Jasmonate ZIM-domain (JAZ) proteins and DELLA proteins are key negative regulators of jasmonates (JAs) and gibberellin (GA) signaling, respectively. In this study, we found JA and GA synergistically promote fiber cell initiation. We characterized the cellular function of a JAZ protein (GhJAZ3), and a DELLA protein (GhSLR1) of cotton (Gossypium hirsutum). GhJAZ3 is specifically expressed in elongating fibers, while GhSLR1 is expressed in different tissues and at a relatively higher level in 3 DPA ovules. GhSLR1 and GhJAZ3 proteins are localized in the cell nucleus. Yeast two-hybrid analysis indicated that GhSLR1, GhJAZ3 and GhDEL65 could interact with each other, and GhSLR1 could also interact with GhBZR1. Overexpression of GhJAZ3 in Arabidopsis increased hypocotyl and root length, leaf trichome length, and plant height, but decreased the number of leaf trichome, while overexpression of GhSLR1 in Arabidopsis decreased hypocotyl length, leaf trichome length and density. Expression of several leaf trichome initiation determinators (GL3, GL2, TTG2 and MYB23) was down-regulated in GhJAZ3 or GhSLR1 transgenic Arabidopsis, while expression of the cell elongation related genes (EXP1, EXP8, EXPL2 and XTH4) was altered in the GhJAZ3 and GhSLR1 transgenic Arabidopsis. Taken together, these results demonstrate that GhJAZ3 and GhSLR1 function in jasmonate and gibberellin mediated epidermal cell differentiation and elongation.  相似文献

Brown cotton fibres are the most widely used naturally coloured raw materials for the eco‐friendly textile industry. Previous studies have indicated that brown fibre pigments belong to proanthocyanidins (PAs) or their derivatives, and fibre coloration is negatively associated with cotton productivity and fibre quality. To date, the molecular basis controlling the biosynthesis and accumulation of brown pigments in cotton fibres is largely unknown. In this study, based on expressional and transgenic analyses of cotton homologs of ArabidopsisPA regulator TRANSPARENT TESTA 2 (TT2) and fine‐mapping of the cotton dark‐brown fibre gene (Lc1), we show that a TT2 homolog, GhTT2‐3A, controls PA biosynthesis and brown pigmentation in cotton fibres. We observed that GhTT2‐3A activated GhbHLH130D, a homolog of ArabidopsisTT8, which in turn synergistically acted with GhTT2‐3A to activate downstream PA structural genes and PA synthesis and accumulation in cotton fibres. Furthermore, the up‐regulation of GhTT2‐3A in fibres at the secondary wall‐thickening stage resulted in brown mature fibres, and fibre quality and lint percentage were comparable to that of the white‐fibre control. The findings of this study reveal the regulatory mechanism controlling brown pigmentation in cotton fibres and demonstrate a promising biotechnological strategy to break the negative linkage between coloration and fibre quality and/or productivity.  相似文献

To verify current thresholds for Bollgard II® cotton in Australia, the impact of Helicoverpa spp. (Lepidoptera: Noctuidae) larvae on yield, development, and quality under various infestation intensities and durations, and stages of growth, was tested using small plot field experiments over two seasons. Infestation with up to 80 eggs m?1 of Helicoverpa armigera (Hübner) and Helicoverpa punctigera Wallengren showed that species, infestation level, and stage of growth had no significant effect on yields of seed‐cotton or lint and on maturity and fibre quality. The duration of infestation of white flowers with H. punctigera neonates (maximum of every day for up to 4 weeks) had no impact on the yield of seed‐cotton or lint, maturity, and fibre quality, but when 100% of flowers were infested (compared with 0 or 50%), seed‐cotton and lint yields were significantly reduced and maturity was delayed. Infestation with up to 18 medium H. armigera larvae m?1 at several plant stages did not significantly affect yields of seed‐cotton and lint, maturity, and fibre quality. A heliocide spray applied on a commercial farm at the current threshold resulted in a significantly higher lint yield, compared with a farm where no spray was applied. In conclusion, Bollgard II® cotton is highly resistant to Helicoverpa spp. infestation.  相似文献

Foliar stomatal movements are critical for regulating plant water loss and gas exchange. Elevated carbon dioxide (CO2) levels are known to induce stomatal closure. However, the current knowledge on CO2 signal transduction in stomatal guard cells is limited. Here we report metabolomic responses of Brassica napus guard cells to elevated CO2 using three hyphenated metabolomics platforms: gas chromatography‐mass spectrometry (MS); liquid chromatography (LC)‐multiple reaction monitoring‐MS; and ultra‐high‐performance LC‐quadrupole time‐of‐flight‐MS. A total of 358 metabolites from guard cells were quantified in a time‐course response to elevated CO2 level. Most metabolites increased under elevated CO2, showing the most significant differences at 10 min. In addition, reactive oxygen species production increased and stomatal aperture decreased with time. Major alterations in flavonoid, organic acid, sugar, fatty acid, phenylpropanoid and amino acid metabolic pathways indicated changes in both primary and specialized metabolic pathways in guard cells. Most interestingly, the jasmonic acid (JA) biosynthesis pathway was significantly altered in the course of elevated CO2 treatment. Together with results obtained from JA biosynthesis and signaling mutants as well as CO2 signaling mutants, we discovered that CO2‐induced stomatal closure is mediated by JA signaling.  相似文献
