丝氨酸内肽酶在黄瓜叶片衰老中的作用   总被引:3,自引:0,他引:3  
采用丝氨酸内肽酶抑制剂和植物生长调节剂处理离体黄瓜叶片,研究了黄瓜叶片暗诱导衰老过程中丝氨酸内肽酶的作用。结果表明,6-BA50μmol/L与丝氨酸内肽酶抑制剂AEBSF能抑制叶片内肽酶活性的升高,延缓蛋白质降解,而ABA50μmol/L则促进了内肽酶活性的升高:其作用效果与AEBSF相反。活性电泳结果显示,黄瓜叶片中检测到6条内肽酶同工酶,其中4条(CEP2、3、4、6)为丝氨酸类型内肽酶,而ABA使丝氨酸内肽酶CEP2、3、4、6的活性明显增强,提示了丝氨酸类型内肽酶在黄瓜叶片衰老过程中具有重要作用。  相似文献

The characterization of senescence-associated endopeptidase (EP) isoenzymes in wheat (Triticum aestivum L. var. Yangmai 158) leaves during dark-induced senescence was performed. It was found that there was much higher endoproteolytic activity in dark-induced wheat leaves than in control. Six endopeptidase isoenzymes (EP1-EP6) were identified by natural gradient杙olyacrylamide gel electrophoresis (PAGE) co-polymerized gelatin in the gel, five of which (EP1, EP2, EP4, EP5 and EP6) were only detected in senescing leaves. Treatment with 6-benzylaminopurine (6-BA) delayed the expression of these EP isoenzymes and abscisic acid (ABA) accelerated it. The activity of EP3 could be detected at a wider range of pH and temperature levels while EP4, EP5 and EP 6 could be only detected at pH 4-5 and 30 45 ℃, EP1 and EP2 at pH 3 5 and 30 ℃ 45 ℃. All of the EP isoenzymes showed high thermal stability, especially EP3, EP5 and EP6 which still had activities even by incubation at 55 ℃ for 1 h. By using different class-specific inhibitors, EP1 and EP2 were characterized as metal-dependent cysteine-proteases, EP4 as a serine-protease.  相似文献

以谷氨酸棒杆菌(Corynebacterium glutamicum) SYPS-062基因组DNA为模板,扩增得到L-丝氨酸脱水酶(L-SerDH)的编码基因sdaA。将其克隆到表达载体pET-28a(+),并在E.coli BL21(DE3)中诱导表达,对纯化的L-SerDH进行了酶活测定,并与来自C.glutamicum ATCC13032的重组L-SerDH进行了比较,结果显示,两种不同菌株来源的重组L-SerDH降解L-丝氨酸的酶比活力差异并不显著。在此基础上敲除菌株SYPS-062 的sdaA基因,探讨该基因对C.glutamicum SYPS-062生长及产酸的影响。通过构建自杀型重组质粒pK18mobsacB-△sdaA,电击转入C.glutamicum SYPS-062中,以同源重组的方式获得了sdaA基因缺失突变株,并用PCR方法对突变株C.glutamicum SYPS-062△sdaA进行了验证。与出发菌株相比,突变菌株生长缓慢,单位菌体L-丝氨酸的产量(YP/X)提高了15.13%。  相似文献

以采自海南陵水的紫萍(Spirodela polyrhiza DW2501-4)为材料,观察紫萍的营养繁殖过程。紫萍DW2501-4培养在0.5倍Hoagland′s液体培养基中,先不断长出新的叶状体,随着营养物质的减少,长出休眠体进入休眠状态。休眠体经过4℃处理7 d后,可在含1%蔗糖的Hoagland′s固体培养基中重新萌发。为了进一步观察叶状体、类休眠体和休眠体的组织结构差异,以及休眠体萌发过程中组织结构的变化,制作了石蜡切片。观察结果显示:紫萍叶状体有数条叶脉,细胞中含少量的淀粉粒,表皮层以下有分层的气室,通常分上下两层,上层气室比下层气室小。类休眠体也有数条叶脉和少量气室,细胞中有较多淀粉粒。休眠体的细胞差异不明显,几乎没有气室,细胞内有大量的淀粉粒,部分细胞含有单宁;随着休眠体萌发,细胞中淀粉粒不断变小,同时分生组织分化出根和新的叶状体。  相似文献

从广西隆安县沼气池里的残渣中筛选到一株能以甲醇为唯一碳源生长的MB200菌株。根据常规形态特征、生理生化性状及16S rDNA基因序列分析将其鉴定为甲基杆菌属(M ethylobacteriumsp.)。其最佳生长条件为:温度32℃、pH值8.0、甲醇体积分数1.25%。建立了MB200生成L-丝氨酸的静息细胞培养系统。确定静息细胞培养的条件为:甘氨酸质量浓度为10 g.L-1,甲醇50 g.L-1,菌体质量浓度为30 g.L-1,pH8.9,于摇床250 r.m in-1,32℃静息培养48 h,L-丝氨酸产量为7.2 g.L-1。  相似文献

以甲基营养型假单胞菌J-12为出发菌株,经硫酸二乙酯(DES)和亚硝基胍(NTG)诱变处理,D-丝氨酸结构类似物平板和高浓度甘氨酸平板定向筛选,获得1株三-丝氨酸高产菌N-13,其发酵液中L-丝氨酸产量较出发菌株提高97.9%。在含有20g/L甘氨酸和7mL/L甲醇的培养基中,L-丝氨酸积累可达4.81g/L。  相似文献

sga基因是大多数Methylobacterium sp.中丝氨酸循环途径里的一个关键酶基因,与单碳化合物的同化及二碳化合物的代谢有着密切的关系. 根据已报道的M.extorquens AM1的sga基因序列,合成寡核苷酸引物,用本实验室保存的甲醇利用菌M.sp. MB200的染色体DNA为模板,通过PCR方法扩增出sga基因,测序分析表明该基因与M.extorquens AM1的sga基因在核苷酸水平上有93%的同源性,氨基酸水平上具有85%的同源性.利用自杀质粒pK18mob构建含有sga基因部分片段的重组质粒,通过三亲本结合后导入原始菌株MB200中,经过进一步同源单交换, sga基因被pK18mob插入突变,利用PCR和Southern杂交进行验证, 选择发生正向突变的突变株MB200sTB用于进一步研究分析.分析结果表明突变菌株的丝氨酸乙醛酸氨基转移酶(SGAT)的酶活性消失,同时失去了在以甲醇为唯一碳源的培养基上生长的能力,不能再合成L-丝氨酸, 但仍能以二碳化合物和多碳化合物进行生长.  相似文献

以一株黄假单胞属(Pseudomonas flava)菌株A3为出发菌株,经过紫外(UV)诱变和硫酸二乙酯(DES)逐级诱变处理和选育,选育出一株能够以甲醇为唯一碳源的兼性甲基营养型菌JW-01(Mth~R、Gly~R)。在含甘氨酸30g/L、甲醇1%的发酵培养基中发酵3d后L-丝氨酸产量为6.2g/L,较出发菌株提高了67.6%。该菌具有较好的传代稳定性。  相似文献

将无根和囊的紫萍(Spirodela polyrrhiza)P143品系半叶状体分别放在含有和不含6-苄基嘌呤的培养液上于长日照条件下培养。在不含6-BA的培养液上培养4d以后,半叶状体开始失绿,光合能力降低;第12天时,半叶状体已接近死亡。但是,培养在含有6-BA的培养液上的半叶状体干重、叶绿素和可溶性蛋白质含量、希尔反应活力、核酮糖-1,5-二磷酸羧化酶/加氧酶的小亚基和其编码基因rbcS mRNA水平显著增加。这些6-BA处理过的半叶状体平均寿命可达80d,几乎等同于完整植物体上叶状体的寿命,因此提出细胞分裂素对于紫萍叶状体生物学功能的维持是必需的。  相似文献

不同浓度铜对紫背萍和青萍色素含量及抗氧化酶系统的影响   总被引:11,自引:0,他引:11  
通过水培试验研究了不同浓度的重金属Cu对紫背萍(Spirodela polyrrhiza)和青萍(Lemna minor)的色素含量和抗氧化酶系统的影响.结果表明,低浓度Cu(0.056 mg·L-1)的处理下,紫背萍和青萍的叶绿素a、叶绿素b和叶绿素(a+b)的含量均出现不同程度的增加,分别高出其对照11%、46%、22%和8%、15%、11%,而在高浓度Cu(0.18~5.60 mg·L-1)的处理下,上述色素含量均显著下降,平均下降幅度分别达63%、62%、65%和46%、44%、45%.紫背萍体内丙二醛(MDA)含量为青萍的2.57倍.两种浮萍的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和过氧化物酶(POD)的活性均随Cu浓度的增加而呈先升后降的趋势,紫背萍体内3种酶活性在Cu浓度仅为0.18 mg·L-1时即达峰值,随后则大幅下降,而青萍体内3种酶活性却在Cu浓度分别升高到0.56、1.0和1.0 mg·L-1时才达到峰值.可见,在Cu胁迫下,紫背萍受Cu毒害较青萍深,且其体内抗氧化酶系统也较青萍敏感.  相似文献

Abstract The developmental process leading to the formation of the abscisic acid (ABA) induced turion of Spirodela polyrrhiza was accompanied by a repression of nucleic acid and protein synthesis. DNA synthesis in the developing lurion (induced by 10−4mol m−3 ABA) was inhibited within 3h of ABA addition, followed by a repression of protein synthesis after 24 h, while RNA synthesis was not inhibited until 3 d. The inhibitory effect of ABA on protein synthesis was found to be selective and the synthesis of several novel proteins appeared to be induced. These effects were specific to ABA-sensitive tissue. The relationship between the changes in the protein and mRNA profiles during the development of the turion was investigated. The rapid general inhibition of protein synthesis at early stages of lurion formation could not be accounted for by the level of translatable mRNA, indicating an effect of ABA at the translational level. The specific alteration to the pattern of in vivo labelled proteins could have resulted, however, from control of the level of specific mRNAs for those particular proteins. Only after 3 d in ABA, when the developing primordium is committed to the turion developmental pathway, is there a total inhibition in the production of mRNA leading to the shutdown of all primary processes and the onset of the irreversible events leading to the dormant state.  相似文献

Phytoremediation potential of the aquatic plant Spirodela polyrrhiza was examined for direct blue 129 (DB129) azo dye. The dye removal efficiency was optimized under the variable conditions of the operational parameters including removal time, initial dye concentration, pH, temperature and amount of plant. The study reflected the significantly enhanced dye removal efficiency of S. polyrrhiza by increasing the temperature, initial dye concentration and amount of plant. Intriguingly, artificial neural network (ANN) predicted the removal time as the most dominant parameter on DB129 removal efficiency. Furthermore, the effect of dye treatment on some physiologic indices of S. polyrrhiza including growth rate, photosynthetic pigments content, lipid peroxidation and antioxidant enzymes were studied. The results revealed a reduction in photosynthetic pigments content and in multiplication of fronds after exposure to dye solution. In contrast, malondialdehyde content as well as catalase (CAT) and peroxidase (POD) activities significantly increased that was probably due to the ability of plant to overcome oxidative stress. As a result of DB129 biodegradation, a number of intermediate compounds were identified by gas chromatography–mass spectroscopy (GC–MS) analysis. Accordingly, the probable degradation pathway of DB129 in S. polyrrhiza was postulated.  相似文献

The changes and characteristics of endopeptidase (EP) isoenzymes in cucumber (Cucumis sativus L.) leaves during dark-induced senescence were investigated by activity staining after gradient-polyacrylamide gel electrophoresis (G-PAGE) containing co-polymerized gelatin as substrate. The results showed that both the chlorophyll and the protein contents of leaves were decreased, and the protein degradation was correlated with the increase of proteolytic activity during the course of leaf senescence. Meanwhile, nine cucumber endopeptidases isoenzymes (CEP) with 140, 120, 106, 94, 76, 55, 46, 39 and 35 kDa molecular weights were detected. Four of these, CEP2, 3, 4 and CEP9 appeared all the time, but the changes of the activity were different during incubation. Another four CEPs (CEP5, 6, 7 and CEP8) whose activities increased with dark-induced time were only detected in senescent leaves. Furthermore, the biochemical properties of these nine CEP were also characterized. All the CEPs had high activities from 35 ℃ to 45 ℃, and the optimum temperature was found to be 40 ℃. However, the activities of CEPs were not detected below 25 ℃ or over 60 ℃. The activity bands appeared at a wide range of pH from 5.0 to 9.0, but the optimum pH was found at 7.0. No CEPs were detected at pH 4 or pH 10. By inhibition analysis we concluded that CEP2, 3, 4 and CEP9 were serine endopeptidases and CEP6 was a kind of cysteine protease. It is suggested that serine endopeptidases might play a major role in cucumber leaf senescence, and for the first time, six senescencerelated endopeptidases (CEP1, 5, 6, 7, 8 and 9) were found in cucumber leaves.  相似文献

Abstract The ultrastructural features of the abscisic-acid-induced turion of Spirodela polyrrhiza are briefly described and a comparison between turion and vegetative frond tissue was made by stereological analysis. The turion is characterized by its small size, reniform shape, and dark-brown coloration; the mesophyll is undifferentiated and totally lacking the substantial acrenchyma development found in the vegetative frond. The turion cells have a smaller vacuole and a denser cytoplasm than the cells of the vegetative frond. Stereological analysis showed that the tissues differed quantitatively only in three main respects: air space formation, vacuolation, and starch and cell wall material accumulation. During development, it is suggested that the cells of the turion, while reaching the same final size as the vegetative frond cells, accumulate numerous starch grains, thick cell walls, and large deposits of tannins and anthocyanin pigment at the expense of the vacuolar expansion characteristic of the normal maturity programme. Certain features of the turion ultrastructure indicate a differential cell sensitivity to ABA.  相似文献

Abstract Using the method of compartmental analysis, the ion fluxes and compartment concentrations of Ca2+, K+ and Cl- have been compared in the untreated vegetative frond and the abscisic acid (ABA) induced turion of Spirodela polyrrhiza. The ABA-induced turion is characterized by reduced Ca2+ exchange across the tonoplast and low vacuolar Ca2+ concentration relative to the vegetative frond. In addition the turion exhibits a higher plasmalemma flux with a correspondingly high Ca2+ concentration in the cytoplasm. The concentration of K+ and Cl- is much lower in the cytoplasm of the ABA-induced turion than in the vegetative frond with the influx/efflux ratio at both the plasmalemma and the tonoplast being less than 1, a finding exhibited also in dormant storage tissue. Treatment of vegetative fronds with ABA for 18 h resulted in a reduced K+ plasmalemma efflux relative to untreated vegetative fronds and a concomitant increase in the cytoplasmic concentration. There was no rapid effect of ABA on Ca2+, K+ or Cl- fluxes through either membrane. These results are consistent with the notion that drastic changes in ion fluxes and concentrations in the turion are a secondary consequence of ABA-induced development, possibly due to prior regulation by ABA of enzymes inherent to processes involved in membrane transport.  相似文献

采用响应面法优化微波提取紫萍黄酮的条件。在单因素实验的基础上,选取乙醇体积分数、微波功率、液料比为影响因子,应用Box-Behnken中心组合法进行三因素三水平的试验设计,以紫萍黄酮得率为响应值,进行响应面分析(RSM),结果表明:微波提取紫萍黄酮的最佳提取条件为乙醇体积分数62%、液料比17∶1(mL/g)、微波功率500 W、微波时间9 min;在优化工艺下,紫萍黄酮的得率达到4.14%。对微波提取的紫萍黄酮进行抗氧化性研究发现紫萍黄酮对DPPH.清除作用较好,其抗氧化活性要优于相同浓度的抗坏血酸(VC)和2,6-二叔丁基对甲酚(BHT)。  相似文献
