Mononuclear cells from human peripheral blood are able to destroy alloantibody-coated target cells of a human lymphoblastoid line. The effector cell is a nonadherent, non-phagocytic, medium sized lymphoid cell. The cell is not detectable in early human fetuses. Its activity is destroyed by x-ray treatment in vitro but is not inhibited by anti-immunoglobulin sera. Its efficiency in antibody-dependent cell-mediated cytotoxicity is rapidly lost in culture and this loss is prevented by stimulation with allogeneic cells.  相似文献

Ozone is a powerful oxidant, reactive to biomolecules. In aqueous solution it decomposes to give hydrogen peroxide, superoxide and hydroxy radicals which can take part in secondary reactions. Ozone is a disinfectant that inactivates both viruses and bacteria. Although other reactions are primarily responsible for the inactivation, cellular DNA is also damaged. Ozone is genotoxic to microorganisms, plants and cell cultures in vitro. The results from in vivo cytogenetic studies with laboratory animals after inhalation exposure are contradictory. Chromosome aberrations in lymphocytes, but not SCEs, have been demonstrated in Chinese hamsters but not in mice. Chromatid deletions were induced in pulmonary macrophages in rats. No cytogenetic effects have been reported for bone marrow cells or spermatocytes. The few experimental and epidemiological studies with human subjects do not allow a conclusion on the cytogenetic effects of ozone in lymphocytes in humans. No life-long cancer studies have been performed with ozone. However, after 4 and 6 months of inhalation exposure, lung adenomas were induced in strain A/J mice, but not in Swiss-Webster mice.  相似文献

1. Secretion from the mucous sublingual gland of the mouse has been investigated and compared with the serous parotid gland. The influence of acetylcholine, noradrenalin and adrenalin on the secretion of glycoproteins (e.g. mucins) and proteins (e.g. amylase) from these glands in vitro, and the involvement of cyclic AMP and Ca2+ has been studied. 2. Secretion from the parotid gland could be stimulated by both acetylcholine and the catecholamines. It appears that cyclic AMP plays an important role in the adrenergic secretory process, but not in the cholinergic-induced secretion. In the latter case, exogenous Ca2+ strongly increased the secretion. 3. Mucin secretion from the sublingual gland could be affected by acetylcholine in the presence of exogenous Ca2+. Noradrenalin and adrenalin induced only a slow mucin secretion and, for this secretory process, exogenous Ca2+ is also required. Though cyclic AMP is present in the sublingual gland, no influence on its level could be detected in this gland after stimulation of the adrenergic beta-receptor, whereas, in contrast to the parotid gland, dibutyryl cyclic AMP induced only a slow secretion. Because it was observed that the sublingual gland of the mouse is not innervated sympathetically, it seems reasonable to suppose that the catecholamines stimulate the mucin secretion from this gland via hormonal receptors and not via the adrenergic beta-receptor. 4. The protein secretion from the sublingual gland could be stimulated by both acetylcholine and the catecholamines. An involvement of cyclic AMP in this process was not observed. Addition of exogenous Ca2+ is less important, as was found for the mucin secretion. So it has been concluded that protein and mucin secretion from the sublingual gland are regulated via different pathways.  相似文献

A Kamuela, Hawaii, population of Drosophila mercatorum was surveyed for enzyme variability. The mean heterozygosity and the proportion of polymorphic loci were estimated as 0.1255 and 0.37, respectively. Neither deviates more than one standard error from their respective means for 43 Drosophila species (Nevo 1978). Heterozygosity was distributed across enzyme categories in much the same manner as observed in other species (Gillespie and Kojima 1968; Johnson 1974), and enzymes associated with glycolysis were about as variable as other enzymes of central metabolism.--The levels of heterozygosity and polymorphism in this population do not seem to have been affected by a low-level capacity for parthenogenesis. The observed parthenogenetic reproduction is not strongly associated with particular allelic variants among viable parthenogenetic adults; however, the capacity to establish a self-sustaining parthenogenetic clone is strongly associated with the phenotype with the most frequent allele at every locus studied. We interpret these results to mean that isozyme variants do not strongly influence viability under total homozygosity (the genetic condition imposed by parthenogenesis), but they do have an impact upon the reproductive biology of parthenogenetic adults.  相似文献

When nuclear families are sampled by an ascertainment procedure whose properties are not known, biased estimates of genetic parameters will arise if an incorrect specification of the ascertainment procedure is made. Elsewhere we have put forward a resolution of this problem by introducing an ascertainment-assumption-free (AAF) method, for nuclear family data, which gives asymptotically unbiased estimators no matter what the true nature of the ascertainment process. In the present paper we extend this method to cover pedigree data. Problems that arise with pedigrees but not with families--for example, the question of which families in a pedigree are "ascertainable"--are also considered. Comparisons of numerical results for pedigrees and nuclear families are also made.  相似文献

The hisT gene, one of six genes in which mutation causes derepression of the histidine operon in Salmonella typhimurium, is shown to code for a protein that is not essential for the growth of the bacteria. This is indicated by the characterization of particular classes of mutations in the hisT gene: amber mutations, frame-shift mutations, and temperature-sensitive mutations that affect repression but not growth. In this study we compared steroid sulfatase levels in XO, XX, and XY mice and carried out a clonal analysis in fibroblast cell cultures from mice heterozygous for the steroid sulfatase deficiency gene and heterozygous at the X-linked electrophoretic phosphoglycerate kinase locus. The combined results indicate that the murine steroid sulfatase locus is not dosage compensated and is not subject to X-inactivation. With respect to X-inactivation, it behaves in a somewhat different way from the closely linked sex-reversed gene and the human steroid sulfatase locus.  相似文献

The AP1 protein, a unique aspermatogenic protein localized in the sperm acrosome, exists as a single polypeptide chain of 136 amino acids, as shown by a single band on gel electrophoresis in sodium dodecyl sulfate and the recovery of the expected 21 to 22 tryptic peptides on peptide mapping. The AP1 protein appears to exist in a compact, highly stable conformation, as shown by its resistance to trypsin hydrolysis. Its aspermatogenic acitivity is not affected by trypsin treatment, by heating at 99 degrees C for 1 h, by 8 M urea, or by acid conditions. After reduction and alkylation, however, the molecule appears to open up, since it becomes hydrolyzable by trypsin and migrates more slowly on gel electrophoresis at pH 2.7 and 8.6. After alkylation, the AP1 protein still migrates as a single band at pH 2.7. The AP1 protein shows microheterogeneity near its isolectric point at pH 8.6; each of five bands shows the same amino acid analysis. Aggregation was not observed following treatment with dimethylsuberimidate. The molecular weight of 15 000, obtained from gel electrophoresis consists of 136 amino acids with a relatively high content of proline, half cystine, glycine, histidine and tryptophan. No galactose, mannose, fucose, glucose, or hexosamines were found; the AP1 protein is thus not a glycoprotein.  相似文献

It is shown by titrating a suspension of rat liver mitochondria with either ADP or an uncoupler that a specific rate of respiration may not have a unique associated value of the protonmotive force. Alternatively, a specific protonmotive force may not be associated with a unique rate of respiration. It seems that the rate of respiration and the protonmotive force are more sensitive to the agents used for the titrations than to each other. Such observations are not easily explained by the chemiosmotic hypothesis. It is, however, possible provided that the proton conductivities, i.e. the rates of dissipation of the protonmotive force, are considered to be different for each of the agents used to titrate the rate of respiration at the same protonmotive force, or vice versa.  相似文献

A mathematical model for an n-species food chain in which the growth and predation functions are not specified is presented and analysed. The stability of such systems under arbitrarily great enrichment is considered, and it is concluded that if growth is limited by nutrient saturation a stable system is in danger of destabilization, but that this is less likely if growth is limited by some other factor such as a shortage of some other metabolite or of space. If the system does not destabilize, all population oscillations should ultimately disappear.  相似文献

The histomorphology of the retrocerebral endocrine organs of the larva of the lemon-butterfly, P. demoleus has been described employing a conventional neurosecretory staining technique. The larval organs lie a little away from the brain and oesophagus and, therefore, are not sub-aortic in position unlike most other insects and the adult of this species itself. There are two long NCCs which innervate other target organs in addition to the CC. The recurrent nerve of the stomatogastric nervous system retains its normal connections with the CC even in absence of the hypocerebral ganglion. Histological evidence suggests that the NSM inside the CC remains intraaxonal without being possibly unloaded in the substance of these organs. Axonal transport of NSM to target organs is also in evidence and though NSM is seen in the NCA, it could not be detected inside the CA.  相似文献

Fifty-five reagents were studied as to their ability to replace iodine in the Gram stain. None gave results as good as iodine. Eight gave usable Gram preparations, and forty-seven gave negative results. Omission of the counterstain resulted in increasing to thirty-three the number of reagents giving differentiation, but this, was not considered a true Gram differentiation. Many oxidizing agents were shown not to be substitutes for iodine; therefore the function of iodine must be more than to serve as an oxidizing agent. Many reagents which formed precipitates with the dye could not replace iodine; therefore factors other than precipitate formation must be involved. However, all agents which were good substitutes for iodine were both good oxidizing and dye precipitating agents. Experiments involving the study of cell membrane permeability showed that Gram-positive cells were less permeable to iodine in alcoholic solution than Gram-negative cells. This difference could not be demonstrated for iodine in aqueous solution. It was concluded that iodine served to form a dye-iodine precipitate (or complex) in the cell. Since Gram-positive cells were less permeable to iodine in alcohol than Gram-negative cells, this resulted in a slower dissolving out of this complex from Gram-positive cells during de-colorization and hence a slower decolorization time. The relative solubilities of dye precipitates in alcohol and in aqueous safranin solution were also indicated as an important factor influencing decolorization. Dyes which formed highly soluble precipitates with iodine could not be used in the Gram stain. It is not proposed that the mechanism of the Gram stain is entirely one of membrane permeability; chemical factors are undoubtedly important and will be discussed in a later paper. However, it is proposed that the chemical and physical factors are closely interrelated in the Gram stain mechanism.  相似文献

An analysis is made of the kin-selection/group-selection debate on the issue of the biological basis of morals. The kin-selection view sees altruism, and morals in the case of humans, as resting solely on genetic factors; in fact, on this view, evolution itself is to take place only through genetic change -- a position which cannot be reconciled with our knowledge that species evolution involves karyotypic change. Morality is thought to stem from a particular gene which at one time was completely absent from the human population but which later entered and spread through it. On the generally accepted conception of kin selection, this 'altruistic' gene is to be responsible not only for apparently altruistic behaviour towards near relatives, but also for parental care. Thus this view, among other things, has the absurd consequence that there was a period directly after humans first came into existence during which we did not care for our young. In contrast, the group-selection view, as developed here, sees morality as stemming from the species' karyotype, and to be passed on karyotypically from species to species through evolution. As suggested by Darwin, morality is thus derived from our social instincts, and is fundamentally directed to members of one's tribe or community, not to members of one's family. In keeping with this perspective, it is suggested here that the biological basis of morals does not concern the continuation of each individual's gene line, but the continuing existence of the individual's community, and thus, indirectly, the survival of the human species, through supporting the continuing instantiation of its karyotype.  相似文献

Utilization of n-heptane by a Pseudomonad was studied in pilot-size butch cultures. Optimal pH and temperature were determined by a factorial design and a medium based upon mineral uptake rates was formulated. High cell yields were obtained by volatilizing heptane in the incoming air and thereby achieving good hydrocarbon dispersion. Hydrocarbon carried by effluent gases was recovered and recycled. In cultures where pH is not controlled, decrease in the electrolytic conductivity of the medium was found to be indicative of viable cells and was used in monitoring bacterial propagation. If not checked, increase in salinity in pH controlled cultures was found to affect cell production negatively. Viscosity changes were not very significant. Heptane to aqueous medium ratio was found to affect oxygen supply to the system due to higher dissolved oxygen concentrations associated with hydrocarbons.  相似文献

In order to study the membrane function of tolerant B antigen-binding cells, tolerance to the trinitrophenyl (TNP) determinant was induced in mice by injecting the reactive form of the hapten, trinitrobenzene sulfonic acid (TNBS). By appropriate transfer experiments, Fidler and Golub (J. Immunol.112, 1891, 1974) had previously shown that this form of tolerance is a B-cell property, induced and expressed in the absence of T cells. Hapten inhibition demonstrated the TNP-specificity of receptors on TNP-donkey erythrocyte(TNP-D)-binding cells in tolerant and nontolerant mice. About 88% of these cells were B cells by immunofluorescence, and the remainder were T cells. In the tolerant mice, challenge with TNP-sheep erythrocytes failed to expand the TNP-binding population, but sheep erythrocyte binders and anti-sheep plaque-forming cells expanded normally. Despite little or no change in TNP-binding cell numbers after tolerance induction, the TNP-binding cells of tolerant animals could not cap their receptors, in contrast to the sheep erythrocyte-binding cells from the same animals which capped normally. Although there is no anti-TNP plaque-forming cell response when tolerogen and immunogen are given simultaneously, capping failure is not evident until 2–4 days after tolerogen exposure. By Day 7, substantial recovery of immune responsiveness had occurred, yet even 12 months after a single dose of tolerogen there was no restoration of capping. Thus despite the association of both capping failure and unresponsiveness with tolerogen exposure, these lymphocyte functional defects appeared not to be causally related.  相似文献

In isolated perfused rat hearts, epidermal growth factor (EGF; 15 nM) increased cellular cyclic AMP (cAMP) content by 9.5-fold. In rat cardiac membranes, EGF also stimulated adenylate cyclase activity in a dose-dependent manner, with maximal stimulation (35% above control) being observed at 10 nM-EGF. Half-maximal stimulation of adenylate cyclase was observed at 40 pM-EGF. Although the beta-adrenergic-receptor antagonist propranolol markedly attenuated the isoprenaline-mediated increase in cAMP content of perfused hearts and stimulation of adenylate cyclase activity, it did not alter the ability of EGF to elevate tissue cAMP content and stimulate adenylate cyclase. The involvement of a guanine-nucleotide-binding protein (G-protein) in the activation of adenylate cyclase by EGF was indicated by the following evidence. First, the EGF-mediated stimulation of adenylate cyclase required the presence of the non-hydrolysable GTP analogue, guanyl-5'-yl-imidodiphosphate (p[NH]ppG). Maximal stimulation was observed in the presence of 10 microM-p[NH]ppG. Secondly, in the presence of 10 microM-p[NH]ppG, the stable GDP analogue guanosine 5'-[beta-thio]diphosphate at a concentration of 10 microM blocked the stimulation of the adenylate cyclase by 1 nM- and 10 nM-EGF. Third, NaF + AlCl3-stimulated adenylate cyclase activity was not altered by EGF. The ability of EGF to stimulate adenylate cyclase was not affected by pertussis-toxin treatment of cardiac membranes. However, in cholera-toxin-treated cardiac membranes, when the adenylate cyclase activity was stimulated by 2-fold, EGF was ineffective. Finally, PMA by itself did not alter the activity of cardiac adenylate cyclase, but abolished the EGF-mediated stimulation of this enzyme activity. The experimental evidence in the present paper demonstrates, for the first time, that EGF stimulates adenylate cyclase in rat cardiac membranes through a stimulatory GTP-binding regulatory protein, and this effect is manifested in elevated cellular cAMP levels in perfused hearts exposed to EGF.  相似文献

Numerous specific-locus experiments designed to test the mutagenic effect of external radiation have yielded, in over 3,600,000 animals observed, altogether 119 presumed mutations involving the c locus. Of these, 55 were viable and albino (cav), 13 were viable and of various intermediate pigment types (cxv), four were subvital (cas and cxs), seven were neonatally lethal albinos (cal), 28 prenatally lethal albinos (cal); 12 died untested. All of the prenatally lethal and at least one of the neonatally lethal c-locus mutations (cal classes) are probably deficiencies that we have analyzed extensively in other experiments. Since absence of the locus mimics albino in phenotype, the intermediates (cxv and cxs groups) probably resulted from intragenic changes. The class of viable albino mutants (cav) might include, in addition to intragenic changes, some extremely small deficiencies. --The effects on viability of c-locus lethals (cal's) in heterozygous condition are not drastic enough to be perceived in stocks of mixed genetic background except in the case of the two longest known deficiencies and a few others. --Analysis of the relation between radiation treatment and type of c-locus mutants obtained shows that the relative frequency of viable mutations, for each germ-cell type, is greater for low-LET than for neutron irradiation; however, the difference for any individual cell type is not significant. The majority (66.7%) of mutations derived from X- or gamma-ray irradiated spermatogonia are viable, and the proportion of "intermediates" among these viables is similar to that among presumed spontaneous c-locus mutations. No significant dose-rate effect on the proportion of lethals could be demonstrated within the set of mutants induced by low-LET irradiation of spermatogonia. Although sets from other germ-cell stages are too small for statistical tests, the results for oocytes are similar, as far as they go. Furthermore, most of the c-locus mutations induced in spermatogonia, even by high-dose-rate X-ray or gamma irradiation, are of a type most likely to result from single-tract events (62% cxv, cxs, and cav; plus 16% presumed deficiencies not involving the closest marker). These results support the view that most of the reduction in mutation frequency at low dose rates is not due to a change in relative proportion of two-track and one-track ionizing events.  相似文献
