选取萌发3~5d、长度3~5cm的中国菟丝子(Cuscuta Chinensis Lam)幼苗,将其分为上、中、下3部分并作为外植体进行离体培养与植株再生研究。结果表明,其上、中部片段更适宜愈伤组织诱导;诱导培养基以添加1mg L^-1 NAA和1mg L^-1 BA的MMS培养基效果最好,此培养基也可用于愈伤组织的继代培养,愈伤组织在上述培养基中已生长一年之久。分化培养基为添加1mg L^-1 BA的MMS培养基,平均每块愈伤组织可以产生2.8株植株。  相似文献

以常用的神经嗜铬细胞瘤PC12细胞株为实验模型,通过比较活性氧(ROS)作用细胞后的细胞活力、凋亡相关蛋白(p53、Bax)水平以及细胞中SOD、GSH、MDA的差异,发现菟丝子提取物不仅能提高ROS损伤的已分化PC12细胞活力,调节细胞中凋亡相关基因的表达,而且还能提高细胞中SOD和GSH的含量,降低MDA水平。由此表明,菟丝子提取物对ROS造成的PC12细胞损伤有一定的保护作用。  相似文献

本文以大强度耐力训练大鼠为模型,对菟丝子对运动训练大鼠睾酮含量、物质代谢及抗运动疲劳能力的影响进行了研究.试验中分别以1.16、2.32、6.96 g.kg-1.d-1的剂量给大鼠灌胃42d,并进行负重游泳实验、血清睾酮等生化指标测定.结果显示,菟丝子各剂量组力竭游泳时间长于运动对照组(T组)(P<0.01);血清睾酮高于T组(P<0.01),血清皮质酮低于T组(P<0.05);各组间血清睾酮与皮质酮比值变化与睾酮变化较为一致;肝糖原(P<0.05)、肌糖原(P<0.01)高于T组;血清尿素氮低于T组(P<0.05);血红蛋白高于T组(P<0.05).从而表明补充菟丝子可以减轻大鼠血睾酮、皮质酮受高强度运动量的影响,维持在正常生理水平;促进蛋白质合成,抑制氨基酸和蛋白质分解,提高血红蛋白含量和糖原的储备,增强抗疲劳能力,具有多靶点、多途径的显著特点.  相似文献

Abstract Metabolic changes in developing Cuscuta campestris Yunck seeds were studied with respect to TCA-soluble phosphate fractions, nucleic acids and the enzymes, viz, acid and alkaline phosphatase, neutral and alkaline FDPase, starch phosphorylase, α-amylase ribonuclease, deoxyribonuclease and phytase. RNA and DNA contents increased steadily, and then remained stationary during the last two stages. The activities of all the enzymes studied increased with development with a sharp fall in the final stage. Interestingly, α-amylase was present throughout the seed development. Phytic acid (16% of the total P) does not seem to function as phosphate reservoir, instead orthophosphate (13% of the total P) and nucleic acids (30% of the total P) may serve this function.  相似文献

Chronic hypoxia protects the heart against injury caused by acute oxygen deprivation, but its salutary mechanism is poorly understood. The aim was to find out whether cardiomyocytes isolated from chronically hypoxic hearts retain the improved resistance to injury and whether the mitochondrial large-conductance Ca2+-activated K+ (BKCa) channels contribute to the protective effect. Adult male rats were adapted to continuous normobaric hypoxia (inspired O2 fraction 0.10) for 3 wk or kept at room air (normoxic controls). Myocytes, isolated separately from the left ventricle (LVM), septum (SEPM), and right ventricle, were exposed to 25-min metabolic inhibition with sodium cyanide, followed by 30-min reenergization (MI/R). Some LVM were treated with either 30 μM NS-1619 (BKCa opener), or 2 μM paxilline (BKCa blocker), starting 25 min before metabolic inhibition. Cell injury was detected by Trypan blue exclusion and lactate dehydrogenase (LDH) release. Chronic hypoxia doubled the number of rod-shaped LVM and SEPM surviving the MI/R insult and reduced LDH release. While NS-1619 protected cells from normoxic rats, it had no additive salutary effect in the hypoxic group. Paxilline attenuated the improved resistance of cells from hypoxic animals without affecting normoxic controls; it also abolished the protective effect of NS-1619 on LDH release in the normoxic group. While chronic hypoxia did not affect protein abundance of the BKCa channel regulatory β1-subunit, it markedly decreased its glycosylation level. It is concluded that ventricular myocytes isolated from chronically hypoxic rats retain the improved resistance against injury caused by MI/R. Activation of the mitochondrial BKCa channel likely contributes to this protective effect.  相似文献

Adult female rats were fed chow only, or chow plus the polysaccharide Polycose presented as a solution (32% w/v), as a powder, or as a powder mixed into the chow diet. The rats fed the Polycose solution overate and gained three times as much weight as did the control rats in the 30-day test period. The rats fed the Polycose powder or mixed diet, on the other hand, did not differ from controls in food intake or weight gain. The solution-fed rats consumed more Polycose than did the powder-fed rats, but the two groups were equivalent in their chow intake. The absolute and percent body fat of the solution-fed rats exceeded that of control rats, as well as that of powder and mixed-diet animals. The percent body fat of the powder and mixed diet groups exceeded that of the control animals. The findings demonstrate that the form of diet presentation has a major impact on the rat's feeding and body weight responses to polysaccharide diets.  相似文献

By the hot water extraction method, galactomannan was extracted (4.5% yield of the seed mass) from the seeds of Chinese honey locust (Gleditsia sinensis Lam). It had a molecular weight of 1230 kDa, and its solutions had a high viscosity [η] of 1064 ml/g and optical activity [α]D of +21.4°. The polysaccharide consists of mannose and galactose residues in the molar ratio 2.69: 1. In the galactomannan macromolecule the backbone is formed by 1,4-β-D-mannopyranose residues, 37% of which are substituted by α-D-galactopyranose at C6. By 13C-NMR-spectroscopy, fragments of differently galactose-substituted mannobiose units were found to be in the galactomannan being studied: Man-Man, (Gal)Man-Man, and Man-Man(Gal) in the ratio of 0.23: 0.47: 0.30.  相似文献

The mucilaginous polysaccharide-complex found in the seeds O. adscendens contains an acidic polysaccharide which has been isolated. It is composed of D-galactose (~ 20%), D-galacturonic acid (~ 35%) and L-rhamnose (~ 39%). Methylation analysis using GC and GC/MS, Smith degradation, isolation of an acidic oligosaccharide from partial hydrolysates, and enzymic hydrolysis using β-D-galactosidase indicated the polysaccharide backbone to be → 4)-GalpA-(1 → 2)-L-rhap-(1 →. Nearly two-thirds of the rhamnopyranosyl units are 0-4 substituted by D-galactopyranosyl non-reducing end groups.  相似文献

A new triterpenoid, 2α, 3β, 20β, 23-tetrahydroxy-urs-12, 19 (29)-dien-28-oic acid, together with eight known triterpenoids, was isolated from the aerial parts of Salvia chinensis. Their structures were identified by spectroscopic analyses. Seeds of the tropical tree Moringa oleifera contain small storage proteins able to flocculate particles in suspension in water. The cDNA encoding one of these flocculent proteins, MO(2.1), was cloned and the recombinant protein was expressed in Escherichia coli. The flocculent activity of the purified recombinant MO(2.1)was assayed on clays and bacteria using light and confocal microscopy and GFP-overexpressing bacteria. We show that MO(2.1)is able to aggregate montmorillonite clay particles as well as gram-positive and gram-negative bacteria. We discuss the use of recombinant proteins to study flocculating properties and improve water purification processes.  相似文献

Background  

Restriction of intracellular diffusion of adenine nucleotides has been studied intensively on adult rat cardiomyocytes. However, their cause and role in vivo is still uncertain. Intracellular membrane structures have been suggested to play a role. We therefore chose to study cardiomyocytes from rainbow trout (Oncorhynchus mykiss), which are thinner and have fewer intracellular membrane structures than adult rat cardiomyocytes. Previous studies suggest that trout permeabilized cardiac fibers also have diffusion restrictions. However, results from fibers may be affected by incomplete separation of the cells. This is avoided when studying permeabilized, isolated cardiomyocytes. The aim of this study was to verify the existence of diffusion restrictions in trout cardiomyocytes by comparing ADP-kinetics of mitochondrial respiration in permeabilized fibers, permeabilized cardiomyocytes and isolated mitochondria from rainbow trout heart. Experiments were performed at 10, 15 and 20°C in the absence and presence of creatine.  相似文献

Evidence is growing of a relationship between atrial dilation and atrial fibrillation (AF), the most prevalent type of arrhythmia. Pulmonary veins, which are important ectopic foci for provoking AF, are of increasing interest in relation to the early development of AF. Here, using single cardiomyocytes isolated from rabbit pulmonary veins, we characterised the stretch-activated currents induced by swelling and axial mechanical stretching. Swelling induced both a stretch-activated nonselective cationic current (NSC) and a Cl(-) current. The swelling-induced Cl(-) current (I Cl,swell) was inhibited by DIDS, whereas the swelling-induced NSC (I NSC,swell) was inhibited by Gd3+. The cationic selectivity of the I NSC,swell was K+ >Cs+ >Na+ >Li+, whilst the PK/PNa, PCs/PNa, and PLi/PNa permeability ratios were 2.84, 1.86, and 0.85, respectively. Activation of the I NSC,swell was faster than that of the I Cl,swell. Given a high K+ concentration in the bath solution, the I NSC,swell showed limited amplitude (<-70 mV). Mechanical stretching induced an immediate Gd3+- and streptomycin-sensitive NSC (I NSC,stretch) that was permeable to Na+, K+, Cs+ and NMDG. Persistent stretching activated a DIDS-sensitive current (I Cl,stretch). The I NSC,stretch, but not the I NSC,swell, was completely blocked by 400 microM streptomycin; therefore, the two currents may not be associated with the same channel. In addition, the type of current induced may depend on the type of stretching. Thus, stretch-induced anionic and cationic currents are functionally present in the cardiomyocytes of the main pulmonary veins of rabbits, and they may have pathophysiological roles in the development of AF under stretched conditions.  相似文献

Block of sodium currents by allapinin (diterpene alkaloid with strong antiarrhythmic properties) was investigated in isolated, voltage clamped rat trigeminal neurons and cultured neonatal rat single ventricular myocytes. Allapinin produces a decrease in sodium current amplitude without any changes in voltage dependent properties. Possible differences between the mechanisms of antiarrhythmic effect of diterpene alkaloids and classic antiarrhythmic agents have been analysed.  相似文献

采用Langendorff灌流心脏和酶解分离的心肌细胞为实验模型,研究铁对心肌的损伤作用,以及过氧化氢对铁的心肌作用的影响及其可能机制.结果显示(1)羟基喹啉铁复合物(Fe-HQ)引起分离心肌细胞舒张期缩短,心肌细胞的收缩幅度和速度降低,离体灌流心脏左室发展压(LVDP)、±dp/dtmax、心率、冠脉流量呈现双相变化;冠脉流出液中乳酸脱氢酶(LDH)、肌酸激酶(CK)释放量和心肌丙二醛(MDA)增高.(2)H2O2可加重Fe-HQ对心脏的损伤,冠脉流出液中LDH、CK释放量和心肌MDA增高,而LVDP、±dp/dtmax和心率明显降低.(3)还原型谷胱甘肽可对抗Fe-HQ+H2O2对心肌的损伤作用,DMSO对Fe-HQ+H2O2致离体心脏损伤无明显作用.结果提示,心肌细胞内铁增加可引起心肌功能受损,H2O2可加重铁对心肌的损伤作用,其主要机制可能与@OH无关,而主要与含巯基的蛋白质受损有关.  相似文献
