A simple, inexpensive design is presented for the rapid conversion of the popular MD-4 Polaroid land camera to a high quality digital gel documentation system. Images of ethidium bromide stained DNA gels captured using the digital system were compared to images captured on Polaroid instant film. Resolution and sensitivity were enhanced using the digital system. In addition to the low cost and superior image quality of the digital system, there is also the added convenience of real-time image viewing through the swivel LCD of the digital camera, wide flexibility of gel sizes, accurate automatic focusing, variable image resolution, and consistent ease of use and quality. Images can be directly imported to a computer by using the USB port on the digital camera, further enhancing the potential of the digital system for documentation, analysis, and archiving. The system is appropriate for use as a start-up gel documentation system and for routine gel analysis.  相似文献

Advances in digital photography have made it an efficient and economically appealing alternative to conventional photography. Nevertheless, as objective observers and clinical photographers, we must realize that all digital cameras are not created equal. Different digital cameras frequently used in plastic surgery practices (Olympus 600DL, Olympus 2500, Sony DSC-D700, Nikon Coolpix 950, and Nikon D1) were evaluated, using a subject photographed with each camera in the identical lighting conditions, to determine inherent differences in quality, color, and contrast of the resultant photographs. Three different lighting conditions were examined: single soft-box lighting, dual studio flash boxes, and operating room lighting with on-camera flash. The same digital settings (program mode, ISO camera default setting, high quality setting with JPEG compression) were used. Each camera was digitally color balanced using an 18 percent gray card. Raw and color-balanced images were viewed side-by-side. The macro-image capabilities of each camera were also examined. Conventional 35-mm photographs using a 105 macro-lens on Kodachrome and Ektachrome slide film were obtained for comparison. All of the digital cameras performed with noticeable differences, but they maintained consistency in the three different lighting conditions. Digital photographs differed most greatly with respect to quality and contrast, which was especially obvious once color balancing was performed. Marked differences in quality and ability were observed with respect to macro-image capabilities. Inherent differences in features among digital cameras produce dramatically different photographic results with regard to color, contrast, focus, and overall quality. With the increasing use of digital photography in plastic surgery journals and presentations, it must be recognized that digital cameras do not all display photographs of similar quality, especially when used to evaluate skin appearance. To standardize digital photography, the surgeon must realize that switching digital cameras is akin to switching film types. Standardization of digital photographs should include image resolution between 1.5 and 2.7 million pixels, ISO default setting, color balancing with an 18 percent gray card and software, consistency in focal distance, JPEG compression of medium-to-high quality, and backgrounds of medium blue or 18 percent gray.  相似文献

In search for a cost effective gel documentation system applicable for different fields of molecular biology, we analyzed the capabilities of a cheap CCD-camera originally designed to capture images for transmission through the internet (web-cam) with regard to gel documentation. The camera was connected to a personal computer by universal serial bus (USB) and used for the documentation of DNA separated on agarose gels and stained by ethidium-bromide using the software provided with the camera. The web-cam provided digital images of sufficient quality for routine documentation and combined the low set-up costs of a Polaroid system with the low running costs of video capture systems, hence is ideal as a start-up system and as augmentation to existing equipment.  相似文献

Digital photography has become available to everybody. The aim of this study was to examine possibility of calculating the width of a missing central incisor using digital photographs. Digital photographs were obtained from 51 dentate subjects using a 3.1 Megapixel digital camera from various distances: 35 cm, 70 cm, 1 m and 1.5 m. For the calculation of the width of maxillary left central incisor (MLI), the following equation was used: MLI(calculated) = Photographic width of MLI x IPD / photographic IPD. Statistical analysis was made (Kolmogorov-Smirnov test, dependent sample t-test, correlation and frequencies) using SPSS 10.0 for Windows. Results revealed no significant difference between the calculated MLI (70 cm, 1 m and 1.5 m distance) and actual MLI, however calculated MLI from 35 cm distance was significantly different from the actual MLI value (p < 0.01). The highest correlation was between calculated MLI (70 cm distance) and actual MLI. However, the highest percentage of results from a distance of 70 cm also fitted within +/- 0.3 mm; +/- 0.5 mm and +/- 1 mm difference from the actual MLI values. However, the results obtained from 1 m distance were also satisfactory. The technique of use of digital photography is of proven value in calculating the width of a maxillary central incisor. The photographs using a simple digital camera should be taken from a distance from 70 cm to 1 m. Saving such photographs in a dental office may eventually be helpful for calculating dimensions of artificial teeth.  相似文献

采用数码相机获取叶片的数字图像,用Photoshop图像处理软件计算叶面积,并与目前常用的剪纸法和叶面积仪测定法进行比较分析。结果表明,本方法和上述传统测定方法测定结果存在极显著线性相关;不同拍摄分辨率、单位叶面积存贮像素个数和拍摄角度对测定结果无显著影响。和其它方法相比,本方法具有准确、快速、成本低廉、适合非破坏性动态连续观测等优点,适用于植物生理生态学研究中叶面积的测定。  相似文献

Because of fears that Polaroid colour prints produced with a non-mydriatic fundus camera may not detect important sight threatening lesions in diabetes a study was conducted comparing retinal images obtained on Polaroid prints taken in “field” conditions with those on 35 mm transparencies and fluorescein angiograms. Almost one in five (22/127) Polaroid prints could not be assessed owing to poor quality compared with 3 (2.4%) 35 mm transparencies and 2 (1.6%) fluorescein angiograms. The pick up rate of microaneurysms, haemorrhages, and hard and soft (cotton wool spots) exudates was equivalent for Polaroid prints and 35 mm transparencies of equivalent quality. In two cases with disc new vessels, however, these were not seen on the Polaroid prints.The widespread use of Polaroid colour prints obtained with a non-mydriatic camera without the necessary operative and interpretive skills further limits the usefulness of the technique.  相似文献

The ability to carry out systematic, accurate and repeatable vegetation surveys is an essential part of long-term scientific studies into ecosystem biodiversity and functioning. However, current widely used traditional survey techniques such as destructive harvests, pin frame quadrats and visual cover estimates can be very time consuming and are prone to subjective variations. We investigated the use of digital image techniques as an alternative way of recording vegetation cover to plant functional type level on a peatland ecosystem. Using an established plant manipulation experimental site at Moor House NNR (an Environmental Change Network site), we compared visual cover estimates of peatland vegetation with cover estimates using digital image classification methods, from 0.5 m × 0.5 m field plots. Our results show that digital image classification of photographs taken with a standard digital camera can be used successfully to estimate dwarf-shrub and graminoid vegetation cover at a comparable level to field visual cover estimates, although the methods were less effective for lower plants such as mosses and lichens. Our study illustrates the novel application of digital image techniques to provide a new way of measuring and monitoring peatland vegetation to the plant functional group level, which is less vulnerable to surveyor bias than are visual field surveys. Furthermore, as such digital techniques are highly repeatable, we suggest that they have potential for use in long-term monitoring studies, at both plot and landscape scales.  相似文献

A method of visualizing patency of arteries and arterial grafts by means of intravenous 99mTc pertechnetate using a Nuclear Chicago Pho/Gamma HP camera is described. Polaroid pictures showing normal arteries in the upper and lower limbs, as well as partial and complete occlusion of arteries and the state of arterial bypass grafts in pre- and postoperative patients are compared with conventional arteriograms. Being concerned with physical anthropometric dimensions to explore avenues of research on hands and their working strategies, a research study was made to develop a suitable method by using available photogrammetric resources. The paper discusses the optimums of details found by this study, viz., the use of a Wild C-40 stereometric camera (for obtaining stereoscopic photographs) and the use of a Wild A7 stereorestitution instrument equipped with digital data acquisition accessories (for obtaining all dimensional information). Three-dimensional data at 21 specific points and for nine specific distances were obtained on each hand of ten persons. Test observations indicate, inter alia, a dimensional accuracy of the order of +/- 0.3 mm obtainable through the working system. Cartographic presentations made in support of the digital data are found to be of help. This also permits to obtain additional data as may be necessary at times. Photogrammetry has the advantage of reducing and maintaining to a minimum all sorts of manipulations necessary in such studies.  相似文献

The aim of the study was to determine the acceptance and perception of Nigerian patients to medical photography. A self‐administered questionnaire was distributed among Nigerian patients attending oral and maxillofacial surgery and plastic surgery clinics of 3 tertiary health institutions. Information requested included patients' opinion about consent process, capturing equipment, distribution and accessibility of medical photographs. The use of non‐identifiable medical photographs was more acceptable than identifiable to respondents for all purposes (P = 0.003). Most respondents were favourably disposed to photographs being taken for inclusion in the case note, but opposed to identifiable photographs being used for other purposes most especially in medical websites and medical journals. Female respondents preferred non‐identifiable medical photographs to identifiable ones (P = 0.001). Most respondents (78%) indicated that their consent be sought for each of the outline needs for medical photography. Half of the respondents indicated that identifiable photographs may have a negative effect on their persons; and the most commonly mentioned effects were social stigmatization, bad publicity and emotional/psychological effects. Most of the respondents preferred the use of hospital‐owned camera to personal camera/personal camera‐phone for their medical photographs. Most respondents (67.8%) indicated that they would like to be informed about the use of their photographs on every occasion, and 74% indicated that they would like to be informed of the specific journal in which their medical photographs are to be published. In conclusion, non‐identifiable rather than identifiable medical photography is acceptable to most patients in the studied Nigerian environment. The use of personal camera/personal camera‐phone should be discouraged as its acceptance by respondents is very low. Judicious use of medical photography is therefore advocated to avoid breach of principle of privacy and confidentiality in medical practice.  相似文献

Accuracy of determining radiation interception, and hence radiation use efficiency, depends on the method of measuring photosynthetically active radiation intercepted. Methods vary, from expensive instruments such as Sunfleck ceptometers to simple methods such as digital photography. However, before universal use of digital photography there is need to determine its reliability and compare it with conventional, but expensive, methods. In a series of experiments at Lincoln, New Zealand, canopy development for barley, wheat, white clover and four forage brassica species was determined using both digital photographs and Sunfleck ceptometer. Values obtained were used to calculate conversion coefficient (Kf/Ki) ratios between the two methods. Digital photographs were taken at 45° and 90° for barley, wheat and white clover and at only 90° for brassicas. There was an interaction of effects of crop and cultivar for the cereal crops. Barley closed canopies earlier than wheat, and ‘Emir’ barley and ‘Stettler’ wheat had consistently higher canopy cover than ‘Golden Promise’ and ‘HY459′, respectively. Canopy cover was consistently larger at 45° than 90° for cereals. However, for white clover, the angle of digital photography was not important. There was also an interaction between effects of species and method of determining canopy cover for brassicas. Photographs gave higher cover values than ceptometer for forage rape and turnip, but the relationship was variable for forage kale and swede. Kf/Ki ratios of 1.0–1.10 for cereals, white clover and forage rape and turnip show that digital photographs can be used to estimated radiation interception, in place of Sunfleck ceptometer, for these crops.  相似文献

利用多尺度遥感影像综合进行全球和区域尺度的土地利用/覆盖变化(LUCC)研究是最近全球变化研究的重要方向之一。本文综合利用野外群落样方、数字相机、ETM+影像、NOAA/AVHRR影像,在遥感、GIS和GPS支持下,对我国北方典型草原区植被盖度进行了综合监测、模拟与分析。结果表明:(1) 利用经处理后的数字相机影像测量盖度的结果准确性较高,可以作为植被盖度测量的标准结果,反映真实的覆盖特征,并用以验证利用其它方法测量结果的精度。(2) 利用野外1 m2样方网格法目视估测的植被盖度结果变化较大,不稳定。本次实验中,与数字相机测量结果相比,样方估测的盖度普遍偏高,平均偏差为9.92%;但两者相关性较好(r2=0.89)。(3) 采用Gutman模型ETM+影像、NOAA/AVHRR影像反演植被盖度的结果与数字相机测量结果偏差分别为7.03%、7.83%,ETM+像元分解NOAA像元后得到的植被盖度与数字相机测量结果偏差5.68%。三者与数字相机测量结果的相关系数r2分别为0.78、0.61和0.76。(4)利用野外实测植被盖度数据直接与NOAA-NDVI影像建立统计模型估算植被盖度的精度较低(r2=0.65),而通过空间分辨率介于两者之间的ETM+影像进行转换后,该精度得到一定的提高(r2=0.80)。利用像元分解的方法提高了大尺度植被盖度监测的精度,是利用遥感数据进行尺  相似文献

Animal coloration has provided many classical examples of both natural and sexual selection. Methods to study color signals range from human assessment to models of receiver vision, with objective measurements commonly involving spectrometry or digital photography. However, signal assessment by a receiver is not objective but linked to receiver perception. Here, we use standardized digital photographs of female rhesus macaque (Macaca mulatta) face and hindquarter regions, combined with estimates of the timing of the female fertile phase, to assess how color varies with respect to this timing. We compare objective color measures (camera sensor responses) with models of rhesus vision (retinal receptor stimulation and visual discriminability). Due to differences in spectral separation between camera sensors and rhesus receptors, camera measures overestimated color variation and underestimated luminance variation compared with rhesus macaques. Consequently, objective digital camera measurements can produce statistically significant relationships that are probably undetectable to rhesus macaques, and hence biologically irrelevant, while missing variation in the measure that may be relevant. Discrimination modeling provided results that were most meaningful (as they were directly related to receiver perception) and were easiest to relate to underlying physiology. Further, this gave new insight into the function of such signals, revealing perceptually salient signal luminance changes outside of the fertile phase that could potentially enhance paternity confusion. Our study demonstrates how, even for species with similar visual systems to humans, models of vision may provide more accurate and meaningful information on the form and function of visual signals than objective color measures do.  相似文献

Linear camera responses are required for recovering the total amount of incident irradiance, quantitative image analysis, spectral reconstruction from camera responses and characterisation of spectral sensitivity curves. Two commercially-available digital cameras equipped with Bayer filter arrays and sensitive to visible and near-UV radiation were characterised using biexponential and Bézier curves. Both methods successfully fitted the entire characteristic curve of the tested devices, allowing for an accurate recovery of linear camera responses, particularly those corresponding to the middle of the exposure range. Nevertheless the two methods differ in the nature of the required input parameters and the uncertainty associated with the recovered linear camera responses obtained at the extreme ends of the exposure range. Here we demonstrate the use of both methods for retrieving information about scene irradiance, describing and quantifying the uncertainty involved in the estimation of linear camera responses.  相似文献

It is of interest to evaluate the distortion of intraoral frontal photographs using various focal lengths. Intra-oral frontal view pictures of 20 subjects were taken using a standard 18-55mm lens of a DSLR camera at three different focal lengths (18mm, 35mm, 55mm). 52 experienced orthodontists were chosen and they were asked to assess the photographs and choose the one with the least amount of distortion. Majority of the orthodontists selected the frontal photograph taken at a focal length of 35mm as the best photo with the least distortion among the three photographs. The photos taken at a focal length of 35 mm showed the least amount of distortion.  相似文献

Assessing environmental impacts on coral reef communities has become a growing discipline. As most corals grow relatively slowly, the common method of monitoring changes in coral communities may limit our ability to identify stressors and stress responses. Since chlorophyll density (amount of chlorophyll a + c2 per unit of coral surface area) in corals may correlate with coral color, the latter has been suggested as an indicator of the natural seasonal changes in undisturbed (“healthy”) corals, as well as an indicator of environmentally-induced stresses in corals, including those related to bleaching. However, the color of underwater objects as perceived through the naked eye or a camera is affected by attenuation of irradiance with depth, changes in spectral properties of underwater light, turbidity and dissolved materials in the water, among other factors. Thus, methodological issues have so far held back the use of color as a quantitative indicator for chlorophyll content. Presented here are two accurate and easy to use methods for quantitative measurements of chlorophyll density from digital photographs of corals. In these methods, the intensities of the red, green and blue channels in digital photographs were compared to measured chlorophyll densities in Stylophora pistillata. Variations in external light, known to bias the true color in underwater images, were eliminated either by photographing corals through a specially built funnel with an internal light source or by mathematically normalizing color channels to a gray reference scale. In both methods, chlorophyll density was highly correlated with the intensity of the red channel, despite large variations in lighting conditions during the photography.These photographic methods enabled the estimation of natural spatial and temporal changes of the chlorophyll density of corals. By predicting chlorophyll density in corals at very low costs, both methods presented here could facilitate the study of large-scale physiological changes in corals.  相似文献

Static photographs are currently the most often employed stimuli in research on social perception. The method of photograph acquisition might affect the depicted subject’s facial appearance and thus also the impression of such stimuli. An important factor influencing the resulting photograph is focal length, as different focal lengths produce various levels of image distortion. Here we tested whether different focal lengths (50, 85, 105 mm) affect depicted shape and perception of female and male faces. We collected three portrait photographs of 45 (22 females, 23 males) participants under standardized conditions and camera setting varying only in the focal length. Subsequently, the three photographs from each individual were shown on screen in a randomized order using a 3-alternative forced-choice paradigm. The images were judged for attractiveness, dominance, and femininity/masculinity by 369 raters (193 females, 176 males). Facial width-to-height ratio (fWHR) was measured from each photograph and overall facial shape was analysed employing geometric morphometric methods (GMM). Our results showed that photographs taken with 50 mm focal length were rated as significantly less feminine/masculine, attractive, and dominant compared to the images taken with longer focal lengths. Further, shorter focal lengths produced faces with smaller fWHR. Subsequent GMM revealed focal length significantly affected overall facial shape of the photographed subjects. Thus methodology of photograph acquisition, focal length in this case, can significantly affect results of studies using photographic stimuli perhaps due to different levels of perspective distortion that influence shapes and proportions of morphological traits.  相似文献

OBJECTIVE--To determine whether non-mydriatic Polaroid retinal photography was comparable to ophthalmoscopy with mydriasis in routine clinic screening for early, treatable diabetic retinopathy. DESIGN--Prospective study of ophthalmoscopic findings according to retinal camera screening and ophthalmoscopy and outcome of referral to ophthalmologist. SETTING--Outpatient diabetic clinics of three teaching hospitals and three district general hospitals. PATIENTS--2159 Adults selected randomly from the diabetic clinics, excluding only those registered as blind or those in wheelchairs and unable to enter the screening vehicle. MAIN OUTCOME MEASURES--Numbers of patients and eyes correctly identified by each technique as requiring referral with potentially treatable retinopathy (new vessel formation and maculopathy) and congruence in numbers of microaneurysms, haemorrhages, and exudates reported. RESULTS--Camera screening missed two cases of new vessel formation and did not identify a further 12 but indicated a need for referral. Ophthalmoscopy missed five cases of new vessel formation and indicated a need for referral in another four for other reasons. Maculopathy was reported in 147 eyes with camera screening alone and 95 eyes by ophthalmoscopy only (chi 2 = 11.2; p less than 0.001), in 66 and 29 of which respectively maculopathy was subsequently confirmed. Overall, 38 eyes received laser treatment for maculopathy after detection by camera screening compared with 17 after ophthalmoscopic detection (chi 2 = 8.0; p less than 0.01). Camera screening underestimated numbers of microaneurysms (chi 2 = 12.9; p less than 0.001) and haemorrhages (chi 2 = 7.4; p less than 0.01) and ophthalmoscopy underestimated hard exudates (chi 2 = 48.2; p less than 0.001). CONCLUSIONS--Non-mydriatic Polaroid retinal photography is at least as good as ophthalmoscopy with mydriasis in routine diabetic clinics in identifying new vessel formation and absence of retinopathy and is significantly better in detecting exudative maculopathy.  相似文献

The use of camera traps is now widespread and their importance in wildlife studies is well understood. Camera trap studies can produce millions of photographs and there is a need for a software to help manage photographs efficiently. In this paper, we describe a software system that was built to successfully manage a large behavioral camera trap study that produced more than a million photographs. We describe the software architecture and the design decisions that shaped the evolution of the program over the study's three year period. The software system has the ability to automatically extract metadata from images, and add customized metadata to the images in a standardized format. The software system can be installed as a standalone application on popular operating systems. It is minimalistic, scalable and extendable so that it can be used by small teams or individual researchers for a broad variety of camera trap studies.  相似文献
