Micrococcus sedentarius, an organism associated with pitted keratolysis, produced two proteinases in culture supernatant fluids, as shown by non-denaturing PAGE with overlaying with a casein substrate. A mixture had optimal activity at pH 10 with azocasein substrate. At pH 7.1 and 8.1 in continuous culture with varying dilution rates high proteinase production occurred at relative specific growth rates (mu rels) 0.39 and 0.77 and biomass concentrations decreased with increasing dilution rate. One proteinase was constitutive and varied little in production with different growth rates. The other proteinase was under control with high production at low growth rates and no production at high growth rates. With varying pH at mu rels 0.39 and 0.77 maximum biomass concentration and proteinase production occurred between pH 8.0 and 9.0 as did the highest specific growth rate. These results support the hypothesis that Mic. sedentarius produces pitting in the stratum corneum when the skin is hydrated and the pH rises above neutrality.  相似文献

Abstract Hyphomicrobium X was grown on a range of mixtures of methylamine and ethanol at various dilution rates in continuous cultures. It was shown that both substrates were utilized simultaneously at all dilution rates below μmax ethanol, the biomass being the sum of the biomass obtained during growth on the single substrates alone. No evidence was observed for a redistribution of the carbon-flow nor the ability to utilize the 'poorer' growth substrate-ethanol, at dilution rates greater than μmax ethanol. Work is presented which suggests that the assimilatory pathway enzymes for either the C1- or the C2-compound are regulated coordinately, but separately from the dissimilatory pathway enzymes associated with that compound.  相似文献

Growth and proteinase production by Micrococcus sp. INIA 528 in a batch-operated laboratory fermentor were investigated, with trypticase soy broth as the basal medium for studies on optimum temperature, pH and medium composition. Maximum growth was recorded at 34°C and pH 715, whereas optimum temperature and pH for proteinase production were 31°C and pH 6.25. Maximum rate of enzyme production occurred during the late log and early stationary phases of growth. Addition of 5.0 g 1-1 yeast extract, 1.0 g 1-1 glucose, 1.0 g 1-1 MgSO4 or 1.0 g 1-1 K2HPO4 to basal medium resulted in a lower enzyme yield, but supplementation of basal medium with 2.5 g 1-1 (NH4)2SO4 increased enzyme production by 45%. A high initial biomass added to fresh broth supplemented with 2.5 g 1-1 (NH4)2SO4 only increased enzyme activity by 19%, compared to the maximum enzyme activity achieved with the standard inoculum.  相似文献

Interactive effects of elevated atmospheric CO2 and arbuscular mycorrhizal (AM) fungi on biomass production and N2 fixation were investigated using black locust ( Robinia pseudoacacia ). Seedlings were grown in growth chambers maintained at either 350 μmol mol−1 or 710 μmol mol−1 CO2. Seedlings were inoculated with Rhizobium spp. and were grown with or without AM fungi. The 15N isotope dilution method was used to determine N source partitioning between N2 fixation and inorganic fertilizer uptake. Elevated atmospheric CO2 significantly increased the percentage of fine roots that were colonized by AM fungi. Mycorrhizal seedlings grown under elevated CO2 had the greatest overall plant biomass production, nodulation, N and P content, and root N absorption. Additionally, elevated CO2 levels enhanced nodule and root mass production, as well as N2 fixation rates, of non- mycorrhizal seedlings. However, the relative response of biomass production to CO2 enrichment was greater in non-mycorrhizal seedlings than in mycorrhizal seedlings. This study provides strong evidence that arbuscular mycorrhizal fungi play an important role in the extent to which plant nutrition of symbiotic N2-fixing tree species is affected by enriched atmospheric CO2.  相似文献

The genus Pectinatus has been often reported in beer spoilage with off-flavours. The bacteria are strictly anaerobic, Gram-negative rods. Propionate and acetate are the main fermentation products from glucose in the two species belonging to the genus, P. cerevisiiphilus and P. frisingensis. Amino acids routinely present at a high level in beer were not growth substrates for both species, and a significant accumulation of succinate was observed with lactate as growth substrate. Both Pectinatus ssp. showed almost identical fermentation balances on glucose. Growth kinetics of both glucose-grown species were unchanged under a N2, H2 or 20% CO2-containing atmosphere. Combinations of culture medium pH values from pH 3·9 to pH 7·2, of glucose levels between 5 and 55 mmol l-1, and of lactate concentrations varied from 4 to 40 mmol l-1 demonstrated that biomass and volatile fatty acids production were proportional to glucose concentration for both Pectinatus species. A significant increase of volatile fatty acid production was measured for both species at the lowest pH values with a lactate or a glucose concentration increase. The maximum biomass production was observed at pH 6·2 for P. cerevisiiphilus , and between pH 4·5 and pH 4·9 for P. frisingensis. Glucose and lactate or pH value were dependent with regard to propionate and acetate production in P. frisingensis. On the other hand, the variations of these three parameters were independent with regard to biomass production for both strains, and to volatile fatty acids production for P. cerevisiiphilus. Addition of ethanol to glucose-grown cultures completely inhibited growth at 1·3 mol l-1 ethanol for P. cerevisiiphilus , and at 1·8 mol l-1 for P. frisingensis.  相似文献

Abstract From chemostat enrichments conducted at dilution rates of 0.025, 0.12 and 0.25 h−1 glutamate- and aspartate-fermenting bacteria were isolated. The dominant aspartate-fermenting strains in all these enrichments belonged to the genus Campylobacter , whereas 3 dissimilar types of glutamate-fermenting bacteria predominated at the different dilution rates. One of these strains was identified as Clostridium cochlearium . The remaining two were designated as strain DKglu16 (glutamate → acetate + propionate + ammonium + carbon dioxide) and DKglu21 (glutamate → acetate + formate + ammonium + carbon dioxide). Grown in continuous culture under glutamate limitation, strain DKglu16 (μmax= 0.13 h−1; K s= 1.9 μM) outcompeted C. cochlearium (μmax= 0.36 h−1; K s= 7 μM) at low dilution rates, but was outgrown at higher rates of dilution (0.044 h−1). In glutamate-limited continuous culture the competitiveness of strain DKglu16 increased considerably when lactate was added to the feed in addition to glutamate.  相似文献

Lactobacillus plantarum was found to produce extracellular polygalacturonase (EC 3.2.1.15.). Maximum enzyme production was obtained in a medium containing 0.5% glucose and 1.5% low methyl-pectin as inducer at 27°C at an initial pH of 6.8. Enzyme production was strongly inhibited by 5 μmol/l NiCl2, 5 μmol/l CoCl2, 5 μmol/l CuSO4, and 10 μmol/l ZnCl2. MnSO4 and MgSO4 at 200 μmol/l and 50 μmol/l respectively seemed to enhance enzyme biosynthesis. The optimal pH and temperature for enzyme activity were 4.5 and 30°C respectively. Enzyme production in batch culture accompanied growth.  相似文献

In batch culture the maximum specific growth rate (mum) and the substrate yield coefficient (Yps) varied according to sugar concentration. When the fermentation was carried out with 1% sugar, mum and Ys were 0.47/h and 42.6%, respectively. The best yields occurred in a chemostat at the pH range of 3.5 to 4.5 and temperature of 30 C. A beneficial effect on Ys was observed when the dilution rate (D) was increased. At a D of 0.55/h, the productivity was 2.38 g/liter per h. The maintenance coefficient attained a value of 0.09 g of sugar/g of biomass per h. Increases of D produced higher protein contents of the biomass. The information obtained indicates that protein production with Candida utilis, using prickly pear juice, should be carried out a high dilution rates where the Ys and protein content of the cell mass are also higher.  相似文献

Abstract Conditions of growth are described for the production of streptomycin by Streptomyces griseus ATCC 12475 using chemically defined minimal medium and complex medium. It was found using batch cultures that early synthesis of the antibiotic occurred during growth in minimal medium but was delayed until the onset of stationary phase in complex medium. This effect was independent of whether spores or vegetative cells were used as inoculum. Stability of streptomycin biosynthesis in continuous culture was dependent on dilution rate and medium employed. Cultures were highly unstable when grown on complex medium but could be maintained in steady states in continuous culture using minimal medium when the dilution rate was increased in a stepwise manner, starting at a dilution rate of 0.02 h−1 (15% of μ max). The effect of changing dilution rate on growth, streptomycin production and the level of streptomycin phosphotransferase was examined using this technique.  相似文献

Aim:  To investigate the effects of feeding and induction strategies on the production of Bm R1 recombinant antigen.
Methods and Results:  Fed-batch fermentation was studied with respect to the specific growth rate and mode of induction to assess the growth potential of the bacteria in a bioreactor and to produce high yield of Bm R1 recombinant antigen. Cells were grown at a controlled specific growth rate (μset) during pre-induction, followed by constant feeding postinduction. The highest biomass (24·3 g l−1) was obtained during fed-batch process operated at μset of 0·15 h−1, whereby lower μset (0·075 h−1) gave the highest protein production (9·82 mg l−1). The yield of Bm R1 was increased by 1·2-fold upon induction with 1 mmol l−1 IPTG (isopropyl-β- d -thiogalactoside) compared to using 5 mmol l−1 and showed a further 3·5-fold increase when the culture was induced twice at the late log phase.
Conclusions:  Combination of feeding at a lower μset and twice induction with 1 mmol l−1 IPTG yielded the best result of all variables tested, promising an improved method for Bm R1 production .
Significance and Impact of the Study:  This method can be used to increase the production scale of the Bm R1 recombinant antigen to meet the increasing demand for Brugia Rapid, a commercial diagnostic test for detection of brugian filariasis.  相似文献

Nitrogen regulation in tylosin production by Streptomyces fradiae NRRL 2702 was studied in chemostat culture using a soluble synthetic medium. The maximum value of specific tylosin formation rate ( q TYL) was 1·13 mg g−1 h−1 at the specific growth rate (μ) of 0·05 h−1, and q TYL decreased with increasing levels of the specific growth rate after reaching a rate of 0·1 h−1. The optimum conditions for tylosin formation were that the specific ammonium ion uptake rate ( q N) and μ were 0·13 mmol g−1 h−1 and 0·05 h−1, respectively. The specific formation rates of threonine dehydratase (TDT) and tylosin were repressed by high levels of specific ammonium ion uptake rate. This study showed the adaptation to chemostat cultures of the nitrogen regulation of tylosin fermentations.  相似文献

The production of acid phosphatases (E.C.3.1.3.2, ACPs) by Aspergillus niger N402A is regulated by specific growth rate, as well as phosphate availability and pH, as demonstrated by studies in continuous flow culture. Specific ACP activity was highest when A. niger was grown at pH 6.3 (64±8 U g−1) or pH 2.8 (99±11 U g−1), at a dilution rate of 0.07 h−1 and phosphate concentrations below 0.46 mM. ACP production was growth correlated for specific growth rates between 0.07 and 0.13 h−1. Four different ACPs, including two phytases, were produced by A. niger N402A. The ACP and the phytase with maximal activities at pH 5.5 were differentially expressed at different culture pH values, with greater production at low pH.  相似文献

Clostridium pasteurianum fermented glucose to acetate, butyrate, CO2 and H2. In batch cultures the fermentation pattern was only slightly affected by culture pH over the range 8·0 to 5·5. The acetate/butyrate ratio was always higher than or equal to one. Between 2·14 and 2·33 mol H2 was produced per mol glucose fermented. At unregulated pH, more butanol and less butyrate was formed. In a carbon-limited chemostat, the steady-state acetate/butyrate ratio was always lower than one. H2 production was approximately 1·70 mol per mol glucose consumed. Substantial amounts of extracellular protein were formed. With decreasing pH, acetate and formate production decreased, while H2 production was highest at pH 6.0. With increasing dilution rate ( D ), the product spectrum hardly changed, but more biomass was formed. Y glucosemax and Y ATPmax were 55·97 and 31·48 g dry weight per mol glucose or ATP respectively. With increasing glucose input the formation of fatty acids and H2 slightly decreased.
Continuous cultures fermented mannitol to acetate, butyrate, butanol, CO2 and H2. With acetate as co-substrate, butanol production and molar growth yields, Y mannitol and Y ATP, markedly decreased, while the butyrate and H2 production increased. The latter reached a value of 2·21 mol H2 per mol mannitol consumed.  相似文献

The response of Phaseolus vulgaris L. cv. Contender grown under controlled environment at either ambient or elevated (360 and 700 μmol mol-1, respectively) CO2 concentrations ([CO2]), was monitored from 10 days after germination (DAG) until the onset of senescence. Elevated CO2 had a pronounced effect on total plant height (TPH), leaf area (LA), leaf dry weight (LD), total plant biomass (TB) accumulation and specific leaf area (SLA). All of these were significantly increased under elevated carbon dioxide with the exception of SLA which was significantly reduced. Other than high initial growth rates in CO2-enriched plants, relative growth rates remained relatively unchanged throughout the growth period. While the trends in growth parameters were clearly different between [CO2], some physiological processes were largely transient, in particular, net assimilation rate (NAR) and foliar nutrient concentrations of N, Mg and Cu. CO2 enrichment significantly increased NAR, but from 20 DAG, a steady decline to almost similar levels to those measured in plants grown under ambient CO2 occurred. A similar trend was observed for leaf N content where the loss of leaf nitrogen in CO2-enriched plants after 20 DAG, was significantly greater than that observed for ambient-CO2 plants. Under enhanced CO2, the foliar concentrations of K and Mn were increased significantly whilst P, Ca, Fe and Zn were reduced significantly. Changes in Mg and Cu concentrations were insignificant. In addition. high CO2 grown plants exhibited a pronounced leaf discoloration or chlorosis, coupled with a significant reduction in leaf longevity.  相似文献

The accepted food yeast Saccharomyces fragilis was grown in batch and chemostat culture on coconut water and on a simulated coconut-water medium containing glucose, fructose, sucrose and sorbitol, to provide kinetic data for a feasibility study of microbial protein production. Analyses of growth on individual and mixed carbon substrates were made to determine sugar assimilation patterns in batch and chemostat cultures on coconut water. Growth on the polyol produced a much reduced specific growth rate, assimilation rate, growth yield and productivity compared to growth on the sugars. In mixed substrate fermentations a sequential utilization of the carbohydrates occurred. Both the monosaccharides repressed invertase synthesis and all three sugars repressed sorbitol assimilation. Complete carbon assimilation was only obtained by prolonged batch fermentation or in chemostat cultures at low dilution rates (<0.10 h-1). Supplementation of coconut water with biotin and nicotinic acid increased biomass yields in chemostat cultures.  相似文献

SUMMARY: A quantitative study of the growth and carbohydrate metabolism of Aerobacter aerogenes in continuous culture with particular reference to 2:3-butanediol production is described. The effects of varying dilution rate, oxygen uptake rate, pH value, temperature and CO2 pressure were studied.
The optimum pH value was 5·0°6·0. The optimum temperature was 35–37° with a dilution rate of 0·2 hr−1. The rate of sugar metabolism was decreased by raising the CO2 pressure.
In continuous culture the maximum diol output rate/1. of working volume exceeded the greatest rate reported for batch culture by a factor of three. The maximum diol concentration and yield as a percentage of the theoretical maximum were lower in continuous culture than in batch culture. Reasons for this are given.
Some variants of the parent organism were found in continuous cultures but they caused no change in the yield of butanediol. There was no sign of loss of butanediol-producing ability by any of the cultures, some of which were maintained for 2,000 hr. For the first few generations the rate of production of butanediol actually increased.  相似文献

The growth behaviour of the thermophilic anaerobic bacterium Thermoanaerobium brockii for the production of its intracellular secondary alcohol dehydrogenase (sADH) has been studied in batch cultures as well as in continuous cultivation with complete cell recycling. In batch culture the maximum specific growth rate, μMAX, was 0·5 h−1, resulting in a cell density of 1·2 g l−1 and an sADH activity of 1·3 units ml−1. Higher glucose concentrations resulted in a decrease in ep cf7 max rs, enzyme productivity as well as biomass yield although an increase in total biomass was achieved. To improve cell density and productivity, continuous culture with complete cell recycling was used, resulting in an increase in cell density by 5 times and in productivity of the sADH by 3 times in comparison to those obtained in batch culture.  相似文献

Abstract. The objective of this study was to investigate the effects of water stress in sweet potato ( Ipomoea batatas L. [Lam] 'Georgia Jet') on biomass production and plant-water relationships in an enriched CO2 atmosphere. Plants were grown in pots containing sandy loam soil (Typic Paleudult) at two concentrations of elevated CO2 and two water regimes in open-top field chambers. During the first 12 d of water stress, leaf xylem potentials were higher in plants grown in a CO2 concentration of 438 and 666 μmol mol−1 than in plants grown at 364 μmol mol−1. The 364 μmol mol−1 CO2 grown plants had to be rewatered 2 d earlier than the high CO2-grown plants in response to water stress. For plants grown under water stress, the yield of storage roots and root: shoot ratio were greater at high CO2 than at 364 μmol mol−1; the increase, however, was not linear with increasing CO2 concentrations. In well-watered plants, biomass production and storage root yield increased at elevated CO2, and these were greater as compared to water-stressed plants grown at the same CO2 concentration.  相似文献

The long-term response of Arabidopsis thaliana to increasing CO2 was evaluated in plants grown in 800 μl l−1 CO2 from sowing and maintained, in hydroponics, on three nitrogen supplies: "low,""medium" and "high." The global response to high CO2 and N-supply was evaluated by measuring growth parameters in parallel with photosynthetic activity, leaf carbohydrates, ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) messenger RNA and protein, stomatal conductance (gs) and density. CO2 enrichment was found to stimulate biomass production, whatever the N-supply. This stimulation was transient on low N-supply and persisted throughout the whole vegetative growth only in high N-supply. Acclimation on low N–high CO2 was not associated with carbohydrate accumulation or with a strong reduction in Rubisco amount or activity. At high N-supply, growth stimulation by high CO2 was mainly because of the acceleration of leaf production and expansion while other parameters such as specific leaf area, root/shoot ratio and gs appeared to be correlated with total leaf area. Our results thus suggest that, in strictly controlled and stable growing conditions, acclimation of A. thaliana to long-term CO2 enrichment is mostly controlled by growth rate adjustment.  相似文献
