The phloem sap of two lucerne (Medicago sativa L.) genotypes inducing differences in pea aphid,Acyrthosiphon pisum Harris (Homoptera: Aphididae) performances (weight, fecundity, survival), was collected by stylectomy (radio frequency microcautery). Sugars and amino acids were assayed. No significant difference in their concentrations could be established between genotypes. It is thus unlikely that a substantial part of the varietal resistance of these lucerne genotypes to pea aphid could be related to the content in sugars or amino acids. The same conclusion was drawn for the sugar/amino acid ratio. The amino acid profiles were globally similar. They nevertheless allowed a discrimination between the two genotypes on a few amino acids (alanine, leucine, isoleucine, arginine, ornithine) only. Moreover, the degree of resistance can be related neither to GABA concentration, nor to that of sulfur amino acids, because these constituants were either absent, or in trace amounts, whatever the lucerne genotype taken into account. Stylectomy parameters revealed some interesting differences in the sap exudation of the two genotypes. Average volume of exudation by stylet and average duration of exudation were respective 2.8 and 2.3 times higher in the susceptible genotype. These results suggest a reduced flow of phloem sap, after puncturing of the sieve-tubes by the aphid stylets on the resistant genotype; this could result in a greater difficulty in phloem sap ingestion by aphids settled on the resistant genotype and thus in suboptimal performances.  相似文献

In order to understand the mechanisms of resistance of two lucerne cultivars (susceptible Resistador and resistant Lahontan clones) to a French biotype of Acyrthosiphon pisum Harris, several biological parameters of this aphid were investigated on sap-copied holidic diets differing either by their amino acid or sucrose concentrations or by the relative proportions of some amino acids.We recorded the quantitative influence of nutrient levels and amino acid profiles on adult survival and reproduction, as well as on larval growth and development. The differences in A. pisum performances on artificial diets may partly by explained by variable ingestion rates, but also by differences in metabolic efficiency of the ingested nutrients, which greatly depends on amino acid composition.Although holidic diet experiments partially confirm the biological assays of resistance on plants, they also point out a lack in our understanding of the factors present in the sap which are involved in phagostimulation in vivo, thus preventing a complete interpretation of observed resistance.
Résumé Sur des milieux holidiques se différenciant par leur concentration en acides aminés ou en saccharose ou par leur spectre en acides aminés et copiant la sève de deux variétés de luzerne (sensible et résistante) nous avons étudié différents paramètres biologiques d'un biotype d'Acyrthosiphon pisum Harris afin de mieux comprendre les mécanismes de résistance à ce puceron.Que ce soit pour le reproduction et la survie des adultes ou pour la croissance et le développment larvaire, nous montrons l'influence spécifique de la concentration en saccharose et en acides aminés ainsi que celle du spectre en acides aminés des régimes artificiels. Ces différences de performances de A. pisum peuvent en partie être expliquées par les différences d'ingestion. Néanmoins pour des rapport saccharose/acides aminés identiques et pour des niveaux d'ingestion égaux, il existe selon les milieux de grandes différences de taux de reproduction et donc d'efficacités métaboliques.Les milieux copiant la sève de variétés sensible et résistante permettent jusqu'à présent d'expliquer une partie des observations faites sur végétal, mais démontrent que l'ignorance de certains facteurs phagostimulants présents dans la plante empêche une interprétation complète des effets observés.

When reared in short days (LD 12:12) at 15°C, apterous Acyrthosiphon pisum gave birth to sexual females (oviparae) exclusively for the first eight days of larviposition. After this time they switched to the production of parthenogenetic females (viviparae). Topical application of juvenile hormones I, II and III to fourth instar or adult ovipara-producers induced the precocious appearance of parthenogenetic females in the progeny sequence. Various forms intermediate between oviparae and viviparae were also produced and repetitive JH-I treatments resulted in a few alatiform progeny. However, many of the JH induced apterous, parthenogenetic females appeared to be normal viviparae and were capble of reproduction. Thus, prenatal treatment of oviparous embryos with JH diverts development towards the viviparous form. JH-I treatment of long-day reared A. pisum had no effect on the type of progeny produced.

Résumé Quand il est élevé sous jours courts (LN 12/12) à 15°C, le type anglais vert d'Acyrthosiphon pisum ne donne naissance qu'à des femelles sexuées (ovipares) pendant la première partie de sa période de reproduction. Ensuite quelques types intermédiaires ovipares/vivipares peuvent apparaitre avant que les pucerons ne bifurquent spontanément vers la production de femelles parthénogénétiques (vivipares). L'application cutanée d'hormones juvéniles (JH I, II, et III) aux larves de quatrième stade ou à des adultes producteurs d'ovipares provoque l'apparition prématurée d'intermédiaires et de vivipares dans la descendance. Les différentes formes intermédiaires produites par des applications répétées de J.H. comprenaient des types ailés ou partiellement ailés. Cependant, les vivipares aptères induits par J.H. étaient morphologiquement normaux et beaucoup étaient capables de se reproduire. Des traitements semblables aux J.H. de vivipares élevées en jours longs (LN 16/8) n'ont pas eu d'effets sur le type de la descendance.On ne sait pas si l'action de JH exogène sur l'induction des vivipares est direct ou indirect. La reprogrammation des embryons, autrement destinés à se développer comme ovipares, est examinée en relation avec notre connaissance du contrôle endocrine du polymorphisme des pucerons.

The effects of parasitism by Praon pequodorum Viereck (Hymenoptera: Aphidiidae) on the survival, development and reproduction of apterous pea aphids, Acyrthosiphon pisum (Harris) (Homoptera: Aphididae), were studied in the laboratory. Ten cohorts of aphids differing in age from 0.5 h to 9 days were tested. At 20°C, pea aphids survived an average of 7 days after parasitization. Parasitized first and second instars died before reaching the adult stage. Third instars died as adults but without producing any offspring. Parasitized fourth instars and adult aphids produced a variable number of offspring; their mean total fecundity increased as an exponential function of the age at parasitization. Host age did not significantly affect the period from the beginning of parasitism to the end of reproduction. Equations are provided to estimate the length of the reproductive period, the total fecundity, the intrinsic rate of increase, and the doubling time as functions of aphid age at parasitization. The data are discussed with regard to host population growth. It is shown that a parasite's potential impact on an aphid population is critically dependent on its preference for and successful oviposition in early host instars.
Résumé Les effets du parasitisme par P. pequodorum Viereck (Hym.: Aphidiidae), sur la survie, le développement et la reproduction des aptères de A. pisum, ont été examinés au laboratoire. L'étude a porté sur 10 cohortes de pucerons dont les âges différaient de 0,5 heure à 9 jours. A 20°C, les pucerons parasités ont survécu en moyenne pendant 7 jours après avoir été parasités. Leur évolution ultérieure dépend au stade auquel ils ont été parasités: au premier et second stades, ils sont morts avant de devenir adultes; au troisième stade, ils sont morts adultes avant de se reproduire; au quatrième stade et adultes, ils ont donné un nombre variable de descendants. La fécondité totale moyenne a crû comme une fonction exponentielle de l'âge au moment où ils avaient été parasités. L'âge de l'hôte n'a pas modifié significativement la durée de la période entre le moment où ils ont été parasités et la fin de la phase de reproduction. Des équations ont été établies en fonction de l'âge des pucerons au moment où ils ont été parasités, pour estimer la longueur de la période de reproduction, la fécondité totale, le taux intrinsèque d'accroissement et le temps de doublement des cohortes de pucerons. Les résultats sont discutés sous l'angle de la dynamique de la population. On en déduit que l'impact potentiel d'un parasite sur une population de pucerons dépend d'abord de ses préférences pour les premiers stades de l'hôte et du succès de sa ponte sur ces stades.

Abstract. A recently developed framework was applied to investigate the responses of newborn pea aphids, Acyrthosiphon pisum (Harris) (Homoptera: Aphididae), to simultaneous variations in dietary suerose and amino acid levels. The location of functional 'targets' for intake and growth were determined experimentally using performance criteria. Behavioural rules employed by insects to approach these targets were investigated by reference to the geometry of arrays of intake and growth across a range of diets. First-instar aphids were fed one of thirty-one chemically-defined diets ranging in sucrose concentration from 200 to 1000 mM and amino acid concentration from 25 to 250 mM. Insect survivorship, dry weight, protein- and carbohydrate-derived growth were high for all diets except those with the lowest nutrient levels. Peaks in final dry weight and protein content identified the intake target as the point reached by larvae fed the 600 mM sucrose, 75 mM amino acid diet. Aphids regulated sucrose primarily by consumption (i.e. behaviourally), whereas amino acids were regulated post-ingestively (i.e. by physiological mechanisms).  相似文献

Summary Pea aphids show intraclonal variability in antipredator behaviour. Among the offspring of a single parthenogenetically reproducing female, some individuals drop from the plant in response to alarm pheromone while others remain on the plant. We demonstrate that this intraclonal behavioural variability can be altered by selection. The proportion of aphids dropping in response to alarm pheromone was significantly greater in lines in which this behaviour was selected than in clonally identical lines in which the opposite phenotype was favoured. This change occurred within one generation and could not be attributed to grand-maternal effects, nor to environmental effects. These results demonstrate the ability of clonal aphids to adapt to changes in the environment within a single generation.  相似文献

The symbiotic bacteria Buchnera contribute to the nutrition of pea aphids, Acyrthosiphon pisum, through the provision of essential amino acids which are lacking in the diet. However, chemically defined diets, containing nutritionally adequate amounts of essential amino acids, fail to rescue aposymbiotic aphids, in which the bacteria have been disrupted with antibiotics. In this study the injection of a mixture of essential amino acids into the haemocoel of aposymbiotic aphids was shown to alleviate, at least partially, the impact of symbiont loss. Specifically, the total amino acid content in the tissues of aposymbiotic aphids was reduced by approximately 40% to levels comparable with symbiotic insects, and there was a 1.7-fold increase in the number of embryos, suggesting that the availability of essential amino acids promotes aphid protein synthesis by rejuvenating the free amino acid pool of aposymbiotic aphids. In addition, a similar effect on the total amino acid content was observed when phenylalanine alone, but not glutamine, lysine or tryptophan, was injected into the haemocoel of aposymbiotic aphids, and there was also a significant increase in the number of embryos following injection of phenylalanine or tryptophan alone. The impact of amino acid injection on the embryo complement of aposymbiotic aphids was limited to an increase in the number of embryos, with no increase in basal embryo size. It is proposed that older embryos may rely on their own complement of symbiotic bacteria for essential amino acid provisioning. Taken together, the data highlight the importance of bacterial provisioning of essential amino acids, particularly the aromatic amino acids, in the intact symbiosis.  相似文献

Twelve lines of Acyrthosiphon kondoi Shinji and Acyrthosiphon pisum (Harris) were sampled from a small lucerne field in Australia and tested to assess genetic variation by measuring their responses to three lucerne cultivars. Seven or eight life history traits were assessed for each species. Differences in the responses of these traits to the cultivars revealed that three to seven genetically distinct clones were represented in the samples of 12 lines for each species. We conclude that the populations of these two aphids are variable in Australia, although they were recently introduced and reproduce asexually. The responses of the aphids to the three cultivars are described. Because of genetic variation in host plant responses of the aphids, cultivars with potential aphid resistance must be exposed to a large number of clones to confirm the presence of useful aphid resistance.
Résumé Douze lignées d'Acyrthosiphon kondoi Shinji et d'A. pisum Harris ont été échantillonnées dans un petit champ de luzerne australien et leur variabilité génétique examinée en comparant les réponses à 3 cultivars de luzerne. De 7 à 8 caractères biologiques avaient été retenus pour chaque espèce. Les différences observées entre les réponses suivant la distribution de ces caractères chez les pucerons, ont montré que 3 à 7 clones génétiquement distincts étaient représentés dans les 12 lignées de chaque espèce. Nous en concluons que les populations de ces deux espèces de pucerons sont polymorphes en Australie, quoiqu'elles se multiplient d'une façon asexuée et aient été récemment introduites. Les réactions des pucerons aux 3 cultivars sont indiquées. Par suite de la variabilité génétique des réactions des plantes hôtes aux pucerons, des cultivars à résistance potentielle élevée devraient être exposés à un grand nombre de clones de pucerons pour confirmer la présence d'une résistance utilisable.

Pea aphids, Acyrthosiphon pisum, reproduce parthenogenetically and are wing-dimorphic such that offspring can develop into winged (alate) or unwinged (apterous) adults. Alate induction is maternal and offspring phenotype is entirely determined by changes in the physiology and environment of the mother. Juvenile hormones (JHs) have been implicated in playing a role in wing differentiation in aphids, however until recently, methods were not available to accurately quantify these insect hormones in small insects such as aphids. Using a novel LC-MS approach we were able to quantify JH III in pea aphids that were either producing a high proportion of winged morphs among their offspring or mainly unwinged offspring. We measured JH III titres by pooling the hemolymph of 12 or fewer individuals (1 μL hemolymph) treated identically. Levels of JH ranged from 30 to 163 pg/μL. While aphids in the two treatments strongly differed in the proportion of winged morphs among their offspring, their JH III titres did not differ significantly. There was also no correlation between JH III titre and the proportion of winged offspring in induced aphids. This supports earlier findings that wing dimorphism in aphids may be regulated by other physiological mechanisms.  相似文献

Two cultures of first instar pea aphids, Acyrthosiphon pisum (Harris) (Homoptera: Aphididae), were subjected to parasitization by Ephedrus plagiator (Nees) and Aphidius ervi Haliday (Hymenoptera: Braconidae) respectively, and a third culture, of fourth instar pea aphids, was subjected to parasitization by E. plagiator. Significant parasitogenic effects on the reproductive system occurred within 24 h of exposure to parasitoids in the form of reductions in both embryo number and the size of the largest embryo, and degenerative changes in the embryos themselves. No evidence was found of direct feeding by the larvae on the host's tissues, until just before larval pupation. The largest embryos of some fourth instar aphids escaped the effects of parasitization probably because they had developed a resistant cuticle by the time of oviposition by the parasitoid. Results suggested that embryonic degeneration was an indirect effect of parasitization due to starvation, interference with the aphid's hormone levels, or both.
Résumé Trois élevages de 100 à 200 premiers stades d'A. pisum, nés à 24 heures les uns des autres, ont été divisés chacun en un lot témoin et un lot expérimental. Deux des élevages expérimentaux ont alors été soumis au parasitisme d'Ephedrus plagiator ou de Aphidius ervi, le troisième élevage a pu se développer jusqu'au 4è stade avant d'être exposé à E. plagiator. Des échantillons de 10 pucerons ont été retirés de chaque lot après 24 heures et ensuite toutes les 48 heures pour être disséqués dans du liquide physiologique. Les nombres d'ovarioles et d'embryons et la taille du plus gros embryon ont été notés. Des cellules développées, issue de la séreuse de l'oeuf de l'hyménoptère se sont disséminées dans l'hémolymphe de l'hôte et ont servi d'aliment aux larves en développement. Seul le dernier stade larvaire des parasitoïdes s'est alimenté sur les tissus de l'hôte et ceci avait lieu juste avant la nymphose. Des effets significatifs se sont traduits après 24 heures par la réduction du nombre des embryons, par la diminution de la taille du plus grand de ceux-ci et par des altérations dans les embryons eux-mêmes.

The parasite Aphelinus asychisWalker (Hymenoptera: Aphelinidae) oviposits in all four instars of the pea aphid, Acyrthosiphon pisum(Harris) (Homoptera: Aphididae). Searching females display a highly stereotyped sequence of behaviors when encountering a host. Once recognized, an aphid is examined and probed by the wasp with the everted ovipositor prior to oviposition. Oviposition success is influenced by aphid behavior that is related to aphid size and expressed through instarspecific escape and defense reactions. Being smaller and less able to defend themselves, first and early-second instars of pea aphid are more susceptible to successful parasitism than third and fourth instars, in that order. Observed patterns of preference by Aphelinus females for particular aphid species and instars reflect the outcome of behavioral interactions between the hosts and the parasites, rather than preference in the strict sense.  相似文献

The osmotic pressure of the body fluids of aphids is lower than in their diet of plant phloem sap. It is hypothesised that aphids reduce the osmotic pressure of ingested food by sucrase-mediated hydrolysis of dietary sucrose to glucose and fructose, and the polymerisation of glucose into oligosaccharides of low osmotic pressure per hexose unit. To test this hypothesis, the impact of the alpha-glucosidase inhibitor acarbose on the sugar relations and osmoregulation of aphids was explored. Acarbose inhibited sucrase activity in gut homogenates and the production of monosaccharides and oligosaccharides in the honeydew of live aphids. Acarbose caused an increase in the haemolymph osmotic pressure for aphids reared on a diet (containing 0.75 M sucrose) hyperosmotic to the haemolymph and not on the isoosmotic diet containing 0.2 M sucrose. It did not affect aphid feeding rate over 2 days, except at high concentrations on 0.75 M sucrose diet, and this may have been a secondary consequence of osmotic dysfunction. Acarbose-treated aphids died prematurely. With 5 microM dietary acarbose, mean survivorship on 0.2 M sucrose diet was 4.2 days, not significantly different from starved aphids, indicating that, although these aphids fed, they were deprived of utilisable carbon; and on 0.75 M sucrose diet, mean survivorship was just 2.8 days, probably as a consequence of osmotic failure. It is concluded that the aphid gut sucrase activity is essential for osmoregulation of aphids ingesting food hyperosmotic to their body fluids.  相似文献

In temperate climates, pea aphids (Acyrthosiphon pisum) produce a single sexual generation each year in response to declining photoperiod and temperature. Mating occurs in the fall and the eggs have an obligatory winter diapause. Genetic recombination during the sexual phase is thought to be an important source of genetic variability within cyclically parthenogenetic aphid populations. Methods for reliably producing sexual forms and hatching the eggs of aphids are therefore central not only to the study of evolutionary change in aphid populations, but also for a general understanding of the origin of agriculturally important variation in destructiveness within pest species.Here, sexual forms of six pea aphid clones were induced in the laboratory and eggs were successfully hatched by creating conditions that closely mimicked those found in field situations. A declining photoperiod was produced by controlling artificial lighting using a timer with variable cycle length. Using these conditions, sexual forms were successfully produced for all six clones tested, which were then mated in all combinations. Eggs were exposed to a daily cycle of freezing and thawing in an incubator under a short-day photoperiod. Egg hatch averaged 60%, but was as high as 89% for some crosses. These methods will permit testing of evolutionary hypotheses and execution of detailed genetic studies of sources of variability within pea aphid populations. They are thus important tools for both evolutionary and agricultural studies.  相似文献

Conclusion Our behavioural data show that the vat resistance in melon is detected early by the aphid during stylet penetration, and is strongly reinforced during phloem feeding. The repulsive effect of phloem sap has been confirmed in vitro, thus demonstrating the presence of extractable chemical factors discriminating the genotypes. The near isogenicity of the two lines is biochemically confirmed by the high degree of homology of their tested composition in many nitrogenous metabolites. Two small peptides are significantly modified when the vat gene is present and they are presently under investigation. The distribution and mobility of infected aphid hosts can have a great effect on the ability of a pathogen to spread throughout a population. The distribution of dead and living pea aphids (Acyrthosiphon pisum) infected with Pandora neoaphidis was compared with that of their healthy conspecifics. Infected aphids were significantly more likely to be found on the undersides of alfalfa leaves and off of the plants than were healthy aphids. These two shifts in microhabitat location have potential costs and benefits for both the host and the pathogen.  相似文献

The effects of two bacterial endosymbionts, designated PASS and PAR, were evaluated on the pea aphid, Acyrthosiphon pisum (Harris) (Hemiptera:Aphididae), in which they occur facultatively, and on the blue alfalfa aphid, A. kondoi Shinji, in which these bacteria have not been found in natural populations. Subclones of pea aphids and blue alfalfa aphids, derived from parent aphid clones that did not contain PASS or PAR, were infected with one or both bacteria, generating PASS- and/or PAR-positive subclones with minimal genetic differences from the parent clones. Under laboratory conditions at 20 °C, PAR consistently reduced the fecundity (by between 19 and 60%) of subclones derived from three different parent pea aphid clones on bur clover, Medicago hispida Gaertn. PAR had intermediate effects on pea aphids reared on sweet pea, Lathyrus odoratus L., and had no significant effect on pea aphids on alfalfa, Medicago sativa L. The effect of PASS was either neutral or negative, depending on parent clone as well as host plant. Also at 20 °C, PASS reduced fecundity (70–77%) and longevity (40–48%), and increased the age of first reproduction (by up to 1.5 days) of blue alfalfa aphid reared on alfalfa and clover. PAR had a less dramatic effect (e.g., 30–39% reduction in fecundity) on these traits of blue alfalfa aphid. In contrast, PAR and PASS increased the fitness of pea aphid subclones of one parent clone reared for three generations at 25 °C on each of the three test plants. Without facultative bacteria, fecundity of the parent clone was reduced to a mean total of < 6 offspring per adult at this elevated temperature, but with PASS or PAR, mean total fecundity of its subclones was > 35. However, this ameliorative effect of facultative bacteria at 25 °C was not found for two other sets of parent clones and their derived subclones. Alate production in pea aphids was significantly increased in large populations of two PASS- and PAR-positive subclones relative to their parent clones. Attempts to transmit PASS or PAR horizontally, i.e., from aphid to aphid via feeding on host plants (bur clover), were unsuccessful.  相似文献

Recent progress in plant transformation for insect resistance has increased the interest in the potential toxicity of proteins towards insect pests. While studies have been targeted to a large array of insect species, phloem-feeding Homoptera have not been investigated yet. The paper describes a routine test for screening toxicity and growth inhibition of purified proteins in artificial diets onAcyrthosiphon pisum (Harris). Twenty-five commercially available proteins of different classes were tested and compared to some non-protein chemicals (an insecticide, an antibiotic …).A. pisum proved to be very sensitive to all proteases tested and to some venoms with general cytolytic properties. A plant lectin, concanavalin A, displayed significant toxicity and growth inhibition, while various proteins such as a soybean proteinase inhibitor, a chitinase, and bovine serum albumin showed measurable impairments of growth only at higher dose (≥250 μg.ml−1). Some proteins were without short-term effect onA. pisum physiology. The influence of these results on aphid-plant interactions are discussed.
Résumé L'effet de protéines alimentaires sur les insectes phloémophages, dont les pucerons, n'a jamais été étudié. Nous proposons ici un test biologique standardisé sur milieu artificiel permettant d'analyser les effets de différentes classes de protéines sur la physiologie d'A. pisum. La validité de ce test est éprouvée (protocole, reproductibilité) et les différentes données récoltées (mortalité et inhibition de croissance) permettent de définir des paramètres toxicologiques tels que concentration létale 50 ou concentration inhibitrice 50. Cette caractérisation toxicologique a été réalisée sur 25 protéines appartenant à des classes différentes, ainsi que plusieurs substances non protéiques utilisées comme témoin de toxicité (insecticide, antibiotique, inhibiteur de synthèse protéique et glucoside phénolique). Les regroupements de protéines par proximités de profils toxicologiques ont été corrélés aux activités biochimiques des différentes protéines. Les implications de ces résultats sur les interactions plante-puceron sont discutées, ainsi que le potentiel d'une stratégie de création de variétés transgéniques résistantes aux pucerons.

Aphids are highly specialized insects that feed on the phloem-sap of plants, the amino acid composition of which is very unbalanced. Amino acid metabolism is thus crucial in aphids, and we describe a novel investigation method based on the use of 14C-labeled amino acids added in an artificial diet. A metabolism cage for aphids was constructed, allowing for the collection and analysis of the radioactivity incorporated into the aphid body, expired as CO2, and rejected in the honeydew and exuviae. This method was applied to the study of the metabolism of eight energetic amino acids (aspartate, glutamate, glutamine, glycine, serine, alanine, proline, and threonine) in the pea aphid, Acyrthosiphon pisum. All these amino acids except threonine were subject to substantial catabolism as measured by high 14CO2 production. The highest turnover was displayed by aspartate, with 60% of its carbons expired as CO2. For the first time in an aphid, we directly demonstrated the synthesis of three essential amino acids (threonine, isoleucine, and lysine) from carbons of common amino acids. The synthesis of these three compounds was only observed from amino acids that were previously converted into glutamate. This conversion was important for aspartate, and lower for alanine and proline. To explain the quantitative results of interconversion between amino acids, we propose a compartmentation model with the intervention of bacterial endosymbiotes for the synthesis of essential amino acids and with glutamate as the only amino acid supplied by the insect to the symbiotes. Moreover, proline exhibited partial conversion into arginine, and it is suggested that proline is probably indirectly involved in excretory nitrogen metabolism. © 1995 Wiley-Liss, Inc.  相似文献
