand Triticum aestivum. For all cereals, increased NO3resulted in increased area of leaves 1–4. Pasture grasseswere supplied either 0.5 or 50 mol m–3 NO3. Increasedapplied NO3 (0.5–5.0 mol m–3) resulted indecreased duration of growth and increased growth rate and finalarea of leaves 1–4 of Bromus wiltdenowii, leaves 2–4ofFestuca arundinaceae and leaves 3 and 4 of Lolium muitiflorum.In addition, length of leaves 3 and 4 of B. witidenowii increasedwith increased NO3. Increased NO3 resulted inincreased area of leaves 2–4 of Dactylis gtomerata andLolium perenne and leaves 3 and 4 of Phalaris aquaiica but hadno effect on extension growth of all three species. Background and Aims The desert legume genus Ammopiptanthuscomprises two currently endangered species, A. mongolicus andA. nanus. Genetic variability and genetic differentiation betweenthe two species and within each species were examined. • Methods Inter-simple sequence repeat (ISSR) marker datawere obtained and analysed with respect to genetic diversity,structure and gene flow. • Key Results Despite the morphological similarity betweenA. mongolicus and A. nanus, the two species are geneticallydistinct from each other, indicated by 63 % species-specificbands. Low genetic variability was detected for both populationlevel (Shannon indices of diversity Hpop = 0·106, percentageof polymorphic loci P = 18·55 % for A. mongolicus; Hpop= 0·070, P = 12·24 % for A. nanus) and specieslevel (Hsp = 0·1832, P = 39·39 % for A. mongolicus;Hsp = 0·1026, P = 25·89 % for A. nanus). Moderategenetic differentiation was found based on different measures(AMOVA ST and Hickory B) in both A. mongolicus (0·3743–0·3744)and A. nanus (0·2162–0·2369). • Conclusions The significant genetic difference betweenthe two species might be due to a possible vicariant evolutionaryevent from a single common ancestor through the fragmentationof their common ancestor's range. In situ growth and development of Neocalanus flemingeri/plumchrusstage C1–C4 copepodites were estimated by both the artificial-cohortand the single-stage incubation methods in March, April andMay of 2001–2005 at 5–6°C. Results from thesetwo methods were comparable and consistent. In the field, C1–C4stage durations ranged from 7 to >100 days, dependent ontemperature and chlorophyll a (Chl a) concentration. Averagestage durations were 12.4–14.1 days, yielding an averageof 56 days to reach C5, but under optimal conditions stage durationswere closer to 10 days, shortening the time to reach C5 (fromC1) to 46 days. Generally, growth rates decreased with increasingstage, ranging from 0.28 day–1 to close to zero but weretypically between 0.20 and 0.05 day–1, averaging 0.110± 0.006 day–1 (mean ± SE) for single-stageand 0.107 ± 0.005 day–1 (mean ± SE) forartificial-cohort methods. Growth was well described by equationsof Michaelis–Menten form, with maximum growth rates (Gmax)of 0.17–0.18 day–1 and half saturation Chl a concentrations(Kchl) of 0.45–0.46 mg m–3 for combined C1–3,while Gmax dropped to 0.08–0.09 day–1 but Kchl remainedat 0.38–0.93 mg m–3 for C4. In this study, in situgrowth of N. flemingeri/plumchrus was frequently food limitedto some degree, particularly during March. A comparison withglobal models of copepod growth rates suggests that these modelsstill require considerable refinement. We suggest that the artificial-cohortmethod is the most practical approach to generating the multispeciesdata required to address these deficiencies.  相似文献

The ‘core-skin’ hypothesis postulates that secondarilythickened plants behave energetically as an inert ‘core’covered by an active ‘skin’, the ‘skin’being two-imensional, the ‘core’ three-dimensional.This would explain the ‘self-thinning ‘or‘–3/2’ rule of plant ecology, that is, the tendencyfor log (dry weight per plant) and log (number of plants perunit area) to progress along a straight line relationship, withslope = – 3/2’. The hypothesis was tested as follows. Plant nitrogen contentwas used as an estimate of the mass of ‘skin’ perplant, and dry weight as an estimate of the mass of the ‘core’.As plants mature the slope of the relationship between y = log(mass of nitrogen per plant) and x = log (mass of dry matterper plant) is expected to decline from an initial value of 1.0towards a final value of 0.66. The intercept of the relationshipis expected to reflect the intrinsic content of ‘skin’per unit of ‘core’. Genotypic variation in thisparameter should cause genotypic differences in the maximumattainable yield of biomass per unit area. The expectations were investigated by fitting the function y= p+qx+r exp – x to 30 sets of data on plant nitrogencontent, plant weight and time in 18 different vegetables. Simplelinear regressions of y on x were fitted to more limited setsof data on weights and nitrogen contents of mature trees. Theexpectations were, with some minor exceptions, confirmed. The photosynthetic responses of eight tree and shrub speciesto simulated sunflecks was measured in the field. The net carbonexchange (NCE) of Corylus avellana and Ulmus glabra increasedwith irradiance up to the maximum irradiance of 230 µmolm–2 s–1. The NCE of Fraxinux excelsior, Hedera helixand the sun and shade forms of Rhododendron ponticum saturatedat about 120 µmol m–2 s–1 whereas the NCEof Ilex aquifolium, Daphne laureola and Fagus sylvatica hadeffectively saturated at 27 µmol m–2 s–1. In all cases the quantum efficiency of NCE could be predictedfrom measurements of chlorophyll fluorescence and the maximumvalue for NCE from measurements of stomatal conductance. Therelationships were combined into a model for predicting NCE/irradiancecharacteristics. A study was made of the influence of grain size variation withinand between diploid, tetraploid and hexaploid wheat, on a numberof seedling growth characters. Differences in grain size within the three ploidy levels appearedto be related to total photosynthetic area and dry weight accretionin the seedling. In the diploids there was a positive correlationbetween seed size and total photosynthetic area (r = +0·99,P < 0·01) and total dry weight (r = +0·84,P < 0·05) of the seedling at 10 weeks after emergence.In the tetraploid and hexaploids, seed size was negatively correlatedwith both total photosynthetic area (r = –0·69,P < 0·05 and r = –0·33, P < 0·05for the tetraploids and hexaploids respectively) and total dryweight (r = –0·69, P < 0·05 and r = –0·59,P < 0·05 for the tetraploids and hexaploids respectively),of the seedlings 10 weeks after emergence. The main physiological distinction between the tetraploids andhexaploids appeared to be the superiority of the hexaploidsin rate of leaf appearance and the lower ratio of expanded tounexpanded leaves in the seedling 10 weeks after emergence.The tetraploids, in turn, appeared to be superior to the diploidsin these two characters. The relationships between photosynthesis and photosyntheticphoton flux densities (PPFD, P-l) were studied during a red-tideof Dinophysis norvegica (July-August 1990) in Bedford Basin.Dinophysis norvegica, together with other dinoflagellates suchas Gonyaulax digitate, Ceratium tripos, contributed {small tilde}50%of the phytoplankton biomass that attained a maximum of 16.7µg Chla 1 and 11.93 106 total cells I–1.The atomic ratios of carbon to nitrogen for D.norvegica rangedfrom 8.7 to 10.0. The photosynthetic characteristics of fractionatedphytoplankton (>30 µm) dominated by D.norvegica weresimilar to natural bloom assemblages: o (the initial slope ofthe P-l curves) ranged between 0.013 and 0.047 µg C [µgChla]–1 h–1 [µmol m s–1]–1the maximum photosynthetic rate, pBm, between 0.66 and 1.85µg C [µghla]–1 h–1; lk (the photoadaptationindex) from 14 to 69 µ,mol m–2 s–1. Carbonuptake rates of the isolated cells of D.norvegica (at 780 µmolm–2 s–1) ranged from 16 to 25 pg C cell–1h and were lower than those for C.tripos, G.digitaleand some other dinoflagellates. The variation in carbon uptakerates of isolated cells of D.norvegica corresponded with PBmof the red-tide phytoplankton assemblages in the P-l experiments.Our study showed that D.norvegica, a toxigenic dinoflagellate,was the main contributor to the primary production in the bloom.  相似文献

We previously showed that plasma membrane Ca2+-ATPase (PMCA) activity accounted for 25–30% of relaxation in bladder smooth muscle (8). Among the four PMCA isoforms only PMCA1 and PMCA4 are expressed in smooth muscle. To address the role of these isoforms, we measured cytosolic Ca2+ ([Ca2+]i) using fura-PE3 and simultaneously measured contractility in bladder smooth muscle from wild-type (WT), Pmca1+/–, Pmca4+/–, Pmca4–/–, and Pmca1+/–Pmca4–/– mice. There were no differences in basal [Ca2+]i values between bladder preparations. KCl (80 mM) elicited both larger forces (150–190%) and increases in [Ca2+]i (130–180%) in smooth muscle from Pmca1+/– and Pmca1+/–Pmca4–/– bladders than those in WT or Pmca4–/–. The responses to carbachol (CCh: 10 µM) were also greater in Pmca1+/– (120–150%) than in WT bladders. In contrast, the responses in Pmca4–/– and Pmca1+/–Pmca4–/– bladders to CCh were significantly smaller (40–50%) than WT. The rise in half-times of force and [Ca2+]i increases in response to KCl and CCh, and the concomitant half-times of their decrease upon washout of agonist were prolonged in Pmca4–/– (130–190%) and Pmca1+/–Pmca4–/– (120–250%) bladders, but not in Pmca1+/– bladders with respect to WT. Our evidence indicates distinct isoform functions with the PMCA1 isoform involved in overall Ca2+ clearance, while PMCA4 is essential for the [Ca2+]i increase and contractile response to the CCh receptor-mediated signal transduction pathway. Feeding in predacious estuarine ciliates was investigated ina series of laboratory experiments using a new method of preylabeling which facilitates microscopic indentification of ingestedprey items. Ingestion rates of Mesodinium pulex, Euplotes vannusand E.woodruffi were estimated using the appearance, insidethe predator, of bacteriovorous ciliates (Metanophrys sp., Cyclidiumsp.and Pleuronema sp ) labeled with fluorescent microspheres. Preyremain motile and have presumably unaltered surface characteristics.Ingestion rates of log-growth phase predators increased withprey density. Mesodinium pulex ingested 0 15–0.32 cellsh–1 over a prey concentration of 60–2300 ml–1.Maximum ingestion rates of E. woodruffi and E. vannus were 4.5and 3.4 cells h–1 respectively, estimated at prey abundancesof 75 and 172 cells ml–1 respectively. Comparisons offeeding rates on prey of different sizes, and the effects ofstarvation, indicated that ingestion is likely limited by differentfactors in ‘raptorial’ (M pulex) and ‘filterfeeding’ (Euplotes spp.) predators.  相似文献

Analysis of the Promoter of the Auxin-Inducible Gene, parC, of Tobacco   总被引:2,自引:0,他引:2  
The auxin-responsive region (AuxRR) in the promoter of the parCgene was analyzed in transgenic tobacco plants in which the5' flanking region of the parC promoter was placed upstreamof the gene for rß-glucuronidase (GUS). The AuxRRwas located between nucleotides (nt) –226 and –54.Detailed dissection of this segment revealed that the presenceof the non-contiguous sequences from nt –226 to –151and from nt –84 to –54 was required for the expressionof the auxin responsiveness of the parC promoter. The sequencefrom nt –226 to –151 was found to contain a sequencewhich resembles the as-1 element in the 35S promoter of cauliflowermosaic virus (CaMV). Although it has been reported that theas-1 element is involved in auxin responsiveness [Liu and Lam(1994) J. Biol. Chem. 269: 668], we showed that introductionof a point mutation into the as-1-like sequence completely eliminatedauxin responsiveness, a result that suggests that the sequenceis indispensable for auxin responsiveness. However, the presenceof the as-1-like sequence alone was not sufficient for auxinresponsiveness, since the segment (nt –226 to –84)that included the as-1-like sequence failed to confer auxinresponsiveness on the core promoter. It is possible that thetwo separately located sequences play specific roles in interactionswith trans-factors that are required for the expression of theauxin responsiveness of the parC promoter. Rhodotorula minuta cells, which have only traces of carotenoidswhen grown in the dark, started carotenoid production with theonset of illumination and the amount increased almost linearlyuntil 70 hr then remained constant thereafter when incubationwas continued under illumination, with the number of cells continuingto increase. The rate of carotenoid production [Vc (µgg–1 hr–1)] depended on the intensity of light [I(ergcm–2 sec–1)], with the relationship of Vc=0.74 logI–1.46. The final carotenoid content [C(µg g–1)]of cells incubated under continuous light was also controlledby the light intensity [I], with the relationship of C=52 logI–81. Control of carotenoid production by light occursas a two-phase process consisting of a temperatureindependentphotochemical reaction and light-independent biochemical reactions. An attempt has been made to separate constituents of marineseston samples: inorganic material, detritus and the algal species,by density gradient centrifugation, without affecting the physiologicalstate of the algae. A relatively inert gradient material, consistingof Percoll, salt and sucrose, was composed. Since the densitiesof detritus and algae as well as those of different algal speciesoften overlapped, only 10 of the 100 samples processed in thecourse of the year showed a reasonable separation. However,an enrichment with respect to one or more species was oftenachieved. Densities of eleven species of marine diatoms andof one dinoflagellate have been determined at different timesof the year. For eight diatom species and for the dinoflagellatethe following specific density ranges were established: Bidduiphiaaurita: 1.18–1.23 g cm–3, Biddulphia sinensis: 1.03–1.08g cm–3, Cerataulina bergonii: 1.03–1.06 g cm–3,Ditylum brightwellii: 1.07–1.13 g cm–3, Rhizosoleniadelicatula: 1.04–1.09 g cm–3, Skeletonema costatum:1.12–1.17 g cm–3, Streptotheca thamensis: 1.04–1.10g cm–3 , Thalassiosira rotula: 1.05–1.10 g cm–3,Peridinium sp.: 1.08–1.12 g cm–3. No seasonal variationin density was demonstrated. Gradients of different compositiondid not influence density measurements.  相似文献
