The ability ofCarnobacterium spp. originally isolated from vacuum-packed, sugar-salted fish to catabolize arginine was examined. All strains were able to produce citrulline, ornithine, and NH3 from arginine, presumably by the arginine deiminase pathway. The metabolism of arginine was concurrent with acid production from glucose for one strain ofCarnobacterium sp. but delayed for one strain ofCarnobacterium piscicola. The arginine catabolism was not inhibited in the presence of 2% glucose for three strains of carnobacteria during growth in test broth and/or shrimp extract. Growth as well as arginine catabolism was delayed for two strains of carnobacteria by lowering the temperature from 9°C to 4°C. A similar result was obtained by incubating one strain ofC. piscicola in CO2. None of the compoundsl-citrulline,l-ornithine hydrochloride, and (NH4)2SO4 had any effect on growth or arginine catabolism of this strain. Neither did pH of the medium affect the time for initiation of arginine catabolism.  相似文献

Summary Proline-producing strains of Serratia marcescens Sr41 were constructed by three rounds of mutagenesis. A strain SP103 which did not degrade l-proline carried the putA mutation leading to lack of proline oxidase. A 3,4-dehydroproline-resistant mutant SP105, derived from strain SP103, carried the dpr-1 mutation which resulted in desensitization of the feedback inhibition of glutamate kinase. Strain SP103 produced 5.5 mg of l-proline per ml of fermentation medium containing sucrose and urea. Growth inhibition by proline analogs was enhanced when succinate was used as a carbon source in the medium. A thiazolidine-4-carboxylate-resistant mutant SP126 derived from strain SP105 produced 20.5 mg of l-proline per ml of medium. The mutation carried by strain SP126 might be distant from dpr-1 and putA mutations on the chromosome. Pyrroline-5-carboxylate reductase was not repressed by proline in S. marcescens Sr41.  相似文献

The enzymatic profiles of 234 wild strains of thermophilic Campylobacters, seven type strains ofCampylobacter species, and 18 reference strains ofCampylobacter species and Campylobacter-like organisms were studied by use of API strips. These strips allow the detection of 56 arylamidases, one transpeptidase, and ten esterases.Forty enzymes were present at least once. The mean number of enzymes per strain was 13. The enzymatic activity was usually weak. Three enzymes were present in all the strains: esterases of butyric and valeric acids, andl-phenylalanine-l-proline arylamidase. A combination of three enzymes provided a good predictive value for the species differentiation ofC. jejuni andC. coli. There were no differences in relation to the geographical origin of the strain nor to the animal species from which it was isolated. The -glutamyl transpeptidase could be used for the biotyping of the strains.A portion of this work was presented at the Second Workshop of Campylobacter Infections, held in Brussels, Belgium, in September 1983.  相似文献

Summary In Serratia marcescens Sr41, l-canavanine was demonstrated to be a weak cell growth inhibitor in minimal medium containing glucose as the sole carbon source. The inhibition of cell growth was enhanced by changing the carbon source from glucose to l-glutamic acid. An arginine regulatory mutant (i.e., argR mutant) in which formation of l-arginine biosynthetic enzymes was genetically derepressed was isolated by selecting for l-canavanine resistance on the glutamate medium. Furthermore, an l-arginine-producing strain was constructed by introducing the mutation leading to feedback-resistant N-acetylglutamate synthase into the argR mutant. The resulting transductant produced about 40 g/l of l-arginine, whereas the wild strain produced no l-arginine and the argR mutant only 3 g/l.  相似文献

Excellent l-proline producers were screened for among sulfaguanidine resistant mutants derived from three typical l-glutamic acid-producing bacteria: Brevibacterium flavum, B. lactofermentum, and C. glutamicum.

The best strain, No. 199, is a sulfaguanidine resistant mutant derived from an isoleucine auxotroph of B. flavum 2247 by nitrosoguanidine. Strain No. 199 produced 35 mg/ml of l-proline after 72 hr of cultivation with 10% glucose as a carbon source. The strain also accumulated purine bases such as adenine, guanine, and hypoxanthine, i.e., degradation products of purine nucleotides. In the mutant, 1.6 ~ 2.0 fold more intracellular ATP was found than that in the parent strain; it is a substrate of glutamate kinase relating to l-proline biosynthesis.

On the contrary, the levels of intracellular glutamic acid, a substrate of glutamate kinase, were similar among these strains.

It was confirmed that the increment of internal ATP, which was important in the l-proline production mechanism, was very effective in the improvement of l-proline producers.  相似文献

The uptake ofl-[3H]arginine into synaptosomes prepared from rat cerebellum and cortex occurred by a high-affinity carrier-mediated process. The uptake of arginine appeared to be potentiated by removal of extracellular Na+, inhibited by high levels of extracellular K+, but not by depolarization with veratridine or 4-amino pyridine. The effect of Na+ removal or K+ elevation did not seem to be due to changes in intracellular Ca2+ or pH. In both brain regions, uptake was significantly inhibited byl-arginine,l-lysine,l-ornithine, andl-homoarginine, but not byd-arginine norl-citrulline. Uptake was also inhibited by NG-monomethyl-l-arginine acetate, but not by NG-nitro-l-arginine methyl ester nor NG-nitro-l-arginine except in the cortex at a concentration of 1 mM. The results indicate that the carrier system operating in synaptosomes showed many of the characteristics of the ubiquitous y+ system seen in many other tissues, although its apparent sensitivity to variations in extracellular Na+ was unusual.  相似文献

Seven clinical isolates and the ATCC strain 35405 ofTreponema denticola, obtained from human periodontal pockets, were studied for peptidase activity with several chromogenic compounds as substrates. The cell sonicates of all strains hydrolyzed phenylazobenzyloxycarbonyl-l-prolyl-l-leucyl-glycyl-l-prolyl-d-arginine (a collagenase substrate), azocasein, and the 2-naphthylamines ofl-proline,l-hydroxyproline,l-pyrrolidine, and benzoyl-l-arginine, but the rates of hydrolysis varied considerably from strain to strain. Fast protein liquid chromatography on gel and anion exchange columns revealed further biochemical differences between the strains. The ATCC strain consistently produced several proline iminopeptidases, whereas four of the clinical isolates yielded high and three yielded low iminopeptidase activity. The ATCC strain and six clinical isolates displayed high benzoylarginine peptidase activity. The use ofN-l-prolyl-2-naphthylamine as substrate revealed more differences between the strains than other substrates. The substrate specificity of the enzymes discovered suggests that they may be important for the nutrition of the organism or in the protection of the organism against chemical defense factors present in the gingival pocket.  相似文献

Feedback inhibition of crude and purified extracts of homoserine dehydrogenase and threonine deaminase activities in the genusBifidobacterium was studied. Homoserine dehydrogenase was partially or completely inhibited byl-threonine, and a marked inhibitory effect byl-isoleucine on threonine deaminase was observed. In the speciesBifidobacterium cuniculi high levels ofl-valine reversed the inhibitory effect ofl-isoleucine. The -aminobutyric acid-resistant mutant Ru 326/106 of the speciesB. ruminale, overproducer ofl-isoleucine, had a derepressed homoserine dehydrogenase and a lesser feedback inhibition byl-threonine. Homoserine dehydrogenase appeared to be in bifids specifically NAD dependent. The regulatory mechanisms of aspartate family amino acid biosynthesis in bifidobacteria was discussed.  相似文献

l-Arginine is produced by bacterial fermentation and is consumed in food flavoring and pharmaceutical industries. A better understanding of arginine metabolism in bacteria could be beneficial for a rational design of recombinant l-arginine producers by genetic engineering. This mini-review illustrated the current status of genes and enzymes for arginine metabolism, including biosynthetic pathways, catabolic pathways, uptake and excretion systems, and regulation. The linkage of polyamine and glutamate metabolism to the arginine network was also discussed, followed by a perspective view on how to construct arginine overproducing strains of bacteria with increasing biosynthesis and excretion and decreasing catabolism and uptake.  相似文献

Staphylococcus saprophyticus strains ATCC 15305, ATCC 35552, and ATCC 49907 were found to require l-proline but not l-arginine for growth in a defined culture medium. All three strains could utilize l-ornithine as a proline source and contained l-ornithine aminotransferase and Δ1-pyrroline-5-carboxylate reductase activities; strains ATCC 35552 and ATCC 49907 could use l-arginine as a proline source and had l-arginase activity. The proline requirement also could be met by l-prolinamide, l-proline methyl ester, and the dipeptides l-alanyl-l-proline and l-leucyl-l-proline. The bacteria exhibited l-proline degradative activity as measured by the formation of Δ1-pyrroline-5-carboxylate. The specific activity of proline degradation was not affected by addition of l-proline or NaCl but was highest in strain ATCC 49907 after growth in Mueller–Hinton broth. A membrane fraction from this strain had l-proline dehydrogenase activity as detected both by reaction of Δ1-pyrroline-5-carboxylate with 2-aminobenzaldehyde (0.79 nmol min−1 mg−1) and by the proline-dependent reduction of p-iodonitrotetrazolium (20.1 nmol min−1 mg−1). A soluble fraction from this strain had Δ1-pyrroline-5-carboxylate dehydrogenase activity (88.8 nmol min−1 mg−1) as determined by the NAD+-dependent oxidation of dl1-pyrroline-5-carboxylate. Addition of l-proline to several culture media did not increase the growth rate or final yield of bacteria but did stimulate growth during osmotic stress. When grown with l-ornithine as the proline source, S. saprophyticus was most susceptible to the proline analogues L-azetidine-2-carboylate, 3,4-dehydro-dl-proline, dl-thiazolidine-2-carboxylate, and l-thiazolidine-4-carboxylate. These results indicate that proline uptake and metabolism may be a potential target of antimicrobial therapy for this organism.  相似文献

Corynebacterium acetoacidophilum RYU3161 was cultivated in al-histidine-limited fed-batch culture. To investigate the effect of cell growth on thel-proline production, 5l fed-batch culture was performed using an exponential feeding rate to obtain the specific growth rates (μ) of 0.04, 0.06, 0.08, and 0.1 h−1. The results show that the highest production ofl-proline was obtained at μ=0.04 h−1. The specificl-proline production rate (Qp) increased proportionally as a function of the specific growth rate, but decreased after it revealed the maximum value at μ=0.08 h−1. Thus, the highest productivity ofl-proline was 1.66 g L−1 h−1 at μ=0.08 h−1. The results show that the production of L-proline inC. acetoacidophilum RYU3161 has mixed growth-associated characteristics.  相似文献

Summary The Na-dependent transport of a number of organic molecules (d-glucose,l-proline,l-alanine,l-phenylalanine) in brush-border membrane vesicles isolated from the intestine of the eel (Anguilla anguilla) was monitored by recording the fluorescence quenching of the voltage-sensitive cyanine dye 3,3-diethylthiacarbocyanine iodide (DiS-C2(5)). The experimental approach consisted of: a) generating an inside-negative membrane potential mimicking in vivo conditions: b) measuring the rate of membrane potential decay (i.e., the rate of fluorescence quenching decay) due to Na-neutral substrate cotransport. Rates of membrane potential decay showed saturation on substrate concentration andK app values (the substrate concentration giving 50% of the maximal rate) were estimated for Na-dependent transport ofd-glucose (0,099mm),l-alanine (0.516mm),l-proline (0.118mm) andl-phenylalanine (2.04mm). The influence of an inside-negative membrane potential on the affinity of the transporter for glucose and for sodium is discussed.  相似文献

The metabolism ofl-proline toN-acetyl-d-glucosamine (GlcNAc) during germ tube formation ofCandida albicans (C. albicans) ATCC 1002 was studied. In uptake experiments, 6.9 nmol ofl-[14C]proline were taken up by 1×106 cells during 3 h of incubation at 37°C. The percentage of germ tube formation was 94 under the same condition. The presence of GlcNAc reduced the uptake ofl-proline to 3.0 nmol. The percentage of germ tube formation was 95 in the presence and absence of GlcNAc. The [3H]GlcNAc uptake was 3.0 nmol and was constant whetherl-proline was present or not. After the preparation of a chitin fraction from germ tubes that were labeled withl-[14C]proline, the radioactivity froml-proline was detected in the glucosamine (GlcN) fraction by thin-layer chromatography (TLC). The metabolism ofl-proline to GlcNAc in chitin during germ tube formation was confirmed in this experiment.  相似文献

Two auxotrophic mutants of Corynebacterium glutamicum were found to produce a large amount of l-proline in the culture medium. High concentration of MgSO4 or MnSO4 in the medium stimulated the l-proline production by an isoleucine auxotroph. Optimum concentration of l-isoleucine was 200 μg/ml, and the higher concentration of l-isoleucine reduced the l-proline production. The auxotroph produced 14.8 mg/ml of l-proline when cultured in a medium containing 12% glucose, 1.7% NH4C1,0.6% MgSO4·7H2O, 0.06% MnSO4·4H2O and 200 μg/ml of l-isoleucine. The other mutant, whose growth responds to the bases of nucleic acids, produced 7 to 13 mg/ml of l-proline in a cane molasses (15%, as glucose concentration)-medium containing 2% of the acid-hydrolyzate of soybean meal. The l-proline production by this mutant increased to a level of 27 to 31 mg/ml when the growth was suppressed by the addition of 4% NH4C1 to the medium, or by the addition of 2 mg/ml of polyoxyethylenestearylamine, a surfactant, to a culture at an appropriate stage of the fermentation.  相似文献

Summary Aminopterin (10 g/ml) was found to inhibit the formation of 5-hydroxymethylcytosine (HMC), a constituent of mildiomycin, without affecting the growth ofStreptoverticillium rimofaciens. This was available for selecting high-producing mutants.d-Cycloserine caused its morphological mutations at high frequency. In addition, mildiomycin (MIL) production varied widely among the strains picked up from colonies that developed on agar medium containing cycloserine at the inhibitory concentration to the growth. Consequently, we selected the mutants which were capable of producing MIL on agar medium containing 10 g/ml of aminopterin, among mutants enriched by cycloserine. A high-producing mutant thus obtained, C R 4 -257, exhibited higher enzymatic activity of the HMC formation and higher resistance todl-serine hydroxamate than the original strain.l-Canavanine resistant mutants were furthermore selected to enhance the biosynthetic activity of the arginine-like moiety of MIL. Among them, we finally obtained an excellent mutant, CVR-48, with an MIL production 2.6 times that of the original strain,S. rimofaciens B-98891.  相似文献

Dye-linked l-proline dehydrogenase catalyzes the oxidation of l-proline in the presence of artificial electron acceptors such as 2, 6-dichloroindophenol and ferricyanide. The enzyme from the hyperthermophilic archaeon Thermococcus profundus was purified and characterized for the first time in archaea by Sakuraba et al. in 2001. In this study, cloning and sequencing analyses of the gene encoding the enzyme and functional analysis of the subunits were performed. The gene formed an operon that consisted of four genes, pdhA, pdhB, pdhF, and pdhX, which are tandemly arranged in the order of pdhA-F-X-B. SDS-PAGE analysis of the purified recombinant enzyme showed four different bands corresponding to (54 kDa), (43 kDa), (19 kDa), and (8 kDa) subunits encoded by pdhA, pdhB, pdhF, and pdhX, respectively, and the molecular ratio of these subunits was determined to be equal. This indicates that the enzyme consists of a heterotetrameric structure. Functional analysis of each subunit revealed that the subunit catalyzed the dye-linked l-proline dehydrogenase reaction by itself and that, unexpectedly, the subunit exhibited dye-linked NADH dehydrogenase activity. This is the first example showing the existence of a bifunctional dye-linked l-proline/NADH dehydrogenase complex. On the basis of genome analysis, similar gene clusters were observed in the genomes of Pyrococcus horikoshii, Pyrococcus abyssi, Pyrococcus furiosus, and Archaeoglobus fulgidus. These results indicate that the dye-linked l-proline dehydrogenase is a novel type of heterotetrameric amino acid dehydrogenase that might be widely distributed in the hyperthermophilic archaeal strain.Communicated by K. The concentration dependence of the influx ofl-lysine in excised roots ofArabidopsis thaliana seedlings was analyzed for the wild-type (WT) and two mutants,rlt11 andraec1, which had been selected as resistant to lysine plus threonine, and to S-2-aminoethyl-l-cysteine, respectively. In the WT three components were resolved: (i) a high-affinity, low-capacity component [K m = 2.2 M;V max = 23 nmol·(g FW)–1·h–1]; (ii) a low-affinity, high-capacity component [K m = 159 M;V max = 742 nmol·(g FW)–1·h–1]; (iii) a component which is proportional to the external concentration, with a constant of proportionalityk = 104 nmol·(g FW)–1 h–1];·mM–1. The influx ofl-lysine in the mutants was lower than in the WT, notably in the concentration range 0.1–0.4 mM, where it was only 7% of that in the WT. In both mutants the reduced influx could be fully attributed to the absence of the low-affinity (high-K m ) component. This component most likely represents the activity of a specific basic-amino-acid transporter, since it was inhibited by several other basic amino acids (arginine, ornithine, hydroxylysine, aminoethylcysteine) but not byl-valine. The high-affinity uptake ofl-lysine may be due to the activity of at least two general amino acid transporters, as it was inhibitable byl-valine, and could be further dissected into two components with a high affinity (K i = 1–5 M; and a low affinity (K i = 0.5–1mM) forl-valine, respectively. Growth ofSelenomonas ruminantium HD4 in medium that contained 21mm d-lactate was stimulated to varying degrees by 10mm l-malate, 10mm fumarate, and 2% (v/v)Aspergillus oryzae fermentation extract (Amaferm). Amaferm treatment caused the greatest growth stimulation. Initial uptake rates (30s) and long-term uptake rates (30 min) ofd-lactate by whole cells ofS. ruminantium were increased in the presence of 10mm l-malate. Amaferm (25 l/ml) also stimulated long-term uptake rates ofd-lactate, whereas fumarate had no effect. Initial uptake ofd-lactate was depressed in the presence of fumarate or Amaferm. When eitherl-malate, fumarate, or Amaferm was included in thed-lactate growth medium, a homosuccinate fermentation resulted and an inverse relationship was observed between growth (protein synthesis) and succinate production. Recent research demonstrated that Amaferm containsl-malate, and this dicarboxylic acid may be involved in stimulatingd-lactate utilization byS. ruminantium.  相似文献

Trans-4-hydroxy-l-proline has been converted to four imino- and carboxyl-blocked derivatives which are suitable for the synthesis of 4-O-glycosyl conjugates. Reaction of these derivatives with 2,3,5-tri-O-benzyl--l-arabinofuranosyl chloride in the presence of a silver zeolite promoter yielded the blocked -furanosyl amino-acid conjugates. Summary A new process (Living Cell Reaction Process) forl-isoleucine production using viable, non-growing cells ofBrevibacterium flavum AB-07 was optimised using ethanol as the energy source and -ketobutyric acid (-KB) as precursor.l-valine also could be produced from glucose at high yield by this process. This process differs from the usual fermentation method in that non-growing cells are used, and the production ofl-isoleucine andl-valine were carried out under conditions of repressed cell division and growth. Minimal medium missing the essential growth factor, biotin was employed as the reaction mixture for the production ofl-isoleucine andl-valine. The productivity ofl-isoleucine andl-valine were 200 mmol·l–1 · day–1 (molecular yield to -KB: 95%) and 300 mmol · l–1 · day–1 (molecular yield to glucose: 80%) respectively. The content ofl-isoleucine andl-valine in total amino acids produced in the each mixture were 97% and 96% respectively.  相似文献
