野外用剥落法采集落叶松(Larixgmelini)根际土与非根际土,分析P浓度的变化,结果表明,落叶松根际土与非根际土全P浓度无明显差异,但根际土有效P却明显高于非根际土.12年生时根际土有效P增加12.6%,40年生时增加23.4%,表明落叶松根际对土壤中的P具有活化作用.落叶松根际土无机P各组分与非根际土亦有差异,表现出根际土OP低于非根际土,AlP、FeP、CaP和NH4ClP则高于非根际土的趋势.落叶松根际土的pH低于非根际土,但落叶松根际并未发生明显酸化.落叶松根际有效P与pH变化相关不显著.  相似文献

【背景】三七根际微生物群落特征与其土传根腐病害密切相关,而针对自然林下根腐三七的相关研究鲜见报道。【目的】比较分析自然林下与田间根腐三七根际土壤微生物群落的组成特征,结合土壤理化性质与酶活性分析,为三七根腐病害防治与仿野生栽培提供科学依据。【方法】采集自然林下与田间根腐三七根际土壤,利用高通量测序技术,分析土壤细菌与真菌群落的物种组成与多样性,并测定土壤理化性质和酶活性。【结果】自然林下与田间根腐三七根际土壤中细菌和真菌群落组成具有明显差异,自然林下根腐三七根际土壤中担子菌门(Basidiomycota)、酸杆菌门(Acidobacteria)和疣微菌门(Verrucomicrobia)的相对丰度较高,而田间根腐三七根际土壤中子囊菌门(Ascomycota)、变形菌门(Proteobacteria)和绿弯菌门(Chloroflexi)的相对丰度较高。在属分类水平,镰刀菌属(Fusarium)是自然林下根腐三七根际土壤中的优势菌群,相对丰度为17.30%,而癣囊腔菌属(Plectosphaerella)是田间根腐三七根际土壤中的优势菌群,相对丰度为22.55%;Candidatus Ba...  相似文献

用剥落分离法采集胡桃楸(Juglans mandshurica)、落叶松(Larix gmelinii)纯林及其混交林根际与非根际土壤并分析有效P含量特性. 结果表明, 落叶松纯林根际土有效P含量较其非根际土高出55.8%,而胡桃楸纯林根际土有效P含量较其纯林仅高出10.1%,表现出落叶松根系对根际P较强的活化能力.树种混交后,借助落叶松根系的作用使混交林中胡桃楸根际土有效P含量较其纯林高出45.2%. 通过P的吸附/解吸及无机P分级等方面,对落叶松根际土壤有效P含量较高的原因进行了分析.  相似文献

从海南热带植物园采集12种药用植物的根际土样,采用选择性分离方法,分离得到400株根际放线菌。使用5种活性筛选模型对分离菌株进行生物活性评价,154株放线菌在一个或多个活性筛选模型中显示为阳性,菌株初筛阳性率达38.5%;根据菌株形态特征并结合代谢产物的生物活性,从中挑选出28株菌进行16S rRNA基因序列分析,发现其分属于链霉菌属、诺卡氏菌属、小单孢菌属和野野村菌属。  相似文献

本文研究了两种熟化水稻土和两种新垦红壤植稻后根际中铁的形态转化。结果表明,水稻土植稻后根际中无定形铁、游离铁、络合态铁及铁的活化度均低于非根际;红壤中除络合态铁的根际内外分布趋势与水稻土相同外,其余均与水稻土相反。穆斯堡尔谱特征表明,根际中氧化铁的四极矩分裂较大,内磁场较低,即根际中氧化铁被活化。差热分析结果表明根际土的持水力强于非根际土。这对根际微生态系统的生态环境保护、物质流的调控及水分的保持和有效利用有重要作用。  相似文献

从10个植物根际土样中分离出315株根际微生物,然后采用泛酸测定平板初筛和摇瓶复筛的方法,从中筛选到7株D-泛酸高产菌株,其中从菜豆根际土壤分离到的菌株C21的D-泛酸产量最高,达到24.1mg/L。最后通过形态和生理生化特性以及16SrDNA鉴定,确定菌株C21为恶臭假单胞菌(Pseudomonas putida)。  相似文献

采用单一碳源回收菌群的方法与ERIC-PCR方法相结合,检测水稻(Oryza sativa L.)根际施用转基因微生物E4(Enterobacteria cloacae E4)后,其根际微生物的群落结构和多样性的变化,进而推测转基因微生物E4在田间施用的安全性。结果表明:转基因微生物E4施用到水稻根际后,水稻根际的代谢指纹图谱和。DNA指纹图谱都发生了改变,采用Sollthelxi blotting检测显示:E4成为根际的优势菌,这对植物生长有利,应该不会造成不利的影响。  相似文献

通过滇中红壤区域现实林分中不同林龄云南松根际与非根际土的pH分析和不同磷处理水平下培养的云南松幼苗根际pH的测定,探讨了云南松根际pH以及不同磷水平下云南松幼苗根际pH变化。结果表明,低磷环境下云南松根际土和非根际土的pH值在3.49~3.79之间;云南松根际土pH明显低于非根际土,二者之间的差异极显著,但在不同林龄组间差异不明显;云南松幼苗能使根系周围pH下降,随着供磷水平的降低,云南松幼苗根系使根际pH降低的能力加强。本试验对云南松的研究结果与对其它一些树种进行研究所取得的结果有所不同,根际酸化是云南松适应低磷环境的有效反应之一。  相似文献

旨在了解转拟南芥紫色酸性磷酸酶基因的磷高效转基因大豆AP15-1的种植对土壤微生物生态的影响。于春秋两季在网室种植转基因大豆AP15-1及其受体YC03-3,在大豆幼苗期、盛花期和成熟期,分别采集根际土和非根际土。平板菌落计数的实验结果显示:AP15-1和YC03-3根际土与非根际土中可培养细菌、真菌和放线菌的数量在不同的季节和不同的生长期是会发生一定的变化,但没有明显的规律;两者根际土间及根际与非根际土间,细菌、真菌和放线菌数量没有明显的差异。BIOLOG生态板实验对碳源代谢特征分析结果显示:同季AP15-1和YC03-3根际土中微生物总体代谢活性要强于非根际土,但AP15-1和YC03-3根际土间及根际与非根际土间,微生物碳代谢活性差异均不显著;并且AP15-1与YC03-3根际土微生物对四大类碳源的代谢也没有明显的偏好。对春秋季成熟期的BIOLOG数据进行生态学丰富度与多样性指数计算分析,也未发现AP15-1和YC03-3根际土及非根际土间存在显著性差异。主成份和聚类分析结果显示:相同季节和相同生长期,AP15-1和YC03-3根际间的差异小于根际与非根际间的差异;季节和大豆生长时期对根际微生物群落造成的影响要大于AP15-1与YC03-3个体差异所产生的影响。研究结果表明转酸性磷酸酶基因的磷高效转基因大豆种植对根际土壤可培养微生物的数量和群落结构没有产生显著的影响。  相似文献

A method is described for sampling rhizosphere soil under newly establishedPicea sitchensis andAcer pseudoplatanus. The technique involves taking soil samples to a depth of 150 mm at 100 mm intervals along transects, each 45° from its neighbour, radiating from the base of the stem. Invertase activities were measured in the soil samples and compared to their activities in fallow and rhizosphere soils. When the field soil was dry, the tree root systems were carefully excavated to retain as many fine roots as possible. The distribution of the soil invertase was matched to the spatial distribution of the roots showing the precise position of the rhizosphere relative to the initial blind soil sampling. Statistics were applied to derive equations for calculating the percentage enzyme activity relative to that found in rhizosphere soil at various locations radiating from the base of the stem. This information was subsequently applied to soil sampled under trees of the same age as those excavated to give a non-destructive method for sampling rhizosphere soil routinely from under a large number of trees.  相似文献

The influence of application of hexachlorocyclohexane (HCH=gamma BHC), to a submerged tropical field soil at rates equivalent to recommended field rates (1–2.5kg a.i./ha) and twice this level, upon the rhizosphere soil nitrogenase, nitrogen fixers, and soil redox potential (Eh) was investigated. The rhizosphere soil from HCH-treated field exhibited significantly higher nitrogenase activity than that from untreated fields. HCH retarded the drop in redox potential of the field soil upto 80 days after transplantation under submerged conditions. Populations of nitrogen-fixingAzospirillum sp. and Azotobacter, to a greater extent, and anaerobic organisms, to a lesser extent, were stimulated in HCH-treated soils. Results indicate the stimulation of heterotrophic nitrogen-fixing bacteria by HCH in submerged paddy soils.  相似文献

在棉麦两熟双高产条件下研究了棉花根际与非根际土壤酶活性和养分含量的变化.结果表明,套作棉土壤脲酶、蔗糖酶、蛋白酶及过氧化氢酶活性随生育进程的变化趋势与单作棉表现一致,但整个生育期套作棉根际与非根际土壤各种酶活性均明显高于单作棉.套作棉根际与非根际土壤养分含量在麦棉共生期低于单作棉或差异较小,而在麦收后则显著高于单作棉.套作棉土壤养分含量随生育进程的变化趋势与单作棉大体相同,但一些养分的吸收高峰晚于单作棉.无论套作棉还是单作棉,根际土壤酶活性和养分含量高于非根际.土壤各养分含量与土壤脲酶、蔗糖酶和蛋白酶活性呈显著(P=0.0,n=32)或极显著(P=0.01,n=32)相关,与土壤过氧化氢酶活性相关不显著.  相似文献

Summary The nitrification rate in the rhizosphere of Suaeda and Arthrocnemum plants growing in saline soils, as affected by microbial populations, temperature, pH, and organic matter, was examined in the field throughout the year. The genera Nitrosomonas and Nitrobacter were most common in the rhizosphere soil. The bacterial counts in the rhizosphere of both plants fluctuated during the study period, reaching peak values during February–March and in August. The nitrate concentration in the rhizosphere soil could be related with the observed increase in the numbers of ammonium-oxidizers and nitrite-oxidizers in the latter part of the study period. The pH of the rhizosphere soils did not have any influence on the nitrification rate at the values measured. The rhizosphere organic content varied between 1.8 and 4% (w/w), showing the continuous availability of organic matter in the soil. The seasonal changes in bacterial populations in the rhizospheres of both plants was described as the result of the combination of several factors.  相似文献

Abstract Transfer of plasmid RP4 between introduced pseudomonads was studied in rhizosphere and non-rhizosphere soil of wheat, in soil chambers and in culture tubes. In both experiments, the presence of growing wheat roots stimulated the occurrence of plasmid transfers in the soil. The plasmid transfer frequencies in rhizosphere soil in the soil chambers were consistently higher than those in rhizosphere soil in the culture tubes, indicating an influence of the experimental set-up.
In the soil chambers, both the survival of introduced donor and recipient strains and the plasmid transfer frequencies decreased drastically at increasing distances from the roots. In addition, plasmid transfer frequencies were influenced by the inoculum densities of both donor and recipient strains; higher frequencies were observed in soil that was initially inoculated with higher cell numbers.  相似文献

Summary From 36 to 71% of bacteria, depending on the sampling site, that were isolated from the soil or rhizosphere of undisturbed prairie soil or reclamation sites of strip-mined grassland areas in western North Dakota were capable of reducing acetylene. These bacteria generally could be divided into two populations; one capable of acetylene reduction under aerobic conditions and another capable of acetylene reduction under anaerobic conditions. The reclamation site to which no topsoil had been applied, pH 8.5, had a bacterial population which generally was capable of higher levels of acetylene reduction than individual bacteria isolated from other sites.  相似文献

【目的】高通量测序技术对研究环境样品中微生物群落组成具有很大的应用价值。土壤微生物群落结构和多样性及其变化在一定程度上反映了土壤的质量。旨在从微生物群落结构的角度阐述环保肥料增效剂对马铃薯根际土壤主要真菌类群结构的影响。【方法】通过高通量测序结果对比分析应用增效剂前后马铃薯根际真菌宏基因组ITS1区,并依据RDP中设置的分类阈值对序列进行物种分类。【结果】测序结果经过质量控制,共获得有效条带437 375条,依据97%的序列相似性做聚类分析,获得全部样品的可分类操作单元(OTUs)共633个。子囊菌的总体数量在所有样品中最多(相对丰度在56.95%-97.23%之间),且处理后呈增加趋势(HY除外),而担子菌门数量在处理后呈下降的趋势。基于真菌ITS1高通量测序结果获得的α指数显示,样品内部处理和对照之间真菌物种多样性有差别。基于真菌ITS1高通量测序获得的β指数显示,处理组与对照组的真菌多样性之间没有差别,这表明真菌多样性之间的差异更多地取决于样品采集地点。【结论】土壤特性是影响真菌种群的重要因素之一,环保肥料增效剂显著改善了土壤真菌的种群结构。  相似文献
