A new bioreactor (liquid-gas two-phase system) was devised for berberine-secreting Thalictrum minus cells immobilized in calcium alginate beads, which were alternately soaked in medium, and exposed to air. The highest yield of berberine (875 mg/l) was obtained by setting the cycle of medium supply and air supply for 30 seconds and 2 minutes, respectively, during a culture period of 30 days. Under such conditions of batch culture, the berberine productivity of immobilized cells was as high as that of freely suspended cells. Furthermore, the rate of berberine production by immobilized cells remained constant at a high value (50 mg/l/day) for a period of 60 days of semi-continuous culture achieved by renewal of medium at intervals of 10 days.  相似文献

Summary A semi-continuous process for the production of human interferon-c (IFN) from E. coli is described. Harvesting of bacteria, removal of cell debris and concentration were performed by tangentialflow microfiltration (cross-flow microfiltration). Purification was done in a single step by a monoclonal-Ab column. In the process, purified material, 3–9×108 units/mg protein was obtained with a 65% overall yield.  相似文献

Summary Endo-1,4-/B-glucanase from a thermophilic fungusMyceliophthora thermophila has been isolated and purified to homogeneity. The molecular weight of the enzyme was 52,000 with a pI of 4.7, pH-optimum 5.0–6.0, and specific activity 61 IU/mg (40°, pH 5.0); this activity is 2.4 times higher than that of the enzyme fromTrichoderma reesei. The new endoglucanase is very thermostable; its half-life at 65°C is 170 hours, which is 300 times higher than that of the enzyme fromT. reesei.  相似文献

Spring wheat (Triticum aestivum L.) zygotic embryos were successfully cryopreserved, without the addition of exogenous cryoprotectants, using only an abscisic acid (ABA) pretreatment. Optimum survival was obtained when embryos were cultured in vitro for 10 days on semisolid Murashige and Skoog (MS) nutrient medium supplemented with 0.5 mg/L (±) ABA prior to cryopreservation. The embryos resumed growth within three days when returned to MS medium devoid of ABA but containing 2mg/L 2,4-dichlorophenoxyacetic acid. The embryogenic calli produced from these embryos exhibited normal plant regeneration on auxin-free media. Changes in dw/fw ratio, as well as the esterified fatty acid and sucrose concentrations correlated positively with the development of tolerance to cryopreservation.NRCC Publication No. 33519  相似文献

Summary The effects of pH, temperature and substrate concentration on the cellulase (B-1,4-endoglucanase) activity ofA. candidus were studied. Maximum enzyme activities were obtained when the concentration of the substrate (CMC) was 6 mg per ml, at pH 4, and temperature 50 C. The enzyme retained 85% of its original activity under optimal conditions of pH and temperature after 36 hours of incubation. The Km constant of the reaction was calculated as 2.3 mg of CMC per ml and the energy of activation of the enzyme as 7.92 k cal per mole.  相似文献

Summary Saccharomyces anamensis having -galactosidase activity, has been immobilized in calcium alginate gel matrix that retained 78.6% enzyme activity to that of native cells. Optimum pH(7.0) was negligibly affected by immobilization. Km values for immobilized and native cells were 119 mM and 102 mM respectively. Protective agents like dithioerythritol, bovine serum albumin, enhance the enzyme activity when added prior to immobilization. Immobilized cells can be stored in refrigeration(4°C) for 42 days without a significant loss of enzyme activity.  相似文献

In vitro regeneration of plants from both cotyledon-, and leaf — derived calli of Lotononis bainesii Paker was studied under defined nutritional, hormonal and environmental conditions. Explants from both, cotyledons from seedilings of 4 days old and fully expanded leaves from mature plants, were cultured on MS medium containing 0.8% agar and supplemented with 0.01, 0.1, and 1 mg/1 concentrations of naphthaleneacetic acid (NAA) and 0.1, 1, and 3 mg/1 levels of benzyladenine (BA) in various combinations. Multiple shoot (on an average 4 shoots per callus) regeneration from primary callus occurred within 15 to 35 days of culture in most of the media tested. Although the best medium for shoot regeneration from cotyledon-derived callus contained NAA and BA at 1, and 0.1 mg/1 levels, respectively, maximal shoot regeneration from leaf-derived calli was achieved by using NAA and BA at 0.01 and 0.1 mg/1, respectively. Roots were induced to differentiate by transferring the regenerated shoots onto a medium lacking growth regulators.Supported by a research fellowship from Consejo Nacional de Investigaciones Científicas y Técnicas (República Argentina).  相似文献

Glycogen synthetase D from the 17,000 × g supernatant of a homogenate of human polymorphonuclear leukocytes has been purified to a specific activity of 7,4 units/mg protein in a single step, chromatography on Concanavalin A bound to agarose (Con A-Sepharose). The overall recovery of the enzyme was 66% and the entire procedure requires only 3–4 hours. After an in vitro D to I conversion, glycogen synthetase I was purified to a specific activity of 11,5 units/mg protein in a similar procedure.  相似文献

A total of 27 monkeys (M. Fascicularis) whose control cycle lengths ranged from 28 to 32 days were used in this study. All the treatments described below started either on day 17 or 18 of the cycle. Six monkeys received daily injections of 20 μg estradiol-17β (E2) for 5 consecutive days. Although a drop in blood progesterone (P) did occur due to this treatment, no shortening of the luteal phase of the cycle was recorded. Seven monkeys received daily injections of 15 mg PGF (prostaglandin-F) for 4 or 5 days. These monkeys also showed a drop in blood P levels; moreover 5 of these monkeys had vaginal bleeding for 2–3 days starting either on day 19 or 20 of the cycle. This bleeding did not appear to be a normal physiological menstrual flow, since all of the monkeys commenced menstrual flow at the expected time. Four monkeys received daily injections of 10 mg P for 3 days. These monkeys also had normal cycle lengths in spite of the treatment. Finally 9 monkeys received daily injections of 20 μg E2 for 3 days, and starting on the third day of E2 treatment these monkeys also received injections of 15 mg PGF for 4 or 5 days. Shortened cycle lengths were recorded in 8/9 monkeys in this group. Six monkeys had 22-day cycles, 2 monkeys had 24-day cycles and the remaining monkey had a cycle length of 26 days. Thus 8/9 monkeys had shortened luteal phases due to sequential treatment of E2 and PGF. The cycle lengths in all the treatment groups were normal subsequent to treatments. These results provide potentially useful information for further studies in the human as a method of contraception.  相似文献

Summary The tryptophan synthase genes,trpA andtrpB, from a moderate thermophile,Bacillus stearothermophilus IFO13737, were expressed efficiently inEscherichia coli. The recombinant tryptophan synthase amounted to 22% of the soluble cellular protein, and was purified to homogeneity by three steps. The enzyme is more thermostable thanE.coli tryptophan synthase, especially the subunit. The enzyme is also more resistant to sodium dodecylsulfate and methanol thanE.coli enzyme.  相似文献

Summary A polygalacturonase from culture filtrates of a strain ofRhizopus stolonifer was purified about 80 fold by ethanol precipitation, followed by ion exchange chromatography (CM-Sepharose 6B) and gel filtration (Sephadex G-100). The purified preparation was homogeneous when examined by PAGE. The enzyme is an endopolygalacturonase with an optimum catalytic activity at pH 5.0 and 45°C, and a molecular weight of 57,000±500 daltons. The activity was stimulated by Fe+++, Mg++, Co++, and inhibited by Mn++ and Zn++. The enzyme was stable in the pH range of 3.0 to 5.0. The purified enzyme was specific for nonmethoxylate polygalacturonic acid, with Km and Vmax values respectively of 0.19 mg/ml and 1.3 mol/g/min. In addition, this enzymatic preparation degraded pectic substances in organge peel.  相似文献

A protocol for regenerating plants from callus-derived protoplasts of Pelargonium x domesticum (rega l geranium cv. Melissa) has been developed. Protoplasts were isolated from leaf-derived callus tissue on MS medium supplemented with 3.0 mg/l naphthalene acetic acid, 2.0 mg/l 6- benzylaminopurine, and 3.0% sucrose. This callus yielded 2.7×105 protoplasts/gram of tissue after a 6 hr incubation in an enzyme solution consisting of 2.0% cellulysin, 0.5% macerase, and 0.5 M sucrose. Protoplasts were plated at 1×105 protoplasts/ml in a mixture (11 v/v) of KMP8/KP liquid medium layered on the same medium solidified with 0.6% agarose. Protoplast division was initiated within 2 days, and colonies of 15 to 50 cells developed 8 wk after plating. Undersulfation observed in the glycosaminoglycans synthesized by cultured skin fibroblasts from a Lowe's syndrome patient[Fukui, S. etal. (1981) J. Biol. Chem. 256, 10313–10318] was found to be caused by elevated degradation of 3′-phosphoadenosine 5′-phosphosulfate (PAPS). The enzyme involved in this degradation was then identified as an enzyme of nucleotide pyrophosphatase (EC 3.6.1.9) nature, cleaving the phosphosulfate linkage. The specific activities were 8 – 24 (mU/mg protein) in patients' fibroblasts, in contrast to 3 in normal and 5 – 14 in heterozygote cells. A possibility is discussed that the elevation of nucleotide pyrophosphatase activity is the primary genetic defect in Lowe's syndrome.  相似文献

A method is described for rapid multiplication from axillary buds of six Mentha species. Nodal segments from one-year old plants were grown on Murashige and Skoog medium (BMS), supplemented with BAP (1.0; 2.0 mg/l) and KIN (1.0 mg/l) and kept at 28 ± 1°C under fluorescent light for 30 days. After this period, several shoots (15–20 shoots per explant) with roots were produced which were placed in soil for further growth.  相似文献

Using a conventional sexual crossing technique, Nicotiana tabacum x N. acuminata was not produced. After the fertilized ovules were cultured for 20 days in a liquid Nitsch H medium, germination was observed. The roots grew rapidly but leaves did not. However, plantlets were produced in an H medium containing Benzyladenine or Kinetin (0.01–0.1 mg/l). The plantlets grew and flowered in a greenhouse. Summary CMCase was produced by 7 strains of Spicellum roseum in both liquid and wheat bran solid substrate cultures. No growth occurred above 35°C. Maximum enzyme production occurred at 30°C, whereas best enzyme activity occurred at pH 5.0 and 50°C. In liquid cultures of S. roseum, NRRL strains 13103, 13104, and 13106 produced activities of ca. 1.1, 1.5, and 1.5 mg glucose per hr/ml culture supernate at 1 week and 2.9, 1.5, and 2.1, respectively at 3 weeks compared to Trichoderma reesei NRRL 11236 (MCG77), which produced activities of 2.8 and 1.3 at 1 and 3 weeks.The mention of firm names or trade products does not imply that they are endorsed or recommended by the U.S. Department of Agriculture over other firms or similar products not mentioned.  相似文献

Plants were regenerated from leaf explants of Centrosema brasilianum cultured in vitro. Callus and buds were produced on Murashige and Skoog medium (MS), 0.8% agar, 0.1 mg/l NAA and 1 mg/l BAP. Regeneration of multiple shoots was achieved by transferring callus onto fresh medium containing 0.01 and 1 mg/l of NAA and BAP, respectively. Shoots formed roots upon transfer to MS with 0.01 mg/l NAA. Plantlets were succesfully transferred to soil. Leaf-derived calli of Centrosema arenarium, C. macrocarpum, C. pascuorum, C. pubescens, and C. virginianum did not produce shoots when cultured in vitro.  相似文献
