The fatty acid composition of the alga Chlorella saccharophila was investigated under different growth conditions. Using glucose as the sole carbon source, heterotrophically-grown Chlorella saccharophila produced a greater proportion of the polyunsaturated fatty acids (C18: 2 and C18: 3) than photosynthetic cultures, with linoleic acid (C18: 2) predominating. An unexpected discovery was the observation that at the lowest glucose concentration (2.5 gl–1) the lipid content of the algae increased to between 36–47% of the cell weight, depending on the temperature. At glucose concentrations of 5 g l–1 or more, the lipid content fell to 10–12% of the cell, although total fatty acid yield was higher due to higher biomass concentrations. Aeration of heterotrophic cultures promoted the production of unsaturated fatty acids compared to non-aerated cultures.  相似文献

A factorial experiment was designed to study the effects of Mg2+, K+, and Na+ on the growth and biochemistry of Chlorella sorokiniana. Raising Mg2+ or K+ concentration in the nutrient medium increased growth rates as well as total N levels and Mg2+ and K+ accumulation by the cells. The total N effect was Mg2+-dependent—if Mg2+ was below a certain level in the medium—increasing the K+ concentration did not raise the total N level of cells. Low nutrient levels of K+ decreased the levels of unsaturated fatty acids (especially 18:1 and 18:3), while increasing the levels of palmitic acid (16:0), total fatty acids, and total lipid. Increasing nutrient K+ concentrations were accompanied by increases in levels of some unsaturated fatty acids, with a concomitant reduction in 16:0, total fatty acids and total lipid. Low Mg2+ levels in the nutrient medium reduced the cellular levels of palmitic acid, total fatty acids, total lipid, and certain unsaturated fatty acids (though this last effect also depended on the nutrient level of K+). These relationships indicate that Mg2+ may be important in the initial steps of fatty acid synthesis, whereas K+ may be necessary for the formation of certain unsaturated fatty acids. Variations in Na+ concentration did not have any significant effect on the growth and biochemistry of C. sorokiniana.  相似文献

The toxic effects of high pressure oxygen on the isolated toad urinary bladder have been studied. Sodium transport in this system is reversibly inhibited by high pressure O2. This inhibition is potentiated by adrenal steroid hormones, and occurs despite both increased glycolytic and Kreb's cycle flux and tissue ATP content. High pressure O2 leads to increased pyruvate/lactate and pyruvate/malate redox couples, as well as to a decrease in the weight percentage of phospholipid long-chain unsaturated fatty acids and [2-14C]pyruvate incorporation into tissue lipid. During recovery from high pressure O2 treatment, [2-14C]Pyruvate incorporation into lipid is increased and the weight percentage of long-chain unsaturated fatty acids increases. These data indicate that high pressure O2 poisoning in this tissue does not result from an inhibition of carbohydrate metabolism, but may result from the formation of toxic lipid peroxides.  相似文献

Analyses were made of the fatty-acid composition of Candida utilis NCYC 321 grown in a chemostat at a dilution rate (equal to growth rate) of 0.1 hr−1 and at temperatures in the range of 30 to 15 C and dissolved oxygen tensions between 75 and <1 mm of Hg. Cells grown under glucose limitation or NH4+ limitation contained mainly C16:0, C16:1, C18:0, C18:1, C18:2, and C18:3 acids as detected by gas-liquid chromatography of methyl esters of the acids from lipids extracted with chloroform-methanol. The relative proportions of these acids varied with the growth temperature and the dissolved-oxygen tension in the culture. A decrease in growth temperature from 30 to 20 C led to an increased synthesis of unsaturated acids in cells grown under either limitation at a fixed-oxygen tension in the range of 75 to 5 mm of Hg. In cultures with a dissolved-oxygen tension of 1 and <1 mm of Hg, a further decrease in temperature to 15 C caused an increased synthesis of unsaturated fatty acids. A decrease in dissolved-oxygen tension led to a diminished synthesis of unsaturated fatty acids in cells grown at a fixed temperature under either limitation. Cells grown at a fixed temperature under glucose limitation synthesized a greater proportion of C16 acids at the expense of C18 acids as the dissolved oxygen tension was decreased from 75 to <1 mm of Hg. A preferential synthesis of C16 acids also occurred as the growth temperature was decreased from 30 to 15 C in cells grown under glucose limitation at a fixed-oxygen tension. The same effect was observed in cells grown under NH4+ limitation when the temperature was lowered from 30 to 20 C; but when the temperature was decreased further to 15 C, the cells synthesized a slightly greater proportion of C18 acids. Synthesis of a large proportion of C16 acids was accompanied by an excretion of pyruvate, and occasionally traces of 2-ketoglutarate, and an increased intracellular accumulation of certain amino acids.  相似文献

Stichococcus, a genus of green algae, distributes in ice-free areas throughout Antarctica. To understand adaptive strategies of Stichococcus to permanently cold environments, the physiological responses to temperature of two psychrotolerants, S. bacillaris NJ-10 and S. minutus NJ-17, isolated from rock surfaces in Antarctica were compared with that of one temperate S. bacillaris FACHB753. Two Antarctic Stichococcus strains grew at temperature from 4 to 25°C, while the temperate strain could grow above 30°C but could not survive at 4°C. The photosynthetic activity of FACHB753 at lower than 10°C was less than that of Antarctic algae. Nitrate reductase in NJ-10 and NJ-17 had its optimal temperature at 20°C, in comparison, the maximal activity of nitrate reductase in FACHB753 was found at 25°C. When cultured at 4–15°C a large portion of unsaturated fatty acids in the two Antarctic species was detected and the regulation of the degree of unsaturation of fatty acids by temperature was observed only above 15°C, though the content of the major unsaturated fatty acid αC18:3 in FACHB753 decreased with the temperatures elevated from 10 to 25°C. Elevated nitrate reductase activity and photosynthetic rates at low temperatures together with the high proportion of unsaturated fatty acids contribute to the ability of the Antarctic Stichococcus to thrive.  相似文献

Fatty acid composition of old and new roots was determined for soybeans (Glycine max [L.] Merr. cv Ransom) at root-zone temperatures of 14, 18, and 22°C during a 26-day period. New roots had a greater concentration of polyunsaturated fatty acids than old roots. The ratio of polyunsaturated to saturated fatty acid concentration in new roots exposed to 14 and 18°C peaked at 16 days and declined, while the corresponding ratio in old roots increased throughout the treatment period. Apparently the response of fatty acid composition in old and new roots to low temperature was mediated by tissue aging or differentiation. These findings were contrary to the concept that modifications in fatty acid composition remain constant at lower temperatures.

The function of root tissues exposed to lower temperature was evaluated with respect to the ability of the root systems to absorb NO3. Over the relatively long periods of exposure, the ability of whole root systems to absorb NO3 was similar at cool and warm temperatures. The effect of cool temperature on functioning of roots appeared to involve reductions in the rates of initiation and differentiation of young root tissues rather than changes in membrane permeability related to alteration of fatty acid composition.

The incorporation of 14CO2 into unsaturated fatty acids during seed development was measured in sunflowers grown in controlled environments with day temperatures of 28°C and night temperatures of 15°C or 22°C. While the average temperatures to which the plants were exposed did not differ greatly, the ratio of linoleic acid to oleic acid synthesized was much greater at a night temperature of 15°C than at 22°C. These results support the proposal (Harris et al. 1978) that the mean minimum temperature experienced during seed development is the major environmental factor influencing the unsaturated fatty acid composition of sunflower seed oil.  相似文献

The effects of long chain unsaturated fatty acids such as linoleic acid on bovine platelets were examined. Not only linoleic acid, but also oleic and linolenic acid, at just below the concentrations causing marked cell lysis, induced an absorbance decrease of the platelet suspension in the presence of Ca2+. Since this absorbance decrease was reversed by the addition of EDTA and moreover aggregate formation was found by macroscopic and microscopic observation, it was concluded that unsaturated fatty acids at just below their lytic concentrations caused platelet aggregation. Unsaturated fatty acids also caused release of adenine nucleotides, but there was a lag time between the release and the aggregation, just as with ADP-induced release, suggesting that the aggregation was independent of the release of ADP. It was revealed that this activation of platelets by unsaturated fatty acids was caused by marked Ca2+ uptake into the cytoplasm, resulting from significant membrane perturbation.  相似文献

Summary Dieary lipids strongly influence the pattern of torpor and the body lipid composition of mammalian hibernators. The object of the present study was to investigate whether these diet-induced physiological and biochemical changes also occur in species that show shallow, daily torpor. Deer mice, Peromyscus maniculatus, were fed with rodent chow (control diet) or rodent chow with either 10% sunflower seed oil (unsaturated diet) or 10% sheep fat (saturated diet). Animals on the unsaturated diet showed a greater occurrence of torpor (80–100% vs 26–43%), longer torpor bouts (4.5 vs 2.25 h), a lower metabolic rate during torpor (0.96 vs 2.25 ml O2·g-1·h-1), and a smaller loss of body mass during withdrawal of food (2.35 vs 3.90 g) than animals on the saturated diet; controls were intermediate. These diet-induced physiological changes were associated with significant alterations in the fatty acid composition of depot fat, leg muscle and brain total lipids, and heart mitochondrial phospholipids. Significant differences in the total unsaturated fatty acid (UFA) content between animals on saturated and unsaturated diet were observed in depot fat (55.7% vs 81.1%) and leg muscle (56.4% vs 72.1%). Major compositional differences between diet groups also occurred in the concentration of n6 and/or n3 fatty acids of brain and heart mitochondria. Fatty acids newly synthesized by Brevibacterium ammoniagenes grown at different temperatures were analyzed. The assay temperature, not the growth temperature, was found to be the major factor affecting the unsaturated/saturated ratio of newly synthesized fatty acids in logarithmic-phase cells. However, in the stationary-phase cells the growth temperature also affected the product profile significantly; cells grown at 7 degrees C produced relatively more oleate and stearate and less palmitate and hexadecenoate when shifted up to 37 degrees C than did cells grown and assayed at 37 degrees C. The unsaturated/saturated ratio as well as average chain length of fatty acids also varied along with the progress of isothermal growth phase. These changes in fatty acid product profiles observed in vivo could be mimicked in vitro assays of the fatty acid synthetase by changing malonyl-CoA concentrations. Our results suggest that the malonyl-CoA concentration is a factor which, in addition to temperature, determines growth-phase-dependent and growth-temperature-dependent changes in the unsaturated/saturated ratios of fatty acids.  相似文献

The photosynthetic responses of four alfalfa (Medicago sativa L.) cultivars to 10 and 22 C air temperatures were examined and the relationship between the photosynthetic response at 10 C and the fatty acid composition of the chloroplast membranes was determined. Chilling-resistant cultivars exhibited moderate reductions in photosynthesis at 10 C, compared to 22 C, and contained a significantly greater percentage of polyunsaturated fatty acids in the chloroplast membrane and a greater double bond index than the chilling-sensitive cultivars. The chilling-sensitive cultivars exhibited severe reductions in photosynthesis at 10 C, compared to 22 C. The reduction in photosynthesis at 10 C is shown to be negatively correlated (r = −0.94) with the double bond index of the chloroplast membranes of the cultivars observed.

The results support the hypothesis that reduced photosynthesis due to chilling temperatures is influenced by the unsaturated fatty acid composition of the chloroplast membrane which affect temperature-induced phase changes in chloroplast membrane lipids.

The binding of cAMP to the chemotactic cAMP receptor in intact Dictyostelium discoideum cells and isolated membranes is strongly inhibited by unsaturated fatty acids. In isolated membranes, cis-unsaturated fatty acids decreased the number of accessible cAMP binding sites, without significantly altering their affinity. Most potent were C18 and C20 cis-poly unsaturated fatty acids, like arachidonic acid, linoleic acid and linolenic acid. Trans-unsaturated fatty acid was less potent than its cis isomer, while saturated fatty acids did not affect the binding of cAMP to receptors at all. Oxidation reactions were not important for the effect of unsaturated fatty acids. When membranes were preincubated with millimolar concentrations of Ca2+, the effect of unsaturated fatty acids was strongly diminished. Mg2+ was ineffective. Ca2+, if presented after the incubation of membranes with unsaturated fatty acids, did not reverse the inhibitory effect. The specificity of the fatty acid effect, and the interference with Ca2+, but not Mg2+, suggest that the properties of the cAMP receptor are changed as a result of alterations in the lipid bilayer structure of the membrane.  相似文献

The mitochondrial β-oxidation system is one of the central metabolic pathways of energy metabolism in mammals. Enzyme defects in this pathway cause fatty acid oxidation disorders. To elucidate the role of 2,4-dienoyl-CoA reductase (DECR) as an auxiliary enzyme in the mitochondrial β-oxidation of unsaturated fatty acids, we created a DECR–deficient mouse line. In Decr−/− mice, the mitochondrial β-oxidation of unsaturated fatty acids with double bonds is expected to halt at the level of trans-2, cis/trans-4-dienoyl-CoA intermediates. In line with this expectation, fasted Decr−/− mice displayed increased serum acylcarnitines, especially decadienoylcarnitine, a product of the incomplete oxidation of linoleic acid (C18:2), urinary excretion of unsaturated dicarboxylic acids, and hepatic steatosis, wherein unsaturated fatty acids accumulate in liver triacylglycerols. Metabolically challenged Decr−/− mice turned on ketogenesis, but unexpectedly developed hypoglycemia. Induced expression of peroxisomal β-oxidation and microsomal ω-oxidation enzymes reflect the increased lipid load, whereas reduced mRNA levels of PGC-1α and CREB, as well as enzymes in the gluconeogenetic pathway, can contribute to stress-induced hypoglycemia. Furthermore, the thermogenic response was perturbed, as demonstrated by intolerance to acute cold exposure. This study highlights the necessity of DECR and the breakdown of unsaturated fatty acids in the transition of intermediary metabolism from the fed to the fasted state.  相似文献

The aim of this paper was to study the effects of temperature and irradiance on the photodegradation state of killed phytoplankton cells. For this purpose, killed cells of the diatom Chaetoceros neogracilis RCC2022 were irradiated (photosynthetically active radiation) at 36 and 446 J · s?1 · m?2 (for the same cumulative dose of irradiation energy) and at two temperatures (7°C and 17°C). Analyses of specific lipid tracers (fatty acids and sterols) revealed that low temperatures and irradiances increased photooxidative damages of monounsaturated lipids (i.e., palmitoleic acid, cholesterol and campesterol). The high efficiency of type II photosensitized degradation processes was attributed to: (i) the relative preservation of the sensitizer (chlorophyll) at low irradiances allowing a longer production of singlet oxygen and (ii) the slow diffusion rate of singlet oxygen through membranes at low temperatures inducing more damages. Conversely, high temperatures and irradiances induced (i) a rapid degradation of the photosensitizer and a loss of singlet oxygen by diffusion outside the membranes (limiting type II photosensitized oxidation), and (ii) intense autoxidation processes degrading unsaturated cell lipids and oxidation products used as photodegradation tracers. Our results may explain the paradoxical relationship observed in situ between latitude and photodegradation state of phytoplankton cells.  相似文献

The higher the growth temperature of Escherichia coli cultures the greater is the proportion of saturated fatty acids in the bacterial phospholipids. When fatty acids are exogenously supplied to E. coli, higher growth temperatures will likewise increase the relative incorporation of saturated fatty acids into phospholipids. One of the steps in the utilization of fatty acids for phospholipid biosynthesis is, therefore, temperature-controlled. The temperature effect observed in vivo with mixtures of 3H-oleate and 14C-palmitate is demonstrable in vitro by using mixtures of the coenzyme A derivative of these fatty acids for the acylation of α-glycerol phosphate to lysophosphatidic and phosphatidic acids. In E. coli extracts, the relative rates of transacylation of palmityl and oleyl coenzyme A vary as a function of incubation temperature in a manner which mimics the temperature control observed in vivo. The phosphatidic acid synthesized in vitro shows a striking enrichment of oleate at the β position analogous to the positional specificity observed in phospholipids synthesized in vivo.  相似文献

Background and Aims Plants growing under elevated atmospheric CO2 concentrations often have reduced stomatal conductance and subsequently increased leaf temperature. This study therefore tested the hypothesis that under long-term elevated CO2 the temperature optima of photosynthetic processes will shift towards higher temperatures and the thermostability of the photosynthetic apparatus will increase.Methods The hypothesis was tested for saplings of broadleaved Fagus sylvatica and coniferous Picea abies exposed for 4–5 years to either ambient (AC; 385 µmol mol−1) or elevated (EC; 700 µmol mol−1) CO2 concentrations. Temperature response curves of photosynthetic processes were determined by gas-exchange and chlorophyll fluorescence techniques.Key Results Initial assumptions of reduced light-saturated stomatal conductance and increased leaf temperatures for EC plants were confirmed. Temperature response curves revealed stimulation of light-saturated rates of CO2 assimilation (Amax) and a decline in photorespiration (RL) as a result of EC within a wide temperature range. However, these effects were negligible or reduced at low and high temperatures. Higher temperature optima (Topt) of Amax, Rubisco carboxylation rates (VCmax) and RL were found for EC saplings compared with AC saplings. However, the shifts in Topt of Amax were instantaneous, and disappeared when measured at identical CO2 concentrations. Higher values of Topt at elevated CO2 were attributed particularly to reduced photorespiration and prevailing limitation of photosynthesis by ribulose-1,5-bisphosphate (RuBP) regeneration. Temperature response curves of fluorescence parameters suggested a negligible effect of EC on enhancement of thermostability of photosystem II photochemistry.Conclusions Elevated CO2 instantaneously increases temperature optima of Amax due to reduced photorespiration and limitation of photosynthesis by RuBP regeneration. However, this increase disappears when plants are exposed to identical CO2 concentrations. In addition, increased heat-stress tolerance of primary photochemistry in plants grown at elevated CO2 is unlikely. The hypothesis that long-term cultivation at elevated CO2 leads to acclimation of photosynthesis to higher temperatures is therefore rejected. Nevertheless, incorporating acclimation mechanisms into models simulating carbon flux between the atmosphere and vegetation is necessary.  相似文献

The fact that molybdenum (Mo) applications can alleviate low temperature stress (LTS) in plants has been widely reported, but the underlying mechanisms are not fully understood. The effects of Mo (0 and 0.15 mg kg?1) on photosynthetic pigments, fatty acids and the chlorophyll–protein complex of wheat seedlings in pot culture were investigated at 0, 2, 4 and 6 days of LTS. Chlorophyll a and b, total chlorophyll and carotenoid contents were significantly enhanced by Mo application. The palmitic acid (C16:0) content and total saturated fatty acids (TSFA) were drastically decreased in wheat cultivar 97003 at 2 and 4 days of LTS and in wheat cultivar 97014 at 2 days of LTS. The linolenic acid (C18:3) content and total unsaturated fatty acids (TUSFA) were significantly increased in both cultivars at 2 and 4 days of LTS. The palmitoleic acid (C16:1) content was also drastically increased in cultivar 97003 at 2 days of LTS, suggesting that Mo induced a greater production of unsaturated fatty acids or the conversion of TSFA to TUSFA. The ratio of unsaturated to saturated fatty acids and the index of unsaturated fatty acids in the thylakoid membranes were enhanced with supplemental Mo, suggesting that Mo might improve the degree of unsaturation. However, no significant differences were observed in the chlorophyll–protein complexes between the +Mo and ?Mo treatments. These results indicated that the alteration of fatty acids induced by Mo application in the thylakoid membranes of wheat contributed to LTS tolerance.  相似文献

The physical state of the membrane lipids, as determined by fatty acid composition and environmental temperature, has a marked effect on both the temperature range within which Acholeplasma laidlawii B cells can grow and on growth rates within the permissible temperature ranges. The minimum growth temperature of 8 °C is not defined by the fatty acid composition of the membrane lipids when cells are enriched in fatty acids giving rise to gel to liquid-crystalline membrane lipid phase transitions occurring below this temperature. The elevated minimum growth temperatures of cells enriched in fatty acids giving rise to lipid phase transitions occurring at higher temperatures, however, are clearly defined by the fatty acid composition of the membrane lipids. The optimum and maximum growth temperatures are also influenced indirectly by the physical state of the membrane lipids, being significantly reduced for cells supplemented with lower melting, unsaturated fatty acids. The temperature coefficient of growth at temperatures near or above the midpoint of the lipid phase transition is 16 to 18 kcalmol, but this value increases abruptly to 40 to 45 kcalmol at temperatures below the phase transition midpoint. Both the absolute rates and temperature coefficients of cell growth are similar for cells whose membrane lipids exist entirely or predominantly in the liquid-crystalline state, but absolute growth rates decline rapidly and temperature coefficients increase at temperatures where more than half of the membrane lipids become solidified. Cell growth ceases when the conversion of the membrane lipid to the gel state approaches completion, but growth and replication can continue at temperatures where less than one tenth of the total lipid remains in the fluid state. An appreciable heterogeneity in the physical state of the membrane lipids can apparently be tolerated by this organism without a detectable loss of membrane function.  相似文献

The radiation-induced oxidation of saturated and unsaturated fatty acids in aqueous solutions has been estimated by measurement of the continuous uptake of oxygen using an oxygen electrode. Chain reactions, initiated by HO radicals, are easily identified to be occurring in the case of unsaturated fatty acids. Other mild oxidation agents, namely (SCN)-.2, Br-.2 and N.3, are also found to be capable of oxidizing the polyunsaturated fatty acids. Evidence is presented that O-.2 may also initiate peroxidation. The oxidation of the polyunsaturated fatty acids is dependent on dose rate, fatty acid concentration, temperature and the presence of antioxidant and other protective agents. Kinetic studies of the reaction of (SCN)-.2 and Br-.2 with linoleic and linolenic acids have been carried out using pulse radiolysis. The bimolecular rate constants for both radical species with the lipids are approx 10(7) mol-1 dm3 s-1, below their critical micelle concentrations, and decrease at higher concentrations due to micelle formation.  相似文献

Ozone-treated cells of the photosynthetic green alga Chlorella sorokiniana var. pacificensis exhibit an exponential decline in viability, as measured by their ability to form colonies on agar plates. Postexposure conditions appear to have little, if any, effect on this rate of decline. Except in young (early exponential phase) cells, culture age did not affect this rate. The decline in cell viability was correlated with the production of malondialdehyde, arising from the oxidative breakdown of an ozonide of unsaturated fatty acid material. The loss of fatty acids is substantiated by gas-liquid chromatography. A loss of 5 × 10-15 moles of fatty acid per cell corresponds to 75% nonviable cells after 50 minutes of ozone exposure.  相似文献
