The flagellar apparatus of Urospora penicilliformis (Roth) Aresch. is unique, or at least very unusual among green algae. The flagellar axonemes are rigid, and contain wing-like projections. There are no central microtubules in the most proximal part of the axoneme. The transition region contains a series of electron dense transverse lamellae rather than a single septum, and lacks a stellate pattern. There is no cartwheel pattern in the proximal part of the basal bodies. The latter are associated with four different types of fibrous elements: ascending striated fibers that attach to an electron dense plate in the papillar center, lateral striated fibers that parallel microtubular roots, fibrous elements that link adjacent basal bodies, and finally two massive striated fibers that descend into the cell, passing closely along the nucleus (system II fibers, or rhizoplasts). Each of the four microtubular flagellar roots is sandwiched between two system I striated structures. The roots are probably equal; they contain proximally four, and distally up to eight microtubules. Based on the zoospore flagellar apparatus, it is concluded that the multinucleate U. penicilliformis is related to the Ulvaphyceae. Finally, a possible explanation in functional terms is given for the peculiar external morphology and behavior of the zoospore.  相似文献

A Feulgen microspectrophotometric analysis of successive generations of Fritschiella tuberosa Iyengar revealed an asexual repeating of one generation rather than an obligate alternation of isomorphic generations as previously described. F. tuberosa regularly reproduces asexually by the release of one quadriflagellate zoospore produced/cell in all parts of the upright and prostrate portions of the thallus. In the vegetative thalli, nuclei in the G1 (pre-synthesis), synthesis, and G2 (post-synthesis) phases of the mitotic cycle are all well-represented.  相似文献

The position of the rare and monotypic genus Karimbolea is clarified. Its tribal affinities are examined using mainly floral characters; the former tentative placement in the tribe Asclepiadeae (Asclepiadaceae) is confirmed. Characters of vegetative and floral morphology are used to determine the position of the genus within the tribe Asclepiadeae. Sarcostemma is identified as the most likely sister taxon of Karimbolea. Triterpenoid data obtained from gas-liquid chromatographic analysis of latex are used to test this hypothesis. This paper forms the first study within the Asclepiadaceae using two independent data sets to clarify the position of a taxon of uncertain affinities.  相似文献

A comparative study of the distribution, habitat, morphology and cytology of diploid (n = 10) and tetraploid races of Z. juniperifolia of the subgenus Diplothrix (COMPOSITAE-Heliantheae) was carried out to elucidate their relationships. There is a positive correlation between chromosome number and many floral and vegetative characters, including pollen size. Chiasmata frequency is about 0.63 and 0.62 per chromosome in the diploid and in the tetraploid respectively. Multivalents are common in the latter. Although attempts to obtain colchicine-induced tetraploids have failed, available morphological, cytological and chromatographic evidence indicate the tetraploid may be of autoploid origin. Alternative ancestries involving other species of the subgenus are discussed. Taxonomically the tetraploid is considered a race of Z. juniperifolia.  相似文献

Atractomorpha echinata gen. et sp. nov. is described from isolates derived from zygotes present in a dry soil sample obtained from Texas. The new genus is distinguished from Sphaeroplea primarily by its pattern of vegetative growth. While Sphaeroplea is distinctly filamentous with numerous coenocytic cells uniseriately arranged, Atractomorpha grows as individual, multinucleate, spindle-shaped cells with sharply pointed extremities. Such cells may vary considerably in length (25–6000 μm, or more) and normally lack septa. In young, rapidly growing cultures the cells often attain lengths of 300–500 μm, but rarely exceed 1800 μm. The new species is further characterized by: (1) the regular formation of biflagellate zoospores in asexual reproduction, (2)anisogamy (occasionally oogamy) and (3) the size and ornamentation of its zygotes. Variations in vegetative morphology are discussed as are conditions for obtaining gametogenesis.  相似文献

Division and plastic remodelling of the highly differentiated chromatophore inStigeoclonium stagnatile (Hazen)Collins is followed in living cells during their life cycle. In contradistinction to the unicellular algae both processes are separated: During the cell division, when the cell is growing, the highly differentiated chromatophore is simply divided without plastic remodelling and its division is finished before the nuclear division starts. The mode of the chloroplast division is identical with that of other algae. In contrast, during zoospore formation plastic remodelling of the chromatophore takes place: The lobed gutter-shaped chromatophore is transformed into a cup-like one which is adapted to the shape of the zoospore. After the zoospore has changed into a germling the cupshaped chromatophore is turned again into the original lobed gutter-like form of the vegetative cells. The precursory chromatophore division with regard to mitosis as well as the uniform mode of chromatophore division in various algae is stressed.

Complete nuclear-encoded small-subunit ribosomal RNA (18S rRNA) coding sequences were determined for the coccoid green algae Dictyochloropsis reticulata (Tschermak-Woess) Tschermak-Woess , Myrmecia astigmatica Vinatzer, and M. bisecta Reisigl, to investigate the taxonomic position of Dictyochloropsis Geitler and of the genus Myrmecia Printz. Phylogenies inferred from these data revealed a sister-group relationship between D. reticulata and certain coccoid green algae that lack motile stages (autosporic coccoids) within the order Microthamniales. The monophyletic origin of the Microthamniales, including autosporic coccoids previously classified in the Chlorophyceae, is clearly resolved by the rRNA sequence data. This finding. shows the considerable taxonomic breadth of that order, whose taxonomic position has been unclear so far. A new class, Trebouxiophyceae, is proposed for this group of green algae. Phylogenetic inferences from the rRNA sequences show paraphyly of the genus Myrmecia. The 18S rRNA sequence data suggest that, among taxa that share similar vegetative cell morphologies, the zoospore characters resolve better the actual genus and species boundaries. Within identical zoospore types, the rRNA data allow further resolution of taxonomic relationships. On the basis of the.se findings, I propose that the genus Friedmannia Chantanachat ± Bold be merged into Myrmecia and that only those species be left in the genus Myrmecia that are identical in particular zoospore characters (i.e. those described in detail for M. israeliensis ( Chantanachat ± Bold) comb, nov.), namely M. astigmatica, M. biatorellae (Tschermak-Woess ± Ptesst) Petersen, and M. israeliensis. Myrmecia bisecta has to be excluded from Myrmecia; its taxonomic position within the Trebouxiophyceae is unclear .  相似文献

Müller 1773 (Volvocales). From a study of clonal isolates, it has been possible to distinguish 2 distinct species, G. sociale (Dujardin) Warming 1876 and G. sacculiferum Scherffel 1904. The former may be divided further, on the basis of vegetative morphology and sexual reproduction, into 2 varieties: var. sociale and var. sacculum var. nov., and a possible third variety, var. majus Hansgirg 1888. A brief discussion of morphology and sexual reproduction (reported for the first time in G. sociale var. sacculum and G. sacculijerum) is presented as is a key to the species and varieties.  相似文献

The oomycete (Peronosporomycete) Chlamydomyzium oviparasiticum, previously recorded as a parasite of rotifer eggs, was found infecting Rhabditis nematodes in a sample of rotting garden compost. For the first time C. oviparasiticum was cultured in liquid media, which enabled more detailed studies of zoospore behaviour and facilitated the use of confocal microscopy. Rhabditis nematodes were successfully re-infected from liquid-cultured inoculum. Light (including video) microscopy and transmission electron microscopy were used to document details of thallus development, zoospore release and resting spore morphology to enable comparison with other oomycete species. This species showed several significant saprolegnialian characters such as the ‘achlyoid’ pattern of spore formation, centrifugal cleavage and structured encystment vesicles. In contrast, spore release into a transient vesicle was a peronosporalean characteristic. The thick-walled resting spores showed relatively poor cytoplasmic preservation and had a thick multi-layered wall. It was still not possible to unequivocably decide whether these were chlamydospores or parthenogenically formed oospores. The phylogenetic significance of these observations is discussed.  相似文献

Cell walls of Chlorococcum oleofadens Trainor & Bold were examined ultrastructurally and chemically. The wall of zoospores has a uniform 30 nm width and a regular lamellar pattern. Zoospores and young vegetative cell walk exhibit periodicities, consisting of 20 nm ridges on the outer layer. Vegetative cell walls have a variable thickness of Up to 800 nm and are composed of multiple layers of electron dense material. Further, vegetative walk contain a microfibrillar material composed predominantly of glucose and presumed to be cellulose. Except for this cellulose, vegetative cell wall chemistry is very similar to that of Chlamydomemas being composed of glycoprotein rich in hydroxyproline. The hydroxyproline in Chlorococcum walls is linked glycosidically to a mixture of hetrooligosaccharides composed of arabinose and galactose, and in one instance, an unknown 6-deoxyhexose. Altogether, the glycoprotein complex accounts for at least 52% of the wall. The amino acid composition of the walls is stikingly similar to those of widely different plant species. Indirect evidence indicates zoospore cell walls are also chemically similar to those of Chlamydomonas, and like them, are cellulose free. Thus a major chemical difference between zoospore and vegetative cell walk of Chlorococcum is the presence of cellulose in the latter. The contribution of this microfibrillar cellulose to the physical properties of the vegetative wall is discussed.  相似文献

A partially petrified impression of Triloboxylon ashlandicum (Aneurophytales) is the first recognized fertile axis of the genus. Identification of the fertile axis rests on the similarity of its anatomy with that of previously described vegetative specimens. Fertile organs replace some vegetative branches along part of the main axis. Fertile organs are twice dichotomized in one plane and bear elongate sporangia arranged pinnately. Vegetative branches differ in that they bear the ultimate appendages of the plant helically. The latter organs dichotomize many times in one plane. Although similar in size and morphology to the ultimate appendages, the fertile organs are homologous by position and vascular supply to the vegetative branches which they replace. Sporangia of T. ashlandicum dehisce longitudinally and terminate in an apiculate tip. Spores are unknown. Fertile organs of T. ashlandicum resemble those of other Aneurophytales and support the earlier placement of Triloboxylon in the order on anatomical grounds. T. ashlandicum differs from other Aneurophytales, however, by bearing vegetative organs at the distal end of its fertile axis.  相似文献

Application and availability of real-time polymerase chain reaction (PCR) assay to detect and quantify the Noctiluca scintillans zoospore were investigated seasonally. Specific primer set for N. scintillans 18S rDNA was designed and applied to real-time PCR assay using the serial dilutions of N. scintillans zoospores. The real-time PCR assays with Ns63F and Ns260R primers were applied to sea water samples collected weekly in Manazuru Port of Sagami Bay, Japan from April 2005 to June 2006. We developed effective DNA preparation steps for collecting the template DNA of N. scintillans zoospore: size fraction and filter concentration of the water samples, fixation with Lugol solution, cell lysis, and purification. This method is useful for the monitoring of the zoospores of N. scintillans, and can also be used for other small and physiologically fragile planktonic cell. Variation in the density of zoospore was successfully detected in the field samples. The peak density of N. scintillans zoospore was observed to occur just before or at the same time as the peak of the vegetative cells. Moreover, zoospores were detected in seawater even when the vegetative cells were not observed. The presence of zoospore was found all year round in the present study. In this regards, this information is essential for the study of the life cycle and seasonal variation of N. scintillans in the coastal waters.  相似文献

The fine structure of the pyrenoid in the mature vegetative cell of Tetracystis excentrica Brown and Bold is described. During zoosporogenesis, the pyrenoid undergoes regression, and the ultrastructure of this process is described in detail. The ground substance undergoes dissolution, and reticulate fibrillar structures appear as well as intruding chloroplast thylakoids. Pyrenoid-associated starch plates diminish, and quantities of starch not associated with the pyrenoid are produced. New pyrenoids appear late in the division cycle after all other major organelles associated with the motile cell have been formed. Zoospore pyrenoids develop in thylakoid-free spaces of the chloroplast which are similar to the DNA-containing regions. The new pyrenoid ground substance, which is loosely fibrillar, arises in close proximity to starch grains which may be formed in the stroma. Then the zoospore pyrenoid produces 2 hemispherical starch plates identical to those in the mature vegetative cell. Zoospore pyrenoids lack the 2 convoluted thylakoids between the starch plates and the ground substance characteristic of those in the mature vegetative cell. Instead, the thylakoids are identical to those of the chloroplast at first, and then develop into a convoluted state in the vegetative cell. Cytochemical tests for DNA, RNA, and protein were made for the cytoplasm, nucleus, nucleolus, and pyrenoid. Conclusive evidence is presented for the presence of RNA in the cytoplasm and nucleolus, DNA in the nucleus, and protein in the pyrenoid. The tests did not conclusively demonstrate the presence or absence of DNA and RNA in the pyrenoid; however, they suggested that small amounts of both DNA and RNA may be present.  相似文献

The ultrastructure of the vegetative cell and zoospore of Characiochloris acuminata Lee et Bold (Chlorangiellaceae, Tetrasporales, Chlorophyceae) is described.

The vegetative cell is distinctive in having numerous contractile vacuoles which are randomly distributed in the cytoplasm and visible through the fissures of the parietal chloroplast. A single pyrenoid, embedded in the chloroplast, is penetrated by cytoplasmic canals which are lined by the chloroplast envelope. The vegetative cell is attached to the substrate or host by two flagellar remnants (retained from the zoospore stage), each of which is ensheathed in a gelatinous tube through the cell wall at the cell base. The basal bodies are apparently abscissed from the flagellar shaft by a unit membrane which becomes continuous with the plasma membrane.

The zoospore is biflagellate, with the flagella equal in length, smooth and longer than the cell body. The flagellar sheath is characteristically undulate and the two flagellar bases are connected by a dense interflagellar fibre. The large nucleus has a conspicuously inflated nuclear envelope and the pyrenoid is similar to that of the vegetative cell.  相似文献

This fine structural study of the quadriflagellate zoospore of Ulothrix zonata (Weber & Mohr) Kützing, with special attention to the flagellar root system, demonstrates that it is very similar to the zoospore of Ulva lactuca L. in several aspects. Common features include the presence of a cruciate root system (4-2-4-2 type), a non-striated band that connects basal bodies, a so-called terminal cap, and system I and system II striated root components. Only slight differences exist, i.e. in the shape of the terminal cap, and in the number and position of the system II root components. It is concluded that the taxonomic affinities of U. zonata lie with the Ulvaphyceae sensu Stewart and Mattox rather than with the Chlorophyceae. Additional support for this conclusion is the discovery of tiny, flat body scales on the zoospore of U. zonata. A summary of the distinctive characteristics of the Chlorophyceae, Charophyceae and Ulvaphyceae reflecting the current state of knowledge is given.  相似文献

Ultrastructure of the motile zoospore has been investigated in Oedocladium catolinianum & Hoffman. An unwalled zoospore is usually produced from the contents of a terminal vegetative cell and consists of two principal regions: a small anterior dome and a larger body region; a ring of flagella marks the juncture of these two areas. Chloroplast inclusions consist of thylakoids, mature and incipient pyrenoids, starch and striated microtubules; no eyespot has been observed. Zoospores appear to possess permanent contractile vacuoles with numerous accessory vacuoles, coated vesicles and occasionally coated tubules. The cytoplasm of the dome contains numerous mitochondria ER and golgi bodies, as well as two distinct types of vesicles. The first contains an electron-dense; granular core and is surrounded by a loose, sinuate membrane. The second vesicle is electron-opaque and is found at the apex of the dome: it contains mucopolysaccharides employed during zoospore adhesion. A complex flagellar apparatus encircles the lower region of the dome. It consists of ca. 30–65 flagella, a ring-shaped fibrous band, flagella roots and additional supporting material. The flagella and roots alternate with one another beneath the fibrous band. The compound flagellar roots consist of two superimposed components: an outer ribbon-like unit composed of three microtubular elements and a single striated inner component. A band of support material lies beneath the proximal end of the basal bodies. It is a continuous fibrous band, although it often appears as three distinct, repetitive units.  相似文献

Alternative evolutionary hypotheses generated from features of vegetative cell morphology and motile cell ultra-structure were investigated using a molecular data set. Complete nuclear-encoded small subunit (18S) ribosomal RNA (rRNA) gene sequences were determined for six species (three each) of the chlorococcalean green algae “Neo chloris” and Characium. Based on motile cell ultra-structure, it was previously shown that both genera could be separated into three distinct groups possibly representing three separate orders and two classes of green algae. 18S rRNA gene sequences were also obtained for three additional taxa, Dunaliella parva Lerche, Pediastrum duplex Meyen, and Friedmannia israelensis Chantanachat and Bold. These organisms were selected because each, in turn, is a representative of one of the three ultrastructural groups into which the Neochloris and Characium species are separable. Phylogenetic analyses utilizing the molecular data fully support the ultrastructural findings, suggesting that the similar vegetative cell morphologies observed in these organisms have resulted from convergence.  相似文献

Understanding spatio‐temporal variability in recruitment is vital to studies of kelp population dynamics. Research on settlement and post‐settlement processes has suggested that arrival of kelp zoospores to suitable substrate is important in limiting kelp recruitment, yet the role of planktonic processes in kelp population dynamics has not been studied due to difficulties in sampling and identifying zoospores. I developed a method to estimate kelp zoospore abundance from in situ plankton samples and used it to study various processes regulating the availability of giant kelp (Macrocystis pyrifera) zoospores for settlement. My studies focused on (1) identifying temporal scales over which zoospore abundance is most variable, (2) describing physical and biological processes that regulate this variability, and (3) determining the relationship between zoospore abundance and settlement. I found that short‐term variability in zoospore abundance (<24 hrs) was not due to changes in supply but rather dispersion, caused by oscillating hydrodynamic forces (e.g. waves). Long‐term variability in zoospore abundance, however, was best explained by the size and density of reproductive adult plants, with zoospore abundance being most variable at the scale of days to months. Changes in adult reproductive condition caused rapid changes in zoospore abundance suggesting that the supply of kelp zoospores is sensitive to environmental regulation of adult physiology. Thus, unlike with marine animals, these results indicate that variability in kelp propagule supply, over scales most likely to affect subsequent settlement and recruitment, is more tightly coupled to demographic and reproductive mechanisms than to physical transport processes.  相似文献

Cattleya coccinea and C. brevipedunculata (Orchidaceae) are closely related species distinguished primarily by geographical distribution, vegetative morphology and flowering period. Both species inhabit high‐elevation regions in south‐eastern Brazil, but are traditionally associated with different habitats, located in cloudy forests and campos rupestres (rocky fields), respectively. We used morphometrics and genetic variation of microsatellite markers to test the occurrence of a hybrid zone between these species located in Parque Estadual do Ibitipoca (PEI), Brazil. Morphological data reveal a continuum of variation between the putative taxa, influenced mainly by characters of leaf, pseudobulb and peduncle. However, genetic data do not support the occurrence of hybridization and introgression in PEI, showing that it is a pure population of C. brevipedunculata. Differences in vegetative characters among individuals from cloudy forests and campos rupestres suggest that morphological variation may be related to phenotypic plasticity in response to environmental light fluctuations, an unknown situation for this species. These results highlight the inconsistency of morphology for the identification of hybrids and the role of vegetative characters as a possible complicating factor for the taxonomy of these species, as they are subject to environmental influence.  相似文献

Elevated irradiance has a profound effect on the successful dispersal and establishment of kelp zoospores, affecting their physiology and viability. The research to date, however, has been on zoospores localized near the benthos, with little attention on the importance of vertical transportation and subsequent exposure to increased irradiance. Therefore, we wanted to investigate the effects of exposure to high irradiance on the reproductive planktonic life‐history stages of kelps Macrocystis pyrifera (L.) C. Agardh and Pterygophora californica Rupr. Zoospores of both species were exposed to different irradiances (75, 275, 575, 1,025 μmol photons · m?2 · s?1) over varying durations (1, 2, 4, 8, 12 h) and subsequently monitored for settlement competency, gametophyte development, and reproductive viability. Settlement success for M. pyrifera was uniform throughout all irradiance × time treatments, while settlement for P. californica decreased with increasing exposure time but not irradiance, although settlement was generally reduced at the highest irradiance level. Following zoospore settlement, germ tube development was visible in the gametophytes of both species within 1 week, although a significant decline of germ tube density in P. californica was observed with increasing irradiance. Similarly, a decrease in germ tube development with increasing exposure was observed across all irradiance levels for M. pyrifera, but irradiance itself was not significant. Further development into embryonic sporophytes was remarkably similar to gametophyte development, suggesting that the effect of exposure of kelp zoospores to high irradiance on subsequent sporophyte production is mediated through gametophyte development as well as zoospore survival.  相似文献
