Although it is universally accepted that protein synthesis occurs in the cytoplasm, the possibility that translation can also take place in the nucleus has been hotly debated. Reports have been published claiming to demonstrate nuclear translation, but alternative explanations for these results have not been excluded, and other experiments argue against it. Much of the appeal of nuclear translation is that functional proofreading of newly made mRNAs in the nucleus would provide an efficient way to monitor mRNAs for the presence of premature termination codons, thereby avoiding the synthesis of deleterious proteins. mRNAs that are still in the nucleus-associated fraction of cells are subject to translational proofreading resulting in nonsense-mediated mRNA decay and perhaps nonsense-associated alternate splicing. However, these mRNAs are likely to be in the perinuclear cytoplasm rather than within the nucleus. Therefore, in the absence of additional evidence, we conclude that nuclear translation is unlikely to occur.  相似文献

This study sought to examine the shear modulus (i.e., an force index) of three quadriceps muscles [i.e., vastus medialis (VM), vastus lateralis (VL), and rectus femoris (RF)] during passive stretching to determine whether epimuscular myofascial force transmission occurs across muscles. Secondly, this study compared the shear modulus between the quadriceps muscles, in both proximal and distal regions. Twelve healthy individuals were assessed during a passive knee flexion maneuver between 0° and 90° of knee flexion with the hip in two positions: flexed (80°) vs. neutral (0°). Muscle electrical activity was also assessed during the testing. No differences were observed between the hip testing positions for myoelectric activity (p > 0.43), and for VL and VM shear modulus (p = 0.12–0.98). Similarly, there were no differences between the proximal and distal regions for all muscles (p = 0.42–0.93). RF showed a higher shear modulus with the hip in the neutral position (p = 0.004). With the hip flexed, the VL showed the greatest shear modulus among the tested muscles (p < 0.025); while with the hip in the neutral position, no differences were observed for shear modulus between VL and RF (p = 0.817). These findings suggest that epimuscular myofascial force transmission (at a muscle belly level) does not occur between the quadriceps muscles when passively flexing the knee until 90°. Whether epimuscular myofascial force transmission occurs in the quadriceps muscles bellies with greater muscle stretch (either through knee flexion or hip extension) remains to be examined.  相似文献

Wolbachia are maternally inherited endosymbiotic bacteria found within many insect species. Aedes mosquitoes experimentally infected with Wolbachia are being released into the field for Aedes‐borne disease control. These Wolbachia infections induce cytoplasmic incompatibility which is used to suppress populations through incompatible matings or replace populations through the reproductive advantage provided by this mechanism. However, the presence of naturally occurring Wolbachia in target populations could interfere with both population replacement and suppression programs depending on the compatibility patterns between strains. Aedes aegypti were thought to not harbor Wolbachia naturally but several recent studies have detected Wolbachia in natural populations of this mosquito. We therefore review the evidence for natural Wolbachia infections in A. aegypti to date and discuss limitations of these studies. We draw on research from other mosquito species to outline the potential implications of natural Wolbachia infections in A. aegypti for disease control. To validate previous reports, we obtained a laboratory population of A. aegypti from New Mexico, USA, that harbors a natural Wolbachia infection, and we conducted field surveys in Kuala Lumpur, Malaysia, where a natural Wolbachia infection has also been reported. However, we were unable to detect Wolbachia in both the laboratory and field populations. Because the presence of naturally occurring Wolbachia in A. aegypti could have profound implications for Wolbachia‐based disease control programs, it is important to continue to accurately assess the Wolbachia status of target Aedes populations.  相似文献

The evidence and arguments for and against the occurrence of endocytosis in fungal hyphae are summarized. The balance of evidence is in favour of the existence of endocytosis. This is supported by an analysis of the recently sequenced Neurospora genome which strongly suggests that this fungus possesses the complex protein machinery required to conduct endocytosis.  相似文献

The process by which a single follicle is selected to ovulate while others regress is unknown in ewes. If the dominant follicle secretes substances that directly inhibit the growth of other follicles, the superovulatory response to the administration of exogenous gonadotrophins may be blunted. Administration of 1250 iu pregnant mares' serum gonadotrophin (PMSG) before or after the emergence of the dominant follicle in the follicular phase, or 1000 iu PMSG in the presence or absence of a large healthy or atretic follicle during the luteal phase did not affect the induced ovulatory response. Comparisons between the ovary with or without the dominant follicle did not reveal any differences in ovulatory response to PMSG. The in-vitro features (i.e. mitotic index, oestradiol and testosterone production) of follicles ipsilateral or contralateral to the dominant follicle during the early and late follicular phases were also similar. If the dominant follicle secretes substances detrimental to the other follicles, this could be mimicked in vitro. Co-culture of small follicles with the largest follicles in a closed system did not reduce their incorporation of 3H thymidine in granulosa cells, compared with small follicles cultured alone. These data suggest that dominance is probably not operative in sheep. The administration of 500 iu of PMSG during the midfollicular phase increased ovulation rate in Merino ewes, indicating that dominance is essentially passive in ewes and can easily be overcome by raising gonadotrophin concentration.  相似文献

Meesia kenyae P. Varde, a species originally described in 1955 from the altimontane elevation on Mt. Kenya in tropical East Africa, is briefly assessed taxonomically and some details of the type material are illustrated. This species is considered to be conspecific with Ceratodon purpureus (Hedw.) Brid., a weedy, cosmopolitan and highly protean species. Because M. kenyae is the only representative of the Meesiaceae in Africa, this taxonomic conclusion results in the exclusion of this small family from the bryoflora of this continent. Accordingly, the Meesiaceae are represented in the tropics only by three species, namely Meesia triquetra (Jolycl.) Ångstr. in Papua New Guinea, M. longiseta Hedw. in Colombia and M. ulei Müll. The hypothesis that respiratory modulation of heart rate variability (HRV) or respiratory sinus arrhythmia (RSA) is restricted to mammals was tested on four Antarctic and four sub-Antarctic species of fish, that shared close genotypic or ecotypic similarities but, due to their different environmental temperatures, faced vastly different selection pressures related to oxygen supply. The intrinsic heart rate (fH) for all the fish species studied was approximately 25% greater than respiration rate (fV), but vagal activity successively delayed heart beats, producing a resting fH that was synchronized with fV in a progressive manner. Power spectral statistics showed that these episodes of relative bradycardia occurred in a cyclical manner every 2-4 heart beats in temperate species but at >4 heart beats in Antarctic species, indicating a more relaxed selection pressure for cardio-respiratory coupling. This evidence that vagally mediated control of fH operates around the ventilatory cycle in fish demonstrates that influences similar to those controlling RSA in mammals operate in non-mammalian vertebrates.  相似文献

Release of 14CO2 during metabolism of [1-14C]glycolate was studied in purified, intact and Triton X-100-solublized peroxisomes isolated from leaves of Secale cereale. Decarboxylation, apparently resulting from H2O2 attack on glyoxylate, was stimulated in both intact and solubilized peroxisomal preparations by the catalase inhibitors aminotriazole and sodium azide. CO2 evolution was also observed in solubilized peroxisomes in the absence of inhibitor when an amino donor was not provided for conversion of glyoxylate to glycine. Loss of CO2 from labelled glycolate in the absence of amino donors was reduced by addition of exogenous catalase to the reaction medium.Intact peroxisomes showed no glycolate decarboxylation whether or not amino donors were supplied. It appears that the CO2 release from glycolate observed in previous investigations may be an artifact of the peroxisomal preparations and the assay systems used, and not a significant factor in photorespiratory metabolism in vivo.  相似文献

Improving total knee replacement (TKR) requires better understanding of the many factors influencing clinical outcomes. Recently, probabilistic studies have investigated the influence of variability for individual TKR activities. This study demonstrates conceptually how probabilistic studies might further provide a framework to explore relationships not just within but between multiple different activities, e.g. intra-operative passive laxity drawer loading and post-operative active gait. Two implants were compared using simulated ISO-gait and passive laxity loading, with factors including mal-positioning and soft-tissue constraint varied using Monte-Carlo analysis. The results illustrate that correlations between different activities can be quantified; this demonstration study suggests further research is justified (with detailed clinically representative models) to explore the relationship between passive and active mechanics for specific in vivo conditions. Probabilistics is a key enabling methodology for achieving this goal. In future, exploring correlations between different activities may facilitate a better holistic understanding of TKR function.  相似文献

Microspectrophotometric analyses combined with model experiments using polyacrylamide films containing different types of purified glycosaminoglycan or glycosaminoglycan and basic protein, have been applied to investigate the chemical characteristics of the glycosaminoglycan moiety present in rat mucosal mast cell granules. The results obtained with these histochemical techniques present evidence of the absence of significant amounts of heparin and the presence of lower sulfated glycosaminoglycan(s) in the granules of these cells.  相似文献

To determine if the usual natriuretic response to ANP could be altered by raising intrarenal levels of adenosine, ANP was administered to normal anesthetized dogs at 100 ng.kg-1.min-1 i.v. before and after the administration of adenosine (3 micrograms.kg-1.min-1) into the left renal artery (n = 8). For each kidney, the group mean delta UNaV in response to ANP was unchanged by the presence of adenosine. However, following intrarenal infusion of adenosine, this unaltered average response for the infused kidney was achieved by either attenuation or exaggeration of the natriuresis to ANP in half the dogs, respectively. When intrarenal levels of extracellular adenosine were elevated by the i.v. infusion of dipyridamole in seven dogs, there was uniform exaggeration of an ANP-induced natriuresis by an average of 145 mu equiv./min. The provision of theophylline by itself (an adenosine antagonist) had no effect on UNaV but prevented the dipyridamole-induced exaggerated natriuresis to ANP. The infusion of adenosine deaminase into one renal artery reduced the natriuretic response to ANP. We conclude that elevated intrarenal levels of adenosine will exaggerate an ANP-induced natriuresis possibly by altering intracytosolic Ca2+.  相似文献

Mice pre-trained in an olfactometer were tested daily on odor detection and discrimination tasks after irrigation of their olfactory epithelium in each naris with 50 microl of 5% zinc sulfate or saline. Anterograde transport of a wheatgerm agglutinin-horseradish peroxidase (WGA-HRP) conjugate from the epithelium to the olfactory bulb was used to assess anatomical connectivity in these and in mice that were used only for histological analyses. One day after treatment, saline controls performed at high levels of accuracy in detecting vapor from solutions of 5-0.01% ethyl acetate and in an odor discrimination task but most ZnSO4-treated mice performed at chance for 5-30 days before showing recovery. Although dense WGA-HRP reaction product was found in the accessory olfactory bulb, there was little or no evidence for axonal transport to glomeruli of the main olfactory bulb in the first 4-8 days after treatment. These results demonstrate that intranasal application of ZnSO4 to mice produces a brief but essentially total disruption of functional connections from the olfactory epithelium to the main olfactory bulb and a corresponding transient anosmia.  相似文献

Image‐based modeling, and more precisely, Structure from Motion (SfM) and Multi‐View Stereo (MVS), is emerging as a flexible, self‐service, remote sensing tool for generating fine‐grained digital surface models (DSMs) in the Earth sciences and ecology. However, drone‐based SfM + MVS applications have developed at a rapid pace over the past decade and there are now many software options available for data processing. Consequently, understanding of reproducibility issues caused by variations in software choice and their influence on data quality is relatively poorly understood. This understanding is crucial for the development of SfM + MVS if it is to fulfill a role as a new quantitative remote sensing tool to inform management frameworks and species conservation schemes. To address this knowledge gap, a lightweight multirotor drone carrying a Ricoh GR II consumer‐grade camera was used to capture replicate, centimeter‐resolution image datasets of a temperate, intensively managed grassland ecosystem. These data allowed the exploration of method reproducibility and the impact of SfM + MVS software choice on derived vegetation canopy height measurement accuracy. The quality of DSM height measurements derived from four different, yet widely used SfM‐MVS software—Photoscan, Pix4D, 3DFlow Zephyr, and MICMAC, was compared with in situ data captured on the same day as image capture. We used both traditional agronomic techniques for measuring sward height, and a high accuracy and precision differential GPS survey to generate independent measurements of the underlying ground surface elevation. Using the same replicate image dataset (n = 3) as input, we demonstrate that there are 1.7, 2.0, and 2.5 cm differences in RMSE (excluding one outlier) between the outputs from different SfM + MVS software using High, Medium, and Low quality settings, respectively. Furthermore, we show that there can be a significant difference, although of small overall magnitude between replicate image datasets (n = 3) processed using the same SfM + MVS software, following the same workflow, with a variance in RMSE of up to 1.3, 1.5, and 2.7 cm (excluding one outlier) for “High,” “Medium,” and “Low” quality settings, respectively. We conclude that SfM + MVS software choice does matter, although the differences between products processed using “High” and “Medium” quality settings are of small overall magnitude.  相似文献

An exact knowledge of the subcutaneous layers in the different regions of the face and neck is important in several surgical disciplines. In the parotid region, a superficial musculoaponeurotic system (SMAS) has been described. The existence of a SMAS as a guiding structure for the surgeon in the other regions of the face and neck has been discussed but is controversial. Therefore, the authors investigated the development of the subcutaneous connective-tissue layers in the different facial regions and in the neck. They studied these regions in 22 human fetuses using the technique of plastination histology and in three newborn and three adult specimens using sheet plastination. In addition, they dissected the neck and face in 10 fresh adult cadavers to identify the SMAS as in the surgical situation. The results show that no SMAS could be detected in any facial regions other than the parotid region. In the parotid region, it is thick and attached to the parotid sheath. However, it becomes very thin, discontinuous, and undissectable in the cheek area. No SMAS can be found in the neck, in which the authors are the first to describe a fascia covering both sides of the platysma. This fascia has close topographical connections to the subcutaneous layers of the adjoining regions. On the basis of these findings, the surgical pathways have to be defined regionally in the face. A "platysma fascia" can be considered as a surgical landmark in the neck. Therefore, the authors conclude that it is not justified to generalize a SMAS as a surgical guiding structure.  相似文献

The objective of this study was to determine whether individuals of Lymnaea stagnalis naturally parasitised with trematode larvae had different thermal preferences from non-parasitised ones, and what was the influence of constant temperature on biology of the host and the parasite.

The snails under study were placed in and oblong thermal gradient and their preferences were recorded during 24 h. Non-parasitised individuals chose a rather high temperature (25 °C). In the group of parasitised snails, only individuals which released cercariae showed statistically significant differences in thermal preference than the non-larvae releasing ones. Parasitised snails kept in constant temperatures lived shorter than non-parasitised ones. At high temperatures, they released more cercariae per day than at low temperatures. Temperature had a great influence on the longevity and viability of cercariae.

The thermal behaviour of snails parasitised with trematode larvae is not a symptom of behavioural fever but probably can play a therapeutic role in the host.  相似文献

When the availability of sperm limits female reproductive success, competition for sperm, may be an important broker of sexual selection. This is because sperm limitation can increase the variance in female reproductive success, resulting in strong selection on females to compete for limited fertilization opportunities. Sperm limitation is probably common in broadcast-spawning marine invertebrates, making these excellent candidates for investigating scramble competition between broods of eggs and its consequences for female reproductive success. Here, we report our findings from a series of experiments that investigate egg competition in the sessile, broadcast-spawning polychaete Galeolaria caespitosa. We initially tested whether the order in which eggs encounter sperm affects their fertilization success at two ecologically relevant current regimes. We used a split-clutch-split--ejaculate technique to compare the fertilization success of eggs from individual females that had either first access (competition-free treatment) or second access (egg competition treatment) to a batch of sperm. We found that fertilization success depended on the order in which eggs accessed sperm; eggs that were assigned to the competition-free treatment exhibited significantly higher fertilization rates than those assigned to the egg competition treatment at both current speeds. In subsequent experiments we found that prior exposure of sperm to eggs significantly reduced both the quantity and quality of sperm available to fertilize a second clutch of eggs, resulting in reductions in fertilization success at high and low sperm concentrations. These findings suggest that female traits that increase the likelihood of sperm-egg interactions (e.g. egg size) will respond to selection imposed by egg competition.  相似文献

Recently, high-flexion knee implants have been developed to provide for a large range of motion (ROM>120°) after total knee arthroplasty (TKA). Since knee forces typically increase with larger flexion angles, it is commonly assumed that high-flexion knee implants are subjected to larger loads than conventional knee implants. However, most high-flexion studies do not consider thigh–calf contact which occurs during high-flexion activities such as squatting and kneeling. In this study, we hypothesized that thigh–calf contact reduces the knee forces during deep knee flexion as the tibio-femoral load shifts from occurring inside the knee towards the thigh–calf contact interface. Hence, the effect of thigh–calf contact on the knee loading was evaluated using a free body diagram and a finite element model and both the knee forces and polyethylene stresses were analyzed. Thigh–calf contact force characteristics from an earlier study were included and a squatting movement was simulated. In general, we found thigh–calf contact considerably reduced both the knee forces and polyethylene stresses during deep knee flexion. At maximal flexion (155°), the compressive knee force decreased from 4.89 to 2.90 times the bodyweight (BW) in case thigh–calf contact was included and the polyethylene contact stress at the tibial post decreased from 49.3 to 28.1 MPa. Additionally, there was a clear correlation between a subject's thigh and calf circumference and the force reduction at maximal flexion due to thigh–calf contact (R=0.89). The findings presented in this study can be used to optimize the mechanical behavior of high-flexion total knee arthroplasty designs.  相似文献

Toxoplasmosis is an anthropozoonotic widespread disease, caused by the coccidian protozoan parasite Toxoplasma gondii. Since there are no data regarding the genotoxicity of the parasite in vivo, this study was designed to evaluate the genotoxic potential of the toxoplasmosis on isogenic mice with normal diet or under dietary restriction and submitted to a treatment with sulfonamide (375 microg/kg per day). DNA damage was assessed in peripheral blood, liver and brain cells using the comet assay (tail moment). The results for leucocytes showed increases in the mean tail moment in mice under dietary restriction; in infected mice under normal diet; in infected, sulfonamide-treated mice under normal diet; in infected mice under dietary restriction and in infected sulfonamide-treated mice under dietary restriction. In liver and brain cells, no statistically significant difference was observed for the tail moment. These results indicated that dietary restriction and T. gondii were able to induce DNA damage in peripheral blood cells, as detected by the comet assay.  相似文献
