The most common wild yeasts infecting pressed baker's yeast in Great Britain are Candida tropicalis, C. krusei, C. mycoderma, Trichosporon cutaneum, Torulopsis candida and Rhodotorula mucilaginosa. Wild yeasts are readily detected and quantitatively estimated by plating infected baker's yeast on lysine agar, which permits of only limited growth of baker's yeast.
Morphology of wild yeast colonies on lysine agar is affected by duration of incubation, location in the agar plate, and sometimes by temperature of incubation, density of infection and numbers of baker's yeast cells present. It is therefore possible to identify each species by at least one characteristic type of colony produced under specified conditions. Cinemicrographic Study of the Development of Subsurface Colonies of Staphylococcus aureus in Soft Agar 下载免费PDF全文
Cinemicrographic studies revealed that the development of elongated subsurface colonies of Staphylococcus aureus in soft agar (<0.2% agar) originated with a colony-forming unit of about 10 to 20 cells. It was then observed that small clusters of 3 to 12 cells broke off from the main colony unit and drifted away under the combined influence of gravity and Brownian motion. Once the downward or slightly sideward motion of the small clusters ceased, the clusters would continue to increase in size; at the same time, additional small clusters broke off, and the cycle was repeated until the entire colony was formed. Displacement and velocity measurements were made on the drifting small clusters. When compared with the dimensional growth rate and geometry of the subsurface colony, these showed that a correlation existed between the movement and velocity of the small clusters and the subsequent colony development. A relationship between the role of gravity reported in these results and the development of spherical colonies after rotation on a clinostat is suggested. 相似文献

研究pH、温度、供氧状况、投菌量、氨氮初始浓度等理化因子对海洋硝化细菌去除氨氮效果的影响。结果表明,各种理化因子对硝化细菌去除氨氮效果均有明显影响,海洋硝化细菌的最佳作用条件是pH 8.5,25℃,好氧,对氨氮的去除效果随着投菌量的增加而增强。在适宜条件下,海洋硝化细菌对海水中氨氮具有较好的去除效果。研究海洋硝化细菌在去除氨氮过程中D IN之间相互转化关系发现,在处理系统中随着NH4 -N含量的降低,NO2--N、NO3--N含量持续增加,大约有66.57%的D IN在海洋硝化细菌作用过程中以其他形式脱离了处理系统。 相似文献

Whole colonies of Bacillus cereus, B. megaterium, B. mycoides CN2495, Corynebacterium hofmanni NCTC1938, Escherichia coli, Lactobacillus acidophilus NCIB1899, Nocardia graminis NCTC4728, Pseudomonas viscosa, and Serratia marcescens were prepared for scanning electron microscopic examination by freeze-drying and metal-coating. The arrangement of individual cells within colonies could be seen. Cells of Bacillus colonies tended to be longer than in liquid culture and irregular in shape and to give the appearance of branching. B. megaterium colonies frequently had a dense covering film. Colonies of gram-negative bacteria consisted of fairly short rods covered by much adherent extracellular material. L. acidophilus had colonies comprised of densely packed, well-oriented rods. C. hofmanni colonies contained coccobacilli, packed together. Correlations were observed between plano-convex colony form and densely packed cells, rough colony form and random arrangement of well-separated microorganisms, and irregular colony edge and tendency of cells to grow out from the colony in filaments. 相似文献

Existing media designed for selective isolation of clinically important members of the genus Yersinia were found to be unsatisfactory for the growth and isolation of Yersinia pestis. We report the development of a new selective agar medium (termed BIN) that supports the growth of Y. pestis. The development of the formulation of this medium was based on a fluorescence screening system designed for monitoring bacterial growth on semisolid media, using a green fluorescent protein-expressing strain. High-throughput combinatorial experiments can be conducted for the quantitative evaluation of the effect of different medium components on growth. Generation of fluorescence plots in this system, using microplates, allowed the quantitative evaluation of the growth rate of Y. pestis EV76 cultures in different agar compositions. The final BIN formulation is based on brain heart infusion agar, to which the selective agents irgasan, cholate salts, crystal violet, and nystatin were introduced. It was found that BIN agar is more efficient in supporting colony formation and recovery of Y. pestis than are the conventional semisolid media MacConkey agar and Yersinia-selective agar (cefsulodin-irgasan-novobiocin agar). The advantage of BIN over other media has been also demonstrated in recovering virulent Y. pestis from the mixed bacterial populations found in decaying carcasses of infected mice. The BIN medium is suggested as a selective medium for isolation and recovery of Y. pestis from various backgrounds. 相似文献

Using the medium of ZoBell as a basal formulation, we developed a medium to determine the oxidative-fermentative metabolism in marine bacteria. Different oxidative and fermentative collection strains, as well as a group of unidentified marine bacteria, were tested on modified ZoBell medium and other conventional media. In general, superior results were obtained with the modified ZoBell medium since it supported good bacterial growth and gave more clear-cut oxidative reactions. 相似文献

In a comparison of Lowenstein's medium with two agar media, 7H10 without and with the addition of Triton WR 1339, 4,100 sputa were planted on all three, yielding 803 (19.6%) positive cultures. The addition of Triton WR 1339 to the 7H10 medium made it possible to observe the cord formation directly on the isolation plates and did not alter, in any respect, the results obtained with the basic 7H10 medium. The agar media produced much earlier and more positive cultures than Lowenstein's medium. A greater number of cultures showed drug resistance on Lowenstein's medium as compared with the agar media. 相似文献

Summary: Bacterial colonies grown on objects embedded in agar sometimes show a tendency to become confluent. Glass coverslips and polytetrafluorethylene (Teflon) discs were inoculated with Staphylococcus lactis and completely embedded in serum agar. If the surface of the agar were flooded with broth before the plates were incubated the colonies had a tendency to run together, but not if the plates were incubated after thorough drying. Drying also prevented confluence occurring when broth had been injected on to the surface of the embedded object. Drying the poured plate probably acts by removing fluid squeezed out at the interface. 相似文献

A decrease in the concentration of K+ ions below 3 mM in agar medium which also contained starch, casein hydrolysate, MgSO4, and K2HPO4 changed the growth pattern of Fusarium solani illuminated in diurnal 12-h light/12-h dark cycles from zonation to a feathery growth mode. In well-aerated broth cultures, good growth of Renibacterium salmoninarum was obtained in a serum-free medium consisting of 1% peptone, 1% yeast extract, and 0.1% l-cysteine (PYC broth). In contrast, serum or charcoal is required for growth on agar medium. Charcoal treatment of broth media, either before bacterial inoculation or during growth, increased the growth of R. salmoninarum, whereas the surfactants Tween 20 and Tween 80 inhibited growth. l-Cysteine was essential for optimal growth. Other organic sulfur compounds, such as d-cysteine, l-methionine, homocysteine, homocysteine thiolactone, and reduced glutathione, supported only lower levels of growth, while cystine and dithiothreitol did not allow growth. 相似文献
