本文旨在通过诱变和耐受性筛选的手段提高放线茵£一PL的产量。以稠李链霉菌Strepto—myces padanus LS-L5为出发菌,经紫外诱变,在以琥珀酸为唯一碳源的培养基上筛选快速生长的菌株,测定突变株的ε-PL发酵水平和遗传稳定性。经初筛和复筛,获得突变株H3,其摇瓶发酵ε-PL产量为0.68dL,较出发菌提高了41%,传代4次产量基本稳定。H3在5L发酵罐中分批发酵,ε-PL产量达到2.0g/L,较出发菌提高了一倍。在固定二氧化碳的能力方面,突变株H3的PEP羧化酶酶活较出发菌株LS-L5的PEP羧化酶酶活提高了1.67倍。  相似文献

氮离子注入选育阿维拉霉素高产菌株的研究   总被引:3,自引:0,他引:3  
以提高产绿链霉菌(Streptomyces viridochromogenes)SV-1产阿维拉霉素(Avilamycin)产量为目的,采用低能氮离子注入技术,辅之以链霉素抗性筛选法进行诱变选育研究。结果表明,“马鞍”区域即注入剂量范围在3×10~(15)~5×10~(15)ions/cm~2诱变效果最佳,菌株的抗药性突变与产量突变密切相关,链霉素抗性筛选法具有可行性。在摇瓶条件下,最终获得稳定性良好,阿维拉霉素产量达到83.5mg/L,较出发菌株提高195%的突变株SVT-45。实验表明,离子注入技术是一种有潜力的微生物诱变育种新方法。  相似文献

本试验评估了赤霉酸生产菌株“4303”的生产性能,并以此菌株为出发菌株,进行了紫外线诱变育种,选出了一株赤霉酸高产菌株UF456。通过试验发现“4303”平均产量为1036±129mg/L,变异系数为0.12,紫外线诱变对菌株产赤霉酸分布呈均匀状态。将“4303”与UF456进行赤霉酸产势比较,结果表明:UF456菌株平均产量较“4303”高28.8%,产势高20%。  相似文献

新型抗生素AGPM产生菌藤黄灰链霉菌的诱变育种   总被引:4,自引:1,他引:3  
从土壤中分离得到一株藤黄灰链霉菌的新菌株099,该菌株能产生一种具有显抗肿瘤活性的新型抗生素AGPM。产生菌经紫外线单因子诱变、紫外线加氯化锂(LiCl)复合诱变等两种诱变方式诱变处理后,其AGPM产量达到了18.7μg/mL,较出发菌株提高了1.2倍。研究发现最忧的紫外辐射时间和氯化锂浓度分别是30-60s和0.05-0.09mol/L,并且由于氯化锂的协同效应,菌株在经氯化锂处理后,对紫外线的敏感性明显增强了。30L发酵罐发酵试验表明,利用高产突变株发酵,AGPM产量能达到18.5μg/mL;而利用出发菌株发酵,AGM产量仅为8.5μg/mL。  相似文献

阿维拉霉素产生菌SV微波诱变效应的研究发现:菌株SV对微波敏感(微波处理60s,SV菌株存活率低于10%),菌落形态变化大。微波诱变处理最佳作用方式为培养皿不加盖、快速冰上冷却,最佳处理时间为50s(其正突变率25.3%)。通过微波诱变处理、阿维拉霉素推理筛选,最终获得一株稳定性良好,阿维拉霉素产量达到21.5mg/L,较出发菌株提高119.4%的突变株SV-15。  相似文献

以根癌土壤杆菌(Agrobacterium tumefaciens)WSHAT12为出发菌株,通过硫酸二乙酯诱变,获得遗传稳定性好的抗L-乙硫氨酸(Eth)突变株WSH-E01,通过进一步的诱变处理,获得L-乙硫氨酸和维生素K3(VK3)双抗性突变株WSH-V01,以双抗性突变株WSH-V01为出发菌株,再进行诱变处理,获得一株X-gal利用能力提高的突变株WSH-X01,与出发菌株WSHAT12相比,突变株WSH-X01的辅酶Q10产量提高幅度达50.6%,同时,对突变株WSH-E01的发酵条件进行优化。出发菌株WSHAT12、突变株WSH-E01、WSH-V01和WSH-X01在优化后的发酵条件下辅酶Q10产量分别达到23.1mg/L、26.8mg/L、29.5mg/L和34.8mg/L。  相似文献

采用超高静压对一株产琥珀酸放线杆菌A3进行诱变育种,进一步提高其生产性能.考察了压力、变压速度、生长期对菌株致死率的影响.在压力为200 MPa,50 MPa/min变压速度的诱变条件下对稳定期菌株进行诱变,筛选到一株突变菌株产琥珀酸放线杆菌B19,琥珀酸产量达到32.2 g/L,比出发菌株A3提高了17.9%,代谢副产物乙酸的产量降低了11.5%,经过6次传代表明突变菌株具有稳定的遗传特性.  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
