The aim of the present study was to analyze the 24-h rhythm in plasma melatonin concentration and the day-night differences in synaptophysin expresion and ultrastructural characteristics of the pinealocytes in developing female sheep. Ewes of three different ages were examined: infantile (1-6 months old), pubertal and early fertile age (9-24 months old) and adult (36-60 months old). Experiments were conducted under natural non-stimulatory (long) and stimulatory (short) photoperiods. The obtained results were similar for both analyzed photoperiods. Plasma melatonin concentration, measured in samples obtained every 4 h, showed a similar pattern in the three age groups, with peak values at 02:00 h and troughs at 14:00 h. Mean value of plasma melatonin levels in 9-24 month-old sheep was significantly greater than that in younger or older sheep. The weight of pineal glands obtained at night (02:00 h) was significantly higher than in daylight (14:00 h). Pubertal and early fertile sheep had the largest pineal glands. The pineal volume, and the total number of pinealocytes per gland of 9-24 months-old sheep differed significantly from that of younger or older sheep. The pineal volume, and the mean volume of pinealocytes was significantly greater in animals killed at night. Number of pinealocytes did not vary between animals killed during daylight or at night. The mean volumen of pinealocytes did not show statistical differences between the age groups. In quantitative ultrastructural analysis of pinealocyte cells, the relative volume of mitochondria, rough endoplasmic reticulum and Golgi complexes was significantly greater in 9-24 month-old sheep and in animals killed at night. The relative volume of lipid droplets was highest in older sheep. Collectively, the data support the existence of developmental changes in pinealocyte morphology and quantity, partially in coincidence with a higher melatonin secretion rate.  相似文献

Photoperiodic manipulation of young European starlings suggests that their reproductive physiology is incapable of responding to a short photoperiod until they are fully grown. This study aimed to determine whether the lack of response to a short photoperiod is reflected in the daily profile of plasma melatonin concentrations. Five-day-old starlings taken from nest boxes showed a significant (p < 0.0001) rhythm in plasma melatonin concentrations, with high values during night. In nestlings hand-reared from 5 days of age on a long photoperiod (LD 16:8), equivalent to natural photoperiod at the time, the amplitude of the daily rhythm in melatonin increased significantly (p < 0.01) with age until birds were fully grown (20 days old). In nestlings reared on a short photoperiod (LD 8:16), the daily melatonin profile remained almost identical to that of long photoperiod birds until they were fully grown. However, after 20 days old, the duration of elevated nighttime melatonin began to extend to encompass the entire period of darkness. In contrast, fully grown starlings transferred from a long to a short photoperiod had partially adapted to the short photoperiod after 5 days; by 10 days, the daily melatonin profile was identical to that of birds held chronically on a short photoperiod. Thus, consistent with responses of reproductive physiology, the pineal of young birds appears to be incapable of perceiving, or adapting to, a short photoperiod.  相似文献

Adult white-footed mice were maintained under either a long photoperiod (LP, LD 16:8, lights out at 2100) or a short photoperiod (SP, LD 8:16, lights out at 1700) for six weeks. Subgroups from each lighting regime were killed at specific times over a 24 hour period. Pineal radioimmunoassayable melatonin levels were significantly elevated at night compared to daytime values. Pineal melatonin content appears to be elevated for a longer period of time in the SP mice than in the LP animals. The apparent increased melatonin production observed in white-footed mice maintained under short and reproductively repressive daylengths may help to explain the ability of chronically available exogenous melatonin to cause gonadal atrophy in this species.  相似文献

The purpose of this study was to determine structural and immunocytochemical changes taking place during the day and at night in developing sheep pineal gland under natural non-stimulatory photoperiods (summer solstice). Additionally, the diurnal cycle of plasma melatonin levels was charted and differences between diurnal and nocturnal pineal melatonin concentrations were analyzed. 36 ewes of three different ages were examined: infants (1-6 months old), pubertal and early fertile age (9-24 months old) and adults (36-60 months old). Plasma and pineal gland melatonin levels were higher in pubertal sheep than in infants or adults. Pubertal sheep pineal glands were also heavier, contained a larger number of pinealocytes and interstitial cells and displayed more evident innervation and vascularisation than infants or adults. There was no difference in the number of pinealocytes and interstitial cells between animals killed during daylight or at night. Gland weight, pinealocyte nuclear profile areas and plasma melatonin concentrations were all significantly higher at night than during the day.  相似文献

Estradiol receptors (ER) in mammary glands of female Holtzman rats, either intact or neonatally pinealectomised and housed in 10:14 or 24:0 L:D (light:dark) schedule were studied at 30, 40, 50 +/- 5 and 65 +/- 5 days of age. Whereas ER were detectable only at the age of 60-65 days in intact rats housed in 10:14 L:D, they were present as early as 30 days onwards in the pineal ablated group. In the 24:0 L:D pinealectomised group, though mammary gland ER were maximum around 40 days of age and temporarily undetectable around day 50, they had stabilized around the age of 60-65 days. The data demonstrates for the first time, the modulation of ER in rat mammary glands in response to varying photoperiods as well as pineal ablation. Earlier reports on incidence of chemically induced mammary tumours have been compared to the receptor modulation.  相似文献

Light regulates a variety of behavioral and physiological processes, including activity rhythms and hormone secretory patterns. Seasonal changes in the proportion of light in a day (photoperiod) further modulate those functions. Recently, short (SP) versus long days (LP) were found to markedly increase light sensitivity for phase shifting in Syrian hamsters. To our knowledge, photoperiod effects on light sensitivity have not been studied in other rodents, nor is it known if they generalize to other circadian responses. We tested whether photic phase shifting and melatonin suppression vary in Siberian hamsters maintained under LP or SP. Select irradiances of light were administered, and shifts in activity were determined. Photic sensitivity for melatonin suppression was examined in a separate group of animals via pulses of light across a 4 log-unit photon density range, with post-pulse plasma melatonin levels determined via RIA. Phase shifting and melatonin suppression were greater at higher irradiances for both LP and SP. The lower irradiance condition was below threshold for phase shifts in LP but not SP. Melatonin suppression did not vary by photoperiod, and the half saturation constant for fitted sigmoid curves was similar under LP and SP. Thus, the photoperiodic modulation of light sensitivity for phase shifting is conserved across two hamster genera. The dissociation of photoperiod effects on photic phase shifting and melatonin suppression suggests that the modulation of sensitivity occurs downstream of the common retinal input pathway. Understanding the mechanistic basis for this plasticity may yield therapeutic targets for optimizing light therapy practices.  相似文献

The impact of photoperiod on the rate and magnitude of N remobilization relative to uptake of inorganic N during the recovery of shoot growth after a severe defoliation was compared over 18 days in two temperate grass species, timothy (Phleum pratense L. cv. Bodin) and meadow fescue (Festuca pratensis Huds. cv. Salten). Plants were grown in flowing solution culture with N supplied as 20 mM NH4NO3 and pre-treated by extending the 11 h photosynthetically significant light period either by 1 h (short-day or SD plants) or 7 h (long-day or LD plants) of very low light intensity, during the 10 days prior to defoliation. Following a single severe defoliation, 15N-labelled NH4+ or NH4++ NO3? was supplied over a 20-day recovery period under the same SD and LD conditions. Changes in the relative contributions of remobilized N and newly acquired mineral N to shoot regrowth were assessed by sequential harvests. Both absolute and relative rates of N remobilization from root and stubble fractions were higher in LD than SD plants of both species, with the enhancement more acute but of shorter duration in timothy than fescue. Remobilized N was the predominant source of N for shoot regrowth in all treatments between days 0 and 8 after cutting; on average more so for fescue than timothy, because the presence of NO3? reduced the proportional contribution of remobilized N to the regrowth of timothy but not of fescue. Net uptake of mineral N began to recover between days 4 and 6 after cutting, with NO3? uptake restarting 1 or 2 days earlier than NH4+ uptake, even when NH4+ was the only form of N supply. LD timothy plants supplied solely with NH4+ were slowest to resume uptake of mineral N. Supplying NO3? in addition to NH4+ after defoliation promoted shoot regrowth rate but not remobilization of N. Rates of regrowth (shoot dry weight production per plant) were not correlated with rates of N remobilization from stubble either over the short-term (days 0–8) or longer term (days 0–18), interpreted as evidence against a causal dependence of regrowth rate on N remobilization under these conditions. The results are discussed in relation to hypotheses for source/sink-driven rates of N remobilization and their interactions with mineral N uptake following defoliation.  相似文献

Plasma melatonin concentrations were measured every 1-2 h over 24 h and plasma luteinizing hormone (LH) concentrations every 15 min over 12 h in domestic gilts reared under artificial light regimens that had previously been used to demonstrate photoperiodic effects on puberty. In Expt 1, the light regimens both commenced at 12 h light: 12 h dark (12L:12D) and either increased (long-day) or decreased (short-day) by 15 min/week until the long-day gilts were receiving 16L:8D and the short-day gilts 8L:16D at sampling. In Expt 2, both light regimens commenced at 12L:12D and either increased (long-day) or decreased (short-day) by 10 or 15 min/week to a maximum of 14.5L:9.5D or a minimum of 9.5L:14.5D before being reversed. Sampling took place when daylength had returned to 14L:10D (long-day) or 10L:14D (short-day). In immature gilts housed at 12L:12D (Expt 1) and in postpubertal (Expt 1) and prepubertal (Expt 2) gilts reared under long-day or short-day light regimens, mean plasma melatonin concentrations were basal (3.6 pg/ml) when the lights were on and increased to peak concentrations greater than 15 pg/ml within 1-2 h after dark, before declining gradually to basal concentrations at or near the end of the dark phase. In prepubertal gilts bearing subcutaneous melatonin implants and reared under long-days (Expt 2), mean plasma melatonin concentration in the 6 h before dark was 91.9 +/- 5.26 pg/ml and 125.0 +/- 6.66 pg/ml 1 h after dark, but this increase was not statistically significant. In Expt 2, the short-day gilts had fewer LH pulses (2.6 +/- 0.25 vs. 4.6 +/- 0.24; P less than 0.01) in the 12-h sampling period than the long-day gilts, but the amplitude of the pulses (2.28 +/- 0.23 vs. 1.26 +/- 0.16 ng/ml; P less than 0.01) and the area under the LH curve (78.8 +/- 5.60 vs. 47.3 +/- 6.16; P less than 0.01) was greater in the short-day gilts. In the short-day, but not in the long-day, gilts LH pulses were more frequent (2.0 +/- 0.0 vs. 0.6 +/- 0.25; P less than 0.01), but had a smaller area (61.9 +/- 7.2 vs. 120.2 +/- 23.6; P less than 0.05) in the 6 h of dark than in the 6 h of light, which together made up the 12-h sampling period.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Environmental experiences during development provide animals with important information about future conditions. Siberian hamsters are photoperiodic rodents that dramatically adjust their physiology and behavior to adapt to seasonal changes. For example, during short winter-like days, hamsters enhance some components of immune function putatively to cope with increasing environmental challenges. Furthermore, early life stress alters the developmental course of the immune system. Overall, immune function is typically suppressed in response to chronic stress, but responses vary depending on the type of stress and components of immune function assessed. This led us to hypothesize that delayed-type hypersensitivity (DTH), an antigen-specific, cell-mediated immune response, would be differentially modulated in hamsters that underwent early life maternal separation (MS) in either short or long photoperiods. At birth, hamsters were assigned to either short (SD; 8 h light/day) or long (LD; 16 h light/day) photoperiods and either daily 3 h MS, daily 15-min brief maternal separation (BMS), or no manipulation from postnatal day 2 through 14. In adulthood DTH was assessed. Hamsters reared in short days enhanced DTH responses. MS and BMS attenuated DTH responses in both short and long days. However, BMS long-day female hamsters did not suppress pinna swelling, suggesting a protective effect of female sex steroids on immune function. As is typical in short days, reproductive tissue was regressed. Reproductive tissue mass was also decreased in long-day MS female hamsters. Furthermore, MS altered photoperiod-induced changes in body mass. Taken together, these findings suggest that manipulations of early life mother-pup interactions in Siberian hamsters result in physiological changes and suppressed cell-mediated immunity.  相似文献

Pineal concentrations of N-acetylserotonin and melatonin and serum levels of melatonin were studied in 3-wk-old (prepubertal), 8-wk-old (adult), and 17-mo-old (senile) male rats. They were adapted to a photoperiod of 12 h light/12 h darkness for a minimum of 1 wk and killed at mid-light and mid-dark. Melatonin and N-acetylserotonin were determined by radioimmunoassay. The concentrations of pineal N-acetylserotonin and melatonin were high in the dark period and low in the light period. Statistical analysis indicated that pineal N-acetylserotonin and melatonin levels per 100 gm body weight declined with age. Similarly, serum melatonin demonstrated diurnal changes in all the age groups studied. In addition, there was a significant reduction in the levels of serum melatonin with age. The parallel patterns of decrease in pineal and serum melatonin levels with age suggest a decline in pineal secretion of melatonin in the older animals.  相似文献

Daily rhythms of nonshivering thermogenesis NST were studied in common spiny mice Acomys cahirinus, acclimated to different photoperiod regimes (16L:8D and 8L:16D) at a constant ambient temperature of 26°C. Noradrenaline NA (1.5 mg/kg subcutaneous) was injected at: 06:00, 12:00, 18:00 and 24:00 h (±15 min). NST was measured as the ratio between the maximal oxygen consumption (VO2) as response to NA — VO2NA and VO2 measured at 26°C — VO2 min. Rectal temperatures TbNA and Tb min respectively were recorded at the end of VO2 measurements. Significant variations in Tb min, TbNA, and NST were revealed, under the two different photoperiod regimes. Significant differences in VO2 min, NST, Tb min and TbNA were also recorded within each photoperiod acclimation group. These results suggest that daily and photoperiod depended variations in the brown adipose tissue activity, presumably emerge from amount of unoccupied receptors or changes in the receptors affinity to NA.  相似文献

The present study examines how the circadian oscillators in the retina and the suprachiasmatic nucleus (SCN) respond to changes in photoperiod. Arylalkylamine N-acetyltransferase (aa-nat) gene expression studied by quantitative RT-PCR revealed that in adult Sprague-Dawley rats kept under different light-dark (LD) cycles for two weeks the temporal pattern of AA-NAT mRNA expression was identical in retina and pineal gland. In both tissues, the time span between the onset of darkness and the nocturnal rise in AA-NAT mRNA expression was 3 h under LD 20:4, 6 h under LD 12:12, and 15 h under LD 4:20. As aa-nat expression in the pineal gland is regulated by the circadian oscillator in SCN, the results suggest that the photoperiodic differences accompanying the seasons of the year are imprinted in more than one oscillator and that this may accentuate the important message regarding 'time of year.'  相似文献

The effect of subcutaneously injected DT gamma E (beta-endorphin, (beta E)2-17) on the pineal melatonin level was compared with that of closely related peptides and the neuroleptic drug haloperidol. As found previously, DT gamma E (3 ng/rat and 300 ng/rat) increased the melatonin levels. Similar doses of DT alpha E (beta E 2-16), DT beta E (beta E 2-31), gamma E (beta E 1-17), alpha E (beta E 1-16) and beta E failed to significantly change the melatonin levels in both the dark and the light phase. Haloperidol in a dose of 300 ng/rat exhibited a similar effect as DT gamma E.  相似文献

N-acetyltransferase (NAT) is believed to be the rate-limiting enzyme in the synthesis of melatonin from serotonin in the pineal gland. Norepinephrine released from sympathetic nerve endings within the pineal gland stimulates NAT activity and, therefore, melatonin synthesis. When an animal is subjected to a stressful stimulus, it would be expected that the increase in plasma stimulus, it would be expected that the increase in plasma catecholamines originating from the adrenal medulla and/or the sympathetic nervous system would result in a stimulation of pineal NAT activity. Adult male rats were given a 1.5cc injection of physiological saline subcutaneously into the back leg. Compared to non-injected controls, animals stressed in this manner were shown to have significantly lower pineal melatonin content 10 min after the saline injection late in the light phase of the light/dark cycle (at 18.30 h-lights on at 07.00 h). To test this more thoroughly, a time course study was conducted during the dark phase (at 02.00 h-5 hours after lights out) when pineal NAT activity and melatonin levels are either increasing or elevated. NAT activity and melatonin levels in the pineal were significantly depressed in stressed animals as compared to controls by 10 min after the saline injection, and remained so until 60 min after injection. By 90 min they had returned to control values. In the next study the nighttime response of the pineal to stress was compared in intact and adrenalectomized rats. Adrenalectomy prevented the changes in NAT activity and melatonin content associated with the saline injection. Some factor, such as a catecholamine or corticosterone from the adrenal, seems to be eliciting the response in the pineal to the saline injection. It is not known if the factor is acting centrally or directly on the pineal gland.  相似文献

The present study examined the effects of photoperiod on spatial and temporal memory in adult Sprague-Dawley rats that were conceived and reared in different day lengths, i.e., short day (SD-8:16 light/dark) and long day (LD-16:8 light/dark). Both male and female LD rats demonstrated increased spatial memory capacity as evidenced by a lower number of choices to criterion in a 12-arm radial maze task relative to the performance of SD rats. SD rats also demonstrated a distortion in the content of temporal memory as evidenced by a proportional rightward shift in the 20 and 60 s temporal criteria trained using the peak-interval procedure that is consistent with reduced cholinergic function. The conclusion is that both spatial and temporal memory are sensitive to photoperiod variation in laboratory rats in a manner similar to that previously observed for reproductive behaviour.  相似文献
