以芥菜型油菜(Brcssica juncea)下胚轴来源的愈伤组织为材料,进行悬浮细胞振荡培养,得到游离的单细胞。这些细胞作浅层液体静置培养,可获得高频率的愈伤组织。在附加有水解乳蛋白200毫克/升、BA 2—3毫克/升及GA,0.1—1毫克/升的MS培养基上,这些愈伤组织分化了芽。分化的芽在无激素或附加有0.01—0.1毫克/升IAA或NAA的MS培养基上长出根,从而发育成完整植株。当对游离细胞获得的再生植株的茎切段愈伤组织,再经上述程序进行细胞培养时,发现愈伤组织诱导率及愈伤组织分化频率均远远高于原供体,从而建立了B.juncea易再生的体细胞无性系。  相似文献

通过诱导玉米芽尖产生胚性愈伤组织,建立起高频率植株再生的玉米芽尖培养实验体系。在MS+1.0mg·L-16-BA+0.2mg·L-12,4-D+500mg·L-1CH的培养基上诱导愈伤组织并继代培养1次后,将愈伤组织转移到Ms+0.5mg·L-16-BA+0.4mg·L-1IBA+500mg·L-1CH的分化培养基上,可形成大量的体细胞胚胎。组织学观察表明,体细胞胚胎主要发生在胚性愈伤组织表面与表层下部。不同基因型的芽尖形成愈伤组织和再生植株的能力不同。  相似文献

为探讨细枝木麻黄(Casuarina cunninghamianaMiq.)愈伤组织分化过程的细胞组织学,对离体培养条件下的愈伤组织进行扫描电子显微镜和石蜡切片观察,分析愈伤组织的细胞分裂、分化以及芽再生的发生过程。结果表明,新鲜外植体培养于愈伤组织诱导培养基上,伤口处的薄壁细胞开始脱分化,培养1周后形成明显的愈伤组织;继续培养2周后,胚性愈伤组织形成,且表层细胞启动分化形成芽原基;培养4周,可肉眼观察到胚性芽原基,数量增多并逐渐分化形成不定芽;培养至第6周,生成不定芽,并大量增殖和分化。因此,细枝木麻黄是通过愈伤组织分化形成胚状体的途径进行植株再生的,为建立细枝木麻黄组织培养高效再生体系提供了理论依据。  相似文献

本文报道了用罗布麻(Apocynum venetam)幼苗的子叶和下胚轴切段诱导出愈伤组织和再生植株。结果表明,愈伤组织的诱导在附加0.5mg/L6-BA和0.1-1mg/L NAA的MS培养基上为最好。在附加0.5mg/L NAA的MS培养基上培养愈伤组织能促进芽的分化,当NAA浓度增加到1mg/L时则能抑制芽的分化。随后在附加0.4mg/L IBA的MS培养基上诱导生根,获得完整再生植株。  相似文献

以新疆天山雪莲的叶片为外植体,分别用不同配方培养基诱导愈伤组织,后进行体胚诱导和分化培养形成再生雪莲植株.结果表明,诱导愈伤组织的最适培养基为MS 2,4-D 0.5 mg/L BA 1.5 mg/L,诱导率可达到100%;愈伤组织转移至MS 2,4-D 0.5 mg/L BA 1.5 mg/L培养基进行继代培养,增殖后的愈伤组织转移到MS 2,4-D 0.2 mg/L的液体培养基后成功诱导出雪莲体胚,出胚率达40%;将体胚接至MS ABA 0.5 mg/L培养基后,结果分化生长出大量的再生雪莲幼苗.  相似文献

以冬凌草叶片为外植体,研究不同浓度激素组合对冬凌草愈伤组织诱导及植株再生的影响,并对不同外植体(茎、叶)诱导愈伤、芽的分化能力及再生植株内主要次生代谢产物的含量进行了比较研究。结果表明:在MS 2.0 mg/L 6-BA 1.0 mg/L NAA培养基上诱导愈伤组织效果较好;在MS 2.0 mg/L 6-BA的培养基上诱导芽的效果较好;叶片和茎段在愈伤诱导培养基上均能产生大量的愈伤组织,但其再分化能力以茎段最好;再生苗生根培养基以0.3 mg/L IBA最好;以叶为外植体诱导的再生植株中冬凌草甲素、迷迭香酸的含量均高于以茎为外植体诱导的再生植株。  相似文献

利用不同激素组合对丝瓜愈伤组织诱导、芽的分化及试管苗快速繁殖进行了研究.结果发现不同外植体均易诱导愈伤组织,但愈伤组织分化芽较难,而外植体在一些激素组合中可直接形成不定芽;利用无菌苗的顶芽及腋芽,培养于附加6 BA1mg L+NAA0.5mg L或NAA1mg L的MS培养基上,获得了芽的快速增殖,形成芽簇,每丛芽数达6~8苗;试管苗于1 2MS+NAA0.5mg L生根培养基中6d生根率达到100%,获得完整再生植株.结果表明采用不同激素组合对丝瓜体细胞形态发生与发育的调控有决定性作用.  相似文献

以红掌盆栽品种‘Avo-Gloria’为试材,以MS+0.2mg·L^-12,4-D为基本培养基,分别在添加1~10mg·L^-16-BA的10种脱分化培养基上,诱导其叶柄外植体产生愈伤组织;再以MS+2mg·L^-16-BA+0.2mg·L^-1 NAA为分化培养基诱导分化不定芽;以MS+0.2mg·L^-1 NAA为生根培养基,从不定芽获得再生植株。结果显示:(1)在MS+0.2mg·L^-12,4-D+8~10mg·L^-16-BA的3种脱分化培养基上可产生9%~10%的绿色、质地较硬的愈伤组织;(2)愈伤组织在MS+2mg·L^-16-BA+0.2mg·L^-1 NAA的分化培养基上,经6~8次继代培养,可获得3%~7%的不定芽,并可生根长成再生植株;(3)再生植株定植3个月后,有3%~7%植株出现红叶变异,此红叶可终生表现为红色。  相似文献

对稀有植物裸果木进行组织培养研究,在MS培养基上裸果木的下胚轴脱分化形成愈伤组织,并进一步分化形成再生植株。激素种类及其浓度是器官脱分化与植株再生的决定因素。诱导下胚轴形成愈伤组织的最适培养基为MS 1mg/L 6-BA 0.5mg/L NAA;芽分化诱导的最适培养基为MS 1 mg/L6-BA;生根的最适培养基为不含任何激素的1/2MS培养基。  相似文献

本文报道的小麦花粉无性系,已继代培养了两年零一个月共27代,目前仍具有分化能力。继代分化固体培养基以:MS附加NAA0.6毫升/升+KT0.2毫克/升为最好。较好的恢复再生培养基为MS液体培养基附加2.4-D 0.6毫克/升+NAA0.2毫克/升+6-BA 2毫克/升+CH500毫克/升予处理三天后转入附加2.4—D0.6毫克/升+KT 2毫克/升+CH 500毫克/升的MS固体培养基,培养十八天后,丧失分化能力已达八个月之久的愈伤组织又恢复了再生能力,分化出植株。通过愈伤组织的细胞学观察,小麦花粉愈伤组织无性系,分化成小植株的途径有二:一是由特化的胚性细胞经多次分裂后发育成胚状体长成小植株;二是由特化的表层细胞经多次分裂后发育成芽苗。胚性细胞发生于愈伤组织的表皮层和表皮附近的细胞层,有的也可在内层已衰老的细胞中产生。胚状体与芽的区别特征是:胚状体具有两极性,两极间有维管组织相连接;芽具有单极性,芽端的维管组织与愈伤组织内维管组织相连接。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
