The European periwinkle snail, Littorina littorea was discovered in Pictou, NS, Canada in 1840. This snail’s subsequent rapid, conspicuous spread south from Pictou along the Canadian maritime coast and then along the New England and mid-Atlantic coast to New Jersey, its virtual absence in pre-European contact deposits, and its close association with human mechanisms of transport from Europe are among the clearest evidence for a human-mediated introduction. However, molecular genetic data have been proposed as evidence that L. littorea’s occurrence in North America was not the result of a human introduction by Wares et al. (Ecol Lett 5:577–584, 2002). Reexamination of these genetic data and reexamination of all other data available reveal that a human-mediated introduction of L. littorea is the simplest explanation of its occurrence in North America. The refutation of the human-mediated introduction of L. littorea was premature.  相似文献

This study forms part of a larger project in which five planimetric parameters have been used to study changes in the digestive epithelium of Littorina littorea under different environmental and physiological conditions. Our aim was to examine the effect of the reproductive cycle on these parameters in order to assess their usefulness as indicators of stress. A one way anova shows that the absolute parameters of mean epithelial thickness (MET), mean diverticulum radius (MDR) and mean luminal radius (MLR) vary significantly depending on the time of the year. This variation is highly correlated to the amount of digestive tissue present and hence negatively correlated to the reproductive state because gonad and digestive tissue volume are inversely related. Consequently, these parameters are not good indicators of stress. However, whereas the absolute size of the digestive acinus varies with the reproductive state of the animal, the MET, MDR and MLR retain the same proportions and therefore the ratios MET/MDR and MLR/MET remain constant. This makes them useful indicators of stress because they are independent of intrinsic variables such as the reproductive cycle.  相似文献

The rapid range southward expansion of the periwinkle Littorina littorea from the Canadian maritimes has fueled a long-running debate over whether this species was introduced to North America by human activity. A reappraisal of the mitochondrial DNA sequence evidence finds considerable endemic allelic diversity in the American population. The degree of endemic genetic diversity is higher than expected from human-mediated colonization, but not so much to suggest that it survived the last glacial maximum in America. Coalescent estimates of population divergence agree that colonization of America preceded European contact. A reappraisal of the ITS nuclear sequence data finds extensive recombination. Taking this recombination into account strengthens the genetic case against human-mediated introduction. Finally, a reappraisal of conflicting allozyme studies from the 1970’s supports a claim of limited divergence between American and European populations. This is consistent with post-glacial colonization, but the allozyme data cannot distinguish between natural or human-mediated colonization. Taken as a whole, the DNA sequence data supports the many sub-fossil reports of an American L. littorea population in the Canadian maritimes that preceded even the first visits by the Vikings.  相似文献

Specimens of the prosobranch Littorina littorea (L., 1758) collected along the East Frisian North Sea coast in summer 1993 exhibited alterations of the pallial oviduct termed as intersex in response to tributyltin (TBT) pollution. The range of TBT body burden was between 150.9 and 1289.5 μg as Sn kg−1 (dry wt.). Five stages of intersex development (0–4) could be distinguished and are documented with scanning electron micrographs. In stages 2–4, which can be found in the direct vicinity of harbours and marinas, the morphological malformations of the oviduct inhibit successful copulation and capsule formation, resulting in sterilization. The intersex index (ISI, calculated as the average intersex stage of a population) and the average prostate length of females were used as parameters for the determination of intersex intensities in the populations. Both indices show significant and positive correlations to the TBT body burden of L. littorea and are promising parameters for TBT biomonitoring. A comparison of TBT bioconcentration factors with populations from England and France indicates that the threshold concentration for intersex development is in the range of 15 ng TBT as Sn/l. Morphometric analyses of the midgut gland revealed no significant differences between sampling stations. In the ovary a retardation and blockage of maturation (atresia) was observed in populations close to harbours. Lytic processes in ovary follicles were observed not only at TBT exposed sites but also at reference stations.  相似文献

A recent increase in the abundance of cattails (Typha spp.) in North American wetlands has been anecdotally linked with hybridization between Typha latifolia and Typha angustifolia. In this study, we used molecular genetic markers (microsatellites) to investigate whether the hybrid lineage (Typha × glauca) is restricted to The Great Lakes region, or exists across a much broader spatial scale. We also investigated the possibility of backcrossing and genetic introgression in natural populations. Parental species could be distinguished from one another based on the distribution of alleles at six microsatellite loci. Species identification based on genetic data corresponded well with species identifications based on leaf width, a key morphological trait that can distinguish the two parental species. We found that hybrids occur in Ontario, Quebec, New Brunswick, and Nova Scotia, but we did not detect hybrids in Maine. F1s are more abundant than backcrossed or intercrossed hybrids, although we also found evidence of backcrossing, particularly in Ontario. This indicates that hybrids are fertile, and are therefore potential conduits of gene flow between the parental species. Further work is needed to determine whether T. × glauca is particularly successful in the Great Lakes region relative to other areas in which the two parental species co-exist, and to assess whether introgression may lead to increased invasiveness in the species complex.  相似文献

The phylogeography of the endangered tideland snail Batillaria zonalis in the Japanese and Ryukyu Islands was analyzed on the basis of nucleotide sequences of a mitochondrial gene for cytochrome oxidase c subunit I (COI). Extremely low genetic diversity was found in populations at both the northern and southern boundaries of the geographic distribution of this species in Japan, i.e., Sendai Bay and Iriomotejima Island, respectively, which might be attributed to the population bottleneck due to historical environmental variations and/or the recent foundation of populations in the marginal part of the inhabitable range. Most populations contained unique rare haplotypes, and significant genetic differentiation on the whole was shown, while no clear geographic genetic structure was detected between the Japanese and Ryukyu Islands or over the distribution area of B. zonalis in Japan, with the exception of significant genetic divergences in Ago Bay in the central part of Honshu and the southern part of Okinawajima Island.  相似文献

For many plant species in eastern North America, short observed seed dispersal distances (ranging up to a few tens of meters) fail to explain rapid rates of invasion and migration. This discrepancy points to a substantial gap in our knowledge of the mechanisms by which seeds are dispersed long distances. We investigated the potential for white-tailed deer (Odocoileus virginianus Zimm.), the dominant large herbivore in much of eastern North America, to disperse seeds via endozoochory. This is the first comprehensive study of seed dispersal by white-tailed deer, despite a vast body of research on other aspects of their ecology. More than 70 plant species germinated from deer feces collected over a 1-year period in central New York State, USA. Viable seeds included native and alien herbs, shrubs, and trees, including several invasive introduced species, from the full range of habitat types in the local flora. A mean of >30 seeds germinated per fecal pellet group, and seeds were dispersed during all months of the year. A wide variety of presumed dispersal modes were represented (endo- and exozoochory, wind, ballistic, ant, and unassisted). The majority were species with small-seeded fruits having no obvious adaptations for dispersal, underscoring the difficulty of inferring dispersal ability from diaspore morphology. Due to their broad diet, wide-ranging movements, and relatively long gut retention times, white-tailed deer have tremendous potential for effecting long-distance seed dispersal via ingestion and defecation. We conclude that white-tailed deer represent a significant and previously unappreciated vector of seed dispersal across the North American landscape, probably contributing an important long-distance component to the seed shadows of hundreds of plant species, and providing a mechanism to help explain rapid rates of plant migration.Electronic Supplementary Material Supplementary material is available in the online version of this article at  相似文献

Biological invasions are typically the outcome of complex patterns of introduction, establishment, and spread, and genetic methods are excellent tools to resolve such histories for non-native organisms. The mealy plum aphid, Hyalopterus pruni, is an invasive pest of dried plum in California. We examined nine microsatellite loci and DNA sequences from three mitochondrial genes (1,148 bp) in populations throughout the native and invaded ranges of H. pruni to assess key invasion parameters, including geographic origins of invasive populations, number of introductions, and levels of genetic diversity and gene flow. Our results provide evidence for multiple invasions of H. pruni into North America, suggesting that aphids in California may have been introduced from Spain, and aphids in the eastern United States and Vancouver, Canada were likely introduced from central or northern Europe. H. pruni populations in California were characterized by low genetic diversity relative to native populations, while the two other North American populations were less genetically impoverished. Gene flow among introduced populations was low, but does appear to occur with some regularity. These findings provide a framework for more detailed studies of H. pruni, but also represent a model for how population genetics approaches can be used to study invasion biology and aid the development of optimized management methods for agricultural pests. Photosynthetic responses were quantified for two Zostera japonica Aschers. and Graebn. populations from the northern and southern limits of distribution exposed to a range of salinities along the Pacific Coast of North America. Plants were collected from Padilla Bay, Washington (northern) and Coos Bay, Oregon, USA (southern) and cultured together in experimental tanks at 3 salinities (5, 20 and 35) under saturating irradiance for 3 weeks. Subsequently, photosynthesis–irradiance (P vs. E curves) relationships for leaf segments from the two populations were assessed using an oxygen electrode system. We found no evidence for diel rhythms in either light saturated photosynthesis (Pmax) or dark respiration (Rd). For the Padilla Bay population, Pmax ranged from 192 to 390 μmol O2 g DW−1 h−1; for the Coos Bay population Pmax ranged from 226 to 774 μmol O2 g DW−1 h−1. Photosynthetic maxima of the Coos Bay plants occurred at a salinity of 20, whereas salinity had no effect on the photosynthetic maxima of the Padilla Bay plants. There were significant differences in leaf tissue Rd among salinity treatments but the two populations responded similarly to salinity. North American populations of Z. japonica are best adapted to intermediate salinities, displaying minimum Rd rates, lower compensation irradiance, higher saturation irradiance, and greater Pmax rates at a salinity of 20. Additionally, the southern population may be better adapted to southward expansion along the Pacific Coast and changes associated with global climate change.  相似文献

Pedigree analysis has clear benefits for the genetic management of threatened populations through the evaluation of inbreeding, population structure and genetic diversity. The use of pedigrees is usually restricted to captive populations and few examples exist of their exclusive use in managing free-ranging populations. One such example is the management of the takahe (Porphyrio hochstetteri), a highly endangered, flightless New Zealand rail at risk from introduced mammalian predators and habitat loss. During the 1980’s and 90’s, as part of the takahe recovery programme, birds were translocated from the sole remnant population in Fiordland to four offshore islands from which introduced predators had been eradicated. The subsequent “island” population, now numbering 83 and thought to be at carrying capacity, has been closely monitored since founding. Detailed breeding records allow us to analyse the island pedigree, which is up to 7 generations deep. Gene-drop analysis indicated that 7.5% of genetic diversity has been lost over the relatively short timeframe since founding (2.1 generations on average; total genetic founders = 31) due to both a failure to equalise founder representation early on and subsequent disproportionate breeding success (founder equivalents = 12.5; founder genome equivalents = 6.6). A high prevalence of close inbreeding will have also impacted on genetic diversity. Predictions from pedigree modelling suggest that 90% genetic diversity will be maintained for only 12 years, but by introducing a low level of immigration from the Fiordland population and permitting the population to grow, 90% GD could be maintained over the next 100 years. More generally, the results demonstrate the value of maintaining pedigrees for wild populations, especially in the years immediately after a translocation event.  相似文献

During the century following its initial introduction in 1886, the Australian tree Melaleuca quinquenervia (Myrtaceae) dispersed from a few introduction points to occupy over 200,000 ha, primarily in historic Everglades wetlands of southern Florida. Cellulose acetate gel electrophoresis (CAGE) was used to investigate the allozyme diversity and population genetic structure of 208 individuals in a dozen populations resulting from this invasion. The analyses showed that these populations have a high (82%) rate of polymorphic loci and an average of 2 alleles/locus. There was substantial heterozygosity (mean He = 0.356), which concords well with recent studies reporting a greater number of introduction events and sources than generally recognized. The introduction history and distributional patterns within Florida have led to geographic structuring (GST = 0.419) in which the Gulf Coast metapopulation has a greater effective number of alleles and greater heterozygosity than the Atlantic Coast metapopulation. The gene diversity in M. quinquenervia was comparable to other tropical woody species. Its strong population divergence was reminiscent of pioneer species and consistent with its status as a plant invader in Florida.  相似文献

Habitat alteration, fragmentation and destruction as a consequence of human impact are a global phenomenon. Here we document changes in genetic variation in the marsh orchid Anacamptis palustris as a consequence of such habitat changes. We examined historical specimens that were collected during the nineteenth and early twentieth centuries, prior to the most recent massive habitat changes affecting this species. Sequences of a hypervariable region in the plastid DNA, located in the tRNALEU intron, from herbarium vouchers were compared with those from a near-exhaustive survey of the extant A. palustris populations on the Italian peninsula. It was found that private haplotypes and alleles found in small, extant populations were once widespread and more common in historic populations and that alleles, once present in historic populations, have gone extinct. In addition, genetic differentiation among populations has increased over time and haplotype frequencies significantly differ among historic and extant populations. These results document that human induced habitat changes have reduced genetic diversity and increased the importance of random genetic drift in this species. It is concluded that the analysis of herbarium specimens may provide important insights into changes of genetic diversity over time and may be critical for correct inference of the evolutionary history of rare and endangered species.  相似文献

Variation in lateral awn length and pulvini development is described for A. pansa and A. gypsophila. The two species are distinguished by lateral awn development. Within each species, forms are based on pulvini development. Three new forms are described (A. pansa f. contracta; A. gypsophila f. gypsophiloides and f. diffusa), A. dissita is reduced to a form of A. pansa, and A. tehuacanensis is considered synonymous with A. pansa f. dissita. In this study, we investigated the genetic diversity of a collection of 136 Medicago truncatula lines from 10 Tunisian natural populations collected in well-defined locations and in various ecological conditions of soil, salinity and water availability. The genetic diversity was evaluated using a set of 18 microsatellites (SSRs), representing the 8 chromosomes of M. truncatula. A neutrality test showed that 7 SSRs were non-neutral with evidence of balancing selection. The 11 neutral SSRs revealed a geographical pooling with the Tunisian Dorsale axis restricting migration of alleles. The 7 non-neutral alleles demonstrate a correlation with rainfall, altitude and salinity environmental variables suggesting that these SSRs are linked to genes involved in water use efficiency, resistance to salinity or adaptation to altitude, and that there is local adaptation of M. truncatula to these variables. This demonstrates that the choice of so-called neutral markers should be carefully evaluated in population genetic studies. This study illustrates the genetic diversity occurring in natural Tunisian populations of M. truncatula and describes the first collection of this species dedicated to natural variation involved in adaptation to the environment. Fatty acid synthesis in bacteria and plants is catalysed by a multi-enzyme fatty acid synthetase complex (FAS II) which consists of separate monofunctional polypeptides. Here we present a comparative molecular genetic and biochemical study of the enoyl-ACP reductase FAS components of plant and bacterial origin. The putative bacterial enoyl-ACP reductase gene (envM) was identified on the basis of amino acid sequence similarities with the recently cloned plant enoyl-ACP reductase. Subsequently, it was unambiguously demonstrated by overexpression studies that theenvM gene encodes the bacterial enoyl-ACP reductase. An anti-bacterial agent called diazaborine was shown to be a specific inhibitor of the bacterial enoyl-ACP reductase, whereas the plant enzyme was insensitive to this synthetic antibiotic. The close functional relationship between the plant and bacterial enoyl-ACP reductases was inferred from genetic complementation of anenvM mutant ofEscherichia coli. Ultimately,envM gene-replacement studies, facilitated by the use of diazaborine, demonstrated for the first time that a single component of the plant FAS system can functionally replace its counterpart within the bacterial multienzyme complex. Finally, lipid analysis of recombinantE. coli strains with the hybrid FAS system unexpectedly revealed that enoyl-ACP reductase catalyses a rate-limiting step in the elongation of unsaturated fatty acids.  相似文献

Didemnum sp. A is a colonial ascidian with rapidly expanding populations on the east and west coasts of North America. The origin of Didemum sp. A is unknown. Populations were first observed on the northeast coast of the U.S. in the late 1980s and on the west coast during the 1990s. It is currently undergoing a massive population explosion and is now a dominant member of many subtidal communities on both coasts. To determine Didemnum sp. A's current distribution, we conducted surveys from Maine to Virginia on the east coast and from British Columbia to southern California on the west coast of the U.S. between 1998 and 2005. In nearshore locations Didemnum sp. A currently ranges from Eastport, Maine to Shinnecock Bay, New York on the east coast. On the west coast it has been recorded from Humboldt Bay to Port San Luis in California, several sites in Puget Sound, Washington, including a heavily fouled mussel culture facility, and several sites in southwestern British Columbia on and adjacent to oyster and mussel farms. The species also occurs at deeper subtidal sites (up to 81 m) off New England, including Georges, Stellwagen and Tillies Banks. On Georges Bank numerous sites within a 230 km2 area are 50-90% covered by Didemnum sp. A; large colonies cement the pebble gravel into nearly solid mats that may smother infaunal organisms. These observations suggest that Didemnum sp. A has the potential to alter marine communities and affect economically important activities such as fishing and aquaculture.  相似文献

A BAC library of 30,228 clones with an average insert size of 102 kb was constructed in the grass Brachypodium sylvaticum. Brachypodium has a simple genome, similar in size and repetitive DNA content to that of rice, and is more closely related than rice both to the major temperate cereals wheat and barley, and to the forage grasses. The library represents 6.6 genome equivalents, implying a 99.9% probability of recovering any specific sequence. The library was arrayed onto two high-density colony filters, which were screened with heterologous DNA probes from rice chromosome nine and from syntenous regions of wheat, barley, maize and oat. The construction of Brachypodium BAC contigs revealed that synteny between rice, wheat and Brachypodium was largely maintained over several regions of rice chromosome nine. This suggests that Brachypodium will be a useful tool in the elucidation of gene content in agronomically important cereal crops, complementing rice as a grass genome model.  相似文献
