Bone mineralization was studied in rats. Animals were divided into three feeding groups: LCP - diet with 13.5% crude protein in DM (5% of gluten, 10% of casein), HCP - diet with 21.2% CP in DM (8% of gluten, 10% of casein), and LSM - diet based on grain meals and meat-bone meal (21% CP in DM). After 28 days feeding, animals were euthanased by cervical dislocation and femur bones were collected, weighed and kept frozen until analyses. Diets with 21% protein (HCP, LSM) significantly increased weight of femur bones. Despite of the substantially higher ash level (7.1%) in the LSM diet than in the LCP diet (3.4%), rats of both groups had the similar bone concentration of Ca (15.7 +/- 1.1 vs. 17.4 +/- 1.1 g/kg) and Zn (178.7 +/- 7.9 vs. 173.0 +/- 8.5 mg/kg). However bone density in LSM rats was significantly higher than in LCP ones. Although rats fed HCP diet had intermediate bone density, the bone concentration of Ca (11.4 +/- 0.5 g/kg) and Zn (145.1 +/- 2.9 mg/kg) was significantly lower, than in animals fed LCP and LSM diets. This was related to the very wide protein/calcium (37:1 g/g) and protein/zinc (5.3:1 g/mg) ratios in HCP diet. Those ratios were narrowest in the LSM diet: 16.2:1 (CP/Ca) and 2.6:1 (CP/Zn). It can be conluded that protein/mineral ratio in a diet is a very important factor in bone development, besides dietary protein and ash contents itselves.  相似文献

Few studies in wild animals have assessed changes in mineral profile in long bones and their implications for mechanical properties. We examined the effect of two diets differing in mineral content on the composition and mechanical properties of femora from two groups each with 13 free-ranging red deer hinds. Contents of Ca, P, Mg, K, Na, S, Cu, Fe, Mn, Se, Zn, B and Sr, Young’s modulus of elasticity (E), bending strength and work of fracture were assessed in the proximal part of the diaphysis (PD) and the mid-diaphysis (MD). Whole body measures were also recorded on the hinds. Compared to animals on control diets, those on supplemented diets increased live weight by 6.5 kg and their kidney fat index (KFI), but not carcass weight, body or organ size, femur size or cortical thickness. Supplemental feeding increased Mn content of bone by 23%, Cu by 9% and Zn by 6%. These differences showed a mean fourfold greater content of these minerals in supplemental diet, whereas femora did not reflect a 5.4 times greater content of major minerals (Na and P) in the diet. Lower content of B and Sr in supplemented diet also reduced femur B by 14% and Sr by 5%. There was a subtle effect of diet only on E and none on other mechanical properties. Thus, greater availability of microminerals but not major minerals in the diet is reflected in bone composition even before marked body effects, bone macro-structure or its mechanical properties are affected.  相似文献

54 bulls of the German Simmental breed were fed either on a high energy level (maize silage ad libitum and 1.8 kg concentrate) or on a low energy level (maize silage restrictively and 1.0 kg concentrate). In dependence on feeding intensity a mean daily weight gain of 870 or 1210 g was obtained. Animals were slaughtered with a live mass of 200 kg, 350 kg, 500 kg, 575 kg and 650 kg. Empty body was divided into 13 cuts and afterwards separated into lean, bone and adipose tissues and tendons. Major mineral element content was determined in these tissues as well as in the noncarcass parts.

In the lean tissue the mean content (200 kg) of 0.3 g calcium, 10 g phosphorus, 1 g magnesium, 2.3 g sodium and 14.8 g potassium/kg dry matter decreased slightly with rising live mass (200–650 kg). The contents of major mineral elements were much higher in bone tissue. For the fattening period from 200 to 650 kg of live mass mean contents of 151.5 g calcium, 71.3 g phosphorus, 3.2 g magnesium, 5.1 g sodium and 1.1 g potassium per kg DM were analysed. Mineral element content of bone tissue increased with rising live mass as well as animals on low feeding intensity showed a higher mineral content than on high energy level. In all, major mineral element content in fat tissue was very low. In noncarcass parts head and legs calcium and phosphorus had analogous to bone tissue the highest concentration. Hide showed a high content of sodium, whereas organs and digestive tract had a high content of potassium and phosphorus. Total mass of major mineral elements in the different tissues increased above all in the fattening period of 200 to 350 kg. In carcass as well as in empty body, mass of calcium and phosphorus was much higher than magnesium, sodium and potassium. Also animals on low feeding intensity showed a higher mass of major mineral elements in carcass and empty body than animals on high energy feeding intensity. The intensively fed bulls had a mean deposition of 12.7 g calcium, 6.9 g phosphorus, 0.37 g magnesium, 1.2 g sodium and 2.1 potassium per 1000 g of empty body weight gain, whereas restrictively fed bulls deposited in average 15.0 g calcium, 7.8 g phosphorus, 0.4 g magnesium, 1.2 g sodium and 2.4 g potassium per 1000 g of empty body weight gain.  相似文献

This study examined the effects of estrogen (17β-estradiol) and testosterone on the growth of long bones in male and female mice, with and without gonadectomy. Weight and nose-to-tail length were determined at 3 weeks of age at time of gonadectomy, 7 days later at the onset of hormone therapy, and throughout the treatment period. Gonadectomized mice exhibited an initial weight gain during the pretreatment period but length was unaffected. Hormone treatment altered weight gain in surgical and intact animals in a gender- and hormone-dependent manner. Estradiol enhanced weight gain in intact mice, but inhibited weight gain in ovariectomized mice. Lower doses of estradiol increased weight gain in orchiectomized mice at early time points. Testosterone increased weight in intact females and males, but not in gonadectomized mice. Estradiol increased nose-to-tail length in intact females at early time points, but inhibited length in ovariectomized females at later times, and it decreased length in intact males. Testosterone increased length in normal females and normal males. Serum Ca was unaffected by ovariectomy, but orchiectomy resulted in decreased levels. Estradiol reduced serum Ca in gonadectomized animals; serum Ca was increased by estradiol treatment in intact females. Changes in tibial bone weight, ash weight and mineral composition, and relative sizes of epiphyseal and metaphyseal bone were gender-, gonadectomy- and hormone-specific. Bone weight was greater in ovariectomized mice. Ash weight per bone was comparable, but there was an increase in Ca and P content with ovariectomy. Estradiol increased bone weight, ash content, and bone Ca and P in ovariectomized and intact females. Orchiectomy alone did not alter bone weight, ash content, or Ca and P, but orchiectomized mice were sensitive to estradiol; all parameters were increased in the orchiectomized animals treated with estradiol. Analysis of the ash content and Ca and P per mg bone, rather than per bone, demonstrated estradiol and testosterone alter net bone formation, but not the amount of mineral per unit bone. Ovariectomy increased hypertrophic cartilage. While estradiol did not alter tibial area in ovariectomized mice, it caused an increase in intact females. The total amount of growth plate cartilage in ovariectomized animals was decreased by estradiol to levels typical of intact animals due to a greater decrease in the hypertrophic cartilage in the ovariectomized mice, as well as a greater increase in metaphyseal bone area. Testosterone had no effect on these parameters in the females. Orchiectomy decreased the amount of growth plate cartilage, but increased the hypertrophic zone. Estradiol increased growth plate cartilage in intact male mice, but decreased it in orchiectomized mice. This difference was also seen in the hypertrophic zone. Total growth plate cartilage and hypertrophic cartilage were increased by testosterone in intact males, whereas metaphyseal and epiphyseal bone area were decreased. The results show for the first time that there is a gender-specific response in both male and female mice to both estradiol and testosterone, whether or not the animals have been gonadectomized. For many parameters, orchiectomized mice behave like females in response to both sex steroids, indicating that the male gonad is needed for mouse bone to exhibit the male phenotypic response to estradiol and testosterone.  相似文献

Two experiments, a performance experiment and a mineral balance study, were conducted on grower-finisher pigs (42 to 101 kg live weight) to investigate the effects of Peniophora lycii phytase enzyme and 25-hydroxyvitamin D3 (25-OHD3) on growth performance, carcass characteristics, nutrient retention and excretion, and bone and blood parameters. The two experiments were designed as a 2 × 2 factorial (two levels of phytase and two levels of 25-OHD3). The four diets were T1, low-phosphorous diet; T2, T1 + phytase; T3, T1 + 25-OHD3 and T4, T1 + phytase + 25-OHD3 diet. In all, 25 μg of 25-OHD3 was used to replace 1000 IU of vitamin D3 in diets T3 and T4. Diets were pelleted (70°C) and formulated to contain similar concentrations of energy (13.8 MJ DE/kg), lysine (9.5 g/kg) and digestible phosphorus (P; 1.8 g/kg). Neither the inclusion of phytase nor 25-OHD3 in the diet had any effect on pig performance. There was an interaction between phytase and 25-OHD3 on calcium (Ca) and P retention (P < 0.01) and on the apparent digestibility of ash (P < 0.01), P (P < 0.001) and Ca (P < 0.001). Pigs offered phytase diets only, had a higher retention of Ca and P and digestibility of ash (P < 0.01), P (P < 0.001) and Ca (P < 0.01) compared with pigs offered unsupplemented diets. However, when the combination of phytase and 25-OHD3 were offered, no effects were detected compared with 25-OHD3 diets only. Pigs fed phytase diets had higher bone ash (P < 0.01), bone P (P < 0.01) and bone Ca (P < 0.05) concentrations compared with pigs offered non-phytase diets. In conclusion, pigs offered phytase diets had a significantly increased bone ash, Ca and P than pigs offered unsupplemented phytase diets. However, there was no advantage to offering a combination of phytase and 25-OHD3 on either bone strength or mineral status compared to offering these feed additives separately.  相似文献

Several animal and clinical studies have shown that phytoestrogens, plant-derived estrogenic compounds, can be useful in treating postmenopausal osteoporosis. Phytoestrogens and phytoestrogen-containing plants are currently under active investigation for their role in estrogen-related disorders. The present study deals with anti-osteoporotic evaluation of phytoestrogen-rich plant Cuminum cyminum, commonly known as cumin. Adult Sprague-Dawley rats were bilaterally ovariectomized (OVX) and randomly assigned to 3 groups (10 rats/group). Additional 10 animals were sham operated. OVX and sham control groups were orally administered with vehicle while the other two OVX groups were administered 0.15 mg/kg estradiol and 1 g/kg of methanolic extract of Cuminum cyminum fruits (MCC) in two divided doses for 10 weeks. At the end of the study blood, bones and uteri of the animals were collected. Serum was evaluated for calcium, phosphorus, alkaline phosphatase and tartarate resistant acid phosphatase. Bone density, ash density, mineral content and mechanical strength of bones were evaluated. Scanning electron microscopic (SEM) analysis of bones (tibia) was performed. Results were analyzed using ANOVA and Tukeys multiple comparison test. MCC (1 g/kg, p.o.) significantly reduced urinary calcium excretion and significantly increased calcium content and mechanical strength of bones in comparison to OVX control. It showed greater bone and ash densities and improved microarchitecture of bones in SEM analysis. Unlike estradiol it did not affect body weight gain and weight of atrophic uterus in OVX animals. MCC prevented ovariectomy-induced bone loss in rats with no anabolic effect on atrophic uterus. The osteoprotective effect was comparable with estradiol.  相似文献

目的研究甲状旁腺激素相关蛋白1-34(PTHrP1-34)对荷瘤小鼠骨代谢的影响,同时观察肿瘤生长情况。方法对照组、模型组、实验组4周龄健康雌性BALB/c小鼠各12只,模型组和实验组采用乳腺癌组织块悬浊液注射法制备小鼠乳腺癌模型,10d后模型组每日予以生理盐水腹腔注射,实验组每日予以PTHrP1-34400μg/(kg.bw)腹腔注射。用药35 d后测全身骨密度(BMD)、股骨灰干重比、骨代谢相关血清指标[钙(Ca)、磷(P)、碱性磷酸酶(ALP)、骨钙素(OC)、Ⅰ型胶原C-末端交联顶端肽β(β-CTX)、骨唾液酸蛋白(BSP)],剥离肿瘤比较体积与质量。结果各组Ca水平差异无显著性(P〉0.05)。与对照组和模型组比较,实验组P、β-CTX、BSP等反映骨吸收的指标显著升高(P〈0.01或P〈0.05),总ALP、BGP等反映骨形成的指标显著降低(P〈0.01或P〈0.05);实验组BMD、股骨灰干重比均显著降低(P〈0.01)。与对照组相比,模型组各指标变化差异无显著性。实验组肿瘤体积和质量显著升高(P〈0.01)。结论在PTHrP1-34的作用下,荷瘤小鼠骨吸收大于骨形成,造成溶骨性骨破坏,骨密度降低,骨破坏释放的细胞因子可能促进肿瘤生长。  相似文献

The study aimed to investigate the effect of various doses of betaine supplemented dietary on Japanese quail performance, carcass characteristics, and blood chemistry. Therefore, 400 seven days old Japanese quails were classified randomly into four equal groups. Each group was subdivided into five replicates of 20 birds each. Four rations were formulated using four different betaine levels (0, 0.75, 1.5 and 2.25 g/kg, respectively) for five successive weeks. All groups received feed and clean water ad-libitum. The results of this trial indicated that the feed intake was lowered in groups fed with betaine (p ≤ 0.05) when compared with the control one. The highest weight gain (p ≤ 0.05) was noticed in groups fed diets BS4 (betaine supplementation at the rate of 2.25 g/kg). No difference among groups was observed in body length, shank length, shank diameter, and keel bone length or breast width. Also, the carcass weight and breast yield were highest (p ≤ 0.05) in the group reared on the BS4 diet. In addition, intestinal length and weight were significantly higher (p ≤ 0.05) in groups fed betaine with a concentration of 2.25 g/kg. Fat weight was lower in the group fed BS4 than in the untreated group. Significantly higher values of high-density lipoprotein (p ≤ 0.05) were observed in the group fed BS4. All groups fed a ration containing betaine showed lower levels of liver enzymes such as alanine amino transferase, alkaline phosphatase, and aspartate amino transferase (p ≤ 0.05) and lowered low-density lipoprotein level. The quails fed BS4 had the greatest growth hormones and insulin (p ≤ 0.05) and the lowest thyroxin level. We concluded that dietary betaine supplementation positively impacts Japanese quail growth performance, carcass traits, and blood chemistry.  相似文献

Wheat infected naturally withFusarium, contaminated mainly with 18.53 mg DON per kg, was added to a total constant wheat proportion of 400g/kg diet. Control and DON contaminated feed was fed for 11 weeks underad libitum and restrictive feeding conditions to 48 pigs of both sexes, which were randomly divided into 4 groups. Effects on performance (live weight range between 26 and 100kg), duration of feed intake and blood parameters were investigated. Parallel to this study, a balance study was carried out to examine the effects on nutrient digestibility and DON metabolism. The group fed the DON contaminated rationad libitum consumed 15% less feed and gained 14% less live weight compared to thead libitum control group, while the feed to gain ratio was unaffected. Under restrictive feeding conditions (DON and control) pigs exhibited 33%, 25% and 10% lower feed consumption, live weight gain and feed to gain ratio, respectively, than the control group fedad libitum. Metabolizability of energy, digestibility of organic matter, crude protein, crude fat and N-retention were significantly increased by 4, 3, 6, 11 and 10%, respectively, in the DON group of the restrictively fed pigs. In average up to 43.2% of the ingested DON, as the parent toxin in both groups, was eliminated with the urine and up to 3.0% with faeces. 目的研究饲料中不同的钙磷含量和钙磷比对生长期实验大鼠体重及骨骼发育的影响。方法采用完全随机化设计方案,取1月龄(100±10)g清洁级Wistar雄性大鼠70只,随机分成7组,每组10只,饲喂7种不同钙磷含量的饲料。自由采食,饮用去离子水,试验期42 d。各组于实验开始和结束时称重。试验结束后,处死动物并分离一侧胫骨和股骨用于实验指标的检测。结果当保持饲料中钙或者磷中的一种含量不变的情况下,调整另一种的含量即改变钙磷比例时,1.2∶1的正常钙磷比例组生长期实验大鼠增重及骨骼发育要明显好于0.4∶1的低钙磷比例组和4∶1的高钙磷比例组(P〈0.05),并且当保持饲料中钙磷比例不变时,即使在低钙低磷和高钙高磷进食水平下,正常钙磷比例组大鼠增重及骨骼发育都较好,但低钙磷比例组和高钙磷比例组大鼠增重及骨骼发育则由于饲料中钙磷含量的变化波动较大(P〈0.05)。结论饲料中钙和磷需按比例添加,如果两者绝对含量相差过大就会影响到动物的增重及骨骼发育,本实验证明饲料钙磷比为1.2∶1时生长期实验大鼠增重及骨骼发育较好。  相似文献

This study aimed to investigate effects of restricted calcium intake on cortical and trabecular bone density in white rats. Low Ca diet was fed for six weeks, and bone density and bone metabolism parameters were assessed in blood. This study was carried out on 12 male white rats aged 12 weeks (Sprague-Dawley; SD). These rats were bred for 1 week and randomly assigned to the standard calcium diet group (SCa group, n = 6) and the low calcium diet group (LCa group; n = 6). The SCa group was given a modified AIN-93M mineral mix (with 0.5% Ca), which was made by adding calcium to a standard AIN93 diet, and the LCa Group was fed a modified AIN-93 Mineral mix (with 0.1% Ca). Femoral BMD and BMC were measured by DEXA in each rat. After trabecular bone was separated from cortical bone, volumetric bone mineral density (vBMD) was measured using pQCT. Serum Ca and P levels were measured as parameters of bone metabolism, and S-ALP, S-TrACP and-Dpd levels were also measured. The results revealed no significant differences in weight, growth rate, feed consumption and feed efficiency between the two groups before and after calcium-restricted diet (p > .05). No significant differences were also observed in bone length and bone mass between the two groups (p > .05). Although bilateral femoral BMDs were not significantly different between the two groups, bilateral femoral BMCs significantly decreased in the LCa group, compared with the SCa group (p = .023, p = .047). Bilateral cortical MDs were not significantly different between the two groups, either. However, trabecular BMD significantly decreased in the LCa group, compared with the SCa group (p = .041). U-Dpd and S-TrACP levels significantly declined in the LCa group, compared to the SCa group (p = .039, p = .010). There were no significant differences in serum Ca and P levels between the two groups (p > .05). However, a significant decrease in urinary Ca level (p = .001) and a significant increase in urinary P (p = .001) were observed in the LCa group, compared to the Sca group. These findings described that six-week low calcium diet led to decreased trabecular bone density, reduced urinary excretion of Ca and increased urinary excretion of P. Mineral requirements of pregnant dairy goats are still not well defined; therefore, we investigated the net Ca, P, Mg, Na and K requirements for pregnancy and for maintenance during pregnancy in two separate experiments. Experiment 1 was performed to estimate the net Ca, P, Mg, Na and K requirements in goats carrying single or twin fetuses from 50 to 140 days of pregnancy (DOP). The net mineral requirements for pregnancy were determined by measuring mineral deposition in gravid uterus and mammary gland after comparative slaughter. In total, 57 dairy goats of two breeds (Oberhasli or Saanen), in their third or fourth parturition, were randomly assigned to groups based on litter size (single or twin) and day of slaughter (50, 80, 110 and 140 DOP) in a fully factorial design. Net mineral accretion for pregnancy did not differ by goat breed. The total daily Ca, P, Mg, Na and K requirements for pregnancy were greatest in goats carrying twins (P<0.05), and the requirements increased as pregnancy progressed. Experiment 2 was performed to estimate net Ca, P, Mg, Na and K requirements for dairy goat maintenance during pregnancy. In total, 58 dairy goats (Oberhasli and Saanen) carrying twin fetuses were assigned to groups based on slaughter day (80, 110 and 140 DOP) and feed restriction (ad libitum, 20% and 40% feed restriction) in a randomized block design. The net Ca, P and Mg requirements for maintenance did not vary by breed or over the course of pregnancy. The daily net requirements of Ca, P and Mg for maintenance were 60.4, 31.1 and 2.42 mg/kg live BW (LBW), respectively. The daily net Na requirement for maintenance was greater in Saanen goats (11.8 mg/kg LBW) than in Oberhasli goats (8.96 mg/kg LBW; P<0.05). Daily net K requirements increased as pregnancy progressed from 8.73 to 15.4 mg/kg LBW (P<0.01). The findings of this study will guide design of diets with adequate mineral content for pregnant goats throughout their pregnancy.  相似文献

The hamster was used as a model for investigating the effect of low, moderate, and high protein intake (12, 18, and 36% casein) on bone mineral content. Animals fed the low level of protein between 3 and 8 months of age had a reduction in the weight of all skeletal components measured, with the exception of the diaphyseal portion of the long bones. Diaphyseal weight and calcium remained significantly lower when expressed as a percentage of body weight. However, urinary calcium excretion was not lower than that of animals consuming an adequate protein intake. Ingesting a high protein diet resulted in a significant increase in urinary calcium excretion, and a reduced amount of both mineral and organic material in the diaphyses. We conclude that long-term consumption of a high protein ration led to the development of a mild osteoporotic condition in the hamster which was limited to the diaphyseal portions of the long bones.  相似文献

The purpose of this study was to evaluate the effects of vanadium absorbed by Coprinus comatus (VACC) treatment on bone in streptozotocin (STZ)-induced diabetic rats. Forty-five Wistar female rats used were divided into three groups: (1) normal rats (control), (2) diabetic rats, and (3) diabetic rats treated with VACC. Normal and diabetic rats were given physiological saline, and VACC-treated rats were administered VACC intragastrically at doses of 0.18 mg vanadium/kg body weight once daily. Treatments were performed over a 12-week period. At sacrifice, one tibia and one femur were removed, subjected to micro computed tomography (micro-CT) for determination of trabecular bone structure, and then processed for histomorphometry to assess bone turnover. Another femoral was used for mechanical testing. In addition, bone samples were collected to evaluate the content of mineral substances in bones. Treatment with VACC increased trabecular bone volume fraction in diabetic rats. Vanadium-treated animals had significant increases in ultimate load, trabecular thickness, and osteoblast surface. However, vanadium treatment did not seem to affect bone stiffness, bone energy absorption, trabecular separation, and osteoclast number. P levels in the femurs of diabetic rats treated with VACC were significantly higher than those of diabetic animals. Ca levels in diabetic and diabetic rats treated with vanadium showed no obvious changes. In conclusion, our results provide an important proof of concept that VACC may represent a powerful approach to treating or reversing diabetic osteopathy in humans.  相似文献

This paper reports that the selective beta(2)-adrenergic receptor agonist clenbuterol affects bone metabolism in growing 3-mo-old male Wistar rats treated over 8 wk. Thirty-two 3-mo-old growing Wistar rats weighing 234 +/- 2 g were assigned to a progressive isometric force, strength-training exercise program plus oral clenbuterol (2 mg x kg body wt(-1) x day(-1)) for 5 days each week, exercise program without clenbuterol 5 days each week, no exercise program plus oral clenbuterol (2 mg x kg(-1) x day(-1)) for 5 days each week, or no exercise without clenbuterol 5 days each week. At the end of 8 wk, lean mass, fat mass, and right total femoral, distal metaphyseal femoral, and diaphyseal femoral bone mineral density were measured by Hologic QDR 4,500 dual X-ray absorptiometry (DEXA) technique. Left femoral bones were harvested after death on day 58, and femoral resistance was determined by three-point bending testing. We found that fat mass was decreased in rats given strength training exercise and decreased further in rats treated with clenbuterol. Lean mass was increased in clenbuterol-treated animals. Strength-training exercise appeared to have no effect on bone mineral density, serum osteocalcin, or urinary deoxypyridinoline. However, clenbuterol treatment decreased femoral length, diameter, bone mineral density, and mechanical resistance. Clenbuterol had no effect on osteocalcin but increased urinary deoxypyridinoline. We concluded that clenbuterol treatment decreased bone mineral density and increased bone resorption independent of the level of exercise rats were given.  相似文献

Two experiments comprising a total of 80 Danish Landrace pigs in the period 20–90 kg live-weight were carried out to study the effect of dietary levels and ratios of Ca and P on serum values of Ca, inorg. P, and alkaline phosphatase and on the development of degenerative arthritis and atrophic rhinitis. In both experiments each kg feed (Table 1) was supplemented with 600 i. u. vitamin D3 and 80 p. p. m. Zn. In the first experiment the Ca:P ratio was 1.2 and the amount of Ca increased from 0.48 % to 1.20 % without influencing daily gain, feed conversion, carcass performance (Table 2) or frequency of atrophic rhinitis (Table 3). The Ca:P ratio in scapula was 2.15. Only when the diet was supplemented with 1.20 % Ga and 1.00 % P the bone quality was histomorphologically adequate (Figs. 1–4). In the second experiment various Ca:P ratios (ranging from 0.1 to 3.0) were compared with the recommended level and ratio of 0.72 % Ga and 0.60 % P. Performance of pigs on the normal level was better than for the other pigs (Table 4). Clinically, various degrees of leg-weakness and -deformation occurred, most pronounced in the high Ca — low P group. Very little lameness and no convulsions were observed. Marked hypocalcemia was not found. Unilateral high Ca intake increased serum Ca significantly, and serum alkaline phosphatase slightly, and decreased serum P (Figs. 5–7). Advanced pericytic osteolysis, poor mineralization and generalized osteodystrophia fibrosa were found in the low Ca groups (Figs. 8–13). The groups showed no differences in frequency of atrophic rhinitis (Table 5), and rickets was not observed. The incidence of degenerative joint lesions was not increased in pigs on the unfavourable Ca:P ratios, and depended apparently more on litters than on diets.  相似文献

OBJECTIVE: The objective of this study was to consider the effects of boron (B) and calcium (Ca) supplementation on mechanical properties of bone tissues and mineral content of the selected bones in rats. METHODS: Adult male Sprague Dawley rats underwent three different treatments with boron and calcium in their drinking water, while taking diet ad libitum for 4 weeks. Rats in the three treatment groups received 2 mg B/d, 300 mg Ca/d, and a combination of 2 mg B+ 300 mg Ca/d, respectively. After the experimental period body weights were recorded and bone mechanical properties were determined on the tibiae, femurs, and fifth lumbar vertebral bones and the mineral contents of these bones was calculated as the ash percentage. RESULTS: Better measurement of bone mechanical properties were observed for boron supplementation. The stiffness of the lumbar vertebral bones tended to increase in all groups and was significant for Ca supplementation. The significant maximal load obtained for boron in all bones indicates higher strength and less strength for apparently a high level of calcium, while this negative defect in the case of lumbar vertebral bones was corrected in the presence of boron. Highest mean energy to maximal load was shown with boron supplementation, demonstrating significant values with Ca group, and lower energy for the lumbar vertebral bones in Ca group in comparison with the controls. Less deformation at the yield points was shown in Ca group. There were no significant differences in ash weights among the four groups. CONCLUSIONS: Additional and longer studies are warranted to further determine the effects of supplemental boron with different calcium levels and possibly other minerals involved in bone mechanical properties in rats.  相似文献

The aim of the present study was to examine long-term effects of low levels of ergot alkaloids on growing bulls. Natural grown ergot with a mean total alkaloid concentrations of 633 mg/kg, and ergotamine (25%), ergocristine (15%) and ergosine (13%) as the most prominent alkaloids, was used. In a dose-response study 38 Holstein Friesian bulls were fed with three different doses of this ergot (0, 0.45 and 2.25 g/kg concentrate corresponding to an average total alkaloid concentration of the daily ration of 0, 69 and 421 microg/kg DM) over a period of approximately 230 days. Live weight, feed intake and health condition were monitored over the entire test period. The bulls were slaughtered at a live weight of approximately 550 kg. Carcass composition and quality were recorded and samples of liver, muscle, kidneys, fat, bile, urine and blood were analysed for ergot alkaloids. Liver enzyme activities and total bilirubin were measured in the blood. Statistically, no significant differences were detectable between the three feeding groups. Mean live weight gain over all groups was 1.41 kg/d with a mean dry matter intake of 7.35 kg/d. No carry over into tissues could be proved out of the experiment. To derive a no-effect level for beef cattle further research including higher ergot doses will be necessary.  相似文献

Two experiments in a 2 x 2 factorial arrangement were conducted to evaluate the effect of crude protein (CP) (130 vs. 200 g/kg) and phosphorus (P) (4.0 vs. 6.0 g total P/kg) level in a phytase supplemented diet (500 FTU [phytase units]/kg) in grower-finisher pigs. Owing to the design of the experiment, as dietary P level increased, there was also an increase in dietary calcium (Ca) level in order to maintain a dietary Ca to P ratio of 1.6:1. In Experiment 1, four diets were fed to 56 pigs (n = 14, initial body weight [BW] 36.7 +/- 4.2 kg) to investigate the interaction between CP and P on growth performance, bone mineralisation and digesta pH. Experiment 2 consisted of 16 entire male pigs (n = 4; offered identical diets to that offered in Experiment 1) for the determination of total tract apparent digestibility and nitrogen (N), P and Ca utilisation. There was an interaction between CP and P level on bone ash, bone P and bone Ca concentrations (p < 0.05). Pigs offered low CP-low P diets had a higher bone ash, P and Ca concentrations than pigs offered high CP-low P diets. However, there was no effect of CP level at high P levels on bone ash, P and Ca concentrations. Pigs offered low P diets had a lower ileal pH compared with pigs offered high P diets (p < 0.05). In conclusion, offering pigs a high CP-low P, phytase-supplemented diet resulted in a decrease in bone mineralisation.  相似文献

Intramuscular injections with botulinum toxin A (BTX-A) lead to a rapid decrease in muscle mass and force, but the effect of this drug on bone development is unclear. In the present pilot study we evaluated the effect of a one-time injection of BTXA in growing rabbits. Twelve young (weight 1.5 kg) New Zealand rabbits were randomly assigned to receive either BTX-A (total dose 8 units per kg body weight) or sodium chloride 0.9% injections into the left quadriceps and gastrocnemius muscles. Both groups continued to gain weight in a similar manner following the injection. However, when the animals were sacrificed at five weeks after the injection, the group receiving BTX-A had a significant deficit (of 10%) in gastrocnemius muscle mass on the injected side, whereas no significant side-difference was found for the quadriceps. BTX-A injections did not affect the length of the tibia. Nevertheless, bone mineral content of the whole tibia, as measured by dual-energy X-ray absorptiometry, was 7% lower in the BTX-A injected side than on the contralateral side. Peripheral quantitative computed tomography showed that this bone mass deficit was larger in the metaphysis than in the epiphysis or diaphysis. In the diaphysis, the bone mass deficit was due to a reduction in cross-sectional bone dimensions, which equally affected the cross-section of the entire bone, the cortical compartment and the marrow space. BTX-A injections did not have a detectable effect on cortical bone mineral density. The bone mass deficit in the diaphysis thus appeared to be caused by a lack of periosteal bone apposition rather than increased endocortical or intracortical resorption. These preliminary data suggest that intramuscular BTX-A injections can have a deleterious effect on the development of bones that are loaded by the injected muscles.  相似文献
