Dear Editor, In the eukaryotic cell, the nuclear envelope separates genomic DNA from the rest of the cell, but ensures a molecu- lar communication between the nucleus and the cytoplasm through the nuclear pore complexes (NPCs). Nuclear trans- port of macromolecules is an energy-dependent process, exemplified by the classical nuclear import pathway. In the cytoplasm, proteins containing classical nuclear localization signals (cNLSs) are recognized by a hetero-dimeric transport receptor (importin-α (Impα):importin-β (Impl3) complex). Impa is employed as an adaptor protein that specifically rec-ognizes the classical nuclear localization signal (cNLS) from the cargo protein (Marfori et al., 2011). Translocation of the trimeric cargo:transport receptor import complex through the NPCs is mediated by transient interactions between Impβ and nucleoporins in the NPC. The directionality of the trans- port is imparted by the asymmetric distribution, between the cytoplasm and the nucleus, of the distinct nucleotide-bound (GTP/GDP) states of the small GTPase Ran (Tran et al., 2007).  相似文献

This study aimed to evaluate the effect of Cd exposure (100 μmol/L) on polar lipid composition, and to examine the level of fatty acid unsaturation in maize (Zea mays L.). In roots, the level of 16:0 and monounsaturated fatty acids (16:1 + 18:1) decreased in phosphatidylcholine (PC) and phosphatidylethanolamine (PE). In contrast, the proportion of unsaturated 18-C fatty acid species showed an opposite response to Cd. The content, on the other hand, of PC, PE, digalactosyldiacylglycerol (DGDG), and steryl lipids increased in roots (2.9-, 1.6-, 5.3-, and 1.7-fold increase, respectively). These results suggest that a more unsaturated fatty acid composition than found in control plants with a concomitant increase in polar lipids may favor seedling growth during Cd exposure. However, the observed increase in the steryl lipid (SL) : phospholipid (PL) ratio (twofold), the decrease in monogalactosyldiacylglycerol (MGDG) : DGDG ratio, as well as the induction of lipid peroxidation in roots may represent symptoms of membrane injury. In shoots, the unsaturation level was markedly decreased in PC and phosphatidylglycerol (PG) after Cd exposure, but showed a significant increase in sulfoquinovosyldiacylglycerol (SQDG), MGDG and DGDG. The content of PG and MGDG was decreased by about 65%, while PC accumulated to higher levels (4.4-fold increase). Taken together, these changes in the polar lipid unsaturation and composition are likely to be due to alterations in the glycerolipid pathway. These results also support the idea that the increase in overall unsaturation plays some role in enabling the plant to withstand the metal exposure.  相似文献

Intraguild predation (IGP) can be an important factor influencing the effective- ness of aphid natural enemies in biological control. In particular, aphid parasitoid foraging could be influenced by the presence of predators. This study investigated the effect of larvae of the predatory hoverfly Episyrphus balteatus DeGeer (Diptera: Syrphidae) and the multicolored Asian ladybird Harmonia axyridis Pallas (Coleoptera: Coccinellidae) on the foraging behavior of the aphid parasitoid, Aphidius ervi Haliday (Hymenoptera: Aphidiidae) in choice experiments using a leaf disc bioassay. Wasp response to chemical tracks left by those predator larvae was also tested. Parasitoid behavior was recorded using the Observer (Noldus Information Technology, version 5.0, Wageningen, the Netherlands). The experiments were conducted under controlled environmental conditions using leaves of the broad bean plant, Viciafaba L. (Fabaceae) with Myzus persicae Sulzer (Homoptera: Aphididae) as the host complex. A. ervi females avoided aphid patches when larvae of either predator were present. A similar avoidance response was shown by A. ervi to aphid patches with E. balteatus larval tracks, whereas no significant response was observed to tracks left by H. axyridis larvae. It was concluded that IG predator avoidance shown by the aphid parasitoid A. ervi may be a factor affecting their distribution among host patches.  相似文献

目的:观察强直电刺激大鼠右侧尾壳核(CPu)时,CPu-海马(HPC)网络癫痫的神经信息编码特征。方法:雄性SD大鼠59只。急性或慢性强直电刺激CPu(acute tetanization of the right CPu or chronic tetanization of the right CPu,ATRC or CTRC)(60Hz.0.4~0.6mA,2s)诱导大鼠癫痫模型。结果:①ATRC可以诱导双侧HPC神经元出现非对称性癫痫相关性单位电活动.增加对侧HPC单位放电时间间隔(Interspike interval,ISI)点分布的分岔角度。②CTRC可以诱导双侧CPu网络出现尖波样连续发放,同侧振荡样网络发作具有明显的相位移动特征;频率变化顺序为70Hz、110Hz、35Hz以及30Hz.与时间呈显著的负相关;振荡波波峰间隔(Interpeak interval,IPI)和波峰振幅逐渐增大,与时间呈显著正相关。③CTRC后加ATRC可以分别诱导双侧CPu网络出现原发性后放电。结论:激活CPu可以跨大脑半球重建双侧CPu—HPC癫痫电网络.其神经信息编码特征可能成为癫痫发生的神经信息学基础。  相似文献

本文通过扫描电镜、透射电镜观察了日本鳗鲡(Anguilla japonica)精子形成的特殊过程及产生细胞器的特殊结构。由精细胞变成精子包括四个特殊阶段,即经过早期、中期、晚期和精子期.最后形成正常成熟的精子.(1)早期阶段:其特征是细胞核由椭圆形逐步变成长条形;在细胞核的一端.有一个大的圆的染色较浅的形状似球形的特殊结构,约占细胞核的三分之一,其内含有少量着色深的颗粒状和线条状物质,外面由质膜包被着与细胞核分开,该结构和细胞核的外层还有一层质膜包着形成一个整体:精子早期阶段没有形成独立的线粒体和中心粒。(2)中期阶段:其特征是细胞核呈长条形.有球形结构的一端成为细胞核的上端,无球形结构的一端成为细胞核的下端,在下端出现鞭毛的原基;球形结构伴随着精子的发生也发生变化,内部逐步分化出中心粒和线粒体等细胞器:在细胞核的中段有明显的溶酶体分布。(3)晚期阶段:其特征是细胞核呈“眉形”或“新月形”.中心粒从球形结构中释放出来形成独立结构.球形结构中只剩下还没有形成独立结构的线粒体:在细胞核的下端鞭毛的原基处长出较长的鞭毛,这时期的精子已具有运动能力。(4)精子期:其特征是细胞核呈圆形,中心粒位于植入窝内,线粒体分布在细胞核的下面.在线粒体的下面有袖套腔形成,此时形成的鞭毛为“9+2”结构。日本鳗鲡精子经过四个时期的变态.才能形成真正成熟的精子。  相似文献

Substrate selectivity of glycerol-3-phosphate acyltransferase (EC 2. 3. 1. 15) of rice (Oryza sativa L.) was explored in a comparative study of acyltransferases from seven plant species. In vitro labeling of acyl carrier protein (ACP) with ^14C or 3H showed that acyltransferase from chill-sensitive plants, such as rice that uses either oleic (18:1) or palmitic acid (16:0) as acyl donor at comparable rates, displays lower selectivity than the enzyme from chill-resistant plants, such as spinach, which preferentially uses oleic acid (18:1) rather than palmitic acid (16:0) as an acyl donor. This may be a result of the size and character of the substrate-binding pocket of acyltransferase. Homology modeling and protein structure-based sequence alignment of acyltransferases revealed that proteins from either chill-sensitive or chill-tolerant plants shared a highly conserved domain containing the proposed substrate-binding pocket. However, the aligned residues surrounding the substrate-binding pocket are highly heterogeneous and may have an influence mainly on the size of the substrate binding pockets of acyltransferases. The substrate selectivity of acyltransferase of rice can be improved by enlarging the substrate-binding pocket using molecular biological methods.  相似文献

A collection of 368 advanced lines and cultivars of spring wheat(Triticum aestivum L.) from Chile, Uruguay, and CIMMYT(Centro Internacional de Mejoramiento de Maíz y Trigo), with good agronomic characteristics were evaluated under the Mediterranean conditions of central Chile. Three different water regimes were assayed: severe water stress(SWS, rain fed), mild water stress(MWS; one irrigation around booting), and full irrigation(FI; four irrigations: at tillering,flag leaf appearance, heading, and middle grain filling). Traits evaluated were grain yield(GY), agronomical yield components,days from sowing to heading, carbon isotope discrimination(△^13C) in kernels, and canopy spectral reflectance. Correlation analyses were performed for 70 spectral reflectance indices(SRI) and the other traits evaluated in the three trials. GY and △^13C were the traits best correlated with SRI, particularly when these indices were measured during grain filling. However,only GY could be predicted using a single regression, with ResearchNormalized Difference Moisture Index(NDMI2: 2,200; 1,100)having the best fit to the data for the three trials. For △^13C, only individual regressions could be forecast under FI(r^2: 0.25–0.37)and MWS(r^2: 0.45–0.59) but not under SWS(r^2: 0.03–0.09).NIR‐based SRI proved to be better predictors than those that combine visible and NIR wavelengths.  相似文献

Felines use their spinal column to increase their running speed at rapid locomotion performance. However, its motion profile behavior during fast gait locomotion has little attention. The goal of this study is to examine the relative spinal motion profile during two different galloping gait speeds. To understand this dynamic behavior trend, a dynamic motion of the feline animal (Felis catus domestica) was measured and analyzed by motion capture devices. Based on the experiments at two different galloping gaits, we observed a significant increase in speed (from 3.2 m.s-1 to 4.33 m.s-1) during the relative motion profile synchronization between the spinal (range: 118.86~ to 168.00~) and pelvic segments (range: 46.35~ to 91.13~) during the hindlimb stance phase (time interval: 0.495 s to 0.600 s). Based on this discovery, the relative angular speed profile was applied to understand the possibility that the role of the relative motion match during high speed locomotion generates bigger ground reaction force.  相似文献

目的:观察家兔心肌缺血/再灌注(I/R)损伤对心肌细胞核膜核苷三磷酸酶(NTPase)动力学及mRNA出核转运的变化。方法:制备家兔离体心脏I/R模型,密度梯度离心法制备心肌细胞核膜,按Tiffany等的方法测定NTPase活性及mRNA的转运率。结果:心肌细胞核膜NTPase在以ATP或GTP为底物时,与对照组比较,I/R组最大反应速率(Vmax)分别降低26%及22%(P<0.01)。Km值分别升高54%及72%(P<0.01);而缺血组Vmax及Km值较对照组无明显差异。以ATP或GTP启动反应时I/R组10min内心肌细胞核mRNA转出率分别较对照组降低27%及32%(P<0.01);而缺血组与对照组相比并无显著性差异(P>0.05)。结论:心肌缺血/再灌注损伤时,心肌细胞核膜受损,核膜上NTPase活性降低,导致细胞核膜mRNA出核受阻,从而可能影响蛋白质的合成表达。  相似文献

The hot-spring snakes,Thermophis, were previously known only from the Tibet Autonomous Region and Western Sichuan, China. During the past two years, three adult hot-spring snakes (2 females, 1 male) were sampled in Shangri-La, northern Yunnan, China, thus expanding their known distribution region towards the southeast. This site is the southeastern-most corner of the Tibetan Plateau and the southernmost tip of the Hengduan Mountains (Mts.). Phylogenetic analysis of mitochondrial (mt) and nuclear (n) DNA segments suggested that the three specimens belong to the genus ofThermophis. Morphologically, the new species is more similar toT. zhaoermii. However, it is distinguished fromT.zhaoermiiin the number of maxillary teeth (15), distance between the two eyes/head width, rostral width/height, mental width/height, in one character limited to female: head width/length, and in four characters restricted to male: occurrence of the reduction from 10 to 8 (8 to 6, 6 to 4) scales in each dorsal row on the tail. There are differences in morphology, genetics (mtDNA, nDNA), and geography between the putative new species andT.zhaoermii,the new species meets our proposed eclectic and feasible "four-differences" rule.  相似文献

In an attempt to explore the relationships between phosphatidylglycerol (PG) molecular species of thylakoid membrane lipids and sensitivities to chilling-induced photoinhibition, PG molecular species, D1 protein, electron transport activities of thylakoid membrane and the potential quantum yield (FvlFm) in rice treated under middle and low photon flux density (PFD) at 11℃ were analyzed by high performance liquid chromatography, enzyme hydrolysis, gas phase chromatography (GC) and so on. Results showed that the major molecular species of PGs in rice thylakoid membrane were 18:3/16:0, 18:3/16:1(3t), 18:2/16:0, 18:2/16:1(3t), 18:1/16:0, 18:1/16:1(3t), 16:0/16:0, 16:0/16:1(3t). There were large differences in the contents of unsaturated PG molecular species such as 18:1-3/16:0-16:1(3t) and saturated PG molecular species like 16:0/16:0-16:1(3t) among japonica cv 9516 0-9516), japonica-indica hybrid F1 j-9516/i-SY63 (ji-95SY) and indica cv Shanyou 63 (i-SY63). J-9516 containing higher contents of unsaturated PG molecular species was manifest in stable D1 protein contents under chill and tolerant to chill-induced photoinhibition. In contrast to j-9516, i-SY63 with lower contents of unsaturated PG molecular species, exhibited unstable D1 protein contents under chill and was sensitive to chill-induced photoinhibition, ji-95SY containing middle contents of unsaturated PG molecular species between those of j-9516 and i-SY63, exhibited mid extent of sensitivity to chill-induced photoinhibition. The losses in D1 protein also account for the inhibition in electron transport activity of thylakoid membrane and the observed decline in FvlFm. The PG molecular species that is efficient in raising chilling-resistant capacity were those containing unsaturated fatty acids, namely, unsaturated PG molecular species. These results implied that the substrate selectivity of the glycerol-3-phosphate acyltransferase in chloroplasts towards 16:0 or 18:1 displayed greatly the difference between japonica and indica rice. Itwas possible to enhance the capacity of resistance to chilling-induced photoinhibition by improving or modifying the GPAT gene.  相似文献
