Kimura mistook ambiguous maximum parsimony codons for wrong codons. The maximum parsimony method performed well as judged by the two classes of serine codons (which can not be connected by silent mutations) on comparing the parsimony codons for serines in human, rabbit, and mouse hemoglobin chains to actual codons determined by nucleotide sequencing. In genealogical reconstructions involving 247 eucaryotic globins, the maximum parsimony distances separating the contemporary sequences show that Kimura's Poisson and Dayhoff's PAM estimates of rate of globin evolution miss most of the superimposed replacements and are therefore seriously in error. Nor is Kimura's constant rate assumption and his belief in a single origin of myoglobin supported. Lamprey myoglobin appears to be most like lamprey hemoglobin, while gnathostome myoglobin seems closest to gnathostome hemoglobin. It was found that the three types of gnathostome globins (Mb, Hb, Hb) evolved between the shark-boney vertebrate and bird-mammal ancestors at a much faster rate than from the latter ancestor to the present. The data indicate that rates were exceedingly fast during the origin of these globin chains because a high proportion of substitutions were adaptive. It was concluded that wherever strong stabilizing selection acts on a protein, somewhere in the past positive Darwinian selection must have spread the amino acid substitutions now being preserved.  相似文献

Summary We have developed a culture system for longterm growth of human lymphokine-activated killer (LAK) cells exhibiting an elevated, wide-spectrum antitumor cytotoxicity. The system allows the exponential growth of monocyte-depleted low-density lymphocytes in the presence of human serum and recombinant human interleukin-2 (103 U/ml), alone or in combination with interleukin-1 or (both at 10 U/ml). Eighteen cultures were established from 18 normal adult donors. The membrane phenotypes of the final LAK cell population, assessed by a panel of monoclonal antibodies (mAb), consist of three main types: (a) NKH-1+, Ti/, Ti/, and CD3 lymphocytes; (b) NKH-1+, Ti/, Ti/+, and CD3+ lymphocytes and (c) NKH-1+, Ti/+, Ti/ and CD3+ lymphocytes. Northern blot analysis showed that all these cell populations express relatively high levels of perforin RNA, particularly cells exhibiting the first phenotype. This culture system may provide a tool for cellular and molecular studies on the mechanisms of antitumor cytotoxicity, as well as the basis for new adoptive immunotherapy protocols in advanced cancer.  相似文献

Methods of intrinsic viscosity () and beam flow birefringence were used to study the effects of some single-charged ions (F, Cl, Br, I, NO 2, NO 3, ClO 4, SCN, CH3COO) on the size and thermodynamic rigidity of a DNA molecule in aqueous solutions of sodium salts in a broad interval of ionic strength when temperature T is changed. It has been shown that the close interactions in a macromolecule and the resulting DNA persistent length a are independent of the type of the salt anion over the whole interval of . On the contrary, the specific volume of the DNA molecule in solution, proportional to the value, is quite sensitive to the anionic composition of the solvent, which is due to the effect of anions and their hydration on the long-range interactions in the macromolecule. The presence of polyatomic and halide anions is manifested differently in the value of DNA. Possible factors responsible for the observed effect and the role of structural alterations of water upon anion hydration are discussed.  相似文献

Effects of light and temperature, on the growth of three freshwater green algae isolated from an eutrophic lake and identified as Selenastrum minutum, Coelastrum microporum f. astroidea and Cosmarium subprotumidumwere studied in batch cultures under non-nutrient limited conditions. Experiments were performed to determine the growth rate over a wide range of light intensities (30–456 mol m–2 s–1) and temperature (15–35°C), using a 15/9 (light/dark) photoperiod cycle. The maximum growth rates and the optimum light intensities at a temperature of 35°C were 1.73 d–1 and 420 mol m–2 s–1for Selenastrum minutum, 1.64 d–1 and 400 mol m–2 s–1 for Coelastrum microporum and 1.00 d–1 and 400 mol m–2 s1 for Cosmarium subprotumidum. The results were fitted with the mathematical models of Steele (1965), Platt & Jassby (1976) and Peeters & Eilers (1978). Steele's function and equation of Platt & Jassby don't describe correctly the relationship between the growth and light intensity. In the opposite, the equation of Peeters & Eilers provides the best fit for the three species.  相似文献

Atriplex gmelini plants were regenerated via organogensis from hypocotyl explants. Callus lines were induced from the hypocotyl explants on Linsmaier and Skoog (LS) medium supplemented with 1 M benzyladenine and 5 M -naphthaleneacetic acid in the dark. Shoots were regenerated from the callus lines on LS medium supplemented with 20 M thidiazuron and 0.1 M -naphthaleneacetic acid under a high-intensity light condition (450 mol m–2 s–1). The regenerated shoots were rooted on LS medium without growth regulators to obtain fully developed plants. We succeeded in transforming Atriplex gmelini from callus lines using Agrobacterium tumefaciens.  相似文献

A key to the management of hillstreams in relation to Fish Species Richness (FSR) and Shannon and Weaver Diversity Index (H) (Shannon and Weaver, 1949. The Mathematical Theory of Communications. Urbana, University of Illinois Press, IL, USA) using a multitude of factors such as altitude, water source, bed gradient, dominant channel materials and habitat type has been proposed on the basis of 10 study sites, belonging to three north Indian rivers namely; Ghaggar, Yamuna and Ganga. The selected streams are located between the altitudes ranging from 380 to 1524m. It has been observed that high altitude streams having steep gradient, boulder dominated substrate with rapids as the main habitat type had lower FSR (3–4) and H (0.55–0.99) as compared to hillstreams located in the lower altitudes having gentle gradient, cobble dominated substrate with pools, riffles, runs, rapids and cascades as the main habitats and have high FSR (6–14) and H (1.67–2.35). Types – A, B and F hillstreams were encountered when the hillstreams are classified after Rosgen (Applied River Morphology, Wildland Hydrology, Co, USA).  相似文献

Nitrate reduction in a non-polluted, coastal marine sediment was measured with an open flow-through system. The recorded rates depended upon nitrate concentration but were largely independent of the weight of sediment (14–35 g) and the dilution rate (0.7–5 h–1). Rate of nitrate uptake followed classical Michaelis-Menten kinetics, and km and Vmax values were equal to 78M and 0.168m mol g–1 hour–1, respectively. These values are in good agreement with those found by the other authors for the same biotope but by different methods. This technique of the open flow-through system is fast, simple, and inexpensive and involves small quantities of sediment (10 g).  相似文献

The direct charge recombination rates from the primary quinone, k AD (D+Q A DQA) and the secondary quinone, k BD (D+Q B DQB), in reaction centers from Rhodobacter sphaeroides were measured as a function of the free energy differences for the processes, G AD 0 and G BD 0 , respectively. Measurements were performed at 21 °C on a series of mutant reaction centers that have a wide range of dimer midpoint potentials and consequently a large variation in G AD 0 and G BD 0 . As –G AD 0 varied from 0.43 to 0.78 eV, k AD varied from 4.6 to 28.6 s–1. The corresponding values for the wild type are 0.52 eV and 8.9 s–1. Observation of the direct charge recombination rate k BD was achieved by substitution of the primary quinone with naphthoquinones in samples in which ubiquinone was present at the secondary quinone site, resulting specifically in an increase in the free energy of the D+Q A state relative to the D+QAQ B state. As –G BD 0 varied from 0.37 to 0.67 eV, k BD varied from 0.03 to 1.4 s–1. The corresponding values for the wild type are 0.46 eV and 0.2 s–1. A fit of the two sets of data to the Marcus theory for electron transfer yielded significantly different reorganization energies of 0.82 and 1.3 eV for k AD and k BD, respectively. In contrast, the fitted values for the coupling matrix element, or equivalently the maximum possible rate, were comparable (25 s–1) for the two charge recombination processes. These results are in accord with QB having more interactions with dipoles, from both the surrounding protein and bound water molecules, than QA and with the primary determinant of the maximal rate being the quinone-donor distance.  相似文献

Carbon and nitrogen stable isotopic data from the primary producers in mangrove ecosystems are needed to investigate trophic links and biogeochemical cycling. Compared with other mangrove species (e.g. Rhizophora mangle) very few measurements have been conducted on the white mangrove, Laguncularia racemosa. The carbon and nitrogen stable isotopic and elemental compositions of L. racemosa were analyzed and compared from Florida and Belize. 13C values of L. racemosa from Florida (mean = –26.4) were slightly higher than those from Twin Cays, Belize (mean = -27.4), which may be due to higher salinity in some parts of the Florida site. There was no difference between the 15N values from L. racemosa from these two sites (Florida mean = 0.6; Belize mean = 0.3), which are indicative of nitrogen derived from nitrogen fixation in a planktonic marine system. However, higher 15N values from L. racemosa at Man of War Cay in Belize (11.4 and 12.3), which is fertilized by roosting marine birds (14.0), illustrate that L. racemosa can sensitively reflect alternative nitrogen sources. Although the isotopic data could not distinguish between Avicennia germinans, R. mangle and L. racemosa in Belize the L. racemosa had considerably higher C/N ratios (46.5 – 116.1) compared with the Florida samples (42.2 – 76.0) or the other mangrove species. Unlike some previous findings from R. mangle, substrate characteristics (e.g. salinity, NH4 +, and H2S) were not related to the isotopic or elemental composition of L. racemosa. 13C, 15N and C/N were analyzed for ecosystem components from L. racemosa habitats at Twin Cays, including other plants (e.g. R. mangle, A. germinans and seagrass), detritus, microbial mats and sediments. Results from mass-balance calculations show that mangrove detritus composes very little of the sediment, which is principally composed of microbial biomass (80 – 90%). Detritus at some sites is also influenced by sources other than that from L. racemosa, including seagrass leaves.  相似文献

The amino acid sequences of the -lactalbumins of the echidna, Tachyglossus aculeatus, and the platypus, Ornithorhynchus anatinus, were compared with each other and with those of 13 eutherian and 3 marsupial species. Phylogenetic parsimony analyses, in which selected mammalian lysozymes were used as outgroups, yielded trees whose consensus indicated that the two monotremes are sister taxa to marsupials and eutherians and that the latter two clades are each other's closest relatives. The data do not support the notion of a Marsupionta (monotreme–marsupial) clade. Pairwise comparison between the -lactalbumins yielded maximum-likelihood distances from which divergence dates were estimated on the basis of three calibration points. The distance data support the view that the echidna and platypus lineages diverged from their last common ancestor at least 50 to 57 Ma (million years ago) and that monotremes diverged from marsupials and eutherian mammals about 163 to 186 Ma.  相似文献

-Glucans were prepared from Agaricus blazei Murill by repeated extraction with hot water. The average molecular weights of -glucans were 30–50 kDa by gel filtration chromatography. Oligosaccharides (AO), derived from hydrolyzing -glucans with an endo--(16)-glucanase from Bacillus megaterium, were mainly di- and tri-saccharides. Though -glucans and AO both showed anti-hyperglycemic, anti-hypertriglyceridemic, anti-hypercholesterolemic, and anti-arteriosclerotic activity indicating overall anti-diabetic activity in diabetic rats, AO had about twice the activity of -glucans with respect to anti-diabetic activity.  相似文献

Summary We examined the transition from ectothermy to endothermy in nestling bank swallows (Riparia riparia) by measuring the peak metabolic response to cold (PMR) in groups of nestlings. Additionally aerobic capacity, as assessed by citrate synthase activity (CS), and contractile function, as assessed by myofibrillar ATPase activity (mATPase) were measured in the pectoralis and mixed leg muscles during development. During the first 65% of their growth (from 2–12 g) bank swallows do not increase their metabolic rate in response to cold (Fig. 1). Between 12 and 16 g the PMR increased from 4 to more than 10 ml O2 (g·h)–1. Citrate synthase activity increased throughout development, starting at 20 moles (min·g fresh mass)–1 in both tissues and increasing to 150 and 50 moles (min·g)–1 in the pectoralis and leg muscles, respectively (Fig. 5). The augmented aerobic capacity combined with large increases in muscle mass undoubtedly contributes to the improved thermoregulatory abilities of older nestlings. However, muscle mass and aerobic capacity increase continuously and do not show the sharp transition noted in PMR. In the leg muscle mATPase activity is constant throughout growth, but in the pectoralis muscle it undergoes an abrupt increase from 0.5 moles (min·mg myofibrillar protein)–1 in animals weighing less than 12 g to 0.9 moles (min·mg)–1 in nestlings weighing more than 15 g (Fig. 6). Summary Biotransformation of benzaldehyde to benzyl alcohol bySaccharomyces cerevisiae immobilized in different support matrices was investigated. Polymers with intrinsic hydrophobic and/or hydrophilic nature as well as mixed hydrophobic and hydrophilic supports were examined both in aqueous and bisphasic aqueous-organic systems. The hydrophobic support material ENTP-2000 or mixed silicone:alginate (50-2550-75) proved to be most suitable not only for nonconventional media but also for conventional aqueous media for production of benzyl alcohol. With ENTP-2000, catalytic activity and maximum yield were 159 mol h–1 g–1 dry weight catalyst and 0.89 mM, respectively, in hexane containing 2% moisture. Corresponding values in aqueous media were 246 mol h–1 g–1 dry weight catalyst and 1.53 mM. With 5050 silicone:alginate, catalytic activity and maximum yield were 177 mol h–1 g–1 dry weight catalyst and 1.18 mM, respectively, in hexane containing 2% moisture. Corresponding values in aqueous media were 192 mol h–1 g–1 dry weight catalyst and 0.8 mM.  相似文献

Overall, 30 strains of hyperthermophilic archaea, representing seven species of the genera Thermococcus, Desulfurococcus, Thermoproteus, and Acidilobus, were tested for the presence of thermostable DNA polymerases. Thermostabilities of the polymerases varied distinctly among the strains within one species. Polymerases of five strains retained 60–100% activity upon incubation of the preparations at 95°C for 120 min. A new DNA polymerase was isolated from the strain Thermococcus litoralis Sh1AM, possessing the enzyme with the most promising properties, and characterized. Molecular weight of the enzyme is 90–100% kDa. The purified DNA polymerase preserved 50% of the initial activity upon incubation at 95°C for 120 min. The polymerase isolated displayed an associated 3–5 exonuclease activity. The error rate when extending a DNA strand was at least twofold lower compared with Taq polymerase. Daily and weekly fluctuations of PAR, UV-A, and UV-B have been continuously monitored for 5 months in Ancient Korinthos, Greece (37°58 N, 23°0 E) using a calibrated instrument based on 3 sharp band sensors. Daily dose ranged between 521–12 006 kJ m-2 for PAR; 52–1, 239 kJ m-2 for UV-A; and 0.66–22.5 kJ m2 for UV-B. Weekly dose ranged between 16 778-81 788 kJ m-2 for PAR; 1 406–8 517 kJ m-2 for UV-A; and 18–151 kJ m-2 for UV-B. UV-B/PAR and UV-A/PAR ratio distribution, however, does not follow closely PAR fluctuations. Generally, the UV-B/PAR and UV-A/PAR ratios were high in bright light conditions (2.1×10-3, 118×10-3) and low in darker weeks (0.9×10-3, 63×10-3. The UV-B/UV-A ratio exhibits smaller fluctuations with season (20x1×10-3, 12×10-3). Attention is drawn to the effects of sudden changes in ambient radiation and to the ratios of UV-B, UV-A, and PAR.  相似文献

Cultures of Haematococcus pluvialis were exposed to mutagens like u.v. and EMS (ethyl methanesulphonate). The results showed that the survival rate decreased with the increase in u.v. exposure time and increase in EMS concentration. These mutants were further screened using inhibitors of the carotenoid biosynthetic pathway viz. diphenylamine (15–90 M), nicotine (160–320 M) and compactin (1.5–3.0 M). The mutants thus obtained showed early enhanced (2.2–3.2-fold) astaxanthin accumulation and also exhibited higher lycopene cyclase activity.  相似文献

Summary The abundance and depth distribution of zooplankton faeces in spring to early summer were investigated along meridional transects (47°W and 49°W) that extended from the Scotia Sea (57°S) across the Weddell-Scotia Confluence and into the Weddell Gyre (62°S). The sea ice edge retreated from 59°30S to 61°S during the study. Faeces were sampled with nets, Niskin bottles and sediment traps and subsequently analysed by light and electron (SEM) microscopy. Krill faecal strings and oval faecal pellets of unknown origin were by far the most important zooplankton faeces and highest concentrations were always found in the Confluence often close to the ice border. Krill faeces were usually more abundant in the uppermost layer (0–50m) where they contributed an average of 130 g dry weight m–3. There was an exponential decrease with depth, with a minimum of 0.6 g dry weight m–3 in the 500–1000 m stratum. Oval pellets were more evenly distributed in the upper 1000 m of the water column, with an average of 9 g dry weight m –3, although there was a small peak (20 g dry weight m–3) in the subsurface layer (50–150 m depth). Consecutive collections (day-night) of krill faeces using drifting sediment traps showed that only the larger strings sank from 50 to 150 m depth. Peritrophic membranes appeared to deteriorate during sinking. Diatoms (in particular Nitzschia and Thalassiosira spp.) contributed by far the bulk of material in krill and oval faeces. In samples collected near or under the pack ice, remains of crustaceans in both krill- and oval faeces were also found.Data presented here were collected during the European Polarstern Study (EPOS) sponsored by the European Science Foundation  相似文献

Wheat for human consumption (140 samples) was collected after harvest from all regions of Bulgaria. The 1995 crop year was characterized by heavy rainfall in the spring and summer months. The internal mycoflora of wheat samples was dominated by Fusarium spp. and Alternaria spp., and storage fungi were rarely present. The samples were analysed for contamination with Fusarium mycotoxins deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-AcDON), 15-acetyldeoxynivalenol (15-AcDON), T-2 Toxin (T-2), diacetoxyscirpenol (DAS), and zearalenone (ZEA), using enzyme immunoassay methods. DON and ZEA were the predominant toxins, with a contamination frequency of 67% and 69%, respectively. The average levels of these toxins in positive samples were 180 g/kg (DON) and 17 g/kg (ZEA), maximum concentrations were 1800 g kg–1 and 120 g kg–1, respectively. Acetyl derivatives of DON, namely 3-AcDON and 15-AcDON, were found in 2.1 % and 0.7% of the samples, at at maximum level of about 100 g kg–1. Only one sample was positive for T-2 (55 g/kg), DAS was not detected. We investigated the carbon supply status in species-rich mediterranean plant communities growing in a bowl-shaped 1-ha CO2 spring area near Sienna, Italy. A geothermic lime-kiln has provided these communities, for as long as historical records are available, with pure CO2 that mixes with ambient air at canopy level to daytime means of 500–1000 ppm CO2. Immediately outside the spring area similar plant communities are growing on similar substrate, and in the same climate, but under ca. 355 ppm CO2. We found no evidence that plants in the CO2 spring area grow faster, flower earlier or become larger. However, we found very large differences in tissue quality among the 40 species studied inside and outside the spring area. Depending on weather conditions, the mean concentration of total non-structural carbohydrates (TNC, sugars and starch) in leaves of herbaceous plants was 38–47% higher in the spring area. Fast growing ruderals growing on garden soil inside and outside the spring area show the same response. Among trees, leaves of the deciduousQuercus pubscens contain twice as much TNC inside as outside the vent area, whereas evergreenQ. ilex leaves show no significant difference. TNC levels in branch wood paralleled leaf values. TNC in shade leaves was also higher. Elevated CO2 had no effect on the sugar fraction, therefore differences in TNC are due to starch accumulation. Leaf nitrogen concentration decreases under elevated CO2. These observations suggest that the commonly reported TNC accumulation and N depletion in leaves growing under elevated CO2 are not restricted to the artificial conditions of short-term CO2 enrichment experiments but persist over very long periods. Such an alteration of tissue composition can be expected to occur in other plant communities also if atmospheric CO2 levels continue to rise. Effects on food webs and nutrient cycling are likely.  相似文献

A 20–40 m pellicular high density (3.7 g cm–3) expanded bed material has been designed for the capture of DNA and other large macromolecules. Anion exchangers fashioned out of these supports exhibited dramatically enhanced DNA binding capacities over commercial anion exchange adsorbents (6 mg ml–1, c.f. 50 g ml–1 at 10% breakthrough), due to a combination of small particle and fuzzy surface architecture created through the coupling of polyethylene imine chains.  相似文献
