The two-stage control system, ensuring the high quality of serological investigations in the network of screening laboratories in Moscow was developed and introduced into practice. At the first stage the entry control of the quality of the test system for the detection of HbsAg, coming to the screening laboratories, is made in the reference laboratory with the use of specially developed "representative" panels, as well as the test systems for comparison ("reference" test systems). Then "minipanels" are formed from specimens included into the "representative" panels, which are used for the evaluation of the quality of laboratory investigations made in the screening laboratories. The high quality of such system for controlling the quality of the detection of HBsAg in the screening laboratories of Moscow is shown.  相似文献

Prevalence rates (PRs) for EFP (schizophrenic, schizoaffective and affective psychoses), with allowance for proband sex and age-of-onset data were studied in a subdivided population from the North-East of the European Region of the USSR. The population includes three subpopulations: a small old religious semi-isolate of Russians ("Rs"), aboriginal Komi people ("Ks")--an ethnic community of Ugro-Finnish lineage, and a mixed group of migrants ("Ms") from various regions of the USSR. The latter is mainly an urban population, while the "Rs" and "Ks" are, on the whole, rural populations. The total PR for EFP was found to be 0.97% for the "Rs", 0.63% for the "Ks" and 0.35% for the "Ms", whereas PRs-0.85-1.15% in other parts of the USSR, mainly for "panmixed" populations in large towns. The lower PRs for EFP in the "Ms" is caused by a backmigration flow involving certain groups of patients; consequently, the mean liability for "Ms" offsprings (as a whole) should also be lower. On the other hand, the lower PRs for EFP in the "Ks" is caused by underpresentation of clinically mild cases of the mental disease (mainly, pseudoneurotic schizophrenia), especially among female patients, probably due to that the so affected persons are sufficiently adapted to the cultural traditions of this rural population. It was shown that in the "Rs" the total PR for "nuclear" and paranoid schizophrenia is 0.68% versus 0.25% in a "panmixed" population. The increase is most likely caused by the high inbreeding level in the "Rs" semi-isolate, and if this is correct, we may suppose that at least one or two recessive genes are involved in the liability to the most heavy forms of schizophrenia. On the other hand, in the "Ms" (as in other "panmixed" populations) positive assortative mating among hereditary-predisposed persons is a more significant factor influencing family transmission of EFP, since the correlation between probands and their spouses is rpp = 0.31 (p less than 0.001) in the "Ms", as compared to rpp = 0.19 (p less than 0.1) in the "Rs". Thus, our general conclusion is that neither the place of inhabitance nor the life mode are the causal factors for EFP, but rather some genetic factors, more accurately, certain sets of specific genes.  相似文献

The most primitive code is assumed to be a GC code: GG coding for glycine, CC coding for proline, GC coding for alanine, CG coding for arginine. The genetic code is assumed to have originated with the coupling of glycine to its anticodon CC mediated by a copper-montmorillonite. The polymerization of polyproline followed when it was coupled to its anticodon GG. In this case the aminoacyl-tRNA synthetase was a copper-montmorillonite. The first membrane is considered to be a sheet formed from polyglycine. As the code grew more complicated, the alternative hydrophobic-hydrophilic polypeptide (alanine-arginine) was coded for by the alternating CG copolymer. This alternating polypeptide (ala-arg) began to function as both a primitive membrane and as an aminoacyl-tRNA synthetase. The evolution of protein structure is tightly coupled to the evolution of the membrane. The a helix was evolved as lipids became part of the structure of biological membranes. Summary Pituitary glands from juvenile (pre-pubertal) and adult male and female rhesus monkeys were examined following immunocytochemical staining with antisera to the beta subunits of ovine luteinizing hormone (LH) and of human thyroid stimulating hormone (TSH). The LH antiserum reacts with a cell that is PAS-positive, occurs singly and is randomly distributed throughout the pars distalis. The diameter of these cells is approximately 11.5 m. They do not seem to vary in number in either juveniles (pre-pubertals) or adults, or in males or females. There appears to be fewer LH cells in the pituitary glands of pregnant and lactating females. In addition to staining cells in the pars distalis, the antiserum also reacts with a population of cells located in the pars tuberalis.The cells that stain with the anti-TSH serum are confined primarily to the pars distalis. They are approximately 15.8 m in diameter and are generally found in groups or clusters located in the anterior and medial regions of the gland. The TSH cells vary in number from one animal to another; however, this variability is unrelated to the age or the sex of the animals. Summary The ultrastructure of the heterocyst and its development from the vegetative cell is described. The ultrastructure of the akinete is also described. The mature heterocyst (still attached to the filament) has an elaborate structure which is distinct from both that of the normal vegetative cell and the akinete (the normal reproductive cell).Despite the extra structural detail seen in the electron micrographs, the observations do not indicate a likely physiological role for the heterocyst. However, since the developing-mature heterocyst has an organised structure, it is probable that it has an active metabolism. In contrast, the detached heterocyst has a highly disorganised structure and, for this reason, it is likely to be metabolically inactive and incapable of germination.  相似文献

Summary Pimozide, a specifie blocker of dopaminergic receptors, was injected for 4 or 9 days in freshwater (FW) eels or eels acclimated to sea water (SW) for 10 or 30 days. The daily dose was 100 or 200 g/100g. Melanophore index values increase in FW and in 1 month-SW injected eels. All the treated fish react by a total or subtotal degranulation of the lead-hematoxylin positive cells in the pars intermedia. These cells were previously identified as -MSH-secreting cells. The MSH cell nuclear area is significantly increased, nucleoli are larger and the endoplasmic reticulum more developed. The intensity of the response is similar in FW and SW eels, but it does not increase with the higher dose. The rapid release of pituitary -MSH is also visualized by immunofluorescence and immunoenzymologic techniques. No effect on the second cell type of the pars intermedia (PAS-positive cell) is detected. The amount of neurosecretory material is often reduced in the neurohypophysis. These results suggest that the hypothalamic inhibitory control of MSH release and synthesis is mediated through dopaminergic fibers in the eel, but other factors cannot be ignored in this regulation.The author is grateful to Dr. P.A.J. Given the relatively small number of microarrays typically used in gene-expression-based classification, all of the data must be used to train a classifier and therefore the same training data is used for error estimation. The key issue regarding the quality of an error estimator in the context of small samples is its accuracy, and this is most directly analyzed via the deviation distribution of the estimator, this being the distribution of the difference between the estimated and true errors. Past studies indicate that given a prior set of features, cross-validation does not perform as well in this regard as some other training-data-based error estimators. The purpose of this study is to quantify the degree to which feature selection increases the variation of the deviation distribution in addition to the variation in the absence of feature selection. To this end, we propose the coefficient of relative increase in deviation dispersion (CRIDD), which gives the relative increase in the deviation-distribution variance using feature selection as opposed to using an optimal feature set without feature selection. The contribution of feature selection to the variance of the deviation distribution can be significant, contributing to over half of the variance in many of the cases studied. We consider linear-discriminant analysis, 3-nearest-neighbor, and linear support vector machines for classification; sequential forward selection, sequential forward floating selection, and the -test for feature selection; and -fold and leave-one-out cross-validation for error estimation. We apply these to three feature-label models and patient data from a breast cancer study. In sum, the cross-validation deviation distribution is significantly flatter when there is feature selection, compared with the case when cross-validation is performed on a given feature set. Summary The method of Glenner et al. for the histochemical demonstration of MAO activity was studied by means of scanning microdensitometry and discrete measurement of optical density (-590 nm) of the liver and brain tissues sections.The experiments indicated that: (1) The optimal time of incubation (the thickness of sections is 15 m) is 60–90 min. (2) The histochemical reaction proceeds with the following substrates: dopamine, noradrenalin, serotonin, and tryptamine. (3) Iproniazid is the best inhibitor for preincubation whereas for simultaneous inhibition the substrate semicarbazide is better. (4) The incubation under the anaerobic conditions caused a considerable decrease of the stain intensity of the sections. We consider these data to indicate that both the aldehydes and acids formed under oxidation may take part in direct reduction of NBT to diformazan. (5) The histochemical reaction for MAO without substrates testifies to the presence of endogenous amines or other redox reactions leading to the reduction of NBT.  相似文献

It is shown that and how it is possible to single out the chain fold which is thermo-dynamically most stable. The suggested approach is based on two physical ideas: A "molecular field" approximation permits to examine all protein structures which belong to the same "folding pattern". Only a limited set of the "potentially stable" folding patterns have to be examined. The general approach is illustrated by calculations of the stable folds for two beta-domains.  相似文献

Summary Myofilaments of striated muscles can be recognized in the electron microscope to be in structural continuity with the outer membrane of the nuclear envelope. The very site of insertion of these myofilaments at the membrane surface frequently appears characterized by a dense basal knob of 85–135 Å. It is hypothesized that this attachment of myofilaments to the nuclear membrane plays a role in mechanically transmitting the contraction of the fiber to the nucleus, thus bringing about the harmonica-like folded appearance of the nucleus which is known for the contracted states of striated, smooth and cardiac muscles.The work was supported in part by the Deutsche Forschungsgemeinschaft.The author is indebted to Miss Sigrid Krien and Miss Marianne Winter for careful technical assistance as well as to Drs. Hepatitis C virus (HCV) is a liver-tropic pathogen with severe health consequences for infected individuals. Chronic HCV infection can progress to cirrhosis and hepatocellular carcinoma and is a leading indicator for liver transplantation. The HCV core protein is an essential component of the infectious virus particle, but many aspects of its role remain undefined. The C-terminal region of the core protein acts as a signal sequence for the E1 glycoprotein and undergoes dual processing events during infectious virus assembly. The exact C terminus of the mature, virion-associated core protein is not known. Here, we performed genetic analyses to map the essential determinants of the HCV core C-terminal region, as well as to define the minimal length of the protein that can function for infectious virus production in trans.Hepatitis C virus (HCV) is a major contributor to the development of human liver diseases, infecting approximately 2% of the population, or 130 million people, worldwide (2). Up to 80% of HCV infections progress to chronic hepatitis and can lead to cirrhosis and hepatocellular carcinoma (38). No vaccine exists to prevent HCV infection, and current treatments are frequently inadequate.HCV is an enveloped virus of the genus Hepacivirus in the family Flaviviridae (30). The single-stranded, positive-sense RNA genome encodes a polyprotein of about 3,000 amino acids, which is processed by viral and host proteases into three structural proteins (the core protein, E1, and E2) and seven presumed nonstructural proteins (p7, NS2, NS3, NS4A, NS4B, NS5A, and NS5B). The core protein is thought to encapsidate the RNA genome within the virion, forming a complex that is surrounded by a host cell-derived lipid bilayer displaying the envelope glycoproteins, E1 and E2. Although not thought to be components of the virion, p7 and NS2 have recently been implicated in the production of infectious virus (5, 12, 29, 33, 39). The remaining nonstructural proteins, NS3 to NS5B, are essential for genome replication and have additional emerging roles in virus assembly. NS3 possesses RNA helicase/NTPase activities and together with its cofactor, NS4A, forms the major viral protease. NS5B is the RNA-dependent RNA polymerase (reviewed in references 16 and 27).The core protein is the first protein produced during translation of the incoming viral genome. A signal sequence in its C-terminal region targets the nascent E1 glycoprotein to the endoplasmic reticulum (ER) membrane and is the substrate for processing by two host proteases. Cleavage by signal peptidase (SP) following core amino acid 191 (31) is thought to precede processing by signal peptide peptidase (SPP) (20, 26), an integral membrane aspartyl protease that cleaves within transmembrane segments (37). The C terminus of the mature, infectious-virion-associated core protein has not been determined, but it is speculated to lie between amino acids 173 and 182 (24, 31). SPP processing has been shown to mobilize the core from the ER membrane and enable it to traffic to lipid droplets (20). These triglyceride-rich storage organelles have recently been shown to be the sites of HCV particle assembly (21). Consistent with this finding, impaired SPP activity leads to decreased HCV infectious titers (34). Dual processing of the core proteins is a common feature of the Flaviviridae family. GB virus B, a hepacivirus, and classical swine fever virus, a related pestivirus, encode core proteins that undergo SP and SPP processing during maturation (8, 35). In the genus Flavivirus, the capsid protein undergoes regulated cleavage by the viral NS2B-3 protease and SP; this stepwise processing has been shown to be essential for proper encapsidation of genomes into infectious particles (3).The development of an infectious cell culture system for HCV has been a major breakthrough in the field (7). Many details of virus morphogenesis and infectivity, however, are still unknown. In this study, we examined the role of the C-terminal portion of the HCV core protein and identified individual amino acids that are essential for infectious virus assembly and core protein stability. Findings from alanine-scanning and transcomplementation studies suggest that at least 177 residues of the core protein are needed to produce infectious particles.  相似文献

Summary Fragile-X syndrome is a major cause of mental retardation in humans. The X-inactivation imprinting model accounts for the unusual pattern of inheritance and expression of this syndrome. According to this model, the fragile-X mutation creates a local block to the attempted reactivation of the mutant X chromosome prior to oogenesis. This local block results in an imprinted fragile-X chromosome that is deleterious in males and in females for whom this chromosome is predominantly the active X chromosome. The imprinted state of the fragile-X mutation is inferred to be stable when transmitted by an imprinted female because the penetrance of the syndrome in sons of affected females is estimated to be 1.0. To provide a more precise estimate of the stability of the proposed fragile-X imprint, we have analyzed published pedigrees that include restriction fragment length polymorphism and cytogenetic data from sibships with mothers who are interpreted as having an imprinted fragile-X allele. We conclude that the fragile-X imprint was stable in 46 out of 48 female meioses. This analysis leads to a preliminary estimate of about 96% for the stability of the imprint through female meiosis. Two imprinted females had progeny who appeared to be carriers of a nonimprinted fragile-X allele. If this interpretation is correct, then reversion from the imprinted to the nonimprinted state, or erasure, can occasionally occur when the mutant fragile-X allele is transmitted by an imprinted female. We discuss the genetic and epigenetic significance of possible female erasure. We request DNA and cytogenetic information from unpublished pedigrees to quantify further the stability, during female meiosis, of the proposed imprinted state of the mutant fragile-X allele.  相似文献

The aim of our investigation is to understand the mechanisms which control the movement of the human arm. The arm is here considered as a redundant system: the shoulder, elbow and wrist joints, which provide three degrees of freedom, combine to move the hand in a horizontal plane, i.e. a two dimensional space. Thus the system has one extra degree of freedom. Earlier investigations of the static situation led to the hypothesis that independent cost functions were attached to each of the three joints and that the configuration chosen for a given target position is that which provides the minimum total cost (Cruse 1986). The aim of the current investigation was to look for measurable values corresponding to the hypothetical cost functions. Experiments using pointers of different lengths attached to the hand showed that the strategy in choosing the joint angles are independent of the limb length. The muscle force necessary to reach a given angle is increased by a spring mounted across a joint. In this situation the angles of the loaded joint are changed for a given target point to give way to the force effect. This leads to the conclusion that the hypothetical cost functions are not independent of the physiological costs necessary to hold the joint at a given angle. The cost functions seem to depend on joint angle and on the force which is necessary to hold the joint in a given position. Cost functions are measured by psychophysical methods. The results showU-shaped curves which can be approximated by parabolas. The position of minimum cost (maximum comfort) for one joint showed no or weak dependency on the angles of the other joints. For each subject these psychophysical cost functions are compared with the hypothetical cost functions. The comparison showed reasonable agreement. This supports the assumption that the psychophysically measured comfort functions provide a measure for the hypothetical cost functions postulated to explain the targeting movements. Targeting experiments using a four joint arm which has two extra degrees of freedom showed a much larger scatter compared to the three joint arm. Nevertheless, the results still conform to the hypothesis that also in this case the minimum cost principle is applied to solve the redundancy problem. As the cost function for the whole arm shows a large minimum valley, quite a large range of arm positions is possible of about equal total costs. The scatter does not result from pure randomness but seems to be mainly produced by the fact that the angles at the end of the movement depend on the value of the joint angles at the beginning of the movement.  相似文献

This paper is a sequel to an earlier paper which proposed an active role for the thalamus, integrating multiple hypotheses formed in the cortex via the thalamo-cortical loop. In this paper, I put forward a hypothesis on the role of the reciprocal, topographic pathways between two cortical areas, one often a higher area dealing with more abstract information about the world, the other lower, dealing with more concrete data. The higher area attempts to fit its abstractions to the data it receives from lower areas by sending back to them from its deep pyramidal cells a template reconstruction best fitting the lower level view. The lower area attempts to reconcile the reconstruction of its view that it receives from higher areas with what it knows, sending back from its superficial pyramidal cells the features in its data which are not predicted by the higher area. The whole calculation is done with all areas working simultaneously, but with order imposed by synchronous activity in the various top-down, bottom-up loops. Evidence for this theory is reviewed and experimental tests are proposed. A third part of this paper will deal with extensions of these ideas to the frontal lobe.  相似文献

The pectinase produced during the anaerobic retting of flax appeared to have a pH optimum between 5.4 and 5.6. Continuous neutralization during the fermentation, however, induced the production of a pectinase with a pH optimum of 7.1.The majority of the pectin splitting bacteria in the acid and in the neutral retting liquors belonged to the speciesClostridium pectinovorum.Pure culture studies withCl. pectinovorum at fixed pH levels showed that the pectinase produced is adapted to the pH of the environment. No differences in pH dependency could be established between the hemicellulases from the acid and the neutral retting liquor.Although a pH of 6.5 is far from optimal for the hemicellulase, the actual hemicellulolytic activity in the neutral pH retting method is greater than the activity of the normal, acid retting liquor. This fact supports the theory that hemicellulase is responsible for losses in fibre quality and quantity in the neutral pH retting method.  相似文献

Brown adipose tissue mitochondria predominantly oxidize fatty acids in order to generate heat for non-shivering thermogenesis, and have an unusually high capacity for net transfer of long-chain fatty acyl groups from the outer to the inner (matrix) compartment. The activities of the "outer" and "inner" carnitine long-chain acyltransferases have been estimated in isolated mitochondria of cold-acclimated guinea pits by the continuous spectrophotometric recording of the redox level of flavoproteins in the acyl-CoA dehydrogenase pathway. This redox level is determined by the intramitochondrial content of acyl-CoA under the selected experimental conditions. The apparent initial rate of the "inner" acyltransferase (palmitoyl-L-carnitine added) is three order of magnitudes higher than the "outer" acyltransferase (palmitoyl-CoA added), and this difference is not influenced by the substrate concentration, pH and reaction temperature. Thus, the "outer" acyltransferase reaction is rate limiting in the transfer of long-chain acyl groups across the inner membrane of these mitochondria and catalyzes a non-equilibrium reaction in the intact organelle. Estimates of the absolute rate of the "outer" long-chain acyltransferase indicate that it exceeds that of rat liver mitochondria by a factor of 20.  相似文献

Summary A method for the demonstration of the activity of phosphoglucomutase in tissues is described. In the histochemical system the enzyme converts the substrate -D-glucose-1-phosphate to -D-glucose-6-phosphate. The resulting -D-glucose-6-phosphate is oxidized by exogenous glucose-6-phosphate dehydrogenase to D-glucono--lactone-6-phosphate, whereby the activity of endogenous NADPH-tetrazolium oxidoreductase reduces Nitro-BT to the slightly soluble diformazan. The problems involved in the histochemical demonstration of phosphoglucomutase are discussed.  相似文献
