Several in vivo and in vitro studies have shown that grape extracts could prevent certain steps in carcinogenesis and a few mechanisms have been proposed for this activity. In this study, the potential antimutagenic activity of methanolic and aqueous extracts from two Greek grape varieties of Vitis vinifera against DNA damage induced by reactive oxygen species (ROS) was assessed as a potential novel chemopreventive mechanism, using Salmonella typhimurium strain TA102. The two grape varieties were Assyrtiko (white grapes) and Mandilaria (red grapes), while the oxidant mutagens used were bleomycin (BLM) and hydrogen peroxide (H(2)O(2)). Since it has been considered that polyphenols present in grapes are their most potent biologically active compounds, we also tested the effects of polyphenol-rich fractions as well as some of the more common grape polyphenols on the activity of the two test mutagens. These polyphenols were quercetin, (+)-catechin, (-)-epicatechin, trans-resveratrol, gallic acid and protocatechuic acid. Almost all extracts showed inhibitory activity against both mutagens. On the other hand, polyphenol-rich fractions as well as individual polyphenols at concentrations found in the extracts either did not diminish or did enhance the activity of the mutagens. These results suggest that the protection of DNA from mutations induced by ROS may be one of the mechanisms accounting for the chemopreventive activity of grape extracts. However, it seems that this protective activity may not be attributed to polyphenols but rather to a synergism of many compounds in the grapes.  相似文献

Vitis vinifera L. cv. ‘Thompson Seedless’ presents a type of stenospermocarpy in grape where fertilization occurs but seeds abort and fail to develop. To unravel the molecular basis for stenospermocarpy in grapes, subtractive hybridization was carried out in order to isolate differentially regulated genes that participate in the seedlessness machinery. Two ‘Thompson’ lines, a seeded and a seedless, were screened during different flower developmental stages. One of the genes, that was differentially expressed between the seeded and seedless lines, was the chloroplast chaperonin 21 (ch-Cpn21). ch-Cpn21 is a 21-kDa co-chaperonin polypeptide formed by two GroES-like domains fused together in tandem. Silencing of ch-Cpn21 in Nicotiana benthamiana plants resulted in leaf stunting, chlorosis, as well as ovary necrogenesis leading to seed abortion. Moreover, organ-specific silencing of ch-Cpn21 only in Lycopersicum esculentum fruits resulted in the development of seedless tomatoes. These results suggest that ch-Cpn21 may play a role in seed abortion in stenospermocarpic grapes.  相似文献

Literature investigations indicate that the grapes have quite complex fertilisation biology. This complexity necessitates extensive investigations to obtain reliable knowledge for both well‐organised hybridisation studies and maximising grape yield. Therefore, this study was conducted to investigate the influences of self‐, free‐ and cross‐pollination on berry and seed characteristics in grape. Five different pollination treatments were applied to ‘Narince’, the most widely known and popular white wine grape in Turkey. Pollen tests indicated that all the cultivars had satisfactory in vitro pollen viability percentages. Free‐pollination produced a significantly higher percentage berry set. Among the pollinizers, the use of pollen of ‘Thompson Seedless’ and ‘Cardinal’ varieties resulted in higher berry set percentage in ‘Narince’. The free‐pollination was also superior in giving the highest weight, length and width of the berry, as well as number of seeds per berry. These findings revealed that there were strong xenial and metaxenial effects in the studied grape cultivars. Among the pollinizer cultivars, the most effective pollinator was ‘Thompson Seedless’. Hence, for better berry set and quality, the use of ‘Thompson Seedless’ as a pollinizer may be an attractive option in both grape production and breeding studies.  相似文献

Exported organic table grapes from the U.S. are currently fumigated with methyl bromide to control quarantined pests such as western flower thrips, Frankliniella occidentalis (Pergande). An alternative treatment which is compatible with organic products is needed. In this study, controlled atmosphere with ultralow oxygen (ULO) treatment was studied to control thrips on harvested table grapes. ULO treatments with < 0.01 ppm oxygen at 3.3 °C for 1, 2, and 3 days were tested for thrips control. The 3-day ULO treatment achieved complete control of thrips. The treatment was also applied to Thompson seedless and Flame seedless grape varieties and grape quality was evaluated two weeks after the treatment. The ULO treatment had no negative impact on the visual quality of grapes. There were no significant differences in overall grape quality or premium quality berries between the ULO treatment and the control. The results indicated that the ULO treatment was effective and safe to control western flower thrips on table grapes and had a potential for commercial use.  相似文献

Pressed grape pomace obtained from the wine production of Cabernet sauvignon (Vitis vinifera) vintage was dried until 9.8% moisture content, ground and submitted to extraction of soluble components from different extraction techniques. Low pressure extractions were performed with ethanol maceration followed by fractionation with n-hexane, dichloromethane, butanol and ethyl acetate. These solvents were furthermore applied for soxhlet extraction. Supercritical fluid extraction (SFE) was also performed to obtain grape pomace extracts by using pure CO(2) and CO(2) with ethanol as co-solvent in concentrations of 10, 15 and 20%w/w. The operating condition used in high pressure extractions was 150bar and 40 degrees C. The antioxidant activity of the grape pomace extracts was determined considering the free radical scavenging assay using 1,1-Diphenyl-2-picrylhydrazyl (DPPH) and was correlated with the total phenol content determined according to the Folin-Ciocalteu method. The results obtained in DPPH tests indicate the highest antioxidant activity of 96.6+/-0.3%AA, with an IC(50) value of 13+/-1, for the extracts obtained with ethyl acetate in solid-liquid extraction. The highest yield values were achieved in soxhlet extraction with ethanol (13.2%w/w) and with butanol (12.2%w/w), and also by SFE with 15% ethanol (9.2%w/w). The lipophilic composition of grape pomace extracts was evaluated by gas chromatography-mass spectrometry with the identification of components like linoleic acid and ethyl linoleate, with important therapeutic activities.  相似文献

Wine grapes are usually harvested in vineyards when they ripen. However, not all of the berries in a vineyard ripen homogeneously because of different microclimates around the clusters and berries. In this study, the influence of berry heterogeneity on the phenolic content and antioxidant capacity of grapes and wines under a continental monsoon climate was evaluated for a new wine grape cultivar Meili (Vitis vinifera L.). The total phenolic, flavonoid, flavanol, and monomeric anthocyanin contents in the skin and wine significantly increased with grape density; however, there was no significant difference in the seeds between the two lower densities. The highest values of DPPH free radical-scavenging activity, cupric-reducing antioxidant capacity, and hydroxyl radical-scavenging activity in the skin, seed and wine were detected for the densest berries. The sum of individual phenolic compounds in skin, seed and wine increased with berry density, though no significant difference for skin was observed between the two higher density classes. Hence, the chemical components of Meili grapes and wines were positively associated with the berry density at harvest under the continental monsoon climate.  相似文献

The Oligomeric proanthocyanidin (OPC) in green and black tea, grape seeds, grapes and wine has raised much attention but that OPC in wild grape seed remains to be intensively investigated. This study investigated the total OPC contents and total antioxidant activity of wild grape seeds and developed an efficient extraction process with various temperatures, solvent compositions and times. Also, a chromatography column packed with the Dia-ion HP-20 resin was used for further purification of the OPC. The total OPC contents were determined with the Folin-Ciocalteu reagent, and the antioxidant activity using total antioxidant potential (TAP) and 1,1-diphenyl-2picrylhydrazyl (DPPH). The yield of final purified OPC was 1.78 (+)-catechin equivalent (CE) g/100 g, with IC50 activities of TAP and DPPH of 31.60 and 15.70 μg/mL. These activities of the final purified OPC were about two times higher than that of the BHA used as a reference sample.  相似文献

The interaction of grape erineum mite (GEM), Colomerus vitis Pagenstecher (Acari: Eriophyidae), with grape was investigated in the laboratory. We studied some plant morphological biochemical features potentially related to vine resistance/tolerance of eight native grapevine cultivars, extensively cultivated in western Iran, and two non-native cultivars. Free-choice experiments indicated that the cultivars Shahani, Flame seedless and Yaghuti were colonized by lower levels of GEM, whereas Muscat Gordo, Gazne and White Thompson seedless hosted denser populations. These differences between cultivars may be due to differential attractiveness to GEM, possibly associated with plant biochemical and morphological traits. In no-choice assays with six grapevine cultivars, mite population development and some cultivar features were assessed. Mite populations grew fastest on Gazne and Muscat Gordo, and slowest on Yaghuti and Shahani. The degree of mite infestation was associated with reduction of leaf area, increase of leaf weight, shortening of shoots and more numerous erinea: these features were larger on the most infested Gazne, whereas morphological features of Shahani and Yaghuti were scarcely affected by GEM infestation. Also trichome type and density of the assayed cultivars appeared to be related to mite density: the most infested cultivars (Gazne and Muscat Gordo) displayed higher ranks of blade and vein hairs and lower ranks of blade and vein bristles and domatia. No correlation was found between mite density and leaf thickness of mature leaves. The amount of leaf waxes was highest in Shahani and Yaghuti, which displayed the lowest mite density, the fewest erinea and the largest leaves. Carbohydrate amount of uninfested leaves was lowest on the least infested Shahani and highest on the most infested Gazne; phenols increased in leaves of Shahani and decreased in those of Gazne after mite infestation. Finally, cultivars also appeared to influence some morphological traits of the mites: larger specimens were detected on White Thompson seedless, Flame seedless and Gazne, whereas smaller mites were found on leaves of the less infested Yaghuti and Shahani. These results indicate that leaf hairiness, leaf wax and carbohydrate contents may be useful tools for a preliminary screening among vine cultivars and help predict resistance/tolerance to GEM. Shahani and Yaghuti seem quite promising for developing grape resistance programs against GEM in western Iran.  相似文献

Grape seeds (GS) are one of the most important by-products of the winery and grape juice industry. GS contain a vast array of health-giving metabolites, including several polyphenols known for their remarkable antioxidant activity. In this study, it has been evaluated the nutraceutical potential of seeds from 15 grapevine cultivars/clones in terms of total phenols, monomeric, dimer, polymeric, gallate esters flavan 3-ols composition and in vitro antiradical activity. Considerable quali-quantitative differences in polyphenol content and AA had been observed among GS from different cultivars/clones collected in 2013 and 2014. In most of the investigated cultivars/clones, the better values of TP, FL and AA were recorded in GS collected in 2014. The polymeric procyanidins were significantly higher in 2014 in all the cultivars, while the gallate esters were significantly higher in 2013. These differences can be attributed to different climatic conditions in 2013 and 2014. In conclusion, the data here reported help to identify the best source of GS for the recovery of bioactive polyphenols and to valorize these waste materials.  相似文献

Indian mustard seeds were defatted by distillation with hexane and the residue extracted with methanol was analyzed for potential antioxidants; ascorbate, riboflavin, and polyphenols. Gallic acid (129.796 μg), caffeic acid (753.455 μg), quercetin (478.352 μg) and kaempferol (48.060 μg)/g dry seeds were identified by HPLC analysis of the extract. DPPH free radical scavenging activity and protection of lipids, proteins and DNA against metal induced oxidation was examined. Defatted mustard seed remnant had excellent free radical scavenging activity and protects biomolecules with IC50 value 2.0–2.25 mg dry seed weight. Significant content of polyphenols in methanol extract of defatted seeds accounts for high antioxidant potential. We are the first to report the detailed analysis of antioxidant composition and protection of biomolecules against oxidative damage by methanol extract of mustard seed remnant after oil extraction.  相似文献

Three hydrolyzable tannins, glansrins A-C, together with adenosine, adenine, and 13 known tannins were isolated from the n-BuOH extract of walnuts (the seeds of Juglans regia L.). Glansrins A-C were characterized as ellagitannins with a tergalloyl group, or related polyphenolic acyl group, based on spectral and chemical evidence. The 14 walnut polyphenols had superoxide dismutase (SOD)-like activity with EC(50) 21.4-190 microM and a remarkable radical scavenging effect against 1,1-diphenyl-2-picrylhydrazyl (DPPH) (EC(50) 0.34-4.72 microM).  相似文献

In both plants and animals, programmed cell death (PCD) is an indispensable process that removes redundant cells. In seedless grapes (Vitis vinifera), abnormal PCD in ovule cells and subsequent ovule abortion play key roles in stenospermocarpy. Metacaspase, a type of cysteine-dependent protease, plays an essential role in PCD. To reveal the characteristics of the metacaspase (MC) gene family and the relationship between metacaspases and the seedless trait, we identified the 6 V. vinifera metacaspases VvMC1VvMC6, from the grape genome, using BLASTN against the 9 known Arabidopsis metacaspases. We also obtained full-length cDNAs by RT-PCR. Each of the 6 grape metacaspases contains small (p10-like) and a large (p20-like) conserved structural domains. Phylogenetic analysis of 6 grape and 9 Arabidopsis metacaspases showed that all metacaspases could be grouped into two classes: Type I and Type II. Each phylogenetic branch shares a similar exon/intron structure. Furthermore, the putative promoters of the grape metacaspases contained cis-elements that are involved in grape endosperm development. Moreover, expression analysis of metacaspases using real-time quantitative PCR demonstrated that VvMC1 and VvMC2 were able to be detected in any tissue, and VvMC3, VvMC4, VvMC5 and VvMC6 exhibited tissue-specific expression. Lastly, in cv. Thompson seedless grapes VvMC1, VvMC3, and VvMC4 were significantly up-regulated at the 35 DAF during ovule development, roughly same stage as endosperm abortion. In addition, the expression trend of VvMC2 and VvMC5 was similar between cv. Pinot Noir and cv. Thompson grape ovule development and that of VvMC6 was sustained in a relatively low level except the expression of cv. Pinot Noir significantly up-regulated in 25 DAF. Our data provided new insights into PCD by identifying the grape metacaspase gene family and provide a useful reference for further functional analysis of metacaspases in grape.  相似文献

Polyphenols are the predominant ingredients in apple seeds. However, few data are available on the phenolic profile or antioxidant activity in apple seeds in previous researches. In this study, low-molecular-weight phenolic compounds and antioxidant activity in seeds, peels, and flesh of seven apple cultivars grown in northwest China were measured and analyzed using HPLC and FRAP, DPPH, ABTS assays, respectively. HPLC analysis revealed phloridzin as the dominant phenolic compound in the seeds with its contents being 240.45–864.42 mg/100 gDW. Total phenolic content (TPC) measured by the Folin–Ciocalteu assay in apple seed extracts of seven cultivars ranged from 5.74 (Golden Delicious) to 17.44 (Honeycrisp) mgGAE/gDW. Apple seeds showed higher antioxidant activity than peels or flesh; antioxidant activity in seeds varied from 57.59 to 397.70 μM Trolox equivalents (TE)/g FW for FRAP, from 37.56 to 64.31 μM TE/g FW for DPPH, and from 220.52 to 708.02 μM TE/g FW for ABTS. TPC in apple seeds was significantly correlated with all three assays. Principal component analysis (PCA) indicated that Honeycrisp was characterized with high contents of total polyphenols and phloridzin. Our findings suggest that phenolic extracts from apple seeds have good commercial potential as a promising antioxidant for use in food or cosmetics.  相似文献

Grapes are rich in phytochemicals with many proven health benefits. Phenolic profiles, antioxidant and antiproliferative activities of twenty-four selected Vitis vinifera grape cultivars were investigated in this study. Large ranges of variation were found in these cultivars for the contents of total phenolics (95.3 to 686.5 mg/100 g) and flavonoids (94.7 to 1055 mg/100 g) and antioxidant activities (oxygen radical absorbance capacity 378.7 to 3386.0 mg of Trolox equivalents/100 g and peroxylradical scavenging capacity14.2 to 557 mg of vitamin C equivalents/100 g), cellular antioxidant activities (3.9 to 139.9 µmol of quercetin equivalents/100 g without PBS wash and 1.4 to 95.8 µmol of quercetin equivalents /100 g with PBS wash) and antiproliferative activities (25 to 82% at the concentrations of 100 mg/mL extracts).The total antioxidant activities were significantly correlated with the total phenolics and flavonoids. However, no significant correlations were found between antiproliferative activities and total phenolics or total flavonoids content. Wine grapes and color grapes showed much higher levels of phytochemicals and antioxidant activities than table grapes and green/yellow grapes. Several germplasm accessions with much high contents of phenolics and flavonoids, and total antioxidant activity were identified. These germplasm can be valuable sources of genes for breeding grape cultivars with better nutritional qualities of wine and table grapes in the future.  相似文献

Summary The European, or bunch grape, Vitis vinifera, is widely grown because of its high fruit quality and its capacity to grow in a wide range of climatic conditions. However, they are susceptible to fungal diseases and insect pests, especially when grown in cool, wet climates. The aim of a number of grapevine breeding programs throughout the world is to develop new varieties resistant to diseases using complex hybrids between European and American species of Vitis. Within these breeding programs it is essential to maintain heterozygosity and desirable hybrids are multiplied by asexual propagation. New approaches to grapevine improvement include the use of protoplast fusion to overcome sexual barriers, however the routine regeneration of plantlets from protoplasts and calluses is difficult. In vitro rescue of ovules from varieties with stenospermocarpic seeds shows considerable promise for breeding new seedless grapes. Eventually the use of plant transformation techniques to insert specific pieces of DNA coding for desirable genetic characteristics will provide opportunities for equipping well known grape cultivars with new characteristics.  相似文献

选取广西、湖南等地野生葡萄,与经典酿酒葡萄比较,研究抗氧化活性和活性物质,同时监测葡萄酒发酵过程中各指标的动态变化,并对不同品种葡萄酒的抗菌性进行研究。结果表明:赤霞珠的酚类含量和抗氧化活性高于野生葡萄和玫瑰香葡萄,但野生葡萄酒的抗菌性能显著优于赤霞珠和玫瑰香葡萄酒。葡萄酒在发酵过程中其抗氧化活性和酚类物质含量均随发酵过程的进行而升高;总抗氧化活性与总酚含量、氧自由基清除能力与原花青素含量成显著正相关,相关系数均大于0.989;总花色苷含量在发酵初期上升,后期下降,葡萄酒颜色变浅。  相似文献

Hypothesizing that seed abortion in stenospermocarpic grapes (Vitis vinifera L.) is caused by high gibberellin levels in the seed during the first stages of its development, we studied the effect of gibberellic acid GA3 and uniconazol (a GAs biosynthesis inhibitor) on this phenomenon. In vitro germination was analyzed in the seedless cultivars Emperatriz and Perlon, which were treated with 60 and 120 mg.-l 1 uniconazol (5 and 15 days before bloom) and 100 mg.-l 1 GA3 (5 days after bloom). In addition, endogenous levels of free gibberellins in flowers and seeds of Emperatriz and Perlon were compared with their seedeed progenitor Emperador. Clusters were harvested at bloom and 20 days after bloom for gibberellin analysis and at commercial maturity for in vitro culture of the seeds. Considerable gibberellin activity was found in the three cultivars, but only small differences were detected between the seedless and the seeded genotypes. Exogenous applications of GA3 had a deleterious effect on seed growth and on in vitro germination. Uniconazol also inhibited in vitro germination, though not affecting the total number of germinating embryos plus those rescued from non-germinating seeds. In conclusion, gibberellins do not appear to be directly involved in seed abortion of the stenospermocarpic cultivars Emperatriz and Perlon, although their participation in a more complex scenario should not be rejected, taking into account that in Perlon germination rates are positively correlated with the number of clusters per plant. Treatments with growth regulators also modified berry number per cluster, berry weight and rachis morphology. Finally, the plant source was a determinant affecting germination rates in vitro.  相似文献
