The patterns of serum luteinizing hormone (LH), follicle stimulating hormone (FSH), progesterone and estradiol-17β during the estrous cycle of six crossbred (Alpine × Nubian × Native) and six native goats showing a 21 day estrous cycle in a semiarid zone of Venezuela are presented. In the crossbred goats, FSH had two significant peaks on Days 19 and 0 (33 ± 8.6 ng ml−1 and 25 ± 6 ng ml−1, respectively); in contrast, native goats only had one significant peak on the day of estrus (22 ± 2 ng ml−1), with the increase beginning on Day 17. During the follicular phase of crossbred goats, estradiol-17β and LH increased to 28 ± 6 pg ml−1 and 23 ± 6.9 ng ml−1, respectively, on Day 0. Prior to Day 0, LH increased to 10.0 ± 4.9 ng ml−1 on Day 18, decreasing to 1.5 ng ml−1 on Day 19, while estradiol-17β was increasing. This relationship between estradiol-17β and LH was not found to exist in native does, which presented a LH peak on Day 0 (30 ± 8 ng ml−1 and 35 ± 10 ng ml−1 in first and second estrus, respectively). LH basal levels were notably higher in native does. The highest concentrations of progesterone (10 and 12 ng ml−1) were detected on Days 12 and 15 in crossbred and native females, respectively. In conclusion, the relationship between estradiol-17β and gonadotropins during the follicular phase in crossbred goats suggests negative and positive feedback effects on both LH and FSH. Serum concentrations of LH were higher in native than in crossbred goats, whereas concentrations of FSH were higher in crossbred does. Thus, genetic factors need to be taken into account when comparing blood levels of gonadotropins in goats raised in tropical semiarid zones.  相似文献

Abstract

Pituitary LH is a key factor in controlling Leydig cell activity. In order to verify how steroids produced by the male gonads could influence testicular insulin binding, different steroids at varying doses were administered. Testosterone propionate (Tp) as well as estradiol-17β (E2-17β) and 5α-dihydrotestosterone (DHT) led to a 50% decrease of insulin binding in testis while no effect was noted when HCG was administered concomitantly with DHT. LH receptors were parallely measured: DHT and E2 -17β significantly depressed testis LH binding. It is concluded that steroids play a role in the regulation of testis membrane insulin receptor via their feedback mechanism on pituitary LH.  相似文献

A series of experiments were conducted to determine the effects of several estrogens upon FSH and LH secretion in immature ovariectomized rats. Groups of animals were castrated at 26 days of age and treated for 5 days post-operatively with various dosages of one of the following steroids: estrad1ol-17β(E2), estradiol benzoate (EB), estrone (E1), equilenin (EQ), 17α-ethinyl estradiol (EE), or mestranol (ME). Uterine weights were recorded and blood taken for radioimmunoassay.Estradiol was able to suppress both FSH and LH within a “physiologic dosage range” (PDR), wherein both gonadotropins were suppressed to intact control levels by a dose which did not stimulate uterine weight higher than intact control weight. EB and ME suppressed LH but not FSH at the PDR; the other steroids suppressed at higher than PDR doses. LH was preferentially suppressed, as compared to FSH, by all 6 steroids. By biological potency the order of activity was E2 = EE ? EB ? ME ? E1 ? EQ. For relative ability to suppress FSH (compared to LH), the order was E1 or E2, ? ME ?EB ? EE ? EQ. At higher doses (near maximum uterine stimulation), e1, E2 and EE produced higher FSH and LH than suppressed levels seen at lower doses; a pharmacologie dosage of E2 caused re-suppression of both gonadotropins.Results indicate that a feedback system is present before puberty and this system is sensitive to very low levels of estrogens. Likewise, there is a potential for positive feedback present for higher estrogen levels, and a complete suppression occurs at pharmacologie levels. Various naturally occurring steroids, synthetic steroid derivatives and non-steroidal hormone agonists and antagonists were assayed as inhibitors of human placental 17β-HSD activities. Microsomal 17β-HSD was inhibited by C18 -,C19- and C21-steroids. Soluble 17β-HSD was highly specific for C18-steroids. In contrast to the soluble activity, the microsomal enzyme also had a strong affinity for ethinylestradiol (KI=0.3 μM) and danazol (KI=0.6 μM); anabolic steroids and norethisterone were weaker inhibitors. Of the non-steroids tested only diethylstilbestrol and o-demethyl CI-680 were inhibitors and they showed a greater affinity for soluble 17β-HSD.KI-values for estradiol-17β, (0.8 μM), progesterone (27.0 μM) and 20α-dihydroprogesterone (1.5 μM) were comparable to reported tissue levels of these compounds, consistent with a possible competition in vivo among naturally occurring C18-, C19-, and C21-steroids for the active site of microsomal 17β-HSD.  相似文献

The steroids 7 alpha,17 beta-dihydroxy-5 alpha-androstan-3-one (7 alpha-hydroxy-Dht), 5 alpha-androstan-3 alpha,7 alpha,17 beta-triol (7 alpha-hydroxy-3 alpha-A'DIOL) and 5 alpha-androstane-3 beta,7 alpha,17 beta-triol (7 alpha-hydroxy-3 beta-A'DIOL) have been synthetized from 7 alpha,17 beta-dihydroxy-4-androsten-3-one (7 alpha-hydroxy-testosterone). The effect of administering 7 alpha-hydroxy-Dht, 7 alpha-hydroxy-3 alpha-A'DIOL or 7 alpha-hydroxy-3 beta-A'DIOL on serum levels of LH, FSH and on ventral prostate and seminal vesicle weight were investigated in gonadectomized adult male rats. Each steroid was administered for seven days in a dose of 300 micrograms per day. No suppression of serum LH or FSH levels was recorded following injections of these 7 alpha-hydroxylated steroids to castrated rats, compared to castrated control rats receiving vehicle only. Administration of 7 alpha-hydroxy-Dht or 7 alpha-hydroxy-3 alpha-A'DIOL to castrated mature rats could maintain ventral prostate and seminal vesicle weights above that of castrated control rats. Administration of 7 alpha-hydroxy-3 beta-A'DIOL to castrated mature rats resulted in ventral prostate weights slightly above castrate control levels, while seminal vesicle weight in such rats were in the same range as castrated control rats. Intraperitoneal administration of testosterone or of 5 alpha-androstane-3 beta,17 beta-diol (3 beta-A'DIOL) to castrated rats maintained activity of the androgen dependent isoenzyme of acid phosphatase in the ventral prostate; 7 alpha-hydroxy-testosterone or 7 alpha-hydroxy-3 beta-A'DIOL showed, however, no effect on this enzymic activity.  相似文献

ABSTRACT: BACKGROUND: The gonadotropins (GtHs), follicle-stimulating hormone (FSH) and luteinizing hormone (LH) are produced in the pituitary gland and regulates gametogenesis through production of gonadal steroids. However, respective roles of two GtHs in the teleosts are still incompletely characterized due to technical difficulties in the purification of native GtHs. METHODS: Native FSH and LH were purified from the pituitaries of adult chub mackerel, Scomber japonicus by anion-exchange chromatography and immunoblotting using specific antisera. The steroidogenic potency of the intact chub mackerel FSH (cmFSH) and LH (cmLH) were evaluated in mid- and late-vitellogenic stage follicles by measuring the level of gonadal steroids, estradiol-17beta (Epsilon2) and 17,20beta-dihydroxy-4-pregnen-3-one (17,20beta-P). In addition, we evaluated the maturation-inducing potency of the GtHs on same stage follicles. RESULTS: Both cmFSH and cmLH significantly stimulated E2 production in mid-vitellogenic stage follicles. In contrast, only LH significantly stimulated the production of 17,20beta-P in late-vitellogenic stage follicles. Similarly, cmLH induced final oocyte maturation (FOM) in late-vitellogenic stage follicles. CONCLUSIONS: Present results indicate that both FSH and LH may regulate vitellogenic processes, whereas only LH initiates FOM in chub mackerel.  相似文献

We have demonstrated previously an ability of estrogen to inhibit ovarian androgen production. We report here further evidence in support of this intraovarian short-loop feedback mechanism. Thecal cells from ovarian follicles of estradiol-17β (E)-treated rats demonstrated an enhanced capability of producing progesterone in response to LH in vitro. In contrast, testosterone production by the same thecal preparations was markedly inhibited by pretreatment with E, suggesting a selective inhibitory action of E at the level of the androgen-producing cells in the ovarian follicle. In a somewhat contrasting experiment in hypophysectomized rats, while simultaneous administration of purified follicle-stimulating hormone (FSH) antagonized an inhibitory action of E on ovarian progesterone production, treatment of the hypophysectomized rats with either E alone or concomitantly with E plus FSH still attenuated ovarian testosterone production by these animals in response to acute LH stimulation. These results are consistent with a direct inhibitory action of estrogen at the level of the ovarian C17α-hydroxylase /C17,20-lyase enzyme system.  相似文献

LH and FSH release of immature male rats was remarkedably enhanced by LH-RH if primed with LH-RH one hour before. The effect of exogenous steroids on the action of the second LH-RH was investigated. C-18, C-19 and C-21 steroids in different doses were tested. Blood samples were collected from the jugular vein immediately before and 30 min after the second LH-RH injection. Serum LH and FSH were determined by respective radioimmunoassays. The concomitant increase of LH and FSH was not induced by all the steroids administered iv. Estrone or 17alpha-hydroxyprogesterone suppressed LH and FSH release. Estradiol-17beta preferentially suppressed LH release. Cortisone, progesterone or dehydroepiandrosterone significantly facilitated FSH release, whiel 20alpha-dihydroprogesterone or 20beta-dihydroprogesterone selectively promoted LH release. The sc injection of most steroids dissolved in oil tended to augment the acute release of LH but not FSH. 20alpha-Dihydroprogesterone was particularly potent in this concern. Dehydroepiandrosterone or androstenedione was effective in maintaining FSH release for a longer period. These data revealed that potentiated LH and FSH release induced by the second LH-RH was readily modified by steroids administered simultaneously.  相似文献

To determine when undifferentiated rabbit granulosa cells first develop the capacity to secrete progesterone, pieces of intact ovaries from neonatal rabbits (newborn—30 days old) and pure granulosa cells from 150–1200 μm follicles at 60–600 days old were cultured in vitro for 6–10 days with human chorionic gonadotropin (HCG), Pergonal (LH/FSH), dibutyryl cyslic AMP (Bu2CAMP), prostaglandin E2, estradiol-17β, and as controls. The culture medium was collected every 2 days, and progesterone, estrone, and estradiol-17β were measured by radioimmunoassay.None of the neonatal ovaries or granulosa cell cultures secreted estrone or estradiol-17β spontaneously or in response to stimulation by gonadotropins, Bu2CAMP, or prostaglandin E2.Control cultures of newborn and 7-day-old ovaries did not secrete progesterone, but ovaries from 17- and 30-day-old rabbits did. Gonadotropins and Bu2CAMP induced progesterone secretion in 7-day-old ovaries and stimulated its production 5-10-fold in ovaries at 17 and 30 days old, but prostaglandin E2 and estradiol-17β were without effect.Granulosa cells from all antral follicles (200–1200 μm) secreted progesterone spontaneously, and its production was stimulated 100–1000-fold with gonadotropins and Bu2CAMP, but not with estradiol-17β or prostaglandin E2. In contrast, granulosa cells from 100–150 μm preantral follicles from 200-day-old animals did not secrete progesterone under these culture conditions.These results demonstrate that rabbit granulosa cells differentiate the capacity to secrete progesterone at the time the primary follicle develops an antrum, and suggest the differentiation process involves the acquisition of the capacity to respond to gonadotropins perhaps by the synthesis or unmasking of gonadotropin receptors.  相似文献

This study was carried out to determine the effects of a transient doe-litter separation on plasma prolactin, LH, FSH, estradiol-17β and progesterone concentrations before artificial insemination and during the subsequent pregnancy. Control does (n=12) had free access to nursing, whereas separated does (n=12) were kept away from their litters for 48 hours before artificial insemination. Both groups were inseminated on day 11 after parturition. Teat stimulation by suckling caused a high increase in prolactin concentrations in separated does (p<0.0001). Basal prolactin concentrations were observed in both groups on days 8 and 18 of pregnancy. No effect of the treatment was detected on LH and FSH concentrations during the sampling period. A rise of estradiol-17β concentrations was observed 48 hours after doe-litter separation, compared to control does and to previous values (p<0.003). Both groups showed low progesterone concentrations before artificial insemination. Pregnant rabbits in both groups showed increased progesterone concentrations on days 8 and 18 of pregnancy. Lower estradiol-17β concentrations were observed in control does on day 18 of pregnancy compared with separated rabbits (p<0.003). The results suggest that a transient separation of nursing does from their litters before artificial insemination may promote high follicular steroidogenesis activity leading to increased estradiol-l7β concentrations. This hormonal change could be a result of several stimulatory actions probably triggered by the absence of suckling episodes and may affect the luteotrophic function during the subsequent pregnancy.  相似文献

Administration of 100 μg of testosterone (T) daily for 14 and 28 days to 7-day castrate rats restored the weight of the ventral prostate to a level which slightly exceeded that of the controls. Ventral prostate weight in groups receiving estradiol-17β (E2) doses of 10, 50, 100, 200, or 500 μg administered simultaneously with 100 μg of T did not differ significantly from intact controls, although the weights were lower at E2 levels greater than 100 μg. Body weights of the castrated rats receiving 100 μg of T did not differ from those of sham castrated controls. However, mean body weights of all groups which received E2 (10 to 500 μg) simultaneously with 100 μg of T were significantly less than (p< .025 or less) those of the sham castrated controls. Analysis of normalized ventral prostate weights, i.e., mg ventral prostate/100 gm body weight, showed that E2 does not antagonize T and revealed a trend which suggested that low levels of E2 (10, 50 and 100 μg) may have enhanced the restorative effects of 100 μg T. Our data indicate that 100 μg of T approaches a physiological dosage for castrated rats and that in contrast to the possible enhancement of its restorative effects on the ventral prostate by low leve E2, its body weight stimulating effects are clearly impaired by E2.  相似文献

Chronic (2-4 days) constant-rate infusions of mammalian gonadotropin releasing hormone (GnRH) were performed in female bullfrogs, Rana catesbeiana. The magnitude and temporal relationship of profiles of plasma follicle-stimulating hormone (FSH), luteinizing hormone (LH) and sex steroids [testosterone (T), estradiol-17 beta (E2) and progesterone (P)] during GnRH infusion were dependent on ovarian stage. However, in all females, the same biphasic increase in plasma gonadotropins was apparent and initial elevations in gonadotropins were accompanied by correlated increments in plasma T and E2. Complete pituitary "desensitization" to chronic GnRH infusion was not observed. Females in early follicular stages were relatively unresponsive to infusions of 1.0-10.0 micrograms/h GnRH; elevations in plasma LH were marginal and FSH was unchanged. Females with fully developed (preovulatory) ovaries were more responsive: infusion of 1.0 micrograms/h GnRH produced significant elevations in plasma LH by 2 h followed by even larger increases ("surges") after 12 h. This LH "surge" was preceded by a decline in plasma T and E2 and was accompanied by abrupt elevations in plasma P and by ovulation. Postovulatory females showed a more gradual and smaller increase in plasma LH. Infusion of GnRH in the female bullfrog establishes a clear relationship between pituitary responsiveness and the ovarian cycle not evident from acute GnRH injection; GnRH was most effective immediately before ovulation. These data are also the first to detail periovulatory changes in plasma gonadotropins and ovarian steroids in an amphibian.  相似文献

This study was undertaken to investigate the effects of supplementation of serum (fetal calf serum), gonadotropins (LH, FSH, prolactin) and estradiol-17 beta (E2) to culture medium during in vitro maturation of bovine cumulus oocyte complexes on subsequent fertilization and development to the blastocyst stage in vitro. Serum supplementation during bovine oocyte maturation was not required but hormonal supplementation, gonadotropins (LH + FSH) and E2, enhanced the fertilizability and developmental ability of bovine oocytes matured in vitro. The addition of prolactin to maturation medium containing LH, FSH, and E2 did not further enhance frequencies of fertilization and development.  相似文献

A method is described for the purification and quantitation of estradiol-17β (E2), estrone (E1), testosterone (T), androstenedione (A) and progesterone (P) from a single plasma sample extraction. A combination of Sephadex LH-20 column chromatography and thin layer chromatography is used to separate the phenolic and neutral steroids; quantitation of the purified steroids is by radioimmunoassay (E2, E1) and competitive protein binding (T,A,P) techniques. The precision and accuracy of this method is comparable with that reported by others. A preliminary investigation of the temporal relationship between estrogens androgens, progesterone and serum LH during five normal and one short luteal phase menstrual cycle is presented.  相似文献

Eleven heifers, between 63 and 197 days of age, were exposed to 18 hr light/day (L) or natural photoperiods (N), beginning October 19, 1979. They were ovariectomized 8 weeks later. LH concentrations after ovariectomy were not affected by photoperiod, but the rate of increase of FSH after ovariectomy was greater (P<0.10) for group L than for group N. Three weeks after ovariectomy, heiters were injected, IV, with 0.1 mug/kg estradiol-17beta. LH concentrations initially decreased after injection. This was followed by a series of pulses larger than those prior to injection. FSH concentrations declined after injection and remained low throughout the sampling period. The net response of LH concentrations to estradiol (mean post-injection concentration minus mean pre-injection concentration) was greater (P=0.05) for group L (4.7 +/- 0.49 ng/ml) than for group N (2.9 +/- 0.37 ng/ml). Photoperiod did not affect the net response of FSH concentrations to estradiol. We concluded that exposing prepubertal heifers to 18 hr light/day during the winter resulted in a greater rate of increase of FSH after ovariectomy and greater estrogen-induced LH release. Because the response of LH to estradiol-17beta differed from the response of FSH, these hormones may be regulated differently.  相似文献

Inhibition of human placental 17β-hydroxysteroid dehydrogenase by C18 and C19 steroids and nonsteroidal alcohols was assayed at pH 9.0 with 17β-estradiol 3-methyl ether and NAD+ as reactants. The nonstaroidal alcohols tested were poor inhibitors. Cyclopentanol and cyclohexanol had Ki values greater than 5 mm. Nonaromatic C18 and C19 steroids with oxygen functions at both positions 3 and 17 gave no detectable inhibition or had Ki, values greater than or equal to 160 μm. 3μ-Hydroxy-5,16-androstadiene, 5-androsten-3β-ol, 1,3,5(10)-estratrien-3-ol, and 1,3,5(10),16-estratetraen-3-ol, steroids lacking a C(17) oxygen function, had Ki values of 1.8, 6.0, 0.04, and 0.17 μm, respectively, demonstrating that both C18 and C19 steroids can bind at the steroid site. Binding specificity is narrowed and binding affinity for nonaromatic steroids weakened by oxygen functions at C(17) or both C(3) and C(17). The structural implications of the specificity data for steroid recognition and complex formation and in vivo control of enzyme activity are discussed.  相似文献

Concentrations of luteinizing hormone (LH), follicle stimulating hormone (FSH) and prolactin (PRL) were measured in jugular blood and those of oestradiol-17β (E217β) in utero-ovarian blood. Samples were taken from five intact gilts every 15 min for 108 h starting between day 15 and day 18 of the oestrous cycle. In the late luteal/early follicular phase, high pulsatile LH secretion, close to one pulse per hour, was observed. This could be the stimulus necessary for the final maturation of the ovarian follicles.Thereafter, frequency and amplitude of pulses, and the baseline value, decreased and were low at least between 36 and 12 h before the preovulatory LH surge. PRL and FSH concentrations also declined. This was probably due to the increase of oestrogen secretion. As E217β concentrations were still high, the surge of LH which was accompanied by increase in FSH and PRL, occurred for approximately 13 to 20 h. While LH and PRL mean levels decreased, FSH concentrations continued to increase. Peaks of PRL were observed during the late luteal/early follicular phase and during the LH discharge. During the period of estrus, each exposure to the boar was immediately followed by one of these peaks, which could play a role in the sexual behavior of the gilt.  相似文献

The steroid secreting activities of dispersed granulosa and theca interna cells from preovulatory follicles of prepubertal gilts 72 h after pregnant mare's serum gonadotropin treatment (750 IU) were compared. The cells were cultured for 24 h with or without steroid substrate (10(-8) to 10(-5) M progesterone, 17 alpha-hydroxyprogesterone, or androstenedione), FSH (100 ng/mL), LH (100 ng/mL), and cyanoketone (0.25 microM, an inhibitor of 3 beta-hydroxysteroid dehydrogenase). Granulosa cells cultured alone secreted mainly progesterone. Theca interna cells secreted mainly 17 alpha-hydroxyprogesterone and androstenedione, with secretion being markedly enhanced by LH. In the presence of cyanoketone, which inhibited endogenous progesterone production, theca interna but not granulosa cells were able to convert exogenous progesterone to 17 alpha-hydroxyprogesterone and androstenedione, and exogenous 17 alpha-hydroxyprogesterone to androstenedione and estradiol-17 beta in high yield. The secretion of the latter steroids from exogenous substrates was unaffected by LH. Theca interna cells secreted more estradiol-17 beta than did granulosa cells in the absence of aromatizable substrate, but estradiol-17 beta secretion by the latter was markedly increased after the addition of androstenedione. These apparent differences in steroid secreting activity between the cell types suggest that the enzymes responsible for conversion of C21 to C19 steroids, i.e., 17 alpha-hydroxylase and C17,20-lyase, reside principally in the theca interna cells. However, aromatase activity appears to be much higher in granulosa cells.  相似文献
