Species composition and density of cladoceran populations changed in Lakes Zürich, St. Mortiz and Baldegg as human populations increased in these watersheds. Lake Zürich sediments became annually laminated in the 1890's as a result of increased organic input as the size of the cities surrounding the lake grew. At the same time, the Bosmina species changed from a oligotrophic form (longispina) to a eutrophic form (longirostris). An increase in Daphnia spp. populations also occurred at this time in the lake's history. Bosmina longispina reappeared in the lake in 1965 as the lake's trophic status changed from eutrophic to mesotrophic due to effective sewage treatment facilities. Annual laminations appear in the Lake St. Moritz sediments about 1910. Shortly thereafter, a shift from B. longispina to B. longirostris occurred. This change in trophic status is associated with increased tourism in the area. Lake Baldegg sediments also show annual laminations beginning in 1885 and a similar shift in the Bosmina species. Other cladoceran remains were too scarce to be useful in interpreting the histories of these lakes.  相似文献

Recent studies describe interactions of pollen surfaces with aerosol particles; pollen surfaces undergo morphological changes and the release of allergens and allergenic fragments from the pollen can be enhanced. Thus allergens from pollen can be found in particle size fractions much smaller than undamaged pollen (<5 μm). This may explain allergic reactions in parts of the lungs which cannot be reached by undamaged pollen. In Switzerland the birch tree (betula verrucosa) major allergen Bet v 1 and the grass (phleum pratense) pollen major allergen Phl p 5 are of particular relevance for inducing pollinosis. In this study aerosols of different aerodynamic diameters were sampled by Andersen-Impactors over 18 months. Sampling areas are subjected to different levels of air pollution (Zürich, Switzerland, urban; Payerne, Switzerland, rural; Davos, Switzerland, alpine). Samples were scanned by electron microscopy and submitted to specific allergen assays (ELISA) for birch pollen major allergen Bet v 1 and grass pollen major allergen Phl p 5 respectively. Particle and major allergen concentrations were highest in Zürich, followed by Payerne and, significantly lower, Davos. Scanning electron microscopy investigations showed interactions of aerosols with pollen surfaces in Zürich and Payerne. The presence of Bet v 1 in smaller aerosol fractions was demonstrated in Zürich and Payerne some weeks before and after birch pollen was counted. PrP knockout mice in which only the open reading frame was disrupted ('Zürich I') remained healthy. However, more extensive deletions resulted in ataxia, Purkinje cell loss and ectopic expression in brain of Doppel (Dpl), encoded by the downstream gene, PRND: A new PrP knockout line, 'Zürich II', with a 2.9 kb PRNP: deletion, developed this phenotype at approximately 10 months (50% morbidity). A single PRNP: allele abolished the syndrome. Compound Zürich I/Zürich II heterozygotes had half the Dpl of Zürich II mice and developed symptoms 6 months later. Zürich II mice transgenic for a PRND:-containing cosmid expressed Dpl at twice the level and became ataxic approximately 5 months earlier. Thus, Dpl levels in brain and onset of the ataxic syndrome are inversely correlated.  相似文献

Cobalt hemoglobin Zürich (alpha 2 beta 263His leads to Arg) has been successfully reconstituted from the apohemoglobin Zürich and cobaltous protoporphyrin IX. The oxygen affinity of cobalt hemoglobin Zurich, as well as that of iron hemoglobin Zürich, were measured in the absence and presence of organic phosphate and Cl-. The overall oxygen affinity of cobalt hemoglobin Zürich was found to be higher and the cooperativity as measured by the n value was smaller than those of cobalt hemoglobin A. Organic phosphate and Cl- affect the oxygen equilibrium properties of cobalt hemoglobin Zürich in a manner similar to that of cobalt hemoglobin A, but to a lesser extant than cobalt hemoglobin A. The EPR spectrum of oxy cobalt hemoglobin Zürich is less sensitive to the replacement of the buffer system from H2O to 2H2O, indicating that the hydrogen bond between the distal amino acid residue and the bound oxygen is not formed in the abnormal beta subunits. The deoxy EPR spectrum of cobalt hemoglobin Zürich is similar to that of deoxy cobalt hemoglobin A, suggesting that the deoxy cobalt hemoglobin Zürich is predominantly in the deoxy quaternary structure (T state).  相似文献

A new gas chromatographic-mass spectrometric method was established that is applicable for the determination of NH4+ utilization and regeneration rates in freshwater. Hollow-fibre modules were used to stop the biogenic nitrogen-fluxes by separating the particulate from the dissolved matter. Incubations were performed in Tedlar bags (polyvinylfluoride), which enabled repeated sample removals through Teflon tubes, making the calculation of nitrogen-fluxes in accordance to Blackburn and Caperon much more reliable. The Berthelot reaction was performed with ammonium and a fragment ion (base peak) of tris-(trifluoroacetyl) 4,4'-dihydroxydiphenylamine was used to determine the at% excess 15N by gas chromatography-mass spectrometry. Nitrogen-flux measurements were made in the epilimnion of the deep, stratified, mesotrophic Lake Zürich, in which the cyanobacterium Planktothrix rubescens was the dominating photoautotrophic micro-organism. The size fraction <20 microm that consisted of heterotrophic bacterioplankton and nanoflagellates, and photoautotrophic pico- and nanoplankton accounted only for a minor part of the ammonium utilization (<25%) and regeneration (< or =25%) rates, whereas the size fraction >20 microm which primarily consisted of Planktothrix rubescens was responsible for the major part. In the eutrophic Lake Au, which is connected to Lake Zürich through a canal, utilization and regeneration rates as high as 700 and 482 nM h(-1) were measured.  相似文献

The paper presents the results of phytocoenological studies of the helophyte vegetation occupying the litoral zone of three North Italian glacial lakes and of the tall-forb communities occurring on the Adige riverside. It uses the method of the Zürich-Montpellier school. The association complex occupying the littoral zone of Lake Caldaro, above all the associations ofCladietum marisci, Caricetum elatae, Juncetum subnodulosi, Schoenetum nigricantis, andSelino-Molinietum caeruleae resembles those of ± mesotrophic, calcium rich prealpine lakes studied in Switzerland, West Germany and Austria. On the other hand, the majority of the communities occurring in the littoral of the great Lake Monticolo are of dystrophic-mesotrophic character, manifested above all in the occurrence ofCaricetum diandrae, Peucedano-Caricetum lasiocarpae, andCaricetum elatae comaretosum, all with boreal distribution. The tall-forb associationLysimachio vulgaris-Filipenduletum has developed in the Adige riverside fromCalthenion and/or fromMagnocaricetalia communities as a result of interruption of mowing.  相似文献

Microcystins (MCs) have a toxic effect on crustacean zooplankton in the laboratory, but there is little or no unequivocal evidence in the literature of their lethal effects on crustacean zooplankton in the field. We used the natural microcystins extracted from Microcystis spp. to test if they could cause any negative effects on crustacean zooplankton. We conducted three experiments in enclosures with water from Lake Taihu, China, and microcystins derived by extraction from Microcystis spp. collected from the lake when the species was in bloom conditions. Initial concentrations of extracellular microcystins (EMCs = MC-RR + MC-LR + MC-YR) ranged from 9.7 to 44.9 μg/L in treatments with microcystin addition. Microcystin concentrations sharply decreased on second day in all the three experiments. EMCs at the end of the experiments varied from only 2.7 to 14.2 % of the levels at the start of the experiments. The dominant species of crustacean zooplankton in the lake were Bosmina longirotris, Ceriodaphnia cornuta, Mesocyclops spp., Limnoithona sinensis, Sinocalanus dorrii and Schmackeria inopinus. ANOVA analysis showed that the density and biomass of cladoceran and copepod did not significantly differ between treatments with microcystin addition and controls. Our results indicate that microcystins derived from lysing Microcystis do not cause any negative effects on crustacean zooplankton.  相似文献

The contamination of lettuce (Lactuca sativa L.) by water‐borne crude extracts of the cyanobacterium microcystin‐producing Microcystis aeruginosa (Kützing) Kützing was investigated. The aim of the study was to determine whether bioaccumulation of microcystins occurs in lettuce foliar tissue when sprayed with solutions containing microcystins at concentrations observed in aquatic systems (0.62 to 12.5 μg · L?1). Microcystins were found in lettuce foliar tissues (8.31 to 177.8 μg per Kg of fresh weight) at all concentrations of crude extracts. Spraying with water containing microcystins and cyanobacteria may contaminate lettuce at levels higher than the daily intake of microcystins recommended by the World Health Organization (WHO), underscoring the need to monitor such food exposure pathways by public authorities.  相似文献

Some species of cyanobacteria synthesize toxins whose concentration during water bloom can reach values dangerous for human and animal health. Planktonic cyanobacteria are the most common and well-studied microcystins producers, hepatotoxic cyclic heptapeptides, whereas microcystin-producing benthic cyanobacteria are less known. In recent years, the mass development of benthic cyanobacteria forming extensive fouling on different substrates has been detected in the littoral zone of Lake Baikal. We found microcystins produced by benthic cyanobacteria in the biofouling on different natural and artificial substrates, including diseased and dead endemic sponges Lubomirskia baicalensis and Baikalospongia spp. collected from the littoral area of Lake Baikal. Microscopic analysis of the biofouling revealed prevalence of representatives of Nostocales and Oscillatoriales with predominance of Tolypothrix distorta that is likely the main microcystin producer in Lake Baikal. According to enzyme-linked immunosorbent assay (ELISA), microcystin concentrations in biofouling were 29.8–3050 μg/kg dry weight. We identified eight microcystin variants using MALDI-TOF/TOF; [Dha7]MC-YR was detected in most samples. The presence of microcystins in biofilms formed on the surface of the artificial substrate by Phormidium autumnale was also recorded. The data obtained demonstrated the necessity to monitor potentially toxic species and concentrations of cyanotoxins in plankton and benthos in the littoral zone of Lake Baikal, especially in the regions with intense tourist and recreational activities.  相似文献

This paper deals with the aerophytic Cyanophyceae from New Caledonia sampled by Prof. Dr A. Guillaumin from Paris, Dr M.G. Baumann-Bodenheim from Zürich, Dr H. Hürlimann from Winterthur and Prof. Baas Becking from Noumea. A total of 124 samples were studied, most of them collected on soil, stones, rocks and barks. The characteristics of these samples are given. Among species, varieties and forms, a total of 85 taxa were recorded. Among them three are new for science, Gloeothece neocaledoniense nov. sp., Hydrocoryne neocaledonica nov. sp. and Scytonematopsis neocaledoniense nov. sp. Cyanobacterial mass occurrences in freshwater lakes are generally formed by Anabaena, Microcystis, and Planktothrix, which may produce cyclic heptapeptide hepatotoxins, microcystins. Thus far, identification of the most potent microcystin producer in a lake has not been possible due to a lack of quantitative methods. The aim of this study was to identify the microcystin-producing genera and to determine the copy numbers of microcystin synthetase gene E (mcyE) in Lake Tuusulanjärvi and Lake Hiidenvesi in Finland by quantitative real-time PCR. The microcystin concentrations and cyanobacterial cell densities of these lakes were also determined. The microcystin concentrations correlated positively with the sum of Microcystis and Anabaena mcyE copy numbers from both Lake Tuusulanjärvi and Lake Hiidenvesi, indicating that mcyE gene copy numbers can be used as surrogates for hepatotoxic Microcystis and Anabaena. The main microcystin producer in Lake Tuusulanjärvi was Microcystis spp., since average Microcystis mcyE copy numbers were >30 times more abundant than those of Anabaena. Lake Hiidenvesi seemed to contain both nontoxic and toxic Anabaena as well as toxic Microcystis strains. Identifying the most potent microcystin producer in a lake could be valuable for designing lake restoration strategies, among other uses.  相似文献

Cyanobacterial harmful blooms (CyanoHABs) that produce microcystins are appearing in an increasing number of freshwater ecosystems worldwide, damaging quality of water for use by human and aquatic life. Heterotrophic bacteria assemblages are thought to be important in transforming and detoxifying microcystins in natural environments. However, little is known about their taxonomic composition or pathways involved in the process. To address this knowledge gap, we compared the metagenomes of Lake Erie free-living bacterioplankton assemblages in laboratory microcosms amended with microcystins relative to unamended controls. A diverse array of bacterial phyla were responsive to elevated supply of microcystins, including Acidobacteria, Actinobacteria, Bacteroidetes, Planctomycetes, Proteobacteria of the alpha, beta, gamma, delta and epsilon subdivisions and Verrucomicrobia. At more detailed taxonomic levels, Methylophilales (mainly in genus Methylotenera) and Burkholderiales (mainly in genera Bordetella, Burkholderia, Cupriavidus, Polaromonas, Ralstonia, Polynucleobacter and Variovorax) of Betaproteobacteria were suggested to be more important in microcystin degradation than Sphingomonadales of Alphaproteobacteria. The latter taxa were previously thought to be major microcystin degraders. Homologs to known microcystin-degrading genes (mlr) were not overrepresented in microcystin-amended metagenomes, indicating that Lake Erie bacterioplankton might employ alternative genes and/or pathways in microcystin degradation. Genes for xenobiotic metabolism were overrepresented in microcystin-amended microcosms, suggesting they are important in bacterial degradation of microcystin, a phenomenon that has been identified previously only in eukaryotic systems.  相似文献

The ability of the Planktothrix rubescens to stratify in Lake Zürich is related to the size and shape of the cyanobacterial filaments. Detailed measurements made in the lake are used in a dynamic computer model of buoyancy regulation to investigate the vertical movements of filaments tracking the depth at which the irradiance would support neutral buoyancy. The movement of the filament lags behind the constantly changing target depth owing to (a) the time taken for the filament to respond to the irradiance by changing its density and (b) the time it takes to move by sinking down or floating up through the water column. The model simulates the stratification depth over a 5-month period of the summer from the continuous measurements of irradiance and weekly measurements of light attenuation and temperature, without any further adjustment over the period. Models using filaments of the size observed in Lake Zürich explain several details of the observed depth changes: smaller planktonic cyanobacteria (e.g. Limnothrix sp.) are unable to migrate fast enough and larger ones (e.g. Anabaena spp.) will overshoot and become entrained in the epilimnion. The model can be used to simulate recruitment of Planktothrix filaments from different depths after vernal stratification. Recruitment of filaments from depths down to 45 m will contribute to the metalimnetic population increase in early July.  相似文献

Objective: To evaluate undergraduate students’ attitude towards the clinical components of the Leipzig (LPEG) and Zürich (ZPEG) Programmes of Education in Gerodontology. Background: Undergraduate student education is the seedbed for conscientious professionals. Extramural clinical education contributes to the formation of positive attitudes. Students in Zürich participate in three clinical activities (in‐house gerodontology clinic, extramural acute geriatrics ward, mobile dental service), in Leipzig they visit a long‐term care facility on six occasions within 4 years. Methods: A structured questionnaire with 10 items was administered to students in Leipzig [n = 34, 70.6% female, mean age 25.8 (SD 3.04) years] at the beginning and after completion of gerodontology training and to students in Zürich [n = 33, 48.5% female, mean age 27.0 (SD 3.28) years] on three occasions after clinical training. Students indicated the degree of their agreement with seven statements presented using a 5‐point scale. A choice of responses which characterised the course was offered for assessment. Results: Close collaboration with dental tutors, while self‐treating patients in the mobile dental service (mobiDent?) attracted the most positive responses. Ratings from students completing their training in Leipzig were less favourable than their initial responses. Conclusion: The lack of a dental service and Leipzig students’ inability to offer treatment in the presence of disease was associated with frustrations. Practical training should go beyond dental examinations at a long‐term care facility and include the opportunity for dental treatment. Personnel and equipment required for mobile treatment exceed resources available at most German dental schools.  相似文献

Background

Harmful algal blooms deteriorate the services of aquatic ecosystems. They are often formed by cyanobacteria composed of genotypes able to produce a certain toxin, for example, the hepatotoxin microcystin (MC), but also of nontoxic genotypes that either carry mutations in the genes encoding toxin synthesis or that lost those genes during evolution. In general, cyanobacterial blooms are favored by eutrophication. Very little is known about the stability of the toxic/nontoxic genotype composition during trophic change.

Results

Archived samples of preserved phytoplankton on filters from aquatic ecosystems that underwent changes in the trophic state provide a so far unrealized possibility to analyze the response of toxic/nontoxic genotype composition to the environment. During a period of 29 years of re-oligotrophication of the deep, physically stratified Lake Zürich (1980 to 2008), the population of the stratifying cyanobacterium Planktothrix was at a minimum during the most eutrophic years (1980 to 1984), but increased and dominated the phytoplankton during the past two decades. Quantitative polymerase chain reaction revealed that during the whole observation period the proportion of the toxic genotype was strikingly stable, that is, close to 100%. Inactive MC genotypes carrying mutations within the MC synthesis genes never became abundant. Unexpectedly, a nontoxic genotype, which lost its MC genes during evolution, and which could be shown to be dominant under eutrophic conditions in shallow polymictic lakes, also co-occurred in Lake Zürich but was never abundant. As it is most likely that this nontoxic genotype contains relatively weak gas vesicles unable to withstand the high water pressure in deep lakes, it is concluded that regular deep mixing selectively reduced its abundance through the destruction of gas vesicles.

Conclusions

The stability in toxic genotype dominance gives evidence for the adaptation to deep mixing of a genotype that retained the MC gene cluster during evolution. Such a long-term dominance of a toxic genotype draws attention to the need to integrate phylogenetics into ecological research as well as ecosystem management.

The fresh water polyp Hydraforms buds which develop a foot at their base and separate from the parent. In the strain H. vulgaris (Zürich), various compounds including phorbolesters, diacylglycerols, cantharidin and Li+-ions were found to prevent foot formation at the bud's base. Therewith, the bud transforms into a branch which persists at the parent. Other strains were found to be unaffected by such treatments. Here we show that a reduced Ca2+-ion concentration of the culture medium causes branch formation in the H. vulgaris (Zürich) strain but not in the other strains tested. However, all strains tested transformed their buds into branches when the medium was enriched with Ba2+ and Sr2+ ions. We suggest that the various treatments either reduce the internal concentration of Ca2+-ions by stimulating Ca2+-ion export or compete with Ca2+-ions at theirtarget. H. vulgaris (Zürich) isthe most sensitive strain tested and appears to have the most efficient Ca2+-pumps. This appears to be necessary forthese animals derived from a lake which is extremely rich in Ca2+ ions.  相似文献

The occurrence of toxic cyanobacterial blooms is a serious problem for fast‐developing countries in Africa, such as Ethiopia, that are struggling with significant degradation of the natural environment and limited access to water of good quality. Research undertaken on Lake Tana in Ethiopia between 2009 and 2011 was intended to assess the seasonal threat from cyanobacteria and to select methods for tracking of this threat in the future. The cyanobacterial genus Microcystis was found to be present throughout the monitoring period, and M. aeruginosa was determined as the dominant species. Moreover, in all samples, toxigenic cyanobacteria with the potential to produce microcystins were detected. High levels of microcystins, ranging from 0.58 to 2.65 μg L?1, were detected each November, which indicates that in the postrainy season, water usage should be limited. The correlation between concentrations of chlorophyll‐a and microcystins suggested that chlorophyll‐a could be used as an indicator of the potential presence of cyanobacterial‐derived hepatotoxins in Lake Tana in the future. Furthermore, for quick quantitative confirmation of the presence of microcystins, a simple and rapid ELISA test was recommended.  相似文献

Lake Naivasha, an important inland water ecosystem and a crucial freshwater resource in the Great African Rift Valley, has displayed clear signals of degradation in recent decades. We studied the phytoplankton composition and biomass levels in the period 2001–2013 and noted a progressive increase in the occurrence of potentially toxic cyanobacteria. Analyses for the presence of cyanotoxins such as microcystins (MC), cylindrospermopsin (CYN) and anatoxin-a (ATX-a) were carried out on samples collected in 2008–2013. Among the cyanotoxins tested, low concentrations of MC were detected in the lake. This is the first record of the occurrence of MC in Lake Naivasha. For the first time, molecular phylogenetic investigations of field clones of cyanobacteria from Lake Naivasha were carried out to establish the taxa of the dominant species. Amplification of the aminotrasferase (AMT) domain responsible for cyanotoxin production confirmed the presence of the mcyE gene belonging to the microcystin synthesis gene cluster in field samples containing Microcystis and Planktothrix species. These findings suggest that toxin producing cyanobacteria could become a threat to users of this over-exploited tropical lake in the near future.  相似文献

Five polymorphic microsatellite loci were isolated in a gregarious endoparasitoid, Cotesia glomerata, using an enrichment protocol. We detected two to 15 alleles per locus in a sample of 50 female wasps collected from a field population near Zürich, Switzerland. The observed heterozygosities ranged from 0.21 to 0.92. These markers are currently being used in our studies of sex determination in this species.  相似文献
