About 95% of the ultraviolet (UV) photons reaching the Earth’s surface are UV-A (315–400 nm) photons. Plant responses to UV-A radiation have been less frequently studied than those to UV-B (280–315 nm) radiation. Most previous studies on UV-A radiation have used an unrealistic balance between UV-A, UV-B, and photosynthetically active radiation (PAR). Consequently, results from these studies are difficult to interpret from an ecological perspective, leaving an important gap in our understanding of the perception of solar UV radiation by plants. Previously, it was assumed UV-A/blue photoreceptors, cryptochromes and phototropins mediated photomorphogenic responses to UV-A radiation and “UV-B photoreceptor” UV RESISTANCE LOCUS 8 (UVR8) to UV-B radiation. However, our understanding of how UV-A radiation is perceived by plants has recently improved. Experiments using a realistic balance between UV-B, UV-A, and PAR have demonstrated that UVR8 can play a major role in the perception of both UV-B and short-wavelength UV-A (UV-Asw, 315 to ∼350 nm) radiation. These experiments also showed that UVR8 and cryptochromes jointly regulate gene expression through interactions that alter the relative sensitivity to UV-B, UV-A, and blue wavelengths. Negative feedback loops on the action of these photoreceptors can arise from gene expression, signaling crosstalk, and absorption of UV photons by phenolic metabolites. These interactions explain why exposure to blue light modulates photomorphogenic responses to UV-B and UV-Asw radiation. Future studies will need to distinguish between short and long wavelengths of UV-A radiation and to consider UVR8’s role as a UV-B/UV-Asw photoreceptor in sunlight.

In sunlight, UVR8 mediates the perception of both UV-B and short-wavelength UV-A radiation with its sensitivity moderated by blue light perceived through cryptochromes.  相似文献

Using quartz optical fibres, penetration of both monochromatic (310 nm) and polychromatic UV-B (280–320 nm) radiation in leaves of Brassica napus L. (cv. Ceres) was measured. Plants were grown under either visible light (750 μmol m−2 s−1 photosynthetically active radiation) or with the addition of 8. 9 KJ m−2 day−1 biologically effective UV-B (UV-BBE) radiation. Results showed that of the 310 nm radiation that penetreated the leaf, 90% was within the intial one third of the leaf with high attenuation in the leaf epidermis, especially in UV-treated plants. Polychromatic UV-B radiation, relative to incident radiation, showed a relatively uniform spectral distribution within the leaf, except for collimated radiation. Over 30% of the UV-screening pigments in the leaf, including flavonoids, were found in the adaxial epidermal layer, making this layer less transparent to UV-B radiation than the abaxial epidermis, which contained less than 12% of the UV-screening pigments. UV-screening pigments increased by 20% in UV-treated leaves relative to control leaves. Densely arranged epicuticular wax on the adaxial leaf surface of UV-treated plants may have further decreased penetration of UV-B radiation by reflectance. An increased leaf thickness, and decreases in leaf area and leaf dry weight were also found for UV-treated plants.  相似文献

In higher plants one of the important functions of the leaf epidermis is the effective screening of ultraviolet-B (280–320 nm, UV-B) radiation, due mostly to phenolic compounds. The assessment of the contribution of this function is necessary for an evaluation of the impact of increasing UV-B radiation. A method is proposed to estimate epidermal transmittance on the basis of chlorophyll fluorescence measurements. Fluorescence of chlorophyll induced by UV-A (320–400 nm, measuring beam centered at 366 nm, half band width 32 nm) or UV-B (measuring beam centered at 314 nm, half band width 18 nm) is compared to that induced by a blue-green measuring light (475 nm, half band width 140 nm). It is shown that the ratios of UV-and blue-green (BG)-induced fluorescence, F(UV-A)/F(BG) and F(UV-B)/F(BG), are relatively constant among leaf samples of various species ( Vicia faba, Spinacia oleracea, Rumex scutatus ) from which the epidermis was removed. In epidermis-free leaves no significant differences were found between adaxial and abaxial leaf sides, suggesting that leaf structure has negligible influence on the F(UV)/F(BG) ratios. On the other hand, fluorescence excitation ratios varied over a vast range when intact leaves from different species and habitats were investigated. Ratios were low in sun leaves and relatively high in shade- and greenhouse-grown leaves. By relating these results to those obtained with epidermis-free leaves, epidermal transmittances for UV-B radiation could be estimated, with values ranging between 1 and 45%. The data demonstrate a large adaptability of epidermal UV-A and UV-B transmittance in higher plants. The proposed method may prove a versatile and relatively simple tool for investigating epidermal UV transmittance complementing established methods.  相似文献

Experiments were conducted on the atrazine-tolerant mutant Stallion and the atrazine-sensitive cv. Paroll of Brassica napus L., which were grown under either visible light or with the addition of UV-B radiation (280–320 nm) for 15 days. The mutant has been shown to be sensitive to high levels of visible light as compared to the atrazine-sensitive cultivar and therefore we wished to determine plant response to UV-B radiation with respect to potential pigment changes, certain anatomical features, radiation penetration and partial photosynthesis. With regard to pigment changes, we were particularly interested in whether the compositional shift in flavonol pigments under enhanced UV-B radiation, previously suggested to favour increased antioxidant activity, is confined to the adaxial epidermis, which generally receives most UV-B radiation or whether the pigment shift is also inducible in the abaxial epidermis. As was to be expected, the penetration of UV-B radiation (310 nm) was lower in the UV-B-exposed plants, which was correlated with an increased amount of UV-screening pigments in the adaxial and abaxial epidermal layers. The main flavonoid glycosides showed the largest shift from kaempferol to quercetin as aglycone moiety in the adaxial epidermal layer. However, in the abaxial epidermal layer the hydroxycinnamic acid (HCA) derivatives and kaempferol glycosides were predominant. Penetration of 430 nm light was higher after UV-B exposure, and probably contributed to the fact that photosynthetic efficiency of photosystem II was unchanged or higher after UV-B exposure. UV-B radiation decreased leaf area in the atrazine-tolerant mutant only. Both cultivars showed an increased leaf thickness after UV-B exposure due to cell elongation mainly of the palisade tissue. This was especially evident in the mutant.  相似文献

Three general classes of photomorphogenic photoreceptors have been characterized in higher plants: phytochrome, a blue light/ultraviolet (UV)-A photoreceptor(s), and a UV-B sensory system(s). Although a great deal is known about phytochrome and the blue light/UV-A photoreceptor(s), little is known about UV-B detection processes. One reason for this is the lack of readily quantifiable morphogenic responses that are specifically induced by UV-B radiation. We have discovered a response to UV-B, upward curling of Brassica napus L. cotyledons, that may be useful for probing the mechanism of UV-B photoreception. The process was initially observed when B. napus seeds were germinated under visible light plus UV-B radiation, but did not occur under visible light alone or visible light plus UV-A. When 5-d-old seedlings grown in visible light were given relatively short exposures of UV-B (100 min of 5.5 [mu]mol m-2 s-1), the curling response was also observed. Development of curling was separated from the application of this UV-B pulse by a 14-h latent period. Pulses of red light, blue light, farred light, and UV-A (100 min of 5.5 [mu]mol m-2 s-1) did not induce curling, indicating UV-B specificity Additionally, these other spectral regions did not reverse or enhance the UV-B-triggered response. The degree of curling showed a log-linear dependence on UV-B fluence (6-40 mmol m-2) and reciprocity with respect to length of exposure and fluence rate. The data indicate that curling is photomorphogenic in nature and may be triggered by a single photoreceptor species.  相似文献

Ultraviolet-B radiation absorbing capacity of leaf hairs   总被引:14,自引:0,他引:14
Pubescence layers with their native structure and orientation were isolated from the leaves of Olea europaea L. and Olea chrysophylla L. They were almost transparent in the visible, but considerable absorptance was evident in the ultraviolet-B region (UV-B), with maximum at 310 nm. Methanolic extracts of hairs from Olea and a variety of other pubescent species consistently showed the existence of UV-screening pigments. Absorptance of trichomes varied, but a trend towards more effective UV-B radiation attenuation in the sub-alpine Verbascum species may be claimed. In all cases, pigments were located within hair cells and in Olea they were characterized as phenolics with considerable flavonoid contribution. It is suggested that leaf hairs, besides other functions, may constitute a shield against UV-B radiation.  相似文献

Leaves of Vicia faba were collected from the field and the greenhouse and transmittance of epidermal peels from adaxial and abaxial sides was determined in the wavelength range from 250 to 800 nm using a spectrophotometer equipped for the measurement of turbid samples. From the same leaves, epidermal transmittance was estimated by a recently developed fluorometric method. Both methods gave highly correlated results with a slope of the regression line between both methods close to 1 and an intercept close to 0. Transmittances at around 310 nm as low as 3% were detected in the adaxial epidermis of field-grown leaves, while transmittance could be as high as 70% in the abaxial epidermis of greenhouse-grown leaves. There was a strong correlation between UV-A (ca. 366 nm) and UV-B (ca. 310 nm) transmittance detected by both methods which could be explained by the pigment composition in methanolic extracts where flavonols accounted for 90% of the absorption at 310 nm in the extract, while hydroxycinnamic acid derivatives which absorb only at the shorter wavelength constituted about 5%. It is concluded that the fluorescence method which allows rapid measurements on intact leaves can provide a quantitative estimate of epidermal transmittance for UV-B (280–320 nm) and UV-A (320–400 nm) radiation. This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

In wild-type (WT) Columbia and Landsberg erecta ecotypes of Arabidopsis thaliana (L.) Heynh., trichomes are present on the adaxial surfaces of all rosette leaves but are absent from the abaxial surfaces of the first-formed leaves. We have determined that both long-day (LD) photoperiod and gibberellin (GA) stimulate trichome formation. WT plants grown in LD conditions produce the first abaxial trichome on earlier leaves than plants grown in short-day (SD) conditions. Photoperiod sensitivity of abaxial trichome formation on WT plants develops gradually over time, reaching the maximum sensitivity about 24 d after germination. Application of gibberellic acid to WT plants growing in SD conditions accelerates the onset of abaxial trichomes. Conversely, application of 20 to 80 mg L-1 paclobutrazol, a GA biosynthesis inhibitor, to wild-type plants suppresses trichome initiation on the abaxial epidermis. The GA-deficient mutants ga1-5 and ga4-1 and the GA-insensitive mutant gai-1 exhibit delayed onset of abaxial trichomes when grown in LD conditions. The null mutant ga1-3 produces completely glabrous leaves when grown in SD conditions. Application of gibberellic acid to glabrous ga1-3 plants consistently induces earlier formation of trichomes on the adaxial epidermis than on the abaxial epidermis, demonstrating a difference between the adaxial and abaxial surfaces in their response to GA with regard to trichome formation.  相似文献

In 1991 a field experiment was established in subarctic heathland at Abisko (68°35'N, 18°82'E), northern Sweden, to investigate the effects of enhanced UV-B (280–315 nm) radiation, simulating 15% ozone depletion, on plants in their natural environment. Leaves of the four dominant dwarf shrubs, the deciduous Vaccinium myrtillus L. and V. uliginosum L. and the evergreen V. vitis-idaea L. and Empetrum hermaphroditum Hagerup were examined after 7 years of UV-B treatment. SEM and ESEM were used to visualize surface features and to determine trichome density. Multiphoton laser scanning microscopy showed that UV-B absorbing compounds were localized in the trichomes of all species. Trichomes varied in size, number and distribution between the species. Enhanced UV-B reduced adaxial trichome density significantly (by approximately 25%) in only one species, V. uliginosum . This effect could be of importance for the UV-B absorbing potential of the adaxial epidermis of V. uliginosum . Epicuticular wax structures were found only on the abaxial surface of V. uliginosum and were unaffected by enhanced UV-B. The cuticular surfaces of all other species were smooth and featureless. Leaf thickness, adaxial and abaxial cuticle thickness varied between the species although there was no apparent effect of enhanced UV-B. It is concluded that long-term enhancement of UV-B has an effect on adaxial trichome density in V. uliginosum , but that there is no general effect on leaf morphology of the other species.  相似文献

The effect of salicylic acid (SA) counteracting the UV-A, UV-B, and UV-C-induced action on pepper (Capsicum annuum L.) plants was studied. For this purpose, the activities of antioxidant enzymes (peroxidase, polyphenol oxidase, ascorbate peroxidase, catalase, and glutathione reductase) were measured. Plants were sprayed with SA and treated with UV-A (320–390 nm), UV-B (312 nm), and UV-C (254 nm) radiation with a density of 6.1, 5.8, and 5.7 W/m2. The activities of antioxidant enzymes were enhanced in leaves in response to UV-B and UV-C radiation. SA treatment moderated an increase in the activities of some antioxidant enzymes (peroxidase, ascorbate peroxidase, catalase, and glutathione reductase) in plants that were treated with UV radiation. The activity of antioxidant enzyme polyphenol oxidase in plants that were treated with UV-B, UV-C, and SA was significantly increased. The aim of the present study was to investigate the possible protective effect of SA treatment on UV-A, UV-B, and UV-C stress.  相似文献

Three-year-old birch (Betula pendula Roth.) seedlings were exposed, in the field, to supplemental levels of UV-B radiation. Control seedlings were exposed to ambient levels of UV radiation, using arrays of unenergized lamps. A control for UV-A radiation was also included in the experiment. Enhanced UV-B radiation had no significant effects on height growth, and shoot and root biomass of birch seedlings. Leaf expansion rate increased transiently in the middle of the growing period in enhanced UV-B- and UV-A-exposed plants; however, final leaf size and relative growth rate remained unaffected. Leaf thickness and spongy intercellular spaces were increased in UV-B-exposed seedlings along with increased density of glandular trichomes. At the ultrastructural level, enhanced UV-B increased the number of cytoplasmic lipid bodies, and abnormal membrane whorls were found. Both enhanced UV-B and UV-A radiation induced swelling of chloroplast thylakoids. Stomatal density and conductance were significantly increased by elevated UV-B radiation. UV-A radiation increased the length and width of stomata, whereas UV-B radiation had only a marginal effect on stomatal size. UV-A and enhanced UV-B radiation attenuated the appearance of necrotic spots in autumn, probably caused by the fungus Pyrenopeziza betulicola, suggesting a direct harmful effect of UV on pathogens or reduced susceptibility to pathogens in UV-exposed seedlings. Secondary metabolite analysis showed increases in (+)-catechin, quercetin, cinnamic acid derivative, apigenin and pentagalloylglucose in birch leaves under enhanced UV-B radiation. Negative correlations between apigenin, and particularly quercetin concentrations and lipid peroxidation levels indicated an antioxidant role of secondary metabolites in birch leaves exposed to UV-B radiation.  相似文献

Sugar beet ( Beta vulgaris L.) plants injected with Cercospora beticota Sace. as well as non-infected plants were grown under visible light with or without ultraviolet-B (UV-B, 280–320 nm) radiation for 40 days. An interaction between UV-B radiation and Cercospora leaf spot disease was observed, resulting in a large reduction in leaf chlorophyll content, dry weight of leaf laminae, petioles and storage roots. Lipid peraxidation in leaves also increased the most under the combined treatments. This was also true for ultraweak luminescence from both adaxial and abaxial leaf surfaces. However, no correlation between lipid peroxidation and ultraweak luminescence was observed. Ultraviolet-B radiation given alone appeared to have either a stimulating effect, giving an increase in dry weight of laminae and reducing lipid peroxidation, or no effect. This lack of effect was seen in the absence of change in dry weight of storage roots and chlorophyll content relative to controls. The :study demonstrated a harmful interaction between UV-B radiation and Cercospom leaf spot disease on sugar beet.  相似文献

Spectral reflectance and transmittance of leaves to ultraviolet irradiation were determined under laboratory conditions for seven species of hardwood trees, namely red oak (Quercus rubra, L), black oak (Q. velutina, Lamarch), white oak (Q. alba, L.), sugar maple (Acer saccharum), Norway maple (A. plantanoides), hickory (Carya tomemtosa), sweetgum (Liquidambar styraciflua), and black oak litter. The experimental system consisted of a solar simulator, an integrating sphere, and a spectroradiometer. Experiments were repeated three to five times for both adaxial and abaxial surfaces of fresh leaves chosen at randomly. The spectral distributions and simple averages of the radiative properties in the wavelength ranges of ultraviolet-B (UV-B, 280–320 nm) and ultraviolet-A (UV-A, 320–400 nm) were determined. The spectral distributions of reflectance were similar between adaxial and abaxial surfaces, although the magnitude varied among tree species. Leaf reflectance was very low for the ultraviolet spectrum in general and varied among species and between adaxial and abaxial surfaces. It was generally higher over the UV-A waveband compared to UV-B, and higher on the abaxial than adaxial surface. The broadband reflectance in the UV-A range (over all species) was 5.0 and 3.9% for abaxial and adaxial surface, respectively, compared to 3.5 and 2.8% in UV-B. The transmittance through leaves was extremely small in the UV-B (<0.1%) and nearly zero in the UV-A spectral range. Consequently, the absorptance of ultraviolet radiation by leaves, as determined from the measured reflectance and transmittance, was quite high, being more than 90% for all the combinations of species and wavebands examined. The reported results are useful for studies requiring spectral radiative properties of the examined leaves with respect to ultraviolet irradiation.  相似文献

Grape (Vitis vinifera cv Silvaner) vine plants were cultivated under shaded conditions in the absence of ultraviolet (UV) radiation in a greenhouse, and subsequently placed outdoors under three different light regimes for 7 d. Different light regimes were produced by filters transmitting natural radiation, or screening out the UV-B (280-315 nm), or screening out the UV-A (315-400 nm) and the UV-B spectral range. During exposure, synthesis of UV-screening phenolics in leaves was quantified using HPLC: All treatments increased concentrations of hydroxycinnamic acids but the rise was highest, reaching 230% of the initial value, when UV radiation was absent. In contrast, UV-B radiation specifically increased flavonoid concentrations resulting in more than a 10-fold increase. Transmittance in the UV of all extracted phenolics was lower than epidermal UV transmittance determined fluorimetrically, and the two parameters were curvilinearly related. It is suggested that curvilinearity results from different absorption properties of the homogeneously dissolved phenolics in extracts and of the non-homogeneous distribution of phenolics in the epidermis. UV-B-dependent inhibition of maximum photochemical yield of photosystem II (PSII), measured as variable fluorescence of dark-adapted leaves, recovered in parallel to the buildup of epidermal screening for UV-B radiation, suggesting that PSII is protected against UV-B damage by epidermal screening. However, UV-B inhibition of CO(2) assimilation rates was not diminished by efficient UV-B screening. We propose that protection of UV-B inactivation of PSII is observed because preceding damage is efficiently repaired while those factors determining UV-B inhibition of CO(2) assimilation recover more slowly.  相似文献

UV and blue light control the expression of flavonoid biosynthesis genes in a range of higher plants. To investigate the signal transduction processes involved in the induction of chalcone synthase (CHS) gene expression by UV-B and UV-A/blue light, we examined the effects of specific agonists and inhibitors of known signaling components in mammalian systems in a photomixotrophic Arabidopsis cell suspension culture. CHS expression is induced specifically by these wavelengths in the cell culture, in a manner similar to that in mature Arabidopsis leaf tissue. Both the UV-B and UV-A/blue phototransduction processes involve calcium, although the elevation of cytosolic calcium is insufficient on its own to stimulate CHS expression. The UV-A/blue light induction of CHS expression does not appear to involve calmodulin, whereas the UV-B response does; this difference indicates that the signal transduction pathways are, at least in part, distinct. We provide evidence that both pathways involve reversible protein phosphorylation and require protein synthesis. The UV-B and UV-A/blue light signaling pathways are therefore different from the phytochrome signal transduction pathway regulating CHS expression in other species.  相似文献

Bacterial endospores are 1 to 2 orders of magnitude more resistant to 254-nm UV (UV-C) radiation than are exponentially growing cells of the same strain. This high UV resistance is due to two related phenomena: (i) DNA of dormant spores irradiated with 254-nm UV accumulates mainly a unique thymine dimer called the spore photoproduct (SP), and (ii) SP is corrected during spore germination by two major DNA repair pathways, nucleotide excision repair (NER) and an SP-specific enzyme called SP lyase. To date, it has been assumed that these two factors also account for resistance of bacterial spores to solar UV in the environment, despite the fact that sunlight at the Earth's surface consists of UV-B, UV-A, visible, and infrared wavelengths of approximately 290 nm and longer. To test this assumption, isogenic strains of Bacillus subtilis lacking either the NER or SP lyase DNA repair pathway were assayed for their relative resistance to radiation at a number of UV wavelengths, including UV-C (254 nm), UV-B (290 to 320 nm), full-spectrum sunlight, and sunlight from which the UV-B portion had been removed. For purposes of direct comparison, spore UV resistance levels were determined with respect to a calibrated biological dosimeter consisting of a mixture of wild-type spores and spores lacking both DNA repair systems. It was observed that the relative contributions of the two pathways to spore UV resistance change depending on the UV wavelengths used in a manner suggesting that spores irradiated with light at environmentally relevant UV wavelengths may accumulate significant amounts of one or more DNA photoproducts in addition to SP. Furthermore, it was noted that upon exposure to increasing wavelengths, wild-type spores decreased in their UV resistance from 33-fold (UV-C) to 12-fold (UV-B plus UV-A sunlight) to 6-fold (UV-A sunlight alone) more resistant than mutants lacking both DNA repair systems, suggesting that at increasing solar UV wavelengths, spores are inactivated either by DNA damage not reparable by the NER or SP lyase system, damage caused to photosensitive molecules other than DNA, or both.  相似文献

Plants exposed to natural solar radiation usually show acclimation responses on a daily and seasonal basis. Many of these responses are complex and modified by interactions with acclimation responses to other climatic factors. While changes in photosynthetically active radiation (PAR, 400-700 nm) are the driving force for many acclimation responses in plants, radiation outside the PAR range is also important. Recently, interest has increased in the potential role of UV-A (320-400 nm) and UV-B (280-320 nm) components of sunlight in plant developmental, physiological and daily acclimation processes. In order to explore the role of UV-B further, Brassica napus L. cv Paroll plants were grown to maturity under 13 kJ d(-1) of biologically effective ultraviolet-B radiation (UV-B(BE), 280-320 nm) plus 800 micromol photons m(-2) s(-1) photosynthetically active radiation (PAR, 400-700 nm) or PAR alone. Leaf anatomy and palisade cell structure were quantified using stereological techniques. The leaves of plants grown under UV-B radiation exhibited an increase in overall leaf width, although no change in leaf anatomy was discerned. Palisade cells in UV-B exposed leaves showed a significant decrease in chloroplast, mitochondrial, starch, and microbody volume density (Vv), while the vacuolar Vv increased compared to cells exposed to PAR only. In UV-B exposed leaves, there was an increase in the appressed and non-appressed thylakoid surface area density (Sv) within the chloroplasts. Since the relative proportion of appressed to non-appressed thylakoid surface area did not change, both thylakoid systems changed in concert with each other. Thylakoid stacks were broader and shorter in leaves subjected to UV-B. In general these responses were similar to those which occurred in plants moved from a high to low PAR environment and similar to mature plants exposed to 13 kJ d(-1) UV-B(BE) for only a short period of time. Although UV absorbing pigments increased by 21% in UV-B exposed leaves, there was no significant difference in chlorophyll a,b or carotenoid content compared to plants exposed to only PAR.
