A variegated position effect on the autonomous gene, purple, has been studied enzymologically in Drosophila melanogaster. Sepiapterin synthase, the enzyme system associated with pr +, was examined for activity in different developmental stages of the fly. The results indicate that T(Y:2) pr c5, cn/prc4 cn flies (flies in which pr + has been translocated and which exhibit variegation) have a reduced amount of enzyme activity as compared with both Oregon-R and pr 1 flies. This reduction in activity was not found in larval stages, which suggests that the inactivation process probably occurs in late larval or early pupal stages. The phenotype of the variegated adult has white eyes with red-colored spots and patches where drosopterins occur. The phenotype of the fly carrying the translocation is modified by the presence of additional Y chromosomes. This extends the observation from other systems that extra heterochromatin acts to suppress the variegated position effect. The advantages of studying the variegation by measuring enzyme activity, as well as the phenotypic expression, are several; for example, the developmental time at which variegation occurs may be estimated even though drosopterin synthesis is not occurring.The Oak Ridge National Laboratory is operated by Union Carbide Corporation for the Department of Energy under Contract No. W-7405-eng-26.  相似文献

Summary The mutagenic efficiency of ionizing radiations has been tested on different lines of Drosophila melanogaster. It has been shown that differential lethal effects are obtained when irradiated females from different lines are mated to flies carrying heterozygous lethal genes. The results seem not to be attributable to differential expression of the lethality in the various crosses performed with the irradiated flies. This might suggest that gene activity is involved in the expression of the mutagenic effects of radiations.  相似文献

【背景】实蝇的竞争可能发生在生活史的各个阶段,但未见卵和蛹的种间竞争的相关报道。【方法】将橘小实蝇与番石榴实蝇的卵及蛹按照相应的比例或龄期分别进行混合培养,分析2种实蝇的卵或蛹混合后的发育历期和存活率是否受到彼此影响。【结果】橘小实蝇与番石榴实蝇的卵同龄等量混合,对彼此卵的历期和孵化率无影响。当橘小实蝇和番石榴实蝇的蛹为新鲜蛹时,分别与比它们蛹龄大1 d的对方蛹混合,蛹的羽化率分别为(87.67±3.61)%和(84.33±2.56)%,显著小于对照,说明在此混合蛹处理中,后化蛹者的发育可能受到先化蛹者的抑制。【结论与意义】总体上,在卵和蛹期,橘小实蝇与番石榴实蝇未产生竞争作用;但不排除蛹期竞争的可能性,这种竞争可能存在于特定的蛹期。  相似文献

A method for maintaining anin vivo culture ofEntomophthora muscae (C) Fres. on its original host, adult carrot flies (Psila rosae F.), is described. The lethal time for adult carrot flies was greatly influenced by temperature, both for infected and for uninfected flies. In the range 8.2°C–20.2°C the LT50 for infected flies was about 5.4 times shorter than the estimated average life-span for uninfected flies. The discharge of primary spores was also strongly dependent on temperature. The total number of primary spores discharged per fly at 100% RH and in darkness ranged between 1.2×104 and 9.6×104 with a mean of 5.1×104.   相似文献

Summary We undertook a deletional analysis of the gypsy retrotransposon in order to determine which sequences of the element are required for its mutagenic effect. We show that a phenotype indistinguishable from that ofy 2 flies can be generated by transformingy flies with a construct containing theyellow gene and a gypsy element located at the same insertion site inyellow as found iny 2 flies. When flies are transformed with similar constructs in which increasing amounts of the 5 transcribed untranslated region of gypsy have been removed, either a partialy 2 revertant or a completely revertant phenotype is obtained. These results yield direct proof that the region of gypsy to which thesu(Hw) protein binds is required for the generation of mutant phenotypes by this retrotransposon.  相似文献

Flies that are homozygous for the recessive autosomal mutation bent wingshave a limited ability to fly and are less tolerant of high temperatures than normal flies in both the egg and puparial stages. The differences between the mutant and normal flies were found sufficient to be the basis of a genetic sexing strain. Genetic sexing strains were created using translocations of the autosome bearing the wild-type allele of bent wings(chromosome 2) to the Y chromosome, and crossing male flies carrying the translocation to mutant bent wingsfemales. In the resulting strain, the females were homozygous for the bent wingsmutation and the males were phenotypically normal for wing characters. Several translocations were recovered after irradiation, but only one translocation involving chromosome 2 was both stable and expressed in a stock that was vigorous enough for long-term viability. Unfortunately, all stocks containing the translocation showed high levels of temperature-dependent lethality, including, inexplicably, both males and females. Translocation stocks showing this effect included bent wings, another second chromosome mutation, white marks, and an otherwise normal stock. This phenomenon is probably rare, as it has not been reported before. It is likely that bent wingscould be suitably used with another translocation.  相似文献

In order to elucidate whether the aphidRhopalosiphum padi is low quality food for the carabid beetleAgonum dorsale, as it has previously been found to be for cereal spiders, we performed a series of experiments using fruit fliesDrosophila melanogaster as a standard alternative prey (‘control prey’): 1) Prey preference, 2) Aphid consumption for satiated and starved beetles, 3) Egg production on different diets and 4) Preference experiments with fruit flies coated with a taste of different prey types. Other alternative prey types used were earthworms and sciarid midges, all potential prey in the field.A. dorsale has a low preference for aphids compared to fruit flies. Apparently there is an upper limit to the consumption of aphids independent of hunger and much below the beetles' total food demand. Egg production on a pure diet of aphids is lower than on a pure diet of fruit flies; it is extremely low on a pure diet of earthworms; the highest fecundity is found on a mixed diet. As fruit flies coated with a taste of aphids or sciarid midges are less prefered than flies covered with a taste of fruit flies there may be a chemical factor, acting through taste, involved in determining prey preference.  相似文献

The effects of the parasitic phorid fly, Pseudacteon tricuspis Borgmeier, on the competitive interactions between the red imported fire ant, Solenopsis invicta Buren, and a native North American ant, Forelius pruinosus (Roger), were investigated in the laboratory. P. tricuspis is a highly host-specific endoparasitoid of S. invicta workers that is currently being reared and released as a biological control agent of S. invicta in the US. We tested the effect of P. tricuspis on the colony growth rate of S. invicta when S. invicta was forced to compete with F. pruinosus for a protein resource (freeze-killed crickets) in laboratory competition arenas. In addition to colony growth rate, we quantified the effect of the phorid flies on the foraging rate of S. invicta. Though S. invicta significantly reduced its foraging rate in the presence of the phorid flies, we did not detect an effect of the flies on colony growth rate. Possible explanations for these results include behavioral compensation by S. invicta for the presence of the flies. We present these laboratory results in light of a literature search indicating that laboratory tests of biological control agent efficacy are good predictors of field efficacy. We conclude that P. tricuspis alone is unlikely to suppress S. invicta populations in the field by reducing their competitive ability.  相似文献

目的 习得性无助(learned helplessness)是指动物经历了不可逃避性刺激后而产生的无助状态。它对随后实验动物的运动能力、生理状态等多方面都会产生影响,也是人类抑郁症的有效动物模型之一。目前已在多种动物模型中建立了研究习得性无助神经机制的行为实验范式,包括黑腹果蝇(Drosophila melanogaster)。之前的研究发现,经历过不可逃避高温惩罚的果蝇在活跃程度等方面有显著改变。然而,果蝇的习得性无助状态是否也影响学习认知能力,目前仍然未知。本文对果蝇空间位置学习能力是否受到习得性无助状态影响进行了研究。方法 此研究中,我们完善实验范式,分别检测果蝇在新范式中的空间位置学习行为和习得性无助行为,以及经过长时程随机刺激后果蝇空间位置学习能力的变化。结果 野生型果蝇可在此范式中展现空间位置学习能力与习得性无助状态,而经过长时程不可逃避刺激训练的果蝇,空间位置记忆有显著下降。结论 此研究完善了果蝇习得性无助行为实验范式,实验结果表明习得性无助对果蝇空间位置记忆存在影响。这将对进一步加深对动物习得性无助行为的理解,进而其发生机制研究起到推进作用。  相似文献

Summary The mus308 mutants of Drosophila have previously been demonstrated to be defective in an enzyme that is designated Nuclease 3 (Boyd et al. 1990b). In this study that enzyme is shown to be present in mitochondria of both wild-type flies and embryos. Since the mus308 mutants are hypersensitive to DNA crosslinking agents, Nuclease 3 is potentially required for resistance of the mitochondrial genome to such agents. In support of this hypothesis, electron microscopic studies of mus308 mutant flies that had been exposed to nitrogen mustard revealed an increased frequency of mitochondrial abnormalities. Further investigation of the defect at the enzymological level revealed that the mutants possess a new nuclease activity that is apparently a modified form of the wild-type protein. In the earlier study, enzyme extracts from mus308 mutants were found to lack an enzyme with a pl of approximately 6.2. More precisely defined assay conditions in this study revealed the appearance of a new nuclease activity with a higher pI in extracts from mutants. This observation, together with the finding that only the normal enzyme form is present in heterozygous individuals, supports the hypothesis that the mus308 locus is not the structural gene for the enzyme. Rather, the mus308 gene product is necessary for Nuclease 3 to assume the lower pI. Nuclease 3 has been partially purified and characterized from wild-type embryos. Its activity is stimulated by Mg++ and ATP. Optimum activity is found at a pH of 5.5 and a NaCl concentration of 50–100 mM. Nuclease 3 exhibits a temperature optimum of 42°C and is insensitive to N-ethylmaleimide. The enzyme is probably membrane-associated because it exhibits a strong tendency to aggregate and detergent is required for full solubilization.  相似文献

Sepiapterin synthase, the enzyme system responsible for the synthesis of sepiapterin from dihydroneopterin triphosphate, has been partially purified from extracts of the heads of young adult fruit flies (Drosophila melanogaster). The sepiapterin synthase system consists of two components, termed enzyme A (MW 82,000) and enzyme B (MW 36,000). Some of the properties of the enzyme system are as follows: NADPH and a divalent cation, supplied most effectively as MgCl2, are required for activity; optimal activity occurs at pH 7.4 and 30 C; the K m for dihydroneopterin triphosphate is 10 µm; and a number of unconjugated pterins, including biopterin and sepiapterin, are inhibitory. Dihydroneopterin cannot be used as substrate in place of dihydroneopterin triphosphate. Evidence is presented in support of a proposed reaction mechanism for the enzymatic conversion of dihydroneopterin triphosphate to sepiapterin in which enzyme A catalyzes the production of a labile intermediate by nonhydrolytic elimination of the phosphates of dihydroneopterin triphosphate, and enzyme B catalyzes the conversion of this intermediate, in the presence of NADPH, to sepiapterin. An analysis of the activity of sepiapterin synthase during development in Drosophila revealed the presence of a small amount of activity in eggs and young larvae and a much larger amount in late pupae and young adults. Sepiapterin synthase activity during development corresponds with the appearance of sepiapterin in the flies. Of a variety of eye color mutants of Drosophila melanogaster tested for sepiapterin synthase activity, only purple (pr) flies contained activity that was significantly lower than that found in the wild-type flies (22% of the wild-type activity). Further studies indicated that the amount of enzyme A activity is low in purple flies, whereas the amount of enzyme B activity is equal to that present in wild-type flies.This work was supported by research grants from the National Institutes of Health (AM03442) and the National Science Foundation (PCM75-19513 A02). Although Leishmania metacyclic promastigotes are generally considered resistant to human complement, studies of in vitro-cultured axenic stationary promastigotes using serum concentrations that approximate physiological plasma conditions indicate complement sensitivity. Natural Leishmania infection is caused by sand fly-inoculated promastigotes, whose complement resistance has not been analyzed systematically. We compared Leishmania susceptibility to human complement in L. infantum promastigotes derived from in vitro cultures and from sand flies. Phlebotomus perniciosus sand flies were fed with axenic promastigotes, L. infantum-infected U-937 cells, or spleen cells from L. infantum-infected hamsters. On selected days post-feeding, flies were dissected and promastigotes isolated; in addition, axenic promastigotes were obtained from culture at equivalent days of growth. In near-physiological serum concentration and temperature conditions, measurement of real-time kinetics of propidium iodide uptake showed that 90% of axenic- and sand fly-derived promastigotes were rapidly killed by complement. We found no substantial differences between promastigotes from axenic culture, those isolated from flies on different post-feeding days, or those generated in flies fed with distinct inocula. The results indicate that Leishmania susceptibility to human complement is independent of promastigote developmental stage in the sand fly mid-gut and in axenic culture.  相似文献

The biochemical action of the Notch locus whose mutants cause morphological aberrations in flies, viz., notches of wings and bristle multiplication, has been analyzed (1) by the addition to the food medium of enzyme inhibitors causing phenocopies of Notch and (2) by comparison of enzyme activity patterns of Notch mutants with different degrees of phenotypic expression. Notch phenocopies were induced by inhibitors of enzyme activities in two biochemical pathways: (1) the de novo pyrimidine synthesis by 5-methylorotate (inhibitor of dihydroorotate dehydrogenase) and (2) the choline shunt by amobarbital (inhibits choline dehydrogenase) and methoxyacetate (inhibits sarcosine dehydrogenase). The inhibition of de novo pyrimidine synthesis prevents the production of deoxyuridine-5-phosphate, the substrate for the synthesis of thymidine-5-phosphate via thymidylate synthase, whereas the inhibition of the choline shunt prevents the production of HCHO groups and glycine, both of which are involved in the synthesis of 5,10-methylenetetrahydrofolate, which is a cofactor of thymidylate synthase. It was already known that the inhibition of the latter enzyme in vivo induces Notch phenocopies. Notch mutants with a strong morphological expression show low enzyme activities for dihydroorotate dehydrogenase and choline dehydrogenase. Both are flavoprotein enzymes linked to the respiratory chain. The correspondence between the low enzyme activities in Notch mutants with a strong morphological expression and the phenocopying effect of antimetabolites on these enzymes in the two biochemical pathways involved strongly suggests that the morphological effects of Notch on flies are a consequence of lowered activities of choline dehydrogenase and dihydroorotate dehydrogenase.  相似文献

The addition of a novel host plant to a phytophagous insect’s diet may result in subsequent host-plant specialisation, and is believed to be a key cause for speciation in this trophic group. In northern Britain, the tephritid fly Tephritis conura has experienced a unique host-plant expansion, from the melancholy thistle Cirsium heterophyllum to the marsh thistle C. palustre. Here, we examine whether the incorporation of C. palustre in the repertoire of British T. conura flies has caused genetic divergence between populations infesting the old host and the novel host, and how British populations differ from populations infesting C. heterophyllum in continental Europe where C. palustre is not infested. No evidence for restricted gene flow among British C. palustre and C. heterophyllum flies was found. Significant differentiation between British and continental T. conura was found at only one allozyme locus, hexokinase, and caused by a new allele, Hex_95. Hexokinase is related to host-race formation in continental European flies infesting C. heterophyllum and C. oleraceum, and might be linked to loci determining host choice. Based on morphological and phenological data from previous studies, we suggest that T. conura in Britain has adapted to the novel host but that host-race formation is impeded by similar plant phenologies.  相似文献

Summary Viruses were detected in 32 of the culture medium supernates from 42Drosophila melanogaster cell lines. The picornaviruses DCV, DPV and DAV, which also have been found in natural and laboratory populations of flies, were detected as well as the diplornavirus DXV which has never been noted inDrosophila stocks. Two of the 21 samples of commercial fetal bovine sera that were examined were found to be capable of causing infections of DXV in virus-freeDrosophila flies. It is inferred that the insects used to initiate the cell lines, the sera used in the culture media, and accidental contaminations account for the presence of viruses in the continously culturedDrosophila cell lines.  相似文献

Electron microscopic analysis of Drosophila melanogaster (w1118) ovarian cells has shown that stressful heat treatment of flies causes the appearance of electron-dense granules and large lysosomes in the cytoplasm of ovarian cells. These changes are not due to the presence of the endosymbiotic bacteria Wolbachia, as these changes were observed in both infected and uninfected flies. Essential envelope disturbances and other structural alterations have been revealed in the bacteria present in the ovarian cell cytoplasm of the flies. Some of the fly embryos died after heat shock; however, the bacteria retain their typical morphology in survived embryos. Endosymbionts did not change their localization in ovarian cells and in early fly embryos; they closely interacted with mitochondria and endoplasmic reticulum after the heat-shock treatment of flies. The performed study has shown that the high temperature affects both the host and the endosymbiont, but does not change the character of their structural interaction. Original Russian Text M.V. Summary Drosophila melanogaster flies were transformed with a yp1-Adh fusion gene with 890 bp of yp1 5 flanking sequence. In an Adh - background these flies show a stage, tissue and sex-specific pattern of alcohol dehydrogenase (ADH) activity characteristic of yolk protein genes. ADH activity is not present in dsx D/dsx pseudomales indicating that this fragment contains sites where the dsx gene product exerts its effect. Transformed male flies do not exhibit ADH activity when injected with 20-hydroxyecdysone while synthesis of native yolk proteins is induced. Thus the hormone inducibility and sex regulation have been separated in this construct.  相似文献

A field experiment with Drosophila melanogasterrevealed that when flies encounter a particular food type soon after emergence, the probability of their subsequently being attracted to such a resource is increased. In this experiment, the length of time flies experienced their postemergence environments was under the control of the flies themselves. The experiment thus realistically mimicked one form of experiential effect that may be important in nature. A theoretical model is developed which shows that enhanced adult preferences for the types of resources fed on as larvae can substantially increase the degree of host-based genetic subdivision within a polyphagous population.  相似文献
