Data on a unique phenomenon of annual involution and neogenesis of thymus gland in hibernating animals are reviewed. In accordance with morphological findings, the annual thymus involution in hibernating animals is close to the age-dependent thymus involution occurring in all mammals once in a lifetime. In opposite, thymus involution in hibernating animals is totally different from the accidental involution. During hibernation, the thymus tissue is substituted by the brown fat tissue. In the spring, thymus gland neogenesis stats with intensive growth of epithelial tissue followed by lymphocyte infiltration and exhaustion of brown tissue. Morphological changes in the thymus gland within the annual cycle were compared with seasonal dynamics of structural and functional changes in peripheral lymphoid organs (spleen, lymphoglandular, peritoneal fluid). A general regularity was observed involving a decreased functional activity of immune cells in autumn, its sharp depression during winter hibernation, and obvious increase in summer with the onset of a season of animal activity. It is supposed that a sharp increase in the tumor necrosis factor (TNF) production observed during short-term awakenings in winter may serve an important link in this unique immune adaptation mechanism. The season changes in cellular TNF secretion suggest a mobilization of protective resources in hibernating animals in autumn and winter, i.e. in seasons when the thymus gland activity is depressed. The annual involution of thymus gland cannot be related to droppings in the environmental or body temperatures, as it comes long before their fall. Additionally, it is not related to ageing, as it occurs already in young hibernating animals. The role of hormones, including melatonine and corticosteroids, in mechanisms regulating thymus gland involution in hibernating animals is discussed.  相似文献

The spleen and thymus have been studied macro- and microscopically in rats (180-200 g body mass) on the 1st, 3d, 5th, 7th, 14th and 28th days of adaptation to a decreased atmospheric pressure in the altitude chamber corresponding to lifting to 5,000 and 7,500 m (after a preliminary gradual acclimatization) and on the 14th, 28th, 42d, 56th days of readaptation. A relative mass of the organs, the white pulp section area--the transversal section area of the spleen ratio, the summation section area of its lymph nodules have been estimated. In the thymus the cortico-medullary index (CMI) has been defined. A relative mass of the spleen increases during the first week of hypoxia, and during adaptation period it somewhat decreases and stabilizes, remaining higher than in the control. At the altitude of 5,000 m the cross section area of the lymph nodules decreases by 17% on the 28th adaptation day and at the altitude of 7,500 m--by 27% beginning from the 14th up to the 28th adaptation days. In the thymus the CMI, after some decrease during the first days of hypoxia at the altitude of 5,000 m, increases and normalizes on the 28th adaptation day, and at the altitude of 7,500 m stabilizes on the 14th - 28th days of hypoxia. When the rats are at the altitudes of 5,000 and 7,500 m the thymus lymphoid tissue reacts more quickly to the hypoxia effect and much sooner normalizes during the readaptation period than does the white pulp of the spleen. The main changes in the lymphoid tissue of the spleen and thymus take place on the 7th - 28th days of hypoxia.  相似文献

By means of histological and morphological methods reaction of mast cells has been studied in the thymus and inguinal lymph nodes of mature non-inbred white male rats, subjected to systematic physical loadings (daily swimming) with increasing time from 5 up to 100 min during 5 months. Morphological changes in the organs studied and manifestation of the mast cell reaction essentially depend on the degree of the animals' adaptation to the loading. In the animals adapted to swimming, decreasing area, occupied by the connective tissue elements, in comparison to that in the control--increasing cortical area, increasing number of lymphoid cells, decreasing number of the mast cells in the inguinal lymph nodes--are noted. When the adaptation of the animals to the loading is insufficient, outgrowth of the connective tissue elements, decreasing cortical zone, impoverishment of the parenchyma with lymphocytes occur. The number of the mast cells increases, many of them are at the state of degranulation.  相似文献

By means of histological and morphometrical methods normal age involution of the thymus has been studied, as well as its changes under conditions of dosed physical loadings. The experiment has been performed in 92 non-inbred white male rats. At adaptation of the organism to the loadings, involution of the gland decelerates, and at an insufficient adaptation--accelerates. This is, probably, the cause of decreasing protective forces of the organism.  相似文献

Summary A histological study has been made of the thymus in mice during acute involution and regeneration following administration of hydrocortisone. The cortex undergoes remarkable changes in the microscopic structure during involution and regeneration. During involution the lymphocytes in the cortex rapidly decrease and are removed. Then a rapid replacement of lymphocytes occurs during regeneration. On the basis of formation and repopulation of lymphocytes the regenerative process of the cortex is divided into seven phases. The reconstitution of the cortex proceeds more rapidly in females than in males. Newly formed lymphocytes take origin from the mesenchymal cells in the cortex. Such mesenchymal cells become distinguishable from epithelial reticular cells during involution. They appear to engulf destroyed lymphocytes and debris during involution and then transform into immature lymphoid cells during early regeneration. The findings may support the recent reutilization concept that destroyed lymphocytes are phagocytized and reutilized by reticular cells in heteroplastic differentiation into immature lymphoid cells. In the cortex PAS-positive sudanophilic cells which are derived from the perivascular and subcapsular connective tissue appear with involutionary changes. They become gradually reduced again with progress of the regeneration of the cortex. During involution the medulla are temporarily filled with lymphocytes migrated from the cortex. The epithelial reticular cells in the medulla are found grouped in cords or clumps in the severely involuted thymus. In the medulla there are two types of PAS-positive epithelial reticular cells; one contains a large, colloid-like, PAS-positive inclusion within the cytoplasm and the other has cytoplasm diffusely filled with PAS-positive substance. During involution and early regeneration, the former type increases while the other shows almost no significant changes. Hassall's corpuscles somewhat increase in frequency during involution and early regeneration.  相似文献

Age-related thymus involution results in decreased T-cell production, contributing to increased susceptibility to pathogens and reduced vaccine responsiveness. Elucidating mechanisms underlying thymus involution will inform strategies to restore thymopoiesis with age. The thymus is colonized by circulating bone marrow (BM)-derived thymus seeding progenitors (TSPs) that differentiate into early T-cell progenitors (ETPs). We find that ETP cellularity declines as early as 3 months (3MO) of age in mice. This initial ETP reduction could reflect changes in thymic stromal niches and/or pre-thymic progenitors. Using a multicongenic progenitor transfer approach, we demonstrate that the number of functional TSP/ETP niches does not diminish with age. Instead, the number of pre-thymic lymphoid progenitors in the BM and blood is substantially reduced by 3MO, although their intrinsic ability to seed and differentiate in the thymus is maintained. Additionally, Notch signaling in BM lymphoid progenitors and in ETPs diminishes by 3MO, suggesting reduced niche quality in the BM and thymus contribute to the early decline in ETPs. Together, these findings indicate that diminished BM lymphopoiesis and thymic stromal support contribute to an initial reduction in ETPs in young adulthood, setting the stage for progressive age-associated thymus involution.  相似文献

Structural-functional organization of the lymphoid organs and functional state of the adrenals have been studied in animals, subjected to cold in early postnatal period, as well as changes of the parameters mentioned to a short and prolonged cooling in mature rats. For the animals increase in the thymus mass and in reproduction rate of lymphocytes in the thymus, spleen and lymph nodes is specific against the background of high corticosteroid secretion. When the control animals are kept in cold for a long time, after the phase of an acute stress, accompanied with hypercorticism and a pronounced lymphatic effect, during the period of an increased cold stability, the high secretion of glucocorticoid hormones is accompanied with a certain activation of thymus-dependent zones in the peripheral lymphoid organs. In the mature rats, subjected to cold at early ontogenesis both stress-reaction to cooling and rearrangement in the regulatory systems studied does not develop at adaptation to cold.  相似文献

In the experiment performed on 70 noninbred white male rats by means of histological and morphometric techniques construction and cell composition of the thymus, spleen and inguinal lymph nodes have been studied under the influence of systematic physical (swimming) loads. They have been applied under conditions of a pharmacological correction (administration of leacadin) and without it. In the animals not given leacadin, the physical loads result in decrease of the immune function in all the organs investigated. This is evident from decreasing size of the thymus, outgrowth of adipose tissue in it, drop in amount of lymphoid nodules++ in the spleen, decline of contents of lymphocytes in all the organs, and in the cords of the inguinal lymph nodes--decline of plasma cells. Application of leacadin prevents appearance of unfavourable changes.  相似文献

Snell's pituitary dwarf mice (dw) were used for studies on the relationship between hypophysis and lymphoid organs. The age-dependent changes of thymus or spleen weights of dwarf mice were compared with those of normal littermates. The suppression of growth of the thymus or spleen in dwarf mice was recognized at 5th day of age. Although involution of the thymus varied among animals, a strong positive correlation was demonstrated between relative thymus weight and body weight in 30 approximately 40 days old dwarf mice. Lymphoid organs of dwarf mice were reconstituted by injection of growth hormone and or thyroxin. Relative thymus weight significantly increased in dwarf mice when the treatment with growth hormone started at 7 days of age, but the same treatment at 3 months of age did not show any effect on the increment of relative thymus weight. On the other hand, the antibody-forming capacitiy against sheep erythrocytes of dwarf mice was significantly increased even when the treatment with growth hormone was started at 3 months of age. A marked increase in the number of lymphoid cells in dwarf mice was observed by treatment with thyroxin, even if treatment was started either at 7 days or 3 months of age. Similar changes were also obtained in the antibody-forming capacity.  相似文献

Progesteron and estradiol, administered in doses equivalent to those used for therapy caused a marked transient reduction of the thymus mass, but did not affect the cellularity of other lymphoid organs. Humoral and cellular immune response of the hormone-treated mice was normal at the time of thymus involution. The same was true for the stem-cell differentiation capacity. The remaining thymus cells after hormone treatment showed increased DNA-synthesis.  相似文献

With the exception of Agnatha, fish possess the functional equivalent of the thymus gland found in higher vertebrates. As in other vertebrates, this gland originates from the pharyngeal pouches and ontogenically is the first lymphoid organ to be infiltrated with lymphoid cells. Histology of the structure may differ from one species to another but the cellular component is basically similar. The (paired) gland is surrounded by an epithelial capsule. Within the gland a framework of reticulo-epithelial cells supports the lymphocytes. The age-related involution process, which characterizes the thymus of higher vertebrates, does not necessarily occur in fish. Nevertheless, thymus growth and function may be modulated by those factors that induce its involution such as aging, season, sexual maturity, and stress. The major role played by the thymus in the immune response of higher vertebrates is presumed to occur in fish. Thymus-derived cell dependent immune reactions have been demonstrated in fish. The cells that mediate these functions are designated as T-like cells. So far, cell surface markers equivalent to those of mammalian T lymphocytes have not been characterized. The T lymphocyte specificities are supposed to be acquired within or via the thymic microenvironment. Unfortunately, there is limited data concerned with the cytological and physiological basis of the maturation of thymus-derived cells. Direct involvement of the fish thymus in defense mechanisms has not been investigated extensively. The gland appears to be weakly protected because of its superficial location and is easily exposed to pathogens. Neoplasia is the main pathologic condition reported in the thymus of fish, with little else having been published regarding thymic pathology.  相似文献

Diabetes in man and rats is accompanied by increased levels of acid hydrolases in plasma and urine, the increase being reversed by insulin. In investigating possible tissue sources of the additional enzymes, the unexpected observation was made that streptozotocin-induced diabetes in rats is accompanied by substantial involution of the thymus and the failure of the spleen to grow at its normal rate. One week after streptozotocin treatment, the average thymus weight of diabetic rats was 30% less than that of age-matched control rats. Eight weeks after the induction of diabetes, only 10 to 20% of the thymus remained. At that time, the spleen weight was only 50% that of age-matched controls. The organ weight changes were paralleled by changes in the organ levels of acid hydrolases. The changes were all reversed by insulin treatment. Since it appeared possible that corticosteroids might be the mediators of the streptozotocin-induced changes, the effects of streptozotocin were determined in adrenalectomized rats. The decreases in thymic and splenic weights again were observed, as well as decreases in most organ hydrolases and increases in most plasma hydrolases. Clearly, the principal streptozotocin effects are not adrenal dependent. Although analyses of hexosaminidase A and B levels in various tissues and plasma suggested that the thymus and spleen may contribute to the additional hydrolases found in diabetic plasma, other factors must also be involved.  相似文献

Diabetes is chronic disease that is accompanied by a rapid thymus involution. To investigate the factors responsible for thymic involution in a model of STZ-induced diabetes, mice were injected with STZ alone or in combination with the cyclooxygenase 2 inhibitor indomethacin (INDO). Thymus weight, glycemia and serum corticosterone were measured, and apoptosis in thymus and thymocyte cultures was analyzed by flow cytometry. Although earlier studies report that streptozotocin (STZ) is toxic to lymphoid tissues, in our experiments even massive doses of STZ did not negatively affect thymocyte cultures. Cultured thymocytes also seemed unaffected by high glucose concentrations, even after 24 h of exposure. Administration of INDO concomitantly with STZ reduced thymic involution but did not prevent the onset of hyperglycemia or reduce established hyperglycemia. When INDO was given before STZ, the same degree of thymic involution occurred; however, hyperglycemia was reduced, although normoglycemia was not restored. INDO also reduced serum corticosterone. Because thymocytes are known to be sensitive to glucocorticoids, this finding suggests that cyclooxygenase 2 inhibition may retard thymic involution by reducing serum glucocorticoids. In conclusion, our results show that STZ and hyperglycemia are not toxic to thymocytes and that cyclooxygenase 2-mediated mechanisms are involved in thymic involution during diabetes.  相似文献

Disability and mortality as consequence of Chagas disease is enormous in South America. Recently, the success of the trypanocidal treatment with benznidazole, the only available drug, has been associated with the host immune response. In the current study, the impact of benznidazole administration immediately after the experimental infection with Trypanosoma cruzi was evaluated in the main lymphocyte populations in lymphoid organs. Untreated mice displayed enlargement of spleen and lymph node related to the increased frequency of T and B lymphocytes, respectively. An intense thymus involution with the depletion of CD4(+)CD8(+) double-positive thymocytes also occurred. Benznidazole treatment led to a partial reversion of the spleen and lymph node enlargement related to changes in the frequency of lymphocyte subsets due to infection. Prevention of thymus involution was achieved, with the profile of thymocyte subsets similar to that of non-infected mice. The parasitic load at the onset of T. cruzi infection seems critical to trigger immune system activation.  相似文献

Thymic involution is a slow physiological process than can be accelerated by some pathological or experimental conditions. In this work we kept male mice under low temperatures in order to observe whether or not thymic involution would be promoted. For this purpose, we carried out a histometrical analysis of the thymus and observed that there was a decrease in both cortical and medullary regions of the thymic lobules. These changes paralleled a loss in the absolute and relative weights of the thymus. It was concluded that low temperatures induce thymic involution even in nonhibernating animals.  相似文献

With the help of antibodies-containing serum reacting with the thymus reticulum epithelial cells components by immunofluorescence method the thymus parenchimal tissue in mice with functional and pathological involution has been detected electively. In spite of large thymus changes in hairless mice with the mutation of gene hrrhy in 14-th chromosome of B10.R109/Y animals the basal cells antigen in epithelial reticulum has been preserved. It permits to estimate thymus involution level of this organ. Low lymphocyte content in the thymus of C57BL/6 mice is accompanied by total decamouflage of the epithelium. The functional thymus involution of pregnant mice is characterized by the luminescence of large number of epithelial cells, the restoration of organ after the delivery--by their few number. The elective detection of thymus epithelium many serve as additional test for the estimation of functional and pathological involution level.  相似文献

The thymus plays an irreplaceable role as a primary lymphoid organ. However, the complicate processes of its development and involution are incompletely understood. Accumulating evidence indicates that non-coding RNAs play key roles in the regulation of biological development. At present, the studies of the circRNA profiles and of circRNA-associated competing endogenous RNAs (ceRNAs) in the thymus are still scarce. Here, deep-RNA sequencing was used to study the biological mechanisms underlying the development process (from 2-week-old to 6-week-old) and the recession process (from 6-week-old to 3-month-old) of the mouse thymus. It was found that 196 circRNAs, 233 miRNAs and 3807 mRNAs were significantly dysregulated. The circRNA-associated ceRNA networks were constructed in the mouse thymus, which were mainly involved in early embryonic development and the proliferation and division of T cells. Taken together, these results elucidated the regulatory roles of ceRNAs in the development and involution processes of the mouse thymus.  相似文献

The stromal cells of the thymus of sham-irradiated and sublethal fission neutron-irradiated CBA/H mice were analyzed with immunohistology, using monoclonal antibodies directed to I-A and H-2K antigens as well as specific determinants for cortical and medullary stromal elements. In the control thymuses, I-A expression in the thymus shows a reticular staining pattern in the cortex and a confluent staining pattern in the medulla. In contrast, H-2K expression is mainly confluently located in the medulla. Whole body irradiation with 2.5 Gy fission neutrons reduces within 24 hr the cortex to a rim of vacuolized "nurse cell-like" epithelial cells, largely depleted of lymphoid cells. The localization of I-A antigens changes in the cortex and I-A determinants are no longer associated with or localized on epithelial reticular cells. Medullary stromal cells, however, are more or less unaffected. A high rate of phagocytosis is observed during the first 3 days after irradiation. About 5 days after irradiation, the thymus becomes highly vascularized and lymphoid cells repopulate the cortex. The repopulation of the thymic cortex coincides with the appearance of a bright H-2K expression in the cortex which is associated with both stromal cells as well as lymphoid blasts. During the regeneration of the thymus, the thymic stromal architecture is restored before the expression of cell surface-associated reticular MHC staining patterns. The observed sequential changes in the thymic microenvironment are related to the lymphoid repopulation of the thymus.  相似文献

Summary Influences of pregnancy and lactation on the thymus were quantitatively and microscopically studied in the mouse. Thytnic involution due to pregnancy occurs in later pregnancy and reaches a maximum at parturition. After parturition the involuted thymus regenerates, but lactation has an inhibitory influence on the regeneration. The possible significance of the thymic changes is discussed particularly in relation to the adrenocortical activity. Histologically the cortex exhibits prominent alterations during the involution and regeneration. The pattern of depletion and repopulation of lymphocytes in the cortex is similar to that in other types of acute involution. The medulla also undergoes microscopic changes which are revealed particularly by the histometric examination of its components.  相似文献

Effect of glucocorticoids on proliferation, death and migration of T-lymphocytes was studied by mathematical methods. A model of the interaction between hormone and thymus T1-lymphocytes is proposed which imitates involution of lymphoid tissue under stress. There were constructed models of migrations of free and radioactive chromium labeled lymphocytes in lymphoid and non-lymphoid organs, blood and lymph at arbitrary concentration of glucocorticoids in the blood. Possible mechanisms of immunodepression due to increased hormone concentrations in the organism are discussed.  相似文献
