The endocrine-disrupting chemical bisphenol A (BPA) is used to manufacture plastics including food containers, and it may leach into these containers. Consumption of BPA that has leached out of plastics may be harmful as recent research highlighted that BPA can induce alterations in the nervous system. In the present work, we studied the effects of BPA on Ca2+ channels in dorsal root ganglion (DRG) neurons. Using whole-cell patch-clamp recordings, we found that I Ca could be reduced by BPA in a concentration-dependent manner. Additionally, BPA shifted the activation curve of calcium currents toward a depolarizing direction and increased the slope factor of the curve. The inactivation curve for the currents was also assessed, and the curve shifted toward the depolarizing direction, although it was not significant. Moreover, inhibitory effects of BPA on the increments of intracellular Ca2+ concentrations ([Ca2+]i) induced by 50 mM KCl were observed in DRG neurons using a laser scanning confocal microscopy assay. Further work revealed that the PKA and PKC pathways may be involved in the inhibitory effects of BPA since the PKA antagonist GÖ-6983 and the PKC antagonist H-89 significantly alleviated the inhibitory effects of BPA on I Ca. As such, the results of the present study provide direct evidence that BPA decreases I Ca and impairs calcium homeostasis, which may be involved in any toxic effects of BPA on DRG neurons.  相似文献

研究低氧、复氧对乳鼠心肌细胞内钙离子浓度的影响,以及牛磺酸在模拟心肌缺血/再灌注(I/R)过程中对细胞内钙的调节作用。采用SD大鼠乳鼠进行心肌细胞培养,建立模拟I/R模型。以Fluo-4/AM荧光指示剂负载,应用激光共聚焦显微镜技术(confocal laser scanning microscope,CLSM)检测心肌细胞钙离子浓度的变化。对照组心肌细胞内钙离子荧光强度(23.71±2.37U)较低;低氧180 min后复氧即刻,钙离子荧光强度开始增加(57.52±8.31U),复氧180 min后钙离子荧光强度(71.13±4.74U)显著增高(P<0.01vs对照组)。而牛磺酸组细胞内钙离子荧光强度较模拟I/R组显著降低[(42.42±4.17U)vs(71.13±4.74U),P<0.01]。心肌细胞缺血/缺氧导致Ca2+超载;模拟I/R Ca2+超载加剧,而牛磺酸有明显减轻心肌细胞模拟I/R时Ca2+超载的作用。  相似文献

Background

Any inflammatory response following corneal transplantation may induce rejection and irreversible graft failure. The purpose of this study is to analyze the anti-inflammatory effect of azithromycin (AZM) following experimental keratoplasty in rats.

Methods

Corneal transplants were performed between Fisher-donor and Lewis-recipient rats. Recipients were postoperatively treated three times daily with AZM, miglyol, ofloxacin or dexamethasone eye drops. As an additional control, AZM was applied following syngeneic keratoplasty. Furthermore, short-term treatments with AZM for seven days perioperatively or with AZM only three days prior to the transplantation were compared to appropriate controls. All transplants were monitored clinically for opacity, edema, and vascularization. Infiltrating CD45+, CD4+, CD8+, CD25+, CD161+ and CD163+ cells were quantified via immunohistochemistry.

Results

AZM significantly promoted corneal graft survival compared with miglyol or ofloxacin treatment. This effect was comparable to topical dexamethasone. No adverse AZM effect was observed. Histology confirmed a significant reduction of infiltrating leukocytes. The short-term application of AZM for three days prior to transplantation or for seven days perioperatively reduced corneal graft rejection significantly compared with the controls.

Conclusions

Along with antibiotic properties, topical AZM has a strong anti-inflammatory effect. Following keratoplasty, this effect is comparable to topical dexamethasone without the risk of steroid-induced adverse effects. Short-term treatment with AZM three days prior to the transplantation was sufficient to promote graft survival in the rat keratoplasty model. We therefore suggest further assessing the anti-inflammatory function of topical AZM following keratoplasty in humans.  相似文献

The annual average degree of phosphorus retention (both TP and OP) in reservoirs was found to depend mainly on the theoretical retention time (RT). The phosphorus retention (RP) is shown for some reservoirs in Czech Republic to be well approximated (standard deviation = 10.9) with RP(TP) = 74.7* (1-exp(-0.0255* RT)) (N = 38, 7 ≤ RT ≤ 604 days). The inclusion of 13 additional reservoirs for which data were assembled from literature resulted in an almost identical relationship: RP(TP) = 76.1* (1-exp(-0.0282* RT)) (N = 51, range of RT nearly unchanged). The retention of soluble reactive phosphorus RP(OP) studied on data from literature seems to be higher than that of TP. Statistical comparison with data for RP(TP) of lakes has shown a significantly lower retention in lakes than in reservoirs for comparable RP, resulting in RP(TP) = 66.0* (1-exp(-0.00419* RT)) (N = 59, 5.8 ≤ RT ≤ 260000 days). The differences between RP(OP) and RP(TP) and differentiation of lakes and reservoirs are discussed and a need for more thorough theoretical analysis stressed. 采用全细胞膜片钳技术观察不同浓度葡萄糖对新生Wister大鼠胰岛β细胞膜上电压依赖性L-型钙离子通道门控特性的影响,即分别用2.8、5.5、16.7和22.2 mmol/L的葡萄糖刺激单个贴壁胰岛β细胞,以Ba2+作为载流子,分析比较葡萄糖对L-型钙通道电流的影响。结果显示:在低糖(2.8 mmol/L)情况下,大鼠胰岛β细胞电压依赖性L-型钙离子通道电流静息膜电位约为-70 mV,钙离子内流不明显,且无明显的时间依赖性关系。在葡萄糖浓度为5.5 mmol/L的条件下,大鼠胰岛β细胞电压依赖性L-型钙离子通道电流在-40 mV激活, +20 mV左右达峰值;高糖(16.7 mmol/L)作用胰岛β细胞后,电压依赖性L-型钙离子通道电流约-40 mV激活,+10 mV左右达峰值,即峰值电位向负方向移动约10 mV;葡萄糖浓度达22.2 mmol/L时,电活动呈持续性去极化,峰值电位增加不明显,提示葡萄糖降低胰岛β细胞电压依赖性L-型钙通道电流的激活电位阈值,促进其开放,钙电流峰值电位增加,随着高糖作用时间的延长,胰岛β细胞容积变大,细胞膜破坏。提示高浓度葡萄糖在一定范围内可以刺激胰岛素的分泌,但浓度过高则可抑制胰岛素的分泌,通过观察葡萄糖刺激的胰岛β细胞胰岛素第一时相分泌的变化,在一定程度上对高糖毒性作用的可能提供了证据。  相似文献

目的研究饲料中不同的钙磷含量和钙磷比对生长期实验大鼠体重及骨骼发育的影响。方法采用完全随机化设计方案,取1月龄(100±10)g清洁级Wistar雄性大鼠70只,随机分成7组,每组10只,饲喂7种不同钙磷含量的饲料。自由采食,饮用去离子水,试验期42 d。各组于实验开始和结束时称重。试验结束后,处死动物并分离一侧胫骨和股骨用于实验指标的检测。结果当保持饲料中钙或者磷中的一种含量不变的情况下,调整另一种的含量即改变钙磷比例时,1.2∶1的正常钙磷比例组生长期实验大鼠增重及骨骼发育要明显好于0.4∶1的低钙磷比例组和4∶1的高钙磷比例组(P〈0.05),并且当保持饲料中钙磷比例不变时,即使在低钙低磷和高钙高磷进食水平下,正常钙磷比例组大鼠增重及骨骼发育都较好,但低钙磷比例组和高钙磷比例组大鼠增重及骨骼发育则由于饲料中钙磷含量的变化波动较大(P〈0.05)。结论饲料中钙和磷需按比例添加,如果两者绝对含量相差过大就会影响到动物的增重及骨骼发育,本实验证明饲料钙磷比为1.2∶1时生长期实验大鼠增重及骨骼发育较好。  相似文献

本文对血清岩藻糖测定方法进行了系统研究。血清用量由200μL减至50μL,显色反应4h内稳定。吸收峰在396nm。岩藻糖浓度40μg/mL内线性良好,批间CV=2.1%。以Sephadex-G200层析,血清岩藻糖主要存在于分子量为500kD的组分中。以本法测得30例健康人血清岩藻糖浓度为588.1±172.0μmol/L。  相似文献

Abstract: A procedure was established for determining the calcium content of mitochondria isolated from rat brain subregions based on changes in fura-2 fluorescence after disruption of the organelles with Triton X-100 and sodium dodecyl sulfate. Mitochondria isolated from the forebrain of normal rats contained 2.5 ± 0.9 nmol of calcium/mg of protein. A 30-min ischemic period produced an approximately twofold increase in the calcium content of mitochondria isolated from the dorsolateral striatum, a region in which most neurons die within 24 h after this period of ischemia. The calcium content of mitochondria from the paramedian cortex, a region in which there are few ischemia-susceptible neurons, tended to be similarly increased, although this difference was not statistically significant. Larger increases (to approximately five times control values) were seen in mitochondria isolated from both regions after 10 min of recirculation. By 1 h of recirculation, mitochondrial calcium had returned close to preischemic control values in both regions. Longer recirculation periods produced no further changes in the calcium content of mitochondria from the paramedian cortex. However, mitochondrial calcium was again increased in the dorsolateral striatum after 6 h (6.5 nmol of calcium/mg of protein) and 24 h (8.7 nmol of calcium/mg of protein) of recirculation. This regionally selective increase in calcium in the dorsolateral striatum preceded the period during which the majority of neurons in this region exhibit advanced degenerative changes. Thus, this increase may be an essential step, albeit a late one, in the development of neuronal loss.  相似文献

Serum phosphorus (P) concentration is associated with coronary artery calcification (CAC) as well as cardiovascular events in patients with chronic kidney disease. It has been suggested that this relationship is extended to subjects without renal dysfunction, but further explorations in diverse races and regions are still needed. We performed a cross-sectional study of 2,509 Korean subjects (Far Eastern Asian) with an estimated glomerular filtration rate of ≥60 ml/min/1.73m2 and who underwent coronary computerized tomography. Serum P concentration was divided into pre-determined 4 categories: ≤3.2, 3.2< to ≤3.6, 3.6< to ≤4.0 and >4.0 mg/dL. Agatston score (AS), an index of CAC, was divided into 3 categories: 0, 0< to ≤100, and >100. A multinomial logit model (baseline outcome: AS = 0) was applied to estimate the odds ratio (OR) for each serum P category (reference: ≤3.2mg/dL). Mean age of subjects was 53.5±9.1 years and 36.9% were female. In the adjusted model, serum P concentration of 3.6< to ≤4.0 mg/dL and >4.0 mg/dL showed high ORs for AS of >100 [OR: 1.58, 95% confidence interval (CI): 1.04–2.40 and OR: 2.11, 95% CI: 1.34–3.32, respectively]. A unit (mg/dL) increase in serum P concentration was associated with 50% increase in risk of AS >100 (OR: 1.50, 95% CI: 1.16–1.94). A higher serum P concentration, even within a normal range, may be associated with a higher CAC in subjects with normal renal function.  相似文献

目的:探讨中药异位康冲剂对小鼠子宫内膜异位症的作用及可能机理。方法:通过对小鼠一般情况、血常规、肝肾功及各主要脏器的组织学观察,观察中药异位康冲剂急性和长期毒副作用。通过一般情况、阴道脱落细胞观察、激素检测、病理组织学检查及血液流变学的改变,了解中药异位康冲剂对小鼠子宫内膜异位症模型的药理作用。结果:急性和长期毒性实验表明,中药异位康冲剂无明显毒性反应。异位康冲剂能抑制大鼠子宫内膜异位模型,降低血清雌二醇水平(P〈0.01)。病理学观察显示异位子宫的内膜腺体及腺上皮明显萎缩,其作用与丹那唑相似。结论:中药异位康冲剂无明显毒副作用,可促进子宫内膜异位组织细胞凋亡、抑制子宫内膜异位组织增殖,为临床用药提供了实验依据。  相似文献

To study the effect of salt (NaCl) on root elongation we developeda device that measures this effect by means of a Linear VariableDifferential Transformer (LVDT). To test the efficacy of thedevice we performed experiments demonstrating that (a) ratesof elongation of primary maize (Zea mays L.) roots were comparableto elongation rates of primary roots growing freely in solutionculture; and (b) chilling and low O2 concentrations of the solutionelicited the expected responses. Inhibition of root elongation by 75 mol m–3 NaCl was gradual.At an iso-osmotic concentration, mannitol did not inhibit rootgrowth, suggesting that the inhibition was not due to osmoticfactors but rather to effects of salt on metabolism. The additionof supplemental Ca (10 mol m–3) ameliorated this stressfulcondition. Timing of the application of Ca was critical. Treatmentwith Ca after addition of NaCl only partially restored growth,but pretreatment with Ca completely prevented the inhibitionof growth by salt stress.
