In phototrophically grown Chlamydomonas cells, ammonium strongly inhibited the utilization of nitrate or nitrite. Under darkness, or in the presence of an uncoupler or inhibitor of the non-cyclic photosynthetic electron flow, the utilization of nitrate, nitrite or ammonium was suppressed. l-Methionine-d,l-sulfoximine (MSX) or azaserine, which blocks the assimilation of ammonium, inhibited the consumption of nitrate, but not nitrite, by the cells. Ammonium produced an immediate inhibition of the permease for nitrate in Chlamydomonas growing with nitrate, while ammonium-grown cells lacked this permease. The synthesis of nitrate-reductase activity was dependent on an active permease. In N-starved Chlamydomonas cells, previously treated with MSX, the permease for nitrate was insensitive to inhibition by ammonium, and a significant amount of nitrate reductase was synthetized. These cells photoproduce ammonium by reducing nitrate. Catabolic repression in the synthesis of inducible enzymes by glucose, fructose, and intermediates of the glycolytic cycle has been observed in many microorganisms. In order to enhance the polygalacturonase (PG) production of Aspergillus niger GJ-2, Saccharomyces cerevisiae J-1 was inoculated to the medium at 12 h of culture, which resulted in a significant improvement of PG production. It was also found that maximum PG activity of 512.7 U/ml was obtained at 37 °C in the mixed culture, which was nearly twofold higher than that of the culture without the inoculation of S. cerevisiae J-1.  相似文献

【目的】近年来,红螯螯虾养殖面积越来越广泛,明确不同规格的红螯螯虾对氨氮和亚硝酸盐的耐受力,有利于提高其养成率,促进其养殖业的健康发展。【方法】在水温24~25℃、p H 7.9~8.0的条件下,研究了氨氮和亚硝酸盐对红螯螯虾幼虾和亚成虾的急性毒性,分析半致死浓度(LC50)和安全浓度(SC)。【结果】总氨氮对红螯螯虾幼虾的24、48、72和96 h LC50分别为188.0、136.15、104.67和88.00 mg·L~(-1),SC为8.80 mg·L~(-1);总氨氮对亚成虾的24、48、72和96 h LC50分别为344.01、270.46、205.15和167.68 mg·L~(-1),SC为16.77 mg·L~(-1);非离子氨对幼虾的24、48、72和96 h LC50分别为10.16、7.35、5.65和4.75 mg·L~(-1),SC为0.48 mg·L~(-1);非离子氨对亚成虾的24、48、72和96 h LC50分别为18.58、14.60、11.08和9.05 mg·L~(-1),SC为0.91 mg·L~(-1);亚硝酸盐对幼虾的24、48、72和96 h LC50分别为46.76、33.88、27.97和22.81 mg·L~(-1),SC为2.28 mg·L~(-1);亚硝酸盐对亚成虾的24、48、72和96 h LC50分别为77.56、59.33、45.41和37.48 mg·L~(-1),SC为3.75 mg·L~(-1)。【结论】红螯螯虾对氨氮的耐受力高于亚硝酸盐,亚成虾对氨氮和亚硝酸盐的耐受力高于幼虾。  相似文献

Addition of nitrite to rapidly growing, nitrogen-fixing filaments ofAnabaena variabilis caused an immediate drop in nitrogenase activity. This was followed by a transient induction of nitrite reductase, recovery of nitrogen fixation and cyanobacterial growth. The effect of NH 4 + on the regulation of NO 3 and NO 2 transport systems in roots of intact barley (Hordeum vulgareL.) seedlings grown in NO 3 or NO 2 was studied. Ammonium partially inhibited induction of both transport systems. The inhibition was less severe in NO 2 -fed than in NO 3 -fed seedlings, presumably due to lower uptake of NH 4 + in the presence of NO 2 . In seedlings pretreated with NH 4 + subsequent induction was inhibited only when NH 4 + was also present during induction, even though pretreated roots accumulated high levels of NH 4 + . This indicates that inhibition may be regulated by NH 4 + concentration in the cytoplasm rather than its total accumulation in roots. L-Methionine sulfoximine did not relieve the inhibition by NH 4 + , suggesting that inhibition is caused by NH 4 + itself rather than by its assimilation product(s). Ammonium inhibited subsequent expression of NO 3 transport activity similarly in roots grown in 0.1, 1.0, or 10 mM NO 3 for 24 h (steady-state phase) or 4 d (decline phase), indicating that it has a direct, rather than general feedback effect. Induction of the NO 3 transport system was about twice as sensitive to NH 4 + as compared to the NO 2 transport system. 【背景】扶桑绵粉蚧是近年来入侵我国的重要检疫性害虫,在我国多个省份均有发生,造成了严重的经济损失。【方法】于2009—2015年对杭州地区扶桑绵粉蚧的发生危害情况进行了实地调查,同时结合mt DNA COI分子标记方法,对扶桑绵粉蚧进行分子鉴定和系统进化分析。【结果】杭州市余杭区、萧山区和临安市3个地区发现扶桑绵粉蚧分布,共调查到寄主植物38科61属68种,以辣椒、茄子、番茄、南瓜、棉花、芝麻、菊花、苦荬菜、大花马齿苋和五色梅受害最严重。系统进化分析表明,杭州地区的扶桑绵粉蚧未发生明显的遗传分化,与我国其他地区扶桑绵粉蚧mt DNA COI基因相似度为99.4%~100%。【结论与意义】本研究为进一步研究扶桑绵粉蚧的遗传进化、可能的侵入途径以及科学防控提供了理论基础。  相似文献

亚硝酸盐胁迫对罗氏沼虾血细胞及其抗氧化酶活力的影响   总被引:2,自引:0,他引:2  
【背景】亚硝酸盐是虾类集约化养殖过程中最常见的毒性污染物之一,研究亚硝酸盐胁迫对罗氏沼虾血细胞的毒性以及抗氧化酶在抗胁迫防御中的作用,能够为罗氏沼虾养殖过程中的亚硝酸盐中毒防治提供理论参考。【方法】以不同浓度(0、1、5和10 mg·L~(-1))的亚硝态氮(NO_2~--N)对罗氏沼虾进行胁迫,于胁迫后的0、6、12、24和48 h取样,应用流式细胞术检测血细胞活性氧(ROS)含量和细胞凋亡率,同时测定血细胞总数(THC)和胞内抗氧化酶活力。【结果】1 mg·L~(-1)NO_2~--N在48 h内对血细胞ROS含量、凋亡率和THC均无显著影响。5 mg·L~(-1)NO_2~--N胁迫24 h,血细胞ROS含量显著上升,THC显著下降,胁迫48 h凋亡率显著提高。10 mg·L~(-1)NO_2~--N胁迫6 h,血细胞ROS含量和凋亡率均显著上升,胁迫12 h THC显著下降。血细胞的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GPx)的活力均不同程度地被NO_2~--N胁迫所诱导,CAT活力主要在胁迫前期提高,而GPx活力在胁迫后期提高。【结论与意义】亚硝酸盐存在浓度和时间毒性效应,一定浓度的亚硝酸盐会诱导虾血细胞产生ROS,这些ROS的过量产生诱导了血细胞发生凋亡,继而导致THC下降,这一氧化胁迫过程可能是亚硝酸盐对罗氏沼虾产生细胞毒性的重要机制之一。抗氧化酶活力的诱导表明抗氧化酶在亚硝酸盐胁迫过程中发挥防御作用。  相似文献

Nitrite reductase (EC 1.6.6.4) prepared from pea roots was found to be immunologically indistinguishable from pea leaf nitrite reductase. Comparisons of the pea root enzyme with nitrite reductase from leaf sources showed a close similarity in inhibition properties, light absorption spectrum, and electron paramagnetic resonance signals. Cells of the obligately lithotrophic species Nitrosomonas europaea and Nitrosomonas eutropha were able to nitrify and denitrify at the same time when grown under oxygen limitation. In addition to oxygen, nitrite was used as an electron acceptor. The simultaneous nitrification and denitrification resulted in significant formation of the gaseous N-compounds nitrous oxide and dinitrogen, causing significant nitrogen loss. In mixed cultures of N. europaea and various chemoorganotrophic bacteria, the nitrogen loss was strongly influenced by the partners growing under oxygen limitation. Under anoxic conditions, pure cultures of N. eutropha were able to denitrify with molecular hydrogen as electron donor and nitrite as the only electron acceptor in a sulfide-reduced complex medium. The increase of cell numbers was directly coupled to nitrite reduction. Nitrous oxide and dinitrogen were the only detectable end products. In pure cultures of N. eutropha and mixed cultures of N. eutropha and Enterobacter aerogenes, ammonium and nitrite disappeared slowly at a molar ratio of about one when oxygen was absent. However, under these conditions cell growth was not measurable.  相似文献

The fatty acid profiles of all described species of the nitrite-oxidizing genera Nitrobacter, Nitrococcus, Nitrospina and Nitrospira were analyzed. The four genera had distinct profiles, which can be used for the differentiation and allocation of new isolates to these genera. The genus Nitrobacter is characterized by vaccenic acid as the main compound with up to 92% of the fatty acids and the absence of hydroxy fatty acids. The genus Nitrococcus showed cis-9-hexadecenoic acid, hexadecanoic acid and vaccenic acid as main parts. Small amounts of 3-hydroxy-dodecanoic acid were detected. The genus Nitrospina possessed tetradecanoic acid and cis-9-hcxadecenoic acid as main compounds, also 3-hydroxy-hexadecanoic acid was detected for this genus. The genus Nitrospira showed a pattern with more variations among the two described species. These organisms are characterized by the cis-7 and cis-11-isomers of hexadecenoic acid. For Nitrospira moscoviensis a specific new fatty acid was found, which represented the major constituent in the fatty acid profiles of autotrophically grown cultures. It was identified as 11-methyl-hexadecanoic acid. Since this compound is not known for other bacterial taxa, it represents a potential lipid marker for the detection of Nitrospira moscoviensis relatives in enrichment cultures and environmental samples. A cluster analysis of the fatty acid profiles is in accordance with 16S rRNA sequence-based phylogeny of the nitrite-oxidizing bacteria.  相似文献

Little is known about the responses of Australian plants to excess metal, including Mn. It is important to remedy this lack of information so that knowledgeable decisions can be made about managing Mn contaminated sites where inhabited by Australian vegetation. Acacia holosericea, Melaleuca leucadendra, Eucalyptus crebra and Eucalyptus camaldulensis were grown in dilute solution culture for 10 weeks. The seedlings (42 days old) were exposed to six Mn treatments viz., 1, 8, 32, 128, 512 and 2048 M. The order of tolerance to toxic concentrations of Mn was A. holosericea E. crebra < M. leucadendra < E. camaldulensis, the critical external concentrations being approximately 5.1, 5.0, 21 and 330 M, respectively. The critical tissue Mn concentrations for the youngest fully expanded leaf and total shoots were, respectively, 265 and 215 g g–1 DM for A. holosericea, 445 and 495 g g–1 DM for M. leucadendra, 495 and 710 g g–1 DM for E. crebra and 7230 and 6510 g g–1 DM for E. camaldulensis. The high tolerance of E. camaldulensis (as opposed to the sensitivity of E. crebra) to excess Mn raises concern about fauna feeding on the plant and is consistent with hypotheses suggesting the Eucalyptus subgenus Symphomyrtus is particularly tolerant of stress, including excess Mn. The results from this paper provide the first comprehensive combination of growth responses, critical external concentrations, critical tissue concentrations and plant toxicity symptoms for three important Australian genera, viz., Eucalyptus, Acacia and Melaleuca, for use in the management of Mn toxic sites.  相似文献

厌氧氨氧化(anaerobic ammonium oxidation, anammox)是微生物学、地质学和环境学领域的重要反应,厌氧氨氧化菌(anaerobic ammonium-oxidizing bacteria, AnAOB)是厌氧氨氧化的驱动器,探明AnAOB的生物学性状对厌氧氨氧化的应用具有重要意义。火山口结构是AnAOB的标志性微观结构,也是AnAOB的重要识别特征。由于迄今没有获得AnAOB纯培养物,相关研究进展缓慢。本文对AnAOB及其所归属的浮霉状菌的火山口结构研究进展作了综述,探讨了火山口结构的形态特征、生理功能和生态意义,得出以下结论:(1) AnAOB的火山口结构均匀分布在细胞表面,其直径约5 nm;(2) AnAOB的火山口结构推测向外可连通细胞外膜和内膜,向内可与厌氧氨氧化体膜相连,对于物质转运及转化具有重要意义;(3)火山口结构具有遗传稳定性,其形成可能与鞭毛脱落相关;(4) AnAOB的火山口结构可能通过促进细胞物质交流、信息通讯等在维持其生态位稳定方面起作用。  相似文献

The effects of decoctions prepared from Antrodia camphorata on antioxidant, nitrite scavenging, and antitumor activities were investigated. Glutathione (GSH) production was 18.2 μM/g of rat liver under the influence of a methanol extract, a result similar to with the effect of silymarin administration. GSH peroxidase activity was about 2.0-fold higher than with silymarin administration. Superoxide dismutase activity was 18.9 U/mg protein, about 2.5-fold higher than the control group. The hot water extract (1000 μg/mL) showed the highest nitrite scavenging activity at 98.1%, similar to those observed with BHA and vitamin E. Among a variety of human cancer cell lines, when the concentration of hot water extract was increased from 31 to 500 μg/mL, the viability of Hep3B cell was decreased from 100 to 60.7%. On the other hand, in the case of methanol extract, it was decreased from 98.4 to 10.0%. These results supported the conclusion that the antioxidant, nitrite scavenging, and antitumor activities of this A. camphorata methanol extract indicate a potential source for the development of various health supplements and pharmaceutical and nutraceutical applications.  相似文献

米氏凯伦藻和东海原甲藻是我国东南沿海地区赤潮的主要优势种。为定量获取米氏凯伦藻和东海原甲藻生长的温度生态幅,根据3个光照水平(28.32,75.06,111.66μmol m~(-2)s~(-1))条件下4个温度水平(18,22,25,28℃)对米氏凯伦藻和东海原甲藻生长特性的室内培养实验结果,并结合Shelford耐受性定律建立了基于温度的米氏凯伦藻和东海原甲藻比生长率的耐受性模型,最后根据前期的研究成果分别获取了米氏凯伦藻和东海原甲藻生长的最适温度、适温范围及耐受温度范围。结果表明,无论是米氏凯伦藻还是东海原甲藻,在相同培养光照条件下,在设定的温度水平范围内,分别存在一个适宜米氏凯伦藻和东海原甲藻的最适生长温度T_(opt),且当T≤T_(opt)时,米氏凯伦藻和东海原甲藻细胞密度和比生长率随着温度的升高而显著增大;而当T≥T_(opt)时,米氏凯伦藻和东海原甲藻细胞密度和比生长率随着温度的升高而显著减小。随着培养光照强度的升高,米氏凯伦藻和东海原甲藻细胞密度和比生长率均呈现"先升后降"的变化趋势。建立的藻类生长温度耐受性模型与谢尔福德耐受定律较为吻合,定量获取米氏凯伦藻在3个光照水平(28.32,75.06,111.66μmol m~(-2)s~(-1))下的最适生长温度分别为22.48,22.37,22.33℃;适温范围分别为17.93—27.03,17.82—26.92,17.78—26.88℃;耐受温度范围分别为13.38—31.58,13.27—31.47,13.23—31.43℃;东海原甲藻在3个光照水平(28.32,75.06,111.66μmol m~(-2)s~(-1))下的最适生长温度分别为22.10,21.99,21.93℃;适温范围分别为17.59—26.61,17.48—26.5,17.42—26.44℃;耐受温度范围分别为13.08—31.12,12.97—31.01,12.91—30.95℃。  相似文献

On transition from nitrogen starvation to ammonia or ammonia/glutamine sufficiency Phormidium uncinatum produces high amounts of H2O2, which is consumed by several oxidative reactions catalyzed by thylakoid membrane bound enzymes. These include: oxidation of glutamine to free hydroxylamine, of ammonia to nitrite, of bound hydroxylamine to nitrite, and dismutation of free hydroxylamine to ammonia and nitrite. 目的和方法 :应用高效液相色谱技术建立一种灵敏的检测不同体液中硝酸盐和亚硝酸盐的方法。结果 :唾液、血清及尿液经过不同方法处理后应用高效液相色谱ODS反相柱分离 ,紫外检测器于 2 10nm检测其中的硝酸盐和亚硝酸盐的含量。整个分离过程少于 7min ,硝酸盐和亚硝酸盐的测定线性范围分别为 0 .7~ 10 0ng、5~ 10 0ng ,最低检测极限分别为 0 .3ng和 2ng。硝酸盐回收率为 99%~ 10 2 % ,亚硝酸盐回收率为 99%~ 10 4 %。测定硝酸盐及亚硝酸盐的精密度分别为 0 .8%和 1.7%。结论 :本法测定硝酸盐和亚硝酸盐简便、灵敏度高、特异性好  相似文献

The utility of the dnaJ gene for identifying Vibrio species was investigated by analyzing dnaJ sequences of 57 type strains and 22 clinical strains and comparing sequence homologies with those of the 16S rDNA gene and other housekeeping genes (recA, rpoA, hsp60). Among the 57 Vibrio species, the mean sequence similarity of the dnaJ gene (77.9%) was significantly less than that of the 16S rDNA gene (97.2%), indicating a high discriminatory power of the dnaJ gene. Most Vibrio species were, therefore, differentiated well by dnaJ sequence analysis. Compared to other housekeeping genes, the dnaJ gene showed better resolution than recA or rpoA for differentiating Vibrio coralliilyticus from Vibrio neptunius and Vibrio harveyi from Vibrio rotiferianus. Among the clinical strains, all 22 human pathogenic strains, including an atypical strain, were correctly identified by the dnaJ sequence. Our findings suggest that analysis of the dnaJ gene sequence can be used as a new tool for the identification of Vibrio species.  相似文献

The adhesion of Nitrosomonas sp. and Nitrobacter sp. cells isolated from fishpond sediment to different solid particles was studied. Nitrosomonas and Nitrobacter cells rapidly attached to particles of bentonite, calcium carbonate, amberlite, and fishpond sediment, however they did not adhere to phenyl-sepharose beads. The nitrifying activity of attached bacteria was greater than the activity of freely suspended cells or the activity of cells which have been detached from CaCO3 particles. The enhancement in the nitrifying activity was rapid and was already observed within the first hour after attachment (which equals only 1/24 to 1/50 of the generation time of Nitrosomonas sp. or Nitrobacter sp. In addition, the survival of the attached bacteria under both anaerobic and under aerobic incubation was extended to weeks, compared to only a few days for the free cells. The presence of substrate (ammonia or nitrite) during the anaerobic incubation period was found not to affect the survival time of the bacteria. Finally, it was found that the attachment of Nitrosomonas and Nitrobacter cells to CaCO3 particles affected the dispersal and sinking rate of these particles.  相似文献

A phylogeny of the lichen family Porinaceae using mitochondrial SSU rDNA sequences is presented, with special focus on foliicolous taxa. Fifty specimens of 28 mostly tropical species, representing eight species groups of Porina as well as the genus Trichothelium, were analysed together with species of other members of Ostropomycetidae, and using Agyriaceae as outgroup. We performed the phylogenetic analyses with a Bayesian approach and under the criterion of maximum parsimony. Four main clades can be distinguished: the P. nitidula-group s. lat. (including Trichothelium, P. papillifera and P. rubescens), the Porina epiphylla-group s. lat. (including the P. radiata-, the P. nucula-, the P. imitatrix- and the P. epiphylla-group s. str.) and two clades of the P. rufula-group. The genus Porina as understood by all recent concepts is paraphyletic, and Trichothelium is nested within the Porina nitidula-group. The non-setose P. repanda forms a monophyletic clade with Trichothelium. The tree does not support a monophyletic origin of substrate preferences or photobiont selection. Species-specific associations with morphologically different trentepohlioid photobionts mapped on the tree suggest that closely related mycobiont species switch between different types of algae.  相似文献
