Whilst reliable estimates of chiasma frequencies can usually not be obtained, the probability (b) of a chromosome arm to be bound by at least one chiasma can often be determined. In the absence of interference this probability equals (1–e –2), where 2 is the average chiasma frequency of the chromosome arm and the average crossover frequency or map length. In the presence of interference is shown to retain its genetic meaning as an additive metric that may describe the chromosome arm or other distinctive chromosome segment in terms of genetic recombination. It is a form of potential map length, comparable to, but numerically different from the regular map length. It is termed provisionally crossing-over potential.A chromosome with armsm andn with crossing-over potentials and will form ring bivalents with a frequency (1–e –2).(1–e –2); open bivalents with a frequency (1–e –2).e –2+(1–e –2).e –2; univalent pairs with a frequencye –2.e –2. Estimates of these frequencies yield equations from which and may be solved. In rye (Secale cereale) their ratio (q) is approximately two and differs from the mitotic arm length ratio of 1.4, indicating localization of chiasmata in the long arms.Graphs are given to show how, with constantq, the relation between the probabilitiesb m andb n of the two arms being bound changes with changing averageb.Data are presented on chiasma frequencies in M I, and compared with the frequencies expected in the absence of interference to give an impression of the degree of interference. The effect of lead on the filtration rate of the zebra musselDreissena polymorpha was investigated, together with the accumulation of Pb in the soft tissues of the mussels. The NOEC-filtration was 116 g.l–1 (0,56 mol.l–1) and the EC50-filtration was 370 g.l–1 (1.79 mol.l–1). The NOEC-accumulation was the concentration found in the control water (1.4g.l–1). These experiments show that the EC50-filtration for Pb is similar to that for Cd, higher than that for Cu and lower than that for Zn. The water quality criteria for lead allow 25 g Pb.l–1 in surface water. This will not cause short-term effects. Long-term effects may, however, occur, since an accumulation of Pb as low as 16 g.l–1 was recorded in this study.  相似文献

The effects of plant growth regulators, light intensity, and end-of-day (EOD) light quality treatments on node and microtuber induction (% of cultures with microtubers) and development (fresh weight of microtubers) in yam (Dioscorea alata L. cv. Oriental) cultures were investigated. Nodal segments were excised from plantlets cultured on tuberization medium containing growth regulators and exposed to various light treatments. Absciscic acid (1 M) stimulated and cytokinins (2.5 M) inhibited microtuber development from yam nodal segments cultured on Mantell's and Hugo's full-strength tuberization medium under 8-h photoperiods. EOD far-red (FR) light inhibited microtuber induction and development and enhanced node formation. EOD FR light effects were nullified by immediately following the FR treatment with red light. This suggested the involvement of phytochrome in these processes. The lowest light intensity evaluated (12 mol m–2 s–1) inhibited microtuber, root and shoot production as compared to light intensities of 42, 72 and 102 mol m–2 s–1. Node explants collected from flowering plants of Vanda spathulata, an endemic and exquisite orchid of Peninsular India and Sri Lanka, were cultured in Mitra medium with combinations of 4.4–88.8 m 6-benzyl adenine (BA) and 0.0–114.2 m indole-3-acetic acid (IAA). Combinations of 44.4 m BA with 17.1 or 28.5 m IAA and 66.6 mM BA with 28.5 or 40.0 m IAA induced maximum formation of 12.6 and 12.1 shoots / node, respectively, in a 6-month period. Subcultured nodal explants produced maximum of 6.1 shoots at combinations of 22.2–44.4 m 21 BA and 5.7–28.5 m IAA. Rooting of shoots occurred in medium containing 75 g l–1 banana pulp and 5.7 m IAA within 3–9 weeks. Plantlets of 2–5 cm length possessing two to five roots established easily in community pots at 80–90% rates without hardening. Community potted plants introduced into forest segments at Ponmudi and Palode in Southern Western Ghats of India established at a rate of 50–70%.  相似文献

The influence of copper (0–32 M) and iron (0–108 M) on growth and astaxanthin production by Phaffia rhodozyma was studied. Copper below 3.2 M increased the astaxanthin content of the cells (from 220 to 287 g g–1) but at the expense of a slightly decreased growth (from 11.3 to 10.2 mg ml–1). In contrast, iron below 1 M decreased both the growth and astaxanthin content of the cells. Using copper limitation instead of toxic respiratory inhibitors to improve astaxanthin production has obvious advantages from the product quality, environmental and process operation points of view.  相似文献

Denitrification processes were measured by the acetylene-blockage technique under changing flood conditions along the aquatic/terrestrial transition zone on the Amazon floodplain at Lago Camaleão, near Manaus, Brazil. In flooded sediments, denitrification was recorded after the amendment with NO 3 (100 mol liter–1) throughout the whole study period from August 1992 to February 1993. It ranged from 192.3 to 640.7 mol N m–2 h–1 in the 0- to 5-cm sediment layer. Without substrate amendment, denitrification was detected only during low water in November and December 1992, when it occurred at a rate of up to 12.2 mol N m–2 h–1 Higher rates of denitrification at an average rate of 73.3 mol N m–2 h–1 were measured in sediments from the shallow lake basin that were exposed to air at low water. N2O evolution was never detected in flooded sediments, but in exposed sediments, it was detected at an average rate of 28.3 mol N m–2 h–1 during the low-water period. The results indicate that under natural conditions there is denitrification and hence a loss in nitrogen from the Amazon floodplain to the atmosphere. Rates of denitrification in flooded sediments were one to two orders of magnitude smaller than in temperate regions. However, the nitrogen removal of exposed sediments exceeded that of undisturbed wetland soils of temperate regions, indicating a considerable impact of the flood pulse on the gaseous turnover of nitrogen in the Amazon floodplain.  相似文献

Summary Zoogloea ramigera 115 was immobilized into beads of calcium-alginate and placed into batch air-bubbled column reactors. In the absence of any added nutrients the immobilized bacterium adsorbed Cd from solutions containing levels of 2 and 20 g ml–1 per day, over a period of 21 and 20 days, respectively. Adsorption of Cd from solutions containing 20 g ml–1 Cd was better than 90% for 16 days. Beads treated with Cd at 2 g ml–1 never adsorbed less than 95% of the metal. Alginate adsorbed Cd as well, but inclusion of cells changed the effectiveness of adsorption. Of a 250 g ml–1 Cd solution, alginate adsorbed 70.4% Cd in 60 min whereas alginate plus cells adsorbed 90.5% in the same time span. Temperature had no effect on adsorption by immobilized cells at levels of 2 and 10 g ml–1 Cd. However at higher concentrations, binding was enhanced as temperature increased.Z. ramigera beads were stable during all treatments and for prolonged periods of time (21 days).  相似文献

The influence of Zn2+ (6.0 × 10–3 –18.0 × 10–3 M) and Cu2+ (4 × 10–4 –1.2 × 10–4 M) in the basal medium on mycelial growth (dry weight), activities of lignin peroxidase (Lip), manganese peroxidase (Mnp), solubilization, and mineralization (14CO2 evolution) of lignin during a period of 3 weeks was studied in Phanerochaete chrysosporium strain MTCC-787. Highest mycelial growth was obtained at 0.6 M Zn2+ and 0.4 M Cu2+ levels. Enzyme activities were found to increase up to the highest levels of both the trace elements. However, Zn2+ had a relatively more stimulatory effect on Lip production and the reverse was true in case of Cu2+. [14C]Lignin solubilization was also promoted by higher levels of both trace elements. Mineralization of [14C]lignin was optimal at 6.0 M Zn2+ and 1.2 M Cu2+. The stimulatory effect of Zn2+ on Lip production was correlated with higher rates of [14C]lignin mineralization.  相似文献

Strawberry (Fragaria ananassaDuch. cv. Fengxiang) plantlets were cultured under two in vitroenvironments for rooting, and then acclimatized under two ex vitroirradiance conditions. At the end of rooting stage plant height, fresh weight and specific leaf area of T1-plants grown under high sucrose concentration (3 sucrose), low photosynthetic photon flux density (30 mol m–2 s–1) and normal CO2 concentration (350–400 l l–1) were significantly higher than those of T2-plantlets grown under low sucrose concentration (0.5), high photosynthetic photon flux density (90 mol m–2 s–1) and elevated CO2 concentration (700–800 l l–1). But T2-plantlets had higher net photosynthetic rate (Pn), effective photochemical quantum yield of PSII (PSII), effective photosynthetic electron transport rate (ETR), photochemical quenching (qP) and ratio of chlorophyll fluorescence yield decrease (Rfd). After transfer, higher irradiance obviously promoted the growth of plantlets and was beneficial for the development of photosynthetic functions during acclimatization. T2-plantlets had higher fresh weight, leaf area, PSII and ETR under higher ex vitroirradiance condition.  相似文献

The effects of light intensity, oxygen concentration, and pH on the rates of photosynthesis and net excretion by metalimnetic phytoplankton populations of Little Crooked Lake, Indiana, were studied. Photosynthetic rates increased from 1.42 to 3.14 mg C·mg–1 chlorophylla·hour–1 within a range of light intensities from 65 to 150E·m–2·sec–1, whereas net excretion remained constant at 0.05 mg C·mg–1 chlorophylla·hour–1. Bacteria assimilated approximately 50% of the carbon released by the phytoplankton under these conditions. Excreted carbon (organic compounds either assimilated by bacteria or dissolved in the lake water) was produced by phytoplankton at rates of 0.02–0.15 mg C·mg–1 chlorophylla·hour–1. These rates were 6%–13% of the photosynthetic rates of the phytoplankton. Both total excretion of carbon and bacterial assimilation of excreted carbon increased at high light intensities whereas net excretion remained fairly constant. Elevated oxygen concentrations in samples incubated at 150E· m–2·sec–1 decreased rates of both photosynthesis and net excretion. The photosynthetic rate increased from 3.0 to 5.0 mg C·mg–1 chlorophylla· hour–1 as the pH was raised from 7.5 to 8.8. Net excretion within this range decreased slightly. Calculation of total primary production using a numerical model showed that whereas 225.8 g C·m–2 was photosynthetically fixed between 12 May and 24 August 1982, a maximum of about 9.3 g C·m–2 was released extracellularly.  相似文献

The aims of this study were to describe the distribution of magnesium (Mg) and its retranslocation within wheat, in order to develop diagnostic procedures for Mg deficiency. Plants were grown in solution culture with both constant supply (0, 5, 10, 20, 40, 80, 160 MMg) and discontinued supply (40 M and 160 M decreased to nil).Magnesium was depleted from old leaves when Mg supply to the roots was halted. However, initial deficiency symptoms occurred on young leaves under constant but inadequate supply, contrasting with previous reports. Magnesium concentrations were also lower in young leaves compared to old leaves. Symptoms of yellowing and necrosis occurred if the leaf tissue contained <1194 gg–1, irrespective of leaf age. The minimum Mg concentration in whole shoots associated with maximum shoot weight was 932 gg–1; for the youngest emerged blade (YEB) it was 861 gg–1. Symptoms were apparent on the young leaf before a reduction in shoot weight was measurable. The concentration of Mg in the YEB and whole shoot were better related to solution Mg concentration than was the Mg concentration in the old leaf.  相似文献

Summary A method for the determination of exchangeable lithium using 0.5M NH4Cl is described. The range of exchangeable Li in the fifty Papua New Guinea (PNG) soils analyzed was 0.002 to 0.409 gg–1 in contrast to five Australian soils which ranged from 0.032 to 0.830 gg–1. The PNG soils were divided into hill and alluvial soils with average exchangeable Li contents of 0.062 and 0.263 gg–1 respectively. No significant correlation between total and exchangeable Li was found in either group of soils althoughr=0.67 for the comined data and was significant at the 5% level. From the analysis of three profiles exchangeable Li was found to be at least twice as high (0.27 gg–1) in surface soils as in subsurface samples (0.10 gg–1). The average value of the deeper subsoil samples was 0.18 ppm.R mode cluster analysis of the data for village garden soils collected on a sampling grid showed that exchangeable Li was more strongly assoicated with Ca and Mg than with pH, 0.05M EDTA soluble Zn, 0.5M NaHCO3 soluble P or exchangeable Na and K. Computer constructed isographs using the analyses of grid samples from a garden illustrated the association between Li, Ca and Mg and the inverse association with Na.The correlation coefficient between Ca and Li in the ash of three food plants (Gnetum gnemon, Hibiscus abelmoschus andStenochlaena plustris) while not significant on an individual basis, was significant when the data was combined suggesting that the association between these elements in the soil may reflect an association in the ash returned to the soil when the garden was cleared. The correlation coefficient between soil exchangeable Li and Li in plant ash was positive, but not significant.Adsorption experiments over a five-day period demonstrated that Li was strongly adsorbed from solution. On average 63–75% of the adsorbed Li was fixed in a form which was not exchangeable with 0.5M NH4Cl or soluble in 0.05M EDTA.  相似文献

Thymidine and leucine incorporation into macromolecules of soil bacteria extracted by homogenization-centrifugation were measured after size-fractionation of the bacterial suspension through different sized filters (1.0, 0.8, 0.6, 0.4 m). The specific thymidine incorporation rate was highest for the unfiltered and 1.0 m filtered suspensions (approximately 10 × 10–21 mol thymidine bacteria–1 h–1), but decreased to 1.39 × 10–21 mol bacteria–1 h–1 for bacteria passing the 0.4 m filter. The proportion of culturable bacteria (percent colony forming units/acridine orange direct counts) also decreased with bacterial cell size from 5.0% for the unfiltered bacterial suspension to 0.8% in the 0.4 µm filtrate. A strong linear correlation (r 2 = 0.995) was found between the specific thymidine incorporation rate and the proportion of culturable bacteria. Leucine incorporation gave similar results to the thymidine incorporation. No effects of cell size on the degree of isotope dilution or unspecific labeling of other macromolecules were found either for the thymidine or the leucine incorporation technique. These data indicate that small bacteria, although more numerous than larger ones, not only constitute a smaller proportion of the soil bacterial biomass than larger bacteria, but also contribute to a lesser degree to carbon transformations in soil.  相似文献

In order to evaluate the ability of the freshwater bivalve Potomida littoralis for use as a biodetector of aquatic pollution, the short-term effects of some pollutants (metals, antifouling product and effluents) on pumping activity of this mussel were studied qualitatively (pumping behaviour) and quantitatively (filtration rate). The method of the flow sensing at the exhalent siphon showed that the normal activity of P. littoralis was characterized by a continuous pattern of pumping with occasional and brief interruptions. When exposed to toxic stress, the exhalent flow immediately shifted to an intermittent pattern with periods of pumping alternating with periods of pause that could last several minutes. The threshold concentrations of heavy metals/antifouling agent inducing this type of activity within 2 h were: 25 g l–1 for copper, 90 g l–1 for cadmium, 5 g l–1 for mercury, 3000 g l–1 for zinc and 700 g l–1 for the antifouling agent Mexel. Both the effluent samples of the urban wastewater of Marrakech city tested were found to be toxic.The filtration rate of control animals ranged from 0.91 to 1.72 l h–1 g–1 animal, while after 3 h exposure to Cu, Cd, Hg and the effluent E1 and E2, filtration rates were decreased by concentrations above the threshold levels set as toxic for the pumping activity.The sensitivity of P. littoralis to pollution and its usefulness as biodetector for rapid toxicity screening is discussed and a procedure for the detection of pollution in Moroccan freshwater is proposed.  相似文献

Nitrification rates were measured along a salinity gradient in the Rhône River estuary, using specific inhibitors (allylthiourea and chlorate) coupled with the measurement of change in nitrite concentration and inorganic carbon uptake by nitrifiers. Rates of ammonium and nitrite oxidation were similar up to 15 practical salinity units (from 1 to 2 mol N oxidized liter-1 day-1). For higher salinities, nitrite and ammonium oxidation rates were 0.14 and 0.23 mol N oxidized liter-1 day-1, respectively. Ammonium oxidizers assimilated 19–150 × 10–3 mol C liter-1 day-1, while nitrite oxidizers fixed 4.8–72.6 × 10–3 mol C liter-1 day-1. The amounts of nitrogen oxidized and C incorporated demonstrated a linear correlation (r 2 > 0.99). The ratio of N oxidized to C incorporated ranged between 14.3 to 12.3 for ammonium oxidizers, and between 31.6 and 29 for nitrite oxidizers, the lower values being measured in seawater. Rhizobium trifolii, R. leguminosarum, andR. hedysarum, grownex planta under anoxic conditions in a chemically defined medium, evolve N2O from NO3 , NO2 , and (NH4)2NO3. The amount of nitrous oxide formed after 96 hours is about 0.2M×mg–1 cells d.w. Large availability of organic matter enhances the production of N2O from nitrate by free-livingR. trifolii in peat/sand mixtures. Denitrification of the above species andR. meliloti was detected also in planta. Nitrous oxide production increases almost linearly from 10–45M×mg–1 nodules d.w. when nitrogen-fixing plants are exposed to increasing concentrations of nitrate (1–12M).  相似文献

Lower concentrations of CuSO4 (25–75 M) in the MS medium supplemented with 0.1 mg l–1 IAA+5.0 mg l–1 Kn+500 mg l–1 CH+10 mg l–1 Cyst hyd enhanced the growth of regenerants of Dioscorea bulbifera L. CuSO4 (75 M) induced an appreciable diosgenin yield in the regenerants compared to those obtained on media without Cu. The presence of Cu thus seems to stimulate diosgenin production. The regenerants also differentiated bulbils on lower concentrations of Cu. At CuSO4 (100 M), however, cultures showed poor growth as well as a low diosgenin yield. Increased proline and protein contents were recorded in cultures grown on Cu-enriched media.  相似文献

In order to maintain axenic seedstock cultures axenically of thecommercially important red seaweed, Porphyra yezoensis, aprocedure was developed for axenic isolation and culture of conchocelis andmonospores. For axenic isolation of the conchocelis, contaminated microalgaewere most effectively removed by filtering contaminated samples through a100-m mesh after sonication. Removal of bacteria and otheralgaewas accomplished using a mixture of 5 agents (0.02% chitosan, 100 gml–1 GeO2, 10 gml–1 ampicillin, 40 gml–1 kanamycin and 200 gml–1 streptomycin). Axenic single colonies wereisolatedfrom a semi-solid medium prepared from 1% transfer gel. After collectingmonospores from the 40–50% density layer on a percoll-gradient, removalofbacteria and fungi from the monospores was accomplished using a mixture of 5antibiotics (3.5 g ml–1 nystatin, 2 mgml–1 ampicillin, 400 gml–1 kanamycin, 50 gml–1 neomycin and 800 gml–1 streptomycin). Synopsis Foraging behaviour of lake charr, Salvelinus namaycush, exposed to 0.5 and 5.0 g l–1 cadmium concentrations for 106–112 d, was examined in a flow-through stream channel using fathead minnows, Pimephales promelas, as prey. Five modes of foraging behaviour were observed and quantified (ending at fixate-orient; ending at approach and/or chase; ending at failed attack; ending at capture and loss; successful consumption). Each mode represented an increasingly progressive end-point from initial sighting of the prey to final consumption. Compared to controls., 0.5 g l–1 Cd-exposed predators captured significantly fewer prey, while 5 g l–1 Cd-exposed predators made fewer attacks. Both Cd-exposed groups consumed fewer prey than controls.  相似文献

In Taxus cuspidata callus, vanadyl sulfate (10 mg l–1) induced a high (146 g g–1 dry wt) production of 10-deacetylbaccatin III in comparison to 7 g g–1 dry wt of the control. The content of paclitaxel in this species increased from 16 g g–1 to 74 g g–1 dry wt when 20 mg phenylalanine l–1 was used. In T. media, p-aminobenzoic acid induced the highest content of 10-deacetylbaccatin III (481 g g–1 dry wt) versus 181 g g–1 in the control. Paclitaxel increased from 89 to 139 g g–1 dry wt after adding chitosan (20 mg l–1) to the cultures.  相似文献
