SYNOPSIS. An ultrastructural investigation has been carried out on 180°-rotated ciliary meridians (inverted meridians) in Tetrahymena pyriformis temperature-sensitive mutant (molb/molb), syngen 1, strain B. The longitudinal, transverse and postciliary microtubular bands, the kinetodesmal fiber, and the parasomal sac, are shown to be disposed at a 180° angle to their normal positions or orientations. Other abnormalities are as folows: the first 2 basal bodies of the inverted meridian fail to organize into “couplets” and the inverted meridian intrudes into the anterior pole region; an extra longitudinal microtubular band is found in one of the cell lines.  相似文献

SYNOPSIS The formation of the contractile vacuole pore (CVP) in Tetrahymena thermophila. genotype molb/molb is temporally and spatially associated with the formation of the fission gap in the CVP meridian. New CVPs arise when fission gaps appear in CVP meridians, the new pores being found anterior to the gaps. When, however, CVP meridians are rotated 180°, the fission gaps develop late. In more than 1/3 of the 180°-rotated CVP meridians, the new CVPs are formed before the appearance of the fission gap. Evidently, the appearance of the gap is not a prerequisite for CVP formation. Nevertheless, mutants exist in which the absence of fission gap and CVP are correlated in some cases and in which the presence of supernumerary fission gap and CVP are correlated in other instances. It is suggested that the 2 developmental events, although not causally related to each other, may be controlled by a common morpho-genetic signal. This commits a certain site (mid-body) along a ciliary meridian to develop the fission gap as well as the CVP; however, after this step of commitment, the appearance of the fission gap is delayed in 180°-rotated CVP meridians.  相似文献

The meridian phenomena are closely associated with the neuro-muscular function. Our experiments show that meridian phenomenon, acupoint stimulation-induced generating-sensations along meridians (GSM), does exist in some volunteers, and the subjects can describe the characteristics of the feelings and the route of GSM. Simultaneously, GSM is often accompanied with progressive myoelectric activity along the meridian which is able to be recorded objectively. Bra-chial plexus anesthesia and regional neuro-muscular block may arrest these responses while the neuro-muscular blocking agent may stop neuromuscular transmission at the motor end-plate rather than check GSM. The facts reveal that GSM probably depends on muscular activity and is involved in the secondary neuro-muscular excitation propagating along the longitudinal axis of muscles. These findings suggest that the myoelectric activities constitute the foundation of secondarily generating-sensation propagating along the longitudinal orientation of limbs  相似文献

Background

The scientific basis for acupuncture meridians is unknown. Past studies have suggested that acupuncture meridians are physiologically characterized by low electrical impedance and anatomically associated with connective tissue planes. We are interested in seeing whether acupuncture meridians are associated with lower electrical impedance and whether ultrasound-derived measures – specifically echogenic collagenous bands - can account for these impedance differences.

Methods/Results

In 28 healthy subjects, we assessed electrical impedance of skin and underlying subcutaneous connective tissue using a four needle-electrode approach. The impedances were obtained at 10 kHz and 100 kHz frequencies and at three body sites - upper arm (Large Intestine meridian), thigh (Liver), and lower leg (Bladder). Meridian locations were determined by acupuncturists. Ultrasound images were obtained to characterize the anatomical features at each measured site. We found significantly reduced electrical impedance at the Large Intestine meridian compared to adjacent control for both frequencies. No significant decrease in impedance was found at the Liver or Bladder meridian. Greater subcutaneous echogenic densities were significantly associated with reduced impedances in both within-site (meridian vs. adjacent control) and between-site (arm vs. thigh vs. lower leg) analyses. This relationship remained significant in multivariable analyses which also accounted for gender, needle penetration depth, subcutaneous layer thickness, and other ultrasound-derived measures.

Conclusion/Significance

Collagenous bands, represented by increased ultrasound echogenicity, are significantly associated with lower electrical impedance and may account for reduced impedances previously reported at acupuncture meridians. This finding may provide important insights into the nature of acupuncture meridians and the relevance of collagen in bioelectrical measurements.  相似文献

Purpose

To identify corneal epithelial- and stromal-thickness distribution patterns in keratoconus using spectral-domain optical coherence tomography (SD-OCT).

Patients and Methods

We analyzed SD-OCT findings in 20 confirmed cases of keratoconus (group 1) and in 20 healthy subjects with corneal astigmatism ≥2 D (group 2). Epithelial and stromal thicknesses were measured at 11 strategic locations along the steepest and flattest meridians, previously located by corneal topography. Vertical mirrored symmetry superimposition was used in the statistical analysis.

Results

The mean maximum keratometry measurements in groups 1 and 2 were 47.9±2.9 D (range, 41.8–52.8) and 45.6±1.1 D (range, 42.3–47.5), respectively, with mean corneal cylinders of 3.3±2.2 D (range, 0.5–9.5) and 3.6±1.2 D (range, 2.0–6.4), respectively. The mean epithelial thickness along the steepest meridian in group 1 was the lowest (37.4±4.4 µm) at 1.2 mm inferotemporally and the highest (59.3±4.4 µm) at 1.4 mm supranasally from the corneal vertex. There was only a small deviation in thickness along the steepest meridian in group 2, as well as along the flattest meridians in both groups. The stromal thickness distribution in the two groups was similar to the epithelial, while the stromal thickness was generally lower in group 1 than in group 2.

Conclusions

SD-OCT provides details about the distribution of corneal epithelial and stromal thicknesses. The epithelium and stroma in keratoconic eyes were thinner inferotemporally and thicker supranasally compared with control eyes. The distribution pattern was more distinct in epithelium than in stroma. This finding may help improve the early diagnosis of keratoconus.

The sub-thecal microtubular cytoskeleton of Amphidinium rhynchocephalum Anissimowa was investigated using indirect immunofluorescence microscopy and transmission electron microscopy. The majority of sub-thecal microtubules are longitudinally oriented and radiate from one of two sub-thecal transverse microtubular bands that lie adjacent to the anterior and posterior edge of the cingulum.Both transverse bands consist of 3–5 microtubules and are loop shaped with one end adjacent to the cell's right edge of the sulcus and the other end adjacent to the fibrous ventral ridge. The posterior transverse microtubular band (PTB) defines the posterior edge of the cingulum and gives rise to numerous posteriorly directed longitudinal microtubular bundles that consist of 1–3 microtubules per bundle. These bundles end at the posterior end of the cell. The PTB also gives rise to the cingular longitudinal microtubules that underlie the cingular groove and terminate at the anterior transverse microtubular band (ATB). The ATB defines the anterior edge of the cingulum and loops around the base of the epicone. This band gives rise to anteriorly directed longitudinal microtubular bundles that terminate in the small epicone of the cell. The longitudinal microtubular root of the flagellar apparatus is directed posteriorly and lies immediately beneath the theca but is distinct from the subthecal microtubule system. A narrow fibrous ridge is ventrally located to the cell's left between the exit apertures of the transverse and longitudinal flagella. In this position, the ventral ridge lies between and also connects with the anterior and posterior transverse microtubular bands. The ventral ridge is also associated with three microtubules that are distinct from other cytoskeletal microtubules. Our results demonstrate that the majority of sub-thecal microtubules originate from one of two microtubular bands associated with the cingulum. The possible role of the fibrous ventral ridge and its associated microtubules is also discussed.  相似文献

Purpose

To determine the biometry of anterior segment dimensions of the human eye on both horizontal and vertical meridians with extended scan depth optical coherence tomography (OCT) during accommodation.

Methods

Twenty pre-presbyopic volunteers, aged between 24 and 30, were recruited. The ocular anterior segment of each subject was imaged using an extended scan depth OCT under non- and 3.0 diopters (D) of accommodative demands on both horizontal and vertical meridians. All the images were analyzed to yield the following parameters: pupil diameter (PD), anterior chamber depth (ACD), anterior and posterior surface curvatures of the crystalline lens (ASC and PSC) and the lens thickness (LT). Two consecutive measurements were performed to assess the repeatability and reproducibility of this OCT. They were evaluated by calculating the within-subject standard deviation (SD), a paired t-test, intra-class correlation coefficients (ICC) and the coefficient of repeatability/reproducibility (CoR).

Results

There were no significant differences between two consecutive measurements on either horizontal or vertical meridians under both two different accommodative statuses (P>0.05). The ICC for all parameters ranged from 0.775 to 0.998, except for the PSC (0.550) on the horizontal meridian under the non-accommodative status. In addition, the CoR for most of the parameters were excellent (0.004% to 4.89%). In all the parameters, only PD and PSC were found different between the horizontal and vertical meridians under both accommodative statuses (P<0.05). PD, ACD, ASC and PSC under accommodative status were significantly smaller than those under the non-accommodative status, except that the PSC at the vertical meridian did not change. In addition, LT was significantly increased when accommodation.

Conclusion

The extended scan depth OCT successfully measured the dimensions of the anterior eye during accommodation with good repeatability and reproducibility on both horizontal and vertical meridians. The asymmetry of lens posterior surface and oval-shaped pupil were found during accommodation.  相似文献

The absolute configuration of the flagellar apparatus of biflagellate zoospores of Enteromorpha flexuosa (Wulfen ex Roth.) J. Agardh ssp. pilifera (Kütz.) Bliding was determined. Viewed from the anterior of the cell, the flagellar apparatus shows 180° rotational symmetry with a counter-clockwise absolute orientation of its components. In longitudinal sections, the posteriorly directed basal bodies form an angle of about 170°–180° to one another. A reduced striated distal fiber connects the two basal bodies. The cruciate microtubular rootlet system has a 4–2–4–2 alternation pattern. Striated microtubule-associated components (SMACs or system I-fibers) and rhizoplasts (or system II fibers) accompany the two-membered rootlets. Striated bands connect the proximal sheaths with the four-Membered rootlets. The bilobate terminal caps do not completely cover the proximal ends of the basal bodies. This is the first ultrastructural study of biflagellate zoospores in a member of the Ulvales.  相似文献

Summary The sub-thecal microtubular cytoskeleton of the dinoflagellatesAmphidinium rhynchocephalum, Gymnodinium sanguineum, andGymnodinium. sp has been investigated by indirect immunofluorescence microscopy. In these cells, the majority of cytoskeletal microtubules lie in the anterior-posterior plane. These longitudinal microtubules clearly originate from one of two radially arranged microtubular bands that correspond in location with the anterior and posterior edge of the cingolar depression. Purpose

To investigate the morphology of corneal caps in femtosecond laser small incision lenticule extraction (SMILE) and its relation to the refractive outcomes.

Methods

A prospective study of fifty-four corneal caps created with VisuMax femtosecond laser were examined using an Fourier-domain optical coherence tomography at 1 day, 1 week, 1 month and 6 months after SMILE. The cap thickness at nine points on each of the four meridians (0°, 45°, 90°, 135°) and the diameter were measured. Cap morphology, changes over time and its correlation with refractive outcomes were assessed.

Results

The mean achieved central cap thickness were (108.74±5.06) µm at 6 months and (107.32±4.81 ) µm at 1 month postoperatively, significantly thinner than that at 1 day (110.81±7.95) µm and 1 week (109.58±7.48 ) µm (P<0.05). The mean diameter on 0° meridian was (7.61±0.07) mm, significantly larger than that on 90° meridian (7.57±0.06) mm (P = 0.001). Cap morphology showed good regularity, except that the differences of points in two pairs were significant at 1 day postoperatively. The uniformity was consistent over time and the central cap thickness was thinner than those in the paracentral and peripheral areas. The refractive outcomes stabilized within 1 month. Uncorrected distance visual acuity (UDVA) was correlated to the central cap thickness at 1 day and 1 week (both rs = 0.33, p<0.05). The uniformity index was correlated with UDVA (rs = 0.34, p<0.05) and corrected distance visual acuity (rs = 0.32, p<0.05) at 1 week postoperatively.

Conclusions

Corneal caps of SMILE are predictable with good reproducibility, regularity and uniformity. Cap morphology might have a mild effect on refractive outcomes in the early stage. Further study should focus on the impact on the visual quality.  相似文献

PurposeTo determine using swept-source optical coherence tomography (SS-OCT) whether there are differences in the location of the anterior lamina cribrosa insertion (ALI) in primary open-angle glaucoma (POAG) patients and healthy subjects.MethodsFifty three eyes from 53 patients with POAG, and 53 eyes from 53 age-matched healthy subjects were included prospectively in Seoul National University Bundang Hospital. Twelve radial line B-scans centered on the optic disc in every half-clock-hour meridian were acquired using SS-OCT. The ALI position was assessed by measuring two parameters: (1) ALI distance (ALID)—the distance from the anterior scleral canal opening (ASCO) to the ALI; and (2) marginal anterior lamina cribrosa surface depth (mALCSD)—the perpendicular distance from the ASCO plane to the anterior lamina cribrosa surface. These parameters were compared between the two groups for each meridian.ResultsBoth ALID (256±54 vs. 209±37 µm, mean ± SD, p<0.001) and mALCSD (232±63 vs. 187±40 µm, p<0.001) were significantly greater in the POAG group than in the normal group. The largest difference was observed at the 6.5 o′clock and 11.5 o′clock meridians for both ALID and mALCSD. Multiple regression analysis revealed a negative correlation between age and both ALID and mALCSD in the control group, and a negative correlation between mean deviation of the visual field test and both ALID and mALCSD in the POAG group.ConclusionsThe ALI was displaced posteriorly in eyes with POAG compared to those of healthy controls. This finding suggests that the posteriorly located lamina cribrosa insertion is an important component of glaucomatous optic nerve excavation.  相似文献

Monocular threshold stimulus intensities (ΔIo, photons) were measured along the 0–180° meridian of human retinae for three observers. The test image was small (= 0.08°) and of short duration (= 0.20 second). ΔIo was found to decrease as the angular distance from the fovea was increased. Actual counts of the number of retinal elements per mm.2 along the 0–180° meridian (Østerberg) were compared with the obtained results. No direct correlation was found to exist between visual sensitivity and the number of retinal elements. Binocular threshold stimuli were also measured along the same meridian. The form of the function relating binocular visual sensitivity and retinal position was discovered to be essentially similar to that for monocular sensitivity, but is more symmetrical about the center of the fovea. The magnitude of the binocular measurement is in each case smaller than that of the monocular threshold stimulus intensity for the more sensitive eye. The ratio is statistically equal to 1.4 (a fact which suggests Piper''s rule). These results are shown to be consistent with the hypothesis that the process critical for the eventuation of the threshold response is localized in the central nervous system. They are not consistent with the view that the quantitative properties of visual data are directly determined by properties of the peripheral retina.  相似文献

使用高灵敏度二氧化碳(CO_2)测定仪对30例健康人12经原穴的皮肤CO_2呼出量(SRC)进行了测定.对所测24个穴位的SRC进行两两相关计算,使用SAS统计软件对其距离矩阵进行聚类分析.类平均法聚类的结果表明,左右同名穴首先聚类,其次是表里经穴和同名经穴,伪t检验还支持将12经原穴分成两大类.Ward法聚类的结果与类平均法接近.同名穴组、表里经穴组、同名经穴组和非特异组的平均相关系数分别为0.814,0.65,0.514和0.379.秩和检验表明各组之间有显著性差异.本结果为传统中医经络理论中的同经左右相关、表里经相关和同名经相关原理提供了科学支持.  相似文献

The ultrastructure of Woloszynskia limnetica Bursa was examined using serial thin section electron microscopy. Sections of W. limnetica reveal numerous chloroplast profiles without any obvious pyrenoids. The extensive pusular complex consists of a "smooth" part and a part lined with electron-dense particles. The nucleus is located in the episome. A stigma (= eyespot) consisting of numerous electron-dense globules is situated beneath the amphiesmal vesicles of the sulcal groove. The longitudinal microtu-bular root extends between the stigma and the amphiesma vesicles. Subthecal fibers occur in conjunction with the microtubules and the stigma. Both flagellar exit apertures are encircled by a broad striated collar, each giving rise to a fiber that extends along the pusular canal opening. The striated collars are interconnected by the ventral ridge fiber. The basal part of the transverse flagellum has, in addition to the normal paraxonemal rod (= striated strand or fiber), a semicircular structure consisting of fibrils. The flagellar apparatus is complex but possesses components typically found in the Dinophyceae. The longitudinal mi-crotubular root is broad and is connected to both striated collars. The transverse basal body gives rise to the transverse microtubular root, which in turn is associated with microtubules that extend to the interior of the cell and with the transverse striated root. The transitional region of both basal bodies possesses a distinctive fibrous ring attached to each microtubular triplet by short fibers that collectively appear as spokes of a wheel. Not unexpectedly, the flagellar apparatus of Woloszynskia limnetica is much like that of the related Woloszynskia sp.; however, some dif ferences were discovered. A phylogenetic relationship between Woloszynskia limnetica, W. coronata ( Wolosz.) Thompson, and W. sp. is indicated based on similarities in pusule and stigma structure .  相似文献

Background:

Acupuncture is commonly used to treat migraine. We assessed the efficacy of acupuncture at migraine-specific acupuncture points compared with other acupuncture points and sham acupuncture.

Methods:

We performed a multicentre, single-blind randomized controlled trial. In total, 480 patients with migraine were randomly assigned to one of four groups (Shaoyang-specific acupuncture, Shaoyang-nonspecific acupuncture, Yangming-specific acupuncture or sham acupuncture [control]). All groups received 20 treatments, which included electrical stimulation, over a period of four weeks. The primary outcome was the number of days with a migraine experienced during weeks 5–8 after randomization. Our secondary outcomes included the frequency of migraine attack, migraine intensity and migraine-specific quality of life.

Results:

Compared with patients in the control group, patients in the acupuncture groups reported fewer days with a migraine during weeks 5–8, however the differences between treatments were not significant (p > 0.05). There was a significant reduction in the number of days with a migraine during weeks 13–16 in all acupuncture groups compared with control (Shaoyang-specific acupuncture v. control: difference –1.06 [95% confidence interval (CI) –1.77 to –0.5], p = 0.003; Shaoyang-nonspecific acupuncture v. control: difference –1.22 [95% CI –1.92 to –0.52], p < 0.001; Yangming-specific acupuncture v. control: difference –0.91 [95% CI –1.61 to –0.21], p = 0.011). We found that there was a significant, but not clinically relevant, benefit for almost all secondary outcomes in the three acupuncture groups compared with the control group. We found no relevant differences between the three acupuncture groups.

Interpretation:

Acupuncture tested appeared to have a clinically minor effect on migraine prophylaxis compared with sham acupuncture.

Clinicaltrials.gov NCT00599586About 6%–8% of men and 16%–18% of women in the United States and England experience migraines, with or without an aura.1,2 A prevalence of 1% has been reported in mainland China,3 compared with 4.7% in Hong Kong and 9.1% in Taiwan.4,5 A recent Cochrane meta-analysis suggests that acupuncture as migraine prophylaxis is safe and effective and should be considered as a treatment option for willing patients.6Although the specific effects acupuncture are controversial, acupuncture, as it is currently practised, clearly differentiates between real acupuncture points and nonacupuncture points. The Chinese Government launched the National Basic Research Program to obtain more data about the specificity of acupuncture points.7Trials from Italy and Brazil8,9 showed that acupuncture was more effective than sham acupuncture in preventing migraines, but other trials have reported no differences.1013 There is no evidence that one acupuncture strategy is more effective than another for treating migraines. According to acupuncture theory, a headache on the lateral side is usually defined as a Shaoyang headache. In Jinkuiyi,14 migraines are said to affect the yang meridians (including the Taiyang, Yangming and Shaoyang meridians). In Lingshu,15 the Shaoyang meridians are said to go through the lateral side of the body, therefore the Shaoyang meridians are thought to be superior for treating migraines. Some points on the Shaoyang meridians are regarded as being more specific for migraines than other points.16Our aim was to investigate whether acupuncture at specific acupuncture points was more efficacious in preventing migraine than sham acupuncture at nonacupuncture points. SYNOPSIS. Clones of 3 strains of the ciliated protozoon Glaucoma chattoni were subjected to corticotypic analysis. In agreement with previous studies on Tetrahymena, many cortical features vary in a systematic way with the total number of ciliary meridians. Most strikingly, the positions of the contractile vacuole pore and the numbers of postoral meridians change in a regular progression, but the patterns of change are distinctive in different strains. Under the growth conditions employed the corticotypes (meridian numbers) in G. chattoni are much less stable than those in T. pyriformis and no preferential “stability sink” is apparent.  相似文献

The three-dimensional structure of the flagellar apparatus in the gonyaulacoid dinoflagellate. Ceratium hirundinella var. furcoïdes (Schröder) Hub.-Pest. was determined using serial section electron microscopy. The flagellar apparatus is quite large and consists of several components. The two basal bodies nearly abut at their proximal ends and are separated by an angle of approximately 120° The broad longitudinal microtubular root extends from the cell's left edge of the longitudinal basal body and bends around the sulcal/cingular depression into the cell's left antapical horn. A transverse striated fibrous root is associated with the transverse basal body and a narrow electron dense extension is present along the anterior edge of the transverse basal body. This study revealed severa1 hitherto unreported fibrous components of the flagellar apparatus that link the various microtubular and fibrous components to themselves and to the two striated collars. A large striated fibrous connective links the two striated collars to one another. This fibrous connective is linked to another striated fibrous connective that originates from the longitudinal basal body and lies perpendicular to the longitudinal microtubular root. The readily identifiable and numerous components of the Ceratium flagellar apparatus are comparable to those of other dinoflagellates. The combined presence of well dpveloped striated collars, a striated collar connective, and a basal body angle of approximately 120° indicates that this flagellar apparatus is most like that described for Peridinioid dinoflagellates. Important similarities are also noticeable between this flagellar apparatus and that of Oxyrrhis marina.  相似文献

Summary Modern microscopical approaches have allowed more accurate investigations of the three-dimensional nature of the dinoflagellate flagellar apparatus (FA) and several other cytoskeletal protein complexes. Our presentation overviews the nature of the dinoflagellate FA and cytoskeleton in a number of taxa and compares them with those of other protists. As with other protists, the FA of the dinoflagellates can be characterized by the presence of fibrous and microtubular components. Our studies and others indicate that the dinoflagellate FA can be expected to possess a striated fibrous root on the basal body of the transverse flagellum and a multimembered microtubular root on the basal body of the longitudinal flagellum. Two other features that appear widespread in the group are the transverse striated root associated microtubule (tsrm) and the transverse microtubular root (tmr). The tsrm extends at least half the length of the transverse striated root while the tmr extends from the transverse basal body toward the exit aperture of the transverse flagellum. In most cases, the tmr gives rise to several cytoplasmic microtubules at a right angle. The apparent conserved nature of these roots leads us to the conclusion that the dinoflagellate FA can be compared to the FA of the cryptomonads, chrysophytes, and the ciliates for phylogenetic purposes. Of these groups, the chrysophytes possess an FA with the most structures in common with the dinoflagellates.
