The infestiveness of heat (56°C/5 min. and 50°C/5 min.), and cold (−10°C and −6°C) exposed Diphyllobothrium latum plerocercoids was studied by administration to golden hamsters (Mesocricetus auratus). The cold exposed larvae were subjected to an exposure, analogous the freezing of fish and which is cut off when the temperature has declined either to −6°C or −10°C. The plerocercoids were administered to the hamsters under inhalation anaesthesia, 5 larvae per hamster. Out of 150 normal control plerocercoids administered on 30 hamsters, 74 adult worms developed, indicating 49 % infestiveness. Tests carried out with 105 56°C/5 min. exposed larvae on 21 hamsters, 45 50°C/5 min. exposed larvae on 9 hamsters, and 100 larvae exposed to −10°C on 20 hamsters all gave negative results, while out of 50 −6°C exposed larvae on 10 hamsters 3 developed to adult worms. The results show that the 56°C/5 min. and −10°C exposed larvae, which in previous studies have been considered to be inactivated on the basis of the resultant permanent immobilization, were not able to develop further in the host. Thus permanent immobility seems to be a reliable criterion of larval death.  相似文献

Eggs of the stonefly, Arcynopteryx compacta, that overwinter in the alpine region of Norwegian mountains, increase their cold-hardiness by dehydration. Eggs enclosed in ice at −22°C survive the loss of about two-thirds of their total water content by shrinkage due to passive diffusion of body water along the concentration gradient. Fully hydrated eggs are killed by freezing at their supercooling point of −26°C, and by direct cooling to −30°C. Dehydrated eggs have a mean supercooling point of −31°C, and survive exposure at −27 and −29°C in ice. Judged from their melting points the eggs do not accumulate low-molecular-weight cryoprotective substances. The difference between freezing and melting points corresponds to a thermal hysteresis of up to 1.8°C. The presence of thermal hysteresis antifreezes may stabilize their supercooled state when enclosed by ice during overwintering. The eggs enter diapause in the autumn, and diapause completion is enhanced both by temperature and time during enclosure in ice.  相似文献

The lean fish Labeo rohita was subjected to liquid nitrogen vapor in a batch freezer to determine its freezing characteristics. The moisture content of the fish muscle tissue was about 75%. The initial and average freezing points were found to be − 1.34 and − 5.25 °C, respectively. The fish slab sample showed a decreased rate of temperature change in that region. The suggested cut-off freezing temperature was − 30 °C, corresponding to about 90% of total water frozen. Continued freezing below − 30 °C resulted in negligible freezing of residual liquid water in the sample. The effects of low-density polyethylene (LDPE) bags and aluminum foil on the freezing times and the overall heat transfer coefficients were sufficiently small that the authors recommended thicknesses of up to 0.1 and 0.5 mm for LDPE bags and aluminum foil, respectively. In these cases there was only 5% increase in freezing time compared to unpackaged fish samples.  相似文献

Lesions caused by plerocercoids of Triaenophorus nodulosus in the second intermediate hosts were studied in fish of an alpine lake, the Königssee. Four species (Arctic charr, Salvelinus alpinus; minnow, Phoxinus phoxinus; burbot, Lota lota: perch, Percafluviatilis) could be shown to be affected, whereas seven other fish species did not have macroscopical or histological signs of invasion. The capsules of T. nodulosus plerocercoids were usually located in liver tissue. Whereas only living parasites were found in charr and minnow, dead and destroyed plerocercoids could be demonstrated additionally in burbot and perch.
The histological picture is characterized by capsules surrounded by connective tissue and by fresh or older migration routes in charr and minnow. Furthermore, larvae migrating through the biliary ductules were found regularly in charr. In burbot and perch, different stages of destruction of the parasitic larvae took place. The ability to reject the parasite by immunological reactions seems to depend on the species of host. Even if heavy changes in the liver tissue could be demonstrated, no negative influences on condition and growth of affected fish could be established.  相似文献

The effect of household smoking, as carried out with three commercially available smoke boxes, on the infestiveness of Diphyllo-bothrium latum was investigated. The temperature changes in fish during the smoking were measured and the effect on tapeworm larvae estimated on the basis of a lethal temperature/time exposure of 56°C/5 min. For all the boxes smoking treatments sufficient to destroy the tapeworm larvae are suggested. The obtained results confirm the conclusion, made in previous studies on other heat preparation methods of fish, that well cooked fish in which all the parts have been subjected to a 56°C/5 min. exposure is safe with regard to the risk of Diphyllobothrium latum infestation.  相似文献

Cancer is the second major cause of death in the world. The problem of post-cancer infertility plays a significant role, because chemotherapy can be gonadotoxic. Cryopreservation of ovarian tissue before cancer therapy with re-implantation after convalescence is the potential key solution to this problem. The aim of this study was to test the viability of cryopreserved human ovarian cortex after long-term cooling in culture medium composed of permeable cryoprotectants. Ovarian fragments from sixteen patients were randomly divided into two groups. After the operation, tissue pieces assigned to both groups were cooled to 5 °C for 22–24 h, frozen and thawed. Group 1 pieces (n = 32) were cooled before cryopreservation in the standard culture medium, and Group 2 pieces (n = 32) were cooled in the freezing medium (culture medium+6% ethylene glycol+6% dimethyl sulfoxide+0.15 M sucrose). Freezing was performed in standard 5 ml cryo-vials with ice formation at −9 °C, cooling from −9 to −34 °C at a rate of −0.3 °C/min and plunging at −34 °C into liquid nitrogen. After thawing in a 100 °C (boiling) water bath, the removal of cryoprotectants was performed in 0.5 M sucrose with 20 min exposure in sucrose and 30 min stepping rehydration. The effectiveness of the pre-freezing cooling of tissue was evaluated by the development of follicles (histology). Six months after the autotransplantation, oocytes from the twenty-seven-year old, hormonally stimulated patient were retrieved and fertilized with her partner sperm through the intracytoplasmic spermatozoa injection (ICSI). For groups 1 and 2, 93.5 ± 1.9% and 96.4 ± 2.0% of the preantral follicles, respectively, were morphologically normal (P > 0.1) (with a tendency toward increasing in quality in Group 2). Six months after the auto-transplantation, two ICSI cycles resulted in the gathering and transplantation of high quality embryos, but no pregnancy had been established. Thirteen months after the auto-transplantation, the patient became spontaneously pregnant and delivered a healthy baby girl at term. Long-term (24 h) cooling of ovarian tissue to 5 °C before cryopreservation in the presence of permeable cryoprotectants simplifies the protocol of cryopreservation and has a tendency of increasing of the cells viability after thawing.  相似文献

In the enclosed fresh-water environmsnt of Hanningfield Reservoir, Essex, England, Anisakis sp. larvae (parasites of marine fish) were found in 55 per cent of 40 brown trout and in 26·2 per cent of 61 rainbow trout. Experimental infection by intubating larvae into the stomach was more successful in brown trout (50·6 per cent recovery rate) than in rainbow trout (27 per cent recovery rate). Some larvae reached the body-cavity as early as 2 h after infection. They penetrated the region between the oesophagus and intestine immediately posterior to the caecal openings. Fewer larvae successfully penetrated the gut wall of brown trout within 24 h at 8°C than at 15 ± 1°C. It appears that the reservoir trout acquired Anisakis by being fed as juveniles on untreated marine fish offal containing live larvae.  相似文献

  • 1 The survival of adult and first-instar Myzus persicae reared at 20°C and 10°C was investigated after brief (1 min) exposure in the absence of plant material to temperatures between −5°C and −25°C, and extended exposures on plants of 1–10 days at a constant 5°C, 3°C and −5°C and a 24 h cycling regime between 5°C (18 h) and −5°C (6 h).
  • 2 Life stage, rearing temperature, period of exposure and temperature regime all had a significant effect on the ability of aphids to survive cold. The effects of life stage and rearing temperature were most noticeable following exposure to cycling temperatures and extended exposures at −5°C, and least apparent after 1 min exposures at lower sub-zero temperatures.
  • 3 Mortality following exposure to temperatures cycling between −5°C and 5°C was greater than that at 3°C (the mean of the cycling temperatures) and less than at a constant −5°C, suggesting that when temperatures fluctuate by a few degrees around 0°C the minimum temperature may affect survival to a greater extent than the mean.
  • 4 These results suggest that an overwintering population of acclimated M.persicae would persist without significant mortality after a period of 7–10 days with −5°C frosts each night.

Eggs and larvae of the carangid fish, Caranx mate (Cuv. & Valenc.), were incubated at various temperature (17.2 to 33.1 °C) and salinity (10 to 42 ‰) combinations in five experiments. The following rates were directly proportional to temperature: embryonic development, yolk absorption, eye and jaw development, and increase in length. Unfed C. mate larvae attained a maximum size at 25 °C and 20 ‰ Eyes and jaws of larvae were functional by the end of the yolk sac stage at all temperature and salinity levels tested.Hatching success and larval survival at the end of the yolk sac stage were generally greater than 50 % between 22° and 32°C. Hatching success and larval survival at the end of the yolk sac stage were reduced at salinity extremes, especially in low temperature-low salinity and high temperature-high salinity combinations. The frequency of morphological abnormalities was also high at extreme temperatures and salinities.The incipient upper thermal TLm for unfed C. mate larvae acclimated to 23.8°C increased from 31.5°C for newly hatched larvae, to 34.2°C for 72 h larvae, but decreased to 32.0°C for starving larvae after the exhaustion of the yolk supply.  相似文献

The single radial diffusion test is an effective method for detection of nuclear polyhedrosis virus infection in Heliothis zea larvae. Virus antigens were detected in some instances 48 hr after the larvae were exposed to virus. Most larvae tested positively for virus antigens 72 hr after exposure to the virus. The tests could be read within 4 hr if the incubation temperature was 35°C, and within 24 hr at 22°C.  相似文献

Laboratory manipulations of ambient temperature were used to investigate the role of temperature in triggering or modulating cold-hardiness adaptations, supercooling-point depression and cryoprotectant accumulation, in larvae of the goldenrod gall moth, Epiblema scudderiana (Clemens), a freeze-intolerant species. Low temperature strongly facilitated cryoprotectant synthesis; larvae subjected to a 1°C per day decrease in temperature showed a major increase in the rate of glycerol synthesis when temperature fell below 5°C with highest rates of synthesis, greater than 90 μmol g−1 d−1, at temperatures between 0 and −10°C. Conversely, abrupt rewarming of larvae from −18 to 23°C in mid-November stimulated a rapid loss of glycerol (from a starting level of 1763 ± 278 μmol/g wet weight) with a half time of only 1.5 days. Supercooling-point depression was not keyed to ambient temperature but appeared to be an endogenous event occurring over the same time interval in laboratory animals held at warm or cold temperatures, as well as in outdoor animals. Rewarming of cold-adapted larvae in November resulted in only a small rise in supercooling point (and did not break diapause) but rewarming in February resulted in a 19°C increase in supercooling point in 4 days, followed rapidly by pupation.  相似文献

This study aimed to investigate temperature effect on physiological and biochemical responses of the marine medaka Oryzias melastigma larvae. The fish were subjected to a stepwise temperature change at a rate of 1 °C/h increasing or decreasing from 25 °C (the control) to six target temperatures (12, 13, 15, 20, 28 and 32 °C) respectively, followed by a 7-day thermal acclimation at each target temperature. The fish were fed ad libitum during the experiment. The results showed that cumulative mortalities were significantly increased at low temperatures (12 and 13 °C) and at the highest temperature (32 °C). For the survivors, their growth profile closely followed the left-skewed ‘thermal performance curve’. Routine oxygen consumption rates of fish larvae were significantly elevated at 32 °C but suppressed at 13 and 15 °C (due to a high mortality, larvae from 12 °C were not examined). Levels of heat shock proteins and activities of malate dehydrogenase and lactate dehydrogenase were also measured in fish larvae exposed at 15, 25 and 32 °C. The activities of both enzymes were significantly increased at both 15 and 32 °C, where the fish larvae probably suffered from thermal discomfort and increased anaerobic components so as to compensate the mismatch of energy demand and supply at these thermal extremes. Coincidently, heat shock proteins were also up-regulated at both 15 and 32 °C, enabling cellular protection. Moreover, the critical thermal maxima and minima of fish larvae increased significantly with increasing acclimation temperature, implying that the fish could develop some degrees of thermal tolerance through temperature acclimation.  相似文献

Ecological aspects of Steinernema diaprepesi isolate SRC were studied to evaluate the species potential as biological control agent of insect pests. Under laboratory conditions, the following aspects were determined: the nematode life cycle, pathogenicity to several arthropods, reproductive capacity, tolerance to desiccation, effect of temperature on survival and infectivity of infective juveniles (IJs), and influence of soil texture and soil water potential on the isolate. The parasitic cycle on last-instar larvae of Galleria mellonella at 25°C was completed 8 days after infection. The nematode showed high virulence to lepidopteran larvae, being limited or nil in the remaining orders of arthropods evaluated. An acceptable offspring production of S. diaprepesi was confirmed in the species G. mellonella and S. frugiperda, suggesting that the isolate would have potential for control of lepidopteran larvae. Optimum temperature for reproduction was 20–25°C. IJs survived exposure to a range of temperatures between 10 and 40°C, with a significant reduction in the number of live IJs at 40°C. The nematodes remained infective at 20–40°C. IJ mortality was 100% on day 6 of exposure to 85% RH. The movement of IJs observed in the soil column experiments revealed that the isolate uses a cruiser-type search strategy. Soil texture and water potential significantly influenced IJ movement, search and penetration of G. mellonella larvae. The efficacy of this isolate was found to be favoured in sandy soils, regardless of the soil water potential.  相似文献

Whole-body cryotherapy (WBC) and partial-body cryotherapy (PBC) are two methods of cold exposure (from −110 to −195 °C according to the manufacturers). However, temperature measurement in the cold chamber during a PBC exposure revealed temperatures ranging from −25 to −50 °C next to the skin of the subjects (using isolating layer placed between the sensor and the skin). This discrepancy is due to the human body heat transfer. Moreover, on the surface of the body, an air layer called the boundary layer is created during the exposure and limits heat transfer from the body to the cabin air. Incorporating forced convection in a chamber with a participant inside could reduce this boundary layer. The aim of this study was to explore the use of a new WBC technology based on forced convection (frontal unilateral wind) through the measurement of skin temperature. Fifteen individuals performed a 3-min WBC exposure at −40 °C with an average wind speed of 2.3 m s−1. The subjects wore a headband, a surgical mask, underwear, gloves and slippers. The skin temperature of the participants was measured with a thermal camera just before exposure, just after exposure and at 1, 3, 5, 10, 15 and 20 min after exposure. Mean skin temperature significantly dropped by 11 °C just after exposure (p<0.001) and then significantly increased during the 20-min post exposure period (p<0.001). No critically low skin temperature was observed at the end of the cold exposure. This decrease was greater than the mean decreases in all the cryosauna devices with reported exposures between −140 °C and −160 °C and those in two other WBC devices with reported exposures between −60 °C and −110 °C. The use of this new technology provides the ability to reach decreases in skin temperature similar to other technologies. The new chamber is suitable and relevant for use as a WBC device.  相似文献

Elevated environmental temperatures associated with anthropogenic warming have the potential to impact host‐parasite interactions, with consequences for population health and ecosystem functioning. One way that elevated temperatures might influence parasite prevalence and intensity is by increasing life cycle completion rates. Here, we investigate how elevated temperatures impact a critical phase of the life cycle of the bird tapeworm Schistocephalus solidus – the growth of plerocercoid larvae in host fish (three‐spined sticklebacks Gasterosteus aculeatus). By 8 weeks post‐infection, plerocercoids recovered from experimentally infected sticklebacks held at 20 °C weighed on average 104.9 mg, with all exceeding 50 mg, the mass considered consistently infective to definitive hosts. In contrast, plerocercoids from sticklebacks held at 15 °C weighed on average 26.5 mg, with none exceeding 50 mg. As small increases in plerocercoid mass affect adult fecundity disproportionately in this species, enhanced plerocercoid growth at higher temperatures predicts dramatically increased output of infective parasite stages. Subsequent screening of thermal preferences of sticklebacks from a population with endemic S. solidus infection demonstrated that fish harbouring infective plerocercoids show significant preferences for warmer temperatures. Our results therefore indicate that parasite transmission might be affected in at least two ways under anthropogenic warming; by enhancing rates of parasite growth and development, and by increasing the likelihood of hosts being able to seek out proliferating warmer microhabitats. Furthermore, our results suggest the potential for positive feedback between parasite growth and host thermal preferences, which could dramatically increase the effects of even small temperature increases. We discuss the possible mechanisms underpinning our results, their likely ecological consequences and highlight key areas for further research.  相似文献

Larvae of the goldenrod gall moth, Epiblema scudderiana (Clemens) utilize a freeze-avoidance strategy for winter survival. Cold-hardiness adaptations of an outdoor population of the species were profiled over the 1984–1985 winter. Over the autumn months supercooling points of the larvae dropped from −13.9±2.3°C to −37.8±2.8°C (the lowest winter temperature recorded was −26°C), water content of the larvae decreased from 57.2±1.2 to 24.8±1.6% of fresh weight, and glycerol content of the larvae rose to an average of 2030 μmol/g wet weight or 18.7% of fresh weight. All parameters stabilized over the mid-winter months. Glycerol production was largely accounted for by the loss of stored glycogen while lipid and protein reserves remained nearly constant over the winter months. Supercooling-point depression and glycerol systhesis both appeared to be initiated after the first overnight exposures to subzero temperatures. Highest rates of glycerol production, about 60 μmol g−1 d−1, were achieved with mean daily temperatures of about 0°C and subzero nights. Glycerol content was rapidly cleared in the spring but only 20% of the resulting carbon was restored as glycogen.  相似文献

The oomycete Leptolegnia chapmanii is among the most promising entomopathogens for biological control of Aedes aegypti. This mosquito vector breeds in small water collections, where this aquatic watermold pathogen can face short-term scenarios of challenging high or low temperatures during changing ambient conditions, but it is yet not well understood how extreme temperatures might affect the virulence and recycling capacities of this pathogen. We tested the effect of short-term exposure of encysted L. chapmanii zoospores (cysts) on A. aegypti larvae killed after infection by this pathogen to stressful low or high temperatures on virulence and production of cysts and oogonia, respectively. Cysts were exposed to temperature regimes between ?12 °C and 40 °C for 4, 6 or 8 h, and then their infectivity was tested against third instar larvae (L3) at 25 °C; in addition, production of cysts and oogonia on L3 killed by infection exposed to the same temperature regimes as well as their larvicidal activity were monitored. Virulence of cysts to larvae and the degree of zoosporogenesis on dead larvae under laboratory conditions were highest at 25 °C but were hampered or even blocked after 4 up to 8 h exposure of cysts or dead larvae at both the highest (35 °C and 40 °C) and the lowest (?12 °C) temperatures followed by subsequent incubation at 25 °C. The virulence of cysts was less affected by accelerated than by slow thawing from the frozen state. The production of oogonia on dead larvae was stimulated by short-term exposure to freezing temperatures (?12 °C and 0 °C) or cool temperatures (5 °C and 10 °C) but was not detected at higher temperatures (25 °C–40 °C). These findings emphasize the susceptibility of L. chapmanii to short-term temperature stresses and underscore its interest as an agent for biocontrol of mosquitoes in the tropics and subtropics, especially A. aegypti, that breed preferentially in small volumes of water that are generally protected from direct sunlight.  相似文献

The aim of this study was to determine if the plerocercoid larvae of Gymnorhynchus gigas, a common cestode of the ray's bream (Brama raii), possess antigenic compounds potentially capable of provoking anaphylactic episodes. A murine experimental model, using BALB/c mice, was developed to study the humoral immune response induced by G. gigas extracts. A highly specific humoral immune response was detected and cross-reactions were not observed between parasite and host antigens. The presence of IgM and IgG3 levels suggest the presence of thymus-independent antigens in the parasitic extract. The IgG antibody class showed the highest levels, with the IgG1 the predominant subclass. These IgG1 levels are in accordance with the supposed presence of a type I allergic reaction after the ingestion of G. gigas plerocercoids parasitizing fish, as well as inducing anaphylaxia in fish. These results indicate that somatic products released from ingested larvae of G. gigas could induce the development of a Th2 response capable of causing allergic disorders.  相似文献

This study of the bed bug, Cimex lectularius, examines tolerance of adult females to extremes in temperature and loss of body water. Although the supercooling point (SCP) of the bed bugs was approximately −20°C, all were killed by a direct 1 h exposure to −16°C. Thus, this species cannot tolerate freezing and is killed at temperatures well above its SCP. Neither cold acclimation at 4°C for 2 weeks nor dehydration (15% loss of water content) enhanced cold tolerance. However, bed bugs have the capacity for rapid cold hardening, i.e. a 1‐h exposure to 0°C improved their subsequent tolerance of −14 and −16°C. In response to heat stress, fewer than 20% of the bugs survived a 1‐h exposure to 46°C, and nearly all were killed at 48°C. Dehydration, heat acclimation at 30°C for 2 weeks and rapid heat hardening at 37°C for 1 h all failed to improve heat tolerance. Expression of the mRNAs encoding two heat shock proteins (Hsps), Hsp70 and Hsp90, was elevated in response to heat stress, cold stress and during dehydration and rehydration. The response of Hsp90 was more pronounced than that of Hsp70 during dehydration and rehydration. Our results define the tolerance limits for bed bugs to these commonly encountered stresses of temperature and low humidity and indicate a role for Hsps in responding to these stresses.  相似文献

During cold exposure, peripheral tissues undergo vasoconstriction to minimize heat loss to preserve the maintenance of a normal core temperature. However, vasoconstricted tissues exposed to cold temperatures are susceptible to freezing and frostbite-related tissue damage. Therefore, it is imperative to establish a mathematical model for the estimation of tissue necrosis due to cold stress. To this end, an explicit formula of finite difference method has been used to obtain the solution of Pennes' bio-heat equation with appropriate boundary conditions to estimate the temperature profiles of dermal and subdermal layers when exposed to severe cold temperatures. The discrete values of nodal temperature were calculated at the interfaces of skin and subcutaneous tissues with respect to the atmospheric temperatures of 25 °C, 20 °C, 15 °C, 5 °C, −5 °C and −10 °C. The results obtained were used to identify the scenarios under which various degrees of frostbite occur on the surface of skin as well as the dermal and subdermal areas. The explicit formula of finite difference method proposed in this model provides more accurate predictions as compared to other numerical methods. This model of predicting tissue temperatures provides researchers with a more accurate prediction of peripheral tissue temperature and, hence, the susceptibility to frostbite during severe cold exposure.  相似文献
