Citrulline is an efficient hydroxyl radical scavenger that can accumulate at concentrations of up to 30 mm in the leaves of wild watermelon during drought in the presence of strong light; however, the mechanism of this accumulation remains unclear. In this study, we characterized wild watermelon glutamate N-acetyltransferase (CLGAT) that catalyses the transacetylation reaction between acetylornithine and glutamate to form acetylglutamate and ornithine, thereby functioning in the first and fifth steps in citrulline biosynthesis. CLGAT enzyme purified 7000-fold from leaves was composed of two subunits with different N-terminal amino acid sequences. Analysis of the corresponding cDNA revealed that these two subunits have molecular masses of 21.3 and 23.5 kDa and are derived from a single precursor polypeptide, suggesting that the CLGAT precursor is cleaved autocatalytically at the conserved ATML motif, as in other glutamate N-acetyltransferases of microorganisms. A green fluorescence protein assay revealed that the first 26-amino acid sequence at the N-terminus of the precursor functions as a chloroplast transit peptide. The CLGAT exhibited thermostability up to 70 degrees C, suggesting an increase in enzyme activity under high leaf temperature conditions during drought/strong-light stresses. Moreover, CLGAT was not inhibited by citrulline or arginine at physiologically relevant high concentrations. These findings suggest that CLGAT can effectively participate in the biosynthesis of citrulline in wild watermelon leaves during drought/strong-light stress.  相似文献

Summary Young seedlings of English Oak, Quercus robur L., and Silver Birch, Betula verrucosa Ehrl., were subjected to a number of consecutive periods during which water was withheld. During one 14-day period leaf-and soil-water potentials and leaf- and root-solute potentials of two groups of plants were sampled at noon of each day. One group of plants was watered every day while water was withheld from the other group. Solute accumulation in roots and leaves of oak seedlings subjected to water stress resulted in maintenance of turgor and high leaf conductance as the soil dried. In birch seedlings turgor was only maintained by stomatal closure at high soil water potential.Fourteen consecutive water stress cycles greatly reduced the growth of birch seedlings but had little effect on oak seedlings other than to alter root morphology. Water stress treatment resulted in the production of long thin roots in this plant. Stomatal behaviour in oak and birch seedlings during the 14-week stress period was consistent with observed changes in leaf water and solute potentials. Daily solute accumulation in oak leaves was presumably responsible for the maintenance of plant growth as water potentials fell.  相似文献

Proteomic analysis of rice leaves during drought stress and recovery   总被引:6,自引:0,他引:6  
Three-week old plants of rice (Oryza sativa L. cv CT9993 and cv IR62266) developed gradual water stress over 23 days of transpiration without watering, during which period the mid-day leaf water potential declined to approximately -2.4 MPa, compared with approximately -1.0 MPa in well-watered controls. More than 1000 protein spots that were detected in leaf extracts by proteomic analysis showed reproducible abundance within replications. Of these proteins, 42 spots showed a significant change in abundance under stress, with 27 of them exhibiting a different response pattern in the two cultivars. However, only one protein (chloroplast Cu-Zn superoxide dismutase) changed significantly in opposite directions in the two cultivars in response to drought. The most common difference was for proteins to be up-regulated by drought in CT9993 and unaffected in IR62266; or down-regulated by drought in IR62266 and unaffected in CT9993. By 10 days after rewatering, all proteins had returned completely or largely to the abundance of the well-watered control. Mass spectrometry helped to identify 16 of the drought-responsive proteins, including an actin depolymerizing factor, which was one of three proteins detectable under stress in both cultivars but undetectable in well-watered plants or in plants 10 days after rewatering. The most abundant protein up-regulated by drought in CT9993 and IR62266 was identified only after cloning of the corresponding cDNA. It was found to be an S-like RNase homologue but it lacked the two active site histidines required for RNase activity. Four novel drought-responsive mechanisms were revealed by this work: up-regulation of S-like RNase homologue, actin depolymerizing factor and rubisco activase, and down-regulation of isoflavone reductase-like protein.  相似文献

Water availability is a critical determinant for the growth and ecological distribution of terrestrial plants. Although some xerophytes are unique regarding their highly developed root architecture and the successful adaptation to arid environments, virtually nothing is known about the molecular mechanisms underlying this adaptation. Here, we report physiological and molecular responses of wild watermelon (Citrullus lanatus sp.), which exhibits extraordinarily high drought resistance. At the early stage of drought stress, root development of wild watermelon was significantly enhanced compared with that of the irrigated plants, indicating the activation of a drought avoidance mechanism for absorbing water from deep soil layers. Consistent with this observation, comparative proteome analysis revealed that many proteins induced in the early stage of drought stress are involved in root morphogenesis and carbon/nitrogen metabolism, which may contribute to the drought avoidance via the enhancement of root growth. On the other hand, lignin synthesis-related proteins and molecular chaperones, which may function in the enhancement of physical desiccation tolerance and maintenance of protein integrity, respectively, were induced mostly at the later stage of drought stress. Our findings suggest that this xerophyte switches survival strategies from drought avoidance to drought tolerance during the progression of drought stress, by regulating its root proteome in a temporally programmed manner. This study provides new insights into the complex molecular networks within plant roots involved in the adaptation to adverse environments.  相似文献

In wild species of almond (Prunus spp.), the activities of ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), and glutathione reductase (GR), as well as the levels of ascorbate/glutathione pools and H2O2 were subjected to water deficit and shade conditions. After 60 days of water shortage, the species were subjected to a rewatering treatment. During water recovery, leaves exposed to sunlight and leaves under shade conditions of about 20–35% of environmental irradiance were sampled. After 70 days without irrigation, mean predawn leaf water potential of all the species fell from −0.32 to −2.30 MPa and marked decreases in CO2 uptake and transpiration occurred. The activities of APX, MDHAR, DHAR, and GR increased in relation to the severity of drought stress in all the wild species studied. Generally, APX, MDHAR, DHAR, and GR were down-regulated during the rewatering phase and their activities decreased faster in shaded leaves than in sun-exposed leaves. The levels in total ascorbate, glutathione, and H2O2 were directly related to the increase in drought stress and subsequently decreased during rewatering. The antioxidant response of wild almond species to drought stress limits cellular damage caused by reactive oxygen species during periods of water deficit and may be of key importance for the selection of drought-resistant rootstocks for cultivated almond.  相似文献

Abstract

Lanthanum is one of the most abundant elements in rare earths enriched fertilizers and is supposed to be one of the main responsible of the effects of such fertilizers on crops. In this work, the effect of lanthanum nitrate on H2O2 production, lipid peroxidation, ascorbate and glutathione content, and on the activity of cytosolic ascorbate peroxidase, dehydroascorbate reductase, monodehydroascorbate reductase, and glutathione reductase in Lycopersicon esculentum L. cv. Marmande during drought stress was evaluated. The results confirmed that treatments of tomato plants with lanthanum nitrate affect the antioxidant cellular defences and that lanthanum toxicity is dependent on the way of treatment. The stimulation of antioxidant systems did not induce any improvement in drought stress responses in tomato but seemed to be only a consequence of the unbalance in cell metabolism due to the treatment with lanthanum nitrate.  相似文献

Two cultivars ofCicer arietinum L. with contrasting sensitivities to water stress, cv. C 214 (relatively resistant) and cv. 130 (susceptible) were compared for their protein content and soluble nitrogen (sol-N) under water stress. During water stress shoots of the cv. C 214 showed an increase (over the control) in both protein and sol-N content. The total nitrogen (total-N) also increased in the cv. C 214 while remaining almost unaffected in the cv. G 130. Cultivar C 214 shoots recorded lower protein/sol-N ratios. The results have been discussed in relation to resistance to water stress.  相似文献

Wheat, staple food crop of the world, is sensitive to drought, especially during the grain-filling period. Water soluble carbohydrates (WSCs), stem reserve mobilization and higher invertase activity in the developing grains are important biochemical traits for breeding wheat to enhance tolerance to terminal drought. These traits were studied for three accessions of Triticum dicoccoides(a tetraploid wheat progenitor species) - acc 7054 (EC 171812), acc 7079 (EC 171837) and acc 14004 (G-194-3 M-6 M) selected previously on the basis of grain filling characteristics. Check wheat cultivars- PBW-343 (a popular bread wheat cultivar for irrigated environments) and C-306 (widely adapted variety for rain-fed agriculture) were also included in this set. Analysis of variance revealed significant genotypic differences for the content of water soluble carbohydrates, activity of acid invertase and alkaline invertase. Acc 7079 was found to be a very efficient mobilizer of water soluble carbohydrates (236.43 mg g-1 peduncle DW) when averaged over irrigated and rain-fed conditions. Acid invertase activity revealed marked genotypic differences between wild and cultivated wheats. Alkaline invertase activity was highest in Acc 7079 when pooled across both the environments. On the whole, acc 7079 qualifies as a suitable donor for enhancing tolerance of bread wheat to terminal drought. The association of physio-biochemical differences observed with grain filling attributes on one hand and molecular markers on the other could be of use in improving wheat for water stress conditions.  相似文献

This study investigated several aspects related to drought tolerance in arbuscular mycorrhizal (AM) soybean plants. The investigation included both shoot and root tissues in order to reveal the preferred target tissue for AM effects against drought stress. Non-AM and AM soybean plants were grown under well-watered or drought-stressed conditions, and leaf water status, solute accumulation, oxidative damage to lipids, and other parameters were determined. Results showed that AM plants were protected against drought, as shown by their significantly higher shoot-biomass production. The leaf water potential was also higher in stressed AM plants (-1.9 MPa) than in non-AM plants (-2.5 MPa). The AM roots had accumulated more proline than non-AM roots, while the opposite was observed in shoots. Lipid peroxides were 55% lower in shoots of droughted AM plants than in droughted non-AM plants. Since there was no correlation between the lower oxidative damage to lipids in AM plants and the activity of antioxidant enzymes, it seems that first the AM symbiosis enhanced osmotic adjustment in roots, which could contribute to maintaining a water potential gradient favourable to the water entrance from soil into the roots. This enabled higher leaf water potential in AM plants during drought and kept the plants protected against oxidative stress, and these cumulative effects increased the plant tolerance to drought.  相似文献

Six lines of sorghum ( Sorghum bicolor L. Moench) with differing drought resistance (IS 22380, ICSV 213, IS 13441 and SPH 263, resistant and IS 12739 and IS 12744, susceptible) were grown under field conditions in the semi-arid tropics and analysed for proline and nitrate reductase activity (NRA; EC 1.6.6.1) during a mid-season drought. The resistant lines accumulated high levels of proline, while the susceptible lines showed no significant proline accumulation. Most of the proline was accumulated after growth of the plants had ceased. In a separate greenhouse experiment, most of the proline was found in the green rather than the fired portions of leaves. The levels returned to that of irrigated controls within 5 days of rewatering. Proline levels increased as leaf water potential and relative water content fell, and there was no apparent difference among the different sorghum lines with change in plant water status. Susceptible lines accumulated less proline than resistant lines as leaf death occurred at higher water potentials. Proline accumulation may, however, contribute to the immediate recovery of plants from drought. Leaf NRA reached high levels at about 35 days after sowing in both the stressed and irrigated plants, after which it declined. The decline in NRA was more pronounced in the stressed than in the irrigated plants and closely followed changes in the growth rate. Upon rewatering, NRA increased several-fold in all the lines and, in contrast to proline accumulation, genotypic differences in NRA were small, both during stress and upon rewatering. The high sensitivity of NRA to mild drought stress was reflected in the rapid decline of activity with small changes in leaf water potential and relative water content. The results are discussed in the light of a possible role for proline during recovery from drought, and the maintenance of NRA during stress and its recovery upon rewatering.  相似文献

Experiments were performed to test the hypothesis that succulents “shift” their method of photosynthetic metabolism in response to environmental change. Our data showed that there were at least three different responses of succulents to plant water status. When plant water status of Portulacaria afra (L.) Jacq. was lowered either by withholding water or by irrigating with 2% NaCl, a change from C3-photosynthesis to Crassulacean acid metabolism (CAM) occurred. Fluctuation of titratable acidity and nocturnal CO2 uptake was induced in the stressed plants. Stressed Peperomia obtusifolia A. Dietr. plants showed a change from C3-photosynthesis to internal cycling of CO2. Acid fluctuation commenced in response to stress but exogenous CO2 uptake did not occur. Zygocactus truncatus Haworth plants showed a pattern of acid fluctuation and nocturnal CO2 uptake typical of CAM even when well irrigated. The cacti converted from CAM to an internal CO2 cycle similar to Peperomia when plants were water-stressed. Reverse phase gas exchange in succulents results in low water loss to carbon gain. Water is conserved and low levels of metabolic activity are maintained during drought periods by complete stomatal closure and continual fluctuation of organic acids.  相似文献

Wild progenitors of common wheat are a potential source of tolerance to biotic and abiotic stresses. We conducted a glasshouse pot experiment to study genotypic differences in response to drought stress in a collection of 180 accessions of Aegilops and Triticum along with one tolerant and one sensitive control variety. Several physiological traits and chlorophyll fluorescence parameters were evaluated. Our findings indicated that drought significantly reduced shoot fresh (59.45%) and dry (50.83%) weights, stomatal conductance (41.52%) and maximum photosynthetic capacity (41.06%), but increased initial fluorescence (28.10%). Drought stress also decreased the chlorophyll content, relative water content and maximum quantum efficiency by 14.90, 12.13 and 11.42%, respectively. Principal component analysis of the 182 individuals identified three components that explained 57.61 and 61.68% of the total variation in physiological and photosynthetic traits under control and stress conditions, respectively. When grouped into the 12 species tested, the three top components explained 78.22% of the total variation under drought. The means comparison, stress tolerance index and biplot analysis identified five accessions with superior tolerance to drought. Remarkably, four species of wild relatives—Ae. cylindrica (DC genome), Ae. crassa (DM genome), Ae. caudata (C genome) and T. urartu (Au genome)—responded well to drought stress with a lower percentage decline for most traits and high values for the first two components. The potential of these species offers further opportunities for analysis at the molecular and cellular levels to confront with drought stress through a physiological mechanism.  相似文献

Cuttings of Populus kangdingensis and Populus cathayana, originating from high and low altitudes in the eastern Himalaya, respectively, were examined during one growing season in a greenhouse to determine their responses to drought stress (soil moisture decreased from 100 to 55 or 25 % field capacity). Compared to control plants grown under 100 % field capacity, those poplars grown under 55 and 25 % field capacity possessed lower increases in height and stem diameter, and higher contents of soluble sugars, free proline, malondialdehyde (MDA) and hydrogen peroxide, and higher activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX) and glutathione reductase (GR). Compared with P. cathayana with greater leaf area, P. kangdingensis with greater root/shoot ratio exhibited lower MDA and H2O2 contents, higher soluble sugar and free proline contents, and higher activities of CAT, SOD, POD, APX and GR. These results suggested that P. kangdingensis was more drought tolerant than P. cathayana.  相似文献

采用CIRAS-2型便携式光合作用系统,设置充足供水、轻度、中度和重度胁迫4种土壤水分处理,研究土壤干旱和强光胁迫对3年生黄栌苗木叶片光合作用的影响.结果表明:土壤干旱胁迫对黄栌叶片的光合性能参数影响显著,随着土壤水分胁迫的加剧,黄栌叶片的光合速率、蒸腾速率和光量子效率明显降低,光补偿点增高,水分利用效率在轻度水分胁迫(土壤相对含水量60%~65%)下最高;在强光(有效辐射强度1000~1800μmol.m-2.s-1)范围内,叶片光合速率和水分利用效率均较高,而对弱光的光能吸收和光量子效率较低,并随土壤水分胁迫加剧明显下降.  相似文献
