The possibility of shortening the thermal sterilization time for cultivating media was demonstrated in chlortetracycline fermentation with an industrial strain ofStreptomyces aureofaciens. The medium was artificially contaminated with a mixture of eight strains of G + and G-bacteria isolated from contaminated industrial fermentors, and the following chemical agents, either alone or in combination, were added: formaldehyde, phenol, dimethylformamide,p-aminosalicylic acid and nitrofurazone. Dimethylformamide was inhibitory even at 0.08%. formaldehyde concentrations higher than 0.05%, Nitrofurazone stimulated chlortetracycline production, The best combination was 0.01% formaldehyde added before, and 2.10-3% nitrofurazone added after short sterilization at 120 °C.  相似文献

Extracts of Pseudomonas C grown on methanol as sole carbon and energy source contain a methanol dehydrogenase activity which can be coupled to phenazine methosulfate. This enzyme catalyzes two reactions namely the conversion of methanol to formaldehyde (phenazine methosulfate coupled) and the oxidation of formaldehyde to formate (2,6-dichloroindophenol-coupled). Activities of glutathione-dependent formaldehyde dehydrogenase (NAD+) and formate dehydrogenase (NAD+) were also detected in the extracts.The addition of d-ribulose 5-phosphate to the reaction mixtures caused a marked increase in the formaldehyde-dependent reduction of NAD+ or NADP+. In addition, the oxidation of [14C]formaldehyde to CO2, by extracts of Pseudomonas C, increased when d-ribulose 5-phosphate was present in the assay mixtures.The amount of radioactivity found in CO2, was 6.8-times higher when extracts of methanol-grown Pseudomona C were incubated for a short period of time with [1-14C]glucose 6-phosphate than with [U-14C]glucose 6-phosphate.These data, and the presence of high specific activities of hexulose phosphate synthase, phosphoglucoisomerase, glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase indicate that in methanol-grown Pseudomonas C, formaldehyde carbon is oxidized to CO2 both via a cyclic pathway which includes the enzymes mentioned and via formate as an oxidation intermediate, with the former predominant.  相似文献

Localization of antigens homologous to excretory-secretory proteins in developing embryos ofSetaria digitata has been carried out by indirect fluorescent antibody test, [14C] labelling studies and Western blotting. Indirect fluorescent antibody test showed binding of excretory-secretory antibodies at perivitelline space. The fluorescent antibody binding was almost absent at small morulae stage and increasing in intensity in the successive developmental stages with maximum at coiled microfilaria stage. Hatched microfilaria did not show the presence of antigens by immunofluorescence. Immuno-complex of excretory-secretory antiserum against “amniotic fluid” collected from developing embryos ofSetaria digitata labelled with [14C] amino acids showed highest radioactivity at coiled and tadpole stages and differed significantly from small morulae, big morulae and hatched microfilaria. Immunoblot analysis of amniotic fluid showed two proteins, 16.5 and 11 kDa, to be highly antigenic. The antigenic protein (11 kDa) content as seen by immuno blotting increased during embryogenesis and decreased at the stage of hatching.  相似文献

The metabolism ofl-proline toN-acetyl-d-glucosamine (GlcNAc) during germ tube formation ofCandida albicans (C. albicans) ATCC 1002 was studied. In uptake experiments, 6.9 nmol ofl-[14C]proline were taken up by 1×106 cells during 3 h of incubation at 37°C. The percentage of germ tube formation was 94 under the same condition. The presence of GlcNAc reduced the uptake ofl-proline to 3.0 nmol. The percentage of germ tube formation was 95 in the presence and absence of GlcNAc. The [3H]GlcNAc uptake was 3.0 nmol and was constant whetherl-proline was present or not. After the preparation of a chitin fraction from germ tubes that were labeled withl-[14C]proline, the radioactivity froml-proline was detected in the glucosamine (GlcN) fraction by thin-layer chromatography (TLC). The metabolism ofl-proline to GlcNAc in chitin during germ tube formation was confirmed in this experiment.  相似文献

Summary A recombinant cosmid carrying the sucrase gene (sacA) was obtained from a colony bank ofE. coli harboring recombinant cosmids representative of theB. subtilis genome. It was shown that thesacA gene is located in a 2 kbEcoRI fragment and that the cloned sequence is homologous to the corresponding chromosomal DNA fragment. A fragment of 2 kb containing the gene was subcloned in both orientations in the bifunctional vector pHV33 and expression was further looked for inB. subtilis andE. coli. Complementation of asacA mutation was observed in Rec+ and Rec- strains ofB. subtilis. Expression of sucrase was also demonstrated inE.coli, which is normally devoid of this activity, by SDS-polyacrylamide gel electrophoresis, specific immunoprecipitation and assay of the enzyme in crude extracts. The specific activity of the enzyme depended on the orientation of the inserted fragment. The saccharolytic activity was found to be cryptic inE. coli since the presence of the recombinant plasmids did not allow the transport of [U14C] sucrose and the growth of the cells.It was shown also that the recombinant cosmid contained part of the neighboring locus (sacP) which corresponds to a component of the PEP-dependent phosphotransferase system of sucrose transport ofB. subtilis.  相似文献

Summary Two loci,ma-l + andry +, necessary for xanthine dehydrogenase activity inDrosophila melanogaster have been studied for dosage effects utilizing deficiencies and duplications induced for this purpose. Comparisons of one, two and three doses ofma-l + in the female or one and two doses in the male indicate that there is no increase in specific enzyme activity with dose. On the other hand, comparisons of one, two and three doses ofry + in the male and female reveal an increase in enzyme activity that is roughly proportional to dose. Since dosage ofry + is limiting, whereas that ofma-l + is not, the final concentration of xanthine dehydrogenase is shown to depend on the number of doses ofry +.The implications of these findings with respect to the hypothesis of dosage compensation and to the mechanism of control of enzyme and protein concentration are discussed.Operated by Union Carbide Corporation for the U.S. Atomic Energy Commission.  相似文献

Summary Organic components leaked fromSorghum bicolor seedlings (‘root exudates’) were examined by recovering14C labelled compounds from root solutions of seedlings inoculated withAzospirillum brasilense, Azotobacter vinelandii orKlebsiella pneumoniae nif-. Up to 3.5% of the total14C recovered from shoots, roots, and nutrient solutions was found in the root solutions. Inoculation with Azospirillum and Azotobacter increased the amounts of14C and decreased the amounts of carbohydrates in the root solutions. When sucrose was added as a carbon source for the bacteria, the increase of14C in the solutions did not occur. Quantities of14C found in the root solutions were proportional to amounts of mineral nitrogen supplied to the plants. Bacterial growth also was proportional to nitrogen levels. When sorghum plants were grown in soil and labelled with14CO2, about 15% of the total14C recovered within 48 hours exposure was found in soil leachates.  相似文献

A galactofuranosyl-containing glycopeptide has been isolated from mycelium ofAscobolus furfuraceus by extraction with water. The glycoconjugate was purified by DEAE-cellulose chromatography followed by gel filtration. A molecular weight of about 20 000 was determined by the latter method using standard dextrans. Neutral sugars accounted for 94.5% of the glycopeptide and were characterized as mannose, galactose, and glucose. Glucosamine was estimated colorimetrically (1.8%). The molar ratio of Man:Gal:Glc:GlcNH2 was 68:32:16:2. A trace amount of total phosphorus (0.2%) was found. The predominant amino acids were threonine and serine. The peptide moiety was labeled with [14C]formaldehyde and the elution of radioactivity was coincident with sugar on gel filtration in the presence of sodium dodecyl sulfate. The peak of radioactivity was retarded on release of galactose by mild acid hydrolysis. These results confirm the sugar-peptide linkage.  相似文献

Net synthesis of [14C]ent-kaurene from [14C]2-mevalonic acid was assayed in cell-free enzyme extracts prepared from Alaska pea (Pisum sativum L.) seedlings throughout 44 h of a regimen consisting of a 16-h day and an 8-h night. Activities generally followed an upward trend during the dark period and a downward trend during the photoperiod. Activity was also assayed in enzyme extracts prepared at intervals during a 12-h photoperiod and a following, continuous 36-h dark period after entrainment of plants to a regimen of 12-h days and 12-h nights.Ent-kaurene synthesis activity again followed an upward trend in enzyme extracts prepared during what would have been the entrainment dark period, and a downward trend during the entrainment photoperiod. The apparent endogenous rhythm ofent-kaurene biosynthesis may have implications for the regulation of gibberellin biosynthesis.  相似文献

The heterobasidiomycetous yeastRhodotorula glutinis was able to grow in medium containing a high concentration of LiCl. This character ofR. glutinis was presumed to be attributable to its ability to incorporate [14C]-adenine and [14C]-leucine into nucleic acids and proteins, respectively, in the presence of LiCl. Intracellular levels of Li+ and Cl ions, production and accumulation of glycerol as an osmoregulator, and respiration in the LiCl-stressed condition were almost the same in the tolerant yeastR. glutinis and the sensitive yeastRhodosporidium sphaerocarpum.  相似文献

Adult longevity, developmental time and juvenile mortality ofEncarsia formosa Gahan (Hymenoptera:Aphelinidae) parasitizing the Poinsettia-strain ofBemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) on Poinsettia (Euphorbia pulcherrima Willd.) were investigated in laboratory experiments at three temperatures: 16 °C, 22 °C and 28 °C. Furthermore, the parasitoid's preference for different larval stages of the whitefly was determined at 24.5 °C. The lifespan ofE. formosa decreased with temperature from one month at 16 °C to nine days at 28 °C. A lower temperature threshold of 11 °C for adult development was found. The development of juvenile parasitoids inB. tabaci lasted more than two months at the lowest temperature, but was only 14 days when temperature was 28 °C. The lower temperature threshold for immature development was 13.3 °C, yielding an average of 207 day-degrees for the completion of development into adults. Juvenile mortality was high, varying from about 50% at 16 °C to about 30% at 22 °C and 28 °C.E. formosa preferred to oviposit in the 4th instar and prepupal stages ofB. tabaci followed by the 2nd and 3rd instars. The preference for the pupal stage was low. The parasitoid used all instars of the whitefly for hostfeeding, with no apparent differences between the stages. The average duration of the oviposition posture was four minutes. Demographic parameters were calculated from life tables constructed from the data. The intrinsic rate of increase (r m) and the net reproductive rate (R 0) increased with temperature from 0.0279 day−1 at 16 °C to 0.2388 day−1 at 28 °C and from about 12 at 16 °C to about 66 at 28 °C, respectively.  相似文献

Summary Uniformly14C labelled glucose, cellulose and wheat straw and specifically14C labelled lignin component in corn stalks were aerobically incubated for 12 weeks in a chernozem soil alongwith15N labelled ammonium sulphate. Glucose was most readily decomposed, followed in order by cellulose, wheat straw and corn stalk lignins labelled at methoxyl-, side chain 2-and ring-C. More than 50% of14C applied as glucose, cellulose and wheat straw evolved as CO2 during the first week. Lignin however, decomposed relatively slowly. A higher proportion of14C was transformed into microbial biomass whereas lignins contributed a little to this fraction.After 12 weeks of incubation nearly 60% of the lignin14C was found in humic compounds of which more than 70% was resistant to hydrolysis with 6N HCl. Maximum incorporation of15N in humic compounds was observed in cellulose amended soil. However, in this case more than 80% of the15N was in hydrolysable forms.Immobilization-remineralization of applied15N was most rapid in glucose treated soil and a complete immobilization followed by remineralization was observed after 3 days. The process was much slow in soil treated with cellulose, wheat straw or corn stalks. More than 70% of the newly immobilized N was in hydrolysable forms mainly reepresenting the microbial component.Serial hydrolysis of soil at different incubation intervals showed a greater proportion of 6N HCl hydrolysable14C and15N in fractions representing microbial material.14C from lignin carbons was relatively more uniformly distributed in different fractions as compared to glucose, cellulose and wheat straw where a major portion of14C was in easily hydrolysable fractions.  相似文献

Several denitrifying Pseudomonas spp., isolated with various aromatic compounds, were tested for the ability to degrade toluene in the absence of molecular oxygen. Four out of seven strains were able to degrade toluene in the presence of N2O. More than 50% of the 14C from ring-labelled toluene was released as CO2, and up to 37% was assimilated into cell material. Furthermore it was demonstrated for two strains that they were able to grow on toluene as the sole carbon and energy source in the presence of N2O. Suspensions of cells pre-grown on toluene degraded toluene, benzaldehyde or benzoate without a lag phase and without accumulation of intermediates. p-Cresol, p-hydroxybenzylalcohol, p-hydroxybenzaldehyde or p-hydroxybenzoate was degraded much slower or only after distinct lag times. In the presence of fluoroacetate [14C]toluene was transformed to [14C]benzoate, which suggests that anaerobic toluene degradation proceeds through oxidation of the methyl side chain to benzoate.  相似文献

Ribosomal proteins labelled with [14C]PCMB, a reversibly reacting sulfhydryl reagent, lose most of their radioactivity when separated by the conventional two-dimensional polyacrylamide gel electrophoresis. Therefore, the two-dimensional method was modified to insure stability of the PCMB-protein complex during the procedure. Proteins ofEscherichia coli ribosomes were separated by the modified method, and those which reacted with [14C]PCMB were identified by counting of stained spots and by autoradiography of dried gels.  相似文献

Isolated hepatocytes from starved rats were loaded with N-[14C]acetylglutamate by preincubating them with [14C]bicarbonate, oleate, NH3, ornithine and lactate. Turnover of N-acetylglutamate in these cells was subsequently measured in an unlabelled medium under conditions of minimal flux (oleate alone present) and maximal flux (oleate, NH3, ornithine and lactate present) through the urea cycle. 1. Direct measurement of the distribution of N-[14C]acetylglutamate across the mitochondrial membrane in the hepatocytes showed that, under the conditions studied, the rate of degradation of total intracellular N-[14C]acetylglutamate was about equal to the rate of efflux of N-acetylglutamate from the mitochondria. 2. In the presence of oleate alone, intramitochondrial N-acetylglutamate decreased because mitochondrial N-acetylglutamate efflux predominated over the synthesis of N-acetylglutamate in the mitochondria. 3. In the presence of oleate, NH3, ornithine and lactate both the rate of synthesis of N-acetylglutamate and the rate of its transport out of the mitochondria were increased when compared with the condition with oleate alone. However, the intramitochondrial concentration of N-acetylglutamate increased because initially the rate of its synthesis exceeded that of its efflux from the mitochondria. Finally, a steady state was reached in which both rates were equal. 4. The data indicate that in hepatocytes from starved rats N-acetylglutamate transport out of the mitochondria takes place at a rate proportional to its intramitochondrial concentration. It is concluded that transport of N-acetylglutamate either occurs by diffusion or is mediated by a transport system with a high Km for intramitochondrial N-acetylglutamate.  相似文献

Abstract: In the presence of substance P (SP; 10 μM), serotonin (5-HT; 1 μM) triggered a cation permeability in cells of the hybridoma (mouse neuroblastoma X rat glioma) clone NG 108-15 that could be assessed by measuring the cell capacity to accumulate [14C]guanidinium for 10-15 min at 37°C. In addition to 5-HT (EC50, 0.33 μM), the potent 5-HT3 receptor agonists 2-methyl-serotonin, phenylbiguanide, and m-chlorophenylbiguanide, and quipazine, markedly increased [14C]guanidinium uptake in NG 108-15 cells exposed to 10 μM SP. In contrast, 5-HT3 receptor antagonists prevented the effect of 5-HT. The correlation (r= 0.97) between the potencies of 16 different ligands to mimic or prevent the effects of 5-HT on [14C]guanidinium uptake, on the one hand, and to displace [3H]zacopride specifically bound to 5-HT3 receptors on NG 108-15 cells, on the other hand, clearly demonstrated that [14C]guanidinium uptake was directly controlled by 5-HT3 receptors. Various compounds such as inorganic cations (La3+, Mn2+, Ba2+, Ni2+, and Zn2+), D-tubocurarine, and memantine inhibited [14C]guanidinium uptake in NG 108-15 cells exposed to 5-HT and SP, as expected from their noncompetitive antagonistic properties at 5-HT3 receptors. However, ethanol (100 mM), which has been reported to potentiate the electrophysiological response to 5-HT3 receptor stimulation, prevented the effects of 5-HT plus SP on [14C]guanidinium uptake. The cooperative effect of SP on this 5-HT3-evoked response resulted neither from an interaction of the peptide with the 5-HT3 receptor binding site nor from a possible direct activation of G proteins in NG 108-15 cells. Among SP derivatives, [D-Pro9]SP, a compound inactive at the various neurokinin receptor classes, was the most potent to mimic the stimulatory effect of SP on [14C]guanidinium uptake in NG 108-15 cells exposed to 5-HT. Although the cellular mechanisms involved deserve further investigations, the 5-HT-evoked [14C]guanidinium uptake appears to be a rapid and reliable response for assessing the functional state of 5-HT3 receptors in NG 108-15 cells.  相似文献

The distribution of12C and14C and dynamics of14C in barley plots were studied after pulse labelling with14CO2. Barley was grown in microplots on a Black soil at Ellerslie and a Gray Luvisol at Breton, Alberta and was sampled on four dates between July 31 and October 20. The quantity of12C in shoots, microbial biomass, and soil was greater at Ellerslie than Breton. Root12C did not differ between sites. There were no significant differences over time in the quantity of12C in any of the pools. On the first sampling date 18.59% of the14C was recovered in the soil-plant system at Ellerslie and 60.82% was recovered at Breton. At Ellerslie 20.35 MBq were recovered in shoots, 0.50 in roots, 0.08 in microbial C and 0.77 in soil while at Breton 27.92 MBq were recovered in shoots, 2.70 in roots, 0.22 in microbial C and 39.3 in soil. The difference between sites in soil14C was due to higher water filled porosity at Breton than at Ellerslie at the time of labelling. Soluble C (gg?1 root C), used as a measure of root exuded C, increased above 70% water filled porosity. There were no significant differences over time in the quantity of14C in any of the pools or in the specific activity of14CO2 released from soil during 10-day laboratory incubation. This indicated that the belowground system was in steady state with a continuous input of14C from labelled root matrial. Differences in specific activity of the various belowground pools revealed that an average of 17% of the microbial C was active at Ellerslie while 43% was active at Breton. Active microbial C (gm?2) was the same at both sites because total microbial C was lower at Breton than at Ellerslie. At Breton some of the14C released under conditions of high water filled porosity at the time of labelling appeared to be stabilized against microbial turnover.  相似文献

The biosynthesis and metabolism of purine alkaloids in leaves ofCamellia ptilophylla (cocoa tea), a new tea resource in China, have been investigated. The major purine alkaloid was theobromine, with theophylline also being present as a minor component. Caffeine was not accumulated in detectable quantities. Theobromine was synthesized from [8-14C] adenine and the rate of its biosynthesis in the segments from young and mature leaves from flush shoots was approximately 10 times higher than that from aged leaves from 1-year old shoots. Neither cellfree extracts nor segments fromC. ptilophylla leaves could convert theobromine to caffeine. A large quantity of [2-14C] xanthine taken up by the leaf segments was degraded to14CO2 via the conventional purine catabolic pathway that includes allantoin as an intermediate. However, small amounts of [2-14C] xanthine were also converted to theobromine. Considerable amounts of [8-14C] caffeine exogenously supplied to the leaf segments ofC. ptilophylla was changed to theobromine. These results indicate that leaves ofC. ptilophylla exhibit unusual purine alkaloid metabolism as i) they have the capacity to synthesize theobromine from adenine nucleotides, but they lack adequate methyltransferase activity to convert of theobromine to caffeine in detectable quantities, ii) the leaves have a capacity to convert xanthine to theobromine, probably via 3-methylxanthine.  相似文献

Summary The assimilation of14CO2 and [2-14C] acetate, [3-14C] pyruvate, [5-14C] -ketoglutarate, [2,3-14C] succinate, [U-14C] glutamate and [U-14C] aspartate was followed in cell suspensions ofNitrosomonas europaea andNitrobacter agilis respectively. There was appreciable incorporation of these substrates even without adding the inorganic nitrogen compounds that are oxidized by these bacteria yielding ATP. In the soluble amino acid fraction most of14C label was recovered in glutamate while in the protein amino acids a more uniform distribution was found. Acetate was rapidly incorporated to a high level in both nitrifying bacteria while inNitrobacter there was a relatively lower uptake of the other substrates especially succinate. High levels of the NAD malate dehydrogenase and NADP isocitrate dehydrogenase were measured but no significant amounts of the other tricarboxylic acid cycle enzymes or NADH oxidase were found. Glutamate decarboxylase was detected in both organisms and the transferase assay for glutamine synthetase indicated a 30-fold higher activity for this enzyme inNitrobacter. The amino acid composition of the water soluble fraction was determined in both bacteria.  相似文献

The impact of inoculation with Paxillus involutus on the utilization of organic carbon compounds by birch roots was studied by feeding [14C]Glu or [14C]malate to the partners of the symbiosis, separately or in association, and by monitoring the subsequent distribution of 14C. Inoculation increased [14C]Glu and [14C]malate absorption capacities by up to eight and 17 times, respectively. Six- and 15-d-old mycorrhizal roots showed about four-fold higher [14C]Glu and [14C]malate absorption capacities compared with 60-d-old mycorrhizal roots, suggesting that the early stages of mycorrhiza formation induced higher requirements for C skeletons. Moreover, the results demonstrated that inoculation strongly modified the fate of [14C]Glu and [14C]malate. It was demonstrated that exogenously supplied Glu and malate might serve as C skeletons for amino acid synthesis in mycorrhizal birch roots and in the free-living fungus. Gln was the major 14C-sink in mycorrhizal roots and in the free-living P. involutus. In contrast, citrulline and insoluble compounds were the major 14C sinks in non-mycorrhizal roots, whatever the 14C source. It was concluded that mycorrhiza formation leads to a profound alteration of the metabolic fate of exogenously supplied C compounds. The ecological significance of amino acid and organic acid utilization by mycorrhizal plants is further discussed.  相似文献
