Abstract.  Infestation of the head louse Pediculus humanus var capitis DeGeer (Phthiraptera: Pediculidae) is an important public health problem in Australia, with up to a third of children infested in some primary schools. Insecticide resistance and inadequate attention to the application instructions of topical pediculicides are common reasons for treatment failure. This study evaluated six popular Australian over-the-counter products against head lice, primarily comprised of different botanical extracts, and compared them with permethrin 1% (Quellada®) and a non-treatment control in order to assess their in vitro efficacy. We also assessed commonly used criteria for evaluating pediculicide efficacy in vitro. All tested products failed to demonstrate high levels of efficacy with the exception of Tea Tree Gel®, which outperformed 1% permethrin. Permethrin had a high level of efficacy, but using stringent criteria 18% of lice were not dead at 3 h, indicating some resistance to Quellada®. Commonly used less stringent criteria were shown to overestimate mortality of head lice as a result of the protective phenomenon of stasis or sham death observed in exposed lice that may recover after some time. Using two different levels of stringency resulted in different rankings of efficacy for most products, with the exception of the first ranked product, Tea Tree Gel®. Rankings of efficacy also varied over time, even within the different assessment criteria. Government regulatory agencies should require standard in vitro tests using stringent mortality criteria, with an observation period of ≥ 6 h, to determine the efficacy of new pediculicides, and only products that cause a minimum mortality rate (e.g. 80%) in head lice collected from the target population should be licensed for sale.  相似文献

The safety and efficacy of treatment with a new pediculicide lotion, Prioderm (0.5% malathion in isopropanol), were compared with those of treatment with Kwellada (1% lindane) shampoo in a randomized trial of children with head lice. The children''s scalps were examined for live lice immediately, 7 days and 4 to 6 weeks following treatment. To determine the in-vitro ovicidal effect of treatment, samples of hair with nits were removed before and immediately following treatment; the subsequent rates of hatching were compared. No live lice were present immediately following either treatment. At 7 days after treatment 2 of the 29 children treated with Prioderm lotion and 4 of the 33 children treated with Kwellada shampoo were infested with live lice, whereas at 4 to 6 weeks after treatment 5 and 3 children respectively were infested. The initial effectiveness of treatment was 93% in the children treated with Prioderm lotion and 88% in those treated with Kwellada shampoo; however, a large difference in efficacy could have been missed owing to the small number of children in the study. Both preparations demonstrated in-vitro ovicidal activity, but neither totally abolished post-treatment hatching. No side effects were reported from either preparation. Because ovicidal activity was incomplete two treatments with the same pediculicides should be given about 7 days apart.  相似文献

Head lice, Pediculus capitis , were collected from children aged 3–12 years in Maale Adumin, a town near Jerusalem, after reports of control failure with the pyrethroid insecticide permethrin. A total of 1516 children were examined: living lice and eggs were found on 12.1% of the children; or another 22.8% of the children only nits were found. Twice as many girls as boys (8.1% v 4%) were infested with lice and or nits. Head lice collected from infested children were exposed to permethrin impregnated filter-papers. Log time probit mortality (ltp) regression lines were calculated for mortality data and compared to ltp lines for a similar collection of head lice made in 1989. The regression lines for the two years were significantly different, with a 4-fold decrease in susceptibility at the LT50 level between 1989 and 1994. The slopes of the lines also suggested that the 1994 population was more heterogenous in its response to permethrin than the 1989 population. In contrast, a laboratory population of body lice (Pediculus humanus) tested with the same batch of permethrin-impregnated papers showed a slight but non-significant increase in susceptibility between 1989 and 1994. The results suggest that resistance to pyrethroids has developed rapidly among head lice since permethrin was introduced in 1991 as a pediculicide in Israel.  相似文献

Abstract.  The improper use of pediculicides containing permethrin has led to the development of resistance. Thus, new alternatives for control are needed. Plant-derived insecticides are attractive alternatives to common chemical insecticides because most of them are environmentally friendly and non-toxic to mammals. The toxic activity of 23 monoterpenoids belonging to several chemical classes was tested against the eggs of permethrin-resistant head lice, Pediculus humanus capitis De Geer (Phthiraptera: Pediculidae). Significant differences in ovicidal action were observed among the tested substances. The most effective chemicals were hydrocarbons and ethers, followed by ketones, alcohols, phenols and esters. A linear relationship between egg mortality and knockdown time (KT50) on adults by the tested components revealed that most of the components were effective on both egg and adult stages. The monoterpenoids described herein are good candidates as effective pediculicides.  相似文献

The toxicity of cinnamon, Cinnamomum zeylanicum, bark essential oil compounds against eggs and adult females of human head louse, Pediculus humanus capitis, was examined using direct contact and vapour phase toxicity bioassays and compared with the lethal activity of their related compounds, benzyl alcohol, cinnamic acid, cinnamyl acetate, 4-hydroxybenzaldehyde and salicylaldehyde, as well as two widely used pediculicides, d-phenothrin and pyrethrum. In a filter-paper contact toxicity bioassay with female lice at 0.25 mg/cm2, benzaldehyde was 29- and 27-fold more toxic than pyrethrum and d-phenothrin, respectively, as judged by median lethal time (LT50) values. Salicylaldehyde was nine and eight times more active than pyrethrum and d-phenothrin, respectively. Pediculicidal activity of linalool was comparable with that of d-phenothrin and pyrethrum. Cinnamomum bark essential oil was slightly less effective than either d-phenothrin or pyrethrum. Benzyl alcohol and (E)-cinnamaldehyde exhibited moderate pediculicidal activity. After 24 h of exposure, no hatching was observed with 0.063 mg/cm2 salicylaldehyde, 0.125 mg/cm2 benzaldehyde, 0.5 mg/cm2 Cinnamomum bark essential oil, 1.0 mg/cm2 (E)-cinnamaldehyde, and 1.0 mg/cm2 benzyl cinnamate. Little or no ovicidal activity was observed with d-phenothrin or pyrethrum. In vapour phase toxicity tests with female lice, benzaldehyde and salicylaldehyde were much more effective in closed containers than in open ones, indicating that the mode of delivery of these compounds was largely due to action in the vapour phase. Neither d-phenothrin nor pyrethrum exhibited fumigant toxicity. Cinnamomum bark essential oil and test compounds described merit further study as potential pediculicides or ovicides for the control of P. h. capitis.  相似文献

After evaluation of presence of nits and/or lice, PARA aerosol(bioallethrin with piperonyl butoxide) was applied to 97 women and 43 girls, regardless of the results of their infestation, living at 2 separate institutions. Twelve days after the first treatment the second treatment was carried out and the presence of head lice was examined for the evaluation of treatment effect. Among total 140 subjects, 98(70.0%) were nit positive and 85(60.7%) were louse positive. The infestation rates of girls by nit(86.0%) and louse(86.0%) were higher than those of women, nit 62.9% and louse 49.5% (p less than 0.01). Also the detection rate of lice among nit positive subjects was higher in girls(100%) than in women(78.7%). On the second evaluation 12 days after treatment, 36(97.3%) out of 37 girls and 46(95.8%) out of 48 women were found louse-free. There was no difference in the treatment efficacy between the women and girls.  相似文献

The sustained speed of kill against Dermacentor reticulatus of two topical combinations, one containing fipronil/amitraz/(S)-methoprene and the other, imidacloprid/permethrin, was evaluated in dogs. Two treated groups and one untreated control group of eight adult Beagle dogs each were randomly formed based on pre-infestation rates and bodyweight. Each treatment was administered topically once on Day 0, according to the recommended label dose and instructions for use. All dogs were infested with 50 adult unfed D. reticulatus starting on Day 1, then weekly, for a total of five weeks. While most studies determine tick efficacy at 48 hours (h), in this study, all remaining ticks were counted and categorized 24 h following each infestation. The numbers of ticks (living or dead) that remained attached on treated dogs were compared to those on the control animals. The percent reduction of attached ticks (disruption of attachment) at 24 h on dogs treated with fipronil/amitraz/(S)-methoprene remained above 92% for four weeks. The reduction of attached ticks at 24 h on dogs treated with imidacloprid/permethrin did not reach 80% during the entire study. The number of ticks attached at 24 h was significantly (p<0.05) lower in the fipronil/amitraz/(S)-methoprene group than in the imidacloprid/permethrin group in assessments on Days 2, 15, 22, 29 and 36. When assessing efficacy based upon live ticks on treated versus control dogs, fipronil/amitraz/(S)-methoprene 24 h efficacy was above 95% for four weeks, decreasing to 77.8% at Day 36. The 24 h efficacy of imidacloprid/permethrin ranged from 56.2% to 86.7% through Day 29, never achieving 90% throughout the study. The 24-hour efficacy of fipronil/amitraz/(S)-methoprene was significantly (p<0.05) higher than imidacloprid/permethrin at all time points, including Day 36.  相似文献

A tape formulation of pyriproxyfen was prepared by coating the chemical onto yellow plastic film. We evaluated the transovarial ovicidal activity of the tape formulation against Bemisia tabaci and investigated the optimal application method and efficacy in a greenhouse. The tape formulation showed high ovicidal activity of over 80% mortality against B. tabaci for 4 days by a 30-second compulsory contact test. The formulation controlled the B. tabaci population on tomatoes by both “sideways spreading application” and “vertical hanging application”. “Sideways spreading application” was considered to be more practical, although there was no significant difference in efficacy between the two application methods. In a large-scale greenhouse trial of “sideways spreading application”, the tape formulation suppressed the B. tabaci population for 77 days. The results suggested that the tape formulation is promising as a tool for IPM because of its high efficacy and lower environmental impact.  相似文献

Human head lice are blood-sucking insects causing an infestation in humans called pediculosis capitis. The infestation is more prevalent in the school-aged population. Scalp itching, a common presenting symptom, results in scratching and sleep disturbance. The condition can lead to social stigmatization which can lead to loss of self-esteem. Currently, the mainstay of treatment for pediculosis is chemical insecticides such as permethrin. The extended use of permethrin worldwide leads to growing pediculicide resistance. The aim of this study is to demonstrate the presence of the knockdown resistance (kdr) mutation in head lice populations from six different localities of Thailand. A total of 260 head lice samples in this study were collected from 15 provinces in the 6 regions of Thailand. Polymerase chain reaction (PCR) was used to amplify the α subunit of voltage-sensitive sodium channel (VSSC) gene, kdr mutation (C→T substitution). Restriction fragment length polymorphism (RFLP) patterns and sequencing were used to identify the kdr T917I mutation and demonstrated three genotypic forms including homozygous susceptible (SS), heterozygous genotype (RS), and homozygous resistant (RR). Of 260 samples from this study, 156 (60.00%) were SS, 58 (22.31%) were RS, and 46 (17.69%) were RR. The overall frequency of the kdr T917I mutation was 0.31. Genotypes frequencies determination using the exact test of Hardy-Weinberg equilibrium found that northern, central, northeastern, southern, and western region of Thailand differed from expectation. The five aforementioned localities had positive inbreeding coefficient value (Fis > 0) which indicated an excess of homozygotes. The nucleotide and amino acid sequences of RS and RR showed T917I and L920F point mutations. In conclusion, this is the first study detecting permethrin resistance among human head lice from Thailand. PCR-RFLP is an easy technique to demonstrate the kdr mutation in head louse. The data obtained from this study would increase awareness of increasing of the kdr mutation in head louse in Thailand.  相似文献

Permethrin resistance has been reported worldwide and clinical failures to commercial pediculicides containing permethrin have likewise occurred. Permethrin resistance in head lice populations from the U.S. is widespread but is not yet uniform and the level of resistance is relatively low (~ 4–8 fold). Permethrin-resistant lice are cross-resistant to pyrethrins, PBO-synergized pyrethrins and to DDT. Nix®, when applied to human hair tufts following manufacturer's instructions, did not provide 100% control when assessed by the hair tuft bioassay in conjunction with the in vitro rearing system. Resistance to permethrin is due to knockdown resistance (kdr), which is the result of three point mutations within the α-subunit gene of the voltage-gated sodium channel that causes amino acid substitutions, leading to nerve insensitivity.A three-tiered resistance monitoring system has been established based on molecular resistance detection techniques. Quantitative sequencing (QS) has been developed to predict the kdr allele frequency in head lice at a population level. The speed, simplicity and accuracy of QS made it an ideal candidate for a routine primary resistance monitoring tool to screen a large number of louse populations as an alternative to conventional bioassay. As a secondary monitoring method, real-time PASA (rtPASA) has been devised for a more precise determination of low resistance allele frequencies. To obtain more detailed information on resistance allele zygosity, as well as allele frequency, serial invasive signal amplification reaction (SISAR) has been developed as an individual genotyping method. Our approach of using three tiers of molecular resistance detection should facilitate large-scale routine resistance monitoring of permethrin resistance in head lice using field-collected samples.  相似文献

The essential oils extracted from Eucalyptus grandis, Eucalyptus camaldulensis, Eucalyptus tereticornis, and the hybrids E. grandisxE. camaldulensis, and E. grandisxE. tereticornis were analyzed by GC-MS, and evaluated for their fumigant and repellent effects on permethrin-resistant head lice. Fumigant activity of both hybrids was higher than that for pure species. E. grandisxE. tereticornis and E. grandisxE. camaldulensis showed KT50 values of 12.99 and 13.63min, respectively. E. grandis, E. camaldulensis, and E. tereticornis showed KT50 values of 25.57, 35.01, and 31.31, respectively. A simple regression analysis revealed a significant correlation between KT50 data and % of 1,8-cineole in these essential oils. Repellency varied from 47.80+/-16% to 80.69+/-6% for the five Eucalyptus essential oils tested. Interspecific hybridization improves the pediculicidal activity of Eucalyptus essential oils.  相似文献

Abstract. Resistance to human body lice, Pediculus humanus humanus L, induced by feeding on rabbits immunized with an extract of louse gut was studied. The mortality of lice fed on immunized rabbits was 73%, significantly higher than that of lice fed on control rabbits (52%) ( P < 0.01). The proportion of dead nymphs and female lice with ruptured guts was significantly higher in lice fed on immunized rabbits (P<0.01). The size of the bloodmeal was 35% greater in female lice fed on control rabbits than on immunized rabbits. Lice fed on immunized rabbits laid 40% less eggs than those fed on the controls, they also demonstrated a significant decrease in the number of eggs per female over time ( P < 0.01). 86% of the eggs laid by lice fed on immunized animals hatched, compared with 92% hatching of eggs laid by the lice fed on control animals (P< 0.01). With the exception of the first bloodmeal the percentage of hatched eggs which were laid between any two bloodmeals was significantly smaller (p<0.01) in the lice fed on immunized rabbits than in the control group. The first nymphal stage of lice fed on immunized rabbits took an average of 5.2 days to moult to the second stage, compared with 4 days for those fed on control rabbits.  相似文献

The efficacy of permethrin against Myobia musculi (Schrank) (Acari:Myobiidae) infestations on mice was evaluated using four different dose delivery methods. In all methods, an attempt was made to deliver 0.5 mg of active permethrin weekly to each mouse. Successful control was achieved with topical or environmental treatment with 0.25% permethrin dust and by dipping the mice into a 0.6 g/l permethrin emulsion. The use of an emulsion applied to the bedding produced the least satisfactory control. Untreated mice had an average infestation rate of 57.3% throughout the study. No significant differences were seen between treated and untreated groups in either body weight or histopathology of the liver, lung, or kidney.  相似文献

Abstract. A village-scale field trial of pyrethroid-impregnated mosquito nets was undertaken in The Gambia, West Africa, in the Mandinka village of Saruja (13o13'N, 14o55'W) during July-November 1989. Nearly all the villagers possessed and used their own bednets. Anopheles gambiae is the main vector of human malaria in the area.
An experimental wash-resistant formulation of permethrin was compared with standard emulsifiable concentrate (EC) formulations of permethrin and lambda-cyhalothrin, versus placebo-treated bednets. Target concentrations of pyrethroids on bednets were permethrin 500mg/m2 and lambda-cyhalothrin 25 mg/m2. The experimental design involved random allocation of a treatment to one net per family. Whereas 68% of people questioned said they washed their nets fortnightly, observations during the 16-week trial period showed that only 4/130 (3%) of nets involved in the trial had been washed as frequently as once per month.
Early morning searches for mosquitoes under bednets (1 day/week for 16 weeks) found significantly more mosquitoes (60% An. gambiae ) in placebo-treated nets than in pyrethroid-treated nets. The numbers found with each of the three pyrethroid treatments did not differ significantly from each other. Insecticidal efficacy of the treatments was tested by bioassays using wild-caught unfed mosquitoes exposed to netting for 3min. Linear regression analysis of bioassay mortality against number of times that a net had been washed by villagers showed that nets impregnated with the wash-resistant permethrin retained their insecticidal properties better than nets impregnated with lambda-cyhalothrin or with the standard permethrin formulation.  相似文献

Control of infestation by cosmopolitan lice (Pediculus humanus) is increasingly difficult due to the transmission of parasites resistant to pediculicides. However, since the targets for pediculicides have no been identified in human lice so far, their mechanisms of action remain largely unknown. The macrocyclic lactone ivermectin is active against a broad range of insects including human lice. Isoxazolines are a new chemical class exhibiting a strong insecticidal potential. They preferentially act on the γ-aminobutyric acid (GABA) receptor made of the resistant to dieldrin (RDL) subunit and, to a lesser extent on glutamate-gated chloride channels (GluCls) in some species. Here, we addressed the pediculicidal potential of isoxazolines and deciphered the molecular targets of ivermectin and the ectoparasiticide lotilaner in the human body louse species Pediculus humanus humanus. Using toxicity bioassays, we showed that fipronil, ivermectin and lotilaner are efficient pediculicides on adult lice. The RDL (Phh-RDL) and GluCl (Phh-GluCl) subunits were cloned and characterized by two-electrode voltage clamp electrophysiology in Xenopus laevis oocytes. Phh-RDL and Phh-GluCl formed functional homomeric receptors respectively gated by GABA and L-glutamate with EC50 values of 16.0 μM and 9.3 μM. Importantly, ivermectin displayed a super agonist action on Phh-GluCl, whereas Phh-RDL receptors were weakly affected. Reversally, lotilaner strongly inhibited the GABA-evoked currents in Phh-RDL with an IC50 value of 40.7 nM, whereas it had no effect on Phh-GluCl. We report here for the first time the insecticidal activity of isoxazolines on human ectoparasites and reveal the mode of action of ivermectin and lotilaner on GluCl and RDL channels from human lice. These results emphasize an expected extension of the use of the isoxazoline drug class as new pediculicidal agents to tackle resistant-louse infestations in humans.  相似文献

For their biological and ecological characteristics, ticks are vectors of the widest variety of pathogens causing tick-borne diseases (TBDs). Little information is available about the ways in which time spent by ticks to feed on hosts, transmission times and TBD prevention are related and it is exclusively limited to laboratory reports on adult stages. In particular, the time required by immature stages to transmit pathogens has not been determined for most TBDs. Considering their importance for animal and human health, effective control of immature ticks is advisable to reduce the damage ticks cause. Recently, the efficacy of a combination of imidacloprid 10%/permethrin 50% was compared with that of fipronil 10%/S-methoprene 9% against ticks when administered to naturally infected dogs. A semi-quantitative method was used to assess the methodological parameters to calculate drug efficacy on immature stages. On adult ticks, the efficacy of both products was high and overall very similar, whereas for the immature stages the combination of imidacloprid 10% and permethrin 50% had a higher efficacy than fipronil 10% and methoprene 9% throughout the observation period (statistically significant difference on day + 28 only). The semi-quantitative method proposed for the evaluation of immature stages can be considered a useful tool for calculating the efficacy of a drug in the field. Due the important role of immatures in the spread of TBDs, the immature tick load should be calculated to assess the efficacy of acaricidal products both under laboratory and field conditions.  相似文献

High resistance ability on insecticides among major mosquito vectors of diseases in Nigeria is of growing concern for severe control strategies. The objective of this study was to assess the susceptibility status of females Anopheles gambiae and Culex quinquefasciatus complexes mosquitoes to permethrin (21.5 μg/bottle-pyrethroids), propoxur(12.5 μg/bottle-carbamate) and malathion (50 μg/bottle organophosphate), in Niger State, North-Central, Nigeria. Anopheline and Culecine larvae were collected from the larval habitats of the studied sites (Bosso, Katcha, Lapai, and Shiroro) larvae and pupae were identified guided by standard keys and reared to adults in troughs. Insecticide susceptibility bioassays were performed according to the CDC bottle bioassay standard operating procedures on 3 days old, sugar-fed female Anopheles and Culex mosquitoes. Post-exposure mortality after 24hr and knockdown values for KDT50 were calculated. Knock-down at 1-hour insecticide exposure ranged (84–96 %) permethrin, (94–100 %) propoxur and (100 %) malathion for An. gambiae and (86–97 %) permethrin, (92–100 %) propoxur and (96–100 %) malathion for Cx. quinquefasciatus. Mortality, after 24hr post-exposure was 100 % in malathion, indicating the high effect of the insecticide. Tested samples were found potentially resistant to permethrin recorded against mosquitoes collected from all study sites, in two locations of the study sites to propoxur and one location site to malathion. All the tested mosquitoes were found to be potentially resistant to permethrin, however, mosquitoes tested in Katcha and Shiroro resist potentially to propoxur. Except, Culex quinqufasciatus from Lapai that partially resist malathion, all the tested mosquitoes were found to be susceptible to malathion, across the study sites.  相似文献

Lice-infected sea trout populations were monitored using fish traps in the Romsdalsfjord (Norway). The reliability and efficiency of this capture technique, which allows estimation of lice infestation rates without killing the fish, was evaluated through a mark–recapture study. A total of 2447 sea trout smolts were captured, tagged and released over a three-year period. There was a considerable variation in capture rates (range: 0.4–17.7 fish per day) and sea lice numbers (number of lice per fish: 2.8–30.3; number of lice per gram body weight: 0.02–0.69) among localities, sampling times and years. Recapture rates of tagged fish with traps, which were low (2% or 0.11 fish per day), showed that the risk for pseudoreplication was minor, in terms of counting lice on the same fish several times. Most of the tagged sea trout (90%) were recaptured within the first two months after release, and no significant variations in lice numbers were found between tagging and recapture. The lack of differences in lice levels between tagging and recapture during the first week after tagging indicated that the method most likely would not significantly underestimate the lice infestations due to loss of lice during handling. Therefore, our results confirm that the use of fish traps is a suitable method for estimation of lice numbers on wild salmonids.  相似文献

The effect of two known rates of repeated blood loss on rainbow trout Oncorhynchus mykiss swimming performance was measured and blood‐feeding rates of sea lice Lepeophtheirus salmonis were calculated to predict the point at which blood ingestion causes anaemia in infected fish. Known quantities of blood were sampled from rainbow trout over a 5 day period followed by critical swimming performance ( U crit) testing. A predictive equation was developed using masses of blood‐feeding sea lice and host blood loss calculated for increasing levels of sea lice infection. Blood loss of 8% total blood volume caused a decrease in U crit for rainbow trout. Total blood volume losses of 3·2% reduced erythrocyte stores, but did not affect fish swimming performance. The predictive feeding rate model suggests that 15–25% of the tissue consumed by sea lice is blood. This consumption of blood at higher sub‐lethal infection levels (≥0·5 sea lice g−1) may cause anaemia and a further decrease in swimming performance. Anaemia would compound the osmotic balance problems due to infection and potentially precipitate the morbidity seen at lethal sea lice levels (0·75–1·0 lice g−1).  相似文献

Red mite field populations from seven naturally infested Italian caged laying poultry farms were investigated for their susceptibility to acaricide formulations available on the market, containing amitraz, carbaryl and permethrin. A minimum of 3,000 mites of all stages were collected from each farm and were tested with five acaricide concentrations (5, 10, 20, 50, 100%) plus an untreated control (0%). Field red mite populations were found to be tolerant even with the highest concentrations with carbaryl and permethrin for six (86%) and three (42%) of the investigated farms, respectively (< 0.05). Furthermore, six (86%) of the investigated farms showed a red mite population susceptible to amitraz at any concentration. Out of the seven field populations tested with amitraz, one population is becoming less tolerant whereas another was the most tolerant to carbaryl and permethrin at any concentration. Data show that the lack of effectiveness of some acaricides is spreading in Europe and call for the adoption of alternative management strategies to avoid development of resistance.  相似文献
