This study describes an efficient and reproducible protocol for in vitro date palm propagation using mature female flowers. It focuses on the promising proliferation capacity exhibited by a number of female flower tissues taken at the final developmental stage. This capacity resided in the ability to preserve minuscule zones in a juvenile state located at the floral organ armpits (sepals and petals). The originality of this method lies in the possibility of propagation of very rare varieties, particularly the genotypes that exist in only one copy without the excision of the plant mother, the source of the tissue collected to be cultivated, which was not the case for all previous methods. The findings revealed that 2,4-D at 1mg/l, most of the varieties tested showed reactivity. The success of this technique was also noted to depend on the concurrent control of various factors pertaining mainly to the hormonal composition of the culture medium and the appropriate time of tissue transfer, which depends on the proliferation state as well as the culture period. This study describes the nature of the proliferation from the mature female flowers and their outcome, particularly those at the origin of embryogenic and budding strains and discusses the advantages of this novel multiplication method as compared to the currently available ones. 相似文献

This study explores and reports on the gain brought to the morphogenetic aptitude of female date palm inflorescences through in vitro hermaphrodism induction. It investigates the main factors involved in the process of sex modification through hormonal induction, such as the floral developmental stage and hormone combination and concentration. It demonstrates that the vestigial stamens (staminodes) of female date palm flowers display a new and high capacity to proliferate under particular in vitro conditions, without blocking carpel's development, leading to morphologically typical hermaphrodite flowers. This de novo activation of repressed stamens was found to occur rapidly. The isolated pollen mother cells appear in the obtained anther's locules and undergo an ordinary microsporogenesis process. The data show that hermaphrodism induction depended heavily on both the growth regulators applied and the flower's developmental stage. They also confirm the early theory that suggests that dioecious plants come from a hermaphrodite ancestor. Such hermaphrodism control can provide new prospects and opportunities for the investigation of the in vitro self-fertilization process. It can also be useful in improving the understanding of the genetic mechanism involved in sex organ development in date palm. 相似文献

A rapid and reliable micropropagation method was established for Spathoglottis plicata. Nodal and leaf explants dissected from 8-month-old pot-grown seedlings were cultured on charcoal-amended Murashige and Skoog
medium supplemented with 16 combinations of α-naphthaleneacetic acid (NAA) and 6-benzylaminopurine (BA) at concentrations
of 0.54–10.74 μm. Regeneration of protocorm-like bodies (PLBs) and subsequent plantlet development were observed from 98.5% of the nodal explants.
Only 6.5% of leaf explants and occasionally some root segments (dissected from regenerated plantlets) were able to produce
PLBs and then plantlets. The optimum plant growth regulator (PGR) combination for maximal PLB regeneration was 5.37 μm NAA and 0.44 μm BA. The best combination of PGR for plantlet development was 2.69–10.74 μm NAA and 8.88 μm BA. The NAA to BA ratios for maximal PLB induction and plantlet development were 12.2 and 0.3–1.2, respectively. Regenerated
PLBs and plantlets, when cut into pieces of less than 1 mm and subcultured onto the above media, regenerated new PLBs and
plantlets in another 3 months.

Seed explants of A. stenosperma were cultured on MS medium supplemented with 6-benzylaminopurine with the aim of rescuing nonviable accessions stored in seed bank conditions. The regeneration potential of leaf explants from in vitro plants derived from embryonic axes was studied by using whole leaflets and leaflet segments. Explants were cultured on Murashige and Skoog (MS) medium supplemented with different concentrations of 6-benzylaminopurine and naphthalene acetic acid. Indirect organogenesis was observed in response to 6-benzylaminopurine, either alone or in association with naphthalene acetic acid, in both explant types. Media supplemented with naphthalene acetic acid as the sole growth regulator induced rhizogenesis in whole leaflets and leaflet segments, with subsequent shoot production directly from the roots. 相似文献

A micropropagation scheme forAnanas comosus Merr. was developed using nodule culture. Nodules were induced from leaf-base or chopped shoot-base explants on modified half-strength MS medium supplemented with 2.69-5.37 M NAA and 4.44 M BA and could be maintained long-term as nodules. The nodules proliferated into more nodules when chopped into pieces of 1–3 mm and placed onto the same medium. They regenerated shoots when transferred to medium supplemented with 0.54–10.74 M NAA and 0.44–8.88 M BA. The regeneration capacity of nodules is higher than that of direct regeneration or callus. Maximum regeneration was obtained from culture medium containing 0.54 M NAA and 0.44 M BA, where shoots could be observed as early as within 2 weeks. Many shoots formed roots in the same medium in which they were regenerated after 10 subcultures, but the best rooting occurred in medium containing 0.54 M NAA and 0.44 M BA. Rooted plantlets ofA. comosus Merr. could be routinely produced at 6-week intervals.Abbreviations

Red palm weevil (Rhynchophorus ferrugineus) is a voracious pest of date palm worldwide. Pakistan ranks sixth in date palm production globally. Losses to date palm plantations in Pakistan sometimes surpass 10%-20%. Most of the traditional management strategies used by farmers have been found insignificant to combat this voracious pest. The entomopathogenic fungi, Beauveria bassiana [QA-3(L) and QA-3(H)] and insecticides, Nitenpyram (Active 10% SL) [NIT (L) and NIT (H)] were applied to larval (2nd, 4th, and 6th), pupal and adult stages of R. ferrugienus. Integration or alone application of fungi with insecticides at different concentration under laboratory conditions. Combined application was depicted additive and synergistic interactions. Contrarily, highest cumulative mortality (100%) was recorded in 2nd instar larvae as compared to later instar larvae at combined application. The maximum pupal and adult mortality remained 89% and 66% respectively after treatment with [QA-3 (H) + NIT (L)]. The combination of B. bassiana at higher concentration whereas Nitenpyram at lower dose was found more lethal to larvae, pupae and adults of R. ferrugineus. This signifies the need of combining B. bassiana and bio-rational insecticides that can reduce the cost of management with least harm to environment and natural enemies. 相似文献

In vitro culture of Venus fly-trap (Dionaea muscipula) was initiated using flower stalk explants. Activated charcoal was required for bud initiation, but omitted in the subculture of regenerated plantlets. Regenerated plants were subsequently used as explant source for investigations concerning effects of source of tissue, etiolation, orientation and illumination of leaf explants on plant regeneration. Etiolation of source plantlets increased the rate of regeneration from explants and decreased explant failure. Generally, adventitious buds developed at the adaxial side and proximal end of an explant. However, when explants were incubated in the dark, 20–30% of bud initiation occurred at the distal end. The site of shoot regeneration on a leaf explant was affected by both illumination and orientation of explants. Placing an explant adaxial side up resulted in the highest rate of regeneration. The most effective condition for plantlet regeneration was found with etiolated petioles incubated with the adaxial side facing the light. 棉花体细胞培养再生植株存在大量的生理变异和可遗传变异,从中分离到一个性状稳定的花器变异体(CHV1)。从花器官形态特征和表面显微特征分析,该变异体的所有花器官都变成了苞叶状器官,但中央数片叶状器官的基部有胎座和胚珠着生。变异体每朵花有苞叶3-7片,苞叶状器官19-41片。苞叶状器官在花梗上的排布介于“轮”与“螺旋”状之间。据花器发育理论和变异体花的生长特性推测,该变异体中控制花器发育的A、B和C功能皆失活。对造成该变异的可能机理和棉花花发育模式进行了分析。该变异材料对研究棉花花发育和体细胞无性系变异的机理有一定价值。 相似文献

An efficient protocol for in-vitro propagation of an important ornamental crop, Petunia hybrida Vilm. Cv. “Bravo” was developed. The explants that were used to carry out the experiment were Leaf segments, nodal segments and shoot tips. Nodal segments recorded highest per cent asepsis followed by shoot tips and leaf segments. Asepsis was found to be highest when the explants were sterilized with Fungicide (Carbendazim) 0.02% for the duration of 30 min followed by 0.1% HgCl2 for duration of 10 min and then ethanol 70% for 10 s. Longer duration of the sterilant treatment showed more necrotic effects on the explants, thus mercuric chloride treatment when given for 5 min proved to be more effective in terms of survival of the explants. Maximum establishment per cent was recorded in Murashige and Skoog (MS) media fortified with BAP (1.5 mg L−1) and IBA (0.5 mg L−1) in shoot tips and nodal segments, i.e. 97.90 and 95.74% respectively. Callus was efficiently induced and developed when PGR amalgamation of BAP (0.1 mg L−1) and 2,4-D (1.5mg L−1) was used. Kinetin at the concentration of 2.0 mg L−1 along with IBA at 0.5mg L−1 recorded highest callus regeneration in both leaf and internodal segment derived callus. Maximum proliferation percent of shoots (97.90%), highest number of shoots (20.50 explant−1) and maximum length of shoot (2.70 cm) was recorded in PGR combination of IBA and BAP both at 0.5 mg L−1 concentration level. Rhizogenesis was recorded to be highest in the MS media containing IBA 1.00 mg L−1. Best hardening media which recorded maximum survival per cent 92.50% was noticed on the media formulation comprised of equal ratio of perlite and vermiculite mix, under poly house conditions. 相似文献

Soluble proteins of the floral organs of some solanaceous species were analysed by SDS-polyacrylamide gel electrophoresis. In general, the protein content of anther and ovary was higher than that of the style which was greater than that of sepal and petal. The polypeptide patterns showed that in different species, each organ-type had many common bands, although they were of varying intensity. Also, proteins unique to an organ-type were not always detected in the various species. Some ‘species-specific’ polypeptides were, nevertheless, identified in one or more floral organs of a species. 相似文献

DNA methylation plays an important role in the regulation of gene expression in eukaryotes. In this study, the extent and patterns of DNA methylation were assessed in date palm mother-plants and their off-shoots using the methylation-sensitive amplified polymorphism (MSAP) technique. Three types of bands were generated using 12 pairs of primers. Type I were present in both ECOR I + HPA II and ECOR I + MSP I lanes; type II were present in ECOR I + HPA II lanes, but not in ECOR I + MSP I lanes; and type III bands were present in ECOR I + MSP I lanes, but not in ECOR I + HPA II lanes. The total numbers of these three types of bands were 782, 55, and 34, respectively. Among these three types of bands, the polymorphic bands were, respectively, 37, 10, and 0. The distribution of polymorphic bands among mother-plants and off-shoots suggests the methylation variation was present in both the mother-plants and off-shoots. Forty- four out of these 47 polymorphic bands show clear difference between mother-plant and off-shoots: 38 were present only in off-shoots and 6 in both mother-plants and off-shoots. Compared to methylation status in mother-plants, the methylation variation during off-shoot growth of date palm can be characterized as a process involving primarily de-methylation. Hypomethylation of DNA in off-shoots, compared with mother-plants, reflects the marked expression of this molecular feature, which may be related to gene expression during off-shoot development. The methylation or de-methylation status of specific loci in the mother-plants and their off-shoots were probably random events. 相似文献

Oryzaephilus surinamensis, a common grain product pest around the world, particularly causing great damage and losses to the date crops available in the storage. It has been traditionally resisted by using organic pesticides despite suspicions of their harmful effects on dates. This study aimed at the evaluation of the usage of ozone gas as an effective pesticide and also focuses on the safety alternatives provided by the respective method as compared to the usage of phosphine as a pesticide. While studying the ozone effects, the O. surinamensis was exposed to the gas for a variety of time scales (2, 4 and 6 h), distinctly for every life stages of the insect such as egg, larva and adults, infested the Phoenix dactylifera L date palm species present in the Siwa oasis. The obtained results showed that the adult insects had high resistance to ozone gas, while the larvae and egg stages were less resistant. The reduction rate of vital insect was 100% in all stages. Start codon analysis of DNA also showed that there are some SCoT primers can be identified and differentiated between the different treatments and control. On the other hand, the percentage of polymorphisms in insects was 50% while in date kernels 25% by six SCoT primers used. This study highlighted higher efficacy and safety regarding the usage of ozone gas in effectively controlling the manifestation of O. Surinamensis and thereby reducing the loss of stored date crops as compared to the phosphine. 相似文献

In this study, using the quadratic saturation 310 D-optimal design method, we examined the effect of kinetin (KT), gibberellic
acid (GA), and naphthalene acetic acid (NAA) on microrhizome production in ginger. The effect of GA on rhizome induction was
larger than that of KT or NAA. Using simulation and optimality selection for tissue culture, we found that concentrations
of GA, KT, and NAA of 1.33–2.35, 0.49–0.66, and 0.62 g/l, respectively, gave a microrhizome weight of over 0.25 g. The optimal
conditions for microrhizome production were 80 g/l sucrose, 2 × MS macro-elements, and 1 × MS micro-elements, with a photoperiod
of 24L:0D (light/dark). At the same time, 100% survival could be achieved on transfer of the in vitro ginger plantlets with

Protein content and amino acids composition of bee-pollens from major pollen floral sources in Al-Ahsa, Saudi Arabia were determined to investigate the nutritive value of pollen protein relative to requirements of honeybees and adult humans. The major pollen sources were alfalfa, date palm, rape, summer squash, and sunflower. Bee-pollens from alfalfa and date palm showed high content of crude protein and amino acid concentrations. Bee-pollen from sunflower had low content of those components. Eighteen amino acids were found in bee-pollens from the five major floral sources. The highest concentrations of individual amino acids valine, leucine, isoleucine, phenylalanine and proline were obtained from alfalfa bee-pollen; lysine, arginine, cysteine, tryptophan and tyrosine from date palm; methionine, histidine, glycine and alanine from summer squash; threonine, serine and glutamic acid from sunflower; and aspartic acid from rape bee-pollen. The amino acid composition obtained from sunflower bee-pollen showed the lowest concentrations of the essential amino acids: isoleucine, leucine, methionine, phenylalanine and valine. Apart from methionine, arginine and isoleucine, the essential amino acids of bee-pollen from alfalfa, date palm, summer squash and rape exceeded the honeybees’ requirements. Methionine was the limiting amino acid in bee-pollens from the five selected sources. Concentrations of essential amino acids in the tested bee-pollens were variable and significantly correlated to their botanical origin of pollen. The date palm Phoenix dactylifera has played an important role in the day-to-day life of the people for the last 7000 years. Today worldwide production, utilization and industrialization of dates are continuously increasing since date fruits have earned great importance in human nutrition owing to their rich content of essential nutrients. Tons of date palm fruit wastes are discarded daily by the date processing industries leading to environmental problems. Wastes such as date pits represent an average of 10% of the date fruits. Thus, there is an urgent need to find suitable applications for this waste. In spite of several studies on date palm cultivation, their utilization and scope for utilizing date fruit in therapeutic applications, very few reviews are available and they are limited to the chemistry and pharmacology of the date fruits and phytochemical composition, nutritional significance and potential health benefits of date fruit consumption. In this context, in the present review the prospects of valorization of these date fruit processing by-products and wastes’ employing fermentation and enzyme processing technologies towards total utilization of this valuable commodity for the production of biofuels, biopolymers, biosurfactants, organic acids, antibiotics, industrial enzymes and other possible industrial chemicals are discussed. 相似文献
