Field surveys over 2 years in contiguous beds of the seagrassesZostera capricorni andPosidonia australis showed that the green algaCodium duthieae was consistently more abundant inZ. capricorni than inP. australis. In 1 year, mature plants were also more abundant at the boundary between the seagrass beds than in either bed. Field experiments and programmes of sampling were used to investigate three potential explanations for the unusual distribution of this alga: (1) that the availability of substrata suitable for attachment of the alga differed between the two seagrass beds; (2) that mortality of matureC. duthieae differed between the seagrass beds; and/or (3) that the intensity of recruitment was different in the two seagrass beds.C. duthieae plants were exclusively epibionts of the bivalveAnadara trapezia. Detailed sampling showed that the abundance ofA. trapezia was similar in both seagrass beds and that the distribution of bivalves suitable as substrata forC. duthieae plants was not obviously related to proximity to the boundary between the beds. Two experiments investigated the survival ofC. duthieae plants in each bed. In the first, matureC. duthieae plants transplanted into theP. australis bed suffered similar rates of mortality to plants which were disturbed and moved within theZ. capricorni bed or which were left undisturbed in theZ. capricorni bed. Fewer of the host bivalves were recovered from theZ. capricorni bed, however, indicating that the mechanism of mortality differed between the beds, hosts being more frequently dislodged in theZ. capricorni bed. Removal of the leaves of the seagrasses had consistently greater effects on near-bottom current velocities in theZ. capricorni bed than in theP. australis bed and significantly increased mortality ofC. duthieae in theZ. capricorni bed. Survival of plants was greater in plots of artificial leaves ofP. australis placed in theZ. capricorni bed than in plots of the naturalZ. capricorni leaves or plots where the natural leaves were removed. Most mortality in theZ. capricorni bed was due to dislodgement of the alga and its bivalve substratum. Corresponding manipulations of leaves in theP. australis bed had consistently smaller effects on survivorship of both the alga and its host. Patterns in the recruitment of the alga most clearly reflected the distribution of adults.C. duthieae recruits were 5 times more abundant in theZ. capricorni bed and at the boundary between the two beds than in theP. australis bed. The results demonstrate how habitat structure, provided by the canopy of leaves of the two species of seagrass, can have contrasting effects on the recruitment and mortality of a macroalga. In the case ofC. duthieae, it appears that the differential pattern of recruitment is the primary determinant of the distribution of adult plants.  相似文献

Habitat preference of juveniles ofLates angustifrons andLates mariae, endemic fish species in Lake Tanganyika, was investigated in the field and in the laboratory. Both species settle on the grass bed after the post-larval pelagic phase.L. angustifrons preferred the short grass bed composed ofVallismeria sp., whileL. mariae preferred the tall grass bed composed ofPotamogeton schweinfurthii. L. angustifrons always rested near the root, keeping a position parallel to the bottom; dark cross bands on the body may be advantageous as camouflage in the short grass bed. On the other hand,L. mariae always stayed in mid-water, keeping a head-down position parallel to a leaf or stem of grass; such posture and longitudinal stripes on the body may be advantageous as camouflage in the tall grass bed. Both species were more active at night, and fed mainly on atyid shrimps.Lates angustifrons shifted its main food from shrimps to fish with growth, as was suggested inL. mariae. Difference in the habitat preference may play an important role in their coexistence.  相似文献

Cellulase was immobilized in a collagen fibril matrix, and no leakage of cellulase from the collagen fibril matrix was observed. The immobilized cellulase was more stable than the native cellulase. The substrate cellulose was hydrolyzed quantitatively with immobilized cellulase. The final reaction product was identified as glucose. Immobilized cellulase was used in a fluidized bed reactor where the pressure drop of the fluidized bed reactor was low and constant. Cellulose was hydrolyzed to glucose by the cellulase-bead fluidized bed reactor. The minimum flow velocity (Umf) was 0.5 cm/sec and the optimum flow velocity of the cellulose hydrolysis was 1 cm/sec.  相似文献

Summary Along the intertidal zone of Kavaratti Lagoon, a seagrass bed, consisting largely of Thalassia and Cymodocea, grows on a substratum of loose coral sand and coral debris. Primary production of this grass bed was estimated from a study of diurnal changes in dissolved oxygen over the bed and experiments were conducted on some isolated plants to determine their rates of oxygen production and consumption. Both these studies showed that photosynthesis of the community during daylight is greater than its respiration during day and night combined.  相似文献

The faunal assemblages of a mussel bed (Mytilus edulis L.) and ambient sandflat were compared to study how a bioherm of suspension feeding organisms affects benthic communities in a tidal flat. During a survey of mussel beds in the Wadden Sea at the island of Sylt (North Sea), a total of 52 macrofaunal species and 44 meiobenthic plathelminth species were detected. They occupied different microhabitats in the mussel bed. 56% of the macrofauna species were dwelling in the sediment beneath the mussels and 42% were epibenthic or epiphytic. The latter were restricted in their occurrence to the mussel bed. Along a transect from the sandflat to the mussel bed the mean species densities of macrofauna did not differ significantly, while abundances were significantly lower in the mussel bed than in the sandflat. The composition of the assemblages shifted from a dominance of Polychaeta in the sandflat to Oligochaeta in the mussel bed. Surface filter-feeding polychaetes of the sandflat (Tharyx marioni) were displaced by deposit feeding polychaetes under the mussel cover (Capitella capitata, Heteromastus filiformis). The total meiobenthic density was lower and single taxa (Ostracoda, Plathelminthes, Nematoda) were significantly less abundant in the mud of the mussel bed. The plathelminth assemblage was dominated by grazing species (Archaphanostoma agile), and differed in community structure from a sandflat aseemblage. An amensalistic relationship was found between the suspension-feeding mussels and suspension-feeding infauna, while deposit-feeders were enhanced. The presence of epibenthic microhabitats results in a variety of trophic groups co-occurring in a mussel bed. This is hypothesized as trophic group amelioration and described as an attribute of heterotrophic reefs.  相似文献

Oil palm frond parenchyma tissue was used as a solid substrate for the production of laccase via solid‐state fermentation using the white rot fungus Pycnoporus sanguineus. With a rectangular aluminium tray as solid‐state fermentation bioreactor, process parameters such as bed height, moisture and supplemented nitrogen (as urea solution) levels were studied and optimized using a statistical design of experiment. The moisture level exerted a significant effect on the process. The interaction effect observed between bed height and supplemented nitrogen level suggested that uniform distribution of supplemented nitrogen into the substrate bed was important. The proposed regression model sufficiently predicted the process response over the experimental range tested. The optimum parameter combination for laccase production was a 3‐cm bed height, 72% w/w moisture and 0.21% w/v supplemented nitrogen. Laccase productivity remained constant when the tray size was increased from 1.4 to 3.4‐fold.  相似文献

Phlebiopsis gigantea fungus used in biological control of root rot is currently cultivated commercially in disposable, sterilizable plastic bags. A novel packed bed bioreactor was designed for cultivating P. gigantea and compared to the plastic bag method and to a tray bioreactor. The spore viability of 5.4 × 106 c.f.u./g obtained with the packed bed bioreactor was of the same order of magnitude as the viabilities obtained with the other cultivation methods. Furthermore, the packed bed bioreactor was less time and space consuming and easier to operate than the tray bioreactor.  相似文献

This study characterized the microbial community and population dynamics in an anaerobic hybrid reactor (AHR) treating cassava starch wastewater. Methanogens and nonmethanogens were followed during the start-up and operation of the reactor, and linked to operational and performance data. Biomass samples taken from the sludge bed and packed bed zones of the AHR at intervals throughout the operational period were examined by 16S rRNA fluorescence in situ hybridization (FISH). The start-up seed and the reactor biomass were sampled during the feeding of the wastewater with a chemical oxygen demand (COD) value of 8 g L−1 and a hydraulic retention time (HRT) of 8 days. These samples were characterized by the predominance of cells with long-rod morphology similar to Methanosaeta spp. Following a sharp operational change, accomplished by increasing the COD concentration of the organic influent from 8 to 10 g L−1 and reducing the HRT from 8 to 5 days, there was a doubling of the organic loading rate, a reduction of the COD removal efficiency, as well as decreased methane content in the biogas and an accumulation of total volatile acids in the reactor. Moreover, this operational change resulted in a significant population shift from long-rod Methanosaeta-like cells to tetrad-forming Methanosarcina-like cells. The distributions of microbial populations involved in different zones of the AHR were determined. The results showed that nonmethanogens became the predominant population in both sludge and the packed bed zone. However, the percentage of methanogens in the packed bed zone was higher than that in the sludge bed zone. This higher percentage of methanogens was likely caused by the fact that the packed bed zone provided a suitable environmental condition with an appropriate nutrient availability for methanogen growth.  相似文献

Two different recombinant human proteins were purified directly from Pichia pastoris whole cell fermentation broth, containing 30–44% biomass (wet weight percent), by strong cation exchange expanded bed adsorption chromatography. Expanded bed adsorption chromatography provided clarification, product purification and product concentration in a single unit operation at large scale (2000-l nominal fermentation volume). The efficiency of expanded bed adsorption chromatography resulted in a short process time, high process yield, and limited proteolytic degradation of the target proteins. The separations were operated using a 60-cm (d) column run at 14 l/min. For one protein, expanded bed adsorption chromatography resulted in an average product recovery of 113% (relative to fermentation supernatant) and a purity of 89% (n=10). For the other protein, the average product recovery was 99% (relative to fermentation supernatant) and the purity was 62.1 (n=10). The fluxes of particulate and dissolved material between bivalve beds and the water column in the Oosterschelde estuary have been measured in situ with a Benthic Ecosystem Tunnel. On mussel beds uptake of POC, PON and POP was observed. POC and PON fluxes showed a significant positive correlation, and the average C:N ratio of the fluxes was 9.4. There was a high release of phosphate, nitrate, ammonium and silicate from the mussel bed into the water column. The effluxes of dissolved inorganic nitrogen and phosphate showed a significant correlation, with an average N:P ratio of 16.5. A comparison of the in situ measurements with individual nutrient excretion rates showed that excretion by the mussels contributed 31–85% to the total phosphate flux from the mussel bed. Ammonium excretion by the mussels accounted for 17–94% of the ammonium flux from the mussel bed. The mussels did not excrete silicate or nitrate. Mineralization of biodeposition on the mussel bed was probably the main source of the regenerated nutrients.From the in situ observations net budgets of N, P and Si for the mussel bed were calculated. A comparison between the uptake of particulate organic N and the release of dissolved inorganic N (ammonium + nitrate) showed that little N is retained by the mussel bed, and suggested that denitrification is a minor process in the mussel bed sediment. On average, only 2/3 of the particulate organic P, taken up by the mussel bed, was recycled as phosphate. A net Si uptake was observed during phytoplankton blooms, and a net release dominated during autumn. It is concluded that mussel beds increase the mineralization rate of phytoplankton and affect nutrient ratios in the water column. A comparison of N regeneration by mussels in the central part of the Oosterschelde estuary with model estimates of total N remineralization showed that mussels play a major role in the recycling of nitrogen.  相似文献

The kelp bed of Gouqi Island lies in the east of the East China Sea, in the Shengsi Archipelago. It is a key component of the island-reef ecosystem. This study evaluated kelp beds on intertidal and subtidal zones from 2004 to 2006. We evaluated seasonal variations in the community structure of macroalgae, and their relationship with water temperature, nitrogen and phytoplankton. The Gouqi Island kelp bed consists mainly of species of Sargassum. Sargassum horneri was the dominant species in subtidal zones, representing 90% of the total biomass. The dominant species in intertidal zones were S. fusiforme, S. horneri, S. thunbergii and Undaria pinnatifida. Fifty-four phytoplankton species were found in the Gouqi Island kelp bed, of which 51 species were found inside the kelp beds, 43 species outside the kelp beds, and 40 species were present both inside and outside the kelp beds. Diatoms were dominant both inside and outside the kelp beds. Except in winter, Skeletonema costatum was dominant, and its abundance in autumn was over 98% of the total abundance. Water temperature was found to be the primary factor influencing the growth of S. horneri in the Gouqi Island kelp bed. S. horneri grew slowly from November to March, and rapidly from March to June. The upper temperature limit for growth of S. horneri was about 18°C. Higher water temperatures can result in a decline in the kelp bed. Nitrogen seemed to be a limiting factor for macroalgal growth. This was particularly true for S. horneri and phytoplankton. Since nitrate is the primary nutrient for S. horneri, its absorption by S. horneri resulted in seasonal changes of nitrate in the ecosystem.  相似文献

A continuous fluidized bed reactor operation system has been developed for ethanol production by Zymomonas mobilis using hydrolysed B-starch without sterilization. The operation system consists of two phases. In the first phase macroporous glass carriers in a totally mixed fluidized bed reactor were filled up totally with a monoculture of Z. mobilis by fast computer-controlled colonization, so that in the subsequent production phase no contaminants, especially lactic-acid bacteria, could penetrate into the carrier beads. In the production phase the high concentration of immobilized Z. mobilis cells in the fluidized bed reactor permits unsterile fermentation of hydrolysed B-starch to ethanol at short residence times. This results in wash-out conditions for contaminants from the substrate. Long-term experimental studies (more than 120 days) of unsterile fermentation of hydrolysed B-starch in the laboratory fluidized bed reactor (2.2 l) demonstrated stable operation up to residence times of 5 h. A semi-technical fluidized bed reactor plant (cascade of two fluidized bed reactors, each 55 l) was operated stably at a mean residence time of 4.25 h. Glucose conversion of 99% of the unsterile hydrolysed B-starch was achieved at 120 g glucose/l–1 in the substrate, resulting in an ethanol concentration of 50 g·l–1 and an ethanol space-time yield of 13 g·l–1·h–1. This is a factor of three compared to ethanol fermentation of hydrolysed B-starch with Z. mobilis in a continuous stirred tank reactor, which can only be operated stably under sterile conditions. Two plant fossil‐bearing beds from the middle Barremian of Belgium were analysed to ascertain how experimental designs affect conclusions regarding palaeodiversity at a local scale. We analysed eight lateral samples per bed taken regularly every 3 m using an exhaustive sub‐sampling method. The Clench equation was used to evaluate the completeness of the taxonomic inventory of the samples and the sampling effort needed to obtain a reliable representation of diversity. The number of replicates needed to obtain the same representation of diversity from different nearby lateral samples of the same bed ranged from 5 to 19. Richness (S), Evenness (J) and the number of equiprobable taxa (2H’) greatly varied between samples from the same bed, even over short distances. Only one of the studied samples was representative of the taxonomic inventory of its bed. Our study shows that 1) the selection bias of the sampling area is reduced by increasing the number of lateral samples taken in a bed, enabling more reliable conclusions about local‐scale diversity; 2) intense sub‐sampling methods are needed to account for statistically independent observations of detailed lateral variation; and 3) sampling methods in palaeodiversity analyses must look for a similar degree of representativeness in samples rather than a homogeneous sample size. Using a sampling effort analysis provides evidence for the completeness of the data set, adjusting the amount of work required. Implementing the Clench equation in palaeodiversity analyses improves the performance of data acquisition in palaeoecological studies and provides a quality test of the data sets derived from them.  相似文献

It has recently been reported that the Eisenia bicyclis beds on the coast of Tomarihama in the Oshika Peninsula in Miyagi Prefecture (northern part of Japan) have been progressively receding from the offshore side of the beds every year. In this study, to determine the factors causing the change in the distribution of E. bicyclis beds as well as the extent of the change, an ecological survey of E. bicyclis and grazing animals was conducted. A 150 m-long fixed line was set up in the rocky coastal area on the Oshika Peninsula in Miyagi Prefecture, and the number of E. bicyclis individuals inhabiting a 1-m width was then measured. In addition, the numbers of individuals of sea urchins (Strongylocentrotus nudus) and abalone (Haliotis discus hannai) were measured. Although the E. bicyclis bed extended to the 64-m point on the fixed line at the time of the survey in August 1999, by July 2001, its lower limit was at the 53-m point; thus, a retreat of the outer edge of the bed of 11 m was observed in 2 years. The density of adult E. bicyclis sporophytes in the bed was reduced from 1.08 to 1.80 individuals (ind.) m−2, respectively. Juvenile sporophytes were also observed, but most of those that had settled near the lower limit of the bed failed to survive to become adult sporophytes. There were few sea urchins, the main grazing animal on the site, inside the E. bicyclis bed (3.5 ind. m−2 in October 2001) while there were many near the bed lower limit and on the deep-site side (12.9 ind. m−2 in October 2001). These results indicate that the high grazing pressure of sea urchins reduced the survival of juvenile sporophytes near the bed lower limit, thus impeding the recruitment of juvenile sporophytes to replace adult sporophytes that died due to senescence, storm action and/or grazing.  相似文献

海水养殖尾水中总氮、总磷超标是引起沿海水体富营养化的主要原因,为研究碱蓬浮床对海水养殖尾水中氮磷的去除效果,该研究设计加入碱蓬(Suaeda salsa)浮床和不加浮床的两组对比实验,通过比较修复前后碱蓬株高、生物量、含水率、根长以及各部位氮、磷的含量变化,以及水体中总氮(TN)和总磷(TP)的去除效果,探究浮床中碱蓬对总氮和总磷的吸收及其生长特性,验证碱蓬浮床对海水养殖废水中氮、磷等的去除能力。结果表明:浮床中碱蓬株高、鲜重、干重、含水率、根长较修复前均有显著增加,说明浮床中盐生植物碱蓬能够适应含海水养殖尾水水培环境;经碱蓬浮床修复,水体中总氮、总磷均明显下降,其中碱蓬对海水养殖尾水中的总氮总磷去除贡献率分别为16.10%和78.15%,浮床中碱蓬会在叶片和根系中积累氮磷。  相似文献

Microcystis aeruginosa immobilized in a natural polymer was tested for its potential to remove Cu2+ ions from aqueous solution in a continuous, downflow packed columnar reactor. Various parameters like flow rate, bed height and contact time required for maximum removal of test metals by the immobilized Microcystis aeruginosa were optimized. An increase in bed height from 2 to 10 cm resulted in an apparent decrease in biosorption capacity from 8.94 to 5.34 mg g–1, but more Cu2+ solution was purified at the higher bed height. Efficiency of metal recovery from Cu2+-loaded biomass and its subsequent regeneration was also determined. Immobilized M. aeruginosa was found to be effective in Cu2+ removal from solution for up to 10 cycles of adsorption–desorption and 1 M HCl is very efficient desorbent for regeneration of Microcystis biomass for reuse.  相似文献

Summary Citric acid production by immobilized of Aspergillus niger in a fluidized bed reactor was performed, evaluating the productivity and the stability of the process when pulsing device was used. The application of a pulsing flow to fluidized bed reactor and the feed nitrogen limited allow to control of bioparticles morphology avoiding bed compactation. When operated at optimum pulsation frequency (0.3 s–1) the stability of the bioreactor was maintained for more than 30 days, increasing the citric acid production in more than 52.2%.  相似文献

The seasonality of standing crop of a Sargassum bed was investigated by conducting monthly sampling from February 1988 to July 1989. Environmental parameters of water movement, salinity, number of daytime minus tides, and water temperature were also measured. An intra-annual pattern of variation in standing crop of Sargassum crassifolium, S. cristaefolium, S. oligocystum, and S. polycystum was observed. Standing crop was generally lowest in February, March, April, or May, and highest in November through January. Sargassum accounted for about 35 to 85% of the monthly algal standing crop of the bed, and the observed variation in overall standing crop of the bed generally reflected the standing crop of Sargassum. The seasonality of the standing crops of the associated algal divisions also followed an annual cycle, but their maximum and minimum standing crops did not coincide with those of Sargassum. Individually, as well as collectively, the standing crops of the Sargassum spp. were poorly correlated with the environmental factors observed.  相似文献

A capture step was developed using the expanded bed adsorption technology to separate a protein of interest on a cation exchanger from a crude Escherichia coli homogenate. This method was developed in bench-top scale using a STREAMLINE 25 column (Amersham Pharmacia Biotech, Sweden) and STREAMLINE SP. The development was based on earlier experiments performed in a packed bed column (SP-Sepharose FF) to investigate the conditions for sample application, wash and elution. The packed bed method was transformed into an expanded bed method by slightly modifying the wash procedure and cleaning in place (CIP). This method was then scaled-up to pilot scale and used for production of the fusion protein according to cGMP.The yield over the step in pilot scale was 70-85% compared with only 30-50% in small scale. Pressure build-up, attachment of biomass to the adsorbent and collapses of the expanded bed were phenomena seen in small scale but not in pilot scale. The scale-up of the step significantly improved the performance of the step.  相似文献
