Of numerous species belonging to the medicinally important genus Hypericum, only H. perforatum L. and H. maculatum Crantz grow widely in Estonia. A comparative biochemical systematics study of hypericins, hyperforins and other phenolics within Hypericum spp. growing in Estonia was performed. For comprehensive metabolomic investigation, 42 samples of H. perforatum and 16 samples of aerial parts of H. maculatum were collected in two consecutive years from various locations; methanolic extracts were prepared from airdried leaves and flowers. The concentrations of a quinic acid derivative, caffeic acid glucoside, vanillic acid glucoside, neochlorogenic acid, chlorogenic acid, catechin, epicatechin, myricetin glucoside, hyperoside, isoquercitrin, rutin, quercetin pentoside, quercitrin, kaempferol glucoside, kaempferol rutinoside, quercetin, hyperforin, adhyperforin, protopseudohypericin, pseudohypericin, and hypericin were determined by LC-DAD-MS/MS. All the aforementioned compounds were detected in both species, although some at very different levels – H. maculatum contained rutin and hyperforins only in trace amounts and overall tended to contain more phenolic compounds. The level of total hypericins was the same for both species. These results constitute a further contribution to the systematic knowledge about the Hypericum spp. Results of principal component analysis (PCA) demonstrated distinct between-years and between-species diversity in the chemical composition of the plants studied. Between-years diversity in Hypericum spp. has not been addressed before.  相似文献

 Cycle sequencing of the ITS region of nuclear ribosomal DNA of L. concolor, L. dauricum and L. maculatum generated surprisingly homogenous sequences from these three species. Analysis of the few (13 out of 639) polymorphic nucleotide sites in the ITS region produced results that do not support the belief that L. maculatum is a hybrid of the two other species. Neighbor-Joining analysis of the genetic distances calculated using the Kimura 2-parameter model of base substitution confirmed the close relationship between L. dauricum and L. maculatum. The phylogenetic tree, in conjunction with the distribution pattern and morphological similarities of the two species, suggest that L. maculatum is derived from the more widely distributed L. dauricum. The results also revealed that there is sufficient molecular divergence between L. maculatum and L. dauricum to support their status as separate species. Ecdysteroids were analysed with radioimmunoassay (RIA) and high-performance liquid chromatography (HPLC) in females of the apterygotous insect Thermobia domestica, which ahs overlapping moulting and reproductive cycles. During each moulting cycle, a peak in the haemolymph concentration of ecdysteroids occurs at day 9 (in the 11-day standard cycle), which can be correlated with apolysis and the beginning of new cuticle deposition. The ovaries show a peak of ecdysteroids at day 5 (i.e. one day before egg-laying), which suggests that these hormones are also involved in the reproductive cycle. In both cases, HPLC analysis combined with RIA suggests that the main ecdysteroid is 20-hydroxyecdysone. This duality in the function of ecdysteroids is discussed.  相似文献

Egg masses of the yellow-spotted salamander Ambystoma maculatum form an association with the green alga “Oophila amblystomatis” (Lambert ex Wille), which, in addition to growing within individual egg capsules, has recently been reported to invade embryonic tissues and cells. The binomial O. amblystomatis refers to the algae that occur in A. maculatum egg capsules, but it is unknown whether this population of symbionts constitutes one or several different algal taxa. Moreover, it is unknown whether egg masses across the geographic range of A. maculatum, or other amphibians, associate with one or multiple algal taxa. To address these questions, we conducted a phylogeographic study of algae sampled from egg capsules of A. maculatum, its allopatric congener A. gracile, and two frogs: Lithobates sylvatica and L. aurora. All of these North American amphibians form associations with algae in their egg capsules. We sampled algae from egg capsules of these four amphibians from localities across North America, established representative algal cultures, and amplified and sequenced a region of 18S rDNA for phylogenetic analysis. Our combined analysis shows that symbiotic algae found in egg masses of four North American amphibians are closely related to each other, and form a well-supported clade that also contains three strains of free-living chlamydomonads. We designate this group as the ‘Oophila’ clade, within which the symbiotic algae are further divided into four distinct subclades. Phylogenies of the host amphibians and their algal symbionts are only partially congruent, suggesting that host-switching and co-speciation both play roles in their associations. We also established conditions for isolating and rearing algal symbionts from amphibian egg capsules, which should facilitate further study of these egg mass specialist algae.  相似文献

Phytoecdysteroids, plant steroids which are analogues of invertebrate steroid hormones, probably contribute to the deterrence of phytophagous invertebrate predators. They also seem to possess antimicrobial activity and several pharmaceutical and medicinal benefits have been ascribed to them. Here. we present a survey of seeds of 16 species of the genus Asparagus (Asparagaceae), including the crop species A. officinalis, for ecdysteroid agonists (including phytoecdysteroids) and antagonists. Seven species were found to contain ecdysteroids with levels ranging from just detectable (A. racemosus and A. sarmentosus) to relatively high (A. laricinus). RP-HPLC/RIA/bioassay has been used to separate positive extracts of four species (A. falcatus, A. laricinus, A. ramosissimus and A. scandens) and analyse the ecdysteroid profiles. The identities of the major ecdysteroids were confirmed by NP-HPLC. Seeds of A. officinalis do not contain detectable levels of ecdysteroids, but leaves, stems and roots contain low levels (detectable by RIA). This indicates that A. officinalis retains the genetic capacity to synthesise ecdysteroids and that future strategies could be developed for enhanced protection of asparagus spears through elevated ecdysteroid levels.  相似文献

The essential oils of the aerial parts of nine species of Hypericum (Hypericum barbatum, Hypericum hirsutum, Hypericum linarioides, Hypericum maculatum, Hypericum olympicum, Hypericum perforatum, Hypericum richeri, Hypericum rumeliacum and Hypericum tetrapterum), collected from different locations in Southeast Serbia, were obtained by steam distillation and analyzed by GC and GC–MS. The essential oils investigated were characterized by a high content of non-terpene compounds and a low content of monoterpenes. The contents of non-terpenes, monoterpenes and sesquiterpenes in oils of the species H. barbatum, H. richeri and H. rumeliacum (section Drosocaprium) were similar and these oils were characterized by high contents of fatty acids. The oils of H. hirsutum and H. linarioides (section Taeniocarpium) contained a high percentage of n-nonane. There were similarities in contents of non-terpenes and sesquiterpenes in oils of species that belong to the section Hypericum (H. maculatum, H. perforatum and H. tetrapterum). The oil of H. olympicum differed from others by higher terpene content. A comparison was also carried out of the chemical composition of the essential oils from flower, leaf and stem of H. perforatum and it revealed that the highest concentration of non-terpene compounds was found in the flower and stem oil, while a high concentration of sesquiterpenes was characteristic for leaf oil. There were significant differences in the concentrations of the same compounds in the essential oils of H. maculatum, H. olympicum and H. perforatum, collected in different years from the same location which could be explained by seasonal differences. All data were statistically processed with principal component analysis and cluster analysis. The main conclusion from the above data is that genetic and environmental factors both play a role in determining the composition of essential oils of the Hypericum species studied.  相似文献

In the Ibaraki population (Japan) of Locusta migratoria, adult locusts produce diapause eggs under short-day (SD) conditions and non-diapause eggs under long-day (LD) conditions. The identity and titre of ecdysteroids in the ovaries and eggs from LD and SD adult females were investigated by RIA/HPLC. Maternal ecdysteroids accumulated in the developing ovaries represented about 90% polar conjugates, 5% free ecdysteroids and 5% non-hydrolyzable metabolites. Before oviposition the quantity of ecdysteroids reached 29.8±1.85 ng 20-hydroxyecdysone equiv. per mg tissue ovaries from LD females and 13.1±3.55 ng 20E equiv./mg in ovaries from SD females. The sum of RIA-positive materials in newly laid eggs was more than three times higher in non-diapause eggs than in diapause eggs. Ecdysteroids present in egg extracts comprised about 85% polar conjugates, 5% free ecdysteroids and 10% non-hydrolyzable metabolites. On the other hand, after diapause termination the amount of ecdysteroids increased drastically. Also, the composition of ecdysteroids differed from that observed during diapause and became comparable to that of non-diapause eggs. The significant differences in the ecdysteroids between non-diapause and diapause eggs may suggest the possible involvement of these compounds in the control of embryonic diapause of this locust.  相似文献

Ecdysteroids were detected in the phylum Nemertea and their physiological role was studied. Radioimmunoassay (RIA) measurements showed ecdysteroid concentrations ranging from 1–47 pg/mg wet weight in several nemertean species from the orders Palaeonemertea, Heteronemertea, and Hoplonemertea. High-performance liquid chromatographic (HPLC) analysis of Paranemertes peregrina displayed peaks of RIA activity with retention times similar to those of authentic ecdysone and 20-hydroxyecdysone standards. Fluctuating ecdysteroid titers were observed in the various life stages of Carcinonemertes errans with the highest concentrations (47 pg/mg wet weight) found in gravid females. RIA of HPLC fractions of Carcinonemertes errans eggs indicated the presence of ecdysteroids (105 pg/mg wet weight). Alterations in the growth of juvenile, male, or female C. errans were not observed when the worms were exposed to 10–7. 10–6, or 10–5 M 20-hydroxyecdysone. However, the eggs of C. errans appeared to be stimulated by 20-hydroxyecdysone. Shorter hatching times were observed in the egg strings exposed to hormone (10–7 to 10–5 M) compared to sea water and cholesterol (10–11 and 10–9 M) controls. Possible physiological roles and the evolutionary significance of ecdysteroids in nemerteans are discussed.  相似文献

Five molecular species of ecdysteroidogenic peptides were isolated from female heads of the mosquito, Aedes aegypti. Three groups of fractions, separated by conventional liquid chromatography, had gonadotropic activity in an in vivo bioassay using autogenous Aedes atropalpus. The active peptides from one of the three groups were purified to homogeneity with ion-exchange and reversed phase HPLC. Aedes atropalpus decapitated at eclosion do not deposit yolk, whereas injection of 12–15 pg of the purified peptides elicited yolk deposition. In an in vitro assay, the same peptides also stimulated ovaries of A. aegypti to secrete ecdysteroids, as measured in a RIA.The purified peptides have a molecular weight between 6500 and 13,000. Amino acid composition analysis of one species revealed 92 amino acid residues, and the number of basic residues substantiated the basic nature of the peptide observed during chromatography. Since the peptides were purified to homogeneity and are functional in both bioassays, we consider the peptides to be “ovarian ecdysteroidogenic hormones”.  相似文献

The levels of ecdysteroids in Sarcophaga bullata were determined by radioimmunoassay (RIA) from the time of larviposition (0 hr) to after the 2nd ecdysis and from late larval to pupal development. Two distinct peaks of ecdysteroid activity were recorded mid-way through the first and second stadia (14 and 34 hr) and two smaller peaks occurred a few hours prior to each ecdysis. A large release of ecdysteroids occurred from 8 hr before and up to 18 hr after formation of the white prepupa. This peak initiated the formation of the prepupa, the tanning of the puparium, larval/pupal apolysis and secretion of the pupal cuticle.Assays for the cuticle tanning hormone, bursicon, in pre-ecdysial larvae were not positive and a possible role for ecdysone in pre-ecdysial tanning of larval cuticular structures is proposed.  相似文献

Intraguild (IG) predator density can alter its effects on intraguild prey populations through several mechanisms, including density-dependent processes that affect IG predator traits such as size or growth that enhance or limit its predatory abilities. We examined whether intraspecific density-dependence altered IG predator traits, as well as the subsequent interspecific effects among its intraguild prey within a larval salamander guild. Four densities of ringed salamanders (Ambystoma annulatum), the IG predator, were combined with the presence/absence of spotted salamanders (A. maculatum), the IG prey, within experimental mesocosms. We modeled the effects of A. annulatum density on both conspecific and heterospecific responses that would be indicative of density-dependent competition and predation, respectively. We also modeled the reciprocal interspecific effects of A. maculatum on A. annulatum. We found that increasing intraspecific density negatively affected morphological traits but not survival of A. annulatum. No interspecific effects of A. maculatum on A. annulatum were observed. Alternatively, traits of A. maculatum showed nonlinear relationships with increasing A. annulatum density. Thinning effects of A. annulatum on A. maculatum were observed, as survival was positively and size negatively related for A. maculatum with IG predator density. The temporal overlap of the IG predator and prey also increased nonlinearly with IG predator density, intensifying the potential encounter rate of the two species. Overall, this study shows that density-dependent processes in IG predators can significantly affect traits of both themselves, as well as IG prey, which could ultimately change whether competition or predation occurs between the two groups.  相似文献

This study compiles the results from an examination of mycorrhizae on root samples from Tuber melanosporum truffières in New Zealand. Samples were taken over 5 years from 328 trees in 43 truffières established with nursery-inoculated trees. Mycorrhizae were analysed using a combination of morphological and molecular techniques, focusing on the identification of Tuber species. Results show that 49% of the trees, and nearly 90% of the truffières, retained T. melanosporum mycorrhizae up to 21 years after planting. Tuber mycorrhizae with spiky cystidia were found on 26.9% of the tested trees: Tuber brumale (5.5%), Tuber maculatum (10.7%), and unidentified Tuber species (10.7%), and were detected in 67% of the truffières tested. T. brumale was found in 28% and T. maculatum in 35% of the truffières. In 56% of the truffières, T. melanosporum was found to occur with spiky Tuber species. The existence of T. brumale and T. maculatum in the same truffière was recorded only once. Forty-four percent of trees examined had Scleroderma-like (SCL) mycorrhizae and 50% of trees hosted other ectomycorrhizal species (OE). For all categories of mycorrhizal species examined, the variation between truffières was greater than variation within each truffière. Overall results indicate that Corylus avellana tends to be more receptive to mycorrhizae of Tuber species than Quercus robur but is not necessarily more productive. In productive truffières, Q. robur appears to host SCL mycorrhizae more often than C. avellana. This is the first study of its scale to analyse the mycorrhizal species associated with T. melanosporum truffières in the Southern Hemisphere.  相似文献

A spectrum of eight pharmacologically important secondary compounds, all putatively belonging to the polyketide pathway (hypericin, pseudohypericin, emodin, hyperforin, hyperoside, rutin, quercetin, and quercitrin) were analyzed in several hypericin-producing species of Hypericum by LC–MS/MS. Different organs such as leaves, stems and roots of wild-grown plants of Hypericum hirsutum L., Hypericum maculatum Crantz s. l., Hypericum montanum L., Hypericum tetrapterum Fr. collected in Slovakia and of Hypericum perforatum L. collected in India were examined individually. Highest contents of hypericin, pseudohypericin, and emodin were found in H. montanum, suggesting that there are alternative species to H. perforatum with high pharmaceutical value. Amounts of hyperforin and quercetin were highest in H. perforatum, whereas highest contents of hyperoside and quercitrin were found in H. maculatum. A significant positive correlation between hypericin and pseudohypericin as well as between hypericin and emodin was observed by Kruskal’s multidimensional scaling (MDS), indicating a parallel enhancement of emodin as a common precursor in the biosynthetic pathways of hypericin and pseudohypericin. Furthermore, MDS combined with principal component analysis (PCA) revealed strong correlations in the occurrence of pseudohypericin and emodin, pseudohypericin and quercitrin, hypericin and quercitrin, emodin and quercitrin, hyperoside and quercitrin, rutin and quercetin, and, hyperforin and quercetin. On the other hand, rutin showed a negative correlation with emodin as well as with quercitrin. Furthermore, hierarchical agglomerative cluster analysis (HACA) clustered hypericin and pseudohypericin, grouping emodin at equal distance from both. Considerable infraspecific variability in secondary compound spectrum and load of different populations of H. maculatum from Slovakia underscores the need for detailed studies of genotypic variation and environmental factors in relation to polyketide biosynthesis and accumulation.  相似文献

We compared floral odour profiles among populations of two Arum species which show different degrees of specificity for their fly pollinators. Insects were collected from inflorescences in four populations of Arum italicum and two populations of Arum maculatum. In six Arum populations, we compared inflorescences odour profiles collected by Solid Phase Micro Extraction (SPME) and analysed by gas chromatography. We confirmed that from a pollination point of view, A. italicum is an opportunist species, as it is mainly pollinated by insects of the families Psychodidae, Chironomidae and Sciaridae, whereas A. maculatum is a specialist species, as it is 90% pollinated by Psychodidae. In all populations, Arum italicum was less attractive to pollinators than Arum maculatum. Floral odour profiles of A. italicum were not geographically structured among populations, suggesting a high gene flow or adaptation to a fluctuant guild of pollinators. On the contrary, odour profiles of A. maculatum varied between the two populations studied suggesting a lower gene flow or adaptation to different local pollinator preferences  相似文献

An LC–ESI-MS analysis was performed to determine the iridoid composition of Lamium album, Lamium amplexicaule, Lamium garganicum, Lamium maculatum, and Lamium purpureum. Nine iridoids known to occur in these species, lamalbid, sesamoside, lamiol, 5-deoxylamiol, shanzhiside methyl ester, caryoptoside, penstemoside, lamioside, and barlerin, could be detected. Confident identification of the individual compounds was achieved by a combination of HPLC retention times, both positive and negative ionization modes in MS and the presence of cluster and fragment ions. Iridoid glucosides new to four of the studied species were reported here for the first time. In most cases, the observed iridoid profiles were consistent and only small intraspecific variations were detected. Some chemosystematic implications from the observed iridoid composition were discussed.  相似文献

In a Japanese population of Locusta migratoria, adult females become reproductively inactive under crowding and long days (LD) and reproductively active under crowding and short days (SD). The identity and titre of ecdysteroids in the haemolymph and ovaries from adult females reared under SD and LD were investigated by RIA/HPLC. The effects of exogenous juvenile hormone (JH) III treatments on the termination of such reproductive arrest and ecdysteroid contents in LD females were also examined. In general, ecdysteroid titres in both haemolymph and ovaries were significantly higher in reproductively active SD females than in reproductively inactive LD females. A clear difference was also observed in oocyte growth between SD and LD individuals. JH III applications (three consecutive topical applications, 150 μg per insect per day from day 3) stimulated ovarian development in LD females and significantly increased the haemolymph and ovarian ecdysteroids to a level comparable to that of reproductively active SD adult females.  相似文献

Using radioimmunoassay (RIA) and high performance liquid chromatography (HPLC), the presence of a complex mixture of free and conjugated ecdysteroids is reported in the embryonated eggs of a mole crab,Emerita asiatica. From an initial low value of 6.5 ng/g egg wet weight in stage I, the total ecdysteroids increased in concentration to 15.2 ng/g egg wet weight in stage III. This was followed by a sharp fall in stage IV, but again increased to 15.0 ng/g egg wet weight in stage VI. After a further decline in stage VII, the total ecdysteroids registered the highest value of 36.2 ng/g egg wet weight in stage VIII. This value, however, declined to a low level in the prehatching stage (IX). The concentration of the free ecdysteroids always predominated over the conjugated ones. The HPLC analysis of free ecdysteroids demonstrated the presence of 20-hydroxyecdysone and ecdysone in the ratio of 2.5. Purified lipovitellin II also contained free and conjugated ecdysteroids. The functional significance of the embryonic ecdysteroids as well as their nature of synthesis and storage within the eggs is discussed in the light of the information available on insect embryogenesis.  相似文献

Ecdysteroidogenesis is essential for arthropod development and reproduction. Although the importance of ecdysteroids has been demonstrated, there is little information on the sites and enzymes for synthesis of ecdysteroids from Chelicerates. Ecdysteroid functions have been well studied in the soft tick Ornithodoros moubata, making this species an excellent candidate for elucidating ecdysteroidogenesis in Chelicerates. Results showed that O. moubata has at least two ecdysteroidogenic enzymes, Spook (OmSpo) and Shade (OmShd). RNAi showed both enzymes were required for ecdysteroidogenesis. Enzymatic assays demonstrated OmShd has the conserved functions of ecdysone 20-hydroxylase. OmSpo showed specific expression in the ovaries of final nymphal and adult stages, indicating O. moubata utilizes the ovary as an ecdysteroidogenic tissue instead of specific tissues as seen in other arthropods. On the other hand, OmShd expression was observed in various tissues including the midgut, indicating functional ecdysteroids can be produced in these tissues. In nymphal stages, expression of both OmSpo and OmShd peaked before molting corresponding with high ecdysteroid titers in the hemolymph. In fed adult females, OmSpo expression peaked at 8–10 days after engorgement, while OmShd expression peaked immediately after engorgement. Mated females showed more frequent surges of OmShd than virgin females. These results indicate that the regulation of synthesis of ecdysteroids differs in nymphs and adult females, and mating modifies adult female ecdysteroidogenesis. This is the first report to focus on synthesis of ecdysteroids in ticks and provides essential knowledge for understanding the evolution of ecdysteroidogenesis in arthropods.  相似文献

The chemical composition and the antimicrobial activity of the essential oil isolated from the aerial parts of Hypericum maculatum Crantz were determined. In total, 109 compounds were identified, with germacrene D (21.5%), nonane (6.5%), (E)‐β‐farnesene (5.3%), δ‐cadinene (4.5%), and ledol (4.4%) as the main constituents. The chemical compositions of this oil and of four previously studied H. maculatum oils were compared using multivariate statistical analyses, viz., agglomerative hierarchical cluster and principal component analyses. Based on the results, the interrelationship among the hitherto studied H. maculatum oil samples, including the oil characterized here, was discussed. The study of the antimicrobial potential of the oil against five bacterial and two fungal strains showed that the oil had mainly moderate antimicrobial effects.  相似文献
