Summary Ten alfalfa cultivars were used to study the effects of nodulation and variations in resistance to alfalfa sickness. The alfalfa
seedlings were planted in sick soil with three treatment,,i.e.: pasteurized sick soil inoculated with Rhizobium ‘Nitragin’ which served as the control, the inoculated non-pasteurized sick
soil and the non-inoculated non-pasteurized sick soil. None of the alfalfa cultivars were immune from the sickness. Cultivar
‘Anik’ fromMedicago falcata was among the most resistant cultivars. Three Phytophthora root rot resistant cultivars including ‘Agate’, ‘Apollo’ and ‘Ramsey’
were not resistant to the disease. Alfalfa inoculated with Rhizobium showed greatly improved seedling growth. Correlation
coefficients showed that those alfalfa cultivars more resistant to alfalfa sickness produced more dry weight. Dry weight increase
due to nodulation (82%) had more than compensated for the loss of dry weight due to alfalfa sickness (33%). The present study
suggested that the poor growth of alfalfa on sick soil was attributed to both the soil borne pathogens and the poor nodulation
of alfalfa, probably due to the absence of effective Rhizobium in sick soil. Highly significant differences were also obtained
among olfalfa cultivars for plant dry weight in the inoculation treatment. Selection for effective Rhizobium strains and for
alfalfa genotypes which are resistant to alfalfa sickness and are high in nitrogen fixation rates could improve alfalfa yield

利用光学和电子显微镜,从组织细胞学水平系统研究了冠盘二胞Marssonina coronaria在苹果抗、感病品种叶片上的侵染过程及侵染后寄主细胞的超微结构特征。结果表明:冠盘二胞的侵入和定殖过程可以分为6个阶段:孢子萌发与芽管形成、附着胞形成、侵入细胞角质层、在叶肉细胞内产生吸器、菌丝在叶肉细胞间和细胞内扩展、分生孢子盘形成。随着菌丝扩展,受侵寄主细胞出现细胞壁加厚,细胞壁降解,质壁分离,叶绿体内淀粉粒、嗜饿颗粒积累,叶绿体基粒片层瓦解,线粒体空泡化等现象。在不同抗性的苹果品种上,分生孢子萌发率差别不明 相似文献

Fall dormancy results in decumbent, slow shoot growth of alfalfa (Medicago sativa L.) in autumn and reduced shoot regrowth rates after herbage removal in summer. Although fall dormancy is used to predict alfalfa adaptation, we possess a poor understanding of the biological mechanisms underlying fall dormancy. Our objective was to examine growth and carbohydrate metabolism of suspension cell cultures derived from contrasting alfalfa cultivars that genetically differed in fall dormancy. Suspension cells were grown in B5h media containing 2% sucrose. Cells derived from fall non-dormant plants accumulated sugars more rapidly after transfer to fresh media and to higher concentrations than did cells derived from fall dormant alfalfa cultivars. Dark respiration rates of cells derived from non-dormant plants were similar to those derived from fall dormant plants when growth was limited at low cell sugar concentrations. However, both cell growth and dark respiration rates increased in cells derived from non-dormant cultivars in response to greater cell sugar concentrations. High growth rates of cells derived from rapid growing, fall non-dormant alfalfa cultivars were associated with rapid sugar uptake and higher cell respiration rates when compared to cells derived from dormant alfalfa cultivars. 相似文献

AbstractBegomoviruses are economically important plant viruses with a wide host range infecting numerous vegetables, legumes and fibre crops worldwide. Pakistan cotton suffered with cotton leaf curl disease (CLCuD) epidemic in the 1990s resulting in huge crop losses. Since then CLCuD has become a continuous threat to cotton industry in Pakistan. Several monopartite begomovirus species and satellite molecules are associated with CLCuD. There is threat of disease spreading to other parts of the world which are currently free from CLCuD due to agricultural trade and spread of vector, Bemisia tabaci. Natural resistance in tetraploid cotton, Gossypium hirsutum against CLCuD is very limited and is not durable under field conditions due to emergence of new viral strains. Genetically engineered resistance towards CLCuD has had limited success due to transformation limitations in cotton and the diversity of begomoviruses. Molecular approaches and conventional breeding are underway for developing cotton with CLCuD resistance. There is need to introgress multiple resistance genes in cotton to produce durable resistance which would lost several years. Chilli is one of the spices used to enhance the flavour and taste of cooked food. Fungal diseases are the main biological constraints in chilli production, and Alternaria leaf spot disease caused by Alternaria alternata is one of the most devastating diseases of chilli. One of the effective and environmentally friendly ways to control this disease is introgress resistance from wild relative/varieties to the cultivated one. The first step towards introgression of resistance genes is to screen the chill germplasm for leaf spot resistance. In the current study, we screened the chilly germplasm and identified the sources of leaf spot resistance, which can be harnessed in resistance breeding programmes. 相似文献

Abstract Fifty-six indigenous and 29 exotic mulberry varieties were screened against powdery mildew, Myrothecium leaf spot, Pseudocercospora leaf spot for a period of three years under field conditions. The percent disease index (PDI) was recorded during the peak season of the diseases. Out of 85 germplasm lines studied four-germplasm lines viz. Thailand lobed (0.43), M. multicaulis (2.63), Italian (2.83) and M. australis (4.56) were found highly resistant; nine lines were resistant; 43 lines were moderately resistant and 29 were susceptible to the disease complex. Powdery mildew showed significant positive correlation with Pseudocercospora leaf spot. Highly resistant varieties may be utilized for future disease resistance breeding programme to evolve multiple disease resistant mulberry varieties. 相似文献

以15个杜鹃花品种为试材,利用扫描电镜观察叶片17项解剖结构,应用变异系数、相关分析和聚类分析对叶片解剖结构进行筛选,采用隶属函数对各品种的耐热性进行综合评价,探讨杜鹃花叶片解剖结构与植株耐热性的关系,为杜鹃花耐热品种选育、引种栽培及栽培区划分提供细胞学理论依据.结果表明: 杜鹃花叶片为典型的异面叶,表皮细胞垂周壁略呈波浪状,气孔为无规则形,仅分布于下表皮,品种之间有16项解剖结构指标差异达到极显著水平,仅下表皮厚度为显著水平;叶片角质层厚度、气孔密度、气孔宽度、栅栏组织厚度和组织结构疏松度是影响耐热性的主要叶片解剖结构性状,而叶片厚度、气孔长度、孔径、气孔开度、海绵组织厚度、栅海比、上下表皮细胞长度和厚度、组织结构疏松度、中脉厚度与植株耐热性的关系不大.15个品种杜鹃花耐热性存在一定差异,其强弱顺序为:松江大桃红>状元红>绿色光辉>粉珍珠>外国红>蓝茵>笔止>大和之春>国旗红>玉玲珑>红珊瑚>宁波红>套瓣朱砂>爱丁堡>琉球红.15个品种杜鹃花抗热性可划分为4类:松江大桃红、状元红和绿色光辉为抗热类型;粉珍珠、外国红、蓝茵、笔止、大和之春、国旗红和玉玲珑为中度抗热类型;红珊瑚、宁波红、套瓣朱砂和爱丁堡为低抗热类型;琉球红为不抗热类型.杜鹃花耐热性的准确评价需参考形态结构、生理生化、遗传等因素,以及杜鹃花高温受害及受害后恢复状况. 相似文献

目的:本研究是为了观察饮食补充锌减轻酒精性肝病损伤的作用及与HNF-4α的关系。方法:选用成年C57BL/6小鼠40只,按随机数字表分为4组(n=10):正常对照组、酒精中毒组、正常补锌组及酒精补锌组,用不同饮食喂养6个月处死,在正常补锌组和酒精补锌组小鼠饮用水中加入硫酸锌,使锌的含量达到75 mg/L。取各组小鼠肝组织进行病理切片及增殖细胞核抗原(PCNA)免疫组织化学染色,RT-PCR检测肝细胞核因子-4α(HNF-4α)含量,Western blot检测肝组织HNF-4α蛋白表达,检测"肝组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量"。结果:酒精中毒组小鼠HNF-4α转录及表达均明显低于正常对照组,差异具有统计学意义(P<0.05),该组小鼠MDA含量增高,SOD活性下降与正常对照组相比差异有统计学意义(P<0.05);而酒精补锌组小鼠PCNA阳性肝细胞数目及HNF-4α蛋白表达水平明显高于酒精中毒组,差异有统计学意义(P<0.05),该组小鼠SOD活性增加,MDA下降,与酒精中毒组相比差异有统计学意义(P<0.05)。结论:长期酒精喂养导致小鼠氧化还原失衡,而补锌可逆转该状态。我们推测饮食补锌可能是通过增加HNF-4α的转录及表达而增强酒精喂养小鼠的肝再生,因此,饮食补锌可能对酒精性肝病有较好的影响。 相似文献

To investigate physiological and biochemical changes of thrips-resistant alfalfa (Medicago sativa L. cv. Gan-nong No. 9), we aimed at clarifying the response mechanisms of alfalfa against thrips. Medicago sativa L. cv. including thrips-resistant Gan-nong No.9 (G9), thrips-susceptible Gan-nong No.3 (G3) and highly thrips-susceptible WL363HQ (363) were infested with different thrips densities (3, 5, 7 and 9-thrips per branch). The quantitative change in specific nutrients, secondary metabolites, defensive and antioxidant enzymes were measured at seedling stage of the three alfalfa cultivars. The results showed that with the increase of thrips densities, the damage indices, SS, Pro, flavonoids, tannin and H2O2 in G9, G3 and 363 were significantly increased, but PPO and SOD significantly reduced, compared with CK. Furthermore, the tannin and lignin contents of G9 were significantly higher compared to 363, but SP content was significantly lower than G3 and H2O2 content which was further significantly less compared to 363. Correlation analysis observed that the damage index of the three alfalfa cultivars showed a significant positive association with SS, Pro, flavone, tannin, and H2O2 (P < 0.01), while damage index and DW, Chl (a, b, a + b), PPO and SOD showed a significant negative correlation (P < 0.01). Based on principal component comprehensive evaluation, the 5-thrips adults per branch were the critical inoculation threshold for G9 against thrips injury because the score was – 0.048. These results revealed that thrips damage significantly increased the contents of SS, Pro, flavonoids, tannins and H2O2, as well as significantly declined the activities of PPO and SOD in the three cultivars (P < 0.05), moreover, thrips-resistant G9 markedly accumulated lignin content, POD and CAT activity, inhibited Chl (a + b, b) and SP biosynthesis to resist thrips damage. 相似文献

Supra-optimal levels of zinc in primary leaves of Phaseolus vulgaris increased the CO2 compensation point and inhibited net photosynthesis. Leaf morphology was modified: mesophyll intercellular area, stomatal slit length and interstomatal distance were reduced, but stomatal density increased. Internal and stomatal conductances to CO2 diffusion decreased. These changes are discussed in relation to the observed effects on leaf gas exchange and to the previously reported inhibition of different photosynthetic and photorespiratory enzymes. 相似文献

Tomato line LA1478 and Pusa Ruby were resistant to tomato leaf curl virus (TLCV) disease. They registered higher plant height, number of branches, total phenol content and yield per plant than the other cultivars. Variety Peto 86 was tolerant to the disease while the other popular tomato cultivars, i.e. Ace, Early Pack, Money Maker, Prichard and Strain B were highly susceptible to the disease. Plant height and number of branches per plant revealed significantly positive association with fruit yield per plant. The disease index of TLCV exhibited significant negative correlations with plant height, total phenol content and fruit yield per plant – 0–4 and 5–25 adult whiteflies were observed on resistant susceptible cultivars. In the case of epiphytically colonisation by Beauveria bassiana conidia, not all developing hyphae on the leaf surface penetrated the whitefly cuticle. Many of the germ tubes elongated to a short distance before terminating its growth. On the other hand, the rapid staining of tomato tissues injected with B. bassiana conidial suspension indicates that the entomopathogenic fungus was established inside tomato tissues until the end time of the trial. The direct injection with the spore suspension yielded high post-colonisation, where the fungus was recovered from sites distant from the point of inoculation. This indicates that the fungus has the potential to move throughout the plant tissues. Laboratory bioassay of tomato whitefly feeding on tomato tissues containing B. bassiana conidial spores indicates that plant endophytic colonisation with entomopathogenic fungi may reduce insect survival on these plants. LT50 values of the test diet were between three and four days. The mortality of Bemisia tabaci was high in the case of endophytically colonisation compared to epiphytically one (90.0% compared to 10.0% during three days) for whiteflies fed tomato tissues containing 1.5 × 107 B. bassiana spores/ml. Application of B. bassiana as an artificial endophyte inside tomato plants can be an important component in the integrated control of tomato whiteflies. The endophytic colonising can achieve biocontrol effect based on induced disease resistance in plant tissues. According the available references, this is the first report on B. tabaci controlling by plant endophytic treatment. 相似文献

为了明确河南省小麦品种的抗叶锈性及抗叶锈基因的分布,为小麦品种推广与合理布局、叶锈病防治及抗病育种提供依据,本研究利用2015年采自河南省的5个小麦叶锈菌流行小种混合菌株,对近几年河南省16个主栽小麦品种进行了苗期抗性鉴定,然后选用12个小麦叶锈菌生理小种对这些品种进行苗期基因推导,同时利用与24个小麦抗叶锈基因紧密连锁(或共分离)的30个分子标记对该16个品种进行了抗叶锈基因分子检测。结果显示,供试品种苗期对小麦叶锈菌混合流行小种均表现高度感病;基因推导与分子检测结果表明,供试品种可能含有Lr1、Lr16、Lr26和Lr30这4个抗叶锈基因,其中先麦8号含有Lr1和Lr26;郑麦366和郑麦9023含有Lr1;西农979和怀川916含有Lr16;中麦895、偃展4110、郑麦7698、平安8号、众麦1号、周麦16、衡观35和矮抗58含有Lr26;周麦22中含有Lr26,还可能含有Lr1和Lr30;豫麦49-198和洛麦23可能含有本研究中检测以外的其他抗叶锈基因。因此,河南省主栽小麦品种的抗叶锈基因丰富度较低,今后育种工作应注重引入其他抗叶锈性基因,提高抗叶锈性,有效控制小麦叶锈病。 相似文献

A real-time PCR assay was used to quantify the relationship in alfalfa and pea between disease severity and the amount of Aphanomyces euteiches detected in roots. The study included isolates of race 1 and race 2 of the alfalfa pathovar of A. euteiches and an isolate obtained from diseased pea. Spearman rank correlations between pathogen DNA content and disease severity index (DSI) ratings were positive ( ? 0.57) and significant (P 0.0007) for individual alfalfa plants, bulked alfalfa plant samples, and individual pea plants. In all experiments, significantly more pathogen was detected in susceptible populations than in resistant populations. The results clearly demonstrate that resistance to A. euteiches in both alfalfa and pea is characterized by a reduction in pathogen colonization relative to levels observed for susceptible reactions. The assay was very specific for A. euteiches, producing very linear assays with DNA extracted from pathogen isolates obtained from alfalfa, pea, and bean. Possible applications of the assay in conjunction with other real-time PCR assays specific to other legume pathogens are discussed in relation to simultaneous disease screening for multiple plant pathogens and the study of microbial population dynamics in mixed plant infections. 相似文献

Isogenic diploid and tetraploid alfalfa (Medicago sativa L.) was studied with molecular markers to help understand why diploid performance and breeding behavior does not always predict that of tetraploids. In a previous study of partially heterozygous alfalfa genotypes, we detected a low correlation between yields of isogenic diploid (2x) and tetraploid (4x) single-cross progenies, and genetic distances were more highly correlated with yields of tetraploids than diploids. These differences may be related to the level of RFLP heterozygosity expected among progenies derived from heterozygous parents at the two ploidy levels. The objectives of this study were to determine the relationships among genetic distance, forage yield and heterozygosity in isogenic 2 x and 4 x alfalfa populations. Four diploid genotypes were chromosome doubled to produce corresponding isogenic autotetraploids, and these genotypes were mated in 4 × 4 diallels to produce 6 single-cross families at each ploidy level for field evaluation. Allele compositions of parents were determined at 33 RFLP loci by monitoring segregation of homologous restriction fragments among individuals within progenies, and these were used to estimate RFLP heterozygosity levels for all single-cross progenies at both ploidy levels. RFLP heterozygosity rankings were identical between progenies of isogenic diploid and tetraploid parents; but significant associations (P < 0.05) between estimated heterozygosity levels and forage yield were detected only at the tetraploid level. Since tetraploid families were nearly 25% more heterozygous than the corresponding diploid families, inconsistencies in the association between molecular marker diversity and forage yields of isogenic 2 x and 4 x single crosses may be due to recessive alleles that are expressed in diploids but masked in tetraploids. The gene action involved in heterosis may be the same at both ploidy levels; however, tetraploids benefit from greater complementary gene interactions than are possible for equivalent diploids.

研究了9个紫花苜蓿品种在黄土高原丘陵沟壑区根系的发育能力,结果表明,紫花苜蓿侧根发生的主要部位在距地表10-20cm的主根段,50cm以下没有侧根发生,品种间侧根发生总数以Sandili和Ameristand201最大,陇东苜蓿最低;品种间根系体积和生物量的垂直分布,除Sandili,Amerigraze401,Ameristand201在土壤20-30cm层的根系体积的生物量大于10-20cm层的根系体积和生物量外,其余品种表现为从土壤表层到深层逐次递减。品种间根系生物量从Ameristand201最大,为170.54g,陇东苜蓿最低,仅为38.25g,是Ameristand201的22.4%;品种占有的地下资源空间Ameristand201和Sandili较大,陇东苜蓿最小,经聚类分析,参试的9个品种可分为3类,其中Sandili,Amerigraze401和Ameristand201在黄土高原丘陵沟壑区根系发育良好。 相似文献

连续4年对中国28个已通过审定的紫花苜蓿品种的产量及质量性状进行了研究。生育期测定结果表明:达初花期所需天数最少的品种有甘农3号、武功、蔚县、新牧1号、新牧2号、公农2号、甘农1号、准格尔、陇东、无棣和晋南。产量性状分析表明:总干物质产量最高和较高的品种为新疆大叶(35955.6kg/hm^2)、武功、甘农3号、公农2号、新牧2号、新牧1号、肇东、图牧2号、草原2号、龙牧801和龙牧803共11个品种(31088.3~33183.4kg/hm^2);叶茎比最高的品种为龙牧801和无棣。依据对4年总干物质产量、3龄总干物质产量、4年平均日增高、4龄叶面积、4龄总枝条数和叶茎比6个产量性状测定值的不加权平均进行生产性能的聚类分析,认为可将28个品种分为极强、较强、中等、较差和差5个类群。极强类群苜蓿生产能力最强,特征为生长迅速,干物质产量、日增高、枝条数均为最高,叶面积最高或较低;叶茎比为中等;达出花期所需天数为少或多。 相似文献

本研究旨在明确山东省12个小麦主栽品种(系)抗叶锈性及抗叶锈基因,为小麦品种推广与合理布局、叶锈病防治及抗病育种提供依据。利用2015年采自山东省的5个小麦叶锈菌流行小种的混合小种对这些材料进行苗期抗性鉴定,然后选用15个小麦叶锈菌生理小种对这些品种(系)进行苗期基因推导,并利用与24个小麦抗叶锈基因紧密连锁(或共分离)的30个分子标记对其进行抗叶锈基因分子检测。结果显示,山东省12个主栽小麦品种(系)苗期对该省2015年的5个小麦叶锈菌混合流行小种均表现高度感病。通过基因推导与分子检测发现,济南17含有Lr16,矮抗58和山农20含有Lr26,其余济麦系列、烟农系列、良星系列等9个品种(系)均未检测到所供试标记片段。此外,本研究还对山东省3个非主栽品种进行了检测,结果发现,中麦175含有抗叶锈基因Lr1和Lr37,含有成株抗性基因;皖麦38只检测到Lr26,济麦20未检测到所供试标记片段。综合以上结果,山东省主栽小麦品种(系)所含抗叶锈基因丰富度较低,尤其不含有对我国小麦叶锈菌流行小种有效的抗锈基因,应该引起高度重视,今后育种工作应注重引入其他抗叶锈基因,提高抗叶锈性。 相似文献

Accessions of wild Lycopersicon spp. and selected Fl hybrid tomato cultivars were compared for their resistance to three whitefly-transmissible geminiviruses: Indian tomato leaf curl virus (ITmLCV) and tomato yellow leaf curl viruses from Sardinia (TYLCV-Sar) and Senegal (TYLCV-Sen). The resistance of different plant lines was expressed in different ways but in most instances a given line reacted similarly to graft inoculation with the three viruses. L. pimpinellifolium LA1478 produced as much virus antigen, assessed by triple antibody sandwich-ELISA, as the susceptible cv. Moneymaker but developed only very mild symptoms and is therefore tolerant of infection. In L. hirsutum LA1777 and L. peruvianum CMV-INRA, very mild or no symptoms developed but antigen concentrations were substantially less than in Moneymaker. L. chilense LA1969 remained symptomless and its antigen concentration was < 1% of that in Moneymaker. Symptoms were mild or barely evident in the Fl hybrid cultivars. Cultivars Tyking and Fiona had antigen concentrations about 5–10% of those of Moneymaker, whereas TY20, Top 21 and Tyger had intermediate antigen concentrations. In a few instances, the extent to which virus accumulation was restricted depended on the challenge virus. Accumulation of TYLCV-Sen in TY20, Top 21 and Tyger was less affected than that of the other two viruses, and accumulation of TYLCV-Sar in accessions LA1777 and CMV-INRA was less affected than that of TYLCV-Sen or ITmLCV. Tissue-printing tests showed that ITmLCV and TYLCV-Sen antigens were confined to phloem tissue. In Tyking, the number of virus antigen-containing phloem traces and the antigen content of individual traces were less than in Moneymaker but the partitioning of antigen between internal and external phloem was unaffected. 相似文献
