On the basis of the synthesis of novel biodegradable amphiphilic MPEG-b-PCL-grafted chitooligosaccharide (COS-g-PCL-b-MPEG) copolymers, supramolecular hydrogels were fabricated rapidly via their inclusion complexation with α-cyclodextrin (α-CD) in aqueous solutions. The graft copolymers were characterized by 1H NMR spectroscopy, gel permeation chromatography (GPC), and fluorescence measurement, and the supramolecular structure of the resultant hydrogels was confirmed by X-ray diffraction measurements. Rheological studies of as-obtained hydrogels indicate that the physical properties could be modulated by controlling the concentration and the graft content of the graft copolymers as well as the molar feed ratio of the graft to α-CD. The in vitro release kinetics studies of bovine serum albumin (BSA) entrapped in the hydrogels show that the drug release profiles are dependent on the supramolecular hydrogel compositions.  相似文献

Graft copolymerization of rosin-(2-acryloyloxy)ethyl ester (RAEE) onto chitosan (Cts) was carried out under microwave irradiation using potassium persulfate as an initiator. The structures, morphology, and thermal properties of the Cts graft copolymer (Cts-g-PRAEE) were characterized by means of FT-IR, XRD, SEM, and TG. Also, Cts and Cts-g-PRAEE copolymer were used as carriers of fenoprofen calcium (FC), and their controlled release behavior in artificial intestinal juice were studied. The results show that the rate of release of fenoprofen calcium from the carrier of Cts-g-PRAEE copolymer becomes very slower than that of Cts in artificial intestinal juice.  相似文献

A novel biodegradable graft copolymer, dextran-g-poly(1,4-dioxan-2-one) (PODEX), was synthesized through the ring-opening polymerization (ROP) of 1,4-dioxan-2-one (PDO) onto a dextran backbone. Initially, dextran was silylated with 1,1,1,3,3,3-hexamethyldisilazane. The grafting from pathway was conducted with various (30–70) PDO/OH feed ratios to obtain PODEX copolymers with a various PPDO graft structures. Graft copolymers were characterized by FT-IR, 1H and 13C NMR, DSC, TGA, and WAXD. Molecular weights of the PODEX copolymers (MW: 94,700–117, 300 Da), glass transition temperature (−29 to −17 °C), melting temperature (82–100 °C), and crystallinity (32–40%) were increased with the content of PPDO. AFM observations revealed that polymeric micelles were spherical and uniform in morphology with around 30–58 nm diameter. Critical micelle concentration (CMC) of self-assembled system was significantly decreased from 3.2 to 1.09 mg/L with the increment of PPDO.  相似文献

A novel thermosensitive and hydrogel was designed and synthesized by graft copolymerization of N-isopropylacrylamide (NIPAAm) with biodegradable carboxymethylchitosan (CMCS). The influence of the content of CMCS grafted on the properties of the resulted hydrogels was examined. The morphology of the hydrogels was observed by scanning electron microscopy (SEM), their thermal property was characterized by differential scanning calorimetry (DSC), thermogravimetric analysis (TGA) and deswelling/swelling kinetics upon external temperature changes. In comparison with the conventional PNIPAAm hydrogels, the resulted hydrogels have improved thermosensitive properties, including enlarged water content at room temperature and faster deswelling/swelling rate upon heating. The strategy described here presents a potential alternative to the traditional synthesis techniques for thermosensitive hydrogels.  相似文献

A new type of chitosan derivative, dehydroabietic acid-modified chitosan (DAMC), was synthesized by the acylation reaction of chitosan with dehydroabietic acid chloride (DHAC) under microwave irradiation. The resulting product (DAMC) was characterized by FT-IR, UV, 1H NMR, X-ray diffraction (XRD), scanning electron microscopy (SEM), thermal gravimetric analysis (TGA), and elemental analysis. The degree of substitution (DS) of DAMC was 16.5%. And chitosan and DAMC were used as carriers of fenoprofen calcium (FC), and their controlled release behavior in artificial intestinal juice was studied. The results showed that the controlled release of FC from the carrier of DAMC is better than that from original chitosan.  相似文献

A new scheme was proposed for synthesizing poly(ethylene glycol)-g-chitosan (PEG-g-CS), where methoxy poly(ethylene glycol) iodide (MPEG-I) (Mn 2000) was used for N-substitution of triphenylmethyl chitosan (TPM-CS) in organic medium. The graft copolymers were obtained by subsequent removal of protecting groups with dichloroacetic acid. By varying PEG-I/TPM-CS feed ratio, the grafting levels (GL) of PEG can be adjusted. The chitosan derivatives were characterized by FTIR, 1H NMR, 13C NMR and DSC. All the copolymers were soluble in water over wide pH range. Furthermore, organosolubility of the hybrids in DMF and DMSO was also achieved when the DS value more than 24%. The lysozyme degradation rate of the copolymers in aqueous neutral medium decreased with the increase of GL value.  相似文献

In this paper, 20 kinds of different 2-(α-arylamino phosphonate)-chitosan (2-α-AAPCS) were prepared by different Schiff bases of chitosan (CS) reacted with di-alkyl phosphite in benzene solution. The structures of the derivatives (2-α-AAPCS) were characterized by FT-IR spectroscopy and elemental analysis. In addition, the antifungal activities of the derivatives against four kinds of fungi were evaluated in the experiment. The results indicated that all the prepared 2-α-AAPCS had a significant inhibiting effect on the investigated fungi when the derivatives concentration ranged from 50 to 500 μg mL−1. Furthermore, the antifungal activities of the derivatives increased with increasing the molecular weight and concentration. And the antifungal activities of the derivatives were affected by their dimensional effect and charge density. Besides, the rule and mechanism of the antifungal activities of them were discussed in this paper.  相似文献

A novel water-soluble chitosan derivative [N-(2-carboxybenzyl)chitosan, CBCS] was synthesized. The chemical structure of CBCS was characterized by FTIR, (1)H NMR and UV spectroscopies. The degree of substitution (DS) of N-2-carboxybenzyl was determined by colloid titration. In different pH buffer solutions, the swelling characteristics of hydrogels based on CBCS (CBCSG) prepared by crosslinking with glutaraldehyde have been studied. Results showed that the swelling ratio (SR) of CBCSG decreased with an increase of the amount of glutaraldehyde, and that CBCSG swelled more significantly in alkaline solution than in acidic medium, showing the lowest SR at pH5.0. The SR of CBCSG increased with the raising of the DS of the N-2-carboxybenzyl group in alkaline solution, but no significant change was observed in an acidic environment. CBCSG showed swelling reversibility when alternately soaked in pH1.0 and 7.4 buffer solutions. Release profiles of fluorouracil (5-FU), a poorly water-soluble drug, from CBCSG were studied under both simulated gastric and intestinal pHconditions. The release was much quicker in pH7.4 buffer than in pH1.0 solution. Results indicated that CBCS could be a potential pH-sensitive carrier for colon-specific drug delivery system.  相似文献

An unreported graft copolymer of N,N-dimethylacrylamide (DMA) with chitosan has been synthesized under nitrogen atmosphere using peroxymonosulphate/mandelic acid redox pair. The effect of reaction conditions on grafting parameters i.e. grafting ratio, efficiency, conversion, add on and homopolymer has been studied. Experimental results show that maximum grafting has been obtained at 1.0 g dm−3 concentration of chitosan, 30 × 10−2 mol dm−3 concentration of N,N-dimethylacrylamide and 7.0 × 10−3 mol dm−3 concentration of hydrogen ion. It has also been observed that grafting ratio, add on, conversion and efficiency increase upto 3.2 × 10−3 mol dm−3 of mandelic acid, 12.0 × 10−3 mol dm−3 of potassium peroxymonosulphate, 150 min of time and 40 °C of temperature. Grafted polymer has been characterized by FTIR spectroscopy and thermogravimetric analysis. Water swelling capacity of chitosan-g-N,N-dimethylacrylamide has been determined. It has been observed that the graft copolymer is thermally more stable than parent backbone.  相似文献

Poly(dimer acid-brassylic acid) [P(DA-BA)] copolymers and poly(dimer acid-pentadecandioic acid) [P(DA-PA)] copolymers were prepared by melt polycondensation of the corresponding mixed anhydride prepolymers. The copolymers were characterized by Fourier transform infrared (FTIR), gel permeation chromatography (GPC), differential scanning calorimetry (DSC), wide angle x-ray powder-diffraction, and thermal gravimetric analysis (TGA). In vitro studies show that all the copolymers are degradable in phosphate buffer at 37 degrees C, and leaving an oily dimer acid residue after hydrolysis for the copolymer with high content of dimer acid. The release profiles of hydrophilic model drug, ciprofloxcin hydrochloride, from the copolymers, follow first-order release kinetics. All the preliminary results suggested that the copolymer might be potentially used as drug delivery devices.  相似文献

Three chemical transformations of oligosaccharide 1-deoxy-1-(4-trifluoroacetamidophenyl)aminoalditols are described. 1) Oxidation with hydrogen peroxide to give the corresponding reducing oligosaccharides. 2) Oxidation with cerium ammonium nitrate to give the corresponding 1-amino-1-deoxyalditols. 3) Treatment with acetic anhydride to give the correspondingN-acetylated derivatives, which are more stable towards oxidation.  相似文献

The polypeptide release factor gene, eRF1, of Blepharisma japonicum (Bj-eRF1) was cloned and sequenced. Its coding region was 1314 base pairs and encodes a protein of 437 amino acids. The cloned gene was expressed in Escherichia coli and the recombinant Bj-eRF1 polypeptide was purified by Ni2+-nitrilotriacetic acid agarose and Superose12 chromatography. Pull-down analysis showed that the recombinant Bj-eRF1 interacts with the heterologously-expressed release factor, eRF3C, of Euplotes octocarinatus.  相似文献

We design and develop chitosan nanoparticles which load two different drugs simultaneously. Aspirin (acetylsalicylic acid, ASA), a hydrophilic drug and probucol (PRO), a hydrophobic drug, are chosen as typical drugs, which are widely used to treat restenosis. The drug loaded chitosan nanoparticles are prepared by gelation of chitosan with tripolyphosphate (TPP) by ionic cross-linking. The physicochemical properties of nanoparticles are investigated by FTIR, transmission electron microscope (TEM), scanning electron microscopy (SEM) and differential scanning calorimetry (DSC). The images show that these particles are spherical in shape with ASA being in the amorphous phase, while PRO is crystalline. The properties of chitosan nanoparticles such as encapsulation capacity and controlled release behaviors of ASA and PRO are evaluated. Experimental results indicate that the loading capacity (LC), encapsulation efficiency (EE) and ASA and PRO release behaviors are affected by several factors including pH, concentration of TPP, chitosan molecular weight (MW) and ASA initial concentration as well as PRO. In vitro release shows that the nanoparticles provide a continuous release. Entrapped ASA is released for more than 24 h and PRO lasts longer for 120 h.  相似文献

In this study, carboxymethyl chitosan was prepared, characterized, and then photo-induced graft copolymerized with poly(ethylene glycol) under a nitrogen atmosphere in aqueous solution using 2,2-dimethoxy-2-phenyl acetophenone (DMPA) as the photo-initiator. The grafting copolymerization process was confirmed and the resulting copolymers were characterized using differential scanning calorimetry (DSC), FTIR spectroscopy, 2D-X ray diffraction, and elemental analysis. The kinetics of the grafting reactions was also studied. Under the applied experimental conditions, the optimum grafting values were obtained at: CMCs = 0.2 g, PEGA = 249 mM, DMPA = 10.4 mM at a 2 h reaction time. Some of the resulting copolymers were selected and used in the presence of methylene bisacrylamide (MBA) as a crosslinking agent to develop pH-responsive hydrogel matrices. The swelling characteristics and the in vitro release profiles of 5-fluorouracil (5-FU), as a model drug, from the hydrogels were investigated. The results revealed that the hydrogel matrices developed in this study can be customized to act as good candidates in drug delivery systems.  相似文献

4-Ethoxy-1-(p-tolyl)-s-triazine-2,6(1H,3H)-dione (TA) promoted mesocotyl growth in dark-grown rice (Oryza sativa L.) seedlings. In cultivars of the japonica type TA alone showed a small promotive effect and TA+gibberellic acid(GA3) had a marked synergistic effect, while in other cultivars, mostly of the indica type, TA alone showed a great promotive effect and TA+GA3 had only an additive effect. In cv. Nato, a typical representative of cultivars showing the second type of response, the concentration of TA giving the greatest growth promotion was around 0.1–0.2 mM. In Nato seedlings treated with TA at 0.1 mM, the mesocotyls continued to elongate for 6 days and reached about 75 mm in length, while the mesocotyls of control seedlings grew to a maximum of about 10 mm and growth was limited to the first 3 days after planting. The TA-induced mesocotyl elongation was mainly the consequence of increased cell multiplication in the meristematic area immediately below the coleoptilar node. GA3, abscisic acid (ABA) and ethylene also stimulated mesocotyl growth in dark-grown Nato seedlings but their effects were much smaller than those of TA. Summary Explosive spermatozoid release inLaminaria digitata is triggered by a pheromone secreted by female gametes. Dependence of the pheromone action on extracellular cations was investigated in defined test media using a pure pheromone preparation. The experimental results indicate successive processes in the sensory system and are summarized in a hypothesis on the release mechanism.
