The light response curves of the acceptor and donor side mechanisms of photoinhibition of Photosystem II were calculated, using Arabidopsis as a model organism. Acceptor-side photoinhibition was modelled as double reduction of QA, noting that non-photochemical quenching has the same effect on the quantum yield of QA double reduction in closed PSII centres as it has on the quantum yield of electron transport in open centres. The light response curve of acceptor-side photoinhibition in Arabidopsis shows very low efficiency under low intensity light and a relatively constant quantum yield above light saturation of photosynthesis. To calculate the light response curve of donor-side photoinhibition, we built a model describing the concentration of the oxidized primary donor P680+ during steady-state photosynthesis. The model is based on literature values of rate constants of electron transfer reactions of PSII, and it was fitted with fluorescence parameters measured in the steady state. The modelling analysis showed that the quantum yield of donor-side photoinhibition peaks under moderate light. The deviation of the acceptor and donor side mechanisms from the direct proportionality between photoinhibition and photon flux density suggests that these mechanisms cannot solely account for photoinhibition in vivo, but contribution of a reaction whose quantum yield is independent of light intensity is needed. Furthermore, a simple kinetic calculation suggests that the acceptor-side mechanism may not explain singlet oxygen production by photoinhibited leaves. The theoretical framework described here can be used to estimate the yields of different photoinhibition mechanisms under different wavelengths or light intensities.  相似文献

The photoinhibition of photosynthesis was investigated on intact attached leaves and isolated thylakoid membranes of Populus deltoides.Our studies demonstrate that in intact leaves photoinhibition takes place under high irradiance which is more pronounced at higher temperatures. No net loss of Dl and other proteins associated with photosystem II (PSII) were observed even after 64 % photoinhibition suggesting that the degradation of polypeptides associated with PSII is not the only key step responsible for photoinhibition as observed by other workers. Electron transport studies in isolated thylakoid membranes suggested water oxidation complex as one of the damaged site during high light exposure. The possible mechanisms of photoinhibition without net loss of D1 protein are discussed.  相似文献

Sap extracted from attached leaves of two-to three-week-old maize plants witt the aid of a roller device was almost devoid of bundle-sheath contamination as judged by the distribution of mesophyll and bundle-sheath markers. The extraction could be done very rapidly (less than 1 s) and the extract immediately quenched in HClO4 or reserved for enzyme assay. Comparison of the contents of metabolites in intact leaves and in the leaf extract allowed estimation of the distribution of metabolites between the bundle-sheath and the mesophyll compartments. Substantial amounts of metabolites such as malate and amino acids were present in the non-photosynthetic cells of the midrib. In the illuminated leaf, triose phosphate was predominantly located outside the bundle-sheath while the major part of the 3-phosphoglycerate was in the bundle sheath. The results indicate the existence of concentration gradients of triose phosphate and 3-phosphoglycerate in the leaf which are capable of maintaining carbon flow between the mesophyll and bundle-sheath cells during photosynthesis. There was no evidence for the existence of a gradient of pyruvate between the bundle-sheath and the mesophyll cells.  相似文献

The photoinhibition of photosynthesis at chilling temperatures was investigated in cold-acclimated and unhardened (acclimated to +18° C) spinach (Spinacia oleracea L.) leaves. In unhardened leaves, reversible photoinhibition caused by exposure to moderate light at +4° C was based on reduced activity of photosystem (PS) II. This is shown by determination of quantum yield and capacity of electron transport in thylakoids isolated subsequent to photoinhibition and recovery treatments. The activity of PSII declined to approximately the same extent as the quantum yield of photosynthesis of photoinhibited leaves whereas PSI activity was only marginally affected. Leaves from plants acclimated to cold either in the field or in a growth chamber (+1° C), were considerably less susceptible to the light treatment. Only relatively high light levels led to photoinhibition, characterized by quenching of variable chlorophyll a fluorescence (FV) and slight inhibition of PSII-driven electron transport. Fluorescence data obtained at 77 K indicated that the photoinhibition of cold-acclimated leaves (like that of the unhardened ones) was related to increased thermal energy dissipation. But in contrast to the unhardened leaves, 77 K fluorescence of cold-acclimated leaves did not reveal a relative increase of PSI excitation. High-light-treated, cold-acclimated leaves showed increased rates of dark respiration and a higher light compensation point. The photoinhibitory fluorescence quenching was fully reversible in low light levels both at +18° C and +4° C; the recovery was much faster than in unhardened leaves. Gas exchange and chlorophyll a fluorescence measurements of expanding and adult leaves of four plant species were compared under field conditions. The pioneer species (PS) tended to have thinner leaves with lower nitrogen content and higher stomatal density compared to forest species (FS). Expanding leaves featured lower photosynthetic pigment contents and gas exchange capacity than adult leaves consistent with an immature photosynthetic apparatus. At the time of maximum irradiance, sun-exposed leaves of both PS and FS showed alteration of initial, variable, and maximum fluorescence as well as their ratios indicating photoinhibition. However, leaves recovered to some extent at predawn, suggesting the activation of photoprotective mechanisms. Sun-exposed leaves had comparable responses to high irradiance.  相似文献

Tomato (C3-plants) and maize (C4-plants) were grown in a nutrient solution to which triacontanol was added twice a week. After about 4 weeks the triacontanol treatment caused a significant increase in the dry weight of the tomato plants. Leaf area and dry weight measurements of tomato leaves at different stages of development showed that the largest increase in growth was obtained when triacontanol treatment was initiated before bud formation. In maize, no effect of the triacontanol treatment on dry wieght was observed. Photosynthesis was inhibited by 27% in young leaves from triacontanol-treated tomato plants and 39% in the controls, when the oxygen concentration was raised from 2% to 21%. In maize no change in photosynthesis could be observed, neither after altered oxygen concentration nor after triacontanol treatment. The difference in the response of C3- and C4-plants to triacontanol indicates that it regulates processes related to photosynthesis.  相似文献

Recovery from photoinhibition of photosynthesis in intact Lemna gibba was studied in presence of the protein synthesis inhibitors chloramphenicol and cycloheximide. Exposure to an irradiance of 1000 mol m-2s-1 in N2 for 90 min induced 80% photoinhibition. The plants recovered photosynthesis when transfered to normal irradiances (210 mol m-2s-1) and air. Chloramphenicol added to the medium was taken up by the plant and reduced photosynthesis slightly. Recovery from photoinhibition was more inhibited than photosynthesis. Cycloheximide was also taken up by the plants and reduced synthesis of light harvesting chlorophyll protein: however, neither photosynthesis nor recovery were much affected. Synthesis of 32-kD chloroplast protein during recovery was inhibited by chloramphenicol, but not by cycloheximide. Synthesis of 32-kD protein was enhanced by 20–210 mol m-2s-1 light. The results support the hypothesis that synthesis of 32-kD protein is important for recovery of photosynthesis after photoinhibition.  相似文献

Effects of photoinhibition on photosynthesis in pea (Pisum sativum L.) leaves were investigated by studying the relationship between the severity of a photoinhibitory treatment (measured as Fv/Fm) and several photoacoustic and chlorophyll a fluorescence parameters. Because of the observed linear relationship between the decline of Fv/Fm and the potential oxygen evolution rate determined by the photoacoustic method, the parameter Fv/Fm was used as an indicator for the severity of photoinhibition. Our analysis revealed that part of the Photosystem II (PS II) reaction centers is inactive in oxygen evolution and is also less sensitive to photoinhibition. Correcting the parameter qP (fraction of open PS II reaction centers) for inactive PS II centers unveiled a strong increase of qP in severely inhibited pea leaves, indicating that the inactivated active centers do no longer contribute to qP and that photoinhibition has an all or none effect on PS II centers. Analysis of qE (energy quenching) demonstrated its initial increase possibly associated with dephosphorylation of LHC II. Analysis of qI (photoinhibition dependent quenching) showed that the half-time of recovery of qI increases steeply below an Fv/Fm of 0.65. This increase of the relaxation half-time corresponds with a decrease of the electron transport rate J and tentatively indicates that the supply of ATP, needed for the recovery, starts to decrease. The data indicate the necessity of correcting for inactive centers in order to make valuable conclusions about effects of photoinhibition on photosynthetic parameters.  相似文献

Pot trial in greenhouse was conducted using cumulic cinnamon soil from North China to study the effects of zinc deficiency on CO2 exchange, chlorophyll fluorescence, the intensity of lipid peroxidation, and the activity of superoxide dismutase (SOD) in leaves of maize seedlings. Zn deficiency resulted in a reduction of net photosynthetic rate and stomatal conductance to H2O. The maximum quantum efficiency of photosystem 2 (PS2) and the PS2 activity were depressed, while the pool size of the plastoquinone molecules was not affected by Zn deficiency. The content of super oxygen anion radical (O2 ·−) and the intensity of lipid peroxidation as assessed by malonyldialdehyde content in Zn-deficient leaves were higher than those in Zn-sufficient leaves. The activity of SOD increased with Zn application. The adverse influence of Zn-deficiency on the light stage of photosynthesis is probably one of possible reasons for the limitation of photosynthetic capacity in maize leaves.  相似文献

Intact leaves of kiwifruit (Actinidia deliciosa (A. Chev.) C.F. Liang et A.R. Ferguson) from plants grown in a range of controlled temperatures from 15/10 to 30/25°C were exposed to a photon flux density (PFD) of 1500 μmol·m−2·s−1 at leaf temperatures between 10 and 25°C. Photoinhibition and recovery were followed at the same temperatures and at a PFD of 20 μmol·m−2·s−1, by measuring chlorophyll fluorescence at 77 K and 692 nm, by measuring the photon yield of photosynthetic O2 evolution and light-saturated net photosynthetic CO2 uptake. The growth of plants at low temperatures resulted in chronic photoinhibition as evident from reduced fluorescence and photon yields. However, low-temperature-grown plants apparently had a higher capacity to dissipate excess excitation energy than leaves from plants grown at high temperatures. Induced photoinhibition, from exposure to a PFD above that during growth, was less severe in low-temperature-grown plants, particularly at high exposure temperatures. Net changes in the instantaneous fluorescence,F 0, indicated that little or no photoinhibition occurred when low-temperature-grown plants were exposed to high-light at high temperatures. In contrast, high-temperature-grown plants were highly susceptible to photoinhibitory damage at all exposure temperatures. These data indicate acclimation in photosynthesis and changes in the capacity to dissipate excess excitation energy occurred in kiwifruit leaves with changes in growth temperature. Both processes contributed to changes in susceptibility to photoinhibition at the different growth temperatures. However, growth temperature also affected the capacity for recovery, with leaves from plants grown at low temperatures having moderate rates of recovery at low temperatures compared with leaves from plants grown at high temperatures which had negligible recovery. This also contributed to the reduced susceptibility to photoinhibition in low-temperature-grown plants. However, extreme photoinhibition resulted in severe reductions in the efficiency and capacity for photosynthesis.  相似文献

Rates of photosynthesis vary with foliage age and typically decline from full-leaf expansion until senescence occurs. This age-related decline in photosynthesis is especially important in species that retain foliage for several years, yet it is not known whether the internal conductance to CO2 movement (g i) plays any role. More generally, g i has been measured in only a few conifers and has never been measured in leaves or needles older than 1 year. The effect of ageing on g i was investigated in Pinus pinaster, a species that retains needle for 4 or more years. Measurements were made in autumn when trees were not water limited and after leaf expansion was complete. Rates of net photosynthesis decreased with needle age, from 8 μmol m−2 s−1 in fully expanded current-year needles to 4.4 μmol m−2 s−1 in 3-year-old needles. The relative limitation due to internal conductance (0.24–0.35 out of 1) was in all cases larger than that due to stomatal conductance (0.13–0.19 out of 1). Internal conductance and stomatal conductance approximately scaled with rates of photosynthesis. Hence, there was no difference among year-classes in the relative limitations posed by internal and stomatal conductance or evidence that they cause the age-related decline in photosynthesis. There was little evidence that the age-related decline in photosynthesis was due to decreases in contents of N or Rubisco. The decrease in rates of photosynthesis from current-year to older needles was instead related to a twofold decrease in rates of photosynthesis per unit nitrogen and V cmax/Rubisco (i.e., in vivo specific activity).  相似文献

Photosynthetic induction times and photoinhibition in relation to simulated sunflecks (sudden increase of irradiance from 20 to 1,500 μmol m−2 s−1) were examined in leaves of co-occurring Fagus lucida (a deciduous tree) and Castanopsis lamontii (an evergreen tree) saplings grown either in a beech forest understory or in an adjacent open site during a late rainy season. Two hypotheses were tested: (1) understory leaves would display faster photosynthetic induction times and greater photoinhibition than open-grown leaves; and (2) evergreen species would have slower photosynthetic induction times and lighter photoinhibition than deciduous species. Times to reach 90% of maximal CO2 assimilation rate (t 90%A ) and stomatal conductance did not differ between species, but showed faster by 3–5 min in open-grown leaves than understory leaves due to higher initial stomatal conductance (g s initial) and induction state 1 min into simulated sunflecks (IS1min) in the former. Our analysis across the published data on photosynthetic induction of 48 broad-leaved woody species again revealed the negative correlations between t 90%A and either g s initial or IS1min, and the similarity of t 90%A and between evergreen and deciduous species. Measurements of maximum PSII photochemical efficiency (F v/F m) indicated that photoinhibition occurred in saplings in any of the growth habitats during sunfleck-induced photosynthetic induction. Despite no interspecific differences in the degree of photoinhibition, understory leaves of both species suffered heavier photoinhibition than open-grown leaves, as indicated by a stronger decrease of F v/F m in the former. Dynamic changes in the quantum yields of PSII photochemistry and ΔpH- and xanthophyll-regulated thermal dissipation and adjustments in the partitioning of electron flow between assimilative and non-assimilative processes were functional to resist photoinhibition. However, such photoinhibition, together with stomatal and biochemical limitations, would decrease carbon gain during simulated sunflecks, particularly in understory leaves.  相似文献

Phosphoenolpyruvate carboxylase (PEPC) was overproduced in the leaves of rice plants by introducing the intact maize C4-specific PEPC gene. Maize PEPC in transgenic rice leaves underwent activity regulation through protein phosphorylation in a manner similar to endogenous rice PEPC but contrary to that occurring in maize leaves, being downregulated in the light and upregulated in the dark. Compared with untransformed rice, the level of the substrate for PEPC (phosphoenolpyruvate) was slightly lower and the product (oxaloacetate) was slightly higher in transgenic rice, suggesting that maize PEPC was functioning even though it remained dephosphorylated and less active in the light. 14CO2 labeling experiments indicated that maize PEPC did not contribute significantly to the photosynthetic CO2 fixation of transgenic rice plants. Rather, it slightly lowered the CO2 assimilation rate. This effect was ascribable to the stimulation of respiration in the light, which was more marked at lower O2 concentrations. It was concluded that overproduction of PEPC does not directly affect photosynthesis significantly but it suppresses photosynthesis indirectly by stimulating respiration in the light. We also found that while the steady-state stomatal aperture remained unaffected over a wide range of humidity, the stomatal opening under non-steady-state conditions was destabilized in transgenic rice. The photosynthetic properties of a yellow lethal mutant, Oy/oy, and two yellow-green mutants of maize which are allelic (a homozygous recessive oy/oy and a heterozygous dominant Oy/+) were examined. Although Oy/oy had little or no chlorophyll or capacity for CO2 fixation compared to normal siblings, it had 28% as much ribulose-1,5-bisphosphate carboxylase oxygenase (Rubisco) activity, and from 40% to near normal activities of C4 cycle enzymes.Both yellow-green mutants had only half as much chlorophyll per leaf area as normal green seedlings in greenhouse-grown plants in winter and spring. However, the absorbance of light by the mutants was relatively high, as their transmittance was only 5 to 8% greater than normal leaves. In winter-grown greenhouse plants, the activities of Rubisco and several C4 cycle enzymes in the mutants were unaffected and similar to those of normal seedlings on a leaf area basis. After allowing for small differences in leaf absorbance, the light response curves for photosynthesis in the mutants were similar on a leaf area basis but much higher on a chlorophyll basis than those of the normal seedlings. In spring-grown greenhouse plants the enzyme activities and photosynthesis rates were about 30% lower per leaf area in the yellow-green mutant leaves compared to the wild type. The maximum carboxylation efficiency (measured under low CO2 and 1000 mol quanta m-2 s-1) in the mutants and normal leaves was similar on a Rubisco protein basis. 研究了6-BA和A5A对渗透胁迫时杨树幼苗叶片光合作用光抑制和活性氧代谢的影响.结果表明,渗透胁迫时杨树叶片净光合速率(Pn)和表观量子效率(AQY)降低,光合作用光抑制加剧,超氧化物歧化酶(SOD)活性升高,抗坏血酸过氧化物酶(APX)活性降低,O2产生加快,H2O2和膜脂过氧化产物丙二醛(MDA)含量升高.6-BA和A5A预处理使胁迫时叶片SOD和APx活性升高。O2生成减少。H2O22和MDA含量降低,同时缓解了光合作用的光抑制.相关分析表明,杨树叶片活性氧水平和MDA含量与Pn和AQY呈负相关.胁迫时杨树叶片活性氧的积累与光合作用光抑制有一定关系,6-BA和A5A对光抑制的缓解作用与其对活性氧清除系统的促进作用有关。  相似文献

Bean (Phaseolus vulgaris L.) plants were grown at two light periods of 8 and 13 h with a similar photon flux density (PFD) giving a daily photon receipt (DPR) of 17.9 and 38.2 mol · m–2, respectively. Shoot growth and leaf area development were followed at regular intervals and diurnal whole-plant photosynthesis measured. Single mature trifoliate leaves were exposed to photoinhibitory treatments at PFDs of 800 and 1400 mol · m–2 · s–1 and at temperatures of 12 and 20°C. Chlorophyll fluorescence and photon yields were measured at regular intervals throughout each treatment. Plants grown in 13 h had significantly greater leaf areas than those grown in 8 h. There were no differences in maximum rates of photosynthesis, photon yields and only minor but significant differences in Fv/Fm for plants in the two treatments, showing photosynthetic characteristics were dependent on PFD but not DPR. A significant decline in photosynthesis and Fv/Fm occurred over the 13-h but little change in photosynthesis for plants in the 8 h, indicating some feedback inhibition of photosynthesis was occurring. Plants grown in 8 h were consistently more susceptible to photoinhibition of photosynthesis at all treatments than 13-h plants. Nevertheless, photoinhibition was exacerbated by increases in PFD, and by decreases in temperature for leaves from both treatments. However, for plants from the 8-h day, exposing leaves to 12°C and 1400 mol · m–2 · s–1 caused photo-oxidation and severe bleaching but no visible damage on leaves from 13-h-grown plants. Closure of the photosystem II reaction-centre pool was partially correlated with increasing extents of photoinhibition but the relationship was similar for plants from both treatments. Gas exchange and fluorescence measurements of attached leaves of water stressed bean, sunflower and maize plants were carried out at two light intensities (250 mol quanta m-2s-1 and 850 mol quanta m-2s-1). Besides the restriction of transpiration and CO2 uptake, the dissipation of excess light energy was clearly reflected in the light and dark reactions of photosynthesis under stress conditions. Bean and maize plants preferentially use non-photochemical quenching for light energy dissipation. In sunflower plants, excess light energy gave rise to photochemical quenching. Autoradiography of leaves after photosynthesis in 14CO2 demonstrated the occurrence of leaf patchiness in sunflower and maize but not in bean. The contribution of CO2 recycling within the leaves to energy dissipation was investigated by studies in 2.5% oxygen to suppress photorespiration. We investigated the physiological effect of night chilling (CN) on potted seedlings of two tropical tree species, Calophyllum polyanthum and Linociera insignis, in Xishuangbanna, southwest China. Seedlings grown under 8, 25, and 50 % daylight for five months were moved to a 4–6 °C cold storage house for three consecutive nights, and returned to the original shaded sites during the day. CN resulted in strong suppression of photosynthesis and stomatal conductance for L. insignis, and reduced photorespiration rates, carboxylation efficiency, and maximum photochemical efficiency of photosystem 2 (PS2) at dawn and midday for both species. CN increased dawn and midday rates of non-photochemical quenching, and the contents of malondialdehyde and H2O2 for both species. CN also induced inactivation or destruction of PS2 reaction centres. The impacts of CN on tropical seedlings increased with the number of CN. Shading could significantly mitigate the adverse effects of CN for both species. After 3-d-recovery, gas exchange and fluorescence parameters for both species returned to pre-treatment levels in most cases. Thus CN induced mainly stomatal limitation of photosynthesis for L. insignis, and non-stomatal limitation for C. polyanthum. C. polyanthum was more susceptible to CN than L. insignis. Fog, which often occurs in Xishuangbanna, could be beneficial to chilling sensitive tropical seedlings in this area through alleviating photoinhibition or photodamage by reducing sunlight.  相似文献
