Competition for sulfate and ethanol among Desulfobacter, Desulfobulbus, and Desulfovibrio species isolated from estuarine sediments was studied in energy-limited chemostats. Desulfovibrio baculatus was the most successful competitor for limiting amounts of sulfate and ethanol, followed by Desulfobulbus propionicus. The success of Desulfovibrio baculatus was dependent on the availability of sufficient iron. Of the three species studied, Desulfobacter postgatei was the least successful competitor for limiting amounts of sulfate. Although stimulating the growth of Desulfobacter postgatei, addition of Ca-saturated illite particles to culture media did not affect the outcome of competition for sulfate. Thus, under sulfate limitation acetate accumulated. This phenomenon was briefly discussed in relation to the flow of electrons during anaerobic mineralization in marine and estuarine sulfate-limited sediments.  相似文献

The presence of calcium-or iron-saturated illite had a positive effect on the conversion of ethanol and acetate by non-starved cultures of Desulfobacter postgatei D.A41, but had no effect on non-starved cultures of Desulfobulbus propionicus Lindhorst and Sesulfovibrio baculatus H.L21. Starvation of these cultures at room temperature induced adhesion of cells of D. baculatus H.L21 to the surface of the clay particles. No adhesion of cells of D. propionicus Lindhorst and D. postgatei D.A41 was ever observed. However, for the three strains studied, the presence of clay particles had a positive effect on conservation of the oxidative capacity of the cultures during starvation.  相似文献

Dried vegetative filaments ofSpirogyra sp.,Vaucheria geminata andNostoc calcicola died within 1/2, 1 and 4 h, respectively; those ofHormidium fluitans, Oscillatoria agardhii andLyngbya martensiana retained under similar storage conditions viability for 3, 5 and 10 d, respectively. The viability of dried vegetative filaments ofL. martensiana, O. agardhii andH. fluitans decreased on storage at 20°C in the dark.L. martensiana andO. agardhii tolerated 0.8 mol/L NaCl. The resistance to desiccation inL. martensiana andO. agardhii exhibited similar dependence as that to frost, to heat and UV light.O. agardhii filaments became slightly broader and their cells developed large number of gas vacuoles when grown in 0.8 mol/L NaCl-containing medium. The water stress imposed on growing algae either on high-agar solid media or in NaCl-containing liquid media reduced hormogonium formation inL. martensiana andO. agardhii, heterocyst and akinete formation inN. calcicola and fragmentation inH. fluitans; it did not induce conjugation inSpirogyra sp. and formation of reproductive organs inV. geminata. In all studied algae the stress reduced at various levels the survival of vegetative parts. Generally, algal body form and composition rather than habitats seem to decide primarily the level of resistance against various stress conditions.  相似文献

Cultures ofStreptomyces tendae andPenicillium expansum grown on Actinomyces and Czapek's media, respectively, were exposed to 5 mg L–1 of manganese, magnesium, iron, cobalt, nickel, copper and zinc, supplied as sulfate salts. Only copper markedly increased geosmin (1, 10-dimethyl-9-decalol), biomass, and spore production. inductively coupled plasma-atomic emission spectrometric analysis ofS. tendae andP. expansum cells did not indicate an accumulation of copper. Both 1 and 5 mg L–1 copper, as copper sulfate, increased total geosmin production in cultures ofS. tendae on several media, but decreased production on others, suggesting that substrate composition affects responses to copper.  相似文献

Summary Hydrogenase and nitrogenase activities of sulfate-reducing bacteria allow their adaptation to different nutritional habits even under adverse conditions. These exceptional capabilities of adaptation are important factors in the understanding of their predominant role in problems related to anaerobic metal corrosion. Although the D2–H+ exchange reaction indicated thatDesulfovibrio desulfuricans strain Berre-Sol andDesulfovibrio gigas hydrogenases were reversible, the predominant activity in vivo was hydrogen uptake. Hydrogen production was restricted to some particular conditions such as sulfate or nitrogen starvation. Under diazotrophic conditions, a transient hydrogen evolution was followed by uptake when dinitrogen was effectively fixed. In contrast, hydrogen evolution proceeded when acetylene was substituted as the nitrogenase substrate. Hydrogen can thus serve as an electron donor in sulfate reduction and nitrogen metabolism.  相似文献

Two of nine sulfate reducing bacteria tested,Desulfobulbus propionicus andDesulfovibrio desulfuricans (strain Essex 6), were able to grow with nitrate as terminal electron acceptor, which was reduced to ammonia. Desulfovibrio desulfuricans was grown in chemostat culture with hydrogen plus limiting concentrations of nitrate, nitrite or sulfate as sole energy source. Growth yields up to 13.1, 8.8 or 9.7 g cell dry mass were obtained per mol nitrate, nitrite or sulfate reduced, respectively. The apparent half saturation constants (K s) were below the detection limits of 200, 3 or 100 mol/l for nitrate, nitrite of sulfate, respectively. The maximum growth rates {ie63-1} raised from 0.124 h-1 with sulfate and 0.150 h-1 with nitrate to 0.193 h-1 with nitrite as electron acceptor. Regardless of the electron acceptor in the culture medium, cell extracts exhibited absorption maxima corresponding to cytochromec and desulfoviridin. Nitrate reductase was found to be inducible by nitrate or nitrite, whereas nitrite reductase was synthesized constitutively. The activities of nitrate and nitrite reductases with hydrogen as electron donor were 0.2 and 0.3 mol/min·mg protein, respectively. If limiting amounts of hydrogen were added to culture bottles with nitrate as electron acceptor, part of the nitrate was only reduced to the level of nitrite. In media containing nitrate plus sulfate or nitrite plus sulfate, sulfate reduction was suppressed.The results demonstrate that the ammonification of nitrate or nitrite can function as sole energy conserving process in some sulfate-reducing bacteria.  相似文献

Veillonella alcalescens contained a membrane-bound lactate oxidase system. Studies on the effect of inhibitors on lactate oxidase showed the participation of non-heme iron, quinone and cytochromesb andd. Superoxide anion radicals ( ) and H2O2 were shown to be formed at lactate oxidation and presumably arose from cyanide- and azide-resistant side chains of the respiratory system. The H+/O ratio withL-lactate as a hydrogen donor was 2.3. When an anaerobic culture growing on lactate was shifted to a high dissolved oxygen tension (d.o.t.=15 kPa) rapid inhibition of growth and lactate conversion occurred. This could be correlated with a rapid inactivation of lactate dehydrogenase. The effects of high d.o.t.'s on lactate dehydrogenase, lactate conversion and growth were reversible. After a shift to low d.o.t.'s (<2.5 kPa) growth ofV. alcalescens continued for one or two doublings whereafter lysis did occur. Acetate and pyruvate were the main fermentation products. P/O ratio's were calculated from molar growth yields and fermentation balances. A P/O value of 0.66 was found after a shift to a very low oxygen supply at which the d.o.t. presumably was zero. Shifts to higher d. o. t.'s gave much lower growth yields. Presumably, under these conditions uncoupling between growth and energy production occurred. Obligate aerobes and anaerobes coexist closely in natural ecosystems. One species representative of each class (Desulfovibrio desulfuricans andParacoccus denitrificans) were selected for investigation in different laboratory growth systems. When incubated together, the aerobe protected the anaerobe against oxygen poisoning allowing the latter to grow and to reduce sulfate. When gas phase oxygen tension was systematically altered in stirred batch cultures, both species grew over a broad range of oxygen concentrations. Similar experiments in a chemostat indicated that conditions were far more exacting. No steady state was possible; however, at one critical oxygen tension the two species entered a stable oscillating cycle that lasted for 12 days. When the two organisms were grown in a gradostat in opposing gradients of sulfate plus oxygen and lactate, they coexisted but at different positions in the system. The actual position of the sulfate reducer was determined by the sulfate concentration in the reservoir.  相似文献

Summary The sequences of two rubredoxins isolated from the sulfate reducing bacteria:Desulfovibrio vulgaris andDesulfovibrio gigas have been elucidated. They have similar sequences but many more differences occur than would be expected from two bacteria of the same genus. Of the 52 sites, only 37 are occupied by identical residues. The primary structures are compared with those of the anaerobic bacteria rubredoxins ofClostridium pasteurianum, Micrococcus aerogenes, Pseudomonas oleovorans andPeptostreptococcus elsdenii: only 12 identities are found, mostly in the two clusters that contain two iron-bound cysteines each. A phylogenetic tree based on the primary structures is presented and possible relations with plant and bacterial ferredoxins are discussed. Summary Hydrogenases are among the main enzymes involved in bacterial anaerobic corrosion of metals. The study of their mode of action is important for a full comprehension of this phenomenon. The three types ofDesulfovibrio hydrogenases [(Fe), (NiFe), (NiFeSe)] present different patterns in the pH dependence of their activity. The periplasmic enzyme fromDesulfovibrio salexigens and the cytoplasmic enzyme fromDesulfovibrio baculatus both have pH optima at 7.5 for H2 uptake and 4.0 for H2 evolution and H+–D2 exchange reaction (measured by membrane-inlet mass-spectrometry). The H2 to HD ratio at pH above 5.0 is higher than 1.0. The periplasmic hydrogenase fromD. gigas presents the same pH optimum (8.0) for the H+–D2 exchange as for H2 consumption. In contrast, the enzyme fromD. vulgaris has the highest activity in H2 production and in the exchange at pH 5.0. Both hydrogenases have a H2-to-HD ratio below 1.0.  相似文献

Apparent half-saturation constants (K m) and maximum uptake rates (V max) for sulfate were determined in four species ofDesulfovibrio of freshwater and marine origin using a35S-sulfate tracer technique. The lowerstK m (5 M) was found in the freshwater speciesDesulfovibrio vulgaris (Marburg) and the highestK m (77 M) in the marine speciesDesulfovibrio salexigens. Maximum specific rates of sulfate uptake (i.e.,V max) were proportional to the growth rates observed in batch cultures. The halophilicDesulfovibrio salexigens did not change itsK m andV max between 1 and 6,000 M SO 4 2- , and apparently did not induce a low-affinity uptake system at high sulfate concentrations. The low half-saturation constants measured for sulfate uptake explain why high rates of bacterial sulfate reduction occur in surface sediments of freshwater lakes, and why sulfate reduction can be a quantitatively important process in anaerobic carbon mineralization in low-sulfate environments. Experiments with mixed cultures ofEscherichia coli, Desulfovibrio vulgaris, andChromatium vinosum were conducted using a synthetic medium with glucose as the substrate. The bacterial number and the changes in the chemical factors were determined during the development of the mixed culture. (i) Fermentation of glucose byE. coli produces organic acids (formic, acetic, lactic, and succinic) and alcohol. The growth-yield constant K (cell dry weight per weight solid substrate) does not exceed 0.05. (ii) In the mixed culture ofE. coli andD. vulgaris, the reduction of sulfate is accompanied by a total consumption of formate, lactate, and alcohol and an increase in the sulfide and acetate content. (iii) When the three physiologically different species are allowed to grow simultaneously, there is no accumulation of catabolites in the medium and the growth yield constant increases to 0.46. Maximum phototrophic production requires the presence of bothE. coli andD. vulgaris. A low substrate concentration and the simultaneous growth of the three organisms are other factors that contribute towards a high output. The biochemical parameters of the medium are influenced to a large extent by the glucose level. The results suggest that the behavior of the strains is different in pure and mixed cultures.  相似文献

The individual influence of sulfate reducing and brown phototrophic sulfur bacteria from Lake Kinneret on the metalimnetic redox conditions was investigated by simulating the hydrochemical and microbiological conditions in a specially designed chemostat. The results show a strong correlation between measured redox values and the prevailing hydrogen sulfide concentration leading to a linear relationship. Changes in this relationship allowed differentiation between the on-going microbial processes, sulfate reduction and sulfide oxidation. The comparison ofin vitro andin situ redox values shows that the results of the simulation experiments agree with the data previously measured in the metalimnion of Lake Kinneret.  相似文献

The effect of iron supply on the growth and nodulation ofLupinus angustifolius L. (Gungurru),Lupinus luteus L. (R-1171) andLupinus pilosus Murr. (P20957) was studied in acid solutions. Plants of the three species were grown together in the same solution and inoculated withBradyrhizobium (Lupinus) WU 425. Plants were then grown with or without applied NH4NO3. The lupin species differed greatly in their sensitivity to low iron concentrations in solution withL. pilosus being most tolerant andL. luteus most sensitive.L. pilosus had the highest iron concentration in tissues and had a higher ratio of iron concentration in the youngest fully expanded leaf blades (YEB) to that in roots than the other two species.L. luteus had higher iron concentrations in roots but lower iron concentration in YEB and shoots than didL. angustifolius. The requirements of internal iron for the maximal chlorophyll synthesis in YEB were 65 μg g-1 forL. angustifolius andL. luteus, and 52 μg g-1 forL. pilosus. In contrast to effects on growth, the three species had similar external iron requirements for nodule formation in roots and for maximal nitrogen concentrations in shoots. The results indicate that iron tolerant lupin species require lower internal and external iron supply and have a greater ability than sensitive species to translocate iron from roots to shoots.  相似文献

Hybridization of highly repeated DNA sequences ofEulemur fulvus mayottensis, Lemur catta, andVarecia has been performed on blots of different species of Lemuridae (L. catta, Hapalemur griseus, Varecia variegata variegata, V. v. rubra, E. macaco macaco, E. coronatus, E. mongoz, andE. rubriventer). The probe ofE. fulvus only hybridized with the differentEulemur species, whereas that ofVarecia hybridized with the two subspecies ofVarecia and that ofL. catta with bothL. catta andHapalemur. These results were used to confirm the classification ofVarecia in a separate genus and to review the separation of theL. catta/Hapalemur group from the other species ofEulemur. Comparison of the migration patterns from DNA fragments of these different species has been used to propose a cladogram of the differentEulemur species.  相似文献

The leaf-mining fliesHydrellia sp. andH. pakistanae Deonier were observed attacking the aquatic weedHydrilla verticillata (L.f.) L.C. (Hydrocharitaceae) in south India. Their life histories were very similar. The duration of the immature stages ofH. pakistanae was 25 to 29 days. The number of leaves ofH. verticillata mined by a single larva varied from 8 to 12. In the laboratory, adults feeding on 50% aqueous solution of honey lived for 6 to 21 days and laid an average of 26 eggs. Several overlapping generations occurred in the field except during the period of drought. BothHydrellia sp. andH. pakistanae were heavily parasitized by the braconidsAdemon sp. nr.decrescens Nees andChaenusa sp.   相似文献

Gibbons are small and arboreal apes restricted to southeastern tropical and southern subtropical regions in Asia. They are distributed from Assam in the northwest, eastward to south China and Vietnam, south through the Malay Peninsula to Sumatra, Borneo, Java, and the Mentawai Islands. Twenty-two genetically distinct populations of gibbons are generally recognized. Gibbons were once widely distributed in China; however, they are now primarily restricted to southern and southwestern Yunnan and Hainan Island. Their widest distribution and greatest species diversity is in Yunnan Province. Gibbons are rare in China and are now listed as a first class endangered species. Four gibbon species inhabit Yunnan Province in southern China. They areH. hoolock, H. lar, H. leucogenys, andH. concolor. There are three subspecies ofH. leucogenys: H. l. leucogenys, H. l. siki, andH. l. gabriellae. OnlyH. l. leucogenys inhabits China. Four subspecies ofH. concolor inhabit China. These areH. c. concolor, H. c. jingdongensis, H. c. furvogaster, andH. c. hainanus. The first three subspecies inhabit Yunnan andH. c. hainanus is found only on Hainan Island.H. lar yunnanensis is the subspecies ofH. lar in southern China.H. hoolock leucogenys is the subspecies ofH. hoolock in southern China.H. concolor jingdongensis, H. concolor furvogaster, andH. lar yunnanensis are all newly proposed subspecies byMa andWang (1986). The data on southern China gibbons presented here is based on an analysis of the skeletons and skins of 49 specimens held at the Kunming Institute of Zoology, Kunming, Yunnan.  相似文献

The effect of NaCl concentration gradients on the growth of a marine species,Vibrio pelagius, and two freshwater and estuarine species,Vibrio cholerae andAeromonas hydrophila, was determined in chemostat cultures containing a mineral medium withd-glucose as the sole source of carbon and energy. Complementary cell density profiles were obtained with the marine and the freshwater-estuarine species at the extreme ends of the NaCl concentration gradients. At the lower NaCl concentrations,V. cholerae andA. hydrophila attained maximal cell densities, while no growth or submaximal cell yields were characteristic ofV. pelagius; the reverse was true at the higher NaCl concentrations. At the intermediate concentrations (100–400 mM) all three organisms were able to grow with cell densities ranging from 80%–100% of the maximum. Competition experiments betweenV. cholerae andV. pelagius within this region (100, 150, and 300 mM) indicated that the lower NaCl concentrations favoredV. cholerae while the higher concentrations favoredV. pelagius. The results suggest that the responses ofV. pelagius, V. cholerae, andA. hydrophila to different concentrations of NaCl are important in determining the environmental distribution of these species.  相似文献
