Biomass acid hydrolysate of oleaginous yeast Trichosporon cutaneum after microbial oil extraction was applied as substrate for bacterial cellulose (BC) production by Komagataeibacter xylinus (also named as Gluconacetobacter xylinus previously) for the first time. BC was synthesized in static culture for 10 days, and the maximum BC yield (2.9?g/L) was got at the 4th day of fermentation. Most carbon sources in the substrate (glucose, mannose, formic acid, acetic acid) can be utilized by K. xylinus. The highest chemical oxygen demand (COD) removal (40.7?±?3.0%) was obtained at the 6th day of fermentation, and then the COD increased possibly due to the degradation of BC. The highest BC yield on COD consumption was 38.7?±?4.0% (w/w), suggesting that this is one efficient bioconversion for BC production. The BC structure was affected little by the substrate by comparison with that generated in classical HS medium using field-emission scanning electron microscope (FE-SEM), Fourier transform infrared, and X-ray diffraction. Overall, this technology can both solve the issue of waste oleaginous yeast biomass and produce valuable biopolymer (BC).  相似文献

Tuna condensate was a better substrate than shrimp-blanching water or effluent from a frozen-seafood plant for growing Rhodocyclus gelatinosus under anaerobic conditions in the light. One strain out of four examined, R7, gave the highest biomass (4.0 g/l), cell yield (0.32 g cell/g COD), and COD removal (78%) in 1:10 (v/v) diluted tuna condensate. Shrimp-blanching water added to the tuna condensate further increased growth rate, biomass and COD removal. Optimal growth was at pH 7.0 and 3000 Lux light intensity. Acetate, pyruvate, glucose, glutamate, propionate or malate added to the tuna condensate did not increase cell yield, carotenoid or bacteriochlorophyll content or biomass protein. A maximum cell mass of 5.6 g/l (containing 50% protein) and 86% COD removal were obtained after 5 days' incubation under optimal conditions.  相似文献

The kinetics of anaerobic digestion of cane molasses distillery slops was investigated using a continuous-flow bioreactor which contained waste tyre rubber as support, to which the microorganisms became immobilized. Hydraulic retention times (HRT) ranging from 1 to 10 days were investigated at an average influent chemical oxygen demand (COD) concentration of 47.7?g/l. The maximum substrate utilization rate, k, and half saturation coefficient, K L, were determined to be 1.82?kg CODremoved/kg VSS day and 0.33?kg COD/kg VSS day. The yield coefficient, Y, and sludge decay rate coefficient, K d, were also determined to be 0.06?kg VSS/kg CODremoved and 0.05?day-1, respectively. Methane production was maximum (6.75?l/l day) at a 2 day HRT corresponding to a biomass loading rate of 2.578?kg COD/kg VSS day. Biogas yield ranged between 0.51?l/g COD (HRT=2 days) and 0.25?l/g COD (HRT=1?day). In addition, the methane percentage in the biogas varied between 70.5% (HRT=10?days) and 47.5% (HRT=1?day). The close relationship between biomass loading rate and specific substrate utilization rate supported the use of Monod equations. Finally, the experimental values of effluent substrate concentration were reproduced with deviations equal to or less than 10% in every case.  相似文献

Intrinsic growth and substrate uptake parameters were obtained for Peptostreptococcus productus, strain U-1, using carbon monoxide as the limiting substrate. A modified Monod model with substrate inhibition was used for modeling. In addition, a product yield of 0.25 mol acetate/mol CO and a cell yield of 0.034 g cells/g CO were obtained. While CO was found to be the primary substrate, P. productus is able to produce acetate from CO2 and H2, although this substrate could not sustain growth. Yeast extract was found to also be a growth substrate. A yield of 0.017 g cell/g yeast extract and a product yield of 0.14 g acetate/g yeast extract were obtained. In the presence of acetate, the maximum specific CO uptake rate was increased by 40% compared to the maximum without acetate present. Cell replication was inhibited at acetate concentrations of 30 g/l. Methionine was found to be an essential nutrient for growth and CO uptake by P. productus. A minimum amount of a complex medium such as yeast extract (0.01%) is, however, required.  相似文献

The anaerobic biodegradation of carbon tetrachloride (CT) was investigated during the granulation process by reducing the hydraulic retention time, increasing the chemical oxygen demand (COD) and CT loadings in a 2l laboratory-scale upflow anaerobic sludge blanket (UASB) reactor. Anaerobic unacclimated sludge and glucose were used as seed and primary substrate, respectively. Granules were developed 4 weeks after start-up, which grew at an accelerated rate for 8 months, and then became fully grown. The effect of operational parameters such as influent CT concentrations, COD, CT loading, food to biomass ratio and specific methanogenic activity (SMA) were also considered during granulation. The granular sludge cultivated had a maximum diameter of 2.1 mm and SMA of 1.6 g COD/g total suspended solid (TSS) day. COD and CT removal efficiencies of 92 and 88% were achieved when the reactor was firstly operating at CT and COD loading rates of 17.5 mg/l day and 12.5 g/l day, respectively. This corresponds to hydraulic retention time of 0.28 day and food to biomass ratio of 0.5 g COD/g TSS day. Kinetic coefficients of maximum specific substrate utilization rate, half velocity coefficient, growth yield coefficient and decay coefficient were determined to be 2.4 × 10–3 mg CT/TSS day–1, 1.37 mg CT/l, 0.69 mg TSS/mg CT and 0.046 day–1, respectively for CT biotransformation during granulation.  相似文献

Summary The effect of substrate concentration (S 0) on the fermentation parameters of a sugar mixture byPichia stipitis Y 7124 was investigated under anaerobic and microaerobic conditions. Under microaerobiosisP. stipitis maintained high ethanol yield and productivity when initial substrate concentration did not exceed 150 g/l; ethanol yield of about 0.40 g/g and volumetric productivity up to 0.39 g/l per hour were obtained. Optimal specific ethanol productivity (0.2 g/g per hour) was observed withS 0=110 g/l. Under anaerobic conditionsP. stipitis exhibited the highest fermentative performances atS 0=20 g/l; it produced ethanol with a yield of 0.42 g/g, with a specific rate of 1.1 g/g per day. Summary Aspergillus terreus NRRL 1960 was grown on porous disks rotating intermittently in and out of the liquid phase. This immobilized fungal cell bioreactor was used to produce itaconic acid from glucose in a continuous operation. The effect of temperature, pH, disk rotation speed, and feed rate on the itaconic acid concentration and volumetric productivity were studied. The highest itaconic acid concentration and volumetric productivity obtained were 18.2 g/l and 0.73 g/l·h, respectively, under the following conditions: temperature at 36°C, pH 3.0, disk rotation speed at 8 rpm, and feed rate at 60 ml/h. Summary The ability of C. guilliermondii and C. parapsilosis to ferment xylose to xylitol was evaluated under different oxygen transfer rates in order to enhance the xylitol yield. In C. guilliermondii, a maximal xylitol yield of 0.66 g/g was obtained when oxygen transfer rate was 2.2 mmol/l.h. Optimal conditions to produce xylitol by C. parapsilosis (0.75 g/g) arose from cultures at pH 4.75 with 0.4 mmoles of oxygen/l.h. Summary The use of a mixed culture of Geotrichum candidum, Hansenula anomala and Candida krusei for microbial protein production and treatment of whiskey distillery spent wash was investigated in both batch and continuous culture. Although capable of assimilating the same substrate constituents in pure culture the organisms used different constituents for growth in mixed continuous culture, allowing a stable mixed population to be established. The relative proportions of the three organisms in the population was dependent on the dilution rate.The mixed culture did not give superior yields or productivities, but did give proportionally greater COD reduction and would be expected to bemore stable and resistant to contamination.At a dilution rate of 0.10 h–1 the mixed culture gave a biomass yield of 4.8 g l–1, containing 55% crude protein, with a COD reduction of 31.5%, while in mixed batch culture a biomass yield of 12.5 g l–1, containing 48% crude protein, was obtained with a COD reduction of 54.9%.  相似文献

Summary Removal of most of the phenolic compounds (88.5%) present in molasses by an aerobic pretreatment with Geotrichum candidum considerably increased the kinetics of anaerobic digestion of this wastewater. Inhibition phenomena were not observed in the range of COD studied (2.3–12.9 g/l). The yield coefficient of methane was 51 % higher than for untreated molasses.  相似文献

Summary Pleurotus sajor-caju 537 was grown on chopped, pasteurized wheat straw non-supplemented and supplemented with formaldehyde-treated soybean, commercial delayed-release nutrient (SpawnMate II SE) or vegetable oil. Yield was 2.1-fold higher for substrate supplemented (12% dry wt) with low-volume formaldehyde-treated soybean as compared to non-supplemented substrate. Mushroom yield from substrate supplemented with commercial nutrient was 1.7-fold higher than yield from non-supplemented substrate. As the supplement level increased, the mushroom yield response increased. The yield ranged from 3.56 kg/m2 for non-supplemented substrate to 7.36 kg/m2 for substrate supplemented (12% dry wt) with formaldehyde-treated soybean. The type of supplement affected in vitro dry matter digestibility (IVDMD) of spent substrate; commercial supplement resulted in higher IVDMD compared to formaldehyde-treated substrate. An opportunity exists for commercial development of a nutrient(s) specifically designed for Pleurotus cultivation.  相似文献

A study of the kinetics of the anaerobic digestion of brewery wastewater was carried out using a 1-litre, continuous-flow, completely-mixed, bioreactor operating at 35°C and containing a saponite-immobilized biomass at a concentration of 6·2 g volatile suspended solids (VSS)/litre. The bioreactor worked satisfactorily in a range of hydraulic retention times from 1·2 to 10 days and eliminated more than 95% of the initial chemical oxygen demand (COD) in all instances.

Guiot's kinetic model was used to determine the macroenergetic parameters of the system, and showed it to have a yield coefficient for the biomass (Y) of 0·080 g VSS/g COD and a specific rate of substrate uptake for cell maintenance (m) of 0·045 g COD/g VSS day.

The experimental results showed the rate of substrate uptake (Rs; g COD/g VSS day), correlated with the concentration of biodegradable substrate (Sb; g COD/litre), through an equation of the Michaelis-Menten type.  相似文献

A mathematical model with a consideration of energy spilling is developed to describe the activated sludge in the presence of different levels of metabolic uncouplers. The consumption of substrate and oxygen via energy spilling process is modeled with a Monod term, which is dependent on substrate and inhibitor. The sensitivity of the developed model is analyzed. Three parameters, maximum specific growth rate (μ max), energy spilling coefficient (q max), and sludge yield coefficient (Y H) are estimated with experimental data of different studies. The values of μ max, q max, and Y H are found to be 6.72 day-1, 5.52 day-1, and 0.60 mg COD mg-1 COD for 2, 4-dinitrophenol and 7.20 day-1, 1.58 day-1, and 0.62 mg COD mg-1 COD for 2, 4-dichlorophenol. Substrate degradation and sludge yield could be predicted with this model. The activated sludge process in the presence of uncouplers that is described more reasonably by the new model with a consideration of energy spilling. The effects of uncouplers on substrate consumption inhibition and excess sludge reduction in activated sludge are quantified with this model.  相似文献

Wastewater from three rubber thread manufacturing industries collected from three different sampling points was characterised. The acidic wastewater (pH = 3.6 to 4.7) contains high levels of COD, BOD, nitrogen and zinc. The average COD:N:P ratio was 100:3.8:0.4 whereas the BOD:N:P ratio was 100:4.5:0.4 with more than sufficient amounts of N and P for anaerobic digestion using the up-flow anaerobic filter. The specific biogas yield ranged between 0.250 to 0.069 l CH4/g COD added when the organic loading rate was altered from 2.0 to 14.0 g COD/l/d respectively. The biogas production rate and the biogas yield rate increased from 8.2 to 42.7 l/d and 0.97 to 1.50 l CH4/l/d respectively, for the same organic loading rate. The N and P utilised ranged between 1.53 to 1.20 and 0.3 to 0.26 mg per 100 mg COD consumed respectively when the organic loading rate was increased from 2.0 to 14.0 g COD/l/d. The optimum COD:N:P ratio obtained for steady state operation of the up-flow anaerobic filter in this study for the rubber thread manufacturing wastewater was 100:1.3:0.3. Attached biomass was responsible for the removal of more than 70% of the COD in the wastewater when PU foam was used as the packing media.  相似文献

A column reactor, in which the bottom two-thirds were occupied by a sludge blanket and the upper one-third by submerged clay rings, was evaluated using slaughterhouse wastewater as substrate. The reactor was operated at 35°C at loading rates varying from 5 g to 45 g chemical oxygen demand (COD) 1–1 × day–1 at an influent concentration of 2450 mg COD 1–1. A maximum substrate removal rate of 32 g COD 1–1 × day–1, coupled with a methane production rate of 6.91 × 1–1 × day–1 (STP), was obtained. This removal rate is significantly higher than those previously reported. The rate of substrate utilization by the biomass was 1.22 g COD (g volatile suspended solids)–1 day–1. COD removal was over 96% with loading rates up to 25 g COD 1–1 × day–1, at higher loading rates performance decreased rapidly. It was found that the filter element of the reactor was highly efficient in retaining biomass, leading to a biomass accumulation yield coefficient of 0.029 g volatile suspended solids g–1 COD, higher than reported previously for either upflow anaerobic sludge-blanket reactors or anaerobic filters operating independently.  相似文献

Summary When more than the minimum number of variables are measured, and measurement error is taken into account, the results of parameter estimation depend on which of the measured variables are selected for this purpose. The reparameterization of Pirt's models for growth produces multiresponse models with common parameters. By using the covariate adjustment technique, a unit variate linear model with covariates is obtained. This allows a combined point and interval estimates of biomass energetic yield and maintenance coefficient to be obtained using standard multiple regression programmes. When this method was applied using form I and form II of the Pirt's models, good combined estimates were obtained and compared. Using data from the literature for Candida lipolytica produced reliable results. It was found that pseudo first order kinetics described the anaerobic digestion of sugar mill wastewater using laboratory-scale fixed bed reactors. A pseudo first order equation described the relationship of COD removal rate with the effluent COD. The values of the maximum COD removal rate and saturation constant were found to be 3.02 g COD/1 day and 0.34 g COD/1 respectively. Based on the similar approach, values of the maximum specific substrate utilization rate, U m, and the coefficient of surface loading saturation, K U, were 0.44 and 0.49 Kg COD/m2 day, respectively. Similar expression was employed to evaluate the effect of the organic volumetric loading rate (B v) and the organic volumetric loading removal rate (B vR). Variations in experimental data from the theoretical values obtained using the model were assessed with respect to the efficiency corresponding to each of the hydraulic retention times and influent concentrations. Theoretical values were calculated with a 15% error band using the kinetic parameters obtained. The experimental values were within this error band in all cases. The production of citric and gluconic acids from fig by Aspergillus niger ATCC 10577 in solid-state fermentation was investigated. The maximal citric and gluconic acids concentration (64 and 490 g/kg dry figs, respectively), citric acid yield (8%), and gluconic acid yield (63%) were obtained at a moisture level of 75%, initial pH 7.0, temperature 30°C, and fermentation time in 15 days. However, the highest biomass dry weight (40 g/kg wet substrate) and sugar utilization (90%) were obtained in cultures grown at 35°C. The addition of 6% (w/w) methanol into substrate increased the concentration of citric and gluconic acid from 64 and 490 to 96 and 685 g/kg dry fig, respectively. Summary The effect of oxygen availability on d-xylose and D-glucose metabolism by Pichia stipitis, Candida shehatae and Pachysolen tannophilus was investigated. Oxygen was not required for fermentation of d-xylose or d-glucose, but stimulated the ethanol production rate from both sugars. Under oxygen-limited conditions, the highest ethanol yield coefficient (Ye/s) of 0.47 was obtained on d-xylose with. P. stipitis, while under similar conditions C. shehatae fermented d-xylose most rapidly with a specific productivity (qpmax) of 0.32 h-1. Both of these yeasts fermented d-xylose better and produced less xylitol than. P. tannophilus. Synthesis of polyols such as xylitol, arabitol, glycerol and ribitol reduced the ethanol yield in some instances and was related to the yeast strain, carbon source and oxygen availability. In general, these yeasts fermented d-glucose more rapidly than d-xylose. The capacity to sense and respond to light is widespread in animals, plants, fungi and bacteria. The effect of light quality on growth and pigment yield of Monascus purpureus was investigated. Incubation in total darkness increased red pigment production from 14. 5 OD/g dry substrate to 22 OD/g dry substrate. In contrast, growth of the fungus in direct illumination resulted in total suppression of pigment production. It was found that both red and blue light influenced pigment yield as well as culture morphology. The authors propose the existence of a light-perception system in Monascus purpureus.  相似文献
