应用透射电镜对矮生菜豆的游离核胚乳、胚乳细胞及珠被绒毡层的超微结构进行了观察,揭示了胚乳细胞化过程中自由生长壁的形成方式。实验表明,在游离核胚乳中存在大量有被小泡,胚乳游离核的外层核膜发生明显外突并以出芽的方式产生有被小泡。初始胚乳细胞壁为自由生长壁,大量有被小泡融合于自由生长壁末端使之不断延伸并产生分枝。与珠被绒毡层相邻的胚乳细胞产生发达的壁内突,具有传递细胞特征;在这些细胞中出现大量的核糖体及粗糙内质网,表现现高度的合成活性。 相似文献

传粉后10 d,薏苡(Coix lacrym a-jobiL.)颖果基部胚乳最外层传递细胞具长而多的壁内突,第二层细胞的壁内突较第一层的短而少,均具瓣裂的细胞核、丰富的线粒体、粗糙内质网、核糖体、产生小泡的高尔基体及与壁内突质膜相连的、含深色物质的囊泡。线粒体分布于壁内突附近或其间。授粉后25 d,第一、二层细胞壁内突发达,几乎充满了细胞,但细胞器可见。第四层传递细胞具树枝状及网状的壁内突,大量线粒体、具质体膜的淀粉粒、脂体存在壁内突附近或壁内突的间隙内。高尔基体常见,仅见很少的片段内质网。第五层传递细胞具短的壁内突、较大的淀粉粒及许多小蛋白质体。两个时期的第一、二层细胞内均未观察到胞间连丝。授粉后25 d,第四层及以上的传递细胞的细胞壁和呈网状的壁内突均含有胞间连丝。还讨论了各种细胞器的作用及各层传递细胞的功能 相似文献

白刺(Nitraria sibirica)胚乳发育经历游离核阶段、细胞化阶段和被吸收解体阶段。游离核胚乳沿胚囊壁均匀排列为一层,胞质浓厚,其中有丰富的质体、线粒体、高尔基体、内质网和各种小泡等细胞器。珠孔区域的胚囊壁具发达的分枝状壁内突,而周缘区域的胚囊壁具间隔的钉状内突,内突周围的细胞质中具多数线粒体和小泡。胚乳细胞化时,初始垂周壁源于核有丝分裂产生的细胞板。在细胞板两端开始壁的游离生长,一端与胚囊壁相连接,另一端向心自由延伸。壁的游离生长依赖于小泡的融合。早期胚乳细胞具大液泡,具核或无核,细胞质中有大量的线粒体,质体缺乏,其壁仍由多层膜结构组成。 相似文献

Triticum aestivum cv. “Yang Mai # 1” was used to study the celI wall formation in free-nuclear endosperm with electronmicroscopy and fluorescence microscopy. During the initiation of cellularization the peg-like wall ingrowths developed, and the freely growing walls gradually cleaved the cytoplasm into small compartments in both regions (ventral and dorsal) of the 'endosperm sac, but in ventral region the anastomose of freely growing wall often occured. The striking resemblance and close connection betwteen nucellar debris and freely growing walls showed the possibility that the disintegrated nucellus played an important role in supporting the wall formation of endosperm during the early stages of cellularization. 相似文献

Summary Endosperm cellularization in Ranunculus sceleratus was studied in terms of the initiation of cell-wall formation in the coenocytic endosperm. The first endosperm cell walls were in an anticlinal position relative to the cell wall of the embryo sac and originated from the cell plates and not from wall ingrowths from the embryo-sac wall itself. Alveolar endosperm was formed 3 days after pollination. Microtubules were associated with the freely growing wall ends of the anticlinal walls and were observed in various orientations that generally ranged from angles of 45 ° to 90 ° to the plane of the wall. They were absent in the regions where vesicles had already fused. These microtubules may function in maintaining the growth and the direction of growth of the anticlinal wall until cellularization is completed. At the site where three neighbouring alveoli share their freely growing wall ends, remarkable configurations of microtubules were observed: in each alveolus, microtubules ran predominantly parallel to the bisector of the angle formed by the common walls. These microtubules may form a physically stable framework and maintain the direction of growth of the wall edges. It is concluded that the growing edge of the anticlinal endosperm wall and its associated microtubules are a special continuum of the original phragmoplast that gave rise to the anticlinal wall. 相似文献

观察了“章丘大葱”胚珠、胚囊的结构,胚及胚乳的发育。胚发育属紫菀型,经历球胚前的原胚、球胚、椭形胚、凹形胚、长棒形胚及弯形成熟胚等各期。宿存助细胞生存到球形胚期。胚乳发育属核型。球胚晚期,在胚褒珠孔端及合点端,胚乳开始细胞化。由于游离核之间出现了垂周壁,由一层胚乳游离核形成了一层无内切向壁的“开放细胞”。“开放细胞”行平周分裂而形成两层细胞,外层为完整细胞,内层为新的“开放细胞”。如此多次分裂,向心增生胚乳细胞,最后将中央细胞填满。初始的垂周壁来自于间期游离核之间的细胞板,非“自由生长壁”但又未见明显的成膜体。初始的平周壁是正常有丝分裂的胞质分裂的结果,与成膜体,细胞板有关。在球胚晚期,胚乳细胞化后,在中央液泡内观察到游离细胞。 相似文献

ABSTRACT Sarcocystis dubeyella n. sp. and S. phacochoeri n. sp. from muscle fibers of the skeletal musculature of two warthogs in South Africa are described by light and and electron microscopy. Sarcocystis dubeyella sarcocysts are macroscopic (up to 12 mm long and 1 mm wide), with a parasite-induced encapsulation of the host muscle fiber in which the plasma membrane of the latter remained unaltered. The sarcocyst wall is characterized by evenly arranged, irregularly semicircular or rectangular villar protrusions (5.0 T. 2.8-11.0 μm) with indented margins and no specific content. Sarcocystis phacochoeri formed filiform microcysts (up to 4 mm long and 0.13 mm wide). Its cyst wall is provided with tightly packed, molarlike villar protrusions (1.6-3.3 T. 1.7-3.3 μm), with smooth margins, hollow on one side, and with longitudinal condensations of the fine granular matrix at various locations in the interior. 相似文献

Previous studies using spectroscopic imaging have allowed the spatial distribution of structural components in wheat endosperm
cell walls to be determined. FT-IR microspectroscopy showed differing changes in arabinoxylan (AX) structure, during grain
development under cool/wet and hot/dry growing conditions, for differing cultivars (Toole et al. in Planta 225:1393–1403,
2007). These studies have been extended using Raman microspectroscopy, providing more details of the impact of environment
on the polysaccharide and phenolic components of the cell walls. NMR studies provide complementary information on the types
and levels of AX branching both early in development and at maturity. Raman microspectroscopy has allowed the arabinose:xylose
(A/X) ratio in the cell wall AX to be determined, and the addition of ferulic acid and related phenolic acids to be followed.
The changes in the A/X ratio during grain development were affected by the environmental conditions, with the A/X ratio generally
being slightly lower for samples grown under cool/wet conditions than for those from hot/dry conditions. The degree of esterification
of the endosperm cell walls with ferulic acid was also affected by the environment, being lower under hot/dry conditions.
The results support earlier suggestions that AX is either delivered to the cell wall in a highly substituted form and is remodelled
through the action of arabinoxylan arabinofuranohydrolases or arabinofuranosidases, or that low level substituted AX are incorporated
into the wall late in cell wall development, reducing the average degree of substitution, and that the rate of this remodelling
is influenced by the environment. 1H NMR provided a unique insight into the chemical structure of intact wheat endosperm cell walls, providing qualitative information
on the proportions of mono- and disubstituted AX and the levels of branching of adjacent units. The A/X ratio did not change
greatly with either the development stage or the growth conditions, but the ratio of mono- to disubstituted Xylp residues increased markedly (by about fourfold) in the more mature samples, confirming the changes in branching levels determined
using FT-IR. To the best of our knowledge, this is the first time that intact endosperm cell walls have been studied by 1H NMR. 相似文献

One of the major factors contributing to the failure of new wheat varieties is seasonal variability in end-use quality. Consequently,
it is important to produce varieties which are robust and stable over a range of environmental conditions. Recently developed
sample preparation methods have allowed the application of FT-IR spectroscopic imaging methods to the analysis of wheat endosperm
cell wall composition, allowing the spatial distribution of structural components to be determined without the limitations
of conventional chemical analysis. The advantages of the methods, described in this paper, are that they determine the composition
of endosperm cell walls in situ and with minimal modification during preparation. Two bread-making wheat cultivars, Spark
and Rialto, were selected to determine the impact of environmental conditions on the cell-wall composition of the starchy
endosperm of the developing and mature grain, focusing on the period of grain filling (starting at about 14 days after anthesis).
Studies carried out over two successive seasons show that the structure of the arabinoxylans in the endosperm cell walls changes
from a highly branched form to a less branched form. Furthermore, during development the rate of restructuring was faster
when the plants were grown at higher temperature with restricted water availability from 14 days after anthesis with differences
in the rate of restructuring occurring between the two cultivars. 相似文献

Summary The first stages of cell wall formation were followed in the root meristems of maize and French bean. Most of the primary wall components (hemicellulose, cellulose and highly methylated pectins) were laid down simultaneously along the cell plate. During young cell wall maturation within the meristem itself, significant topochemical alterations, coupled with the addition of new polysaccharides, produced complete redistribution of wall material leading to the progressive appearance of a proper middle lamella. Thus the formation of a pectic middle lamella does not precede the deposition of primary walls. It is delayed until the new partition joins to the mother cell wall.Abbreviations DMSO

Summary The effect of 2,6-dichlorobenzonitrile on cytokinesis of meristematic cells of onion root during both treatment and recovery has been studied by electron microscopic techniques. 2,6-dichlorobenzonitrile interferes with cell plate formation in such a way that Golgi apparatus vesicles of treated cells appear to be different than controls and seem to coalesce as anomalous partial cell plates. During recovery, an apparently normal progression of cytokinesis is observed and abnormal portions of the cell plate are retained. Nuclear constrictions are observed frequently during recovery as a result of temporal alterations in cytokinesis. Our results show that 2,6-dichlorobenzo-nitrile induces anomalous and/or incomplete cell plates, which might be caused by an altered function of Golgi apparatus. 相似文献

Summary The structural and functional continuum of the plant apoplast is the first site of contact with a pathogen and plays a crucial role in initiation and coordination of many defence responses. In this paper, we present an overview of the involvement of the plant apoplast in plant-pathogen interactions. The process of infection of French bean (Phaseolus vulgaris L.) plants byColletotrichum lindemuthianum is analysed. The ultrastructural features of plant defence responses to fungal infection are then compared with those observed in plants or cell suspensions treated with various elicitors. Changes in cell walls and in whole plant cells responding to infection seem to be highly similar in all systems used. Model systems of French bean and white lupin (Lupinus albus L.) are then utilised to provide some biochemical characteristics of oxidative reactions in the apoplast evoked by elicitor treatment. The species specificity of various mechanisms generating reactive oxygen species is discussed, and some details of pH-dependent H2O2-generating activity of peroxidases are demonstrated. As its exocellular nature is an important feature of the oxidative burst, the major consequence of this event, i.e., the oxidative cross-linking of wall components during the papilla formation and strengthening of the walls, is analysed. Finally, the possible involvement of other wall-associated and developmentally regulated H2O2-generating mechanisms, like amine and oxalate oxidases, in plant defence is demonstrated.
