Anaerobic bacteria and anoxic sediments from soda lakes produced electricity in microbial fuel cells (MFCs). No electricity was generated in the absence of bacterial metabolism. Arsenate respiring bacteria isolated from moderately hypersaline Mono Lake (Bacillus selenitireducens), and salt-saturated Searles Lake, CA (strain SLAS-1) oxidized lactate using arsenate as the electron acceptor. However, these cultures grew equally well without added arsenate using the MFC anode as their electron acceptor, and in the process oxidized lactate more efficiently. The decrease in electricity generation by consumption of added alternative electron acceptors (i.e. arsenate) which competed with the anode for available electrons proved to be a useful indicator of microbial activity and hence life in the fuel cells. Shaken sediment slurries from these two lakes also generated electricity, with or without added lactate. Hydrogen added to sediment slurries was consumed but did not stimulate electricity production. Finally, electricity was generated in statically incubated “intact” sediment cores from these lakes. More power was produced in sediment from Mono Lake than from Searles Lake, however microbial fuel cells could detect low levels of metabolism operating under moderate and extreme conditions of salt stress.  相似文献

Lead, Copper, Zinc and Manganese were measured in surficial sediments, chironomid larvae, tubificid worms and two species of bivalve molluscs (Mytilaster lineatus and Corbicula fluminalis) from the Anzali wetland, Iran.No distinct relationship was observed between heavy metal levels and percentage fine fraction in sediments. The pattern of Mn accumulation was parallel to trends of organic matter variation. There were highly significant differences between sampling sites in contents of heavy metals but no significant differences between seasons. Significant differences were found in bioaccumulation of Lead and Zinc in three size categories of chironomid larvae. Lead was higher in smaller M. lineatus, while the reverse was observed for Copper.  相似文献

Levels of p,p-DDT and its metabolites in surficial sediments of Tagus and Sado Estuary and lagoon Ria Formosa varied one or two orders of magnitude. In general, p,-DDE and p,-DDD were the dominant metabolites and p,-DDT was a minor component. While concentrations of p,-DDT, p,-DDD and p,-DDE were hardly related to loss on ignition, concentrations of total DDT (tDDT) in surficial sediments of each studied system were positively correlated to that parameter. Higher contaminated samples were excluded from these relationships: a few sediment samples from the Tagus outlet channel, most of the sediments from the upper Sado Estuary, and some particulate matter samples collected in sediment traps in the adjoining area to the Ria Formosa. Concentration of tDDT appears, thus, to be related to sedimentary organic matter of these estuarine systems except near the actual sources of pollution. On the basis of the relationship slopes, one may conclude that lower Tagus Estuary is more contaminated than Ria Formosa and lower Sado Estuary.  相似文献

The distribution of diatom frustules in the surficial sediments of lakes and ombrotrophic peat pools in Atlantic Canada are examined especially in relation to pH and bog lake succession. Sedimentary diatom associations for peat pools, strongly acidic lakes, and weakly acidic lakes are characterized. These associations and the derived diatom-pH calibration functions will facilitate improved paleoecological interpretation of natural ontogenetic pathways and assessments of recent anthropogenic impacts.  相似文献

A comparison of the present chironomid fauna of four shallow, humic lakes with the subfossils of surficial sediments is made. The chemistry and benthos of the lakes investigated varied greatly. Results indicate a reasonable correlation between biocenoses and thanatocenoses, but a marked under-representation ofProcladius is apparent. It is suggested that, at least in shallow, humic lakes,Procladius be excluded in attempts to reconstruct chironomid communities from subfossils.  相似文献

1. Thirty small upland lakes in Cumbria, Wales, Scotland and Northern Ireland were each visited once during June and July 2000. From each lake, samples of surface sediment epilithon, macrophytes and total dissolved nitrogen (TDN) were collected for nitrogen stable isotope analysis. As part of a wider programme, samples were also collected for chemical analysis and bioassays. 2. Considerable variation was found in δ15N values in all measured nitrogen compartments. Some regional variation was evident but was generally weak. Sediment and epilithon δ15N were positively correlated with δ15N of TDN, suggesting that baseline nitrogen isotope ratios influence those in some organic matter compartments in the lakes. 3. Sediment δ15N was higher when inorganic nitrogen concentration in the water was low, possibly reflecting reduced isotope fractionation under these conditions. However, this was not the case for epilithon or macrophytes. Sediment δ15N values were also negatively related to annual nitrogen deposition. 4. Sediment, epilithon and macrophyte δ15N values all showed significant relations to nutrient limitation in the lakes as determined by algal bioassays. We suggest that sediment δ15N might be developed as a simple integrating measure of the degree of nitrogen limitation in lakes.  相似文献

A radioisotope method was devised to study bacterial respiratory reduction of arsenate in sediments. The following two arsenic-rich soda lakes in California were chosen for comparison on the basis of their different salinities: Mono Lake (approximately 90 g/liter) and Searles Lake (approximately 340 g/liter). Profiles of arsenate reduction and sulfate reduction were constructed for both lakes. Reduction of [73As]arsenate occurred at all depth intervals in the cores from Mono Lake (rate constant [k] = 0.103 to 0.04 h(-1)) and Searles Lake (k = 0.012 to 0.002 h(-1)), and the highest activities occurred in the top sections of each core. In contrast, [35S]sulfate reduction was measurable in Mono Lake (k = 7.6 x10(4) to 3.2 x 10(-6) h(-1)) but not in Searles Lake. Sediment DNA was extracted, PCR amplified, and separated by denaturing gradient gel electrophoresis (DGGE) to obtain phylogenetic markers (i.e., 16S rRNA genes) and a partial functional gene for dissimilatory arsenate reduction (arrA). The amplified arrA gene product showed a similar trend in both lakes; the signal was strongest in surface sediments and decreased to undetectable levels deeper in the sediments. More arrA gene signal was observed in Mono Lake and was detectable at a greater depth, despite the higher arsenate reduction activity observed in Searles Lake. A partial sequence (about 900 bp) was obtained for a clone (SLAS-3) that matched the dominant DGGE band found in deeper parts of the Searles Lake sample (below 3 cm), and this clone was found to be closely related to SLAS-1, a novel extremophilic arsenate respirer previously cultivated from Searles Lake.  相似文献

RibR is a minor cryptic flavokinase (EC 2.7.1.26) of the Gram-positive bacterium Bacillus subtilis with an unknown cellular function. The flavokinase activity appears to be localized to the N-terminal domain of the protein. Using the yeast three-hybrid system, it was shown that RibR specifically interacts in vivo with the nontranslated wild-type leader of the mRNA of the riboflavin biosynthetic operon. This interaction is lost partially when a leader containing known cis-acting deregulatory mutations in the so-called RFN element is tested. The RFN element is a sequence within the rib-leader mRNA reported to serve as a receptor for an FMN-dependent 'riboswitch'. In RibR itself, interaction was localized to the carboxy-terminate part of the protein, a segment of unknown function that does not show similarity to other proteins in the public databases. Analysis of a ribR-defective strain revealed a mild deregulation with respect to flavin (riboflavin, FMN and FAD) biosynthesis. The results indicate that the RNA-binding protein RibR may be involved in the regulation of the rib genes.  相似文献

The cladoceran faunas from 13 northern German lakes (the Plön district) of different trophic levels were analysed using the bosminid and chydorid remains in the surficial sediment as an integrated sample of the total lake fauna. In the dimictic lakes, a shift occurred in the composition of planktonic Cladocera from Bosmina coregoni, including morphs with a mucro, to predominance of Chydorus sphaericus and Bosmina c. thersites as trophic state increased. In the benthic Cladocera (Chydoridae), there was a decrease in species diversity, in the percentages of phytophilous species and in the portion of species that are typical of clear water lakes, but there was an increase both in the number of mud dwellers and species that are typical of polluted lakes. No positive correlation was found between mean depth of the lakes and the planktonic/littoral ratio.  相似文献

Two emergence trap designs were tested in Mono Lake, California, to measure in situ hatching of Artemia monica cysts on the lake bottom. One design incorporated a removable sample bottle; the other had a catch tube which was pumped from the surface. Both traps rested on the bottom and had a narrow gap between the collecting funnel and bottom flange to allow the chemical conditions within the trap to be similar to those outside. This gap was open during April and May but, because some animals entered from outside the area enclosed by the trap, the gap was covered with 400 µm or 800 µm screen during June and July. The two trap types without screens sampled a station in oxic water 7 m deep similarly in April and May 1985. Mean daily hatching rates from April to May 1985 ranged from 720 to 25 340 shrimp m-2 day-1. In contrast, mean daily hatching rates during the same period at a station in anoxic water 21 m deep were from 3 to 138 shrimp m-2 day-1. June and July hatching rates in the shallow station were lower than in the spring, usually less than 1000 shrimp m-2 day-1.  相似文献

Aeroterrestrial filamentous green algae of the genus Klebsormidium (Klebsormidiales, Streptophyta) are typical components of biological soil crusts, which occur worldwide in arid and semiarid habitats including alpine regions. In the present study, Klebsormidium crenulatum (Kütz.) Lokhorst was isolated from an alpine soil crust above the timberline of the Austrian Alps. Growth responses, photosynthetic performance, and desiccation tolerance were measured under controlled laboratory conditions. K. crenulatum exhibited optimal growth and the highest photosynthetic efficiency under relatively low photon fluence densities (30 and 21.9 μmol photons · m?2 · s?1, respectively), indicating low‐light requirements. It grew in a narrow range of salinities between 1.2 and 15 practical salinity units (psu), pointing to a pronounced stenohaline response pattern. Increasing temperatures from 5°C to 40°C led to different effects on photosynthetic oxygen evolution and respiratory oxygen consumption in K. crenulatum. While at low temperatures (5°C–10°C) photosynthesis was relatively high, respiration was not detectable or was at a very low level. Conversely, at the highest temperature of 40°C, photosynthesis was inhibited, and respiration unaffected, indicating strong differences in temperature sensitivity between both physiological processes. K. crenulatum was capable of photosynthesizing efficiently for up to 2.5 h under desiccation, followed by a decrease to 15% of the initial value after 3 h. Complete recovery took place within 2 h after rehydration. All ecophysiological data explain the widespread abundance of K. crenulatum in soil crusts of the alpine regions of the European Alps.  相似文献

Regulation of river flow and the amount of winter rainfall are the major factors affecting the water temperature of the spawning grounds, for green sturgeon in the Klamath River. During the primary spawning period of green sturgeon, mid-April to June, the water temperature may vary from 8 to 21°C. To estimate the potential implications of this modified thermal regime, we examined the survival and development in three progeny groups of green sturgeon embryos from zygote to hatch, at constant incubation temperatures (11–26°C). Temperatures 23–26°C affected cleavage and gastrulation and all died before hatch. Temperatures 17.5–22°C were suboptimal as an increasing number of embryos developed abnormally and hatching success decreased at 20.5–22°C, although the tolerance to these temperatures varied between progenies. The lower temperature limit was not evident from this study, although hatching rate decreased at 11°C and hatched embryos were shorter, compared to 14°C. The mean total length of hatched embryos decreased with increasing temperature, although their wet and dry weight remained relatively constant. We concluded that temperatures 17–18°C may be the upper limit of the thermal optima for green sturgeon embryos, and that the river thermal regime during dry years may affect green sturgeon reproduction.  相似文献

Assuming that the inshore and offshore waters of Lake Victoria are impacted differently by human activities in its catchment, this study investigated the water quality dynamics of the lake. A total of 29 stations were sampled in 2005–2008 for dissolved oxygen (DO), pH, Secchi transparencies, temperature, turbidity, chlorophyll a, NO3, SRSi, TN and TP. There was a decreasing trend of the measured parameters towards offshore sites, except for Secchi transparency and NO3, which increased towards the offshore waters. DO concentrations (mean ± SD) varied between 6.97 ± 0.57 mg O2 l–1 and 5.80 ± 0.72 mg O2 l–1 in the inshore and offshore waters, respectively. Turbidity values were comparatively higher in the inshore (3.73 ± 2.21 NTU) than the offshore waters (2.19 ± 1.81 NTU). Chlorophyll a concentrations varied between 17.36 ± 6.13 µg l–1 and 8.09 ± 4.38 µg l–1 in the inshore and offshore waters, respectively. Increases of unsustainable human activities in the lake and its catchments, plus increased degradation of wetlands, are among the causes of the observed water quality changes. In order to be fruitful and sustainable, the management of Lake Victoria and its catchment needs to take an ecosystem approach, and to involve all key stakeholders.  相似文献

Water levels in many reservoirs typically fluctuate seasonally, but the effects of re-inundation of exposed sediments on nutrient dynamics in the water column are poorly known. This study concerns the seasonal differences in the potential of sediments from two Australian reservoirs, after having undergone different degrees of in situ desiccation, to release P under aerobic conditions. Differences were determined between biotic and abiotic P release, and results were also examined in relation to sediment chemistry. The two reservoirs, Carcoar Dam and Lake Rowlands, demonstrated different patterns of P release involving an interactive complex of P release mechanisms. Sediment chemistry at the reservoir margins was important because of the higher concentrations of N, P, Fe and Mn in Lake Rowlands. Physical and chemical processes influenced P uptake and release due to desiccation and oxidation of sediments and were of greater importance in Carcoar Dam. Abiotic P release from sterilised sediments was greater than from unsterilised sediments where both biotic and abiotic processes were apparent. Biotic P uptake and release were especially marked in Lake Rowlands where large macrophyte beds provided a rich source of organic matter. Little seasonal difference in P release was detected. The increased P release from dried sediments has ramifications for internal P loading into reservoirs and for the calculation of P budgets. For managers of reservoirs where large expanses of sediment are exposed during drying, it may be better to maintain high water levels, where possible, during the summer by modifying drawdown practices.  相似文献

1. We used first‐order kinetic parameters of biological oxygen demand (BOD), the constant of aerobic decomposition (k) and the asymptotic value of BOD (BODult), to characterise the lability of organic carbon pools in six lakes of different trophic state: L. Naroch, L. Miastro and L. Batorino (Belarus), L. Kinneret (Israel), L. Ladoga (Russia) and L. Mendota (U.S.A.). The relative contributions of labile and refractory organic carbon fractions to the pool of total organic carbon (TOC) in these lakes were quantified. We also determined the amounts of labile organic carbon within the dissolved and particulate TOC pools in the three Belarus lakes. 2. Mean annual chlorophyll concentrations (used as a proxy for lake trophic state) ranged from 2.3 to 50.6 μg L−1, labile organic carbon (OCL = 0.3BODult) from 0.75 to 2.95 mg C L−1 and k from 0.044 to 0.14 day−1. 3. Our data showed that there were greater concentrations of OCL but lower k values in more productive lakes. 4. In all cases, the DOC fraction dominated the TOC pool. OCL was a minor component of the TOC pool averaging about 20%, irrespective of lake trophic state. 5. In all the lakes, most (c. 85%) of the DOC pool was refractory, corresponding with published data based on measurements of bacterial production and DOC depletion. In contrast, a larger fraction (27–55%) of the particulate organic carbon (POC) pool was labile. The relative amount of POC in the TOC pool tended to increase with increasing lake productivity. 6. Long‐term BOD incubations can be valuable in quantifying the rates of breakdown of the combined particulate and dissolved organic carbon pools and in characterising the relative proportions of the labile and recalcitrant fractions of these pools. If verified from a larger number of lakes our results could have important general implications.  相似文献

Short-term effects of irradiance (0-1560 micromol photons m(-2) s(-1)), temperature (10-25 degrees C), and salinity (40-160) on oxygenic photosynthesis and oxygen consumption in a hypersaline mat (Salin-de-Giraud, France) were investigated with microsensors under controlled laboratory conditions. Dark O(2) consumption rates were mainly regulated by the mass transfer limitations imposed by the diffusive boundary layer. Areal rates of net photosynthesis increased with irradiance and saturated at irradiances >400 micromol photons m(-2) s(-1). At low irradiances, oxygen consumption increased more strongly with temperature than photosynthesis, whereas the opposite was observed at saturating irradiances. Net photosynthesis vs. irradiance curves were almost unaffected by decreasing salinity (100 to 40), whereas increasing salinities (100 to 160) led to a decrease of net photosynthesis at each irradiance. Dark O(2) consumption rates, maximal gross and net photosynthesis at light saturation were relatively constant over a broad salinity range (60-100) and decreased at salinities above the in situ salinity of 100. Within the range of natural variation, temperature was more important than salinity in regulating photosynthesis and oxygen consumption. At higher salinities the inhibitory impact of salinity on these processes and therefore the importance of salinity as a regulating environmental parameter increased, indicating that in more hypersaline systems, salinity has a stronger limiting effect on microbial activity.  相似文献

Electron transport system (ETS) activity, CO2 evolution, O2 consumption, N2-fixation (C2H2 reduction) and methanogenesis were appropriately measured in aerobic and anaerobically incubated sediment at 4, 10 and 20 ° C to better characterize these activities under different incubation conditions. ETS activity was always higher in the aerobically incubated sediment at all three incubation temperatures, whereas (C2H2 reduction was always greater in the anaerobic sediment. Carbon dioxide evolution was detected only in the aerobic sediment at 10 and 20 ° C but not at 4 ° C. Methane evolution in anaerobic sediment increased gradually with an increase in the incubation temperature.  相似文献

Abstract Simultaneous measurements of sulfate reduction and acetate oxidation using 35S and 14C tracers showed that acetate was the main energy substrate for the sulfate-reducing bacteria in Lake Eliza sediments. Sulfate reduction rates calculated from acid-volatile sulfide data only, correlated with acetate oxidation at around 0.5:1. However, the rates calculated from acid-volatile plus pyrite sulfur data correlated with acetate oxidation at a ratio of around 1:1. Molybdate completely inhibited sulfate reduction but acetate oxidation was not totally inhibited. From 10 to 15% of acetate oxidation was not attributable to the sulfate-reducing bacteria. There was rapid accumulation of acetate, within the first 12 h of incubation. Acetate, propionate and butyrate accumulated in the presence of molybdate.  相似文献
