The skin of the silver marmoset, Callithrix (= Mico) argentata Linnaeus, has many outstanding characteristics. The unusually thin epidermis and dermis are generally devoid of melanotic melanocytes. The surface of the interramal gular region is peppered with small, elevated papillae; that of the corpus penis is characterized by keratinized, proximally directed hollow spines in whose cavities are organized nerve end-organs and capillary loops. Cholinesterase-reactive papillary nerve end-organs and Meissner corpuscles populate all volar friction surfaces. A highly vascular dermis and subcutaneous fat pad partially contribute to the characteristic rubicund color of the face, external ears, and anogenital region. Many body surface hairs are accompanied by the classic Haarscheibe of Pinkus. Sinus hairs are plentiful; a circumscribed vibrissa-bearing eminence is located on the ventral ulnar aspect of each wrist. Alkaline phosphatase-positive, stellate-shaped cells are found on the outer root sheath and epithelial sac of active and quiescent hairs in all cephalic regions; such cells are not argyrophilic, aurophilic, or osmiophilic. Hairs grow in linear perfect sets of 3 to 5 follicles; they do not grow singly on the cheek. Large sebaceous glands contain alkaline phosphatase in their peripheral acini; extensive fields of gigantic multilobular sebaceous glands contribute to the distinct, opaque-white color of the scrotal skin. Numbers of apocrine sweat glands are diminished in cephalic regions but one gland occurs with each hair follicle in the axilla, scrotum, suprapubic region, and ulnar eminence. Relatively avascular eccrine sweat glands are confined to the volar friction surfaces. Their secretory coils contain sporadically distributed glycogen and phosphorylase and surrounding nerves are much more reactive for acetyl- than butyrylcholinesterase.  相似文献

The skin of the white-browed capuchin (Cebus albifrons), although basically similar to that of the squirrel monkey (Saimiri sciureus), contains several outstanding peculiarities: (1) both the epidermis and dermis of the general body surface are devoid of melanotic melanocytes; (2) the skin of the prehensile tail has no modified, glabrous friction surface; (3) the prehensile surface of the tail has no specialized nerve end-organs; (4) cholinesterase-positive, papillary nerve end-organs rest beneath the epidermal ridges of volar skin; and (5) both the clear cells and dark cells of the eccrine glands contain glycogen but neither shows phosphorylase activity.  相似文献

The pigmy marmoset — Callithrix (= Cebuella) pygmaea Spix — is the second detailed study of the members of the family Callithricidae. It is closely allied to the red-mantled tamarin — Saguinus (=Tamarinus) fuscicollis illigeri Spix — and shares some of its characteristics with both Prosimii and Anthropoidea. The epidermis and dermis contain moderate numbers of concurrent, melanotic melanocytes. The dermis is rich in elastin. Hair follicles grow in groups of three or four over the general body surface, and one apocrine gland is associated with each grouping. Arrectores pilorum muscles are well developed. On the ventral ulnar wrist are sinus hairs associated with apocrine glands. Most hair follicles have nerve end-organs around them that are reactive for acetylcholinesterase and alkaline phosphatase. There is a large aggregation of sebaceous glands in the suprapubic region. The large sebaceous glands in the eyelid, face, and external genitalia are surrounded by cholinesterasereactive nerves. Apocrine glands are found over most of the hairy skin except the brow, scalp and back; a large grouping of them is present in the sternal region. Only the secretory coils of apocrine glands in the external genitalia are invested with butyrylcholinesterase-rich nerves. Eccrine glands are confined to the volar surfaces of the pes and manus. They have dark cells with abundant glycogen and clear cells with neither glycogen nor PAS-reactive material. The nerves around the eccrine secretory coil are reactive only for acetylcholinesterase.  相似文献

The skin of the pigmy bushbaby (Galago demidovii), the smallest existing prosimian, is largely similar to that of the other African Lorisidae, although this animal has certain peculiar features. The very thin epidermis contains alkaline phosphatase-reactive dendritic cells which resemble those in the other bushbabies and the potto. The hair follicles of this animal are similar to those of the lesser bushbaby, while the sebaceous glands are different from those of the other bushbabies in being reactive for alkaline phosphatase. The histological and histochemical properties of the sweat glands are similar to those of the other bushbabies.  相似文献

The skin of the golden spider monkey (Ateles geoffroyi) has many histological and histochemical similarities to that of the woolly monkey (Lagothrix lagotricha) and howler monkey (Alouatta caraya); however, this monkey possesses certain peculiar properties such as large sebaceous glands, a combined distributional pattern of eccrine and apocrine sweat glands, and abundant alkaline phosphatase in the sebaceous glands, apocrine and eccrine sweat glands. In brief, the anatomical and histochemical properties of the skin of this animal are more similar to those of the howler monkey than to the woolly monkey. In addition, the skin of these three Ceboids falls phylogenetically between that of the Cercopithecoidea and Pithecoidea.  相似文献

The close resemblance between the skin of Goeldi's marmoset (Callimico goeldii) and that of the genus Saguinus (Hoffmannsegg) suggests that C. goeldii be placed in the family Callithricidae (Thomas, '03) as originally proposed by Pocock in 1920 and not in the family Cebidae (Swainson, 1835). Recent karyological evidence supports this point of view. The skin is characterized by (1) a melanin-laden epidermis and aggregates of melanocytes in the upper dermis; (2) nonglandular skin in the gular region distinguished by a sparsely haired, pebbly, granular surface; (3) cholinesterase-positive Meissner corpuscles and papillary nerve end-organs; (4) groups of 3 to 5 hair follicles in linear perfect sets; (5) sinus hairs on the ventral ulnar wrist; (6) manubrial, sternal, genitopubic, and circumanal sebaceous-apocrine glandular concentrations rich in phosphorylase and alkaline phosphatase; (7) eccrine glands, restricted to volar friction surfaces, whose clear and dark secretory cells contain phosphorylase but no glycogen; and (8) eccrine glomeruli surrounded by many acetyl- but few butyrylcholinesterase-reactive nerves.  相似文献

Data on 84 characteristics of the skin of 36 species of primates were extracted from a series of articles describing the histological and histochemical properties of the skin of primates. The data were subjected to a cluster analysis. The results were in reasonably good agreement with orthodox primate taxonomies although some exceptions were apparent. The species clustered into four main groups approximately comparable to Prosimii, Cercopithecoidea, Ceboidea, and Hominoidea. The internal arrangements of the Prosimii, Cercopithecoidea, and Hominoidea are commensurate with standard taxonomic practice. Within the Ceboidea, however, the Atelinae and Alouattinae tend to group with the Hominoidea, Aotus and Saimiri show variable placements, and Callimico groups with the Callithricidae.  相似文献

We investigated the genotoxic properties of a number of extracts from Tunisian traditional medicinal plants with the bacterial VITOTOX test in Salmonella typhimurium and the alkaline comet assay in human C3A cells. Ethyl acetate and methanol extracts from Marrubium alysson L. and Retama raetam (Forsk.) Webb and methanol extracts from Peganum harmala L. were investigated. Toxicity was furthermore studied with the neutral red uptake test that served for dose-finding.All extracts showed antigenotoxic properties against 4-nitroquinoline-oxide (4-NQO) and benzo(α)pyrene in the VITOTOX test, except the methanol extracts from R. raetam where antigenotoxicity was not found against the mutagen 4-NQO (in the absence of S9). The ethyl acetate extract from R. raetam was found mutagenic with the VITOTOX test in the absence of S9, whereas both ethylacetate and methanol extracts of M. alysson L. induced DNA damage according to the alkaline comet assay in C3A cells.  相似文献

The state of amino acid residues in alkaline protease of Bacillus No. 221 and that of subtiiisin BPN’ were compared by spectrophotometric tiiration of tyrosine residues and by several reagents: β-naphtoqumone-4,6-disulfonic acid and monochlorofluoroquinone for amino groups, H2O2-dioxane for tryptophan, glyoxal for arginine, and tetranitromethane for tyrosine.

The reactivity of both proteases was fairly similar to those reagents.

The helix content of alkaline protease of Bacillus No. 221 (37%) was higher than that of subtilisin BPN’ (20%).

The Km and Vmax of alkaline protease of Bacillus No. 221 toward ATEE and BTEE were obtained from Lineweaver-Burk plot and compared with those of α-chymotrypsin and subtiiisin BPN’.  相似文献

Summary Fetal spinal ganglia and spinal cord segments with adhering spinal ganglia were explanted on collagen-coated coverslips. They were investigated with enzyme histochemical methods for the existence of hydrolases and dehydrogenases up to 54 days of cultivation.Alkaline phosphatase was located in arachnoid cells and in mantle cells (satellite cells). Butyrylcholinesterase and alpha-glycerophosphate-menadione-tetrazolium reductase were found in mantle cells. Acetylcholinesterase and indoxylesterase were active in the whole neuron; acid phosphatase and alpha-naphthylacetate esterase were restricted to the perikarya.During the process of cell differentiation in vitro alkaline phosphatase decreased in mantle cells. Acid phosphatase became diminished distinctly in spinal ganglion cells. The other neuronal enzymatic activities remained unaltered during the whole period of cultivation. Proliferated Schwann cells were conspicuous by their activity for butyrylcholinesterase. In newly formed myelin sheaths arylsulphatase was active. Lactate dehydrogenase was contained in the perineurium which had developed. The alkaline proteases of B. subtilis NRRL B3411, B. pumilis, and B. licheniformis have been isolated by fractionation followed by ion exchange chromatography and their homogeneity demonstrated. General enzyme properties of the B. sublitis NRRL B3411 alkaline protease have been studied and attempts made to differentiate a group of alkaline proteases. It is clear that the alkaline proteases known as Subtilisins or Subtilopeptidases are not, exclusive to B. subtilis but are common to many Bacilli and therefore the generic name Bacillopeptidases has been proposed. It is clear too that on the basis of the effect of pH on activity, amino acid composition, esterase activity, and immunological cross-reactions the Bacillopeptidases can be divided into two groups or types: (a) Bacillopcptidase A (Subtilisin A or Subtilopeptidase A) which includes Subtilisin Carlsberg, B. licheniformis, and B. pumilis alkaline proteases; ( b ) Bacillopeptidase B (Subtilisin B or Subtilopeptidase B) which includes B subtilis NRRL B3411, Subtilisin Novo, Subtilisin BPN' (Nagarse), alkaline protease Daiwa Kasei, and (probably) B. subtilis var. amylosacchariticus. At present, no further differentiation is possible and whether or not the enzymes within group A or B are identical remains an open question. Methods for examination of crude enzyme mixtures or fermentation beers are described and from the examination of a number of crude enzymes and fermentation beers it appears that organisms producing Bacillopeptidase A do not produce neutral protease or amylase, while organisms producing Bacillopeptidase B produce a neutral protease and amylase as well.  相似文献

The skin of Saguinus (= Oedipomidas) oedipus Linnaeus, is basically similar to that of the red-mantled tamarin, Saguinus (= Tamarinus) fuscicollis Spix; it has several peculiarities: (1) a circumscribed tuft of vibrissae on the ulnar aspect of the wrist; (2) an accumulation of apocrine glands over the sternum; and (3) an extensive posterior abdominal field of gigantic sebaceous glands admixed with large apocrine glands, better developed in the female. The epidermis, dermis, hair follicles, sebaceous ducts, and apocrine excretory ducts are all heavily pigmented. Hairs are arranged in linear perfect sets; the epithelial sac of quiescent follicles is devoid of glycogen and phosphorylase. Eccrine sweat glands are restricted to the volar friction surfaces and contain no glycogen. Only the coiled excretory ducts of the eccrine glands contain phosphorylase. All cutaneous nerve fibers are more reactive for acetylthan butyrylcholinesterase.  相似文献

Summary Specitinomycin-resistant mutants of Bacillus subtilis show three different types of alterations in sporulation ability. Class 1 mutants can both grow and sporulate in the presence of spectinomycin. Class 2 mutants can grow in the presence of spectinomycin, but are unable to sporulate in either the presence or absence of spectinomycin. Class 3 mutants have a conditional phenotype, and are able to sporulate in the absence of spectinomycin, but not in its presence. The ability of these strains to produce alkaline phosphatase, a biochemical marker for early sporulation events, is correlated with the ability to sporulate in the presence or absence of antibiotic. All of the spectinomycin-resistance mutations could be genetically linked to the cysA marker, and a mutational alteration of a protein of the 30S ribosomal subunit has been identified in one of the Class 3 strains (Spc1–11). Fine-structure mapping of the spectinomycin resistance mutation of strain Spc 1–11 confirmed its location in the cluster of genes for ribosomal components on the B. subtilis genetic map. Genetic analysis indicated that the properties of the Class 1 and Class 2 mutants result from more than one mutation. The spectinomycin-resistance and altered sporulation properties of the two Class 3 mutants probably result from a single genetic lesion.  相似文献

Indigo is an insoluble blue dye historically used for dyeing textiles. A traditional approach for indigo dyeing involves microbial reduction of polygonum indigo to solubilize it under alkaline conditions; however, the mechanism by which microorganisms reduce indigo remains poorly understood. Here, we aimed to identify an enzyme that catalyzes indigo reduction; for this purpose, from alkaline liquor that performed microbial reduction of polygonum indigo, we isolated indigo carmine-reducing microorganisms. All isolates were facultative anaerobic and alkali-tolerant Bacillus spp. An isolate termed AO1 was found to be an alkaliphile that preferentially grows at pH 9.0–11.0 and at 30–35 °C. We focused on flavin-dependent azoreductase as a possible enzyme for indigo carmine reduction and identified its gene (azoA) in Bacillus sp. AO1 using homology-based strategies. azoA was monocistronic but clustered with ABC transporter genes. Primary sequence identities were < 50% between the azoA product (AzoA) and previously characterized flavin-dependent azoreductases. AzoA was heterologously produced as a flavoprotein tolerant to alkaline and organic solvents. The enzyme efficiently reduced indigo carmine in an NADH-dependent manner and showed strict specificity for electron acceptors. Notably, AzoA oxidized NADH in the presence, but not the absence, of indigo. The reaction rate was enhanced by adding organic solvents to solubilize indigo. Absorption spectrum analysis showed that indigo absorption decreased during the reaction. These observations suggest that AzoA can reduce indigo in vitro and potentially in Bacillus sp. AO1. This is the first study that identified an indigo reductase, providing a new insight into a traditional approach for indigo dyeing.

The black-collared tamarin represents the first detailed study of a member of the family Callithricidae. Although certain features are unique, some of its characteristics resemble those of Prosimii and those of Anthropoidea. The epidermis is moderately pigmented, and the dermis contains numerous elestic fibers. Scattered melanocytes are found throughout both layers. Hair follicles grow in groups of 3 to 4 over the general body surface, including the muzzle. One apocrine gland is associated with each hair group. In the periinguinal region is a large concentration of gigantic sebaceous glands. Apocrine glands occur over the entire hairy skin. Aggregations of these glands on the ventral wrist (in conjunction with sinus hairs) and chest represent the ulnar gland and suprasternal gland, respectively. Apocrine secretory coils of the ulnar gland and external genitalia are surrounded by melanotic, dendritic melanocytes. Eccrine sweat glands are confined to the volar surfaces of the pes and manus. Cholinesterase-reactive granules are dispersed throughout the cytoplasm of the myoepithelial cells.  相似文献

Net inward transport of Cl in the absence of an electrochemical potential difference was demonstrated in the skin of two species of frog, R. pipiens and R. esculenta under conditions of low (2 mM) Cl concentration in the bathing solutions. The electrical potential profile of skins of R. pipiens was examined with microelectrodes under conditions in which the inside solution was negative relative to the outside solution. This reversal of the normal potential difference was found to arise as a result of potential changes across the outward facing electrical barriers in the skin. The reversed potential difference appears to arise, at least in part, as a result of the inward Cl transport. The effect may be due either to electrogenic Cl transfer or to variations in internal composition of the epithelial cells arising as a result of Cl transport.  相似文献

Both acid and alkaline invertases were present in immature juice sacs of satsuma mandarin (Citrus‘Unshu Marcovitch”) fruit, in which sugar content was low. Maturing and mature juice sacs, in which sugar content increased steadily with time, were characterized by the presence of alkaline invertase and the absence of acid invertase. When the immature juice sacs were homogenized with 0.2 M sodium phosphate-citrate buffer (pH 8.0), almost all of the acid invertase activity was found in the solubilized fraction, whereas almost all of the alkaline invertase activity was present in the insoluble fraction. The distribution of alkaline invertase between the solubilized and insoluble fractions changed with the development of fruit. The acid invertase had a molecular weight of 69,000, optimum pH of 4.8–5.3, and Km value for sucrose of 7.3 mM. The alkaline invertase had a molecular weight of 200,000, pH optimum of 7.2–7.7, and Km value of 35.7 mM. The hydrolysing activities of both enzymes for raffinose were considerably less than those for sucrose. The alkaline invertase had lower activity for raffinose than the acid invertase.  相似文献

Summary Skins fromRana temporaria, investigated with microelectrode techniques in the absence of Na uptake across the outer border (Na-free epithelial solution or amiloride), were found to be permeable to K+ at the apical membrane in 10–20% of the experiments. Full development of the K+-permeable state requires the absence of Na+ uptake for certain periods of time, which suggests that the K+-permeability of the apical membrane is higher at lower intracellular [Na]. The addition of Ba++ reduces the K+-permeability of the apical membrane. These skins may provide a model for the study of transcellular K+ movements.  相似文献

Studies with the induced lysogens of λS +R+, λS-R+, λS+R- and λS-R- phages have shown that while theS gene product is essential for the action of intracellularR gene product to release the periplasmic alkaline phosphatase in the presence of EDTA, the latter gene product can bring about this effect while acting onEscherichia coli cells from outside, in the absence of functionalS gene product; chloroform, could help the intracellularR gene product in effecting bacterial lysis in the absence ofS gene product. These result support the premise that theS gene product facilitates theR gene product in crossing the cytoplasmic membrane into the periplasmic space such that the latter can act on the peptidoglycan layer of the host cell thus causing both the release of alkaline phosphatase and cell lysis.
